背景技术

“营养组合物”意为满足对象至少部分营养需求的物质或制剂。在本公开通篇可互换地使用的术语“营养物”、“营养配方”、“肠内营养物”、“营养组合物”和“营养添加剂”,指的是肠内配方、口服配方、用于婴儿的配方、用于小儿对象的配方、用于儿童的配方、成长乳(growing-up milk)和/或用于成人(诸如哺乳的或怀孕的女性)的配方的液体、粉末、凝胶、糊剂、固体、浓缩物、悬浮液或即食形式。
术语“肠内的”意为通过或在胃肠道或消化道中。“肠内施用”包括口服喂食、胃内喂食、经幽门施用或在消化道中的任何其它施用。
“小儿对象”意为小于13岁年龄的人。在一些实施方案中,小儿对象指的是小于8岁的人对象。在其它实施方案中,小儿对象指的是年龄在1岁和6岁之间的人对象。在还进一步的实施方案中,小儿对象指的是年龄在6岁和12岁之间的人对象。因此,小儿对象包括婴儿和儿童。

“婴儿配方”意为满足婴儿的至少部分营养需求的组合物。在美国,婴儿配方的内容物由联邦法规规定,在21 C.F.R.第100、106和107部分阐述。这些法规规定了试图模仿人母乳的营养和其它性能的大量营养物、维生素、矿物质和其它成分水平。
术语“成长乳”指的是意在为了支持年龄在约1岁和约6岁之间的儿童的正常成长和发育,用作多样化饮食的一部分的营养组合物的广义分类。

通过定义,对于儿童“营养完整的”的营养组合物将提供对儿童成长所需的定性地和定量地足够量的碳水化合物、脂质、必需脂肪酸、蛋白质、必需氨基酸、条件必需氨基酸、维生素、矿物质和能量。
如对营养物应用的,术语“必需的”指的是不能通过人体合成足够用于正常成长和维持健康的量的任何营养物,并且其因此,必须通过膳食提供。应用于营养物的术语“条件必需的”意为当足够量的前体化合物不能通过身体内源合成出现时,在这种条件下营养物必须通过膳食提供。

本公开提供了包括至少一种游离氨基酸的营养组合物。在某些实施方案中,本文中提供的组合物,在通过对象摄取组合物后,促进或诱导了饱腹感。同样地,组合物可用于健康的体重增加、防止或减少超重、肥胖和/或糖尿病的发生。例如,当对象是婴儿时,本组合物可用于促进健康的体重增加,并且建立在以后生活中减少婴儿超重、肥胖或糖尿病风险的终生膳食习惯。在某些实施方案中,营养组合物包括蛋白质源、脂肪源、碳水化合物源和至少一种核苷酸,其中蛋白质源包括至少一种游离氨基酸,其中至少一种游离氨基酸是谷氨酸、牛磺酸、谷氨酰胺、丙氨酸或其任意组合。在更特别的实施方案中,组合物包括谷氨酸、牛磺酸、谷氨酰胺和丙氨酸。

在其它实施方案中,组合物包括每100kcal约1mg至约25mg、每100kcal 约2mg至约15mg或每100kcal约3mg至约13mg的谷氨酰胺。在某些实施方案中,组合物包括每100kcal约1mg至约25mg、每100kcal 约2mg至约15mg或每100kcal 约5mg至约10mg的牛磺酸。在其它特别的实施方案中,组合物包括每100kcal 约0.25mg至约20mg、每100kcal 约0.5mg至约10mg或每100kcal 约1mg至约5mg的丙氨酸。
蛋白质摄取对对象成长的效果还取决于对象的消化能力。例如,在出生的第一周,婴儿的消化能力没有达到其完全能力。氨基酸在未成熟的消化系统中可以更容易地被吸收,从而促进了饱腹感,并且因此促进在这些较早的周里更健康的体重和体重增加率。此外,喂食游离氨基酸可以增加吸收至血清中的比例,并且因此减少总配方摄取,而对身体组成和长期健康提供有益的效果。最后,婴儿可以在生命早期学习健康的热量摄取,并且因此提供了终生的健康进食习惯。
在本文中提供的营养组合物可以任选地进一步包括至少一种氨基酸,并且更特别地,至少一种核糖核苷酸,诸如5’-核糖核苷酸。没有被任何特定理论束缚,相信包含5’-核糖核苷酸可以增强本组合物的饱腹感促进效果。例如,5’-核糖核苷酸已经显示出是对GPCRT1R1+T1R3鲜味受体(umami receptor )的变构激动剂(Xu, H. 等, PNAS, 2004, 101:14258-14263)。如果鲜味受体信号是观察到饱腹感效果的原因,则5’-核糖核苷酸可以增强该效果。在特别的实施方案中,5’-核糖核苷酸可以是单磷酸肌苷、单磷酸鸟苷或其组合。更特别地,5’-核糖核苷酸可以是在10:1至1:10的比例之间,更通常地在接近1:1的比例的单磷酸肌苷、单磷酸鸟苷的组合。由Sucrogen Bioethanol出售的Ribotide®(CAS No.80702-47-2)是包括单磷酸肌苷和单磷酸鸟苷(特别地肌苷酸二钠和鸟苷酸二钠)的可商购获得的食品成分,并且可用做本组合物中的5’-核糖核苷酸的来源。纯化合物还可以从例如Yamasa Corporation,Biochemicals Division获得。5’-核糖核苷酸二钠增味剂(flavorenhancer)还通过E数字E635 表征,鸟苷酸二钠(E626)和肌苷酸(E630)二钠的组合。这些产品可以任选地来源于酵母自溶物、鱼或肉。
美国营养科学协会的生命科学研究部关于婴儿配方营养需求的评价(The LifeSciences Research Office (LSRO), American Societies for Nutritional SciencesAssessment of Nutrient Requirements for Infant Formulas (J Nutr 1998; 128(Supp):2059S-2298S)推荐,游离核苷酸的水平,包括在婴儿配方中作为核苷酸前体的可获得的核苷、核酸(DNA和RNA),被限制在每100 kcal最多为16mg(是它们在人乳中上限的相似值)。在2006年12月22日有关婴儿配方和后续配方的欧盟委员会指令 2006/141/EC和修改的指令1999/21/EC中规定可以添加胞苷、尿苷、腺苷、鸟苷和肌苷的5’-单磷酸,并且核苷酸的总浓度应当不超过每100kcal 5mg。因此,在一些实施方案中,5’-核糖核苷酸的量在每100kcal 约0.05 mg至约16mg、每100 kcal 约0.1mg至约5mg或每100kcal约0.2mg至约1.5mg之间范围。得到最优的核糖核苷酸水平的另一个途径是作为谷氨酸水平的重量百分比,例如谷氨酸的约0.1%-10%,或谷氨酸的约1%-5%。
在特别的实施方案中,营养组合物包括蛋白质源、脂肪源、碳水化合物源和至少一种核苷酸,其中蛋白质源包括全蛋白源和包括谷氨酸、牛磺酸、谷氨酰胺和丙氨酸的游离氨基酸的混合物。在一些实施方案中,游离氨基酸的量可以是在本文中描述的量。例如,营养组合物可以包括每100kcal 约2mg至约180mg的谷氨酸,每100kcal 约1mg至约25mg的谷氨酰胺,每100kcal 约1mg至25mg 的牛磺酸和每100kcal 约0.25mg至约20mg 的丙氨酸。在某些实施方案中,前述组合物可以进一步包括至少一种5’-核糖核苷酸,诸如单磷酸肌苷、单磷酸鸟苷或其混合物。

因此,本公开还提供了在对象中促进饱腹感的方法,其包括对对象肠内施用按本文描述的组合物。组合物可以是营养完整的,使得其可以是对象的唯一营养来源。例如,在某些实施方案中,组合物是儿童营养产品,诸如婴儿配方或成长乳。本公开进一步提供用于在对象中减少超重、肥胖和/或糖尿病发生的方法,其包括对对象施用在本文中描述的营养组合物。对象可以是婴儿、儿童或甚至成年对象,但是在特别的实施方案中,对象是婴儿。例如,在某些实施方案中,对婴儿施用本公开的婴儿配方,将减少在以后生活中肥胖和/或糖尿病的发生。

在一些实施方案中,本公开提供了设计用于1-3岁和/或4-6岁年龄儿童的基于强化乳的成长乳,其中成长乳支持成长和发育和终生健康。在一些实施方案中,本公开提供了适合用于年龄范围在0至12个月、或0至3个月、0至6个月或6至12个月的婴儿的婴儿配方。
除了至少一种氨基酸外,本营养组合物的蛋白质源还可以包括本领域通常使用的其它蛋白质源,例如,脱脂乳、乳清蛋白、酪蛋白、大豆蛋白、全蛋白、水解蛋白质等。可用于实施本公开的牛乳蛋白质源包括,但不限于,乳蛋白粉、乳蛋白浓缩物、乳蛋白分离物、脱脂乳固体、脱脂乳、脱脂干乳、乳清蛋白、乳清蛋白分离物、乳清蛋白浓缩物、甜乳清、酸乳清、酪蛋白、酸酪蛋白、酪蛋白酸盐(例如,酪蛋白酸钠、酪蛋白酸钠钙、酪蛋白酸钙)和其任意组合。在还另一个实施方案中,蛋白质源可以添加含谷氨酰胺肽。
在某些实施方案中,除了至少一种氨基酸,蛋白质源还包括至少一种全蛋白源。更特别地,在某些实施方案中,蛋白质源不包括水解蛋白或部分水解蛋白。示例性的全蛋白源包括酪蛋白、乳清蛋白(包括乳清蛋白分离物或乳清蛋白浓缩物)或大豆蛋白。在特别的实施方案中,全蛋白源包括酪蛋白、乳清蛋白或其组合,而在其它实施方案中,全蛋白源包括大豆蛋白。

碳水化合物源可以是在本领域中任何使用的,例如,乳糖、葡萄糖、果糖、玉米糖浆固体、麦芽糊精、蔗糖、淀粉、大米糖浆固体等。在营养组合物中的碳水化合物的量通常可以在约5g/100 kcal和约25g/100 kcal之间变化。
在一个实施方案中,营养组合物可以含有一种或多种益生菌。在该实施方案中任何本领域中已知的益生菌是可接受的。在特别的实施方案中,益生菌可以选自任何乳酸杆菌(Lactobacillus)种类,鼠李糖乳杆菌( Lactobacillus rhamnosus)GG(例如,ATCC号53103)、双歧杆菌(Bifidobacterium)种类,长双歧杆菌(Bifidobacterium longum)(例如,AH1205或AH1206)和动物双歧杆菌亚种(Bifidobacterium animalis subsp. lactis)BB-12(DSM号10140)、婴儿双歧杆菌(Bifidobacterium infantis)(例如35624)或其任意组合。
如果被包括在组合物中,益生菌的量可以在每克营养组合物约1 x 104至约1 x1012 个集落形成单位(cfu)变化。在另一个实施方案中,益生菌的量可以在每克营养组合物约1 x 106至约1 x 1012 cfu变化。在还另一个实施方案中,益生菌的量可以在每克营养组合物约1 x 106至约1 x 109 cfu,或在每克营养组合物约1 x 109至约1 x 1012 cfu变化。在还另一个实施方案中,益生菌的量可以为每克营养组合物至少约1 x 106 cfu。

更具体地,可用于本公开的益生元可以包括聚葡萄糖、聚葡萄糖粉、乳果糖、乳果寡糖、棉子糖、寡聚葡萄糖、菊粉、寡聚果糖、寡聚异麦芽糖、大豆寡聚糖、乳果寡糖、寡聚木糖、寡聚壳糖、寡聚甘露糖、 寡聚阿拉伯糖(aribino-oligosaccharide)、唾液酸寡糖、寡聚岩藻糖(fuco-oligosaccharide)、寡聚半乳糖和龙胆寡糖(gentio-oligosaccharide)。
在一个实施方案中,在营养组合物中存在的益生元总量可以为约1.0 g/L组合物至约10.0 g/L组合物。例如,在一些实施方案中,聚葡萄糖(PDX)可以以约1.0g/L至10.0g/L在营养组合物中包括。在其它实施方案中,PDX的量为约2.0g/L至约8.0g/L。
在某些实施方案中,至少20%的益生元可以包括低聚半乳糖(GOS)、(PDX)或其混合物。 在实施方案中,PDX和GOS具有在约9:1和1:9之间的PDX:GOS比例。在另一实施方案中,PDX:GOS的比例可以为约5:1至1:5。在还另一实施方案中,PDX:GOS的比例可以在约1:3和约1:3之间。在进一步的更特别的实施方案中,比例可以是约1:1或4:1。在其它实施方案中,PDX:GOS组合的量可以在约 2.0 g/L和8.0 g/L之间。在特别的实施方案中,PDX:GOS组合的量可以为约PDX 2 g/L 和GOS 2 g/L 。至少20%益生元可以包括低聚半乳糖(“GOS”)、聚葡糖或其混合物。在实施方案中,在营养组合物中GOS和/或聚葡萄糖各自的量可以在约1.0g/L至约4.0 g/L的范围内。
本公开的营养组合物可以含有包括二十二碳六烯酸的长链多不饱和脂肪酸(LCPUFA)的来源。其它合适的LCPUFA包括,但不限于,α-亚麻油酸、γ-亚麻油酸、亚麻油酸、亚麻酸、二十二碳六烯酸(DHA)、二十碳五烯酸(EPA)和花生四烯酸(ARA)。
在实施方案中,特别地如果营养组合物是婴儿配方,使用DHA和ARA两者添加营养组合物。在该实施方案中,ARA:DHA的重量比例可以在约1:3和约9:1之间。在特别的实施方案中,ARA:DHA的比例是从约1:2至约4:1。

此外,β-葡聚糖是良好耐受的,并且不会在小儿对象中产生或导致过度气体、腹胀、气胀或腹泻。向用于小儿对象的营养组合物,诸如儿童配方、成长乳或其它儿童营养产品的β-葡聚糖的添加,将通过针对入侵病原体增加抵抗力改善对象的免疫应答,并且因此维持或改善整体健康。
在实施方案中,本公开的营养组合物包括胆碱。胆碱是对细胞正常功能必要的营养物。它是膜磷脂质的前体,并且其加速了乙酰胆碱(涉及记忆存储的神经递质)的合成和释放。此外,虽然不希望受到这个或任何其它理论束缚,相信膳食的胆碱和二十二碳六烯酸(DHA)协同地作用以促进磷脂酰胆碱的生物合成并且因此促进在人对象中的突触发生。此外,胆碱和DHA可以展现出促进树突棘形成的协同效应,其对建立的树突连接的维持是重要的。在一些实施方案中,本公开的一种或多种营养组合物包括每份约40mg胆碱至每8 oz.份约100mg。

还可以将一种或多种维生素和/或矿物质以足够提供对象的每日营养需求的量添加至营养组合物中。本领域普通技术人员理解维生素和矿物质需要将是多种多样的,例如,基于对象的年龄。例如,婴儿可以具有与年龄在1岁和13岁之间的儿童不同的维生素和矿物质需求。因此,实施方案不意图将营养组合物限制在特别的年龄组,但是,相反地,提供了可接受的维生素和矿物质组分的范围。

在其它实施方案中,组合物可以任选地包括,但不限于一种或多种以下矿物质或其衍生物:硼、钙、醋酸钙、葡萄糖酸钙、氯化钙、乳酸钙、磷酸钙、硫酸钙、氯化物、铬、氯化铬、吡啶甲酸铬(chromium picolonate)、铜、硫酸铜、葡萄糖酸铜、硫酸铜、氟化物、铁、羰基铁、三价铁、富马酸亚铁、正磷酸铁、铁研制剂(iron trituration)、多糖铁、碘化物、碘、镁、碳酸镁、氢氧化镁、氧化镁、硬脂酸镁、硫酸镁、锰、钼、磷、钾、磷酸钾、碘化钾、氯化钾、醋酸钾、硒、硫、钠、多库酯钠、氯化钠、硒酸钠、钼酸钠、锌、氧化锌、硫酸锌及其混合物。矿物质化合物的非限制性示例性衍生物包括任何矿物质化合物的盐、碱性盐、酯和螯合物。
可以将矿物质以盐的形式添加至成长乳或其它儿童营养组合物中,所述盐诸如磷酸钙、甘油磷酸钙、柠檬酸钠、氯化钾、磷酸钾、磷酸镁、硫酸亚铁、硫酸锌、硫酸铜、硫酸锰和亚硒酸钠。可以添加本领域中已知的额外的维生素和矿物质。
在实施方案中,每份维生素A、C和E、锌、铁、碘、硒和胆碱,儿童营养组合物可以含有对于任何给定国家最大膳食推荐的约10%至约50%,或对于一组国家平均膳食推荐的约10%至约50%。在另一个方案中,每份B族维生素,儿童营养组合物可以提供对于任何给定国家最大膳食推荐的约10-30%,或对于一组国家平均膳食推荐的约10-30%。在还另一个实施方案中,在儿童的营养产品中的维生素D、钙、镁、磷和钾的水平可以对应在乳中发现的平均水平。在其它实施方案中,儿童营养组合物中的每份其它营养物可以以对于任何给定国家最大膳食推荐的约20%,或对于一组国家的平均膳食推荐的约20%存在。
本公开的儿童营养组合物可以任选地包括一种或多种以下调味剂,包括,但不限于,风味提取物、挥发油、可可粉或巧克力调味料、花生酱调味料、饼干碎屑、香草或任何可商购获得的调味料。可用的调味料的实例包括,但不限于,纯茴芹提取物、仿制香蕉提取物、仿制樱桃提取物、巧克力提取物、纯柠檬提取物、纯柑橘提取物、纯薄荷提取物、蜂蜜、仿制菠萝提取物、仿制朗姆酒提取物、仿制草莓提取物或香草提取物;或挥发油,诸如滇荆芥油、月桂油、佛手柑油、香柏油、樱桃油、肉桂油、丁香油或薄荷油;花生酱、巧克力调味料、香草饼干屑、奶油糖果、太妃糖及其混合物。调味剂的量可以根据使用的调味剂很大地变化。可以按照本领域已知的选择调味剂的类型和量。
本公开的营养组合物可以任选地包括一种或多种乳化剂,其可以被添加用于最终产品的稳定性。合适的乳化剂的实例包括,但不限于,磷脂酰胆碱(例如,来自蛋或大豆),α乳清蛋白和/或单-和二-甘油酯,和其混合物。其它乳化剂对技术人员是显而易见的并且某种程度上,合适的一种或多种乳化剂的选择取决于配方和终产品。
本公开的营养组合物可以任选地包括一种或多种还可以添加以延长产品保质期的防腐剂。合适的防腐剂包括,但不限于,山梨酸钾、山梨酸钠、苯甲酸钾、苯甲酸钠、EDTA钙二钠和其混合物。
本公开的营养组合物可以任选地包括一种或多种稳定剂。对在本公开的营养组合物实践中使用的合适的稳定剂包括,但不限于,阿拉伯胶、盖提胶、刺梧桐胶、黄蓍树胶、琼脂、帚叉藻聚糖、瓜尔豆胶、结冷胶、槐树豆胶、果胶、低甲氧基果胶、明胶、微晶纤维素、CMC(羧甲基纤维素钠)、甲基纤维素、羟丙基甲基纤维素、羟丙基纤维素、DATEM(单-和二甘油酯的二乙酰酒石酸酯)、葡聚糖、角叉菜胶和其混合物。
本公开的营养组合物可以提供最低限度的、部分的或全部的营养支持。组合物可以是营养添加剂或正餐替代品。组合物可以是,但不需要是营养完全的。在实施方案中,本公开的营养组合物是营养完全的并且含有脂质、糖、蛋白质、维生素和矿物质的合适的类型和量。脂质或脂肪的量通常地可以在约2g/100 kcal至约7g/100 kcal变化。蛋白质的量通常地可以在约1g/100 kcal至约5g/100 kcal变化。碳水化合物的量通常地可以在约8g/100kcal至约14g/100 kcal变化。
在一些实施方案中,本公开的营养组合物是成长乳。成长乳是意在用于超过1岁年龄(通常地1-6岁年龄)的儿童的基于强化乳的饮料。它们不是医疗食品并且不意在作为正餐替代品或添加剂以解决特别的营养缺陷。相反地,成长乳是按照作为对多样化膳食的补充剂的意图设计的,以提供儿童实现全部必需维生素和矿物质、大量营养物以及额外的功能膳食组分(诸如具有声称的健康促进性能的非必须营养物)的连续的每日摄取的额外保证。
根据本公开的婴儿配方或成长乳或其它营养组合物的确切组成可以按市场不同变化,取决于当地规范和目标群体的膳食摄入信息。在一些实施方案中,根据本公开的营养组合物由乳蛋白质源组成,诸如全乳或脱脂乳,额外添加糖和甜味剂以实现期望的感官性能,并且添加维生素和矿物质。脂肪组合物通常地来源于乳原料。总蛋白质可以以匹配人乳、牛乳或更低值的总蛋白质为目标。碳水化合物总量通常地以提供尽可能少的糖(诸如蔗糖或果糖)来实现可接受的味道为目标。通常地,将维生素A、钙和维生素D以达到地区牛奶的营养贡献的水平添加。另外的,在一些实施方案中,维生素和矿物质可以按每份提供约20%膳食参考摄入(DRI)或20%每日量值(DV)的水平添加。此外,营养值可以在市场之间变化,取决于目标群体确定的营养需求、原材料贡献和区域规范。
小儿对象可以是儿童或婴儿。例如,对象可以是年龄在0至3个月、约0至6个月、0至12个月、3至6个月或6至12个月范围的婴儿。对象可以可选地是年龄在1至13岁、1至6岁或1至3岁范围的儿童。在实施方案中,组合物可以在产前地、在婴儿期期间和在儿童期期间施用至小儿对象。
提供实施例以阐述本公开的营养组合物的一些实施方案,但是不应当被理解为对其有任何限制。考虑到本文中公开的营养组合物或方法的说明或实施,在本文权利要求范围内的其它实施方案将是对本领域技术人员显而易见的。说明书连同实施例被意图认为只是示例性的,本公开的范围和精神通过实施例之后的权利要求指示。
实施例
在该实施例中,在人乳中发现的最充足的4种氨基酸(谷氨酸、谷氨酰胺、牛磺酸和丙氨酸)的效果被通过将它们按每100 kcal 2.1g 蛋白质或每100 kcal 1.8g 蛋白质添加至婴儿配方进行检验。添加至配方中的游离氨基酸的水平按如下:每100 kcal配方,3 mg丙氨酸、23 g谷氨酸、7.6 g牛磺酸和9g 谷氨酰胺。灵长类新生儿接受这些配方4个月,在期间收集配方摄取量和代表它们成长的多种参数。灵长类新生儿的对照组接受没有添加氨基酸的相同配方。

Claims (15)
1.包括蛋白质源、脂肪源、碳水化合物源和至少一种核苷酸的营养组合物,
其中所述蛋白质源包括:(1)全蛋白源和(2)包括谷氨酸、牛磺酸、谷氨酰胺和丙氨酸的游离氨基酸的混合物,其中,所述谷氨酸的量为每100kcal 所述组合物约5 mg至约120mg,所述谷氨酰胺的量为每100kcal 所述组合物约1 mg至约25 mg,所述牛磺酸的量为每100kcal 所述组合物约1 mg至约25mg,并且所述丙氨酸的量为每100kcal 所述组合物约0.25 mg至约20mg。
2.权利要求1的所述营养组合物,其中所述谷氨酸作为谷氨酸盐提供,其中所述盐是钾盐、钙盐、镁盐、钠盐、铵盐或其组合。
3.权利要求2的所述营养组合物,其中谷氨酸盐不是钠盐。

6.权利要求1的所述营养组合物,其中所述至少一种核苷酸的量在每100 kcal约0.05mg至约16 mg、每100kcal 约0.1 mg至约5mg或每100kcal 约0.2 mg至约1.5mg的范围。
7.权利要求1的所述营养组合物,其中在所述组合物中的蛋白质的量为每100kcal 约1.4g至约3.5g。
8.权利要求7的所述营养组合物,其中所述蛋白质源不包括水解的或部分水解的蛋白质。
