Abstract
本发明涉及中药领域,尤其涉及一种抗幽门螺旋杆菌的中药组合物及其制备方法。所述抗幽门螺旋杆菌的中药组合物,按重量份数计,包括:辣木叶5~25份;丁香1~20份;甘草3~20份;西蓝花5~30份;黄芪1~10份;卵黄免疫球蛋白1~10份。将上述六种物质组合在一起,相互作用,能降低中药的毒副作用,抑制或杀死幽门螺旋杆菌效果更佳且具有提高机体免疫力的功效。该中药组合物是通过单独制备的复合中药提取物混合而成,可以作为食品的补充剂、添加剂或药物成分使用。

技术领域
本发明涉及中药领域,尤其涉及一种抗幽门螺旋杆菌的中药组合物及其制备方法。
背景技术
幽门螺旋杆菌(Helicobacter pylori,Hp)是一种革兰氏阴性杆菌,螺旋形,微需氧,对生长条件要求十分苛刻,幽门螺旋杆菌主要分布在胃粘膜组织中,胃液的强酸性,幽门螺旋杆菌很聪明的利用它的螺旋状结构,钻透胃黏膜表面的黏液,寄生在粘黏液中靠近胃黏膜上皮的相对中性的环境中。1983年首次从慢性活动性胃炎患者的胃粘膜活检组织中分离成功,是目前所知能够在人胃中生存的唯一微生物种类。
幽门螺旋杆菌感染引起的疾病包括胃炎、消化道溃疡、淋巴增生性胃淋巴瘤等,其中最严重的就是胃癌。有资料显示,中国13亿人口中,肠胃病患者就有1.2亿,消化性溃疡发病率10%,慢性胃炎发病率30%,是全世界当之无愧的“胃病大国”。此外,我国也是胃癌的高发区,每年新发现40万胃癌患者,占世界胃癌发病人数的42%。慢性胃病患者中Hp的检出率约为50%-95%,定植在胃黏膜表面的Hp通过产氨、分泌空泡毒素A等物质引起细胞损害,并能引发强烈的炎症反应和抗原诱导反应。
幽门螺旋杆菌感染率在不同人群中各不相同,其导致的疾病的多样性提示了其致病机制的复杂性,幽门螺旋杆菌的主要传播途径有粪-口、口-口、密切接触及动物源性等。目前治疗幽门螺旋杆菌的方法主要是基于抗生素的"三联"或"四联"疗法,使用抗生素不仅增加了菌株的耐药性,而且毒副作用大,因此本领域技术人员致力于寻找一种具有抗幽门螺旋杆菌作用、副作用小的替代物。
发明内容
近来的许多研宄证实很多中药具有抑菌的作用,有学者研究发现大黄、生姜、木香、苦参、石菖蒲根、儿茶、芦荟、白草果及芍药对幽门螺旋杆菌均有抑制作用。著名老中医张镜仁教授的经验方清胃冲剂(主要由徐长卿、连翘、赤苟、玄胡、旋覆花、平地木、炙甘草等组成)及双黄连口服液均发现可体外抑制幽门螺旋杆菌。国内有学者等针对脾胃湿热者幽门螺旋杆菌检出率较高这一临床情况,采用健脾清热化湿的健脾清化汤(主要由党参、茯苓、白术、黄连、大黄、蒲公英、丹参、甘草组成)进行体外抑杀幽门螺旋杆菌的实验研宄,结果显示:黄连、大黄有明显抑杀幽门螺旋杆菌作用,甘草、丹参、党参、公英等有抑制幽门螺旋杆菌生长的功效,与单味中药相比,健脾清化汤抑杀幽门螺旋杆菌更具明显优势。本发明根据文献报道并结合大量的实验研究,得到了一个能抑制幽门螺旋杆菌的中药组合物,其成分包括甘草、丁香、辣木叶以及西蓝花。
甘草又叫国老,甘草的主要活性成分为甘草酸、甘草黄酮,研宄指出甘草酸和甘草黄酮微克量级水平即具有一定的幽门螺旋杆菌抑杀作用。丁香可调节胃酸分泌,修复受损的胃粘膜,减少溃疡生成。辣木叶具有降糖降脂、抗肿瘤、抗氧化等多种功效,可增强人体免疫力、延缓衰老及预防疾病发生。黄芪具有抗炎作用,能杀灭幽门螺旋杆菌,提高机体免疫功能,抑制胃酸分泌。西蓝花具有杀菌抗癌的功效,还可提高机体免疫力。卵黄免疫球蛋白(IgY)具有较高的抗菌免疫活性,可抑制致病性大肠杆菌、沙门氏菌、金黄色葡萄球菌等常见肠道致病菌的生长,最近的研究表明,其对幽门螺旋杆菌杀灭的功能。而且该蛋白具有较高的耐热耐冷的特性,并且对胃蛋白酶较敏感,但经胰蛋白酶消化后仍保留了大部分的对抗原的结合力。动物及婴儿喂养实验证实,该IgY安全无毒,并可帮助机体增强免疫功能.提高抵抗细菌感染的能力,完全可作为具有增强免疫功能的食品功能因子。因此这五味中药组合在一起,外加卵黄囊蛋白,共同发挥抑杀幽门螺旋杆菌,提高机体免疫力的功效。
具体的,本发明采用以下技术方案来实现:
本发明一方面公开了一种抗幽门螺旋杆菌的中药组合物,按重量份数计,包括:辣木叶5~25份;丁香1~20份;甘草3~20份;西蓝花5~30份;黄芪1~10份;卵黄免疫球蛋白1~10份。
优选的,按重量份数计,包括:辣木叶15份;丁香20份;黄芪15份;甘草20份;西蓝花20份;卵黄免疫球蛋白10份。
本发明第二个方面公开了一种制备上述的抗幽门螺旋杆菌的中药组合物的方法,包括:
a.称取辣木叶、丁香、黄芪、甘草及西蓝花;
b.分别将辣木叶、丁香、黄芪、甘草及西蓝花进行水提、浓缩和干燥反应,分别得到它们的提取物;
c.将步骤b得到的提取物混合均匀得到混合物;
d.将卵黄免疫球蛋白加入步骤c得到的混合物中,再次混匀即得到中药组合物。
应当理解,本发明不限于上述步骤,还可以包含其他的步骤,例如在步骤a之前、步骤a和b之间、步骤b和c之间、步骤c和步骤d之间、步骤d之后,还包含其他额外的步骤,而不超出本发明的保护范围。
优选的,所述的制备方法,包括:
a.称取辣木叶、丁香、黄芪、甘草及西蓝花;
b.将辣木叶破碎后用8-14倍质量的水,煎煮提取1-2次,每次1-3h次,得到辣木叶提取液过滤后接着进行浓缩、醇沉和干燥反应后制得辣木叶提取物;
c.将丁香煎煮或回流提取1-2次,每次1-2h得到黄芪提取液,丁香提取液经过滤、浓缩和干燥反应后制得丁香提取物;
d.将黄芪煎煮或回流提取1-2次,每次1-2h得到黄芪提取液,黄芪提取液经过滤、浓缩和干燥反应制得黄芪提取物;
e.将甘草煎煮或回流提取1-2次,每次1-2h得到甘草提取液,甘草提取液经过滤、浓缩、干燥等工艺制得甘草提取物;
f.将西蓝花煎煮或回流提取1-2次,每次1-2h得到西蓝花提取液,西蓝花提取液经过滤、浓缩、干燥等工艺制得西蓝花提取物;
g.将步骤a-e中得到的辣木叶提取物、丁香提取物、黄芪提取物、甘草提取物和西蓝花提取物混合均匀,接着加入卵黄免疫球蛋白再次混匀即得中药组合物。

进一步的,在本发明的一些实施例中,步骤c-f中,所述丁香、黄芪、甘草和西蓝花均采用6-10倍质量的水或体积分数为60%~80%的乙醇煎煮或回流提取。
本发明第三个方面公开了上述制备方法得到的中药组合物。
本发明第四个方面公开了上述中药组合物在制备治疗幽门螺旋杆菌感染的药物中的应用。
本发明第五个方面公开了一种中药制剂,所述中药制剂包括上述的中药组合物及其药学上可接受的辅料。
优选的,所述中药制剂为对胃损伤有辅助保护功能的保健食品。
更优选的,所述辅料为糊精、麦芽糊精、β-环状糊精、可溶性淀粉、赤藓糖醇、罗汉果甜苷、异麦芽酮糖醇中的任意一种或任意组合。
应当理解,本发明所保护的辅料并不限于上述几种,本领域技术人员可以根据需要,选择任意合适的辅料来完成本发明的技术方案,并均在本发明的保护范围之内,
更优选的,所述中药制剂的剂型为片剂、颗粒剂、胶囊剂、散剂、丸剂或口服液。

本发明具有如下优点或者有益效果:

实施例1:
本实施例公开了一种制备抗幽门螺旋杆菌的中药组合物的方法,具体步骤如下:
1.称取辣木叶15g、丁香20g、黄芪15g、甘草20g,西蓝花20g以及卵黄免疫球蛋白10g;
2.制备辣木叶提取物:将辣木叶破碎,加入150g水,提取2次,2h/次,提取液过滤、浓缩、醇沉、干燥等工艺制得辣木叶菇提取物,辣木叶提取物的提取得率为15%;
3.制备丁香提取物:将丁香用200g水提取2次,每次1.5h,提取液经过滤、浓缩、干燥等工艺制得丁香提取物,所得丁香提取物的提取得率为20%;
4.制备黄芪提取物:将黄芪用480g水提取2次,每次1.5h,提取液经过滤、浓缩、干燥等工艺制得黄芪提取物,所得黄芪提取物的提取得率为25%;
5.制甘草提取物:300g水提取2次,每次1.5h,提取液经过滤、浓缩、干燥等工艺制得甘草提取物,所得甘草提取物的提取得率为20%;
6.制备西蓝花提取物:将西蓝花用600g水提取2次,每次1h,提取液经过滤、浓缩、干燥,所得西蓝花提取物的提取得率为30%;
7.将辣木叶提取物、丁香提取物、黄芪提取物、甘草提取物、西蓝花提取物,混合均匀后,加入卵黄免疫球蛋白10g,混匀即得中药组合物。
实施例2:
本实施例公开了一种制备抗幽门螺旋杆菌的中药组合物的方法,具体步骤如下:
1.称取辣木叶25g、丁香10g,黄芪10g、甘草20g、西蓝花30g及卵黄免疫球蛋白5g;
2.制备辣木叶提取物:将辣木叶破碎,加入250g水,提取1次,2h/次,提取液过滤、浓缩、醇沉、干燥等工艺制得辣木叶提取物,辣木叶提取物的提取得率为18%;
3.制备丁香提取物:将丁香用100g水提取1次2h,提取液经过滤、浓缩、干燥等工艺制得丁香提取物,所得黄芪提取物的提取得率为20%;
4.制备黄芪提取物:将黄芪用100g水提取1次2h,提取液经过滤、浓缩、干燥等工艺制得黄芪提取物,所得黄芪提取物的提取得率为25%;
5.制备甘草提取物:将甘草用200g水提取1次2h,提取液经过滤、浓缩、干燥等工艺制得甘草提取物,所得甘草提取物的提取得率为25%;
6.制备西蓝花提取物:将西蓝花用300g水提取1次1.5h,提取液经过滤、浓缩、干燥,得西蓝花提取物的提取得率为30%。
7.将辣木叶提取物、丁香提取物、黄芪提取物、甘草提取物、西蓝花提取物混合后加入加入卵黄免疫球蛋白5g,混匀即得中药组合物。
实施例3:
本实施例公开了一种制备抗幽门螺旋杆菌的中药组合物的方法,具体步骤如下:
1.称取辣木叶15g、丁香20g、黄芪15g、甘草20g,西蓝花20g以及卵黄免疫球蛋白10g;
2.制备辣木叶提取物:将辣木叶破碎,加入420g水,提取2次,1h/次,提取液过滤、浓缩、醇沉、干燥等工艺制得辣木叶提取物,辣木叶提取物的提取得率为20%;
3.制备丁香提取物:将丁香用200g体积分数为70%的乙醇提取2次,每次1h,提取液经过滤、浓缩、干燥等工艺制得黄芪提取物,所得丁香提取物的提取得率为30%;
4.制备黄芪提取物:将黄芪用150g体积分数为70%的乙醇提取2次,每次1h,提取液经过滤、浓缩、干燥等工艺制得黄芪提取物,所得黄芪提取物的提取得率为30%;
5.制备甘草提取物:将甘草用200g体积分数为70%的乙醇提取2次,每次1h,提取液经过滤、浓缩、干燥等工艺制得甘草提取物,所得甘草提取物的提取得率为35%;
6.制备西蓝花提取物:将西蓝花用200g体积分数为70%的乙醇提取2次,每次1h,提取液经过滤、浓缩、干燥,得到西蓝花提取物,所得西蓝花提取物的提取得率为35%;
7.将辣木叶提取物、丁香提取物、黄芪提取物、甘草提取物、西蓝花提取物,混合均匀后,加入卵黄免疫球蛋白10g,混匀即得中药组合物。
实施例4
本实施例将实施例3制备得到的抗幽门螺旋杆菌的中药组合物用于临床研究。
研究对象:经胃镜、组织学检查确诊的Hp阳性的患者100例;将这100例患者分为两组,每组各50名患者:一组服用实施例3制备得到的抗幽门螺旋杆菌的中药组合物,一组按照传统的四联疗法治疗。
服用方式:将中药组合物冲调为饮料或将组合物拌入其它饮食中服用,每日三次,每次15g。
治愈标准:临床症状、体征全部消失,胃镜、组织学检查为Hp阴性。
治疗效果:连续服用一周后,经检测发现:实施例3中组合物患者50例中有40例显示Hp阴性(有效率80%);而四联疗法患者50例中有25例显示阴性(有效率为50%)。连续服用二周后,实施例3中组合物患者50例中有50例显示Hp阴性(有效率100%);而四联疗法患者50例中有48例显示阴性(有效率为96%)。从实验中可以得出,实施例3制备得到的中药组合物与传统“四联疗法”相比,具有治疗效果更好、疗程短且制备方法更简单等优点。

1.一种抗幽门螺旋杆菌的中药组合物,其特征在于:按重量份数计,包括:辣木叶5~25份;丁香1~20份;甘草3~20份;西蓝花5~30份;黄芪1~10份;卵黄免疫球蛋白1~10份。
2.根据权利要求1所述的抗幽门螺旋杆菌的中药组合物,其特征在于:按重量份数计,包括:辣木叶15份;丁香20份;黄芪15份;甘草20份;西蓝花20份;卵黄免疫球蛋白10份。
3.一种制备权利要求1-2所述的抗幽门螺旋杆菌的中药组合物的方法,其特征在于:包括:
a.称取辣木叶、丁香、黄芪、甘草及西蓝花;
b.分别将辣木叶、丁香、黄芪、甘草及西蓝花进行水提、浓缩和干燥反应,分别得到它们的提取物;
c.将步骤b得到的提取物混合均匀得到混合物;
d.将卵黄免疫球蛋白加入步骤c得到的混合物中,再次混匀即得到中药组合物。
4.根据权利要求3所述的制备方法,其特征在于,包括:
a.称取辣木叶、丁香、黄芪、甘草及西蓝花;
b.将辣木叶破碎后用8-14倍质量的水,煎煮提取1-2次,每次1-3h次,得到辣木叶提取液过滤后接着进行浓缩、醇沉和干燥反应后制得辣木叶提取物;

d.将黄芪煎煮或回流提取1-2次,每次1-2h得到黄芪提取液,黄芪提取液经过滤、浓缩和干燥反应制得黄芪提取物;
e.将甘草煎煮或回流提取1-2次,每次1-2h得到甘草提取液,甘草提取液经过滤、浓缩、干燥等工艺制得甘草提取物;
f.将西蓝花煎煮或回流提取1-2次,每次1-2h得到西蓝花提取液,西蓝花提取液经过滤、浓缩、干燥等工艺制得西蓝花提取物;
g.将步骤a-e中得到的辣木叶提取物、丁香提取物、黄芪提取物、甘草提取物和西蓝花提取物混合均匀,接着加入卵黄免疫球蛋白再次混匀即得中药组合物。
5.根据权利要求4所述的制备方法,其特征在于,步骤c-f中,所述丁香、黄芪、甘草和西蓝花均采用6-10倍质量的水或体积分数为60%~80%的乙醇煎煮或回流提取。
6.根据权利要求3-5任意一项所述制备方法得到的中药组合物。
7.一种中药制剂,其特征在于,所述中药制剂包括权利要求1-2或权利要求6所述的中药组合物及其药学上可接受的辅料。
