本发明公开了一种克氏原螯虾适宜蛋脂比配合饲料及其制备方法,所述饲料包括如下主要组份及对应质量份:鱼粉20~30份;麸皮8~10份;豆粕6~10份;菜粕6~10份;棉籽粕7~10份;全麦粉6~10份;混合油脂2~4份;大豆卵磷脂1~2份;淀粉1‑1.5份;磷酸二氢钙1~2份;食盐0.1~0.5份;多维1~2份;多矿2~3份;微晶纤维素5~10份;甜菜碱1~2份。通过合理配置蛋白质/脂肪比,优化营养组份,开发出环保、高效克氏原螯虾配合饲料。

技术领域
本发明属于饲料领域,具体涉及一种克氏原螯虾适宜蛋脂比配合饲料及其制备方法。
背景技术
克氏原螯虾(Procambarus)俗称小龙虾,隶属甲壳纲十足目爬行亚目螯虾科原螯虾属。克氏原螯虾因其杂食性、生长速度快、适应能力强,在当地生态环境中形成绝对的竞争优势,近年来克氏原螯虾在我国已经成为重要的经济养殖品种,但是尚缺乏高效、专用的克氏原螯虾饲料,严重制约鳌虾养殖业的快速发展。
申请号2013102165625的中国专利申请公开了一种克氏原螯虾饲料及其制备方法,该饲料包括肉骨粉8~12份,豆粕12~18份,啤酒酵母0.2~0.6份,棉籽6~10份,玉米粉22~28份,蝇蛆粉20~25份,枯草芽孢杆菌0.8~1.2份,硫酸锌1~1.5份,磷酸钙2~3份。该饲料制备时添加了枯草芽孢杆菌降低克氏原螯虾的发病率,但仅考虑到蛋白质,纤维,脂类和矿物等营养成分的丰富性,未考虑作为核心营养成分的蛋白质以及蛋白脂肪比,但蛋白质以及蛋白质脂肪比是配合饲料质量和经济效益的重要指标。
申请号2016105910271的中国专利申请公开了一种克氏原螯虾环保沉性膨化饲料及制备方法。该饲料由一定比例的蛋白源、脂肪源、糖源原料,复合维生素、复合矿物质、抗氧化剂、诱食剂和粘合剂组成。步骤是:A、将饲料原料分别粉碎、过筛后和粘合剂混合均匀,边搅拌边加脂肪源原料,混合均匀后边搅拌边加水,制成饲料基料;B:将饲料基料输入双轴挤压膨化机中,切粒电机打开,挤压膨化机的操作:打开电闸、按下主电机,让饲料基料熟化后膜孔挤出,进行挤压膨化,得到半颗粒饲料,对颗粒饲料进行熟化,得到膨化颗粒饲料。该申请着重解决了克氏原螯虾饲料在水中的稳定性以及适口性问题,对于饲料的营养均衡性、以及克氏原螯虾对饲料蛋白与脂肪的需求以及蛋白与脂肪水平对克氏原螯虾的影响却未做太多考量。
张家宏等采用4×3双因子试验,探讨饲料中不同蛋白质和脂肪水平对克氏原螯虾成活率、生长、饵料系数、肌肉氨基酸组成的影响。结果表明:饲料中的蛋白质和脂肪水平显著影响克氏原螯虾的增重率(833.67%~1091.38%)、饵料系数(1.53~1.96);在蛋白质和脂肪水平分别为28%和6%的饲料组中,虾的增重率、特定生长率最高,饵料系数最低,差异性显著;饲料中必须氨基酸随饲料蛋白水平提高而增加,克氏原螯虾肌肉中必须氨基酸呈先上升后下降的趋势。24%蛋白质和6%脂肪的饲料组的必须氨基酸和总氨基酸含量最高,32%蛋白质和4%脂肪的饲料组最低,28%蛋白质和6%脂肪的饲料组中4种呈味氨基酸含量均较高。该研究采用的饲料配方为包含鱼粉、豆粕、面粉、麸皮、玉米粉、豆油、三七糠、多维和多矿的基础配方,可以为实际应用提供参考,但不具有实际应用价值。
以此为基础,本发明开发出一种克氏原螯虾适宜蛋脂比配合饲料,并提供了一种克氏原螯虾适宜蛋脂比配合饲料的制备方法。
发明内容
针对现有问题的不足,本发明的目的是提供一种克氏原螯虾适宜蛋脂比配合饲料及其制备方法。通过合理配置蛋白质/脂肪比,优化营养组份,开发出环保、高效的配合饲料。
本发明解决其技术问题采用的技术方案是:

作为本申请的优选技术方案,所述混合油脂为大豆油和鱼油按1:1的体积比混合而成。
一种克氏原螯虾适宜蛋脂比配合饲料的制备方法,包括如下步骤:
(1)按配方称取麸皮、豆粕、菜粕、棉籽粕以及微晶纤维素,加水混合均匀;接种乳酸杆菌和酵母菌,乳酸杆菌和酵母菌的接种量均为3~5%,接种同时添加蛋白酶,蛋白酶的添加量为底物总质量的2~4%,35~40℃发酵2-4d,干燥,得混合物A;
(2)按配方称取鱼粉、全麦粉和淀粉,混合均匀后,加压蒸汽条件下搅拌10~15min,干燥后,调湿12~24h,再送入挤压机150~180℃挤压膨化,膨化后粉碎过40目筛,得混合物B;
(3)称取磷酸二氢钙、食盐、多维、多矿和甜菜碱混合均匀,得混合物C;
(4)称取混合油脂和大豆卵磷脂,搅拌机搅拌5~10min,使油脂乳化,再加入步骤(1)的混合物A、步骤(2)的混合物B和步骤(3)的混合物C,混合均匀后调质熟化,挤压造粒,再经烘干和冷却,即得。

本发明利用菌类处理蛋白原料,将原有的蛋白类、多糖类抗营养因子等降解或者破坏,大分子降解为较小的片段,同时还会产生具有未知生长因子的发酵产物,使得发酵产物具有较高的营养价值和可利用性。
利用蛋白酶处理蛋白原料,在不破坏氨基酸组成和不降低蛋白质的营养价值的前提下将蛋白原料降解,分解成多肽和小肽,同时消除抗营养因子,提高酶解蛋白的适口性,增加诱食剂,提高了饲料的消化吸收率,最终提高酶解蛋白的整体营养水平。
利用加压蒸汽处理、调湿后挤压膨化处理鱼粉、全麦粉和淀粉,使其中蛋白质、淀粉和纤维素变性,蛋白质、淀粉和纤维素发生解聚,提高饲料营养价值,克氏原螯虾的摄取率。

本发明提供的一种克氏原螯虾适宜蛋脂比配合饲料及其制备方法,与现有技术相比,具有以下有益效果:
(1)本发明提供的克氏原螯虾适宜蛋脂比配合饲料通过合理配置蛋白质/脂肪比,优化营养组份,开发出环保、高效的配合饲料;
(2)本发明提供的克氏原螯虾适宜蛋脂比配合饲料的制备方法,通过对麸皮、豆粕、菜粕、棉籽粕以及微晶纤维素的发酵,提高饲料的利用率;
(3)本发明提供的克氏原螯虾适宜蛋脂比配合饲料的制备方法,通过加压蒸汽处理,调湿以及加压膨化,使蛋白、淀粉和纤维素发生解聚,提高饲料的营养价值。

实施例1:
一种克氏原螯虾适宜蛋脂比配合饲料,包括如下主要组份及对应质量份:鱼粉20份;麸皮8份;豆粕10份;菜粕7份;棉籽粕10份;全麦粉6份;大豆油2份,鱼油2份;大豆卵磷脂1份;淀粉1份;磷酸二氢钙2份;食盐0.1份;多维1份;多矿3份;微晶纤维素5份;甜菜碱1份;
上述克氏原螯虾适宜蛋脂比配合饲料的制备方法,包括如下具体步骤:
(1)按配方称取麸皮、豆粕、菜粕、棉籽粕以及微晶纤维素,加水混合均匀;接种乳酸杆菌和酵母菌,乳酸杆菌和酵母菌的接种量均为3%,乳酸杆菌和酵母菌的活菌数均≥500亿个/g,接种同时添加碱性蛋白酶,碱性蛋白酶的添加量为底物总质量的2%,37℃发酵2d,干燥,得混合物A;
(2)按配方称取鱼粉、全麦粉和淀粉,混合均匀后,3个大气压加压蒸汽条件下搅拌10min,60℃干燥后,调湿12h,再送入挤压机150℃挤压膨化,膨化后粉碎过60目筛,得混合物B;
(3)称取磷酸二氢钙、食盐、多维、多矿和甜菜碱混合均匀,得混合物C;
(4)称取混合油脂和大豆卵磷脂,搅拌机搅拌5min,使油脂乳化,再加入步骤(1)的混合物A、步骤(2)的混合物B和步骤(3)的混合物C,混合均匀后调质熟化,挤压造粒,再经烘干和冷却,即得。

(1)按配方称取麸皮、豆粕、菜粕、棉籽粕以及微晶纤维素,加水混合均匀;接种乳酸杆菌和酵母菌,乳酸杆菌和酵母菌的接种量均为4%,乳酸杆菌和酵母菌的活菌数均≥500亿个/g,接种同时添加碱性蛋白酶和风味蛋白酶,且碱性蛋白酶和风味蛋白酶的加入比例为2:1,蛋白酶的添加量为底物总质量的3%,35℃发酵4d,干燥,得混合物A;
(2)按配方称取鱼粉、全麦粉和淀粉,混合均匀后,6个大气压加压蒸汽条件下搅拌15min,80℃烘干后,调湿24h,再送入挤压机180℃挤压膨化,膨化后粉碎过60目筛,得混合物B;
(3)称取磷酸二氢钙、食盐、多维、多矿和甜菜碱混合均匀,得混合物C;
(4)称取混合油脂和大豆卵磷脂,搅拌机搅拌10min,使油脂乳化,再加入步骤(1)的混合物A、步骤(2)的混合物B和步骤(3)的混合物C,混合均匀后调质熟化,挤压造粒,再经烘干和冷却,即得。

2.饲养管理:选择体质健康、平均体重为7.23±0.76g的克氏原螯虾300尾,随机分为5个组,每个组3个重复,每个重复20尾虾,投放在15个养殖池(100cm×80cm×60cm)中国,每个重复放入一个池子。饲养条件控制在溶氧量>5mg/L,pH值为7.2~8.0,养殖温度为24~27℃,养殖用水每天更换1/4。每天投喂3次,时间分别为早上8:30;中午13:30,下午18:30,投饵量约为虾体重的4%,投饵后2h清理残饵和粪便。饲养时间80d。
3.生长性能:包括成活率、相对增重率、饲料系数、相对增长率,计算公式如下:
成活率(%)=100×终末尾数/初始尾数;
相对增重率(%)=100×(终末体重-初始体重)/初始体重;
饲料系数=饲料摄取总量/(终末体重-初始体重);
相对增长率(%)=100×(终末体长-初始体长)/初始体长
试验结果如表2所示。

由表2可见,成活率、相对增重率、相对增长率和饲料系数,本发明的实施例1-4都明显优于对照组,说明本发明的饲料确实可以促进克氏原螯虾机体的快速健康生长。
4.对消化酶的影响
消化酶:主要测量克氏原螯虾肠道和肝胰脏的蛋白酶、淀粉酶(AMS)和脂肪酶(LPS)活力,采用试剂盒(南京建成生物工程研究所)测定。

由表4可知,饲料中蛋白质脂肪比例对克氏原螯虾肠道脂肪酶、蛋白酶和肝胰腺脂肪酶、蛋白酶有显著影响,随着饲料蛋脂比的升高,克氏原螯虾肠道脂肪酶活性逐渐降低,肠道蛋白酶活性逐渐升高;饲料蛋脂比对克氏原螯虾肠道淀粉酶没有影响。与对照组相比,本申请的脂肪酶活力低于对照组,蛋白酶活力要高于对照组,因为克氏原螯虾肠道蛋白酶活性在一定范围内随着饲料中蛋白含量的升高而升高,超过一定范围蛋白酶活性反而会降低。

3.权利要求1所述的克氏原螯虾适宜蛋脂比配合饲料的制备方法,其特征在于,包括如下步骤:
(1)按配方称取麸皮、豆粕、菜粕、棉籽粕以及微晶纤维素,加水混合均匀;接种乳酸杆菌和酵母菌,乳酸杆菌和酵母菌的接种量均为3~5%,接种同时添加蛋白酶,蛋白酶的添加量为底物总质量的2~4%,35~60 ℃发酵2-4 d,干燥,得混合物A;
(2)按配方称取鱼粉、全麦粉和淀粉,混合均匀后,加压蒸汽条件下搅拌10~15 min,干燥后,调湿12~24 h,再送入挤压机150~180 ℃挤压膨化,膨化后粉碎过60目筛,得混合物B;
(3)称取磷酸二氢钙、食盐、多维、多矿和甜菜碱混合均匀,得混合物C;
(4)称取混合油脂和大豆卵磷脂,搅拌机搅拌5~10 min,使油脂乳化,再加入步骤(1)的混合物A、步骤(2)的混合物B和步骤(3)的混合物C,混合均匀后调质熟化,挤压造粒,再经烘干和冷却,即得。
4.根据权利要求3所述的克氏原螯虾适宜蛋脂比配合饲料的制备方法,其特征在于,所述步骤(1)中蛋白酶选择碱性蛋白酶、风味蛋白酶、木瓜蛋白酶和胰蛋白酶中的一种或多种。
