Abstract
本发明提供一种CpG寡脱氧核苷酸(CpG‑ODN),CpG‑ODN包含一个或多个重复的GTCGTT序列、一个或多个重复的GTT序列、以及一个或多个重复的TTTT序列,其中至少一个重复的GTCGTT序列是编码在GTT序列及TTTT序列之间。本发明还包含CpG‑ODN的免疫组成物及其在制备治疗或预防宿主疾病的免疫反应的药物用途。
Description
技术领域
本发明涉及CpG-寡脱氧核苷酸(CpG-ODN)、包含其的免疫组成物以及藉其诱发免疫反应的方法,具体而言,是涉及一种用于诱发TLR9-活化免疫反应和/或一种TLR-21-活化免疫反应且包含GTCGTT片段的CpG-ODN、包含其的免疫组成物以及藉其诱发该些免疫反应的方法。
背景技术
CpG-寡脱氧核苷酸(CpG-ODN)为已知具有潜力的免疫调控者,其诱发发炎性细胞介素及辅助型T细胞1(Th1)极化免疫反应,导致抗原呈现细胞中的共刺激分子的表现以及增加的B细胞、T细胞、自然杀手细胞、及其他免疫细胞的活化情况。
通常,不同的CpG-ODN基于其组成及长度而具有种族特异性活性。举例而言,含有两重复的GACGTT片段且长度为20个核苷酸的CpG-1826(SEQ ID NO:4)相较于含有三重复的GTCGTT片段且长度为22个核苷酸的CpG-2007(SEQ ID NO:11),对小鼠细胞的活化较为有力,而对人类细胞的活化效率较低。因此,已有设计并人工合成用于诱发特定物种中特定受体的免疫反应的不同种类的CpG-ODN。
另一方面,通过CpG-PDN活化的目标受体的选择亦为降低成本及普及以CpG-ODN为基础之治疗的重点。许多研究者已发现,由于类铎受体(TLR)在先天免疫系统中所扮演的重要角色,利用其作为在宿主内通过CpG-ODN诱发的目标可为较佳的选择。举例而言,斑马鱼(Daniorerio)、河豚(Takifugu rubripes)、以及鸡(Gallus gallus)中的TLR21显示出其可被CpG-ODN所活化(Brownlie R.et al.Molecular immunology.2009;46(15):3163-70)。
然而,不同的物种表现不同类型的TLR,其可在适用于对应物种之不同类型的CpG-ODN的开发上造成非必要的成本。因此,开发可诱发复数种类型的TLR的,亦即可诱发不同物种之免疫反应,且具有较佳活化效果的单一CpG-ODN为上述问题实质上的解决方法。
发明内容
在本发明中,表现TLR21及TLR9两者的石斑鱼细胞及表现TLR9的哺乳类细胞系用于本发明的CpG-ODN的活化效应测试中,以呈现该CpG-ODN为有力于通过活化不同类型的TLR以诱发不同物种中免疫反应。
在本发明中,提供一种CpG寡脱氧核苷酸(CpG-ODN),其是用于诱发宿主内TLR9-活化免疫反应、TLR21-活化免疫反应或其组合,包含:一个或多个重复的GTCGTT序列、一个或多个重复的GTT序列、以及一个或多个重复的TTTT序列,其中至少一个重复的GTCGTT序列是编码在GTT序列及TTTT序列之间。

具体实施方式
为便于使审查员理解本发明的技术特征、内容、优点及效应,本发明之实施例、附图说明及表格将于下文中详细说明。图示仅用于示例及辅助说明书的内容,并非用于以任何方式限制本发明之实施,在此先与说明。
本发明中所使用之用于实施及用于证实实质效果的方法、制备及取得之材料将于以下详述。
在本发明的实施例中,石斑鱼的头肾细胞及脾细胞是根据以下步骤分离。在含有0.2g/L三卡因(Tricaine)的水中麻醉点带石斑鱼及鞍带石斑鱼,并无菌地将其头肾及脾脏移除。接着将器官和缓地剁碎并与均质化缓冲液(标准Hank’s平衡盐溶液(sHBSS),补以15mM HEPES、10%胎牛血清(FBS)、1x抗生素-抗真菌剂(Antibiotic-Antimycotic)及50U/ml的肝素)通过70-μm滤网。接着,将均质的组织悬浮液置入8mL蒸馏水及1mL的10x磷酸盐缓冲生理盐水(PBS)中。移除大型的碎片,并将细胞悬浮液离心并利用1x PBS清洗两次。在Leibovitz’s L-15细胞培养基中使沉淀的细胞重新悬浮(Gibco,Carlsbad,CA,USA)。
根据以下步骤收集并培养C57BL/6小鼠的骨髓衍生巨噬细胞(BMDM)及脾细胞。BMDM方面,取自胫骨和股骨的小鼠骨随细胞系在完全的改良杜氏伊格尔培养基(Dulbecco’s Modified Eagle Medium,DMEM)与30%L929条件培养基中培养5天,并接续以DMEM培养基与10%FBS培养。在脾细胞方面,其在取得后于RPMI1640培养基与10%FBS中培养。再者,人类周边血液单核球细胞(PBMC)是培养于RPMI1640培养基与10%FBS中。
鞍带石斑鱼的TLR21cDNA(ggTLR21cDNA)是由以下步骤分子选殖(clone)。利用TRIzol由石斑鱼脾细胞中纯化全RNA,且利用III第一股合成系统(III First-Strand Synthesis System)(Invitrogen,Carlsbad,CA,USA)得到cDNA文库,上述是已知于先前技术(Yeh D.W.et al.Proceedings of the NationalAcademy of Sciences of the United States of America.2013;110(51):20711-6)。为选殖ggTLR21cDNA,一对正向及反向引子(5’-GAACAGATTCCTGTACCATGTTCATC-3’(SEQ IDNO:51)及5’-GCTTGTATGAATTGTCACACTGCAC-3’(SEQ ID NO:52))是基于点带石斑鱼TLR21(osgTLR21)的5’-及3’-未转译区域(GenBank:GU198366.2)而设计。含有5’-及3’-未转译区域两者的ggTLR21 cDNA及完整编码区域是利用聚合酶连锁反应(PCR)放大从鞍带石斑鱼脾脏第一股cDNA文库所选殖。进一步地,选殖的ggTLR21cDNA序列是登录至GenBank数据库(登录号:KM024068)。
TLR21的胺基酸序列的多重比对是利用ClustalW2进行。TLR21蛋白的构造模型是以TLR13为模板以SWISS MODEL预测。
osgTLR21及ggTLR21的表现结构是由衍生自点带石斑鱼及鞍带石斑鱼脾脏产生的第一股cDNA文库对应的蛋白质编码区域的PCR放大所产生。设计用于编码osgTLR21(GU198366.2)及ggTLR21(KM024068)区域的基于5’-及3’-端cDNA序列之正向及反向引子,并将其在C端亚选殖放大的DNA片段至具有FLAG标识之框架中的PEF6载体。用于人类及小鼠TLR9的表现载体是通过已述方法生产(Liu J.Comparative immunology,microbiologyand infectious diseases.2012;35(5):443-51)。
在本发明的实施例中,TLR21及TLR9活化分析是根据以下步骤进行。人类胚胎肾脏(HEK)293细胞是在DMEM与10%FBS中生长。接着,进行先前技术(Yeh D.W.etal.Proceedings of the National Academy of Sciences of the United States ofAmerica.2013;110(51):20711-6)所载的步骤。计算与未刺激控制组的倍率差异,以作为相对的荧光素酶活性。
基因表现的反转录定量PCR(RT-qPCR)分析是根据以下步骤进行。将细胞以不同的CpG-ODN处理4小时。接着,通过TRIzol纯化全RNA并利用如:SuperScript III第一股合成系统(Invitrogen,Carlsbad,CA,USA)进行转录。进一步地,RT-qPCR是利用合适的方法或套件运作,例如,用于基因表现分析的ABI PRISM 7900HT序列检测系统(ABI PRISM 7900HTSequence Detection System)及KAPA快速qPCR套件(KAPA fast qPCRkit)(KK4605)。mRNA的表现是标准化至β-肌动蛋白。如下述,在表1中,比较例的序列是用于解释制备过程及用于与本发明的例子的CpG-ODN(SEQ ID NO:2及SEQ ID NO:3)比较,以显示其效果上的差异。进一步的,用于RT-qPCR的引子序列列于表2及表3。

十二烷基硫酸钠聚丙烯酰胺凝胶电泳(SDS-PAGE)及免疫墨点分析是根据以下步骤进行。通过含完整的通用型蛋白酶抑制剂(complete protease inhibitor cocktail)裂解缓冲溶液(100mM NaCl、50mM Tris-Cl(pH 7.5)、0.5mM乙二胺四乙酸(EDTA)、1%乙基苯基聚乙二醇(octylphenoxypolyethoxyethanol)(NP-40))裂解细胞。接着以SDS-PAGE分离细胞裂解物并转移至聚偏二氟乙烯(polyvinylidene fluoride,PVDF)膜上。下一步,将膜与标识抗体共同培养,再与山葵过氧化酶(horseradish peroxidase,HRP)耦合二次抗体培养,其免疫活化带是通过化学冷光HRP底物(chemiluminescent HRP substrate)(Immobilon Western;Millipore,Temecula,CA,USA)及UVP生物光谱影像系统(UVPBioSpectrum Imaging System)可视化。
人类细胞介素的生产结果是通过以下步骤测量。以不同CpG-ODN处理PBMC 24小时,并收集培养系统的上清液以用于细胞介素测量。肿瘤坏死因子-α(TNF-α)、介白素(IL)-1β、IL-12p70及干扰素(IFN)-γ的产生是通过ELISA套件测量。
再者,本揭露所有结果是以三次独立实验的平均值±标准偏差(mean±SD)表示。统计分析是通过学生t检验(Student’s t-test)进行,显著性水瓶为P<0.05。
在此,数个实施例与结果及本发明的附图一并描述,以便于使本发明提供的CpG-ODN更易于理解。
在本发明的实施例中,ggTLR21可通过上述方法选殖。来自osgTLR21A及osgTLR21B的cDNA可根据上述文献(Li Y.W.et al.Cryptocaryon irritans infection.Fish&shellfish immunology.2012;32(3):476-81)选殖。其编码的蛋白质序列间最大的不同是osgTLR21B在C端的四个胺基酸残基的缺少,如图1B到图1F所示。基于预期的点带石斑鱼与鞍带石斑鱼间同一基因核苷酸序列的高相同度,发明人基于osgTLR21的5’及3’未转译区域设计两个引子,以选殖ggTLR21。此结果得到与osgTLR21A同源但与osgTLR21B不同源之完整长度的ggTLR21cDNA。
接着,可分析本发明所使用的石斑鱼TLR21的特性。为比较石斑鱼TLR21与其他TLR,本发明利用ClustalW2对照其与不同种斑马鱼TLR的蛋白质序列,并建构如第2A图所示的进化树,其中该些蛋白质序列及代号系由GenBank数据库取得。如结果所示,ggTLR21及osgTLR21A间的蛋白质相同度为98.26%,ggTLR21及osgTLR21B间为98.15%,斑马鱼(zeb)TLR21及ggTLR21间为54.44%,且zebTLR22及ggTLR21A间为28.30%。TLR22为进化树中最接近TLR21者,而TLR13及TLR20f则是次接近的。
再者,如图2B所示,利用来自不同鱼类及禽鸟类物种的TLR21蛋白质序列谱系分析显示石斑鱼TLR21与横带石鲷(Oplegnathus fasciatus)TLR21最为相关,次者为牙鲆(Paralichthys olivaceus)及大菱鲆(Scophthalmus maximus)TLR21,且与鸡(原鸡(Gullas gullas))、鸭(绿头鸭(Anas platyrhynchos))以及鹅(鸿雁(Anser cygnoides))关系较远。
另一方面,亦分析不同石斑鱼物种间TLR21的结构及序列。就结果而言,ggTLR21包含979胺基酸残基,其中osgTLR21A及osgTLR21B分别包含979及975。此三个蛋白质序列的对照显示其包含细胞外结构域(胞外结构域(ectodomain)、跨膜结构域、以及Toll/IL-1(TIR)胞质液(cytosolic)结构域。该等在胞外结构域中亦具有23重复的富亮氨酸重复(LRR)及C端富亮氨酸重复(LRR-CT)。osgTLR21及ggTLR21的胞外结构域间仅有741个中的14个胺基酸残基不同。先前技术中,已解析不同TLR胞外结构域的数个三维结构,其中TLR13与TLR21最为谱系相近。因此,发明人以TLR13为模板,通过SWISS MODEL软件预测ostTLR21及ggTLR21胞外结构域的三维结构,以进一步探讨其差异。如第1A图所示,此显示该些胞外结构域除在osgTLR21中315-330胺基酸区域挤出的小螺旋结构,是相对地相似于马蹄形螺线体(solenoid)三维结构,其中由左至右系为osgTLR21(黑)、ggTLR21(白)及两者之迭加(黑:osgTLR21;白:ggTLR21)预测的胞外结构域结构。(N:胞外结构域的N端;C:胞外结构域的C端)。参照第1B图到第1F图,哺乳类TLR讯号传导所需的三个片段Box 1-3系保留在全三种石斑鱼TLR21的TIR结构域。然而,osgTLR11B与osgTLR21A及ggTLR21之不同点在于其在Box3后的C端缺少四个胺基酸残基的区域(星号:相同残基;单点:保留型取代;两点:高保留型取代)。
接着,为探讨这些石斑鱼TLR21间的结构差异转译出其对于不同类型的CpG-ODN刺激的活性差异,发明人以该些TLR21及核因子(NF)-κB控制之荧光素酶报告基因与用于其之表现载体共转染HEK293细胞,以建立以细胞为基础的活化试验。接着,发明人以如表1所示之不同的六元片段CpG-ODN及序列处理该些细胞,并测量受诱发的荧光素酶报告基因活性。如图3第(I)部分至第(IV)部分,其显示osgTLR21A、TLR21B及ggTLR21,通过含有GTCGTT六元片段(亦即,CPG-2006[SEQ ID NO:7]及CpG-2007)的CpG-ODN得到相同的活化程度,但对含有GACGTT及AACGTT六元片段的CpG-ODN(亦即,CpG-1826、CpG-2000[SEQ ID NO:5]及CpG-HC4040[SEQ ID NO:6])无反应。此结果支持全三种石斑鱼TLR是具有功能性的,且这些最小的结构差异并不显著影响其对CpG-ODN的刺激。
根据上述结果,为开发强活化石斑鱼TLR21之CpG-ODN之意图,发明人裁切CpG-2006及CpG-2007的长度,以仅留下此三段GTCGTT六元片段的左两段或右两段,以分别产生CpG-261(SEQ ID NO:8)及CpG-27(SEQ ID NO:12)以及CpG-262(SEQ ID NO:9)及CpG-272(SEQ ID NO:13)。再者,发明人亦裁切其长度以仅留下中间复制段(表1),以产生CpG-263(SEQ ID NO:10及CpG-273(SEQ ID NO:14)。接着,这些CpG-ODN的活性是通过以osgTLR21及ggTLR21细胞为基础的活化试验测定,并与CpG-2006及CpG-2007比较。如图4第(I)部分至第(III)部分所示之结果,其显示CpG-272对于osgTLR21及ggTLR21具有最佳活性。因此,发明人进一步生产CpG-2721(SEQ ID NO:15)、CpG-2722(SEQ ID NO:2)、CpG-2723(SEQ ID NOs:16)、CpG-2724(SEQ ID NO:17)及CpG-2725(SEQ ID NO:18)(表1)。进行以细胞为基础的进一步试验,且结果显示CpG-2722对osgTLR21及ggTLR21具有最好的活性,如图5第(I)部分至第(III)部分所示。
由于活化石斑鱼TLR21的CpG六元片段已从上述结果得到,发明人进一步探讨CpG-2722的CpG六元片段的数量及结构是否会影响石斑鱼TLR21的活化。在上述结果中,含有两重复GTCGTT六元片段的19个核苷酸长的CpG-2722具有最佳的活性。为定义是否两重复的片段皆为其活性所需,发明人反转其在5’-及3’-的CpG六元片段中的CpG双脱氧核苷酸,以产生CpG-2726(SEQ ID NO:19)、CpG-2727(SEQ ID NO:3)及CpG-2728(SEQ ID NO:20)(表1)。如图6第(I)部分至第(III)部分的osgTLR21及ggTLR21活化试验结果所示,显示CpG-2727的活性系与CpG-2722一样好,而CpG-2726及CpG-2728是无法活化osgTLR21及ggTLR21石斑鱼TLR21。这些结果支持仅依重复之5’CpG六元片段仅为CpG-2722的活性所需。进一步裁切CpG-2722,通过从其5’端移除三个核苷酸以产生CpG-2729(SEQ ID NO:21),通过从3’-CpG六元片段移除三个核苷酸以产生CpG-2730(SEQ ID NO:21),以及通过从5’端及3’-CpG六元片段两者移除三个核苷酸以产生CpG-2731(SEQ ID NO:1)(表1)。
此外,测试这些CpG-ODN的活性,其结果显示于第7图第(I)部分至第(III)部分。这些结果显示,三个5’端核苷酸为CpG-2722强活化osgTLR21及ggTLR21所需,且尽管3’CpG六元片段非为CpG-2722活性所需,该些核苷酸是为维持CpG-2722的19个核苷酸长度的活性所需。
另外,如图8第(I)部分至第(IV)部分所示,发明人测试CpG-2722及CpG-2727在人类(h)及小鼠(m)的活性,以测定他们是否对石斑鱼TLR21具有特异性。有趣的是,在hTLR9活化试验,根据结果,得到CpG-2722及CpG-2727之活性与CpG-2006及CpG-2007者一样好,在mTLR9活化试验,虽然CpG-2722及CpG-2727的活性不像CpG-1826者一样好,而是已对小鼠细胞之活化而言为优化,两者对mTLR9的活性比CpG-2006及CpG-2007更好。
此外,发明人探讨CpG-2722及CpG-2727在石斑鱼、人类、及小鼠细胞中诱发细胞介素产生的效果,比较其在该些细胞的活性,如图8中第(I)部分至第(IV)部分之以细胞为基础的实验所示。由点带石斑鱼纯化头肾细胞及脾脏细胞,以不同CpG-ODN处理,以测定细胞介素IL-1α、IL-6、IL-8、及IFNγ的诱发。与如图9第(I)部分至第(IV)部分及图10第(I)部分至第(IV)部分所示的以细胞为基础的试验相符,显示CpG-2722及CpG2727对于点带石斑鱼中细胞介素生产的活化相较于CpG1826、CpG-2006及CpG-2007具有较大的效果。另外,进行ELISA分析,显示CpG-2722及CpG-2727与CpG-2006及CpG-2007在人类PBMC中诱发细胞介素生产的潜力相同,而CpG-1826在该些细胞上具有较弱的活性,如图11第(I)部分至第(IV)部分所示。再者,发现CpG-2722及CpG-2727在小鼠皮细胞及BMDM中诱导细胞介素表现的潜力高于CpG-2006及CpG-2007,如图12第(I)部分至第(IV)部分及图13第(I)部分至第(IV)部分所示。该些发现支持为石斑鱼TLR21开发的CpG-2722及CpG-2722亦可有效地活化人类及小鼠细胞的免疫反应。
根据上述实施例及结果,更多本发明的细节可由以下段落描述及解释。
一般而言,TLR9及TLR21为对于CpG-ODN的细胞受体。TLR-9表现于哺乳类及鱼类两者,且强化地及种族特异性地活化此TLR的CpG-ODN的所需结构已有较多研究。在现今,对人类优化的CpG-ODN已探讨其作为免疫佐剂及过敏、感染性疾病及癌症免疫治疗之应用。相似地,在先前技术中,CpG-ODN亦显示其发挥在含有TLR21之鸡、鸭、鱼的免疫刺激活性,其支持CpG-ODN在兽医领域作为免疫调控技及疫苗佐剂的潜力。然而,鲜少有研究探讨CpG-ODN活化TLR21的所需结构,且是否有结构可活化TLR9及TLR21两者仍不清楚。因此,在本发明中,发明人开发用以强活化因生长快速且高价而被视为亚洲的重要水产物种之石斑鱼中TLR21的CpG-ODN,并探讨CpG-ODN、石斑鱼TLR21及人类与小鼠TLR9之间的交互作用。
发明人发现含有GTCGTT片段的CpG-ODN,如CpG-2006及CpG-2007等,可活化石斑鱼TLR21,而含有GACGTT及AACGTT片段者,如CpG-1826、CpG-2000及HC4040等则不可。相似地,先前已示zebTLR9广泛的辨识具有不同CpG六元片段的CpG-ODN,含有GACGTT或AACGTT片段者对TLR9具有较佳的活性,而含有GTCGTT片段者对zebTLR21具有较佳的活性(Yeh D.W.etal.Proceedings of the National Academy of Sciences of the United States ofAmerica.2013;110(51):20711-6))。因此,石斑鱼TLR21具有与zebTLR21相似的CpG六元片段辨识轮廓。
在本发明中,提供对石斑鱼TLR21相对于CpG-2006及CpG-2007具有较佳活性的两个CpG-ODN。CpG-2722具有19个脱氧核苷酸的长度且含有两个GTCGTT片段,并在其之间具有四个相隔的核苷酸。而CpG-2727是CpG-2722的修改,是将GTGTT片段中的CpG-双脱氧核苷酸反转。因此,CpG-2722的良好活性支持一重复的GTCGTT是足以强活化石斑鱼TLR21。在开发强活化兔子TLR9之CpG-ODN的先前文献,发现CpG-ODN的长度亦影响其活性(Chuang TH,etal.PloS one.2014;9(9):e108808)。因此,含有与CpG-2006及CpG-2007相同类型的CpG六元片段的CpG-2722及CpG-2727但具有不同长度及CpG六元片段数量的事实支持为使CpG-ODN强活化TLR21而设计的关键结构与为使TLR9强活化者相同。
在上述实施例及结果中,亦探讨CpG-2722及CpG-2727在应含有TLR9及TLR21两者的由点带石斑鱼头肾及脾脏分离出的细胞中的活性。此显示虽然与用于石斑鱼TLR21的活化轮廓不完全相同,CpG-2722及CpG-2727在这些细胞中细胞介素的生产上展现出比CpG-2006、CpG-2007及CpG-1826更强的效果。因此,CpG-2722及CpG-2727亦可对石斑鱼TLR9具有好的活性,且出现TLR9及TLR21两者偕同地调控石斑鱼中CpG-ODN的免疫刺激活性,如同先前显示之通过具有GTCGTT片段的CpG-ODN调控zebTLR9及zebTLR21者(Yeh D.W.etal.Proceedings of the National Academy of Sciences of the United States ofAmerica.2013;110(51):20711-6)。可选地,此可支持TLR21是为石斑鱼细胞中CpG-ODN的主要受体。
另一方面,人类及小鼠TLR9已显示具有种族特异性配体辨识特性,其中人类TLR9被CpG-2006及CpG-2007强活化,但被CpG-1826若活化,以及小鼠TLR9被CpG-1826优先活化。在此,发明人发现在人类TLR9的活化及人类PBMC的细胞介素产生方面,CpG-2722及CpG-2727具有与CpG-2006及CpG-2007相同的强度。再者,这两个CpG-ODN亦可比CpG-2006及CpG-2007在小鼠TLR9及小鼠细胞内细胞介素的产生具有较佳的效果。这支持了这两个开发的CpG-ODN的结构可更佳地覆盖人类及小鼠TLR的种族特异性配体辨识特性,且亦可共享来自于不同物种的用于TLR9及TLR21的活化的共同结构。因此,这两个CpG-ODN可具有作为一个范围之物种的免疫调控剂或疫苗佐剂的潜力。
总括而言,本发明提供一种用于诱发宿主的TLR9-活化免疫反应、TLR21-活化免疫反应或其组合的CpG-ODN,其中CpG-ODN包含一或多个重复的GTCGTT序列、一或多个重复的GTT序列、以及一或多个重复的TTTT序列。此外,至少一重复的GTCGTT序列是编码于GTT序列及TTTT序列之间。优选地,TTTT可直接编码于GTCGTT之后,亦即,可为GTCGTTTTTT序列(SEQID NO:1)
优选地,对TLR9及TLR21具有较佳活性的CpG-ODN为上述结果中显示较佳效果的CpG-2722(SEQ ID NO:2)及CpG-2727(SEQ ID NO:3)。
通过本发明的CpG-ODN活化免疫反应的宿主可为表现TLR9及/或TLR21的物种,如人类、禽鸟类、啮齿类、及鱼类或其他适当的物种。
同样地,本发明的CpG-ODN可与载体、及/或赋形剂或任何其他医药上可接受的添加物结合作为免疫组成物,以增强其实用性或生物兼容性。较佳地,免疫组成物亦可与抗原结合,以达到治疗、特异性标靶、或其他额外功能的增强。优选地,抗原可为HBsAg、李斯特菌溶血素O(listeriolysin O)、顶膜抗原1(apical membrane antigen 1)、MART1及利什曼原虫(leishmanial)。
本发明的CpG-ODN及免疫组成物亦可用于活化宿主之免疫反应的方法。方法可包含制备有效剂量的CpG-ODN或包含有效剂量的CpG-ODN的免疫组成物,以及施予制备好的产物至宿主中。
在本发明之活化宿主之免疫反应的方法中,有效剂量可优选地为0.01mg/kg~20mg/kg、更优选地为0.1mg/kg~20mg/kg、更优选地为0.2mg/kg~10mg/kg、更优选地为0.5mg/kg~5mg/kg。
优选地,本发明之宿主可为具有治疗或预防包含选自以下疾病所组成之群组中的存活个体:如乳癌、前列腺癌、黑色素瘤、淋巴瘤、非小细胞肺癌、基底细胞癌、胶质母细胞瘤、卵巢癌等癌症、以及如B型肝炎病毒、炭疽芽孢杆菌、疟疾、肺炎链球菌、单纯疱疹病毒及流感病毒等传染性疾病
进一步地,在本发明的方法中,施予可为从口腔施予、通过注射手段施予或其他任何常见的施予方法。

Claims (13)
1.一种CpG寡脱氧核苷酸,其特征在于,是用于诱发宿主内TLR9-活化免疫反应、TLR21-活化免疫反应或其组合,包含:
一个或多个重复的GTCGTT序列;
一个或多个重复的GTT序列;以及
一个或多个重复的TTTT序列;
其中,至少一个重复的GTCGTT序列是编码在GTT序列及TTTT序列之间。
2.如权利要求1所述的CpG寡脱氧核苷酸,其特征在于,包含如SEQ ID NO:1所示的序列。
3.如权利要求1所述的CpG寡脱氧核苷酸,其特征在于,长度为19个核苷酸。
4.如权利要求1所述的CpG寡脱氧核苷酸,其特征在于,包含如SEQ ID NO:2所示的序列。

7.一种免疫组成物,其特征在于,是用于诱发TLR9-活化免疫反应、TLR21-活化免疫反应或其组合,包含:

9.一种免疫组成物在制备用于诱发免疫反应的药物的用途,其特征在于,该免疫反应是用于治疗或预防宿主的疾病,该用途包含:
制备用于诱发TLR9-活化免疫反应、TLR21-活化免疫反应或其组合的且包含有效剂量的CpG寡脱氧核苷酸的该免疫组成物,其中该CpG寡脱氧核苷酸包含:
一个或多个重复的GTCGTT序列;
一个或多个重复的GTT序列;以及
一个或多个重复的TTTT序列;
其中,至少一个重复的GTCGTT序列是编码在GTT序列及TTTT序列之间;以及
施予该免疫组成物至该宿主。
10.如权利要求9所述的用途,其特征在于,该宿主是选自由人类、禽鸟类、啮齿类及鱼类所组成的群组中。
11.如权利要求9所述的用途,其特征在于,该有效剂量为0.01mg/kg体重至20mg/kg体重。
12.如权利要求9所述的用途,其特征在于,该疾病是选自由以下所组成的群组中:乳癌、前列腺癌、黑色素瘤、淋巴瘤、非小细胞肺癌、基底细胞癌、胶质母细胞瘤、卵巢癌、
以及由B型肝炎病毒、炭疽芽孢杆菌、疟疾、肺炎链球菌、单纯疱疹病毒、流感病毒或其组合引起的传染性疾病。
13.如权利要求9所述的用途,其特征在于,该施予包含从口腔施予或通过注射手段施予。
