Abstract
本发明涉及褐藻寡糖制品及其提取方法和肉鸡饲料制品,其中褐藻寡糖制品的提取方法包括:在褐藻酸钠中添加水,制成均匀的褐藻酸钠胶液;将褐藻酸钠胶液与盐酸溶液混合,水解产生沉淀;离心取沉淀物;将沉淀物倒入碳酸氢钠溶液中分级;取上清液醇沉,收集沉淀;将收集的沉淀物重复进行分层步骤和醇沉步骤多次,至清除杂质;干燥,得到褐藻酸钠制品;饲料制品中含有一定量的本发明褐藻寡糖。本发明的褐藻寡糖制品及其肉鸡饲料制品,能显著提高肉鸡的抗氧化能力和免疫力,有助于修复应激造成的肉鸡肠道损伤,提高肉鸡的抗病能力,降低肉鸡死淘率,提高养殖经济效益。

技术领域
本发明属于动物营养与饲料科学技术领域,特别涉及褐藻寡糖制品及其提取方法和肉鸡饲料制品。
背景技术
肉鸡胃肠道的微生物区系对其早期生长阶段的营养、健康和生长性能等方面至关重要。盲肠缺乏健康的微生物群落状态被认为是肉仔鸡容易触发细菌感染的一个主要因素。肉仔鸡的肠道菌群受其日粮营养的影响,肠道食糜的化学组成在很大程度上决定了肠道微生物的组成和群落状态。日粮改变会导致肠道菌群发生相应的变化,而日粮组成、肠道微生物及其两者之间的交互关系又可影响肉鸡的肠道发育、肠黏膜结构和黏液成分。
大量报道已证明褐藻寡糖具有益生元的作用,能够改善肠道微生物菌群结构,日粮添加褐藻寡糖能够减少肉仔鸡盲肠中沙门氏菌的定植和改善鸡的肠道屏障,因此褐藻寡糖在饲料领域中的应用较为广泛。
然而,由于多数褐藻寡糖具有相对分子质量大,粘度大,水溶性低的特点,因此较难被仔鸡肠胃吸收,有时为达到预定效果,往往需要进行大量添加,但仔鸡由于肠胃较为虚弱,因此反过来又会引起其他类生长性问题,因此普通的褐藻寡糖类产品在动物饲料中的理论价值很高,但实际利用率却并不高,且存在一定的定量难度。
发明内容
本发明的目的是为解决以上问题,本发明提供一种总糖含量高、易被仔鸡肠胃吸收的褐藻寡糖制品及其制备方法和肉鸡饲料制品。
根据本发明的一个方面,提供褐藻寡糖制品的提取方法,包括以下步骤:在褐藻酸钠中添加水,制成均匀的褐藻酸钠胶液;将褐藻酸钠胶液与盐酸溶液混合,水解产生沉淀;离心取沉淀物;将沉淀物倒入碳酸氢钠溶液中分级;取上清液醇沉,收集沉淀;将收集的沉淀物重复进行分层步骤和醇沉步骤多次,至清除杂质;将沉淀物干燥得到藻酸钠制品。
其中,褐藻酸钠胶液的质量分数为2%-15%,盐酸溶液的浓度为0.2-1.2mol/L。
其中,水解温度为70℃-150℃,水解时间为10-15h。
其中,分级步骤包括,将沉淀物溶解于浓度为碳8-10g/L酸氢钠溶液中,使用盐酸调节pH值至2-3,使溶液分层。
其中,醇沉步骤包括,将上清液加入95%乙醇中产生沉淀,乙醇的体积为上清液的3-4倍,离心取沉淀,抽滤除去残留的乙醇和水。
其中,离心步骤中,离心转速为4000-6000r/min,离心时间为10-15min;干燥步骤中,干燥方式为烘干,温度为50℃-60℃。
根据本发明的另一方面,提供提取方法制取的褐藻寡糖制品。
根据本发明的第三发明,提供包含该褐藻寡糖制品的肉鸡饲料制品,包括以下重量份数的原料:褐藻寡糖制品0.05-0.2份、玉米50-65份、豆粕25-35份、鱼粉0-3份、大豆油1-4份、磷酸氢钙1-3份、石粉0.5-1.5份、食盐0.2-0.5份、DL-蛋氨酸0.05-0.3份、维生素矿物质预混料0.5-1.5份。
其中,维生素矿物质预混料是指提供每公斤配合饲料:维生素A 9000IU、维生素D33000IU、维生素E 24.0mg、维生素K3 1.8mg、维生素B1 2.0mg、维生素B2 5.0mg、维生素B63.0mg、维生素B12 0.1mg、烟酸40.0mg、泛酸15.0mg、叶酸1.0mg、生物素0.05mg、氯化胆碱500mg、铁80.0mg、铜20.0mg、锌90.0mg、锰80.0mg、碘0.35mg、硒0.30mg。
其中,该肉鸡饲料制品包括以下重量份数的原料组成:褐藻寡糖制品0.20份、玉米58.40份、豆粕32.50份、鱼粉2.05份、大豆油1.55份、磷酸氢钙1.70份、石粉1.50份、食盐0.30份、DL-蛋氨酸0.30份和维生素矿物质预混料1.50份。
其中,肉鸡饲料制品包括以下重量份数的原料组成:褐藻寡糖制品0.10份、玉米62.83份、豆粕28.95份、大豆油3.50份、磷酸氢钙2.55份、石粉0.95份、食盐0.50份、DL-蛋氨酸0.12份和维生素矿物质预混料0.50份。
本发明具有以下有益效果:
1.本发明褐藻寡糖制品的制备方法简洁易操作、成本较低,褐藻寡糖制品分子质量小,水溶性好,总糖含量最高可达81.35%,特性黏数可低至6.45,pH值为3.5-4,肠道吸收性能好,能显著改善肉鸡的氧化能力、提高免疫能力和调节肠道菌群结构,具有促生长和降低死淘的效果。
2.本发明的肉鸡饲料制品,选料合理,配伍性好,营养丰富,与本发明的褐藻寡糖配合,能显著提高肉鸡平均日增重和饲料利用率,耗料增重比可达1.6左右,提高了肉鸡的生长性能。
3.本发明的配合饲料选用了特定褐藻寡糖制品,显著改善了肉鸡的肠道菌群平衡,提高了肉鸡的抗氧化能力和免疫力,降低了肉鸡死淘率,提高养殖经济效益。

实施例1褐藻寡糖制品X1的制备方法
(1)水解:将市售购得的褐藻酸钠加水溶胀至质量分数为2%的均匀胶液,随后在0.2mol/L的盐酸溶液中,70℃条件下水解h,水解结束后在4000r/min条件下离心10min,取沉淀物。
(2)分级:将上述沉淀物溶解于8g/L的碳酸钠溶液中,并用盐酸溶液调节pH值为2.15,溶液分层得到黄色清液和沉淀,取上清液。
(3)醇沉:在上述上清液中加入3倍体积的95%乙醇,在4000r/min条件下离心10min,取沉淀物,抽滤除去残留的乙醇和水。样品溶于8g/L的碳酸钠溶液中,并用盐酸溶液调节pH值为2.15,得到上清液重复上述操作3次,收集沉淀。
(4)干燥:将收集得到的上述沉淀在50℃条件下热风循环烘箱烘干,即可得到褐藻寡糖制品X1。
检测:将得到的褐藻寡糖制品通过硫酸-苯酚法测定其总糖含量,并通过粘度计(具体步骤均为参照中国药典乌氏粘度法进行)和酸度计分别测定其特性粘数和pH值。经测定,得到的所述褐藻寡糖制品总糖含量平均为75.59%,特性黏数为8.13,pH值为3.57。
实施例2褐藻寡糖制品X2的制备方法
(1)水解:将市售购得的褐藻酸钠加水溶胀至质量分数为8%的均匀胶液,随后在0.6mol/L的盐酸溶液中,100℃条件下水解10h,水解结束后在4000r/min条件下离心12min,取沉淀物。
(2)分级:将上述沉淀物溶解于10g/L的碳酸钠溶液中,并用盐酸溶液调节pH值为2.85,溶液分层得到黄色清液和沉淀,取上清液。
(3)醇沉:在上述上清液中加入4倍体积的95%乙醇,在4000r/min条件下离心12min,取沉淀物,抽滤除去残留的乙醇和水。样品溶于8g/L的碳酸钠溶液中,并用盐酸溶液调节pH值为2.85,得到上清液重复上述操作2次,收集沉淀。
(4)干燥:将收集得到的上述沉淀在55℃条件下热风循环烘箱烘干,即可得到褐藻寡糖制品X2。
检测:将得到的褐藻寡糖制品通过硫酸-苯酚法测定其总糖含量,并通过粘度计(具体步骤均是中国药典乌氏粘度法进行)和酸度计分别测定其特性粘数和pH值。经测定,得到的所述褐藻寡糖制品总糖含量平均为81.35%,特性黏数为6.45,pH值为4.03。
实施例3褐藻寡糖制品X3的制备方法
(1)水解:将市售购得的褐藻酸钠加水溶胀至质量分数为15%的均匀胶液,随后在1.2mol/L的盐酸溶液中,150℃条件下水解15h,水解结束后在4000r/min条件下离心10min,取沉淀物。
(2)分级:将上述沉淀物溶解于8g/L的碳酸钠溶液中,并用盐酸溶液调节pH值为2.15,溶液分层得到黄色清液和沉淀,取上清液。
(3)醇沉:在上述上清液中加入3倍体积的95%乙醇,在6000r/min条件下离心15min,取沉淀物,抽滤除去残留的乙醇和水。样品溶于10g/L的碳酸钠溶液中,并用盐酸溶液调节pH值为3,得到上清液重复上述操作4次,收集沉淀。
(4)干燥:将收集得到的上述沉淀在60℃条件下热风循环烘箱烘干,即可得到褐藻寡糖制品X3。
检测:将得到的褐藻寡糖制品通过硫酸-苯酚法测定其总糖含量,并通过粘度计(具体步骤均为参照中国药典乌氏粘度法进行)和酸度计分别测定其特性粘数和pH值。经测定,得到的所述褐藻寡糖制品总糖含量平均为73.21%,特性黏数为7.13,pH值为4。
实施例4肉仔鸡(1-21日龄)前期料Y1
本发明的一种肉仔鸡前期料,由按重量份数的下述原料组成:实施例2所得的褐藻寡糖制品X2 0.20份、玉米58.40份、豆粕32.50份、鱼粉2.05份、大豆油1.55份、磷酸氢钙1.70份、石粉1.50份、食盐0.30份、DL-蛋氨酸0.30份和维生素矿物质预混料1.50份。
其中,维生素矿物质预混料是指提供每公斤配合饲料:维生素A 9000IU、维生素D33000IU、维生素E 24.0mg、维生素K3 1.8mg、维生素B1 2.0mg、维生素B2 5.0mg、维生素B63.0mg、维生素B12 0.1mg、烟酸40.0mg、泛酸15.0mg、叶酸1.0mg、生物素0.05mg、氯化胆碱500mg、铁80.0mg、铜20.0mg、锌90.0mg、锰80.0mg、碘0.35mg、硒0.30mg。
肉仔鸡前期料Y1的制备方法为:将玉米、豆粕粉碎过1.8mm孔径筛,按配方称重,并按比例加入实施例2所得的褐藻寡糖制品X2、鱼粉、大豆油、磷酸氢钙、石粉、食盐、DL-蛋氨酸和维生素矿物质预混料,混合搅拌均匀即可。
实施例5肉仔鸡(22-42日龄)后期料Y2
本发明的一种肉仔鸡后期料,由按重量份数的下述原料组成:实施例2所得的褐藻寡糖制品X2 0.10份、玉米62.83份、豆粕28.95份、大豆油3.50份、磷酸氢钙2.55份、石粉0.95份、食盐0.50份、DL-蛋氨酸0.12份和维生素矿物质预混料0.50份。
其中,维生素矿物质预混料是指提供每公斤配合饲料:维生素A 9000IU、维生素D33000IU、维生素E 24.0mg、维生素K3 1.8mg、维生素B1 2.0mg、维生素B2 5.0mg、维生素B63.0mg、维生素B12 0.1mg、烟酸40.0mg、泛酸15.0mg、叶酸1.0mg、生物素0.05mg、氯化胆碱500mg、铁80.0mg、铜20.0mg、锌90.0mg、锰80.0mg、碘0.35mg、硒0.30mg。
肉仔鸡后期料Y2的制备方法为:将玉米、豆粕粉碎过2.0mm孔径筛,按配方称重,并按比例加入实施例2所得的褐藻寡糖制品X2、大豆油、磷酸氢钙、石粉、食盐、DL-蛋氨酸和维生素矿物质预混料,混合搅拌均匀即可。
实验例含褐藻寡糖肉鸡饲料对肉鸡生长指标的影响
1.1试验设计和日粮
使用本发明肉仔鸡前期料Y1和肉仔鸡后期料Y2的肉仔鸡(即试验组)与对照组在生长性能、死淘率、免疫指标、抗氧化指标、肠道菌群等方面进行比较,其中对照组饲喂的饲料为Y1′和Y2′。Y1′和Y2′中分别包含与饲料Y1和Y2内褐藻寡糖制品X2含量相同的市售饲料级褐藻寡糖成品(购自青岛海之林生物科技开发有限公司),且其他成分和配比与前、后期饲料Y1和Y2相同。
选用216只1日龄、平均初始体重为43.80±0.19g的AA商品达肉仔鸡公雏,随机分为2个处理组,即Y1Y2饲喂组和对照组,每个处理组6个重复,每个重复18只鸡。整个饲养期分为两个阶段:1-21日龄为前期,22-42日龄为后期。
1.2饲养管理
试验鸡采用三层笼养(90×60×40)cm,喂给干粉料,滴头式饮水器。自由采食和饮水,自动控温、控湿。育雏期前3天室温控制在33℃,以后每周下降3℃至24℃恒温,相对湿度45%-55%,1-42d持续24h光照(10-20lux)和通风。按照肉仔鸡常规免疫程序免疫和饲养管理,每日观察鸡群健康和精神状况。所有肉仔鸡在试验的第7天和第21天接种灭活新城疫苗,第14天和第28天接种灭活法氏囊疫苗。按时打扫,保持禽舍清洁卫生。
1.3测定指标及方法
1.3.1生长性能和死淘率:试验开始时,称量并记录肉仔鸡的初始体重。于试验第42天,早上空腹称重、节料并记录死亡情况。计算平均日增重、采食量、料重比和死淘率。
1.3.2血清免疫指标:在试验结束时每个处理组随机挑选6只接近平均体重的肉鸡,心脏采血10mL于非抗凝真空采血管中,室温下倾斜放置30min后,3000r/min离心10min,得到血清测定其免疫球蛋白IgA、IgG和IgM的含量。
1.3.3血清抗氧化指标:在试验结束时每个处理组随机挑选6只接近平均体重的肉鸡,心脏采血10mL于非抗凝真空采血管中,室温下倾斜放置30min后,3000r/min离心10min,得到血清测定其总抗氧化能量、谷胱甘肽、谷胱甘肽过氧化物酶和丙二醛。
1.3.4肠道微生物菌群:与试验结束时,每个处理组随机挑选6只接近平均体重的肉鸡,屠宰后迅速打开腹腔,找到盲肠,并结扎,将带有食糜的结扎肠段,迅速放入液氮中,冷冻后置-80℃冻存,用于测定食糜中总好氧菌、总厌氧菌、大肠杆菌和乳酸杆菌数量。
2结果与分析
2.1含褐藻寡糖制品的肉鸡饲料对肉鸡生长性能和死淘率的影响

由表1数据可知,饲用饲料Y1和Y2的肉鸡,试验末重和平均日增重均高于对照组,料重比有所下降,且肉鸡的死淘率具有更明显的改善,表明含有本发明的褐藻寡糖制品X2的肉鸡饲料Y1和Y2能够更加有效地降低料重比和死淘率,能够更加有效地增重。
2.2含褐藻寡糖制品的肉鸡饲料对肉鸡血清免疫指标的影响

由表2数据可知,饲用饲料Y1和Y2的肉鸡,肉鸡血清中免疫球蛋白G水平得到了明显的提高,但免疫球蛋白A和免疫球蛋白M水平在两个组之间基本没有差异,表明含有本发明的褐藻寡糖制品X2的肉鸡饲料Y1和Y2能够更加显著地提高肉鸡的免疫力。
2.3含褐藻寡糖制品的肉鸡饲料对肉鸡血清抗氧化指标的影响

由表3数据可知,饲用饲料Y1和Y2的肉鸡,血清总抗氧化能力、谷胱甘肽含量和谷胱甘肽过氧化物酶活性均得到了较为明显的提高,而脂质氧化产物丙二醛含量低于对照组。表明含有本发明的褐藻寡糖X2的肉鸡饲料Y1和Y2,能够更加显著地提高肉鸡的抗氧化能力,减少肉鸡的氧化应激反应。
2.4含褐藻寡糖制品的肉鸡饲料对肉鸡盲肠内容物微生物菌群的影响

由表4数据可知,饲用饲料Y1和Y2的肉鸡,盲肠内容物中总好氧菌和总厌氧菌数量没有明显差异,但显著降低了有害菌大肠杆菌数量,并增加了有益菌乳酸杆菌数量。表明含有本发明的褐藻寡糖X2的肉鸡饲料Y1和Y2能够更显著地提高肉鸡肠道中有益菌增殖,降低有害菌繁殖,改善肠道微生物菌群结构更为有效。
综上,含有本发明的褐藻寡糖生物制品的肉鸡饲料,可显著改善肉鸡肠道微生物菌群结构,增强肉鸡的免疫能力和抗氧化能力,提高肉鸡的生长性能和降低死淘率。相对于具有相同成份和配比的普通市售褐藻寡糖成品鸡饲料,其在改善肉鸡氧化能力、提高免疫能力和调节肠道菌群结构方面具有更为积极的作用。
以上所述,仅为本发明较佳的具体实施方式,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,可轻易想到的变化或替换,都应涵盖在本发明的保护范围之内。因此,本发明的保护范围应以所述权利要求的保护范围为准。
Claims (10)
1.褐藻寡糖制品的提取方法,其特征在于,包括以下步骤:
在褐藻酸钠中添加水,制成均匀的褐藻酸钠胶液;
将褐藻酸钠胶液与盐酸溶液混合,水解产生沉淀;
离心取沉淀物;
将沉淀物倒入碳酸氢钠溶液中分级;
取上清液醇沉,收集沉淀;
将收集的沉淀物重复进行分层步骤和醇沉步骤多次,直至清除杂质;
将沉淀物干燥,得到藻酸钠制品。
2.如权利要求1所述的提取方法,其特征在于,
褐藻酸钠胶液的质量分数为2%-15%,盐酸溶液的浓度为0.2-1.2mol/L。

离心步骤中,离心转速为4000-6000r/min,离心时间为10-15min;干燥步骤中,干燥方式为烘干,温度为50℃-60℃。
7.如权利要求1~6中任意一项所述的提取方法制取的褐藻寡糖制品。
8.包含如权利要求7所述的褐藻寡糖制品的肉鸡饲料制品,其特征在于,包括以下重量份数的原料:
褐藻寡糖制品0.05-0.2份、玉米50-65份、豆粕25-35份、鱼粉0-3份、大豆油1-4份、磷酸氢钙1-3份、石粉0.5-1.5份、食盐0.2-0.5份、DL-蛋氨酸0.05-0.3份、维生素矿物质预混料0.5-1.5份;
所述维生素矿物质预混料是指提供每公斤配合饲料:维生素A 9000IU、维生素D33000IU、维生素E 24.0mg、维生素K3 1.8mg、维生素B1 2.0mg、维生素B2 5.0mg、维生素B63.0mg、维生素B12 0.1mg、烟酸40.0mg、泛酸15.0mg、叶酸1.0mg、生物素0.05mg、氯化胆碱500mg、铁80.0mg、铜20.0mg、锌90.0mg、锰80.0mg、碘0.35mg、硒0.30mg。
9.如权利要求7所述的肉鸡饲料制品,其特征在于,所述肉鸡饲料制品包括以下重量份数的原料组成:
褐藻寡糖制品0.20份、玉米58.40份、豆粕32.50份、鱼粉2.05份、大豆油1.55份、磷酸氢钙1.70份、石粉1.50份、食盐0.30份、DL-蛋氨酸0.30份和维生素矿物质预混料1.50份。
10.如权利要求7所述的肉鸡饲料制品,其特征在于,所述肉鸡饲料制品包括以下重量份数的原料组成:
褐藻寡糖制品0.10份、玉米62.83份、豆粕28.95份、大豆油3.50份、磷酸氢钙2.55份、石粉0.95份、食盐0.50份、DL-蛋氨酸0.12份和维生素矿物质预混料0.50份。
