SPSS20.0 software is adopted for data processing, the difference between groups is analyzed by adopting one-factor variance, and the significance difference with P less than 0.05 is considered to have statistical significance.
4 results of the experiment
(1) Determination of dehairing weight
As can be seen from Table 1: compared with a blank control group, the depilation phenomenon of other experimental mice occurs to different degrees, and the significant difference (P <0.01) exists in other groups except the significant difference of example 2 (P < 0.05); compared with the model group, the depilation phenomenon of the mice in the groups of examples 2 and 4 is improved, wherein the depilation phenomenon of the mice in the group of example 2 is improved obviously (P <0.01) compared with the model group, and the depilation phenomenon of the mice in other groups of examples has no significant difference and has no improvement trend on alopecia.
(2) Determination of the Final Hair/vellus Hair ratio in mice
As can be seen from table 2, the terminal hair/vellus hair ratio of the model group was significantly decreased (P <0.01) compared to the blank control group; examples 1, 3 did not change the terminal hair/vellus hair ratio compared to the model group, and examples 2, 4 both increased the terminal hair/vellus hair ratio, with the improvement effect of example 2 being more pronounced (P < 0.01).
(3) The scalp massage cream of the invention, example 2 (earthworm (50000) + fermented soya beans), has a significant improvement effect on seborrheic alopecia, and example 4 also has an improvement effect, but the effect is not as significant as that of example 2.

Experimental example 6: influence of scalp massage cream on hair follicle number of mice in anagen phase
1. Experimental Material
Rosin was purchased from Shenzhen Jitian chemical Co., Ltd.; paraffin wax was purchased from (opav glue wax products); adult female C57BL mice, weighing 18-22g, clean grade, were selected as pink skin and the hair was of resting age. Purchased from beijing weitonglihua experimental animal technology ltd [ license number: SCXK (Jing) 2017-0149 ], with a room temperature of 23 ℃, a humidity of 50-60%, ventilation for 2 times per day, each time lasting 1 hour, and free movement, drinking and eating of mice according to their natural life pattern.
2. Experimental methods
2.1 Experimental groups
60 female C57BL mice were randomly divided into 6 groups, negative control group, scalp massage cream group examples 1-4, and were housed in cages, and the negative control group was coated with a blank medium.
2.2 animal model preparation and administration methods
Mixing rosin and paraffin wax 1:1, heating to melt, uniformly coating on the back of a mouse, wherein the coating area is about 2cm multiplied by 3cm, cooling to solidify, and then removing the hair to remove back hair (the growth period of the hair is confirmed to be in the resting period). The next day after depilation, topical skin is directly applied with cotton swab for administration, and the administration is performed 1 time per day with dosage of 1ml each time, and is continuously performed for 20 days. On day 20, the same location of skin tissue blocks on the backs of each group of mice were taken, fixed with 10% formalin, paraffin embedded sections approximately 4-5 microns thick, HE stained, high power field (× 400) number of hair follicles averaged, and statistically processed.
3. Statistical analysis of data
SPSS20.0 software is adopted for data processing, the difference between groups is analyzed by adopting one-factor variance, and the significance difference with P less than 0.05 is considered to have statistical significance.
4 results of the experiment
(1) Effect of Hair follicle Density
The number of hair follicles in each group of mice on day 20 of the experiment is shown in Table 3. Examples 1, 3 had no effect on the number of hair follicles in C57BL mice compared to the blank control group (P > 0.05); however, examples 2 and 4 can increase the number of hair follicles in the blank mice (P <0.01), wherein the effect of example 2 is very obvious, and the hair follicles in the C57BL6 mice can be induced to enter the anagen phase, the anagen phase is prolonged, and the catagen phase is delayed, so that the hair follicle growth promoting effect is realized.
(2) The scalp massage cream of the present invention, example 2 (earthworm (50000) + fermented soya beans), had a significant effect on hair follicle growth, and the improvement of example 4 also had an improvement effect, but the effect was not significant as in example 2.

Claims (2)
1. A preparation method of a composition for treating alopecia is characterized in that the composition for treating alopecia comprises earthworm protein cut-off liquid and fermented black bean extract;
the preparation method of the composition for treating alopecia comprises the following steps:
weighing Lumbricus, adding secondary distilled water 3 times of Lumbricus, homogenizing to obtain homogenate, adjusting pH to 6.5-6.7, centrifuging at 20-30 deg.C by adding into secondary distilled water centrifuge via tubular mode, and collecting supernatant;
filtering the homogenate supernate obtained in the step two by adopting an ultrafiltration membrane with the molecular weight cutoff of 5 ten thousand to obtain earthworm protein cut-off liquid;
thirdly, fermented soya beans fermentation:
cleaning semen glycines, draining, soaking in the solution of Lumbricus protein for 10-18 hr, and sterilizing at 120 deg.C to obtain fermentation medium;
inoculating 2ml of bacillus subtilis with the concentration of 10 in every 250g of fermentation substrate7-109Mixing CFU/ml composite solution of Bacillus subtilis and Lumbricus protein solution, shaking, and culturing at 4-8 deg.C for 72-120 hr to obtain Lumbricus protein solution and fermented semen Sojae Preparatum extract.
2. The preparation method of the composition for treating alopecia is characterized in that the composition for treating alopecia also comprises a preservative, a humectant, a nutrition conditioner and a pH regulator;
the preservative accounts for 0.3 to 0.6 percent of the total mass of the alopecia treating composition, the humectant accounts for 20 to 30 percent of the total mass of the alopecia treating composition, the nutrition conditioner accounts for 40 to 50 percent of the total mass of the alopecia treating composition, and the pH regulator accounts for 0.2 to 0.3 percent of the total mass of the alopecia treating composition;
the preparation method of the composition for treating alopecia comprises the following steps:
weighing earthworm, adding secondary distilled water, homogenizing for several times, regulating pH value to 6.5-6.7, centrifuging at 20-30 deg.c in a tubular centrifuge, and collecting supernatant;
filtering the homogenate supernate obtained in the step two by adopting an ultrafiltration membrane with the molecular weight cutoff of 5 ten thousand to obtain earthworm protein cut-off liquid;
thirdly, fermented soya beans fermentation:
cleaning semen glycines, draining, soaking in the solution of Lumbricus protein for 10-18 hr, and sterilizing at 120 deg.C to obtain fermentation medium;
inoculating 2ml of bacillus subtilis with the concentration of 10 in every 250g of fermentation substrate7-109Mixing CFU/ml composite solution of Bacillus subtilis and Lumbricus protein solution, shaking, and culturing at 4-8 deg.C for 72-120 hr to obtain Lumbricus protein solution and fermented semen Sojae Preparatum extract;

the preservative is methyl paraben, benzoic acid, benzyl alcohol or potassium sorbate;
the humectant is glycerol, propylene glycol, polyethylene glycol or sorbitol;
the pH regulator is sodium hydroxide or potassium hydroxide;
the nutrition conditioner is one or a mixture of more of higher alcohol, white oil, cetyl alcohol and polyether.
