发明领域和背景

受伤的器官或组织可经由全器官移植替换。然而,主要的障碍限制了为此目的使用的外科手术,包括供体的缺乏和需要使用免疫抑制药物以防止移植器官排斥。通过植入聚合物支架、功能细胞或它们在细胞接种的支架中的组合,组织工程已作为一种改善或恢复受损组织或器官的功能或形状的有希望的方法而出现。组织工程支架的成功取决于制造设计、选择的材料,且取决于对支架的理想特性的准确理解。
在过去的二十年里,为了各种器官中的组织再生,使用天然材料或者合成材料设计和研究了各种支架。然而,组织工程正成为生物学和生物医学研究的重要研究工具,为3D培养提供各种可能性,并且从而桥接在2D培养和动物模型之间的间隙。

本发明的具体实施方案的描述
本发明,在其一些实施方案中,涉及电纺脱细胞的细胞外基质在制造支架中的用途。
在详细解释本发明的至少一个实施方案之前,要理解本发明在其应用中并不一定限制于以下描述中提出的或通过实施例举例说明的细节。本发明能够具有其它实施方案,或以各种方式实施或执行。
为组织修复和再生疗法开发的支架需要考虑组织或器官天然的适当结构和功能特征。天然ECM是为此目的的有前景的候选物,因为每个组织都包含独特的大分子组合和3D结构,其为细胞提供必要的信号和机械支持。
近年来,有提议使用完全脱细胞的细胞外基质(ECM)作为组织工程的最终生物材料,因为它是天然细胞环境的最接近的模拟物,它是生物活性的、可生物降解的和生物相容的。
然而,来自脱细胞ECM的支架难以控制或细微调节所需要的性质。它们缺乏重现性,通常具有差的机械性质,这限制了它们的使用。
为了克服这些限制,本发明人现在提出用于制造生物支架的电纺脱细胞ECM。这将不仅为细胞提供了归因于ECM的天然环境,而且还提供对纤维ECM网络的结构的控制,允许设计具有特定性质(诸如降解性、密度和机械强度)的支架。
在简化本发明以实践时,本发明人生产从ECM制作的支架,所述ECM从单个器官(胰腺或心脏)分离。
如下文和随后的实施例部分所说明的,本发明人表明在支架上接种的细胞具有类似于其来源的器官的天然ECM的多肽组成 – 表1-2。支架的平均纤维直径的范围在300-1500 nm。此外,一旦水化,纤维组构和直径变得类似于其来源的器官的天然ECM (图11A-B)。本发明人还证实,在支架上接种的细胞能够存活至少4周(图13、17、19和23)。依据本公开的方法制造的支架被证明是非免疫原性的(图25-27)。
因此,根据本发明的一个方面,提供一种产生支架的方法,其包括:
(a)在有机溶剂中溶解器官的脱细胞的细胞外基质(ECM),以生成脱细胞ECM的溶液;和
(b)将溶液电纺到固体表面上,从而产生支架。
本文所用的短语“脱细胞ECM”指细胞外基质,其支持组织构造(如,天然组织)并经受脱细胞过程(即,从器官除去所有细胞),从而完全没有任何细胞组分。
脱细胞ECM通常包含多种多肽(如胶原)。例如,脱细胞ECM包含胶原α-2(I)、胶原α-1(III)和胶原α-1(I)。另外地,脱细胞ECM还可包含胶原α-2(IV)、胶原α-1(V)和胶原α-1(II) (或其片段)。优选地,在本发明的这个方面的脱细胞ECM中,胶原α-2(I)、胶原α-1(III)和胶原α-1(I)的量大于胶原α-2(IV)、胶原α-1(V)和胶原α-1(II)。
另外地,脱细胞ECM还可包含较小量的胶原α-2(VI)、胶原α-3(VI)和胶原α-1(VI)或胶原α-1(IV) (或其片段)。优选地,在本发明的这个方面的脱细胞ECM中,胶原α-2(I)、胶原α-1(III)和胶原α-1(I)的量大于胶原α-2(VI)、胶原α-3(VI)和胶原α-1(VI)或胶原α-1(IV)。
本文所用的短语“完全没有任何细胞组分”指自然(如,天然)组织中存在的细胞组分的超过90 %、91 %、92 %、93 %、94 %、95 %、96 %、97 %、98 %、99 % (如,100 %)缺失。本文所用的,短语“细胞组分”指组成细胞的细胞膜组分或细胞内组分。细胞组分的实例包括细胞结构(如,细胞器)或包含在其中的分子。这样的实例包括,但不限于组织的细胞中存在的细胞核、核酸、残余核酸(如,片段化的核酸序列)、细胞膜和/或残余细胞膜(如,片段化的膜)。应该意识到,由于从组织中除去了所有的细胞组分,这样的脱细胞基质在植入受试者中时不能诱导免疫学反应。
本文所用的短语“细胞外基质(ECM)”指由组织细胞产生和分泌到周围的胞外空间和/或介质中的材料的复杂网络,并且其通常与组织的细胞一起赋予组织其机械和结构特性。一般来说,ECM包括纤维元件(特别是胶原、弹性蛋白或网硬蛋白)、细胞粘附多肽(如,纤连蛋白、层粘连蛋白和粘着糖蛋白),和空间填充分子[通常为葡糖胺聚糖(GAG)、蛋白聚糖]。
根据本发明的实施方案,脱细胞ECM源自心脏或胰腺组织。
本发明人所构思的其它组织包括脑组织、骨组织、肌肉、肝、肾、血管、肺和胎盘。
在其它实施方案中,脱细胞ECM不源自脂肪组织
在另外的其它实施方案中,脱细胞ECM不是MATRIGEL或源自分离的基底膜。
ECM可源自自体或非-自体受试者(如,由于生成的脱细胞基质的非免疫原性,其源自同种异体或甚至异种组织)。组织从受试者[如,动物,优选哺乳动物,例如猪、大鼠、猴或黑猩猩,或者作为选择,死亡的人(死后不久)]中取出并例如在无菌盐水溶液(0.9 % NaCl,pH = 7.4)或磷酸盐缓冲盐水(PBS)中洗涤,所述溶液或盐水可补充有抗生素例如青霉素/链霉素250个单位/ml。虽然可使用完整组织,但对于几种应用,组织的区段可以切割,例如切片。这样的组织区段可以具有不同的尺寸,这取决于使用的原始组织和所需的应用。
为去除围绕组织和供养组织的血管,组织可于室温下通过在大量(如,50 ml/每克组织区段)的EDTA溶液(0.5-10 mM, pH-7.4)中搅动来进行洗涤。
接着,组织经受高渗或低渗缓冲液,从而获得组织内增加的细胞间空间。
本发明使用的高渗缓冲液可以是溶质浓度高于存在于组织内的细胞质和/或细胞间液体的溶质浓度的任何缓冲液或溶液[如,高于0.9 % (w/v)的NaCl浓度]。由于渗透作用,用高渗缓冲液孵育组织导致组织内的细胞间空间增加。
根据另一种实施方案,过乙酸被用来使组织脱细胞化。
优选地,根据本发明的这个方面的方法使用的高渗缓冲液包括浓度高于0.9 %(w/v),优选高于1 % (w/v),优选在1-1.2 % (w/v)的范围内,如1.1 % (w/v)的氯化钠(NaCl)。
优选地,根据本发明的这个方面的方法使用的低渗缓冲液包括浓度低于0.9 %(w/v),低于0.8 % (w/v),低于0.7 % (w/v),优选在0.6-0.9 % (w/v)的范围内,如0.7 %(w/v)的氯化钠(NaCl)。
根据本发明的这个方面,组织经受高渗或低渗缓冲液一段时间,所述时间导致生物效应,即细胞皱缩,其导致组织内的细胞间空间增加。
根据一个特定的实施方案,组织与高渗缓冲液(如1.1% w/v)接触,随后与低渗缓冲液(如0.7 % w/v)接触。这个程序可以重复两个或更多个周期。
优选地,根据本发明的这个方面的方法使用的低渗缓冲液包括浓度低于0.9 %(w/v),低于0.8 % (w/v),低于0.7 % (w/v),优选在0.6-0.9 % (w/v)的范围内,如0.7 %(w/v)的氯化钠(NaCl)。
在用高渗/低渗缓冲液孵育后,使组织进一步经受酶蛋白水解消化,其消化组织内的所有的细胞组分,但保留ECM组分(如,胶原和弹性蛋白),因此产生表现原始组织ECM的机械和结构性质的基质。应该意识到,要采取措施在消化组织的细胞组分的同时保留ECM组分。这些措施在下文进一步描述并包括,例如,调整消化溶液内的活性成分(如,胰蛋白酶)的浓度以及孵育时间。
根据本发明的这个方面的蛋白水解消化可使用各种蛋白水解酶进行。合适的蛋白水解酶的非-限制性实例包括从各种来源例如从Sigma (St Louis, MO, USA)获得的胰蛋白酶和胰酶。根据本发明的这个方面的一个优选实施方案,蛋白水解消化使用胰蛋白酶进行。
用胰蛋白酶消化优选在范围从0.01至0.25 % (w/v),更优选0.02-0.2 % (w/v),更优选0.05-0.1 (w/v)的胰蛋白酶浓度,甚至更优选约0.05 % (w/v)的胰蛋白酶浓度下进行。例如,含有0.05 %胰蛋白酶(w/v; Sigma)、0.02 % EDTA (w/v)和抗生素(青霉素/链霉素250个单位/ml), pH = 7.2的胰蛋白酶溶液可被用来有效地消化组织的所有细胞组分。
应该意识到,为了有效消化组织的所有细胞组分,每个组织区段可以40 ml消化溶液/每克组织的比例,放在含有消化溶液(如,如上所述的胰蛋白酶溶液)的单独容器里。优选地,在消化溶液中的时候,缓慢地搅拌组织区段(如,以约150 rpm),以使消化溶液完全渗透到组织的所有细胞中。
应该注意到,消化溶液的浓度和其中的孵育时间取决于要处理的组织类型和所用的组织区段的大小,本领域技术人员能够依据所需的组织大小和类型调整条件。
优选地,组织区段孵育至少约20小时,更优选至少约24小时。优选地,消化溶液被替换至少一次,以使在消化溶液中的总孵育时间为至少40-48小时。
接着,从组织除去细胞组分。从组织除去消化的组分可使用各种洗涤溶液进行,例如洗涤剂溶液(如,离子和非离子洗涤剂例如SDS Triton X-100、Tween-20、Tween-80),其可从例如Sigma (St Louis, MO, USA)或Biolab (Atarot, Israel, Merck Germany)获得。
优选地,根据本发明的这个方面的方法使用的洗涤剂溶液包括TRITON-X-100 (从Merck获得)。为了有效除去所有消化的细胞组分,TRITON-X-100以在0.05-2.5 % (v/v),更优选在0.05-2 % (v/v),更优选在0.1-2 % (v/v)的浓度范围,甚至更优选以1 % (v/v)的浓度提供。
优选地,为了最佳结果,洗涤剂溶液还包括氢氧化铵,其与TRITON-X-100一起帮助破裂和溶解细胞核、骨骼蛋白和膜。
优选地,氢氧化铵以0.05-1.5 % (v/v)的浓度,更优选以0.05-1 % (v/v)的浓度,甚至更优选以0.1-1 % (v/v) (如,0.1 %)的浓度提供。
TRITON-X-100和氢氧化铵在洗涤剂溶液中的浓度可取决于要处理的组织的类型和大小而变化,本领域技术人员能够依据所用的组织调整这样的浓度。
用洗涤剂溶液孵育组织(或组织区段)可持续数分钟至数小时,甚至数天,这取决于组织的类型和大小和所用洗涤剂溶液的浓度,本领域技术人员能够调整这样的孵育时间。优选地,用洗涤剂溶液孵育进行至少24-72小时。根据一个实施方案,执行用洗涤剂溶液孵育2-4个周期,直至未观察到泡沫,以使总孵育时间可以在约150-200小时之间。
虽然如上所述,用洗涤剂溶液孵育能够除去细胞核、蛋白和膜,但根据本发明的这个方面的方法任选地包括从组织除去核酸(以及残余核酸)的另外的步骤,从而获得无核酸的组织。本文所用的短语“无核酸的组织”指如使用常规方法(如,分光光度法、电泳)测定的,99 %以上的任何核酸或其片段不存在的组织。这样一个步骤使用DNase溶液(也任选使用RNase溶液)。合适的核酸酶包括DNase和/或RNase [Sigma, Bet Haemek Israel, 20 μg/ml,在Hank平衡盐溶液(HBSS)中]。
上述洗涤剂溶液优选通过将ECM在水或盐水中多次清洗(如,至少10次洗涤,每次30分钟,和2-3次洗涤,每次24小时),直至基质中没有明显的洗涤剂溶液来去除。
任选地,然后对脱细胞ECM灭菌。脱细胞ECM的灭菌可使用本领域已知的方法(如70%乙醇)进行。
通常地,为了进行脱细胞ECM的溶解,将其冷冻(如在液氮中),切成小片(如粉碎、压碎或碾碎),然后冻干。
然后使冻干的脱细胞ECM溶于有机溶剂中。
本发明人所构思的示例性有机溶剂包括,但不限于丙酮、N,N-二甲基甲酰胺(DMF)、二乙基甲酰胺、氯仿、甲乙酮、乙酸、甲酸、乙醇、1,1,1,3,3,3-六氟-2-丙醇(HFIP)、四氟乙醇、二氯甲烷(DCM)、四氢呋喃(THF)、三氟乙酸(TFA)、樟脑磺酸、二甲基乙酰胺、异丙醇(IPA)及其混合物。
本发明人所构思的脱细胞ECM在有机溶剂中的示例性浓度是在0.005 g/mL-0.5g/mL之间,例如约0.05 g/mL。
根据特定的实施方案,有机溶剂是HFIP。
为帮助溶解过程,溶解后或与溶解同时,可以使脱细胞ECM匀化。
通常地,匀化在刚性研磨介质的存在下进行,所述介质优选为具有小于约10 mm(如在2-10 mm之间),且更优选在2-7 mm之间的平均大小的球形或微粒形式。不认为研磨介质的材料的选择是关键的。氧化锆(例如用氧化镁稳定的95% ZrO)、硅酸锆、陶瓷、不锈钢、氧化钛、氧化铝、用钇稳定的95% ZrO、玻璃研磨介质和聚合物研磨介质是示例性的研磨材料。

根据这个方面,脱细胞ECM可源自任何组织例如心脏组织、胰腺组织、血管组织、肌肉组织、肝组织、肾组织、脑组织、骨组织、肺和胎盘。
优选地,脱细胞ECM源自胰腺组织或心脏组织。
当脱细胞ECM源自心脏组织时,脱细胞ECM维持天然心脏ECM的蛋白组成,或至少具有与天然心脏ECM类似的蛋白组成。因此,例如脱细胞ECM的大部分胶原为I型和III型胶原,因为那是心脏组织的最丰富的胶原。
在另一个实施方案中,脱细胞ECM缺乏VI型胶原。

可掺入本发明的支架中的蛋白包括,但不限于细胞外基质蛋白、细胞粘附蛋白、生长因子、细胞因子、激素、蛋白酶和蛋白酶底物。因此,示例性的蛋白包括血管内皮衍生生长因子(VEGF)、激活素-A、视黄酸、表皮生长因子、骨形态发生蛋白、TGFβ、肝细胞生长因子、血小板衍生生长因子、TGFα、IGF-I和II、造血生长因子、肝素结合生长因子、肽生长因子、红细胞生成素、白细胞介素、肿瘤坏死因子、干扰素、集落刺激因子、碱性和酸性成纤维细胞生长因子、神经生长因子(NGF)或肌肉形态发生因子(MMP)。所用的具体生长因子应适合所需的细胞活性。生长因子的大家族的调节效果是本领域技术人员熟知的。
本发明的支架可用细胞接种,包括例如原代细胞、培养细胞、细胞的单细胞悬浮液、细胞簇(如胰岛)、包含在组织和/或器官中的细胞等。
细胞可通过静态加载,或者更优选通过在搅拌的烧瓶生物反应器中接种(支架一般悬挂在固体支撑物上),在旋转壁容器中接种,或使用细胞在生物反应器的介质中的直接灌注,接种在支架中。通过后一种(直接灌注)技术,在整个支架中达到最高的细胞密度。
细胞可直接接种到支架上,或者作为选择,细胞可与凝胶混合,然后将凝胶吸收到支架的内部和外部表面并可填充支架的一些孔。在硬化前,毛细力将凝胶保留在支架上,或者凝胶可以被允许在支架上硬化,以变得更能自我支持。或者,细胞可与任选地包含细胞外基质组分的凝胶形式的细胞支持基质合并。示例性的凝胶是得自Becton-Dickinson的MatrigelTM。MatrigelTM是从EHS小鼠肿瘤提取的可溶性基底膜基质(Kleinman, H. 实施例1
材料和方法
脱细胞程序:根据先前公布的一个方案21(Chaimov 2016, PMID: 27476611),使猪心脏或胰腺ECM脱细胞并灭菌。简言之,分离健康的商业屠宰称重猪的组织,用于脱细胞程序。程序包括具有以下阶段的两个周期:交替的高渗/低渗NaCl溶液;使用胰蛋白酶的酶法处理;和用Triton-X-100的洗涤剂清洗。
ECM溶解:在液氮中冷冻脱细胞的灭菌ECM,在低温组织研磨机(BioSpec,Bartlesville, OK, USA)中粉碎,随后置于冷冻干燥机中直至干燥。使冷冻干燥的ECM溶于六氟异丙醇(HFIP)中至0.05 g/ml的浓度,在PrecellysTM 24珠粒均化器(BertinTechnologies, Rockville, MD, USA)上,以6000 rpm使用ZrO珠粒匀化,直至溶液呈现匀相。在匀化后,胰腺ECM经超声处理直至3分钟。然后在电纺前,使溶液通过玻璃纤维过滤。
电纺条件:
只将PEO加入到猪心脏(pcECM)溶液中,获得0.1质量% PEO的最终溶液。采用定制的电纺装置对猪胰腺ECM (ppECM)或pcECM/PEO溶液进行电纺。用8-9 kV的电压对纤维进行电纺,并收集在覆盖旋转铝盘(直径9 cm和宽度11 mm)的聚乙烯薄膜上。毛细管是23规格钝针,从针到收集器的距离是6-10 cm,流速是0.5-1 ml/hr。收集纤维直至观察到一个厚层。然后将该层从表面剥落并放在干燥的环境中。通过在水-基溶液中洗涤,从pcECM基质除去PEO,同时使pcECM纤维保持完整。
SEM:使用配备有Schotkky场发射电子枪的FEG Zeiss Ultra Plus高分辨率扫描电子显微镜(HR-SEM, Zeiss, Jena, Germany)使样品可视化,以提供克服了未涂层非导电样本的荷电的高水平亮度。显微镜还配备有电荷补偿系统以进一步提高这个效果。使用胶粘碳双面胶带将样品固定在铝制刺桩(stabs stubs)上,使用in-lens型和SE2型探测器的组合,以1.3 kV加速电压捕获图像。纤维直径的分析通过获得从支架的6个随机选择区域获取的图像间随机选择的20次测量的平均值测定,所述图像至少放大5kX。数字图像使用ImageJ软件(版本1.49)和Porometric软件(PhenomWorld)评价分析。
使用配备有温控样本固定架(Deben UK Ltd., Suffolk, UK)的Phenom ProXdesktop SEM (PhenomWorld, Eindhoven, Netherlands),使湿支架可视化。使用组织冷冻介质(Ted Pella, Inc., CA, USA),将样品固定铝刺上,并冷却至-24℃。以15 kV加速电压捕获图像。
接触角:ECM电纺纤维支架的接触角通过将直径近1 mm的液滴置于表面上来测定。然后用Artcam 130 MIBW摄像机(Artray Co. Ltd., Tokyo, Japan)捕获静态图像。然后使用MatLab R2014a软件(Mathworks, Natick, MA, USA)开发的特定程序计算接触角。
FTIR:电纺ECM支架和脱细胞ECM的傅立叶变换红外光谱(FTIR),使用配备有SmartiTR Attenuated Total Reflectance (ATR)金刚石板的Thermo 6700 FTIR仪器,以500-3500 cm-1的波数范围(以4 cm-1的分辨率扫描64次,n=4)记录。数据使用OMNICTM系列软件(版本8, Thermoscientific)评估。蛋白的二级结构通过通过其I中波段的傅里叶去卷积来确定。
TGA:热重分析(TGA)数据使用TGA-Q5000系统(TA Instruments, USA)获得。在氮气氛下,以20℃ min-1的速率加热样品,从RT至最终温度600℃。数据使用TA UniversalAnalysis Software (TA Instruments, New Castle, DE, USA)分析。
电纺ECM支架组成:蛋白组成的分析在Proteomics Center, Technion – IsraelInstitute of Technology进行。样品用胰蛋白酶消化,生成的肽通过LC-MS/MS分析。此后,肽混合物通过HPLC分级并经电喷雾到离子陷阱质谱仪上,以测定蛋白质量。通过碰撞引起的离解使肽进一步片段化,并再次分析,以供另外的分析和鉴定。利用ProteomeDiscovererTM软件(Thermo-Scientific),对比UniProt数据库中的猪部分来分析和鉴定肽。
机械测试:电纺3D纤维支架的抗拉性质使用动态机械分析仪(DMA) Q800 (TAInstruments, New Castle, DE, USA)在环境条件下测定。支架被切成4.3 ± 0.2 mm x6.7 ± 0.3 mm的矩形样本,垂直固定在生理缓冲液中的两个机械夹持单元之间。然后应用2 % min-1的伸长率直至屈服点。使用TA Universal Analysis 2000 v. 4.5 A软件(TAInstruments)记录应力-应变曲线,并从图外推最终应力、应变和最终应力和在2%应变时的杨氏模量。
电纺(ES)-pcECM支架上培养的细胞:将支架切成圆形(D = 0.18 cm),置于96-孔板,并使用紫外光灭菌。加入培养基,板然后被移入一个加湿的孵育器(37℃, 5% CO2)中。1小时后,洗掉培养基,用新鲜培养基替换,将板返回孵育器过夜以供润湿。第二天,接种细胞。
将人骨髓间充质干细胞(hMSC, Lonza, Basel, Switzerland)接种(10,000个/支架)并培养1个月。在补充有FCS (10%)、青霉素-链霉素(1%)、两性霉素B (0.4%)和碱性成纤维细胞生长因子(5 ng mL-1)的αMEM中培养hMSC。每隔一天更换培养基。依据制造商的方案,使用AlamarBlue™试剂(AbD Serotec, Kidlington, UK),评估细胞生存能力。接种hiPSC(30,000个/支架)并培养1个月。hiPSC在mTeSRTM基础培养基中培养并补充有mTeSRTM 1 5x补剂。每天替换培养基。从24小时龄的Wister大鼠分离新生心肌细胞。将切除的心脏切碎,于37℃,通过在200U mL-1 RDB/PBS-G (PBS、青霉素-链霉素、0.1% D-葡萄糖)中温和搅动10min,分离心细胞。细胞悬浮液以1000 rpm离心5 min,悬浮于补充有5%胎牛血清、5% DHS、1%青霉素-链霉素和0.4%两性霉素B和1mM CaCl2的F-10营养混合物中。将细胞悬浮液预铺板在培养皿中并孵育1 hr,以允许成纤维细胞附着。收集非-附着的富含心肌细胞的细胞悬浮液,如前所述离心,并再悬浮于培养基。将新生心肌细胞接种在支架(200,000个细胞/支架,n=12)上并在补充的F-10营养混合物中培养3周。每隔一天更换培养基。
光片荧光显微术(LSFM):为研究在电纺ECM支架上培养的细胞的形态学,用FDA(对于活细胞),和用Hoechst 33258 (对于DNA)染色细胞。然后将支架包埋在低熔点琼脂糖凝胶(Bio-Rad, Haifa, Israel)中并插入1 mL注射器内。然后使用光片荧光显微术(LSFM),使用配备有具有两个对齐的物镜的双面光束照明(用于完整的高分辨率的3D成像)、两个相机和用于活样品的完整孵育隔室的Lightsheet Z.1 (Zeiss),观察样品
组织学:将细胞-接种的支架固定在PFA (4%)中20 min,在PBS中洗涤并在Tissue-TekTM OCT化合物中冷冻,在玻片上横切成薄片(10 µm)并染色。在染色前,将玻片固定在冷的MeOH (4℃)中20 min。固定后,在DDW中洗涤玻片3-5次,以除去所有的OCT化合物,并用苏木素和伊红(H&E)染色。依据制造商的方案,使用cTn1、肌节α-辅肌动蛋白和连接蛋白-43第一抗体,对心脏标记进行免疫荧光染色。通过倒置相差显微术(Eclipse TE2000-E, NikonInc.)观察玻片。
Ca2+成像:在Pluronic F-127 (Molecular Probes)的存在下,以2:1的稀释度,用5mM fluo-4荧光Ca2+指示剂(Molecular Probes)加载细胞,以允许记录细胞内Ca2+-瞬态(全-细胞[Ca2+]瞬态)。对于起搏,将支架平铺在带有场模拟电极(RC-37FS; WarnerInstruments)的35-mm光板(Matek)上,并使用刺激隔离装置(SIU-102; WarnerInstruments),通过施加5 ms-超阈值双极刺激脉冲直至50 mA起搏。细胞内Ca2+-瞬态使用安装在配备有X60水物镜的正置BX51WI Olympus显微镜上的Zeiss激光扫描共焦成像系统(Fluo-view; Olympus)记录。数据利用基于MatLab的定制编写软件分析。
基于pcECM的支架的MSC重塑:通过在基于pcECM的支架上接种的细胞的ECM重塑-相关基因的表达用实时RT-PCR定量研究21天。研究了以下基因:胶原I (α1链)、胶原III (α1链)、基质金属蛋白酶2 (MMP2)和金属蛋白酶的1型组织抑制剂(TIMP1)。总RNA在不同的时间点,依据生产商的说明书,使用Tri-试剂(Sigma-Aldrich)从接种的细胞中分离,并在PTC-200 PCR循环仪中使用VersoTM cDNA试剂盒(Thermo-Scientific)逆转录。引物经设计以特异性扩增基因的cDNA,如下所示:

体外免疫原性分析:巨噬细胞刺激被用来评估ECM电纺支架在体外的免疫原性。测量分泌的氧化氮(NO)的水平和促炎细胞因子的表达二者。RAW细胞系(TIB-71™; ATCCManassas, VA, USA)在24-孔细胞培养板中接种并在补充有10 % FCS、1 %青霉素-链霉素和0.4 % 两性霉素B的1 mL高葡萄糖DMEM中培养。接种后第二天,培养基用低血清培养基(2% FCS)替换。当细胞达到70 %汇合时,使细胞暴露于以下物质:电纺ECM支架(1 mg),作为阴性对照的PLGA (1 mg),或作为阳性对照的LPS (1 µg/mL)。在16 h孵育后,使用常规的Griess Reagent Assay (参考),测量作为在培养基中的游离稳定的亚硝酸盐形式(NO2 -)的分泌的NO水平。未处理的细胞也用作阴性对照(基础NO分泌)。另外地,总RNA依据制造商的方案,使用Tri-试剂(Sigma Aldrich),从接种的RAW巨噬细胞分离,并在PTC-200 PCR循环仪中使用VersoTM cDNA试剂盒(Thermo-Scientific, Waltham, MA, USA)逆转录。然后用以下特异性引物,分离的RNA被用于实时(RT-) PCR分析,以对促炎细胞因子TNF-α和IL1-β的表达进行定量。

体内免疫原性研究:根据Israeli Animal Welfare (Protection andExperimentation) Law,在获得Technion’s Animal Care Committee的允许后,进行免疫原性实验。ECM电纺纤维支架的免疫原性潜力在体内通过皮下植入另外进行评价。小鼠被分成两组:一组接受电纺ECM支架,而另一组接受电纺PLGA支架作为阴性对照。每组分成三个时间点(1周、2周和4周)。每一组每个时间点有5只小鼠。
在手术当天,6-周龄C57BL小鼠(Pharma Medis Ltd, Holon, Israel)用300 µL腹膜注射氯胺酮(100 mg/kg) (Vétoquinol, Lure, France)和赛拉嗪(5 mg/kg) (PhibroIsrael, Beit Shemesh, Israel)进行麻醉,在右侧腹剃毛,随后为了镇痛经由皮下注射给予150 µL丁丙诺啡(0.05 mg/kg) (Vet Market, Shoham, Israel)。在手术过程中,异氟烷/氧混合物(2-3 %异氟烷)通过给气面罩给予,用于维持。小鼠被置于加热垫(37℃)上,在剃毛的右侧腹上进行皮下切口,通过切口插入相应的支架。缝合切口,将小鼠放入富氧的温度控制(X℃)环境中。按常规监测小鼠,并在各自的时间点处死。血样在尸检后从心脏和/或股动脉采集以供全血计数(CBC),腹股沟淋巴结,分析支架。匀化淋巴结,提取mRNA并逆转录,如前所述定量促炎细胞因子(TNFα和IL-1β水平正常化至GAPDH)。
结果
ECM溶解和电纺
如在WO 2006095342A2和在本文上面的材料和方法章节中所述,从脱细胞的猪组织的切片获得ECM,其避免使用SDS并允许改进ECM生物活性的保存。在液氮中冷冻ECM,在低温组织研磨机中研磨,并冻干。使用生成的粉末以供溶解。
使冻干的ECM溶于HFIP至0.05 g/mL浓度并向pcECM溶液中加入0.1质量% PEO。无PEO时,pcECM电纺导致纤维和枝状物的组合(图1A);而在有PEO时,获得均质纤维的网络(图1B)。通过在水-基介质中洗涤从基质容易地除去PEO,并保持pcECM纤维完整。
支架形态学和特征鉴定:HR-SEM图像被用来检查ECM电纺纤维支架内的纤维的形态学和直径(图2A-D、图3A-B)。在较低的放大倍率(0.2-1 kX)下,电纺pcECM支架的纤维表现为随机分散的,具有极少的非纤维集合体,其被怀疑是没有完全溶解的胶原片(图2A-B)。在较高的放大倍率(5 kX)下,显现两类纤维,一类是相当厚的,而另一类是薄的。在甚至更高的放大倍率(10 kX)下,各个纤维的多孔结构的形态学是可见的(图2D)。根据imageJ分析,较大纤维的直径是约734 ± 228 nm,范围从300至1500 nm,显示出与天然ECM10相一致的正态分布(图5)。较薄纤维的直径是约43 ± 7.9 nm,其仍然落入在天然ECM中所见到的纤维直径的范围内。电纺pcECM支架的孔径的范围在1-30 μm2之间,平均7.6± 1.7 μm2 (图6)。
ECM电纺纤维支架的接触角通过在表面滴1 mm DDW的液滴来观察,并捕获静态图像。分析使用自设计的MatLab程序执行。电纺pcECM和ppECM支架的水接触角分别是96.33± 2.12°或74.3。在不到5分钟后,电纺pcECM的接触角下降至60°(图7A-B和图8)。
ECM电纺纤维支架组成:因为蛋白组成是有利于支架的生物活性和机械性质的主要因素,首先通过比较从脱细胞pcECM获得的FTIR谱与ES-pcECM支架的那些谱,评价蛋白组成的可能改变(图9A-B)。两种材料显示出在接近和超过3000 cm-1的酰胺振动,这是肽基的特征,和500-1700 cm-1之间的振动,这是氨基酸侧链的特征,在脱细胞pcECM和ES-pcECM支架之间无显著差异。而且,每个二级结构的类似的百分率用傅里叶去卷积来测定。进行蛋白质组质谱分析以确保在溶解或电纺过程中蛋白未损失。因为胶原是ECM中最丰富的蛋白,并提供必要的抗拉强度和粘弹性,对胶原含量进行了全面检查(表1、表2和图3)。

表2提供了电纺猪胰腺ECM (ppECM)支架的胶原组成,如蛋白质组分析所示。
所有的胶原类型依据它们的质谱结果,根据其丰度水平分组;水平1表示最丰富的胶原。当检测到显著的倍数降低时,胶原就被归入随后的水平。例如,水平2中的所有胶原的丰度水平不到水平1中的所有胶原的1/3。水平3胶原的丰度不到水平1中的1/20和水平2中的1/3,而水平4胶原的丰度不到水平1中的1/100,水平2中的1/60,和水平3中的1/8。这些分组允许对pcECM电纺支架的胶原组成进行更深入的分析。水平1仅含有I和III型胶原,这是期望的,因为I和III型胶原(通常在一起发现)是心脏ECM组织内最丰富的胶原(> 90 %)15。它们是维持组织结构和支持心肌细胞的原纤维、间质胶原类型。
水平2和3含有不太丰富的原纤维形成胶原(II和V),和存在于基底膜中的网络形成胶原(IV)。水平3还含有VI型胶原,其提供组织原纤维胶原并使它们锚定在基底膜上的微丝网络。胶原α-2 (VI)是显示pcECM电纺支架比脱细胞pcECM显著倍数降低的唯一胶原。一般来说,已确定除了胶原α-2 (VI)外,在脱细胞pcECM和pcECM电纺支架之间的胶原含量没有显著的倍数差别。
TGA分析(图10),比较了支架的特征性热降解与脱细胞pcECM的特征性热降解。两种材料的分解起始(Tonset)相似:259.4℃ (支架)和259.6℃ (脱细胞pcECM),表示ES-pcECM支架的生产过程,和特别是pcECM在HFIP中的溶解,没有降解胶原。
电纺pcECM的自组装:在润湿ES-pcECM支架后,电纺纤维经历了自组装,因此不需要合成交联剂(图11)。自组装的程度依据初始润湿温度以及随后的孵育温度而变化。当ES-pcECM支架维持于37℃时,获得最大程度的自组装,其中ES-pcECM自组装产生一种各向同性的、多孔的、有序的结构,其最清晰和独特地类似于天然pcECM的微结构。维持于24℃的ES-pcECM支架显示出较低程度的自组装,而维持于4℃的那些显示出最低的自组装,具有电纺纤维的各向异性的结构仍然是很明显的。增加初始湿化后的孵育温度(从4℃和24℃)至37℃增加它们的自组装;然而,未实现完全自组装,电纺纤维在某种程度上仍然存在。这些研究提供了关于ES-pcECM支架的能力的大量数据,这用其它天然聚合物的电纺没有观察到。
电纺pcECM支架的机械性质:在解决心脏再生问题时,设计的支架的机械性质是特别重要的。在生理学溶液中将电纺pcECM支架的机械性质与天然组织的那些比较(图12 A-D)。应力和最大应力的应变在电纺pcECM支架和天然心脏组织中是相似的(p>0.05)。支架的杨氏模量(E) (0.29±0.007 MPa)也比得上天然组织的杨氏模量(0.22±0.007),虽然有统计学差异(p<0.05)。这样的结果证实,电纺pcECM支架的自组装足以获得为心脏再生所需的机械性质,而无需合成的交联剂。
在pcECM电纺纤维支架上培养hMSC:细胞对pcECM电纺纤维支架的附着和在其上增殖的能力根据人间充质干细胞(hMSC)的生存能力评价。生存能力研究证实,pcECM电纺支架支撑hMSC,其保持80,000个细胞/支架的平均密度达到4周(图13)。
hMSC描绘正常的拉长形态和支架贯穿(图14A-D)。在支架上培养1个月后出现粘连的存活hMSC也得到光片荧光显微术(LSFM)分析的支持(图15A-B)。苏木素和伊红(H&E)组织学分析表明,细胞已经整合到基质中(图15C)。
接种的hMSC重塑电纺pcECM支架的能力通过分析它们的ECM-重塑相关基因的表达来评价(图16)。在天然ECM和电纺ECM两种支架中,胶原I表达与基础水平比较在接种后3、7和14天增加10-20倍。这样的升高的表达到21天时降低。胶原III表达的最大增加对于天然ECM在第3天观察到(13倍),而对于电纺ECM支架在第21天观察到(18倍)。TIMP1 (胶原酶抑制剂)最大表达对于电纺支架在第7天观察到(25倍),而对于天然ECM在第14天观察到(7倍)。MMP2,一种指示剂胶原酶,在天然ECM和电纺ECM支架二者中在第7天增加,并在第14天达到最大表达。
人诱导的多能干细胞(hiPSC)在电纺ECM支架上接种,在培养3周后表现出汇合的基质(图17),通过小部分的死亡细胞(碘化丙啶,粉红色)证实令人印象深刻的存活水平。CryoSEM图像显示呈规则集群形态的粘着hiPSC (图18A-C),和H&E分析显示它们在基质内的整合(图18D)。
在电纺pcECM支架上接种的新生大鼠心肌细胞(rCM)在21天后使用SEM和LSFM评价(图19A-B),显示了正常的球状形态学。H&E染色证实细胞在基质内的整合(图19C)。接种的rCM对典型的功能心脏蛋白心肌钙蛋白I (cTn1)、肌节α-辅肌动蛋白和连接蛋白-43染色阳性(图20A-C),显示收缩功能以及通过功能缝隙连接的细胞-细胞耦合。这种收缩功能还通过接种的支架的自发性跳动得到了证明,其在接种后不到2天启动。约2周后,利用Ca2+成像对同步跳动支架进行分析以记录跳动和测试电耦合。在不同速率的起搏(使用场刺激)过程中评估的动作电位特性(图21)进一步证实电纺pcECM支架可支持细胞的完整同步电活动。
另外类型的细胞―hiPSC-衍生的心肌细胞(hiPSC-CM)―在电纺pcECM支架上的培养揭示接种后14天和3周的高存活水平(图22、图23)。通过接种的支架的自发性跳动也证明了收缩功能,其在接种后不到24 hr启动。约2周后,同步跳动支架使用Ca2+成像分析以测试电耦合。如在图24中所见到的,在1 Hz速率的起搏(使用场刺激)过程中获得全-细胞[Ca2+]瞬态,显示为线扫描示踪,并进一步确认电纺pcECM支架可支持细胞的完整同步电活动。
免疫原性研究
pcECM电纺支架的免疫原性在体外和体内检查。用粉碎的pcECM支架、PLGA (阴性对照)、脂多糖(LPS-阳性对照)刺激RAW巨噬细胞。非-刺激的细胞也用作阴性对照。与刺激的材料一起孵育16 h后,评价分泌的氧化氮(NO)水平(图25A)。在pcECM支架、PLGA和非-刺激的细胞之间没有观察到分泌的NO水平的显著变化(P > 0.05)。然而,与pcECM支架和两个阴性对照比较,用LPS刺激细胞的分泌的NO水平显著更高(P < 0.01),这证实LPS是成功的阳性对照。在刺激后16 h,还用实时(RT-) PCR分析评价了RAW巨噬细胞的TNF-α和IL1-β的促炎细胞因子表达(图25B, C)。获得与NO分泌类似的结果。对于用pcECM支架刺激的RAW巨噬细胞,促炎细胞因子TNF-α的表达显示出1.66 ± 0.64倍增加,而对于分别用PLGA和LPS刺激的RAW巨噬细胞显示出0.73 ± 0.65倍和67.5 ± 32.6倍增加(相对于非-刺激的细胞)。只有用LPS刺激的RAW巨噬细胞显示的TNF-α表达倍数增加可被认为高度显著(P < 0.01)。对于用pcECM支架刺激的RAW巨噬细胞,促炎细胞因子IL1-β的表达显示出2.69 ± 1.79倍增加,而对于分别用PLGA和LPS刺激的RAW巨噬细胞显示出0.25 ± 0.26倍和18,124 ±14,581倍增加(相对于非-刺激的细胞)。只有用LPS刺激的RAW巨噬细胞显示的IL1-β表达的倍数增加可被认为高度显著(P < 0.01)。
pcECM电纺纤维支架的免疫原性通过皮下植入另外进行体内评价。将小鼠分成两组:一组接受pcECM支架,而另一组接受电纺PLGA支架作为阴性对照。在植入后1周、2周和4周,处死小鼠。淋巴结检查显示在所有试验组均未发现有肿胀或刺激。此外,淋巴结中促炎细胞因子TNF-α和IL1-β的表达显示在相同周的ES-pcECM支架组和ES-PLGA支架组之间没有显著差异(图26A-B)。全血计数(CBC)揭示,对于所有时间点,ES-pcECM支架处理组与PLGA阴性对照组比较,白细胞(WBC)、红细胞(RBC)、血细胞比容、血红蛋白、平均红细胞容积(MCV)、平均红细胞血红蛋白(MCH)、MCH浓度(MCHC)、中性粒细胞和淋巴细胞的水平均无增加(图27A-I)。
总之,这些免疫原性研究证实pcECM电纺纤维支架,虽然取自猪来源并溶解在危险的有机溶剂中,都是非-免疫原性的。因此,它们是使用生物相容性心脏支架修复受损组织的候选物。

13.权利要求4-12的任一项的方法,其中所述合成聚合物是PEO。
