Abstract
本发明属于保健品加工领域,本发明公开了一种组合物,它是由绿茶提取物和中药提取物按重量百分比为绿茶提取物0.25‑4.5%,中药提取物95.5‑99.75%制成,其制备方法:取配比重量较小的绿茶提取物按等量递加方法依次逐步递加加入中药提取物,充分研磨至均匀,过筛即得。本发明利用绿茶提取物具有抗氧化、清除自由基的有益效果,与具有益气化瘀,调经祛斑,滋养肌肤作用的中药组合,对防治自由基对肌肤的损伤,促进血液循环,增进肌肤细胞发育具有较好的辅助防治效果。所述组合物按常规工艺制成片剂、胶囊剂、颗粒剂、口服液或酒品作为制备保健品的用途或作为制备化妆品中的添加剂的用途。

技术领域
本发明涉及药物制剂和/或保健食品加工领域,具体涉及一种由绿茶提取物和中药提取物的组合物和用途。
背景技术
中医学对茶叶资源的研究历史悠久,随着科学技术和保健食品的发展,对这些资源的研究取得了很大的进展。例如,对茶叶从“提神解乏、明目利尿、消暑解热”等方面的认识发展到利用茶叶提取物进行降血糖、降血脂、防止心血管疾病、抗衰老、抗肿瘤、抗突变的研究;中国茶类的演变,在几千年的历史长河中,经历了咀嚼鲜叶、生煮羹饮、晒干收藏、蒸青做饼、炒青散茶,乃至白茶、黄茶、黑茶、乌龙茶、红茶、再加工茶等多种茶类的发展过程。时至今日,中国的茶类已形成了六大基本茶类和再加工茶。基本茶类为绿茶、红茶、青茶(乌龙茶)、黑茶、黄茶和白茶六大类型。再加工茶是以上面六大类茶或鲜叶通过蒸压造型、提取调味、窨茶等措施加工成的,主要包括花茶、紧压茶、萃取茶、香味果味茶、保健茶、含茶饮料和速溶茶等。
茶叶的化学成分
1、茶叶的基本成分
鲜叶中的化学成分随茶树品种、气候条件和栽培条件而变化,成品茶的化学成分随原料(鲜叶)的化学组成与加工工艺及条件而变化,一般来说,成品茶的化学组成较鲜叶更为复杂。目前,人们从鲜叶中检测到的化学成分约500多种,主要有茶多酚、咖啡碱、蛋白质、维生素、氨基酸、糖类、类脂等,还有K、Na、Fe、Cu、P和F等28种矿物元素。茶树鲜叶这些成分大致分为:水分(75%-78%)和干物质(22%--25%);其中分为无机物(3.5%-7.0%)和有机物(93%-96.5%);其中无机物(3.5%-7.0%)分为水溶性部分和水不溶性部分;有机物分为蛋白质(20%-30%)、氨基酸(1%-5%)、生物碱(3%-5%)、茶多酚(20%-35%)、碳水化合物(20%-25%)、有机酸(3%-5%)、脂质(4%-5%)、色素(0.6-1%)、香气成分(0.005%-0.03%)、维生素(0.6%-1.0%)、皂苷类(0.07%-0.1%)、甾醇(0.04%-0.1%)。它们对茶叶的香气、滋味、颜色以及营养、保健起着重要的作用。茶作为饮料,体现其生理功能的主要是水溶性成分,茶叶水溶物约占茶叶干重的48%,主要包括30%多酚物质、7%氨基酸、4%生物碱、3%糖类和4%矿物质组分;不溶于水的部分约占茶叶干重的52%,主要是22%纤维素、16%蛋白质、8%脂类物质、4%果胶、0.5%淀粉及1.5%色素。
2、茶叶的功效成分
关于茶叶的功效,早在2000对年前已被公认。但由于科学技术的限制,茶在药用上很大程度还属于经验性质的。20世纪后期以来,茶叶生物化学研究的不断深入以及医学研究的参与,使得对茶叶的功效成份及其功效有了进一步的了解。
目前已经证实,茶叶中和人体健康有关系密切的组分,主要有以下几类。
(1)茶叶生物碱 茶叶生物碱是一类嘌呤的甲基衍生物,包括了咖啡碱、茶碱和可可碱,这3种茶叶生物碱功能基本相似,但含量以咖啡碱最高,一般为茶叶干重的2%-5%。
(2)多酚类化合物 可溶性的多酚类化合物在红茶中的含量约为干重的10%-20%,它主要由儿茶素类、黄酮类化合物、花青素和酚酸组成,以儿茶素类化合物含量最高,约占茶多酚总量的70%。儿茶素类中主要包括表儿茶素(EC)、表没食子儿茶素(EGC)、表儿茶素没食子酸酯(ECG)和表没食子儿茶素没食子酸酯(EGCG)。这是茶叶药效的主要活性组分。业已证明,它们具有防止血管硬化、防止动脉粥样硬化、降血脂、消炎抑菌、防辐射、抗癌、抗突变等多种功效。茶多酚具有预防中风和艾滋病、并能抑制过氧化脂质的形成,提高体内SOD和GSH-PX等酶系的活力。茶多酚的这些生理活性均在不同程度上与其具有活泼的酚羟基,能清除自由基有关。
(3)维生素类 茶叶含有丰富的维生素类。维生素B族的含量一般为茶叶干重的100-150mg/kg左右。菸酸(维生素B5)的含量是B族中含量最高的,约占B族中含量的一半,它可以预防癞皮病等皮肤病。
(4)矿质元素 茶叶中含有多种矿质元素,如磷、钾、钙、镁、锰、铝、硫等。这些矿质元素中的大多数对人体健康是有益的,茶叶中的氟元素含量很高,平均为100-200mg/kg左右,远高于其他植物,氟元素对预防龋齿和防治老年骨质疏松有明显效果。
(5)氨基酸 茶叶中的氨基酸种类已报道有25种,其中茶氨酸的含量最高,占氨基酸总量的50%以上。众所周知,氨基酸是人体必需的营养成分。
(6)茶多糖 茶多糖是茶叶复合物多糖的简称。根据对茶叶复合物多糖的紫外、红外及气相色谱的分析,茶叶复合多糖是一类由糖类、果胶及蛋白质等组成的复合物,含多糖类30.92%、蛋白质17.87%、水分8.89%、灰分16.48%及果胶12.92%等。茶叶复合多糖中多糖部分是由不同单糖组成的杂多糖,其单糖组成成分及比例为Ara:Xyl:Fuc:Glc:Gal=5.52:2.21:6.08:4.20:41.99。
(7)其他 除了上述这些主要组分外,茶叶中还含有一些次要的活性组分,它们的含量虽然不高,但却具有独特的药效。如茶叶中的二苯胺,具有降血糖的功效;茶叶中的一氨基丁酸具有降血压的作用。
茶叶的生理功能
茶的保健作用是多方面的,其主要作用可归纳如下:
1、兴奋提神;2、利尿;3、止痢和预防便秘;4、防龋作用;5、助消化作用;6、明目作用;7、抗衰老作用;8、减轻吸烟对人体的毒害;9、消炎灭菌作用;10、醒酒作用;11、对重金属毒害的解毒作用;12、抗辐射作用;13、预防高血压作用;14、降血脂和抗动脉粥样硬化;15、降血糖和对糖尿病的疗效;16、抗癌、抗突变作用;17、其他功能:茶还可以作为预防胆结石、肾结石和膀胱结石形成的药物,作为支气管炎和感冒时的发汗药物和增进呼吸作用的药物,可以预防痛风和消除人体中有害的盐类及毒素的积累,治疗瘰病,防治各种维生素的缺乏等。
根据学者们的研究其茶多酚的生理活性均在不同程度上与具有活泼的酚羟基,能清除自由基有关,其多酚类化合物主要由儿茶素类、黄酮类化合物、花青素和酚酸组成,其中以儿茶素类化合物含量最高,约占茶多酚总量的70%,儿茶素类中主要包括表儿茶素(EC)、表没食子儿茶素(EGC)、表儿茶素没食子酸酯(ECG)和表没食子儿茶素没食子酸酯(EGCG)。这也是茶叶药效的主要活性组分。不同的儿茶素类化合物的抗氧化活性,以具有较多数量取代羟基的酯型儿茶素较强,其活性次序按下列程度递减:EGCG>EGC>ECG>EC。科学家实验证明,用绿茶中的茶多酚或EGCG喂饲小白鼠后,发现可抑制皮肤粒线体中脂氧化酶的活性和脂质过氧化作用,同时肝脏和小肠中谷胱甘肽-S-转换酶的活性增强,起着抗氧化的效应。
由于现代人们生活节奏加快,生活压力大,生活结构的变化,加之环境污染给现代生活中一些特有的疾病也相应的呈现增多趋势。如因气血不足,气血阻滞或气虚血瘀等所致的内分泌紊乱,黄褐斑(黧黑斑)、雀斑、蝴蝶斑,皮肤面黄粗糙等病例明显增加,直到目前也未有确切疗效的治疗药物。利用绿茶提取物成分中具有特殊的抗氧化和清除自由基作用的功效,结合我国传统的中医药理论,与具有益气化瘀,调经祛斑,滋养肌肤的中药组合,将绿茶提取物这种天然活性物质与传统医学和现代制药工艺技术有机结合,组成一种新的药物组合物,使该组合物能在上述疾病的治疗中起到辅助的防治作用。
发明内容
本发明的目的在于提供一种由绿茶提取物和一种中药提取物组成的药物组合物。
本发明的目的还在于提供一种药物组合物在制备辅助调节益气化瘀、调经祛斑、滋养肌肤药物中作为制备保健品的用途。
本发明的目的还在于提供一种药物组合物在制备化妆品中的添加剂的用途。
本发明提供的一种药物组合物,其特征在于,它是由绿茶提取物和中药提取物按重量百分比为绿茶提取物0.25-4.5%,中药提取物95.5-99.75%制成,所述的中药提取物按以下方法制备:将人参150g,当归450g,川穹100g,黄芪550g加入浓度为75%(v/v)的乙醇12500g在38-42℃下浸泡168小时,滤取浸泡提取液,回收乙醇得浸提液保存备用,另将药渣分别按药材重量8倍、5倍、3倍加水煎煮3次,煎煮时间为第1次2小时,第二次1小时,第三次0.5小时,合并3次煎煮液,经过滤,经浓缩至比重为1.0的浓缩液时,再与上述回收乙醇后的浸提液合并,再进一步浓缩成稠膏,干燥至含水量小于5%w/w的浸膏。
该中药浸膏在下文中称作“本文(或上文)所述的中药提取物”。
本发明组合物的配制方法,可取配比重量较小的绿茶提取物按等量递加方法依次逐步递加加入中药提取物,充分研磨至均匀,过筛即得。
上述所述的干燥采用本领域公知的干燥技术条件下的干燥手段,如真空干燥、冷冻干燥或热风低温干燥等,在考虑到避免或减少中药提取物中有效成分的流失的条件,选择避免高温的干燥方法均适用于本发明中的干燥处理。
所述的药物组合物按常规工艺制成片剂、胶囊剂、颗粒剂、口服液作为制备保健品的用途。
所述的药物组合物作为制备酒制品的用途。
所述的药物组合物作为制备化妆品中的添加剂的用途。
本发明所述的绿茶提取物是由山茶科茶叶(Camelliia sinensis)干燥嫩叶提取制得,其性状:黄棕色粉末。其检查指标为:(1)干燥失重:≤8%;(2)灰分:≤2%;(3)重金属:≤百分之二十;(4)农残:≤百分之五;(5)溶剂残留:≤0.5%;(6)咖啡碱:≤3%。含量指标:茶多酚≥99%,其中儿茶素总量≥70%,EGG≥3%,EGCG≥30%,GCG≥15%,ECG≥10%。粒度100%通过80目。
人参,甘,微苦,微温。归脾、肺、心肾经。具有大补元气,复脉固脱,补脾益肺,生津养血、安神益智功能。人参大补元气,元气充则心气得养,心神得宁,心智得聪,而具安神益智的功效,用于治疗心气不足或心肾气虚、心脾两虚的失眠、健忘等。人参甘温,大补元气,益气以生血,具气血双补之效,故可用治脾胃气虚,化源不足,血虚萎黄之证。
当归,甘、辛,温。归肝、心、脾经。具有补血活血,调经止痛,润肠通便功能,用于血虚萎黄,眩晕心悸,月经不调,经闭痛经,虚寒腹痛,风湿痹痛,跌扑损伤,痈疽疮疡,肠燥便秘。当归甘温,养血补虚,为补血良药,故可用治心肝血虚,血色萎黄,眩晕心悸。
川穹,辛,温。归肝、胆、心包经。具有活血行气,祛风止痛功能。川穹辛散温通,功能活血行气,有“血中气药”(《本草汇言》)之称。既活血祛瘀通脉,善治心脉瘀阻、胸痹心痛;又行气开郁止痛,为肝郁气滞之胸肋刺痛所多用。川穹善于“下调经水”(《本草汇言》)、活血调经,优为月经不调、痛经、经闭等证之要药;亦治癥瘕腹痛,具有活血祛瘀、消癥散结之功。
黄芪,甘,微温。归肺、脾经。具有补气升阳,固表止汗,利水消肿,生津养血,行滞通痹,托毒排脓,敛疮生肌功效,用于气血乏力,食少便溏,中气下陷,腹泻脱肛,便血崩漏,表虚自汗,气虚水肿,内热消渴,气虚萎黄之证。黄芪甘微温,益气升阳,盖阳生阴长,气旺血生,故有补气生血之功,长用于劳倦内伤,气亏血虚,血虚阳浮,肌热面赤,脉大无力之血虚发热证。及思虑过度,劳伤心脾,气血双亏,面色萎黄,心悸失眠者。
以上4种中药组成制备提取物,按照中医药理论证明具有益气化瘀,调经祛斑,滋养肌肤作用,按现代中药理论研究,如组份中当归为主药之一,其有效成分阿魏酸能减少H2O2含量,并与磷脂酰乙醇胺结合抑制氧化反应和自由基反应,拮抗自由基对组织的损害,促进血液循环,增进肌肤细胞发育的作用。阿魏酸不仅能淬灭自由基。而且还能调节人体生理机制,抑制产生自由基的酶,促进清除自由基的酶的产生。
利用绿茶提取物生物活性成分与中药组合提取物能产生不同成分、不同作用机制的协同治疗作用,能强化其药物的抗氧化和清除自由基作用。本发明利用绿茶提取物具有抗氧化、清除自由基的有益效果,与具有益气化瘀,调经祛斑,滋养肌肤作用的中药组合,对防治自由基对肌肤的损伤,促进血液循环,增进肌肤细胞发育具有较好的辅助防治效果。通过绿茶提取物与本发明中药组合的提取物,组成新的药用组合物,能够充分发挥绿茶提取物中的有益成分和中药成分有益效果,特别是增加了绿茶提取物具有的特殊抗氧化和清除自由基的功效,强化了药物对皮肤损伤、色斑、癣疾病及皮肤色素沉着等具有良好的调理功能,为本发明药物组合物在保健食品、化妆品中的应用提供了理论基础。尤其是抗氧化、清除自由基和身体机能调理功能更为显著,因此产品既可以作为保健食品内服食用或饮用,也可作为天然的美容护肤产品外用。本发明克服了目前中药组合成分功效不理想的问题,而且产品使用方便,口感较好,工艺简单,生产效率高,质量稳定,可广泛用于保健品、酒品、化妆品的生产。其中的绿茶提取物市场已有质量可控、品质可靠的产品供应,且产品易购,能够满足本发明实施的产业化需求。
本发明的一种药用组合物,可以根据需要配制成不同的口服剂型。可以将药用组合物与药学上可接受的辅料混合配制成片剂,胶囊剂,颗粒剂,口服液、酒制品等,其中胶囊剂可制备成软胶囊剂和硬胶囊剂;其中片剂优选泡腾片,咀嚼片,口含片,因为泡腾片,咀嚼片,口含片与普通片剂相比有以下几点优势:(1)服用方便,可嚼服、含服,可化水,由于配制这些片剂都要加入一定量的糖份作为赋形剂,因而口感好,人们乐意服用,泡腾片加水自溶后犹如一杯饮料,使人们乐于饮用;(2)吸收快,因为经口腔或泡饮使药物成分离子化,使之吸收快,生物利用度高;(3)便于携带,方便运输;其中所述的酒品可制成白酒、葡萄酒、黄酒和啤酒制品。本发明一种药用组合物,还可以根据需要配制成化妆品。

通过以下实施例是对本发明作进一步的说明,但对本发明的范围不构成任何限制。
实施例1
中药提取物的制备
分别称取净药材人参150g,当归450g,川穹100g,黄芪550g,置于提取罐中,加入浓度为75%的乙醇12.5kg,在40℃条件下密闭浸泡168小时,浸泡期间每隔6小时进行搅拌1次,浸泡结束后,滤取浸泡提取液,将浸泡提取液回收乙醇得浸提液另器保存备用;将醇浸提后的药渣加入纯净水10公斤,加热煎煮2小时,滤取煎煮液,药渣再加入纯净水6.25公斤,加热煎煮1小时,滤取煎煮液,再将药渣加纯净水3.75公斤,加热煎煮0.5小时,滤取煎煮液,将3次煎煮液合并,过滤,将滤液浓缩至比重为1.0的浓缩液时,再与上述浸提液合并,再进一步浓缩成稠膏后,置于真空干燥机内进行真空干燥至含水量为4.5%,即得中药提取物。
实施例2
中药提取物的制备
分别称取净药材人参150g,当归450g,川穹100g,黄芪550g,置于提取罐中,加入浓度为75%的乙醇12.5kg,在38℃条件下密闭浸泡168小时,浸泡期间每隔6小时进行搅拌1次,浸泡结束后,滤取浸泡提取液,将浸泡提取液回收乙醇得浸提液另器保存备用;将醇浸提后的药渣加入纯净水10公斤,加热煎煮2小时,滤取煎煮液,药渣再加入纯净水6.25公斤,加热煎煮1小时,滤取煎煮液,再将药渣加纯净水3.75公斤,加热煎煮0.5小时,滤取煎煮液,将3次煎煮液合并,过滤,将滤液浓缩至比重为1.0的浓缩液时,再与上述浸提液合并,再进一步浓缩成稠膏后,置于真空干燥机内进行真空干燥至含水量为4.8%,即得中药提取物。
实施例3
中药提取物的制备
分别称取净药材人参150g,当归450g,川穹100g,黄芪550g,置于提取罐中,加入浓度为75%的乙醇12.5kg,在42℃条件下密闭浸泡168小时,浸泡期间每隔6小时进行搅拌1次,浸泡结束后,滤取浸泡提取液,将浸泡提取液回收乙醇得浸提液另器保存备用;将醇浸提后的药渣加入纯净水10公斤,加热煎煮2小时,滤取煎煮液,药渣再加入纯净水6.25公斤,加热煎煮1小时,滤取煎煮液,再将药渣加纯净水3.75公斤,加热煎煮0.5小时,滤取煎煮液,将3次煎煮液合并,过滤,将滤液浓缩至比重为1.0的浓缩液时,再与上述浸提液合并,再进一步浓缩成稠膏后,置于真空干燥机内进行真空干燥至含水量为4.0%,即得中药提取物。
实施例4
药物组合物的制备
称取绿茶提取物2.5g,置乳钵中,按等量递加方法加入实施例1制得的中药提取物2.5g,研磨至均匀,再等量递加中提取物5g,研磨均匀,按依次等量递加中药提取物997.5g至全量为1000g,充分研磨均匀,过筛,即得。

称取绿茶提取物45g,置乳钵中,按等量递加方法加入实施例1制得的中药提取物45g,研磨至均匀,再等量递加中提取物90g,研磨均匀,按依次等量递加中药提取物955g至全量为1000g,充分研磨均匀,过筛,即得。

所述的药物组合物按常规工艺制成片剂、胶囊剂、颗粒剂、口服液或酒品作为制备保健品的用途或作为制备化妆品中的添加剂的用途。

药物组合物的制备
称取绿茶提取物15g,置乳钵中,按等量递加方法加入实施例3制得的中药提取物15g,研磨至均匀,再等量递加中提取物30g,研磨均匀,按依次等量递加中药提取物985g至全量为1000g,充分研磨均匀,过筛,即得。
所述的药物组合物按常规工艺制成片剂、胶囊剂、颗粒剂、口服液或酒品作为制备保健品的用途或作为制备化妆品中的添加剂的用途。

可按常规方式生产含有下列组份的酒:

50度白酒 970g
取药物组合物加入50度白酒中,充分搅拌,然后放置室温处,密闭贮存21天后,过滤,即得。

可按常规方式生产含有下列组份的酒:

称取实施例6药物组合物 50g
蜂蜜 120g
柠檬酸 11g
22度白酒 加至 1000g
取药物组合物、柠檬酸和蜂蜜分别依次加入适量的22度白酒中使溶,然后加入余量的白酒,搅拌均匀,置于室温暗处,静置1周后,经过滤,分装,密封即得。

可按常规方式生产含有下列组份的酒:

称取实施例4药物组合物 25g
葡萄酒 加至 1000g
取药物组合物加入适量葡萄酒中使溶,然后加入余量的葡萄酒,搅拌均匀,置于3-5℃环境下,静置24小时,过滤,装瓶,封口即得。
Claims (5)
1.一种绿茶提取物中药提取物组合物,其特征在于,它是由绿茶提取物和中药提取物按重量百分比为绿茶提取物0.25-4.5%,中药提取物95.5-99.75%制成,
其制备方法:取配比重量较小的绿茶提取物按等量递加方法依次逐步递加加入中药提取物,充分研磨至均匀,过筛即得;其中所述的中药提取物按以下方法制备:将人参150g,当归450g,川穹100g,黄芪550g加入浓度为75%v/v的乙醇12500g在38-42℃下浸泡168小时,滤取浸泡提取液,回收乙醇得浸提液保存备用,另将药渣分别按药材重量8倍、5倍、3倍加水煎煮3次,煎煮时间为第1次2小时,第二次1小时,第三次0.5小时,合并3次煎煮液,经过滤,经浓缩至比重为1.0的浓缩液时,再与上述回收乙醇后的浸提液合并,再进一步浓缩成稠膏,干燥至含水量小于5%w/w的浸膏。
2.如权利要求1所述绿茶提取物中药提取物组合物按常规工艺制成片剂、胶囊剂、颗粒剂、口服液或酒品作为制备保健品的用途。
3.如权利要求1所述的绿茶提取物中药提取物组合物作为制备化妆品中的添加剂的用途。
4.根据权利要求2所述的组合物的用途,用于配制泡腾片,咀嚼片,口含片。
5.根据权利要求2所述的组合物的用途,用于配制白酒、葡萄酒、黄酒和啤酒。
