Description

Raw material formula for preventing hair loss and growing hair
Technical Field
The invention relates to the technical field of cosmetic raw materials, in particular to a raw material formula for preventing alopecia and growing hair.
Background
In cosmetics, a plurality of raw materials can be used for preventing hair loss and nourishing hair under the permission of laws and regulations, most of the raw materials are used for preventing hair loss and nourishing hair on the basis of one single or two or three mechanisms, the functional effect of the raw materials applied to the cosmetics is not satisfactory, otherwise, the raw materials have the defects or the defects, such as unstable raw materials or difficult addition to a formula system, great confusion to a formulator, easy color change, or great irritation to the scalp, high allergy rate, no effect when the raw materials are used by a consumer, and the like. Different raw materials of different manufacturers are tried to be refined into a perfect functional raw material through different experimental researches and verifications. The whole idea is as follows: first, the alopecia is analyzed in combination with the knowledge in medical field, and the following description about the alopecia is obtained:
alopecia is a common disease in dermatology and frequently occurs. Clinically, the alopecia areata, senile alopecia and chemotherapy alopecia can be classified into seborrheic alopecia, alopecia areata, senile alopecia and the like, and the incidence rate of alopecia areata and seborrheic alopecia is the highest. The cause of alopecia is explained in many ways in the international medical community. The result is the same regardless of the etiology, namely atrophy and degeneration of hair follicles, the atrophic and degenerated hair follicles do not participate in the circulation of the body, and cannot absorb nutrients of the surrounding scalp, and the hairs do not grow naturally. Therefore, it is critical to allow the hair follicle to regain its function. The atrophy and degeneration of the hair follicle is represented by the reduction or cessation of the proliferation of hair follicle cells (mainly, Outer root follicle cells or ORS), which deteriorates the hair growth environment in the entire hair follicle, inhibits the division of hair mother cells to prevent the synthesis of hair, keeps the hair follicle in a resting stage, and gradually causes permanent (irreversible) death. With the development of science and technology, the development of western biology has more and more comprehensive understanding on the mechanism and action factors of hair growth, and the hair regrowth has feasibility by regulating the proliferation of scalp hair follicle cells through biotechnology.
Meanwhile, the traditional Chinese medicine treats alopecia on the basis of dialectical treatment, and various functional natural active ingredients are determined and applied to the field of alopecia prevention and hair growth along with the development of extraction and separation technologies on the basis of natural plant traditional Chinese medicine theory. The modern tissue engineering theory and the biological technology can explain the action mechanism of the active ingredients, so as to screen the functional ingredients for treating alopecia more effectively. In contrast, the synthetic additive has faster and more obvious effect, but also has more side effects, so the natural active hair-growing ingredient is a hot spot of current research.
The above is some discussions about alopecia, and in our country, cosmetics are prohibited to add any western medicine component to achieve the purpose of preventing alopecia. Most of the existing anti-drop raw materials aim at one or two symptoms, so the effect is not obvious in practical application. In medical terms, there are times when medication is not prescribed symptomatically.

Anti-drop effect
The method comprises the following steps:
establishing a mouse alopecia model: mixing rosin and paraffin wax 1:1, heating to melt, uniformly coating the mixture on the back of a mouse, wherein the coating area is about 2cm multiplied by 3cm, cooling to solidify, and removing the back hair. The anagen phase of the hair was confirmed to be in the telogen phase.
Animal grouping: the next day of depilation, selecting mice with smooth and undamaged depilation regions, randomly dividing the mice into 4 groups, counting 6 animals in each group, and respectively forming experimental groups: adding 8% of essence of hair loss preventing and hair growth promoting materials; positive control group: a serum of 5% minoxidil was administered; ③ group of matrices: the essence is given; fourthly, negative control group: no smearing of the product.
Smearing: the product is applied to local skin directly with cotton swab, 3 times daily, each time with a dose of 0.1ml, and continuously for 20 days.
And observing and recording results: observing the change of the skin and the hair color of the mice every day, and recording the time from smearing to blackening of the skin color of the hair-removed area of the back of each animal; the negative control group recorded the time from day 1 after depilation to the beginning of skin darkening in the depilated area; the hair removal zone begins to darken to full hair. On days 4, 12 and 20, skin tissue blocks at the same positions on the backs of the mice of each group are taken, fixed by 10% formalin, cut into sections embedded by paraffin, the sections are about 4-5 microns thick, stained by HE, and observed under a staining light microscope. The observation indexes are as follows: general histological examination: observing the skin integrity, whether reactions and degrees of congestion, edema, inflammatory cell infiltration and the like exist; ② counting hair follicles, counting hair follicles of 3 high power fields (x 400) per case, taking the mean value and performing statistical treatment.
Statistical analysis: the t-test of the mean of the two samples was performed using sps.11 statistical software.
Results
Effect of 8% essence of hair loss prevention and hair growth promoting material on hair growth cycle: firstly, the local skin of the experimental group of mice gradually appears black in 5-6 days after epilation, the hair enters the growth period, new hair grows out after 10 days, the skin color changes into pink (rest period) through gray (catagen) in 20 days, and the skin black and gray last for about 15 days; secondly, local skin of the positive control group of mice gradually becomes black on the 5 th day after epilation, hairs grow into a growth period, new hairs grow out after 9 days, the skin color is changed into pink (rest period) through gray (catagen) at 20 days, and the skin black and gray last for 15 days; ③ the mice in the substrate group and the negative control group are all changed from pink to black on the local skin 6 days after epilation, new hair grows out after 10 days, the skin color is changed from pink to pink through gray at 19 days, and the skin black and gray are continuously maintained for 13 days.
Histological examination:
on the 4 th day of the experiment, the back skin of each group of mice is cut, and the subcutaneous blood vessels of the experimental group and the positive control group are rich and thick; this was not observed in the matrix group and the negative control group, as shown in FIG. 1
On the 12 th and 20 th days of the experiment, the back skin of each group of mice is cut, and is sliced by paraffin, and observed by HE staining: the skin structure of each group of mice is complete, the covered squamous epithelium is complete, the cuticle layer is not thickened, and inflammatory cells are not infiltrated in the dermis. The hair follicles of the animal skin of the experimental group and the positive control group are developed more completely and more in quantity, and melanin is more obvious than that of the substrate group and the negative control group. There were no significant differences in the morphology of the hair follicles in each group, as shown in FIG. 2.
The effect of the essence of 8% hair loss prevention and hair growth promoting raw material on the density of hair follicles: the number of hair follicles in each group of mice on day 20 of the experiment is shown in table 1. The mice of the experimental group, the positive control group and the negative control group have very significant difference statistically, P is less than 0.001, and the mice of the matrix group have no significant difference compared with the negative control group, wherein P is more than 0.05. And compared with the positive control group, the experimental group has no significant difference, and P & gt is 0.05.

Mice are an animal model for studying the hair cycle, with periodic growth of hair, a meristematic, a catagen and a telogen phase. The hair of the mouse is black, melanocytes in the skin are uniformly distributed on hair follicles, the melanocytes are synthesized and secreted only in the hair growth period, and the melanocytes are transferred to keratinocytes, so that the skin is black; during the hair anaplastic phase, melanin synthesis is reduced and the skin gradually changes from black to grey; there is no melanin synthesis during telogen and the skin is pink. Histologically, the change in skin color in the mice reflected different periods of the hair growth cycle. When the mouse is bred in a good state for 6-8 weeks, all hair follicles enter a resting state (skin pink) from a growth phase (skin black), and do not automatically enter the growth phase under a physiological state, but can be induced to enter the growth phase after artificial hair removal, the skin color turns black after 5-6 days of hair removal, the black color turns grey again after 12 days of hair removal (a regression phase), and the skin turns pink after 2 days of hair removal (a resting state).
Minoxidil is a medicine for treating various alopecia and has been proved by research, and can make hair enter anagen in advance, prolong anagen period, shorten telogen period and enlarge hair follicle. Therefore, 5% minoxidil essence is selected as a positive control in the study.
The experiment finds that compared with a negative control group mouse, the skin color of the experimental group mouse is changed from pink to black in advance, 24H is changed from black to gray in delay, the hair follicle growth period time is about 72H longer than that of the negative control group, and the result is similar to that of the positive control group. The essence of 8 percent of the hair loss preventing and hair growing raw material DepilationFair is consistent with minoxidil, so that the hair follicle can be induced to enter a growth phase, the growth phase time is prolonged, and the hair follicle is delayed to enter a catagen phase. We also found that on day 4 of the experiment, the skin of the back of each group of mice was excised, and that the subcutaneous blood vessel network of the experimental group and the positive control group was abundant and the blood vessels were thick; this was not observed in the matrix group and the negative control group. It is presumed that the essence of hair loss preventing and hair growth promoting material depilationFair has the same effect of promoting the vascular proliferation as minoxidil. Meanwhile, on the 20 th day of the experiment, the numbers of the hair follicles of the mice of the experimental group, the positive control group and the negative control group are observed to have very significant difference statistically, and P is less than 0.001; the stroma group has no significant difference compared with the negative control group, and P & gt is 0.05; compared with the positive control group, the experimental group has no significant difference, and P & gt is 0.05. Therefore, the hair loss preventing and hair restoring material DepilationFair has the effect of promoting the growth of hair follicles, and the effect is equivalent to that of minoxidil.
Hair growth and restoration effects
In vitro testing:
the method comprises the following steps: 1 female volunteer of 45 years old smears essence of 8 percent of hair loss prevention and growth promotion raw material DepilationFair every day for three times every day for lasting use for 90 days, and the effect is shown in figure 3: the results show that: the hair is obviously strengthened and thickened, which shows that the hair loss preventing and growing raw material has the hair growing and repairing effect.
The research confirms that the hair loss preventing and hair growing raw material DepilationFair for the experiment consists of 16 active ingredients, and the functions and the efficacies of the active ingredients are also proved. According to the formula of the raw materials for preventing hair loss and nourishing hair, the raw materials adopt the plant extract, and the hair loss preventing and fixing effects of the plant extract are represented by increasing the expression of the hair hyalin and the keratin, reshaping the hair root structure, preventing scalp aging and minimizing hair loss; the expression of collagen is increased, and the environment of hair follicles is changed, so that the beautiful appearance of the hairs is restored, and the hair follicles are healthier; the extract can also help maintain the hair growth cycle, optimize cell-to-cell and cell-to-matrix adhesion, increase protein expression, and promote hair growth and hair follicle renewal.
