Abstract

本发明涉及一种2‑甲基柠檬酸高产基因工程菌及其构建方法,属于基因工程技术领域。本发明的2‑甲基柠檬酸高产基因工程菌是由苏云金芽胞杆菌BMB171基因组中敲除2‑甲基柠檬酸脱水酶基因prpD而得到的。本发明还提供了上述2‑甲基柠檬酸高产基因工程菌的构建方法,以苏云金芽胞杆菌BMB171为出发菌株,利用基于归巢内切酶I‑SceI诱发染色体同源重组机制的基因敲除体系成功获得2‑甲基柠檬酸脱水酶(PrpD)基因无痕缺失突变株ΔprpD。通过液相色谱和质谱的检测发现,与出发菌株BMB171相比,ΔprpD菌株胞内2‑甲基柠檬酸浓度提高了217倍,且与化学法合成的2‑甲基柠檬酸相比,本发明法所产生的2‑甲基柠檬酸空间构型专一。

一种2-甲基柠檬酸高产基因工程菌及其构建方法
技术领域
本发明涉及基因工程技术领域,具体涉及一种2-甲基柠檬酸高产基因工程菌及其构建方法。
背景技术
苏云金芽胞杆菌(Bacillus thuringiensis,Bt)是一类杆状、产芽胞的革兰氏阳性细菌,广泛分布于土壤、植物叶表、植物根际等环境中,也是重要的昆虫病原菌。Bt最显著的特点是在形成芽胞的过程中产生由杀虫晶体蛋白组成的伴胞晶体,因而被广泛地用作微生物杀虫剂,具有杀虫活性高、杀虫谱广、对人畜安全、环境友好等优点。同时,Bt因培养周期短、中心代谢旺盛,是构建各种工程菌株的理想宿主。而苏云金芽胞杆菌无晶体突变株BMB171拥有遗传背景清晰、转化效率高、质粒兼容性及稳定性强等诸多优势,在蛋白质异源表达、高产小分子代谢物等方面极具潜力。
2-甲基柠檬酸是2-甲基柠檬酸循环的中间代谢物。2-甲基柠檬酸的合成是在2-甲基柠檬酸合成酶PrpC的催化下,以丙酰-CoA和草酰乙酸为底物缩合而成;其降解则是在2-甲基柠檬酸脱水酶PrpD的作用下脱水形成2-甲基顺乌头酸。
大量实验表明,细菌胞内2-甲基柠檬酸的积累能够产生细胞毒性,抑制细菌生长。例如,在肠道沙门氏菌中,prpD基因的缺失,导致该菌在含有丙酸的培养基中的生长受到明显的抑制,暗示着2-甲基柠檬酸能够抑制抑制细菌的生长;2-甲基柠檬酸可以显著抑制糖异生的关键酶果糖-1,6-二磷酸酶的活性,直接导致细菌的生长受阻。这些实验结果不仅丰富了2-甲基柠檬酸的生理功能,而且为新型抑菌物质的开发提供理论指导。
目前,市售的2-甲基柠檬酸药品均通过化学合成制得。而化学合成法往往选择性差,导致合成的2-甲基柠檬酸实为其四种对映异构体组成的混合物。同时,化学合成法工艺复杂、后续纯化难度极大,直接导致2-甲基柠檬酸药品价格高于黄金(仅仅10mg就需要1500元左右)。由此可见,利用合成特异性极强的生物法生产高纯的2-甲基柠檬酸具有重要经济价值。
因此,需要开发一种简单、高效并且成本低的生产2-甲基柠檬酸的生产方法。
发明内容
为解决上述问题,本发明提供一种2-甲基柠檬酸高产基因工程菌及其构建方法。
本发明解决上述技术问题的技术方案如下:一种2-甲基柠檬酸高产基因工程菌,是由苏云金芽胞杆菌BMB171基因组中敲除2-甲基柠檬酸脱水酶基因prpD而得到的。
所述2-甲基柠檬酸脱水酶基因prpD的核苷酸序列如SEQ ID NO:1所示。
本发明的有益效果是:本发明的2-甲基柠檬酸高产基因工程菌的2-甲基柠檬酸的产量比出发菌株苏云金芽胞杆菌BMB171提高了210倍,产量大大提升,降低了2-甲基柠檬酸的生产成本,且本发明生产的2-甲基柠檬酸空间构型专一。
本发明还提供了一种上述2-甲基柠檬酸高产基因工程菌的构建方法,其特征在于,包括以下步骤:
(1)选择含有壮观霉素抗性基因、归巢内切酶I-SceI特异识别作用位点、多克隆位点MCS、能在苏云金芽胞杆菌BMB171中复制的温敏型初始质粒,获取将要敲除的prpD基因上下游同源片段依次克隆到初始质粒中获得敲除载体;
(2)将敲除载体转入苏云金芽胞杆菌BMB171中,筛选获得发生第一次同源重组使质粒整合到受体菌染色体上的单交换菌株;
(3)将含有卡那霉素抗性基因、能够编码I-SceI酶的诱导质粒导入所述单交换菌株,在选择平板上选择获得第二次同源重组并实现靶基因prpD敲除的苏云金芽胞杆菌BMB171突变菌株;
(4)挑取经PCR验证的靶基因prpD敲除的苏云金芽胞杆菌BMB171突变菌株在无选择压力的条件下转接3~5次连续传代培养,获得质粒丢失的靶基因prpD敲除的苏云金芽胞杆菌BMB171突变菌株,即为2-甲基柠檬酸高产基因工程菌。
在上述技术方案的基础上,本发明还可以做如下改进。
进一步,所述步骤(1)的具体步骤为:通过PCR技术,以出发菌株苏云金芽胞杆菌BMB171的总DNA为模板,分别扩增prpD靶基因上游和下游长度为0.7~1kb的同源序列片段,将他们依次连接到初始质粒上获得敲除载体。
进一步,所述初始质粒为pRP1028,其核苷酸序列为SEQ ID NO:2所示。
进一步,所述步骤(2)中将敲除载体转入苏云金芽胞杆菌BMB171中的方法为三亲本杂交法,将敲除载体转入苏云金芽胞杆菌BMB171中的具体步骤为:将所述敲除载体转化大肠杆菌DH5α,获得含有敲除载体的供体菌,然后将供体菌与苏云金芽胞杆菌BMB171在辅助菌株的作用下将敲除载体转入苏云金芽胞杆菌中。
进一步,所述诱导质粒为pSS4332,其核苷酸序列为SEQ ID NO:3。
本发明的构建方法的有益效果是:本发明率先将无痕基因敲除技术应用于苏云金芽胞杆菌BMB171,可以将prpD基因完全敲除并且不残留任何抗生素抗性基因,本发明能够快速、稳定、高效的敲除prpD,还可以安全用于大规模工业生产。
本发明还提供了一种利用如权利要求1-2所述的2-甲基柠檬酸高产基因工程菌生产2-甲基柠檬酸的方法,其特征在于,包括以下步骤:
(1)将2-甲基柠檬酸高产基因工程菌于28℃,200rpm下培养12~18h,收集菌液;
(2)将收集的菌液离心获得菌体并利用超声破碎以及有机溶剂前处理获得2-甲基柠檬酸粗品。
进一步,所述步骤(2)的具体步骤为:将收集的菌液离心弃去上清,加入去离子水重悬,加入的去离子水与原菌液的体积比为2~4:7,置于冰浴水中用超声破碎仪破碎,离心收集上清,加入8~10倍体积的丙酮剧烈震荡30~90s,12000rpm离心5min,除去上清,将得到的棕黄色沉淀在烘箱中烘干,然后用去离子水重悬棕黄色沉淀,去离子水与原菌液的体积比为3~10:700,加入乙腈,乙腈与去离子水的体积比为2:3,剧烈震荡,12000rpm离心5min,取上清,加入5~10倍的乙腈,剧烈震荡,12000rpm离心5min,弃去上清,将沉淀置于烘箱中烘干,即得到2-甲基柠檬酸粗品。
采用上述方法的有益效果是2-甲基柠檬酸的纯化效率高。

本发明采用I-SceI酶介导的基因无痕敲除技术对苏云金芽胞杆菌BMB171染色体中的prpD基因进行无痕敲除,然后以lacZ为报告基因构建转录融合载体考察prpC基因在BMB171中的转录特性,并利用构建的高产2-甲基柠檬酸的工程菌生产2-甲基柠檬酸。
实施例1高产2-甲基柠檬酸的工程菌构建
1、敲除载体pRP1028-prpD-U-D的构建
以苏云金芽胞杆菌BMB171为出发菌株,根据prpD的基因序列及其上下游DNA序列,分别设计上下游同源片段引物,其中prpD的核酸序列如SEQ ID NO:1所示,

利用cross-over PCR将prpD基因上游臂同源片段prpD-U和prpD基因下游臂同源片段prpD-D连接在一起,获得prpD-U-D片段。第一轮PCR的配置体系为:上游臂同源片段prpD-U 0.5μL,下游臂同源片段prpD-D 0.5μL,高保真PCR聚合酶0.2μL,5×buffer 4μL,dNTP 1μL,补足ddH2O至20μL。第一轮PCR程序为:94℃预变性5min,94℃变性30s,50℃退火40s,72℃延伸1.5min,循环次数10次。该轮PCR得到的产物可直接作为第二轮PCR的模板。第二轮PCR的配置体系为:第一轮PCR产物5μL,引物prpD-U-F 2μL,引物prpD-D-R 2μL,高保真PCR聚合酶1μL,5×buffer 20μL,dNTP 2μL,补足ddH2O至100μL。PCR程序:94℃预变性5min,94℃变性30s,50℃退火40s,72℃延伸1.5min,循环次数30次。第二轮PCR产物进行琼脂糖凝胶电泳并割胶回收纯化得到prpD-U-D片段。
初始质粒的抗生素抗性需要选用苏云金芽胞杆菌不具有的抗生素抗性,并且具有归巢内切酶I-SceI特异识别作用位点,选用本实验室保存的质粒pRP1028,其核苷酸序列如SEQ ID NO:2所示,pRP1028质粒具有壮观霉素抗性,且在苏云金芽胞杆菌中为温敏型,该质粒转入苏云金芽胞杆菌内28℃时可以自主复制,在37℃时无法复制。

使用MulI和KpnI限制性内切酶双酶切初始质粒pRP1028,酶切体系为:86μLpRP1028,2μL MulI,2μL KpnI,10μL buffer K,充分混匀。将盛有酶切体系的1.5ml离心管放入37℃水浴锅中,6h后取出离心管加入10×loading buffer终止酶切反应,琼脂糖凝胶电泳并回收目的条带。使用相同的酶切体系酶切prpD-U-D片段,20h后加入10Xloadingbuffer终止酶切反应,琼脂糖凝胶电泳并回收目的条带。各取2μL回收的pRP1028 DNA和prpD-U-D DNA利用琼脂糖凝胶电泳分析各自浓度,确定连接体系的添加量。连接体系中prpD-U-D和载体pRP1028的分子摩尔比为4~10较适宜,T4DNA连接酶1μL,T4连接酶buffer1μL,40%PEG30001μL总体积10μL置于16℃连接过夜,将连接产物与E.coli DH5α感受态细胞混合均匀,冰上放置30min,42℃热激1min,立即冰上放置2min,再加入900μL LB培养基于37℃摇床中恢复培养1h,之后取100μL菌液涂布在含有壮观霉素(Spc,100μg/mL)的LB固体平板上,37℃倒置培养过夜。待菌落长出后,用无菌牙签挑取适量菌体打散在盛有40μLddH2O的1.5mL离心管中,沸水中煮10min,12,000rpm离心1min后取上清作为PCR反应的DNA模板。PCR配置体系为:DNA模板1μL,引物prpD-U-F、prpD-D-R各1μL,Easy Taq DNA聚合酶0.2μL,10×buffer 2μL,dNTP 0.8μL,补足ddH2O至20μL。PCR程序:94℃预变性5min,94℃变性30s,50℃退火40s,72℃延伸1.5min,循环次数30次。PCR完成后,取5μL产物用琼脂糖凝胶电泳检测,将阳性克隆送至北京擎科新业生物技术有限公司测序,测序正确的转化子即为含有敲除载体pRP1028-prpD-U-D的转化子,将测序结果正确的转化子保存甘油管。
2、第一次单交换流程及筛选
用三亲本杂交法将敲除载体pRP1028-prpD-U-D转入苏云金芽胞杆菌BMB171中,筛选获得发生第一次同源重组并使质粒整合到受体菌染色体上的单交换菌株。三亲本杂交法使用的辅助菌株为含有辅助质粒pSS1827的E.coli DH5α,质粒pSS1827具有Amp抗性,其核苷酸序列为SEQ ID NO:10所示。
活化受体菌株(苏云金芽胞杆菌BMB171)、辅助菌株(含有辅助质粒pSS1827的E.coli DH5α,Amp抗性)和整合菌株(上述制备的含有敲除载体pRP1028-prpD-U-D的E.coliDH5α,Spc抗性),活化的方法为:分别划线与固体LB平板上,过夜培养使其形成典型的单菌落,挑取单菌落接种于5mL液体LB培养基中,200r/min振荡过夜培养,其中BMB171的培养温度为28℃,大肠杆菌培养温度为37℃。各取1mL的上述三株菌,用无抗性的LB液体培养基洗涤三次,最后用300μL的LB培养基重悬(尽量使重悬后的三株菌的OD值保持一致),然后将三株菌混合在一起,并充分混匀,取200μL的混合菌液滴于LB固体平板上,28℃静置培养1天,用枪头将长好的菌落刮尽并重悬于500μL的LB培养基中,分别涂布于4个有抗性的LB平板(Spc 300μg/mL和Pmx 60units)上,其中Pmx为多粘菌素,能够抑制大肠杆菌等革兰氏阴性菌的生长,可以将辅助菌株筛除,于28℃静置培养直至有红色菌落(质粒pRD1028可以编码红色荧光蛋白)出现为止,一般为一星期左右。
挑取红色的单菌落于液体有抗性的LB培养基中(Spc 300μg/mL和Pmx 60units),28℃过夜活化。然后转接两次,37℃培养,每次培养7h,以消除未整合到BMB171基因组上的质粒pRP1028-prpD-U-D,转接完毕后,划线于固体LB平板(Spc 300μg/mL和Pmx 60units)中,37℃培养1天。最后挑取红色的单菌落,则得到第一次单交换菌株。

活化上述得到的第一次单交换菌株、辅助菌株(含有辅助质粒pSS1827的E.coliDH5α,Amp抗性)和表达菌株(含诱导质粒pSS4332的E.coli DH5α,Kan抗性),各取1mL的上述三株菌,用无抗性的LB液体培养基洗涤三次,最后用300μL的LB培养基重悬(尽量使重悬后的三株菌的OD值保持一致),然后将三株菌混合在一起,并充分混匀,取200μL的混合菌液滴于LB固体平板上,28℃静置培养1天,用枪头将长好的菌落刮尽并重悬于500μL的LB培养基中,分别涂布于2个有抗性的LB平板(Kan 50μg/mL和Pmx 60units)上,培养1~2天,用接种环分别划取每个平板上的菌落,然后划线于另2个相同抗性的LB平板上,培养1~2天,得到单菌落。挑取得到的单菌落,分别点板于两个不同抗性的LB固体平板(抗性分别为Spc 300μg/mL和Kan 50μg/mL)上培养,挑取Kan抗性下生长,但Spc抗性下不生长的单菌落于液体LB培养基中活化,抽提总DNA进行PCR和测序验证,PCR的上下游引物分别为prpD-I-F和prpD-I-R,其中prpD-I-F和prpD-I-R的引物序列分别为SEQ ID NO:12和SEQ ID NO:13,SEQ IDNO:12的序列为ATGCATTATCACATGCGTGGA,SEQ ID NO:13的序列为TCTAATCTCTCTTTAATTTC。取6株菌进行PCR验证,结果如图1所示,阳性对照孔道CK+出现大小为600bp的prpD基因的特异性目的DNA条带,而4号与6号孔道中未出现相应的条带,说明4#和6#菌株中prpD基因被成功敲除。
将验证正确的菌株于28℃不断传代培养以获得最终消除多余的表达质粒pSS4332,prpD完全敲除的目的菌株。
实施例2苏云金芽胞杆菌中2-甲基柠檬酸合成时间点的确定
1、转录融合载体pHT1K-PprpC-lacZ的构建及提取
转录融合载体选择含有lacZ的质粒pHT1K,来自本实验室保存的质粒,质粒pHT1K-lacZ含有氨苄青霉素(Amp)抗性和红霉素(Ery)抗性,其核苷酸序列如SEQ ID NO:11所示。
以BMB171总DNA为PCR扩增反应的模板,利用引物PprpC-lacZ-F和引物PprpC-lacZ-R扩增获得prpC启动子片段。其中引物PprpC-lacZ-F和引物PprpC-lacZ-R的序列分别为SEQ IDNO:8和SEQ ID NO:9所示:

PCR配置体系为:DNA模板1μL,引物PprpC-lacZ-F 2μL,引物PprpC-lacZ-R 2μL,高保真PCR聚合酶1μL,5×buffer 20μL,dNTP 2μL,补足ddH2O至100μL。PCR程序:94℃预变性5min,94℃变性30s,50℃退火40s,72℃延伸30s,循环次数30次。琼脂糖凝胶检测并纯化回收目的条带,使用NcoI和BamHI限制性内切酶双酶切pHT1K-lacZ载体(Amp抗性),酶切体系为:86μL pHT1K-lacZ,2μL NcoI,2μL BamHI,10μL buffer K,充分混匀,6h后加入10×loading buffer终止酶切反应并进行琼脂糖凝胶检测纯化回收。使用相同的酶切体系酶切prpC启动子片段。20h后加入10×loading buffer终止酶切反应并进行琼脂糖凝胶电泳检测纯化回收目的条带。将两个连接诶片段进行连接,转化E.coli DH5α菌株并筛选得到含有转录融合载体pHT1K-PprpC-lacZ的转化子,将含转录融合载体pHT1K-PprpC-lacZ的E.coliDH5α菌株(Amp抗性,浓度100μg/mL)接种于5mL LB液体培养基中,37℃振荡培养过夜。用质粒小提中量试剂盒(天根生化科技(北京)有限公司)提取质粒DNA。提取步骤为:取5mL菌液于离心管,12,000rpm离心1min,弃上清液;用250μL溶液P1充分溶解菌体;加入350μL溶液P2,温和上下翻转使菌体充分裂解;加入350μL溶液P3,轻轻翻转混匀,12,000rpm离心10min;将上清液全部转至树脂回收柱中,12,000rpm离心10min,弃掉收集管中废液;向树脂柱中加入700μL Washing Buffer,12,000rpm离心1min,弃掉收集管中废液;向树脂柱中加入500μL Washing Buffer,12,000rpm离心1min,弃掉收集管中废液;12,000rpm离心2min弃尽残留液体,向树脂膜上滴加60-100μL Elution Buffer,2min后12,000rpm离心1min收集得到的液体即为提取的转录融合载体pHT1K-PprpC-lacZ。
2、BMB171感受态细胞的制备及电转化
挑取BMB171单菌落接种于5mL液体LB培养基中,于28℃,200rpm振荡过夜培养;按1:100的接种量将活化好的菌液转接至100mL LB培养基中,然后于28℃,200rpm振荡培养至OD值为0.2;将培养好的菌液冰浴30min后,用100mL离心管以4,000rpm,5min冷冻离心收集菌体;加入10mL SG buffer重悬菌体,4,000r/min,5min冷冻离心收集菌体,重复洗涤3次;最后加入2mL SG buffer重悬菌体,每管分装100μL,放置于-80℃保存,即成感受态细胞;取100μL的感受态细胞,加入5μL的载体pHT1K-PprpC-lacZ,轻轻混匀,随后加入电转杯中,于冰上冰浴10min;用吸水纸吸干电转杯外壁水分后,至电转仪上进行电脉冲处理。电脉冲电压为1.25kV;电转完毕后,迅速加1mL LB培养基于电转化杯中,并轻轻混匀,然后转移至一无菌的EP管中,置于28℃,200rpm振荡恢复培养2h;随后取100μL培养基涂布于红霉素(Ery,浓度为25μg/mL)的LB固定平板中,28℃倒置培养过夜。红霉素抗性平板中长出的菌落表示pHT1K-PprpC-lacZ已成功电转至BMB171中,可用于后续β-半乳糖苷酶活性的测定。
2、β-半乳糖苷酶活性的测定
GYS培养基配方:葡萄糖1g;酵母提取物2g;硫酸铵2g;K2HPO4·3H2O,0.655g;MgSO4·7H2O,0.041g;MnSO4·H2O,0.0378g;CaCl2,0.08g;调节pH至7.8-7.9,定容至1L,115℃高温高压灭菌30min;
Z Buffer配方:0.06mol/L Na2HPO4,0.04mol/L NaH2PO4,0.01mol/L KCl,0.001mol/L MgSO4,0.05mol/Lβ-巯基乙醇,pH 7.0。β-巯基乙醇要在用前加入。
将携带有pHT1K-PprpC-lacZ质粒的BMB171菌株(命名为BMB171/pHT1K-PprpC-lacZ)接种至GYS培养基中,从培养的第6h开始至24h结束,每间隔2h,取2mL样品,12,000r/min离心2min,弃上清;加入1mL Z Buffer悬浮菌体;置于冰浴水中,用超声波细胞破碎仪(宁波新芝生物科技股份有限公司)破碎5min(超声破功率:150W,超声波工作1.0s,超声波停止1.5s,如此反复,共计5min);随后,12,000rpm离心5min,去除细胞碎片和不溶物;取50μL样品用Bradford法测定蛋白质浓度;另取一定体积的上清液,转移到新的离心管中,用ZBuffer补足到1mL;混匀后,置于37℃水浴锅中平衡5min。向样品中加入200μL已预热到37℃的ONPG溶液(4mg/mL),混匀后置于37℃水浴锅中开始反应,并记录反应起始时间t0;待样品出现淡黄色(A420=0.6-0.9),加入0.5mL Na2CO3溶液终止反应,记录反应终止时间t;用可见分光光度计(上海美谱达仪器有限公司)测定样品的A420和A550;样品β-半乳糖苷酶活性可由如下进行计算:

结果如图2所示,表明prpC的转录活性在对数期(9h之前)处于比较低的水平,进入稳定期后(从9h开始),活性逐步增强,并在较长时间内(12~18h)一直维持在高活性水平。BMB171/pHT1K-lacZ为空白对照菌株。
实施例3苏云金芽胞杆菌胞内2-甲基柠檬酸的提取及含量测定
1、提取方法
采用超声波破碎结合有机溶剂除杂前处理的技术获得2-甲基柠檬酸粗品。具体步骤为:将BMB171或ΔprpD菌株在GYS培养基中培养至6h或12h,收集35mL菌液,12,000r/min离心2min,弃上清,加入10mL ddH2O悬浮菌体;置于冰浴水中,用超声波细胞破碎仪(宁波新芝生物科技股份有限公司)破碎30min(超声破功率:250W,超声波工作1.0s,超声波停止1.5s,如此反复,共计30min);随后,12,000rpm离心5min,收集上清;取3.5mL上清液,加入31.5mL丙酮,置于振荡器上剧烈振荡1min,12,000rpm离心5min,弃掉上清;将有棕黄色沉淀的离心管放入50℃烘箱中充分烘干,加入150μL ddH2O充分溶解棕黄色沉淀;随后,加入100μL乙腈,置于振荡器上剧烈振荡,12,000rpm离心5min,取200μL上清至一新的离心管中;加入1mL乙腈,置于振荡器上剧烈振荡,12,000rpm离心5min,弃尽上清,将沉淀置于50℃烘箱中,所得沉淀即为2-甲基柠檬酸粗品。
2、2-甲基柠檬酸含量的测定
使用HPLC-UV和LC-Q-TOF分别测定12h时BMB171和ΔprpD胞内2-甲基柠檬酸含量。用100μL ddH2O充分溶解2-甲基柠檬酸粗品,并用孔径为0.22μm的尼龙膜滤器除去残留细微杂质,滤得的清澈液体即为可直接上机进行高效液相色谱和质谱检测。如图3所示,A图为2-甲基柠檬酸标准品的色谱图;图B为ΔprpD样品的色谱图;图C为BMB171样品的色谱图。2-甲基柠檬酸标准品谱图含有两个特征峰(保留时间分别为6.75min和8.25min),这是由于2-甲基柠檬酸标准品为其对映异构体混合物所致(见图3A)。而ΔprpD样品只在6.75min处出现特征峰,说明生物体只合成特定构型的2-甲基柠檬酸(见图3B)。但是BMB171样品在相应保留时间处无特征峰出现(见图3C),可能是由于该样品中2-甲基柠檬酸浓度较低,液相色谱检测法无法检出。该实验结果说明,ΔprpD样品中2-甲基柠檬酸浓度高于BMB171样品。
HPLC-UV检测2-甲基柠檬酸
液相色谱型号为UltiMate3000;选用C18分析柱(4.6mm×250mm,3.5μm;Agilent)。流动相为90%ddH2O+10%甲醇+0.1%甲酸;HPLC-UV参数设置为:柱温箱25℃,进样体积10μL,流速1mL/min,检测波长210nm。
LC-Q-TOF检测
液相色谱仪型号为Agilent 1260,Q-TOF质谱仪型号为G6530;选用ZORBAX SB-Aq分析柱(2.1mm×150mm,5μm;Agilent)。流动相为90%ddH2O+10%甲醇+0.1%甲酸;LC参数设置为:柱温箱30℃,进样体积5μL,流速300μL/min,紫外检测器波长210nm;Q-TOF参数设置为:负离子模式,毛细管电压4000V,辅助气压力50psi,干燥气流速9L/min,鞘气温度350℃,扫描范围m/z 50~800。利用Agilent MassHunter Qualitative Analysis Softwareversion B.06.00软件和峰面积法计算样品中2-甲基柠檬酸的相对含量。首先,利用m/z205.03从总离子流图谱(TIC)中提取2-甲基柠檬酸离子组分(允许m/z单边20ppm质量摆动),获得提取离子流图谱(EIC);再将EIC中2-甲基柠檬酸特征离子峰进行积分获得峰面积,由于峰面积大小与2-甲基柠檬酸浓度成正比,即可通过峰面积大小得到BMB171和ΔprpD样品中2-甲基柠檬酸的相对含量,同时根据2-甲基柠檬酸的标准曲线计算BMB171和ΔprpD样品中2-甲基柠檬酸的含量。结果显示,在GYS培养基中培养至6h,BMB171和ΔprpD样品中均无2-甲基柠檬酸(前面结果显示2-甲基柠檬酸合成酶PrpC只在稳定期开始表达,因此6h所对应的对数生长期的菌株胞内并未开始合成2-甲基柠檬酸);当培养至第12h时BMB171样品和ΔprpD样品中2-甲基柠檬酸的浓度分别为0.032mg/L和7mg/L,如图4所示,说明与出发菌株BMB171相比,ΔprpD菌株胞内2-甲基柠檬酸浓度提高了217倍。本发明的2-甲基柠檬酸高产基因工程菌能够极大的提高2-甲基柠檬酸的产量,降低其生产成本。

Claims (8)

1.一种2-甲基柠檬酸高产基因工程菌,其特征在于,是由苏云金芽胞杆菌BMB171基因组中敲除2-甲基柠檬酸脱水酶基因prpD而得到的,所述2-甲基柠檬酸脱水酶基因prpD的核苷酸序列如SEQ ID NO:1所示。
2.一种如权利要求1所述的2-甲基柠檬酸高产基因工程菌的构建方法,其特征在于,包括以下步骤:
(1)选择含有壮观霉素抗性基因、归巢内切酶I-SceI特异识别作用位点、多克隆位点MCS、能在苏云金芽胞杆菌BMB171中复制的温敏型初始质粒,获取将要敲除的prpD基因上下游同源片段依次克隆到初始质粒中获得敲除载体;
(2)将敲除载体转入苏云金芽胞杆菌BMB171中,筛选获得发生第一次同源重组使质粒整合到受体菌染色体上的单交换菌株;

(4)挑取经PCR验证的靶基因prpD敲除的苏云金芽胞杆菌BMB171突变菌株在无选择压力的条件下转接3~5次连续传代培养,获得质粒丢失的靶基因prpD敲除的苏云金芽胞杆菌BMB171突变菌株,即为2-甲基柠檬酸高产基因工程菌。
3.根据权利要求2所述的2-甲基柠檬酸高产基因工程菌的构建方法,其特征在于,步骤(1)的具体步骤为:通过PCR技术,以出发菌株苏云金芽胞杆菌BMB171的总DNA为模板,分别扩增prpD靶基因上游和下游长度为0.7~1kb的同源序列片段,将上述同源序列片段依次连接到初始质粒上获得敲除载体。
4.根据权利要求3所述的2-甲基柠檬酸高产基因工程菌的构建方法,其特征在于,所述初始质粒为pRP1028,其核苷酸序列为SEQ ID NO:2所示。
5.根据权利要求2所述的2-甲基柠檬酸高产基因工程菌的构建方法,其特征在于,所述步骤(2)中将敲除载体转入苏云金芽胞杆菌BMB171中的方法为三亲本杂交法,将敲除载体转入苏云金芽胞杆菌BMB171中的具体步骤为:将所述敲除载体转化大肠杆菌DH5α,获得含有敲除载体的供体菌,然后将供体菌与苏云金芽胞杆菌BMB171在辅助菌株的作用下将敲除载体转入苏云金芽胞杆菌中。
6.根据权利要求2所述的2-甲基柠檬酸高产基因工程菌的构建方法,其特征在于,所述诱导质粒为pSS4332,其核苷酸序列为SEQ ID NO:3。
7.一种利用如权利要求1所述的2-甲基柠檬酸高产基因工程菌生产2-甲基柠檬酸的方法,其特征在于,包括以下步骤:
(1)将2-甲基柠檬酸高产基因工程菌于28℃,200rpm下培养12~18h,收集菌液;
(2)将收集的菌液离心获得菌体并利用超声破碎以及有机溶剂前处理获得2-甲基柠檬酸粗品。
8.根据权利要求7所述的生产2-甲基柠檬酸的方法,其特征在于,所述步骤(2)的具体步骤为:将收集的菌液离心弃去上清,加入去离子水重悬,加入的去离子水与原菌液的体积比为2~4:7,置于冰浴水中用超声破碎仪破碎,离心收集上清,加入8~10倍体积的丙酮剧烈震荡30~90s,12000rpm离心5min,除去上清,将得到的棕黄色沉淀在烘箱中烘干,然后用去离子水重悬棕黄色沉淀,去离子水与原菌液的体积比为3~10:700,加入乙腈,乙腈与去离子水的体积比为2:3,剧烈震荡,12000rpm离心5min,取上清,加入5~10倍的乙腈,剧烈震荡,12000rpm离心5min,弃去上清,将沉淀置于烘箱中烘干,即得到2-甲基柠檬酸粗品。
