Abstract

本发明提供了一种改性胶原,制备方法包括以下步骤:A.由表没食子儿茶素没食子酸酯交联胶原膜;B.加载骨形态发生蛋白‑2至步骤A得到的胶原膜,清洗,干燥,即得。本发明将胶原膜以表没食子儿茶素没食子酸酯改性,再加载骨形态发生蛋白‑2,进而得到促骨再生性能良好的加载了骨形态发生蛋白‑2的表没食子儿茶素没食子酸酯改性胶原复合膜。

一种改性胶原及其制备方法
技术领域
本发明属于生物材料领域,具体涉及一种改性胶原及其制备方法。
背景技术
牙科手术中常需要应用引导骨再生手术进行骨增量。胶原是细胞外基质的一种成分,已 经广泛应用于引导骨再生术中。胶原膜有较好的生物兼容性和较低的抗原性,并能促进成骨 细胞黏附、增殖、迁移和分化,也能有阻挡成纤维细胞快速入侵骨组织生长位点、隔开软组 织与硬组织的生长等屏障功能。
胶原(Collagen)是细胞外最重要的水不溶性纤维蛋白,是构成细胞外基质的骨架。胶原在 哺乳动物体内含量丰富,约占人体或其他动物蛋白的20%-30%。主要存在于软骨、皮肤及肌 腱等组织中,在动物体内起支持、保护及连接等多种作用。由于胶原具有良好的生物相溶性、 无毒、弱抗原性、可降解性及安全性等优点,因此可以作为天然生物材料。胶原膜作为可吸 收性膜,它具有低抗原性,生物相容性好,可参与组织愈合过程,降解速率可根据需要调节 等诸多优点。
表没食子儿茶素没食子酸酯是一种茶多酚,拥有抗癌、抗氧化、抗炎、抗纤维化和促骨 再生的作用。并且,表没食子儿茶素没食子酸酯能提高胶原的热稳定性,并保护胶原不被胶 原酶降解,因此它可以用于胶原膜的交联。
骨形态发生蛋白-2在骨引导、骨再生中有重要作用。骨形态发生蛋白-2通过促进多能干 细胞向骨再生细胞分化,从而得到促骨再生的作用。美国食品与药品监督管理局也批准了相 应的材料可以在临床上使用。
发明内容
本发明的目的是针对现有技术中存在的不足,提供一种改性胶原及其制备方法。
为了达到上述目的,本发明采用了下列技术方案:
一种改性胶原,制备方法包括以下步骤:
A.由表没食子儿茶素没食子酸酯交联胶原膜;
B.加载骨形态发生蛋白-2至步骤A得到的胶原膜,清洗,干燥,即得。
作为优选,所述的胶原膜为牛来源I型胶原制备得到。
作为优选,所述步骤A中,表没食子儿茶素没食子酸酯和胶原膜重量比为1:100。
作为优选,所述步骤A中,交联是将胶原置于表没食子儿茶素没食子酸酯中改性60min,lXPBS缓冲液冲洗。
作为优选,所述步骤A中,胶原膜是在-26℃温度下冷冻过夜;在-53℃、1Pa条件下,真 空冷冻干得到。
作为优选,所述步骤B中,加载为按照100mg:1ml的比例,将胶原膜浸泡在浓度为500ng/ml的骨形态发生蛋白-2的PBS溶液中,浸泡时间为24小时。
作为优选,所述步骤B中,干燥为将制备好的改性胶原膜置于-20℃条件下冷冻干燥过夜。
本发明的优点在于:
本发明将胶原膜以表没食子儿茶素没食子酸酯改性,再加载骨形态发生蛋白-2,进而得到促 骨再生性能良好的加载了骨形态发生蛋白-2的表没食子儿茶素没食子酸酯改性胶原复合膜。 本发明提高了以前胶原膜的生物性能,具有很好的应用前景,有望于应用在引导骨组织再生 手术(GBR)中,为引导再生术等相关增量手术提供了新选择。

为了使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例,对本发明进行 进一步详细说明。应当理解,此处所描述的具体实施例仅仅用以解释本发明,并不用于限定 本发明。
实施例1:一种改性胶原,制备方法包括以下步骤:
A.由表没食子儿茶素没食子酸酯交联胶原膜;
B.加载骨形态发生蛋白-2至步骤A得到的胶原膜,清洗,干燥,即得。
实施例2:一种改性胶原,制备方法包括以下步骤:
A.按照重量比为1:100的比例,将表没食子儿茶素没食子酸酯和牛来源I型胶原制备的胶原 膜交联胶原膜,交联是将牛来源I型胶原置于表没食子儿茶素没食子酸酯中改性60min,lXPBS 缓冲液冲洗;
B.按照100mg:1ml的比例,将胶原膜浸泡在的骨形态发生蛋白-2浓度为500ng/ml的PBS溶 液中,浸泡时间为24小时;清洗,将制备好的改性胶原膜置于-20℃条件下冷冻干燥过夜, 即得。

Claims (7)

1.一种改性胶原,其特征在于制备方法包括以下步骤:
A.由表没食子儿茶素没食子酸酯交联胶原膜;
B.加载骨形态发生蛋白-2至步骤A得到的胶原膜,清洗,干燥,即得。
2.根据权利要求1所述的一种改性胶原,其特征在于:所述的胶原膜为牛来源I型胶原制备得到。
3.根据权利要求1所述的一种改性胶原,其特征在于:所述步骤A中,表没食子儿茶素没食子酸酯和胶原膜的重量比为1:100。
4.根据权利要求1所述的一种改性胶原,其特征在于:所述步骤A中,交联是将胶原置于表没食子儿茶素没食子酸酯中改性60min,lX PBS缓冲液冲洗。
5.根据权利要求1所述的一种改性胶原,其特征在于:所述步骤A中,胶原膜是在-26℃温度下冷冻过夜;在-53℃、1Pa条件下,真空冷冻干得到。
6.根据权利要求1所述的一种改性胶原,其特征在于:所述步骤B中,加载为按照100mg:1ml的比例,将胶原膜浸泡在浓度为500ng/ml的骨形态发生蛋白-2的PBS溶液中,浸泡时间为24小时。
