Abstract
本发明公开了一种可以减轻口臭的组合物及其应用,该组合物包括复合益生菌,所述复合益生菌包括唾液乳杆菌、双歧杆菌、鼠李糖乳杆菌、嗜酸乳杆菌、罗伊氏乳杆菌、唾液链球菌和约氏乳杆菌,其中,每克所述组合物中至少含有唾液乳杆菌20亿、双歧杆菌10亿、约氏乳杆菌20亿、鼠李糖乳杆菌40亿、嗜酸乳杆菌20亿、罗伊氏乳杆菌20亿、唾液链球菌20亿。该组合物还可广泛应用于口臭治疗药物、口腔护理制品、美容组合物、食品或保健品的制备中。与现有技术相比,本发明方案的组合物能够同时治疗口源性口臭和口源外口臭,具有治疗效果好且应用前景广泛等优点。
Description
技术领域
本发明涉及医学、微生物学和营养学领域,具体涉及一种可以减轻口臭的组合物及其应用。
背景技术
口臭是一个令人不快的问题,其根源是身体健康出了状况,也可能是因心理健康不佳导致的。口臭患者通常不敢与他人近距离交往,已严重影响人们的正常社际交往和情感交流。此外,世界卫生组织(World Health Organization,WHO)也已将口臭作为一种疾病。根据相关调查显示,中国口臭患病率高达22%-50%不等。
口臭通常是由细菌引起,这些细菌常常会在分解含硫氨基酸后释放出挥发性硫化物(Volatile Sulphur Compound,VSCs),包括硫化氢、二甲基硫、甲硫醇等。口臭的发生是由于产臭菌在口腔中过度增殖,影响了口腔正常菌群如唾液链球菌的生长,导致口腔菌群生态失调。然而并不是所有的口臭都来自于口腔,在口腔之外,如消化道中,也存在导致口臭的病因,如幽门螺旋杆菌(Helicobacter pylori,HP)等。幽门螺旋杆菌是一种微量需氧的螺旋形革兰氏阴性菌,它定植于人体胃黏膜,该致病菌的存在与各种胃病相关——从轻微的胃部不适感到其他严重的症状(Marshall等人,(1984)The Lancet 1984;323:1311–1315)。同时幽门螺旋杆菌常见于口腔牙周袋中,感染幽门螺旋杆菌被认为是引起口臭的重要原因(Suzuki等人,(2008)J Med Microbiol 57(Pt 12):1553-1559)。幽门螺旋杆菌能产生VSCs,对幽门螺旋杆菌的有效治疗显著改善了口臭症状(Kinberg等人,(2010)Can JGastroenterol 24(9):552-556)。因此,根除幽门螺旋杆菌不管是对于口臭还是胃病治疗都具有十分重要的意义。
已知治疗口臭的基本原理是减少产臭菌数量,降低VSCs浓度。目前治疗方法主要有两种:1,机械方法,包括清洁舌体、牙周洁治、清洁牙间隙及必要的刷牙和漱口等;2,生物化学方法,主要是指使用各种抗菌剂来抑制产臭菌的增殖,包括使用氯己定、金属离子和西吡氯铵等。这些方法能短期改善口臭,但长期疗效欠佳。而且使用生物化学方法因含有抗菌剂,长期使用还会导致口腔菌群失调。口臭治疗后,各种细菌重新在口腔内定植、生长、增殖,一旦产臭菌重新增殖为口腔中优势菌群,口臭即可复发,故有效地恢复口腔内菌群正常的生态平衡是治疗口臭的关键。此外,通常口臭是由口源内和口源外的细菌共同引起的,若只是单一性的治疗口源性口臭或口源外口臭均难以获得彻底治疗的效果,然而,现有技术中,能同时兼顾治疗幽门螺旋杆菌等口源外口臭的治疗方法研究却相对较少。近年来,很多学者开始探索更有效地预防及治疗口臭的方法,其中采用益生菌进行治疗已逐渐成为研究的热点。以益生菌本身的抑制致病菌以及与致病菌进行竞争的能力,有效地改善口腔微生物菌群平衡并形成一种可持续的菌群分布,同时兼具对抗幽门螺杆菌感染的作用,调节机体的免疫机能,通过改善胃肠道微生态环境,达到有效根治口臭的目的。
发明内容
本发明所要解决的技术问题是:提供一种能够同时缓解口源性口臭及幽门螺旋杆菌等口源外口臭的含有益生菌组合物及其应用。
为了解决上述技术问题,本发明采用的技术方案为:一种可以减轻口臭的组合物,包括复合益生菌,所述复合益生菌包括唾液乳杆菌、双歧杆菌、鼠李糖乳杆菌、嗜酸乳杆菌、罗伊氏乳杆菌、唾液链球菌和约氏乳杆菌。其中,每克所述组合物中至少含有唾液乳杆菌20亿、双歧杆菌10亿、鼠李糖乳杆菌40亿、嗜酸乳杆菌20亿、罗伊氏乳杆菌20亿、唾液链球菌20亿、约氏乳杆菌20亿。
进一步地,每克所述组合物中活菌数含量为100亿~300亿。
进一步地,所述组合物还包括绿茶粉末。
进一步地,所述组合物还包括山药粉末和/或白术粉末。
进一步地,所述复合益生菌还包括短乳杆菌、酵母和瑞士乳杆菌中的一种或多种。
进一步地,所述组合物还包括辅料。
进一步地,所述辅料包括木糖醇和/或低聚糖。
本发明的有益效果在于:本发明方案的组合物包括多种益生菌,通过多种益生菌的复配,使得该组合物通过调节和维持口腔菌群稳定,同时组合物中添加有能够协同减少胃黏膜内幽门螺旋杆菌数量的鼠李糖乳杆菌、约氏乳杆菌、嗜酸乳杆菌、双歧杆菌和唾液乳杆菌等。本发明方案既可以缓解口腔源口臭,也可缓解口腔外口臭;与此同时,该组合物还能够调节肠道微生态,改善胃内黏膜发炎等情况;此外,本发明方案组合物中益生菌还能够在口腔和胃肠道定植,一旦黏附定植后,将会在口腔硬组织(牙釉质、牙本质和牙骨质)和/或软组织以及胃肠道表面形成一层保护层阻止致病菌的黏附,效果更持久。罗伊氏乳杆菌和唾液乳杆菌可以协同与表皮细胞相互作用,进而改变抗炎症效应因子的释放来调节宿主免疫反应,既可减少牙龈炎,也可以减轻胃炎。
本发明还包括一种可以减轻口臭的复合制剂,包括上述组合物。
优选地,所述制剂的形态为片剂、溶液或凝胶。
优选地,所述制剂为含片或口香糖。
本发明还包括上述组合物在口臭治疗药物、口腔护理制品、美容组合物、食品、保健品的制备中的应用。
本发明的有益效果在于:本发明方案的组合物通过构建良好的口腔和胃肠道微生态环境,同时缓解口源性口臭和口源外口臭,彻底解决口臭问题,具有广泛的应用前景和价值。

一种可以减轻口臭的组合物,包括复合益生菌,所述复合益生菌包括唾液乳杆菌、双歧杆菌、鼠李糖乳杆菌、嗜酸乳杆菌、罗伊氏乳杆菌、唾液链球菌和约氏乳杆菌,其中,每克所述组合物中至少含有唾液乳杆菌20亿、双歧杆菌10亿、鼠李糖乳杆菌40亿、嗜酸乳杆菌20亿、罗伊氏乳杆菌20亿、唾液链球菌20亿、约氏乳杆菌20亿。

本发明的实施例一为:
一种用于改善口臭状况的益生菌组合物,每克组合物中包含唾液乳杆菌20亿、双歧杆菌10亿、约氏乳杆菌20亿、鼠李糖乳杆菌40亿、嗜酸乳杆菌20亿、罗伊氏乳杆菌20亿、唾液链球菌20亿、短乳杆菌20亿,木糖醇含量280毫克,绿茶粉末50毫克,山药粉末50毫克。该组合物形态为含片,可以较快时间在口腔内释放。
人体试验
试验过程:选择20例真性口臭患者(12例男性,8例女性;平均年龄42.3±8.9岁,25-55岁之间),这些患者因为口臭问题初次来诊所就诊,患者全部自愿参加该试验,并且在参加试验前的三个月时间内没有服用任何抗生素。
试验方法:将试验对象随机分为两组,每组10人且两组中男女人数相等,分别为测试组(A组)和对照组(B组),试验周期均为两周。A组每天进行常规口腔卫生护理(刷牙刷舌等),同时在中餐和晚餐后各服用本发明提供的益生菌组合物含片1克剂量;B组每天进行常规口腔卫生护理(刷牙刷舌等),同时进行抗幽门螺旋杆菌的标准三联疗法(奥美拉唑+阿莫西林+克拉霉素,疗程为两周)。
两周后,对受试者进行口臭等级分级评估及碳14呼气试验,分别评价口臭减轻程度及幽门螺旋杆菌的治疗效果。具体结果如下表1所示:

口臭评级:0,无;1,不确定;2,轻微;3,中度;4,重度;5,严重。
碳14呼气试验:结果值≤100为HP阴性,≥100为HP阳性。
分别统计计算A、B组实验前后的口臭评分平均值绘制柱状图,如图1所示;分别统计计算A、B组HP转阴率百分比例绘制柱状图,如图2所示。由图1和2可以看出,本发明方案对口臭的治愈率高于传统的标准三联疗法,本发明方法既能治疗口源性口臭又能解决口源外口臭问题,而传统的标准三联疗法只能对口源外口臭有所缓解,两者比较具有显著的统计学差异(图1中“***”代表P<0.001)。
对受试者六个月后进行回访,在试验结束后至回访日两组受试者均未再服用组合物或抗生素,仅进行常规的刷牙刷舌护理,统计结果如下表2所示:

从上表2可以看出,本发明方案中的复合益生菌已成功定植,因此,对口臭症状具有持续性的缓解效果,而对照组则出现了部分复发的情况。
本发明实施例二为:
一种用于改善口臭状况的益生菌组合物,每克组合物中包含唾液乳杆菌22亿、双歧杆菌13亿、约氏乳杆菌25亿、鼠李糖乳杆菌41亿、嗜酸乳杆菌25亿、罗伊氏乳杆菌20亿、唾液链球菌20亿、短乳杆菌20亿,木糖醇含量300毫克,绿茶粉末40毫克,山药粉末60毫克。该组合物形态为含片,可以较快时间在口腔内释放。
对比例一为一种用于改善口臭状况的益生菌组合物,其与实施例二的区别在于:不含绿茶粉末,其他组分均与实施例二相同。
对比例二为一种用于改善口臭状况的益生菌组合物,其与实施例二的区别在于:不含山药粉末,其他组分均与实施例二相同。
人体试验
试验过程:选择24例真性口臭患者(12例男性,12例女性;平均年龄32.3±4.5岁,20-50岁之间),这些患者因为口臭问题初次来诊所就诊,患者全部自愿参加该试验,并且在参加试验前的三个月时间内没有服用任何抗生素。
试验方法:将试验对象随机分为四组,每组6人且男女各半,分别为测试组(C组)和对照组(D和E组),试验周期均为两周。各组受试者每天进行常规口腔卫生护理(刷牙刷舌等),同时在中餐和晚餐后各服用本发明提供的益生菌组合物含片1克剂量。
两周后,对受试者进行口臭等级分级评估及碳14呼气试验,分别评价口臭减轻程度及幽门螺旋杆菌的治疗效果。具体结果如下表3所示:

从上表3中可以看出,本发明方案对口臭治疗具有良好的效果,未加绿茶粉末的对照例组受试者口臭缓解程度较实施例组要差,但幽门螺旋杆菌的治疗效果仍较好;而未加山药粉末组口臭缓解程度和幽门螺旋杆菌的治疗效果均不稳定;因此,本发明方案中各成分对缓解口臭及幽门螺旋杆菌具有不可替代的协同作用。
综上所述,本发明提供的一种可以减轻口臭的组合物及其应用,该组合物通过改善人体口腔及胃肠道微生物菌群平衡并形成一种可持续的健康微生态,以达到有效地抑制口腔及胃肠道产臭菌的生长,最终达到有效消除口臭、维护口腔及胃肠道健康之目的。
