Abstract
本发明涉及微生物萃取物及其应用技术领域,具体是一种四氢茄红素的应用及其制备方法,所述的四氢茄红素为用作抑制紫外线诱发皮肤内胶原蛋白分解的添加物。本发明的有益效果是,四氢茄红素能有效抑制紫外线诱发皮肤内胶原蛋白的分解,且可以促进胶原蛋白的生成,从而减缓皮肤老化。另外,本发明的四氢茄红素可以大量生产,解决了传统从番茄中萃取四氢茄红素产量低的问题,满足市场需求。

技术领域
本发明涉及微生物萃取物及其应用技术领域,具体是一种四氢茄红素的应用及其制备方法。
背景技术
皮肤老化可分为自然老化和外部因素造成的老化。自然老化可能是受遗传因素的影响,外部因素造成的老化则是由环境因素作用于“内部”所引起的老化,比如日晒(紫外线)和环境污染物(香烟、空气污染等)。其中约有80%的老化是由日晒所造成,由日晒引起的皮肤老化被称为光老化(Photoaging),主要是由紫外线所引起。由于地球臭氧层日益被破坏,亦提高了地球上的紫外线辐射强度,长期不当暴露于如此环境中,皮肤容易受到伤害。
目前,已知茄红素在人体内可以发挥抗氧化的作用,其抗氧化和抗老化的效果远远高于β-胡萝卜素、维生素E,并可以消除造成人体疾病和老化的自由基,特别是单线态氧(Single oxygen)。
虽然茄红素是一种强抗氧化物,但人体不会制造茄红素,必须由外界补充,加上人体内抗自由基系统会随着年龄的增加而衰退,过多的自由基会引发很多种疾病,所以适时适量补充茄红素可以减少疾病的发生和增强体能。
目前,四氢茄红素一般都是从番茄中萃取,产量太小,满足不了市场的需要,因此,迫切需要一种能大量生产四氢茄红素的技术,满足市场需求。
发明内容
本发明的目的在于克服现有技术的不足,提供一种解决上述技术问题的方法。
为了达到上述目的,本发明提供了一种四氢茄红素的应用,所述的四氢茄红素为用作抑制紫外线诱发皮肤内胶原蛋白分解的添加物。
优选地,所述的四氢茄红素为用作促进皮肤内胶原蛋白生成的添加物。
优选地,所述的四氢茄红素为用于制备动物饲料、人类食品、化妆品或保养品的添加物。
优选地,所述的四氢茄红素通过抑制紫外线诱发正常皮肤纤维母细胞内间质性胶原酶的活性,从而降低胞外基质的降解,达到降低胶原蛋白的分解。
优选地,包括如下步骤:
(A)将野生原生光合菌放入培养皿中培养出含有四氢茄红素的光合菌;
(B)将含有四氢茄红素的光合菌接种到发酵槽进行大量培养;
(C)培养出的原生光合菌,采用油脂萃取方法,萃取出四氢茄红素。
本发明的有益效果是,四氢茄红素能有效诱发正常皮肤纤维母细胞内间质性胶原酶(MMP1)的活性,降低胞外基质的降解,有效抑制紫外线诱发皮肤内胶原蛋白的分解,促进胶原蛋白的生成,从而减缓皮肤老化。另外,本发明的四氢茄红素可以大量生产,解决了传统从番茄中萃取四氢茄红素产量低的问题,满足市场需求。

具体实施方式
以下结合附图和具体实施例,对本发明做进一步说明。
参见图1,原生光合菌的传统生产流程,甲基辛烯二甲基辛烯焦磷酸(Geranylgeranyl diphosphate)在菌种B的作用下,生成八氢番茄红素(Phytoene),该八氢番茄红素在菌种I的作用下,生成六氢番茄红素(Phytofluene),接着生成ζ-胡萝卜菌(ζ-carotene),然后生成四氢番茄红素(Neurosporene),最后生成茄红素(Lycopene),该茄红素在菌种C的作用下,生成紫菌红素(Rhodopin),该紫菌红素在菌种D的作用下,生成3,4-二脱氢紫菌红素(3,4-didehydrorhodopin),该3,4-二脱氢紫菌红素在菌种F的作用下,生成脱水紫菌红醇(Anhydro-rhodovibrin),该脱水紫菌红醇在菌种C的作用下,生成紫菌红醇(Rhodovibrin),该紫菌红醇在菌种D的作用下,生成单脱甲基螺菌黄素(Monodemethyl-spirilloxanthin),该单脱甲基螺菌黄素在菌种F的作用下,生成螺菌黄素(Spirilloxanthin)。其中四氢番茄红素在菌种C的作用下,还可以生成羟基四氢番茄红素(hydroxyneurosporene),该羟基四氢西红柿红素在菌种F的作用下,生成甲氧基四氢番茄红素(Methoxyneurosporene),该甲氧基四氢番茄红素在菌种D的作用下,生成球形烯(Spheroidene),该球形烯在菌种C的作用下,生成羟基球形烯羟(Hydroxyspheroidene)。在菌种A的作用下,生成球形酮(Spheroidenone)。
本发明中,首先将野生原生光合菌(Rhodobacter sphaeroides)选至培养皿中进行培养,再将繁殖出含有四氢茄红素的光合菌接种到发酵槽进行大量培养,最后利用油脂萃取方法,萃取出四氢茄红素。
细胞株与培养条件
以正常皮肤细胞株作为模式。将人类正常皮肤细胞株(Detrolt 551细胞)加入到含10%FBS的DMEM培养基,放置在37℃含有5%CO2培养箱中培养,至8分满后,分别加入不同浓度的四氢茄红素,在16-24h作用后,对Detrolt 551细胞进行细胞毒性及抑制紫外线诱发胶原蛋白分解的分析。
细胞存活率分析(MTT viability assay)
利用MTT细胞存活率分析球形酮对Detrolt 551细胞的影响,MTT是经细胞内粒腺体去氢酶(Dehydrogenase)的分解,由透明无色产生蓝色甲臜(Formazan)晶体,细胞死亡则酵素失活,故蓝色结晶的多寡可判断细胞的存活力。首先将细胞置于24孔微量平盘,每孔有1ml新鲜的培养液约含104cells,培养隔夜后,对细胞存活力进行测定。在测定前,先将MTT以未含有FBS的DMEM培养液稀释成0.2mg/ml,将1ml MTT稀释液加入到每孔细胞中,在37℃下培养1小时,在显微镜下,可观察到细胞内有蓝色结晶生成。将24孔的微量平盘倒盖,移去培养液,用1ml的二甲基亚砜(DMSO)溶解,均匀摇晃5分钟直到看不到晶体后,在492nm读取其吸光值。将浓度为1μM,2μM,10μM及20μM的四氢茄红素分别加入到Detrolt 551细胞中,培养24小时后,利用MTT细胞存活率分析四氢茄红素对Detrolt 551细胞的影响。由图2可知,当浓度到达20M时,Detrolt 551细胞存活率仍然有95%,表明四氢茄红素对于Detrolt 551细胞株的细胞存活无显著抑制作用。
细胞紫外线伤害模式
由于紫外线照射皮肤细胞,会增加细胞脂质过氧化程度、DNA断裂与自由基的产生。将用强酸性电解水(AEW)处理的皮肤细胞放置在UV箱内,温度保持在23~25℃,采用放射波长为365nm的UV灯照射,每隔十分钟将细胞轻微摇荡并变换照射位置,然后加入0.5mM的二丁基羟基甲苯(BHT)终止反应。UV照射剂量通过测定UV灯与细胞之间的暴露剂量,再乘以照射时间换算总暴露UV的剂量。
在本发明中,以采用0J/cm2、5J/cm2、10J/cm2、15J/cm2及20J/cm2,五种不同照射剂量的UVA对皮肤细胞进行照射。由图3可知,皮肤细胞经UVA照射后,胶原蛋白会随着UVA照射剂量增加而下降。当UVA照射剂量为15J/cm2,胶原蛋白显著减少。随着UVA照射剂量增加,胶原蛋白减少的程度越明显。当UVA照射剂量为20J/cm2时,胶原蛋白蛋白明显减少,因此,本发明采用UVA 20J/cm2照射Detrolt 551 cells,观察不同浓度四氢茄红素对胶原蛋白分解的影响。
细胞胶原蛋白含量分析
胶原蛋白分解前,胞外基质(Procollagen)会先降解;且第一型胶原蛋白的胞外基质含量最多,故本发明选用细胞内第一型胞外基质蛋白质进行西方转渍法分析(Westernblot analysis)。将细胞加入20%十二烷基磺酸钠(SDS),1mM苯甲基磺酸氟(PMSF)中,置于冰上,采用超声波将细胞震破,放置于离心机中,在温度为4℃,转速为12000rpm时,离心20min,收集清液,以伯乐蛋白定量测试盒(Bio-Rad protein assay kit)定量蛋白质。取40g蛋白质样品进行十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分析,然后再转渍到硝酸纤维素膜(NC-membrane)上,用5%脱脂阻断液进行阻断后,先与第一抗体进行过夜反应后,再与第二抗体反应1h,然后利用化学冷光套组(ECL kit)冷光呈色,最后用成像分析仪(Matrox Inspector 2.1)作定量分析。
由图4可知,与无照射细胞内胶原蛋白含量相比,经20J/cm2UVA照射后细胞内胶原蛋白含量降低达70%,把不同浓度的四氢茄红素加入到20J/cm2UVA照射后细胞上,24h后,测量细胞内胶原蛋白的含量,发现,当四氢茄红素浓度为2μM时,细胞内胶原蛋白的含量开始增加,当四氢茄红素浓度为20μM,细胞内胶原蛋白的含量超过UVA照射前的量。由此可知,四氢茄红素不仅能抑制紫外线对正常皮肤纤维母细胞内胶原蛋白的分解,而且能促进胶原蛋白的生成。
四氢茄红素对间质性胶原酶(MMP1)活性的影响
由图5可知,与无照射细胞内MMP1含量相比,经20J/cm2 UVA照射后细胞内MMP1活性增加达70%,把不同浓度的四氢茄红素加入到20J/cm2UVA照射后细胞上,24h后,测量细胞内MMP1的含量,发现,当四氢茄红素浓度为2μM时,细胞内MMP1活性显著降低,而当四氢茄红素浓度为5μM时,降低至照射UVA前细胞内MMP1活性。结合图6,对应不同条件下的MMP1显微镜图,可知,四氢茄红素能显著抑制紫外线诱发正常皮肤纤维母细胞内MMP1的活性,从而降低胞外基质的降解,达到降低胶原蛋白的分解,减缓皮肤老化。
以上已对本发明创造的较佳实施例进行了具体说明,但本发明创造并不限于所述的实施例,熟悉本领域的技术人员在不违背本发明创造精神的前提下还可以作出种种的等同的变型或替换,这些等同变型或替换均包含在本申请权利要求所限定的范围内。

2.根据权利要求1所述的四氢茄红素的应用,其特征在于,所述的四氢茄红素为用作促进皮肤内胶原蛋白生成的添加物。
3.根据权利要求1或2所述的四氢茄红素的应用,其特征在于,所述的四氢茄红素为用于制备动物饲料、人类食品、化妆品或保养品的添加物。
4.根据权利要求1所述的用于制备四氢茄红素的应用,其特征在于,所述的四氢茄红素通过抑制紫外线诱发正常皮肤纤维母细胞内间质性胶原酶的活性,从而降低胞外基质的降解,达到降低胶原蛋白的分解。
5.一种用于制备如权利要求1-3任一项所述的四氢茄红素的方法,其特征在于,包括如下步骤:
(A)将野生原生光合菌放入培养皿中培养出含有四氢茄红素的光合菌;
(B)将含有四氢茄红素的光合菌接种到发酵槽进行大量培养;
(C)培养出的原生光合菌,采用油脂萃取方法,萃取出四氢茄红素。
