本发明公开了一种黄芪多糖脂质体的制备方法,步骤如下:将卵磷脂、胆固醇置于干燥洁净的茄形瓶中,加入无水乙醇超声溶解,于40‑50℃水浴中减压蒸发除去乙醇,使磷脂类膜材在茄形瓶壁上形成均匀的脂质薄膜;将APS溶解在纯化水中制得水相,44‑46℃水浴预热,将其缓慢滴注于茄形瓶内,边滴加边振荡,至薄膜从茄形瓶壁上完全脱落溶解,旋转水合70‑90min;将所得混悬液超声20‑30min,过微孔滤膜,即得黄芪多糖脂质体混悬液,4℃冰箱冷藏即可。该方法简便、快捷、易操作,制备的黄芪多糖脂质体包封率高,4℃下贮存较稳定,且具有良好的缓释作用。

技术领域
本发明涉及一种黄芪多糖脂质体的制备方法。
背景技术
黄芪是常用大宗药材,始载于《神农本草经》。具有利水消肿、补气升阳、生津养血、固表止汗等功效。黄芪多糖(Astragalus polysaccharide,APS)是黄芪的主要活性成分,具有广泛药理活性,包括免疫调节、抗衰老、抗病毒、抗氧化、抗肿瘤、抗辐射等作用。作为潜在胰岛素敏化剂用于Ⅱ型糖尿病的治疗
目前黄芪多糖的常规剂型在使用过程中存在着缺陷,比如药物在达到作用部位之前,大部分已被降解代谢,因此需要大剂量连续给药。脂质体是一种新型靶向给药载体,无害无毒、具有靶向释药、缓释、可生物降解、降低不良反应、可提高药物治疗指数等优势,因此,长期受到医学界的高度关注,成为医学界一直以来的研究热点。针对黄芪多糖口服吸收差、体内半衰期短、生物利用度低等缺点,本发明利用薄膜蒸发超声法,将药物黄芪多糖载于其中,制备成黄芪多糖脂质体。
发明内容
本发明的目的在于提供一种黄芪多糖脂质体的制备方法。
本发明通过下面技术方案实现:
一种黄芪多糖脂质体的制备方法,包括如下步骤:将7-9份卵磷脂、10-20份胆固醇置于干燥洁净的茄形瓶中,加入55-65份无水乙醇超声溶解,于40-50℃水浴中减压蒸发除去乙醇,使磷脂类膜材在茄形瓶壁上形成均匀的脂质薄膜;将10-20份APS溶解在65-75份纯化水中制得水相,44-46℃水浴预热,将其缓慢滴注于茄形瓶内,边滴加边振荡,至薄膜从茄形瓶壁上完全脱落溶解,旋转水合70-90min;将所得混悬液超声20-30min,依次过0.45μm微孔滤膜3次,0.22μm微孔滤膜3次,即得黄芪多糖脂质体混悬液,4℃冰箱冷藏即可;各原料均为重量份。
优选地,所述的制备方法中,于45℃水浴中减压蒸发除去乙醇。
优选地,所述的制备方法中,45℃水浴预热。
优选地,所述的制备方法中,将所得混悬液超声25min。
本发明技术效果:
该方法简便、快捷、易操作,制备的黄芪多糖脂质体包封率高,4℃下贮存较稳定,且具有良好的缓释作用。

下面结合实施例具体介绍本发明的实质性内容。
实施例1
一种黄芪多糖脂质体的制备方法,包括如下步骤:将8份卵磷脂、15份胆固醇置于干燥洁净的茄形瓶中,加入60份无水乙醇超声溶解,于45℃水浴中减压蒸发除去乙醇,使磷脂类膜材在茄形瓶壁上形成均匀的脂质薄膜;将15份APS溶解在70份纯化水中制得水相,45℃水浴预热,将其缓慢滴注于茄形瓶内,边滴加边振荡,至薄膜从茄形瓶壁上完全脱落溶解,旋转水合80min;将所得混悬液超声25min,依次过0.45μm微孔滤膜3次,0.22μm微孔滤膜3次,即得黄芪多糖脂质体混悬液,4℃冰箱冷藏即可;各原料均为重量份。
实施例2

一种黄芪多糖脂质体的制备方法,包括如下步骤:将9份卵磷脂、20份胆固醇置于干燥洁净的茄形瓶中,加入65份无水乙醇超声溶解,于50℃水浴中减压蒸发除去乙醇,使磷脂类膜材在茄形瓶壁上形成均匀的脂质薄膜;将20份APS溶解在75份纯化水中制得水相,46℃水浴预热,将其缓慢滴注于茄形瓶内,边滴加边振荡,至薄膜从茄形瓶壁上完全脱落溶解,旋转水合90min;将所得混悬液超声30min,依次过0.45μm微孔滤膜3次,0.22μm微孔滤膜3次,即得黄芪多糖脂质体混悬液,4℃冰箱冷藏即可;各原料均为重量份。
该方法简便、快捷、易操作,制备的黄芪多糖脂质体包封率高,4℃下贮存较稳定,且具有良好的缓释作用。
