本发明涉及一种牛蒡苷泡腾片的制备方法,属于食品加工技术领域。
背景技术
牛蒡苷是一种木脂素类化合物,为无色结晶性粉末,熔点110~112℃。作为重要的天然化合物,牛蒡苷具有很多的药理学作用。一直以来,其都因疏散风热、祛痰止咳、清热解毒广泛应用于中医方剂当中。随着对于牛蒡苷的研究的深入,人们发现牛蒡苷具有抗肿瘤、免疫调节及抗炎、抗菌抗病毒、抗糖尿病、抗氧化、肝保护等多种药理作用。其抗癌活性,抗病毒活性,免疫调节作用可以将其应用于癌症治疗或辅助治疗药物;同时其抗糖尿病作用也是值的深入研究和有开发应用价值的。另外,牛蒡苷对于治疗男性性无能(即阳痿)和男性不育具有一定的药理学作用,综上所述,对于牛蒡苷的研究具有广阔的市场前景。
发明内容
针对现有技术的不足,本发明提供一种方法设计合理、可操作性强、加工过程中营养及功能性成分损失小、食用方便的一种牛蒡苷泡腾片的制备方法。
为了实现上述目的,本发明提供了一种牛蒡苷泡腾片的制备方法,包括以下步骤:
(1)打粉:将牛蒡根用组织粉碎机进行粉碎,过100目筛,得牛蒡粉;
(2)酶解:将牛蒡粉用碱性乙醇溶液在微波条件下进行提取,得到含有牛蒡苷粗品的提取液;所述碱性乙醇的pH值为9~11,微波功率为500~700W,设定温度为35~39℃,原料∶提取液=1∶(3~5)m/v,微波提取时间为15~20min;
(3)水解:提取液在微波条件下进行水解;
(4)中和:向提取液中加入碱性水溶液,调节提取液的pH值为中性;

(7)浓缩:将含有牛蒡苷的乙醇溶液进行减压浓缩,得到浓缩液;
(8)干燥:浓缩液采用鼓风烘干,得到牛蒡苷;
(9)制酸粒:将7.0~12.0g乳糖、4.0~8.0g甘露醇、10.0~15.0g步骤(8)制备得到的牛蒡苷、2.0~4.5g柠檬酸、1~2g羧甲基纤维素钠混合均匀,喷洒5%~10%的聚乙烯吡咯烷酮乙醇溶液1.0~7.0ml作粘合剂,溶剂乙醇的浓度为50%~85%,制软材,过30~40目筛制粒,置于40~80℃烘箱中干燥3~5h,用40~60目和50~80目筛整粒,形成酸颗粒;
(10)制碱粒:将6.5~8.5g碳酸氢钠、7.0~12.0g乳糖、4.0~8.0g甘露醇、10.0~15.0g步骤(8)制备得到的牛蒡苷、1~2g羧甲基纤维素钠混合均匀,喷洒5%~10%的聚乙烯吡咯烷酮乙醇溶液1.0~7.0ml作粘合剂,溶剂乙醇的浓度为50%~85%,制软材,即可过30~40目筛制粒,然后置于40~80℃烘箱中干燥3~5h,用40~60目和50~80目筛整粒,形成碱颗粒;
(11)成品:将4~10g酸颗粒、3~9g碱颗粒、3~8g PEG6000与0.4~0.7g微粉硅胶、0.2~0.6g甜蜜素混合均匀,上压片机进行压片,每片片重在0.2~0.4g。

实施例1
一种牛蒡苷泡腾片的制备方法,包括以下步骤:
(1)打粉:将牛蒡根用组织粉碎机进行粉碎,过100目筛,得牛蒡粉;

(2)酶解:将牛蒡粉用碱性乙醇溶液在微波条件下进行提取,得到含有牛蒡苷粗品的提取液;所述碱性乙醇的pH值为10,微波功率为600W,设定温度为37℃,原料∶提取液=1∶4m/v,微波提取时间为18min;
(3)水解:提取液在微波条件下进行水解;
(4)中和:向提取液中加入碱性水溶液,调节提取液的pH值为中性;
(5)萃取:向提取液中加入正己烷进行萃取,摇匀后静置,分液,得到水相;
(6)纯化:将水相先用大孔吸附树脂进行吸附,然后用乙醇溶液对大孔吸附树脂进行解析,得到含有牛蒡苷的乙醇溶液;所述大孔吸附树脂型号为HPD-100、D3520、X-5、S-8、AB-8、D201或D301-G,所述乙醇浓度为50%;
(7)浓缩:将含有牛蒡苷的乙醇溶液进行减压浓缩,得到浓缩液;
(8)干燥:浓缩液采用鼓风烘干,得到牛蒡苷;
(9)制酸粒:将9.5g乳糖、6.0g甘露醇、12.5g步骤(8)制备得到的牛蒡苷、3.0g柠檬酸、1.5g羧甲基纤维素钠混合均匀,喷洒7.5%的聚乙烯吡咯烷酮乙醇溶液4.0ml作粘合剂,溶剂乙醇的浓度为70%,制软材,过30~40目筛制粒,置于60℃烘箱中干燥4h,用40~60目和50~80目筛整粒,形成酸颗粒;
(10)制碱粒:将7.5g碳酸氢钠、9.5.0g乳糖、6.0g甘露醇、12.5g步骤(8)制备得到的牛蒡苷、1.5g羧甲基纤维素钠混合均匀,喷洒7.5%的聚乙烯吡咯烷酮乙醇溶液4.0ml作粘合剂,溶剂乙醇的浓度为70%,制软材,即可过30~40目筛制粒,然后置于60℃烘箱中干燥4h,用40~60目和50~80目筛整粒,形成碱颗粒;
(11)成品:将7g酸颗粒、6g碱颗粒、7.5g PEG6000与0.55g微粉硅胶、0.4g甜蜜素混合均匀,上压片机进行压片,每片片重在0.3g。
实施例3
一种牛蒡苷泡腾片的制备方法,包括以下步骤:
(1)打粉:将牛蒡根用组织粉碎机进行粉碎,过100目筛,得牛蒡粉;
(2)酶解:将牛蒡粉用碱性乙醇溶液在微波条件下进行提取,得到含有牛蒡苷粗品的提取液;所述碱性乙醇的pH值为11,微波功率为700W,设定温度为35℃,原料∶提取液=1∶5m/v,微波提取时间为15min;
(3)水解:提取液在微波条件下进行水解;
(4)中和:向提取液中加入碱性水溶液,调节提取液的pH值为中性;
(5)萃取:向提取液中加入正己烷进行萃取,摇匀后静置,分液,得到水相;
(6)纯化:将水相先用大孔吸附树脂进行吸附,然后用乙醇溶液对大孔吸附树脂进行解析,得到含有牛蒡苷的乙醇溶液;所述大孔吸附树脂型号为HPD-100、D3520、X-5、S-8、AB-8、D201或D301-G,所述乙醇浓度为50%;
(7)浓缩:将含有牛蒡苷的乙醇溶液进行减压浓缩,得到浓缩液;
(8)干燥:浓缩液采用鼓风烘干,得到牛蒡苷;
(9)制酸粒:将12.0g乳糖、8.0g甘露醇、15.0g步骤(8)制备得到的牛蒡苷、4.5g柠檬酸、2g羧甲基纤维素钠混合均匀,喷洒10%的聚乙烯吡咯烷酮乙醇溶液7.0ml作粘合剂,溶剂乙醇的浓度为85%,制软材,过30~40目筛制粒,置于80℃烘箱中干燥3h,用40~60目和50~80目筛整粒,形成酸颗粒;
(10)制碱粒:将8.5g碳酸氢钠、12.0g乳糖、8.0g甘露醇、15.0g步骤(8)制备得到的牛蒡苷、2g羧甲基纤维素钠混合均匀,喷洒10%的聚乙烯吡咯烷酮乙醇溶液7.0ml作粘合剂,溶剂乙醇的浓度为85%,制软材,即可过30~40目筛制粒,然后置于80℃烘箱中干燥3h,用40~60目和50~80目筛整粒,形成碱颗粒;
(11)成品:将10g酸颗粒、9g碱颗粒、8g PEG6000与0.7g微粉硅胶、0.6g甜蜜素混合均匀,上压片机进行压片,每片片重在0.4g。
通过上述实施例制备的泡腾片,方便携带,产品稳定性好,本发明生产工艺简便易行、经济高效、工业化程度高、对资源的利用率高,牛蒡苷纯度高,且具有显著的抗氧化活性,具有极大的市场开发潜力。
Claims (6)
1.一种牛蒡苷泡腾片的制备方法,其特征在于,包括以下步骤:
(1)打粉:将牛蒡根用组织粉碎机进行粉碎,过100目筛,得牛蒡粉;
(2)酶解:将牛蒡粉用碱性乙醇溶液在微波条件下进行提取,得到含有牛蒡苷粗品的提取液;所述碱性乙醇的pH值为9~11,微波功率为500~700W,设定温度为35~39℃,原料∶提取液=1∶(3~5)m/v,微波提取时间为15~20min;
(3)水解:提取液在微波条件下进行水解;
(4)中和:向提取液中加入碱性水溶液,调节提取液的pH值为中性;
(5)萃取:向提取液中加入正己烷进行萃取,摇匀后静置,分液,得到水相;
(6)纯化:将水相先用大孔吸附树脂进行吸附,然后用乙醇溶液对大孔吸附树脂进行解析,得到含有牛蒡苷的乙醇溶液;
(7)浓缩:将含有牛蒡苷的乙醇溶液进行减压浓缩,得到浓缩液;
(8)干燥:浓缩液采用鼓风烘干,得到牛蒡苷;
(9)制酸粒:将7.0~12.0g乳糖、4.0~8.0g甘露醇、10.0~15.0g步骤(8)制备得到的牛蒡苷、2.0~4.5g柠檬酸、1~2g羧甲基纤维素钠混合均匀,喷洒5%~10%的聚乙烯吡咯烷酮乙醇溶液1.0~7.0ml作粘合剂,溶剂乙醇的浓度为50%~85%,制软材,过30~40目筛制粒,置于40~80℃烘箱中干燥3~5h,用40~60目和50~80目筛整粒,形成酸颗粒;
(10)制碱粒:将6.5~8.5g碳酸氢钠、7.0~12.0g乳糖、4.0~8.0g甘露醇、10.0~15.0g步骤(8)制备得到的牛蒡苷、1~2g羧甲基纤维素钠混合均匀,喷洒5%~10%的聚乙烯吡咯烷酮乙醇溶液1.0~7.0ml作粘合剂,溶剂乙醇的浓度为50%~85%,制软材,即可过30~40目筛制粒,然后置于40~80℃烘箱中干燥3~5h,用40~60目和50~80目筛整粒,形成碱颗粒;
(11)成品:将4~10g酸颗粒、3~9g碱颗粒、3~8g PEG6000与0.4~0.7g微粉硅胶、0.2~0.6g甜蜜素混合均匀,上压片机进行压片,每片片重在0.2~0.4g。
2.根据权利要求1所述的一种牛蒡苷复合速溶营养粉,其特征在于:所述步骤(2)中,所述碱性乙醇的pH值为10,微波功率为600W,设定温度为37℃,原料∶提取液=1∶4m/v,微波提取时间为18min。
3.根据权利要求1或2所述的一种牛蒡苷泡腾片,其特征在于:所述步骤(6)中,所述大孔吸附树脂型号为HPD-100、D3520、X-5、S-8、AB-8、D201或D301-G,所述乙醇浓度为50%。
4.根据权利要求1或2所述的一种牛蒡苷泡腾片,其特征在于:所述步骤(9)中,将9.5g乳糖、6.0g甘露醇、12.5g步骤(8)制备得到的牛蒡苷、3.0g柠檬酸、1.5g羧甲基纤维素钠混合均匀。
5.根据权利要求1或2所述的一种牛蒡苷泡腾片,其特征在于:所述步骤(10)中,将7.5g碳酸氢钠、9.5.0g乳糖、6.0g甘露醇、12.5g步骤(8)制备得到的牛蒡苷、1~2g羧甲基纤维素钠混合均匀。
6.根据权利要求1或2所述的一种牛蒡苷泡腾片,其特征在于:所述步骤(11)中,将7g酸颗粒、6g碱颗粒、7.5g PEG6000与0.55g微粉硅胶、0.4g甜蜜素混合均匀,上压片机进行压片,每片片重在0.3g。
