NK/T细胞淋巴瘤(natural killer/T-cell lymphoma,NK/TCL)是一类CD56阳性/胞浆CD3阳性的淋巴细胞恶性增殖性疾病,好发于亚洲地区,与Epstein-Barr病毒(EB病毒)感染密切相关。在过去二十年间,一系列分子生物学研究报道了与NK/TCL疾病发生进展相关的多种遗传学及分子生物学异常。比较基因组研究(Comparative genomichybridization,CGH)报道了一系列疾病相关DNA基因拷贝数变化(copy numberalterations),包括2q5,13q4,10q3,21q2,3q2,5q2和17q2等染色体区段的扩增,以及1p4,17p4,12q3,13q2和6q1等染色体区段的缺失;全外显子测序研究(whole exomesequencing,WES)报道了以DDX3X,TP53,MGA,JAK-STAT信号分子(STAT3和STAT5B),表观遗传学调控分子(MLL2,EP300,ARID1A和ASXL3)等为代表的一系列肿瘤相关的重现性突变。全基因组表达谱(comprehensive genome-wide gene expression profiling,GEP)研究发现,细胞周期相关信号通路及信号分子,如PLK1,CDK1,Aurora-A等,抗凋亡信号通路及信号分子,如MYC,TP53,NF-kB p50等,均存在显著异常调控。启动子及位点特异性甲基化研究发现,NK/TCL中存在显著的高甲基化,表现为以抑癌基因及BCL2L11(BIM),DAPK1,PTPN6(SHP1),TET2,SOCS6和ASNS等基因的转录抑制。microRNA芯片检测,发现miR-101,miR-26a,miR26b,miR-28-5和miR-363存在显著异常表达,通过调控靶基因MUM1,BLIMP1和STMN1等,引起细胞周期、TP53和MAPK等信号通路异常。全基因组关联研究(genome-wideassociation study,GWAS)发现HLA-DPB1的基因组多态性与疾病发生密切相关。然而,上述研究均为基于患者临床标本的组学研究结果,NK/TCL致病分子机制尚待阐明。由于致病机制不明确,缺乏特异性的治疗靶点和标准化治疗策略,NK/TCL患者的疗效预后极差。即使应用当前最优的治疗策略——基于门冬酰胺酶的抗代谢治疗联合放疗,早期患者的5年生存率仅为72%(155例患者统计数据),晚期患者的5年生存率仅为50%(87例患者统计数据)。NK/TCL呈高度恶性,侵袭性强,常规化疗耐药,预后极差,进展期患者五年生存率仅为20%,已成为严重危害人民健康的疾病。制约NK/TCL发病机制和靶向治疗研究的最主要因素是缺乏可供开展分子生物学研究的NK/TCL动物肿瘤模型。
动物肿瘤模型主要用于分子生物学(包括致病机制研究、治疗靶点新药筛选)研究。人类疾病十分复杂,临床积累的经验在时间和空间上都存在局限性,以人作为实验对象来研究疾病发生机制存在许多伦理和方法上的限制。动物肿瘤模型极大地促进了肿瘤分子生物学研究的进展:1.动物模型与人类具有较高的同源性,且繁殖速度快、样本数目多,可复制性高,实验结果可与人类疾病进行比较研究,快捷有效地认识人类疾病的发生发展规律;2.动物模型可以特异性的进行遗传学修饰,严格控制实验条件,增强实验结果的科学性和可比较性,有助于全面认识疾病的本质;3.动物模型可改变自然条件下不可能或不易改变的因素,尤其在潜伏期长、病程长和发病率低的疾病研究中具有极大的应用价值。
人源肿瘤异种移植模型(patient-derived xenograft model,PDX模型)是指将患者的肿瘤组织取下后,接种在免疫缺陷的动物(目前主要是小鼠)体内,小鼠成瘤以后进一步传代以待后续研究。构建的肿瘤模型既保留了肿瘤患者的组织学和遗传学特征,又维持了肿瘤的异质性。PDX模型能帮助科学家更好地研究肿瘤异质性和遗传复杂性,更深入探究肿瘤发生机制及发现潜在治疗靶点,还可以帮助预测患者对治疗药物可能产生的反应,包括疗效、毒副作用、吸收程度等,因此被广泛的应用到肿瘤药物研发当中。相比传统肿瘤细胞异种移植动物模型,应用PDX模型进行一些治疗方法临床应用潜力的评估会更加精确,它能提供更好的临床前药物疗效测试及分析。近年来,随着肿瘤精准医学概念的提出,对PDX小鼠模型的需求和利用正日益扩大,尤其是借助于PDX小鼠模型进行肿瘤有效药物的筛选和评估,并逐步将其应用于临床上指导肿瘤患者的个体化治疗。PDX模型结合临床数据、基因组图谱以及药效数据可以增加药物特异性,应用于肿瘤患者个体化治疗,提高临床治疗成功率。
斑马鱼(Zebrafish,Danio rerio)是一种国际性的生物医学研究模式生物,具有以下特点:1.成鱼体积小,仅3-4cm;繁殖周期短,仅3个月;养殖成本低。产卵量大可用于大规模研究;2.鱼卵和胚胎自身具有胚胎学和遗传学的突出特征。发育早期3-7天呈透明样,便于观察,免疫系统成熟较晚,便于成瘤;3.斑马鱼生物信息,如全基因组测序信息等完善。因此,利用斑马鱼作为人类疾病的动物模型,其独特的生物学、基因组学、遗传学优势及其高度保守的疾病信号通路,使其成为以表型驱动的“正向”及“反向”遗传学(Forward andReverse Genetics)研究,及活体内先导药物筛选研究的最佳模式生物之一。斑马鱼作为新型的模式生物,与细胞和小鼠相比,具有高通量、短周期和易操作等优势。然而,斑马鱼人源肿瘤异种移植模型(Zebrafish patient-derived xenograft model,zPDX模型)的构建及相关分子生物学研究尚处于起步阶段。
关于NK/TCL的动物模型,人源肿瘤模型或斑马鱼肿瘤模型领域的研究仍是一片空白。
发明内容
为了克服现有技术中所存在的问题,本发明的目的在于提供一种斑马鱼NK/TCL肿瘤模型的构建方法及其应用。
为了实现上述目的以及其他相关目的,本发明采用如下技术方案:
本发明的第一方面,提供一种斑马鱼NK/TCL肿瘤模型的构建方法,包括如下步骤:将人源NK/TCL细胞注射到斑马鱼胚胎中,培养成瘤。
本发明中,所述NK/TCL是指NK/T细胞淋巴瘤(natural killer/T-celllymphoma)。
进一步地,所述方法中,所述斑马鱼胚胎是生长了48个小时的斑马鱼胚胎。
进一步地,所述方法中,是将人源NK/TCL细胞注射到斑马鱼胚胎的卵黄囊中。
进一步地,所述方法中,所述人源NK/TCL细胞为人源原代NK/TCL细胞。
进一步地,所述方法中,所述人源NK/TCL细胞为NK/TCL患者术后取得或NK/TCL患者活检取得。
进一步地,每个斑马鱼胚胎中注射人源NK/TCL细胞5个以上。

本发明的第三方面,提供一种筛选NK/TCL治疗药物的方法,包括步骤:将备选物质施用于前述构建方法构建获得的斑马鱼NK/TCL肿瘤模型,若能抑制斑马鱼NK/T细胞淋巴瘤的生长,则可以作为潜在NK/TCL治疗药物。
与现有技术相比,本发明具有如下有益效果:
本发明经过研究首次以斑马鱼作为模式生物成功构建了NK/TCL人源肿瘤异种移植模型。实验操作简便可行,实验周期短,消耗肿瘤细胞极少,可重复性高,可广泛应用于分子生物学研究及靶向药物筛选等多种领域。

实施例1斑马鱼NK/TCL肿瘤模型的构建
1.材料和方法
1.1试剂
Bambanker购自WAKO公司,用于肿瘤组织的冻存。
Liberase购自Sigma公司,用于肿瘤组织消化。
Dil购自Invitrogen公司,用于肿瘤细胞的荧光染色。
1.2肿瘤组织消化及肿瘤细胞染色
患者术后或活检新鲜的NK/T细胞淋巴瘤组织切成1mm*1mm大小,置于Bambanker溶液中,冻存(于﹣80℃冰箱中保存)。
取实验所用的NK/T细胞淋巴瘤组织经Liberase消化成单细胞悬液计数,用PBS按照1*10^7/mL密度重悬后,用Dil染料进行染色,NK/TCL细胞在荧光显微镜下观察呈红色荧光。
1.3人源肿瘤细胞显微注射
取48小时的斑马鱼胚胎,人工破膜麻醉后置于显微注射板上,将1.2中染色的人源NK/TCL细胞(约100个)加入显微注射针中,显微镜下操作将原代NK/TCL细胞注射进入卵黄囊中。
注射结束后即可、24小时或48小时于荧光显微镜下观察成瘤情况。
2.结果
2.1斑马鱼成瘤情况如图1所示:本研究发现,注射结束后即时观察结果和24小时后,可明确看到斑马鱼卵黄囊内成瘤的肿瘤组织,比较注射结束后24小时和48小时,肿瘤组织有明显增大,说明肿瘤组织具有显著的恶性增殖能力。
经统计分析,上述构建方法的成瘤率为100%。通过病理切片观察,病理切片结果显示,肿瘤组织病理切片的病理类型、免疫组化染色、分子靶标检测均与人体NK/TCL肿瘤相似。说明本发明斑马鱼NK/TCL肿瘤模型构建成功。
另外重复上述构建方法十次,注射结束后即时观察结果和24小时后,均可明确看到斑马鱼卵黄囊内成瘤的肿瘤组织,比较注射结束后24小时和48小时,肿瘤组织均有明显增大,说明肿瘤组织具有显著的恶性增殖能力。经统计分析,重复上述构建方法10次,成瘤率均为100%。通过病理切片观察,病理切片结果显示,肿瘤组织病理切片的病理类型、免疫组化染色、分子靶标检测均与人体NK/TCL肿瘤相似。说明本发明斑马鱼NK/TCL肿瘤模型及其构建方法的可用性。
本研究首次成功构建了人源NK/TCL肿瘤模型,也就是说本发明的斑马鱼NK/TCL肿瘤模型是首次构建成功的NK/TCL动物模型。实验操作简便可行,实验周期短,消耗肿瘤细胞极少,可重复性高,可广泛应用于分子生物学学研究及靶向药物筛选等多种领域。
此外,本发明的斑马鱼NK/TCL肿瘤模型属于患者原代肿瘤组织的肿瘤移植模型(Patient-derived xenograft模型,PDX模型),具有如下应用:(1)通过在一定接种细胞数目条件下的成瘤率判断肿瘤细胞的恶性程度;(2)通过观察接种0h、24h、48h不同时相肿瘤大小,可用于肿瘤细胞增殖活性的观察。
3.讨论
在模型构建过程中,我们遇到了以下困难:
(1)肿瘤组织消化部分:
利用Liberase将肿瘤组织消化成单细胞悬液的实验方法,我们也进行了很多摸索。如果消化不充分,细胞损失会很大,但是如果消化时间过久,可能会导致肿瘤细胞的活性受损。因此,合适的消化时间和消化条件极大的影响了成瘤效率。在实验过程中,一方面,在消化前我们将肿瘤组织尽可能的切割成小块,合适的小块范围是(0.5~5.0)mm*(0.5~5.0)mm大小,以保证其余Liberase的充分接触,各部分细胞的消化效果能够尽可能同步。另一方面,我们会每隔5-10分钟在显微镜下动态观察消化效果,及时终止消化反应。经过试验发现,合适的消化时间范围是5-30分钟。合适的消化条件范围是室温到37℃。
(2)肿瘤细胞染色部分:
Dil染色过程中我们希望尽可能的充分有效持久的对肿瘤细胞进行染色以便进行后续观察,另一方面,我们希望能够尽可能降低噪点干扰。为了解决上述问题,我们对不同类型细胞通过设置Dil浓度梯度及染色时间梯度进行摸索观察,寻找最优的Dil浓度和染色时间。针对本发明的NK/T细胞,优选的Dil浓度范围是1-10uM。优选的Dil染色时间范围是1-15分钟。另一方面,我们发现染色结束后将细胞冰上静置后可有效减少非特异性结合。此外,染色结束后1*PBS多次清洗细胞也可极大改善噪点干扰。
(3)显微注射部分:
因为注射所用的是48h的斑马鱼胚胎,和常规注射用的单细胞胚胎存在较大不同,主要在于鱼的大小形状,卵黄囊的质地和韧性,这些因素都会极大的影响肿瘤细胞注射。在实验过程中,由于鱼的大小形状改变,我们首先对注射用的平板进行调整,不同于单细胞注射的凹槽形平板,我们采用平面平板,但是注射时由于缺乏支撑面,鱼容易滑动给注射带来了极大不便,在此基础之上,我们又通过滤纸尽量吸干平板内的水分,将一组多只鱼汇聚起来互相支撑的办法解决注射过程中鱼滑动的问题。另一方面,由于水分的缺乏,时间过久鱼的一般状态会变成很容易死亡,因此我们注射过程中严格控制一批注射的斑马鱼数目和注射时间,通常熟练操作情况下,一批小于100只鱼、15min以内分钟内完成注射。例如一批20只鱼、3分钟内完成注射。另外,卵黄囊的质地和韧性变化,导致显微注射针头的粗细有严格要求,由于48h小时的斑马鱼胚胎的卵黄囊较硬,针头过细无法穿透,过粗又会导致斑马鱼的卵黄囊爆裂和鱼的死亡。因此,我们通过反复摸索,在每次注射前对针头进行人工修剪,并通过预实验确保合适的粗细程度后再开始注射。经过摸索发现,合适的针头直径范围是0.1-1mm。
此外,由于纤维注射的针较细,注射肿瘤细胞不同于常规注射其他DNA或RNA物质,上样很困难。我们尝试了很多方法,一方面,使用了特制的特细上样Tip头进行上样,减少细胞悬液从针尾进样到针头处的时间;另一方面,通过调整细胞悬液的密度改变悬液的粘稠性,双管齐下彻底解决了上述问题。经过摸索发现,上样Tip头可采用(EppendorfRacks,0.1-10uL,PCR clean,Sterile)。合适的细胞悬液的密度范围是小于等于1*108/mL。
(4)注射肿瘤细胞数目:
研究中我们发现注射肿瘤细胞的数目不同会影响斑马鱼成瘤率。为寻找最合适的注射数目,通过梯度设置不同数目细胞进行注射,在不同类型的细胞中反复试验摸索,我们发现当肿瘤细胞在5个以上即可成瘤,100个以上肿瘤细胞数目时,肿瘤细胞斑马鱼成瘤率可达到90%以上。

实施例2斑马鱼NK/TCL肿瘤模型在药物筛选中的应用
采用实施例1的方法建立斑马鱼NK/TCL肿瘤模型,建模24h后通过荧光显微镜观察确认成瘤情况后,确认建模成功。选取45条,随机分组,用抗淋巴瘤化疗药物多柔比星(Doxorubicin)进行治疗。
1.分组及用药
对照组:20条,直接给予不含药物的egg water进行培养,培养液体积4mL。
药物治疗组:
将多柔比星(Doxorubicin)溶解于egg water中,分别形成浓度为0.02ng/mL和0.04ng/mL的药物溶液;
第一治疗组,10条,采用含药物的egg water进行培养,给药浓度为0.02ng/mL,培养液体积2mL;
第二治疗组,15条,采用含药物的egg water进行培养,给药浓度为0.04ng/mL,培养液体积3mL。
2.观察指标及方法
用药后每日观察计数NK/TCL斑马鱼模型的生存率。
3.结果
如图2所示,用药组(包括第一治疗组和第二治疗组)的NK/TCL斑马鱼的生存时间显著延长。而对照组egg water无作用。
此外,抗淋巴瘤化疗药物多柔比星(Doxorubicin)对于用药组(包括第一治疗组和第二治疗组)的肿瘤生长具有抑制作用,抑瘤率分别为9.94%和21.05%。而对照组eggwater无作用。
说明抗淋巴瘤化疗药物多柔比星(Doxorubicin)有效。充分说明,本发明的NK/TCL斑马鱼模型在筛选NK/TCL药物中成功应用。

Claims (7)
1.一种斑马鱼NK/TCL肿瘤模型的构建方法,包括如下步骤:将进行了标记的人源原代NK/TCL细胞注射到斑马鱼胚胎的卵黄囊中,培养成瘤。
2.根据权利要求1所述的方法,其特征在于,所述方法中,所述斑马鱼胚胎是生长了48个小时的斑马鱼胚胎。
3.根据权利要求1所述的方法,其特征在于,所述方法中,所述人源原代NK/TCL细胞进行了染色标记的人源原代NK/TCL细胞。
4.根据权利要求1所述的方法,其特征在于,所述方法中,所述人源原代NK/TCL细胞进行了荧光染色标记的人源原代NK/TCL细胞。
5.根据权利要求1所述的方法,其特征在于,所述方法中,取斑马鱼胚胎,人工破膜麻醉后置于显微注射板上,然后将人源原代NK/TCL细胞加入显微注射针中,显微镜下操作将所述人源原代NK/TCL细胞注射到斑马鱼胚胎中。
6.如权利要求1-5任一项所述斑马鱼NK/TCL肿瘤模型的构建方法用于NK/TCL治疗药物筛选的用途。
7.一种筛选NK/TCL治疗药物的方法,包括步骤:将备选物质施用于如权利要求1-5任一项所述构建方法构建获得的斑马鱼NK/TCL肿瘤模型,若能抑制斑马鱼NK/T细胞淋巴瘤的生长,则可以作为潜在NK/TCL治疗药物。
