After the extracting solution is obtained, centrifuging the extracting solution, and taking supernate to obtain refined solution; the centrifuging device is preferably an ultracentrifuge; the centrifugation time is preferably 20-40 min, and more preferably 30 min; the rotating speed of the centrifugation is preferably 7000-8000 r/min, and more preferably 7500 r/min.
After the refined liquid is obtained, concentrating the refined liquid to obtain an extract; the concentration mode is preferably reduced pressure concentration; the pressure of the reduced pressure concentration is preferably-80 kPa to-40 kPa, and more preferably-60 kPa; the relative density of the extract obtained by the reduced pressure concentration is preferably 1.1-1.3, and more preferably 1.2.

In the present invention, the first drying method is preferably vacuum drying; the first drying temperature is preferably 60-90 ℃, and more preferably 80 ℃; the first drying time is preferably 1-3 h, and more preferably 2 h; the crushing mode is preferably grinding and crushing; the granularity of the first sieve is preferably 90-110 meshes, and more preferably 100 meshes; when the soft material is prepared, starch slurry with volume concentration of 10% is preferably adopted as an adhesive; the granulation is preferably carried out by sieving with a 14-mesh sieve by adopting an extrusion method; the second drying mode is preferably fluidized bed drying; the second drying temperature is preferably 60-90 ℃, and more preferably 80 ℃; the second drying time is preferably 10-60 min, and more preferably 30 min; the granularity of the second screening is preferably 14-18 meshes, and more preferably 16 meshes.
The granules prepared by the invention have the advantages of good taste, uniform granularity, consistent color and luster, and no phenomena of moisture absorption, softening, caking, deliquescence and the like.
In the invention, when the health food is granules, the dosage of the traditional Chinese medicine composition is preferably 3-9 g/day, and more preferably 6 g/day.
The invention provides a preparation method of a health food tablet, which comprises the following steps:
a) mixing flos Lonicerae, Perillae herba, rhizoma Ligustici Chuanxiong, cortex Lycii and rhizoma Cyperi with water or ethanol water solution, soaking and extracting, and filtering to obtain extractive solution;
b) concentrating the extracting solution obtained in the step a) to obtain an extract;
c) mixing the extract obtained in the step b) with auxiliary materials, carrying out wet granulation, and tabletting the obtained granules to obtain tablets; the auxiliary materials are the same as the auxiliary materials of the granules, and the details are not repeated here.
In the invention, honeysuckle, perilla, rhizoma ligustici wallichii, cortex lycii radicis and rhizoma cyperi are mixed with water or ethanol water solution for reflux extraction, and the extract is obtained by filtration; the volume ratio of the total mass of the honeysuckle, the perilla, the ligusticum wallichii, the cortex lycii radicis and the rhizoma cyperi to the extraction solvent is 1g (10-30) mL, preferably 1g (15-25) mL, and more preferably 1g (16 mL); the volume concentration of the ethanol water solution is 30-80%, preferably 40-75%, and more preferably 70%. In the invention, the extraction temperature is preferably 80-100 ℃, more preferably 85-95 ℃, and most preferably 90 ℃; the extraction frequency is preferably 1-3 times, and more preferably 2 times; the extraction time is preferably 90-240 min, and more preferably 150 min; the filtration mode is preferably plate-and-frame filter pressing.
After the extracting solution is obtained, concentrating the extracting solution to obtain an extract; the concentration mode is preferably reduced pressure concentration; the pressure of the reduced pressure concentration is preferably-80 kPa to-40 kPa, and more preferably-60 kPa; the relative density of the extract obtained by the reduced pressure concentration is preferably 1.1-1.3, and more preferably 1.15.
After the extract is obtained, the auxiliary materials and the extract are mixed, wet granulation is carried out, and the obtained granules are tabletted to obtain the chewable tablet. In the present invention, the wet granulation method is the same as the above-mentioned method for preparing granular health food, and is not described herein again.
In the invention, when the dosage form of the health food is a tablet, the dosage of the tablet is preferably 4-12 g/day, and more preferably 8 g/day.
The invention provides a preparation method of powder of health food in the scheme, which comprises the following steps:
A) mixing flos Lonicerae, Perillae herba, rhizoma Ligustici Chuanxiong, cortex Lycii and rhizoma Cyperi with water or ethanol water solution, soaking and extracting, and filtering to obtain extractive solution;
B) centrifuging the extracting solution in the step A), and taking a supernatant to obtain a refined solution;
C) carrying out spray drying on the refined solution in the step B), and mixing with auxiliary materials to obtain powder; the auxiliary material is a food auxiliary material, preferably xylitol and/or L-arabinose, and more preferably xylitol.
In the invention, honeysuckle, perilla, rhizoma ligustici wallichii, cortex lycii radicis and rhizoma cyperi are mixed with water or ethanol water solution for soaking and extraction, and the extract is obtained by filtering; the mass-volume ratio of the honeysuckle, the perilla, the ligusticum wallichii, the cortex lycii radicis and the rhizoma cyperi to the water or ethanol water solution is 1g (10-30) mL, preferably 1g (15-25) mL, and more preferably 1g (16 mL); the volume concentration of the ethanol water solution is 30-80%, preferably 40-75%, and more preferably 70%. In the invention, the extraction temperature is preferably 80-100 ℃, more preferably 85-95 ℃, and most preferably 90 ℃; the extraction frequency is preferably 1-3 times, and more preferably 2 times; the extraction time is preferably 50-120 min, and more preferably 60-90 min; the filtration mode is preferably plate-and-frame filter pressing.
After the extracting solution is obtained, centrifuging the extracting solution, and taking supernate to obtain refined solution; the centrifuging device is preferably an ultracentrifuge; the centrifugation time is preferably 20-40 min, and more preferably 30 min; the rotation speed of the centrifugation is preferably 8000-12000 r/min, and more preferably 10000 r/min.
After the refined liquid is obtained, spray drying is carried out on the refined liquid to obtain a dried product, and the dried product is mixed with auxiliary materials to obtain powder; the temperature of the spray drying is preferably 100-140 ℃, and more preferably 120 ℃; the mass ratio of the dried product to the auxiliary material is preferably 100: 7.
In the invention, when the dosage form of the health food is powder, the dosage of the powder is preferably 2-6 g/day, and more preferably 4 g/day.
The following examples are provided to describe the preparation method of the antioxidant Chinese medicinal composition and health food thereof in detail, but they should not be construed as limiting the scope of the present invention.
Example 1
Weighing 3kg of honeysuckle, 3kg of perilla, 3kg of ligusticum wallichii, 7kg of cortex lycii radicis and 6kg of rhizoma cyperi, adding 330L of water, decocting and extracting for 120min, filtering, mixing filter residues with 330L of water, and decocting for 90min again. Mixing the two filtrates to obtain extractive solution. Centrifuging the extractive solution with ultracentrifuge at 7500r/min for 30 min. Concentrating the supernatant under reduced pressure of-60 kPa to obtain extract with relative density of 1.2.
Mixing the extract with 4.4kg dextrin, and performing primary drying at 90 deg.C. Pulverizing, sieving with 100 mesh sieve, making soft mass with 10 vol% starch slurry as binder, extruding, sieving with 14 mesh sieve, granulating, drying at 80 deg.C, sieving with 16 mesh sieve, and grading to obtain granule.
Example 2
Weighing 45kg of honeysuckle, 1kg of perilla, 1kg of ligusticum wallichii, 1kg of cortex lycii radicis and 1kg of rhizoma cyperi, mixing with 245L of 70 volume percent ethanol water solution, carrying out reflux extraction at 85 ℃ for 60min, filtering, mixing filter residue with 245L of 70 volume percent ethanol water solution, and carrying out reflux extraction again at 80 ℃ for 120 min. Mixing the two filtrates to obtain extractive solution. Concentrating the extractive solution under reduced pressure of-40 kPa to obtain extract with relative density of 1.15. Mixing the extract with 6kg of dextrin, 3kg of lactose and 0.2kg of sodium cyclamate, and performing primary drying at 90 ℃. Pulverizing, sieving with 100 mesh sieve, making into soft material with 50% ethanol water solution as wetting agent, squeezing, sieving with 14 mesh sieve, granulating, drying at 60 deg.C, sieving with 16 mesh sieve, grading, and tabletting.
Example 3
Weighing 50kg of honeysuckle, 20kg of perilla, 10kg of ligusticum wallichii, 30kg of cortex lycii radicis and 20kg of rhizoma cyperi, adding 1300L of water, decocting and extracting for 90min, filtering, adding 1040L of water into filter residues, and decocting and extracting for 60 min. Mixing the two filtrates to obtain extractive solution. Centrifuging the extractive solution with ultracentrifuge at 10000r/min for 30 min. Collecting supernatant as refined solution. Spray drying the refined solution at 120 deg.C, and adding xylitol 7 wt% of the dried product to obtain powder.
Example 4 Each index of the granules prepared in example 1 was measured
The fluidity, the particle size distribution and the water content of the granules are measured according to the relevant requirements of Chinese pharmacopoeia. The measurement results of various indexes of the granules are shown in table 1; the results of the quality inspection of the granules of the present invention are shown in Table 2.

As can be seen from the results in table 2, the quality of the granules prepared in example 1 meets the requirements of "chinese pharmacopoeia".
Example 5 study of antioxidant function of the granules of example 1
80 male Kunming mice with the weight range of 18-22 g are selected and provided by the experimental animal center of Yanbian university. The test animals are adaptively fed for one week before the experiment, and the next experiment is carried out after the physical signs are well shown.
Establishing a D-galactose oxidative damage mouse model: injecting D-galactose 120 mg/kg BW into abdominal cavity of 80 mice to form an oxidative damage model, wherein the injection amount is 0.01mL/g for 1 time per day, observing hair color change and action characteristics of the mice, and continuously administering for 6 weeks. Modeling principle: the injection of D-galactose in large amount for a long time can generate excessive active oxygen and increase the content of free radicals, thereby generating a large amount of oxidation products, causing the reduction of the functions of organelles in a body, and generating various diseases.
Randomly grouping the modeled mice into 3 tested dose groups and 1 model control group based on daily dose of human, and intragastrically administering the granules prepared in example 1 with different concentrations in 3 tested dose groups of a low dose group, a medium dose group and a high dose group, wherein the dosages are 0.24g/kg BW, 0.48g/kg BW and 0.96g/kg BW respectively, and the administration times are 1 time/day and are respectively equal to 5, 10 and 20 times of the dosage taken by human; the model control group was intragastrically filled with the same volume of distilled water. D-galactose was continuously injected into the abdominal cavity of the mice while the gavage administration was performed. After the experiment, the animals were sacrificed and the eyeball bled. The mice were weighed at 0d, 10d, 20d, 30d after administration and the MDA content, SOD activity, protein carbonyl content, and GSH content were determined.
(1) The results of the effect of the samples on the body weight of the mice are shown in table 3 and fig. 1. The results show that: the weight of the mice is continuously increased after the administration for 0-30 d, and the weight of the mice after the administration for 0, 10, 20 and 30d has no significant difference compared with the model control group in each administration group, which indicates that the weight of the mice is not affected by the test sample.

(2) The results of the effect of the samples on the MDA content in the serum are shown in table 4, fig. 2, showing: compared with the MC group, the MDA content of the LG, the MG and the HG group is reduced, and the obvious difference (p is less than 0.05) is provided, which indicates that the tested sample can obviously reduce the MDA content in vivo and reduce the peroxidation damage after the mice are subjected to the oxidative damage. The result shows that the medicine has the function of reducing lipid peroxidation, and the result of the index of the experiment item is positive.

(3) The results of the effect of the samples on the content of protein carbonyl in serum are shown in table 5 and fig. 3, and the results show that: compared with the MC group, the protein carbonyl content of the LG, the MG and the HG group is obviously reduced and has significant difference (p is less than 0.05), which indicates that the granules prepared in example 1 have the capability of repairing the oxidative damage of the protein. The result of the index experiment is positive.

(4) The results of the effect of the samples on the activity of SOD in serum are shown in Table 6 and FIG. 4, and the results show that: compared with the MC group, the SOD activities of the LG, the MG and the HG group are increased, and the determination result has significance (p is less than 0.05). Along with the increase of the dosage, the SOD activity is increased, which shows that the tested sample can improve the activity of the antioxidant enzyme, and the result of the experimental index is positive.

(5) The results of the effect of the samples on the serum GSH content are shown in table 7, fig. 5, and show: compared with the group of LG, MG and HG, the content of GSH is reduced, and the determination result has significance (p is less than 0.05). With increasing dose, GSH content of three groups, LM, MG, HG, varied significantly. It was shown that the GSH content in vivo was significantly increased in mice given the granules by gavage compared to the model control group, with the highest increase in the high dose group. The test sample is shown to have the function of increasing antioxidant substances, and the result of the experimental index is negative.

From the above, the results of the four indexes of MDA content, SOD activity, protein carbonyl content, and GSH content all show positive, and it can be determined that the antioxidant animal experiment result of the granule prepared in example 1 is positive.
From the above embodiments, it can be seen that the present invention provides a Chinese medicinal composition with an antioxidant function and a method for preparing a health food thereof, wherein granules of the health food have the effects of reducing the content of protein carbonyl and the content of MDA, and increasing the anti-SOD activity and the content of GSH, and the results show that the health food is helpful for mice to repair peroxidation damage caused by D-galactose. According to the evaluation and judgment standard of the health food function, the traditional Chinese medicine composition and the health food thereof can be determined to have the antioxidant function.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.

Claims (9)

1. A traditional Chinese medicine composition with an antioxidant function is composed of the following raw materials in parts by weight: 3 parts of honeysuckle, 3 parts of purple perilla, 3 parts of ligusticum wallichii, 7 parts of cortex lycii radicis and 6 parts of rhizoma cyperi.
2. An antioxidant health food comprising the Chinese medicinal composition of claim 1.
3. The antioxidant health food as claimed in claim 2, wherein the health food is in the form of powder, granule or tablet.
4. The method for preparing granules in health food according to claim 3, comprising the steps of:
1) Mixing honeysuckle, perilla, ligusticum wallichii, cortex lycii radicis and rhizoma cyperi with an extraction solvent, soaking and extracting, and filtering to obtain an extracting solution, wherein the extraction solvent is water or an ethanol water solution;
2) Concentrating the extracting solution obtained in the step 1) to obtain an extract;
3) Mixing the extract obtained in the step 2) with auxiliary materials, and performing wet granulation to obtain granules, wherein the auxiliary materials are food auxiliary materials.
5. The preparation method of the granules according to claim 4, wherein the volume ratio of the total mass of the honeysuckle flower, the perilla, the ligusticum wallichii, the cortex lycii radicis and the rhizoma cyperi in the step 1) to the extraction solvent is 1g (10-30) mL.
6. The preparation method of the granules according to claim 4 or 5, wherein the volume concentration of the ethanol aqueous solution in the step 1) is 30-80%.
7. The preparation method of the granules according to claim 6, wherein the auxiliary materials in the step 3) comprise a filler and a flavoring agent.
8. A method for preparing a powder for health food according to claim 3, comprising the steps of:
A) mixing honeysuckle, perilla, ligusticum wallichii, cortex lycii radicis and rhizoma cyperi with an extraction solvent, soaking and extracting, and filtering to obtain an extracting solution, wherein the extraction solvent is water or an ethanol water solution;
B) centrifuging the extracting solution in the step A), and taking a supernatant to obtain a refined solution;
C) and B), after spray drying the refined liquid in the step B), mixing the refined liquid with auxiliary materials to obtain powder, wherein the auxiliary materials are food auxiliary materials.
9. The method for preparing a powder according to claim 8, wherein: the auxiliary material is xylitol and/or L-arabinose.
