Modern pharmacological experimental research shows that the lily extract and the effective components thereof have various pharmacological activities, including cough relieving, phlegm eliminating, tranquilizing, hypnosis, immunoregulation, anti-tumor, anti-oxidation, anti-inflammation, anti-hypoxic stress injury, anti-depression, blood sugar reducing, bacteriostasis and the like.
Dendrobium officinale is a dried stem of an orchid plant, is a rare traditional Chinese medicine, is listed as the head of the nine-large Chinese mesona in the Collection, is well known as the gold in the medicine, and has the effects of promoting the production of body fluid, tonifying stomach, nourishing yin, clearing heat, prolonging life and enhancing immunity. The book records dendrobium nobile in Shen nong Ben Cao Jing: "sweet and mild". Mainly injure the middle-jiao, remove arthralgia, descend qi, tonify five internal organs, strengthen yin and make the stomach and intestine weak, and can be taken for a long time. The dendrobium nobile lindl has the functions of nourishing skin, reducing blood sugar, resisting tumor, protecting liver, benefiting gallbladder, tonifying five internal organs, strengthening body, nourishing yin, and enhancing the functions of spleen and stomach after being taken for a long time, thereby enabling the human body to be flexible, enhancing the immunity and prolonging the life.
The effect of improving sleep of the product is verified through experiments;
preparing a product for experiment;
the needed raw materials of the wild jujube seed extract, the angelica extract, the lily extract and the dendrobium officinale extract can be purchased from commercial sources.
The above raw material extract can be prepared by the following method.
1. Preparation of wild jujube seed extract
Extracting semen Ziziphi Spinosae material selected by quality standard test with 8 times of 70% ethanol under reflux at 70 deg.C for two times, each for 1.5 hr, and filtering; mixing filtrates, concentrating under reduced pressure at 60 deg.C and 0.09MPa for 1 hr, and concentrating to obtain soft extract; then freeze-drying in vacuum to obtain dry extract, wherein the sample temperature is-24 ℃, the cold hydrazine temperature is-96 ℃, the pressure is 1Pa, and the time is 24 h; the dry extract is crushed and sieved by a 60-mesh sieve to obtain extract powder for later use.
2. Preparation of Angelica sinensis extract
Extracting radix Angelicae sinensis with 6 times of water at 90 deg.C twice for 2 hr, and filtering; mixing filtrates, concentrating under reduced pressure at 80 deg.C and 0.09MPa for 3 hr, and concentrating to obtain soft extract; then freeze-drying in vacuum to obtain dry extract, wherein the sample temperature is-24 ℃, the cold hydrazine temperature is-96 ℃, the pressure is 1Pa, and the time is 24 h; the dry extract is crushed and sieved by a 60-mesh sieve to obtain extract powder for later use.
3. Preparation of Lily extract
Extracting radix Angelicae sinensis with 7.5 times of 80% ethanol at 60 deg.C for 3 hr twice, and filtering; mixing filtrates, concentrating under reduced pressure at 60 deg.C and 0.09MPa for 1 hr, and concentrating to obtain soft extract; then freeze-drying in vacuum to obtain dry extract, wherein the sample temperature is-24 ℃, the cold hydrazine temperature is-96 ℃, the pressure is 1Pa, and the time is 24 h; the dry extract is crushed and sieved by a 60-mesh sieve to obtain extract powder for later use.
4. Preparing dendrobium officinale extract
Extracting radix Angelicae sinensis with 80 times of water at 90 deg.C for two times (each for 1 hr), and filtering; mixing filtrates, concentrating under reduced pressure at 80 deg.C and 0.09MPa for 2 hr, and concentrating to obtain soft extract; then freeze-drying in vacuum to obtain dry extract, wherein the sample temperature is-24 ℃, the cold hydrazine temperature is-96 ℃, the pressure is 1Pa, and the time is 24 h; the dry extract is crushed and sieved by a 60-mesh sieve to obtain extract powder for later use.
5. Mixing the obtained mixed spina date seed extract, the angelica sinensis extract, the lily extract and the dendrobium officinale extract according to nine different formulas obtained by orthogonal experiments;
the nine formulations are as follows:
example 1: 10g of spina date seed, 6g of angelica, 6g of lily and 6g of dendrobium officinale
Example 2: 12.5g of spina date seed, 9g of angelica, 9g of lily and 6g of dendrobium officinale
Example 3: 15g of spina date seed, 12g of angelica, 12g of lily and 6g of dendrobium officinale
Example 4: 15g of spina date seed, 6g of angelica, 9g of lily and 9g of dendrobium officinale
Example 5: 10g of spina date seed, 9g of angelica, 12g of lily and 9g of dendrobium officinale
Example 6: 12.5g of spina date seed, 12g of angelica, 6g of lily and 9g of dendrobium officinale
Example 7: 12.5g of spina date seed, 6g of angelica, 12g of lily and 12g of dendrobium officinale
Example 8: 15g of spina date seed, 9g of angelica, 6g of lily and 12g of dendrobium officinale
Example 9: 10g of spina date seed, 12g of angelica, 9g of lily and 12g of dendrobium officinale
And (3) verification experiment: functional evaluation for sleep improvement of the present invention
1 materials and methods
1.1 sample: the product obtained by mixing the formulas of the above 9 examples
1.2 Experimental animals: the clean-grade ICR mice are female, have the weight of 18-22 g, and are provided by Shanghai Jihui laboratory animal feeding company Limited, and the total number of the mice is 100.
1.3 dose selection: in this experiment, 100 animals were randomly divided by body weight into 10 experimental groups, one negative control group (distilled water) and 9 example groups, each of which was 10 animals. The samples were weighed daily for one week after acclimatization, and the samples were gavaged for 30 days at 10-fold dosage in human body according to "health food testing and evaluation specifications" (2003 edition) and orally at 2.0mL/100 gBW.
1.4 Main reagents: pentobarbital sodium (analytically pure, Shanghai Banyan biotechnology Limited)
1.5 test methods:
1.5.1 direct sleep test: after the stomach is not irrigated again, whether the sleep phenomenon appears is observed, and the sleep takes the disappearance of the righting reflex as an index. When the mouse is placed in the back lying position, the mouse can turn over to the right position immediately, if the mouse cannot turn over for more than 60s, the turning over reflection is considered to disappear, and the mouse enters sleep. The recovery of the righting reflex is the awakening of the animal, the period from disappearance of the righting reflex to recovery is the sleeping time of the animal, and the number of sleeping animals and the sleeping time of the control group and each dosage group are recorded.
1.5.2 test for prolonging the sleep time of sodium pentobarbital: after the last gastric lavage for 15min, 50 mg/kg is injected into the abdominal cavity of each group of animals-1bw the injection amount of the sodium pentobarbital is 0.2mL/20gbw, and whether the test sample prolongs the sleep time of the sodium pentobarbital is observed by taking the disappearance of the righting reflex for more than 1 minute as a sleep-falling judgment standard.
1.5.3 sub-threshold dose hypnotic test of sodium pentobarbital: injecting 40 mg/kg into abdominal cavity of each group of animals after last intragastric administration for 15min-1The injection amount of bw sodium pentobarbital is 0.2mL/20gbw, the number of animals falling asleep within 30 minutes is recorded by taking the disappearance of righting reflex for more than 1 minute as the standard for judging falling asleep.
1.5.4 pentobarbital sodium sleep latency test: injecting 40 mg/kg into abdominal cavity of each group of animals after last intragastric administration for 15min-1The injection amount of bw sodium pentobarbital is 0.2mL/20gbw, the influence of the sleep latency of the sample sodium pentobarbital is observed by taking the disappearance of righting reflex for more than 1 minute as the sleep onset judgment standard.
2 results of the test
2.1 the effect of the test substances on sleep time results are shown in Table 1;

As can be seen from Table 1, no direct sleep was observed in any of the groups. In the test for the effect of prolonging the sleep time of sodium pentobarbital, the animals in groups 1, 2, 3, 4 and 6 had an increased sleep time compared to the control group, and the difference was statistically significant (P < 0.05). In the test of the effect on the sleep incidence of the subthreshold dose of sodium pentobarbital, the difference in the sleep incidence of the animals in groups 1, 2, 3, 4, 5 and 6 was statistically significant (P <0.05) compared to the control group. In the test of the effect on sleep latency of sodium pentobarbital, the animals in groups 1, 2, 3, 4 and 6 had a shorter time to sleep latency compared to the control group, and the difference was statistically significant (P < 0.05). The health care product prepared in example 1 of the present application is suggested to have the effect of improving the sleep function of animals according to the judgment standard of "health food inspection and evaluation technical specification" (2003 edition).
And (4) analyzing results: from the above results, the optimal ratio of the active components of the invention is: 10g of spina date seed, 6g of angelica sinensis, 6g of lily and 6g of dendrobium officinale.
Experiment II, a formula component experiment proves that the four components have a synergistic effect;

And (4) analyzing results: according to the results, the optimal formula of the product for improving sleep is the formula of the wild jujube seed, the Chinese angelica, the lily and the dendrobium officinale, and the four medicinal materials have synergistic effects and cannot be used.
The invention adopts a plurality of medicinal and edible Chinese herbal medicines to extract effective components, combines the characteristics of the traditional Chinese medicine with the research of modern medicine, utilizes the different efficacies of the four components to generate synergistic action, and forms a compound product with the functions of nourishing yin, promoting the secretion of saliva or body fluid, and calming the heart and tranquilizing the mind.

Claims (6)

The product for improving sleep is characterized by comprising the following raw materials in parts by mass: 10 parts of spina date seeds, 6 parts of Chinese angelica, 6 parts of lily and 6 parts of dendrobium officinale; the wild jujube seed is a wild jujube seed extract, wherein the wild jujube seed extract is prepared by heating, refluxing and extracting wild jujube seed raw materials by using 70% ethanol in an amount which is 8 times that of the wild jujube seed raw materials, filtering and concentrating the extract for later use; the angelica is an angelica extract which is prepared by taking 6 times of angelica raw material and carrying out water extraction, filtration and concentration for later use; the lily is a lily extract, and the lily extract is obtained by extracting a lily raw material with 7.5 times of 80% ethanol, filtering and concentrating for later use; the dendrobium officinale is a dendrobium officinale extract, and the dendrobium officinale extract is prepared by taking a dendrobium officinale raw material, soaking and extracting the dendrobium officinale raw material with 80 times of water, filtering and concentrating for later use.
2. The method for preparing a product for improving sleep as claimed in claim 1, comprising the following steps:
preparing materials according to a raw material formula; the formula comprises the following raw materials in parts by mass: 10 parts of spina date seeds, 6 parts of Chinese angelica, 6 parts of lily and 6 parts of dendrobium officinale;
preparing the wild jujube seed extract, extracting,
heating and refluxing semen Ziziphi Spinosae raw material with 8 times of 70% ethanol, filtering, and concentrating to obtain semen Ziziphi Spinosae extract;
preparing an extract of Chinese angelica root,
soaking radix Angelicae sinensis in 6 times of water, extracting, filtering, and concentrating to obtain radix Angelicae sinensis extract;
preparing a lily extract, extracting the lily with water,
extracting Bulbus Lilii raw material with 7.5 times of 80% ethanol, filtering, and concentrating to obtain Bulbus Lilii extract;
preparing the dendrobium officinale extract, namely preparing the dendrobium officinale extract,
taking a dendrobium officinale raw material, soaking and extracting the dendrobium officinale raw material with 80 times of water, filtering and concentrating for later use to obtain a dendrobium officinale extract;
mixing the extracts to obtain the final product.

the specific process for preparing the spina date seed extract comprises the following steps:

the specific process for preparing the angelica extract comprises the following steps:
extracting radix Angelicae sinensis with 6 times of water at 90 deg.C for two times (2 hr each time), and filtering; mixing filtrates, concentrating under reduced pressure at 80 deg.C and 0.09MPa for 3 hr, and concentrating to obtain soft extract; then freeze-drying in vacuum to obtain dry extract, wherein the sample temperature is-24 ℃, the cold hydrazine temperature is-96 ℃, the pressure is 1Pa, and the time is 24 h; pulverizing the dry extract, and sieving with 60 mesh sieve to obtain extract powder.
