背景技术

单环刺螠,俗名:海肠、海肠子,属于螠虫动物门、螠纲、无管螠目、刺螠科、刺螠属,是一种营养价值极高的底栖、无脊椎海洋生物。单环刺螠体内含有大量人体所需要的氨基酸、微量元素和维生素,尤其富含纤维蛋白溶解酶、纤溶酶原激活物、胶原酶等三种酶系,是预防高血压、动脉硬化和消除疲劳的有效保健食品,单环刺螠体内所含的单环刺螠激酶具有治疗脑血栓、心肌梗塞等血栓性疾病的功能,所含有大量的不饱和脂肪酸具有增强免疫力、提高记忆力的功效,还具有抗血栓、防止动脉硬化的作用;此外含有丰富的羟氨酸,具有美容养颜、抗衰老的作用。目前,单环刺螠开发的产品报道很少,因此开展单环刺螠资源的综合利用,开发安全、营养、保健的单环刺螠产品,对于提高平民百姓的生活质量和保健水平,具有重要意义。
申请号为200610098354.X的中国专利公开了一种防治心脑血管疾病的中药组合物及其制备方法,但该发明为一种中药,药效较强,不适合平常保健食用,而且口感较差。
申请号为201410215610.3的中国专利公开了一种单环刺螠内脏糖胺聚糖的制备方法及其应用,涉及了该成分可进一步加工成预防和调节血脂的食品、保健品及药品,但是该制备方法所得成分单一,营养不均衡,口感不佳。
发明内容
为了解决上述技术问题,本发明提供了一种单环刺螠溶血栓功能保健饮料及其制备方法,配方合理,营养均衡,口感好,作为饮料长期饮用,具有溶血栓、降血压、提高免疫力的作用。
本发明的具体技术方案为:每1000重量份该饮料由以下重量份的原料制成:400~650份单环刺螠提取液,50-100份中草药提取液,6~12份调配料,0.5~0.8份复合维生素,余量为水。
单环刺螠体内所含的单环刺螠活性成分具有治疗脑血栓、心肌梗塞等血栓性疾病的功能,所含有大量的不饱和脂肪酸具有增强免疫力、提高记忆力的功效,还具有抗血栓、防止动脉硬化的作用;此外含有丰富的羟氨酸,具有美容养颜、抗衰老的作用,还具有抗癌、防癌、免疫及防辐射功效。
作为优选,所述中草药提取液由以下重量份的中草药原料制成:11~17份党参,18~26份马齿苋,16~23份枸杞,12~20份绞股蓝,12~18份菟丝子,8~14份仙茅,6~12份远志,6~12份杜仲和5~10份红景天。
该中草药提取液具有降血压、清热利湿、益精血、强筋骨、提高免疫力等功效,与单环刺螠活性成分辨证配伍,在作用上相互佐助复配,能够提高单环刺螠活性成分的溶血栓效果,同时其药效温和,不易产生其他副作用,适合长期饮用。

3)灭酶:步骤2)酶解结束后,灭酶,过150~250目筛,冷却至室温后,得单环刺螠酶解提取液。
采用3中蛋白酶相结合酶解,大大增加了蛋白质的水解率,能够促进单环刺螠活性成分的析出,提高其生物利用率,充分发挥保健品的保健营养价值。

3)精滤:将均质后的饮料先经250~300目双联过滤器过滤,再经硅藻过滤机精滤;
4)杀菌:调配均质过滤的单环刺螠保健饮料进行高温灭菌,灭菌温度115~121℃,时间为15~20 min;
5)无菌灌装封口:灭菌后单环刺螠保健饮料在无菌灌装生产线中灌装封口。
均质使各种营养成分均匀化,防止产品出现分层沉淀,使产品稳定性得到进一步的保障。
与现有技术对比,本发明的有益效果是:本发明将单环刺螠提取液、中草药提取液、调配料和复合维生素一起做成保健饮料,产品绿色健康新颖,而且营养均衡丰富,色香味俱全,食用方便,保健功效显著,符合营养保健的健康理念;制备方法简单实用,科学合理,既能保证饮料的口感,又能保持原有营养保健成分不流失,适合工厂化生产应用;同时,单环刺螠资源还未综合利用,产品附加值较高;因此本发明提供了一种配方合理、营养均衡、无异腥味的具有良好保健效果的单环刺螠溶血栓功能保健饮料。
具体实施方式
下面结合实施例对本发明作进一步的描述。
实施例1
将鲜活单环刺螠放入淡水吐沙后,将内脏翻出,用清水洗干净,再置于组织捣碎机中,加入6倍单环刺螠原料重量的水,在2℃下,破碎匀浆20min,制得单环刺螠匀浆液;将所得单环刺螠匀浆液,按3g/L、3g/L、1.5g/L比例分别添加食品级菠萝蛋白酶、中性蛋白酶、风味蛋白酶,调节匀浆液的pH值8.2,酶解温度为40℃,酶解时间200min;酶解结束后,酶解单环刺螠匀浆液加热到93℃,灭酶5min,过250目筛,冷却至室温后,得单环刺螠酶解提取液,置于2℃下备用。
将单环刺螠提取液、中草药提取液、调配料、复合维生素按比例混合搅拌均匀;将调配好的饮料放置在均质机中,在均质压力30MPa,温度50℃下均质,将均质后的饮料先经300目双联过滤器过滤,再经硅藻过滤机精滤以除去其中的杂质及不溶物;调配均质过滤的单环刺螠保健饮料进行高温灭菌,灭菌温度为121℃,时间为15min;灭菌后单环刺螠保健饮料在无菌灌装生产线中灌装封口。
实施例2
将鲜活单环刺螠放入淡水吐沙后,将内脏翻出,用清水洗干净,再置于组织捣碎机中,加入3倍单环刺螠原料重量的水,在0℃下,破碎匀浆30min,制得单环刺螠匀浆液;将所得单环刺螠匀浆液,按2.5g/L、2.5g/L、2.5 g/L比例分别添加食品级菠萝蛋白酶、中性蛋白酶、风味蛋白酶,调节匀浆液的pH值7.0,酶解温度为35℃,酶解时间240min;酶解结束后,酶解单环刺螠匀浆液加热到90℃,灭酶6min,过150目筛,冷却至室温后,得单环刺螠酶解提取液,置于0℃下备用。
将单环刺螠提取液、中草药提取液、调配料、复合维生素按比例混合搅拌均匀;将调配好的饮料放置在均质机中,在均质压力20MPa,温度60℃下均质,将均质后的饮料先经250目双联过滤器过滤,再经硅藻过滤机精滤以除去其中的杂质及不溶物;调配均质过滤的单环刺螠保健饮料进行高温灭菌,灭菌温度为115℃,时间为20min;灭菌后单环刺螠保健饮料在无菌灌装生产线中灌装封口。
实施例3
将鲜活单环刺螠放入淡水吐沙后,将内脏翻出,用清水洗干净,再置于组织捣碎机中,加入5倍单环刺螠原料重量的水,在4℃下,破碎匀浆10min,制得单环刺螠匀浆液;将所得单环刺螠匀浆液,按3.2g/L、2.0g/L、2g/L比例分别添加食品级菠萝蛋白酶、中性蛋白酶、风味蛋白酶,调节匀浆液的pH值7.5,酶解温度为50℃,酶解时间120min;酶解结束后,酶解单环刺螠匀浆液加热到95℃,灭酶3min,过200目筛,冷却至室温后,得单环刺螠酶解提取液,置于4℃下备用。
将单环刺螠提取液、中草药提取液、调配料、复合维生素按比例混合搅拌均匀;将调配好的饮料放置在均质机中,在均质压力25MPa,温度55℃下均质,将均质后的饮料先经300目双联过滤器过滤,再经硅藻过滤机精滤以除去其中的杂质及不溶物;调配均质过滤的单环刺螠保健饮料进行高温灭菌,灭菌温度为118℃,时间为18min;灭菌后单环刺螠保健饮料在无菌灌装生产线中灌装封口。
本发明实施例1-3的产品外观为清透的棕红色液体,味甜,微酸。通过动物试验进行毒理学试验及安全性评价。小鼠急性经口毒性试验结果表明,本保健饮料对小鼠急性经口最大耐受剂量大于高剂量组88mg/kg·bw;遗传毒性试验小鼠骨髓细胞微核试验和小鼠精子畸形试验两项试验结果均为阴性,表明该保健饮料无致突变作用;由大鼠30d喂养试验可知,大鼠体征行为正常、体态健康、血液及血液生化指标也无异常变化,因此88mg/kg·bw的剂量(人体推荐量的176倍)属于安全剂量;遗传毒性试验和大鼠30d喂养试验结果均为阴性,因此确定本保健饮料属无毒类食品,人可以安全放心食用。

Claims (10)

1.一种单环刺螠溶血栓功能保健饮料,其特征在于,每1000重量份该饮料由以下重量份的原料制成:400~650份单环刺螠提取液,50-100份中草药提取液,6~12份调配料,0.5~0.8份复合维生素,余量为水。
2.如权利要求1所述的单环刺螠溶血栓功能保健饮料,其特征在于,所述中草药提取液由以下重量份的中草药原料制成:11~17份党参,18~26份马齿苋,16~23份枸杞,12~20份绞股蓝,12~18份菟丝子,8~14份仙茅,6~12份远志,6~12份杜仲和5~10份红景天。
3.如权利要求1所述的单环刺螠溶血栓功能保健饮料,其特征在于,所述调配料各组分的质量百分含量为:13~18%甜叶菊糖,12~16%香菇多糖,10~15%低聚木糖,12~16%甘露糖醇,5~10%柠檬酸,4~8%乳酸钙,余量为蜂蜜。
4.如权利要求1所述的单环刺螠溶血栓功能保健饮料,其特征在于,所述复合维生素每1000 g由下列重量的各组分混合而成:6~10g维生素A,10~16g维生素D,32~48g维生素E,4~7g维生素K3,3~8g维生素B1,15~20g维生素B2,0.5~1.5g维生素B6,1.0~1.5g维生素B12,180~245g维生素C,90~125g肌醇,1~1.5g生物素,2.0~3.0g叶酸,28~36g烟酸,余量为葡萄糖。
5.如权利要求1所述的单环刺螠溶血栓功能保健饮料,其特征在于,所述单环刺螠提取液的制备方法为:
1)将鲜活单环刺螠放入淡水吐沙后,将内脏翻出,用清水洗干净,再置于组织捣碎机中,加入3~6倍单环刺螠原料重量的水,破碎匀浆,制得单环刺螠匀浆液;
2)酶解:将步骤1)所得单环刺螠匀浆液,按2.5~3.2g/L、2.0~3.0g/L、1.5~2.5g/L比例分别添加食品级菠萝蛋白酶、中性蛋白酶、风味蛋白酶,酶解;
3)灭酶:步骤2)酶解结束后,灭酶,过150~250目筛,冷却至室温后,得单环刺螠酶解提取液。
6.如权利要求5所述的单环刺螠溶血栓功能保健饮料,其特征在于,所述单环刺螠提取液的制备方法步骤1)中破碎匀浆时间为10~30min,温度为0~4℃。
7.如权利要求5所述的单环刺螠溶血栓功能保健饮料,其特征在于,所述单环刺螠提取液的制备方法步骤2)中酶解温度为35~50℃,酶解时间为90~180min,酶解pH值为7.0~8.2。
8.如权利要求5所述的单环刺螠溶血栓功能保健饮料,其特征在于,所述单环刺螠提取液的制备方法步骤3)中灭酶温度90~95℃,灭酶时间3~6min。
9.如权利要求5所述的单环刺螠溶血栓功能保健饮料,其特征在于,所述单环刺螠提取液的制备方法步骤3)中单环刺螠提取液置于0~4℃下保存。
10.一种权利要求1所述单环刺螠溶血栓功能保健饮料的制备方法,其特征在于,该方法包含以下步骤:
1)调配:将单环刺螠提取液、中草药提取液、调配料、复合维生素按比例混合搅拌均匀;
2)均质:将调配好的饮料放置在均质机中,在均质压力20~30MPa,温度50~60℃下均质;
3)精滤:将均质后的饮料先经250~300目双联过滤器过滤,再经硅藻过滤机精滤;
4)杀菌:调配均质过滤的单环刺螠保健饮料进行高温灭菌,灭菌温度115~121 ℃,时间为15~20 min;
5)无菌灌装封口:灭菌后单环刺螠保健饮料在无菌灌装生产线中灌装封口。
