Abstract
本发明公开了一种治疗乳腺炎的中药组合物。由如下重量份数的原料制成:芍药8‑12份,附子15‑30份,瓜蒌12‑18份,茜草根3‑8份,密叶黑藓3‑8份,牛膝3‑8份。本发明制备的治疗乳腺炎的中药组合物,能有效治疗乳腺炎,治愈率高,复发率低,副作用小,芍药,附子,瓜蒌作为君药,药效高,茜草根作为臣药,起到增强药效的作用,密叶黑藓作品为佐药,起到减毒,减小副作用的功效,牛膝作为使用,起到调和作用。本发明的中药组合物制备方法简单,原料来源广,适合大规模工业化生产。

技术领域
本发明属于妇科病中药制备技术领域,具体涉及一种治疗乳腺炎的中药组合物及其制备方法。
背景技术
乳腺炎,是由于细菌入侵乳房而引起乳腺结缔组织和导管内发生化脓感染的一种炎症性疾病。乳腺炎多见于初产妇,因为初产妇的乳腺导管欠通畅,而且缺乏哺乳经验,故易患乳腺炎。乳腺炎在哺乳期、怀孕期、非哺乳期、非怀孕期都有发生,但是最多见于产后未满月的哺乳妇女性。
随着医学的不断发展,对乳腺炎病因病机的认识不断深化。总结对乳腺炎的认识的发展变化,其发病原因主要集中在乳汁游积、肝郁胃热、感受外邪三方面。根据乳腺炎不同的临床表现,在中医望闻问切四诊基础上对乳腺炎进行辨证分型,临床上主要分为三型;气滞热塞型、热毒识盛型,正虚邪恋型,分别多见于郁滞期、成胳期、溃后期。
中药治疗乳腺炎,有多种配方,如林毅教授强调乳腺炎以消为贵,以通为用,治以瓜萎牛蒡汤合透脓散加减。许芝银教授主张从“气”而论,分期治之,郁滞期采用蒲公英、青皮、橘叶、橘核、牡丹皮、赤巧药、漏芦等配伍,成脓期采用皇、皂角刺、炮山甲药物等配伍排脓,并及时切开引流或针管穿刺抽脓。然而上述中药配方的治疗效果不佳。
专利201410151736.9公开了一种治疗乳腺炎的中药组合物。由下述重量配比的中药原料组成:乳香10-25g,没药10-30g,炮山甲10-40g,昆布10-30g,海藻10-30g,砂仁5-35g,蒺藜花10-40g,木莲藤10-35g,水八角10-30g。专利201510003217.2公开了一种治疗乳腺炎的药物及其制备方法。它是由以下重量份的原料药组成:大葱150-250份、花椒10-20份、烧酒5-20份、小麦粉15-20份。专利201410070946.5公开了一种治疗急性乳腺炎的药物及其制备方法,它主要是由蒲公英、泽兰、瓜蒌皮、金银花、地丁、苦参、白芷、王不留行、紫花地丁、麦芽、陈皮、大青叶、麻黄等药物按一定重量配比制备而成。上述中药组合物虽然都有一定的疗效,但是,治疗效果都不佳,治愈率低,复发率高,副作用高。
发明内容
本发明的目的在于提供一种治疗妇科常见病乳腺炎的中药组合物及其制备方法。
一种治疗乳腺炎的中药组合物,由如下重量份数的原料制成:芍药8-12份,附子15-30份,瓜蒌12-18份,茜草根3-8份,密叶黑藓3-8份,牛膝3-8份。
所述治疗乳腺炎的中药组合物还包括萨卡巴豆根3-8份。
上述治疗乳腺炎的中药组合物的制备方法,按照如下步骤进行:
(1)称取干芍药根8-12份,干附子根15-30份,瓜蒌干果实12-18份,密叶黑藓干叶片3-8份,粉碎,过30-50目筛,装入渗漉筒,采用1-2倍重量的乙醇溶液溶胀0.5-1.5小时,再加6-12倍的乙醇溶液进行渗漉,流速1-3mL/min,得到渗漉液;
(2)称取茜草根3-8份,粉碎,过40-80目筛,加5-10倍重量份数的水回流提取3次,合并滤液,得到茜草根滤液;
(3)称取牛膝干茎段3-8份,粉碎,过40-80目筛,加入8-14倍重量份数的50-70%乙醇溶液,超声波提取30-60min,过滤,得到牛膝滤液;
(4)合并渗漉液,茜草根滤液,牛膝滤液,减压低温蒸干制成治疗乳腺炎的中药组合物。
所述乙醇溶液为60-80%的乙醇水溶液。
所述超声波提取,超声波的功率为200-400W。
所述减压低温蒸干的压强为10-20kPa,温度为8-16℃。
步骤(2)中还加入萨卡巴豆(Croton sacaquinba Croizat)根3-8份,同茜草根一起回流提取。
所述中药组合物可以做成片剂,崩解剂,水剂,膏剂或者其他药学上可接受的剂型。
本发明的中药组合物方解如下:
芍药:块根由根颈下方生出,肉质,粗壮,呈纺锤形或长柱形,含有芍药甙和安息香酸,具有疏肝清热、通孰消肿的功效;为君药。
附子:附乌头而生者为附子,如子附母也,乌头如芋魁,附子如芋子,附子具有回阳救逆,补火助阳,补益气血之功效;为君药;其含乌头碱,有微毒。
瓜蒌:属多年生攀缘草本,药用部位为其干燥的果实,具有疏肝清热、通孰消肿的功效;为君药。
茜草根:多年生攀援草本,长1-3米;功专活血,清血中之热,一以通壅积之瘀,斯血循故道而不横;为臣药。
密叶黑藓:拉丁文名Andreaea densifolia mitt.,丛生于高山或寒地裸露的花岗岩石上,植物体多呈紫黑色或灰黑色,叶细胞多有粗疣,与附子共提取能解乌头碱之毒,为佐药。
牛膝:为苋科、牛膝属多年生草本植物,高70-120厘米;根圆柱形,直径5-10毫米,常生长在山坡林下;其根茎部含皂甙,并含脱皮甾酮和牛膝甾酮,入药有逐瘀通经、补肝肾、强筋骨等效用;本发明中其为调和药,为使药。
本发明的有益效果:本发明制备的治疗乳腺炎的中药组合物,能有效治疗乳腺炎,治愈率高,复发率低,副作用小,芍药,附子,瓜蒌作为君药,药效高,茜草根作为臣药,起到增强药效的作用,密叶黑藓作品为佐药,起到减毒,减小副作用的功效,牛膝作为使用,起到调和作用。本发明的中药组合物制备方法简单,原料来源广,适合大规模工业化生产。
具体实施方式
下面结合具体实施例对本发明做进一步说明。
实施例1
一种治疗乳腺炎的中药组合物,由如下重量份数的原料制成:芍药100g,附子200g,瓜蒌150g,茜草根50g,密叶黑藓60g,牛膝50g。
上述治疗乳腺炎的中药组合物的制备方法,按照如下步骤进行:
(1)称取干芍药根100g,干附子根200g,瓜蒌干果实150g,密叶黑藓干叶片60g,粉碎,过40目筛,装入渗漉筒,用765mL的70%乙醇溶液溶胀1小时,再加4080mL的70%乙醇溶液进行渗漉,流速2mL/min,得到渗漉液;
(2)称取茜草根50g,粉碎,过60目筛,加400mL的水回流提取3次,合并滤液,得到茜草根滤液;
(3)称取牛膝干茎段50g,粉碎,过60目筛,加入500mL的60%乙醇溶液,超声波提取50min,超声波的功率为300W,过滤,得到牛膝滤液;
(4)合并渗漉液,茜草根滤液,牛膝滤液,减压低温蒸干制成治疗乳腺炎的中药组合物;所述减压低温蒸干的压强为15kPa,温度为12℃。
实施例2
一种治疗乳腺炎的中药组合物,由如下重量份数的原料制成:芍药80g,附子150g,瓜蒌120g,茜草根30g,密叶黑藓30g,牛膝30g。
上述治疗乳腺炎的中药组合物的制备方法,按照如下步骤进行:
(1)称取干芍药根80g,干附子根150g,瓜蒌干果实120g,密叶黑藓干叶片30g,粉碎,过30目筛,装入渗漉筒,用380mL的60%乙醇溶液溶胀0.5小时,再加3380mL的60%乙醇溶液进行渗漉,流速1mL/min,得到渗漉液;
(2)称取茜草根30g,粉碎,过40目筛,加150mL的水回流提取3次,合并滤液,得到茜草根滤液;
(3)称取牛膝干茎段30g,粉碎,过40目筛,加入250mL的50%乙醇溶液,超声波提取30min,超声波的功率为200W,过滤,得到牛膝滤液;
(4)合并渗漉液,茜草根滤液,牛膝滤液,减压低温蒸干制成治疗乳腺炎的中药组合物;所述减压低温蒸干的压强为10kPa,温度为8℃。
实施例3
一种治疗乳腺炎的中药组合物,由如下重量份数的原料制成:芍药120g,附子300g,瓜蒌180g,茜草根80g,密叶黑藓80g,牛膝80g。
上述治疗乳腺炎的中药组合物的制备方法,按照如下步骤进行:
(1)称取干芍药根120g,干附子根300g,瓜蒌干果实180g,密叶黑藓干叶片80g,粉碎,过50目筛,装入渗漉筒,用1360mL的80%乙醇溶液溶胀1.5小时,再加8160mL的80%乙醇溶液进行渗漉,流速3mL/min,得到渗漉液;
(2)称取茜草根80g,粉碎,过80目筛,加800mL的水回流提取3次,合并滤液,得到茜草根滤液;
(3)称取牛膝干茎段80g,粉碎,过80目筛,加入1120mL的70%乙醇溶液,超声波提取60min,超声波的功率为400W,过滤,得到牛膝滤液;
(4)合并渗漉液,茜草根滤液,牛膝滤液,减压低温蒸干制成治疗乳腺炎的中药组合物;所述减压低温蒸干的压强为20kPa,温度为16℃。
实施例4
一种治疗乳腺炎的中药组合物,由如下重量份数的原料制成:芍药100g,附子200g,瓜蒌150g,茜草根50g,密叶黑藓60g,牛膝50g,萨卡巴豆根50g。
上述治疗乳腺炎的中药组合物的制备方法,按照如下步骤进行:
(1)称取干芍药根100g,干附子根200g,瓜蒌干果实150g,密叶黑藓干叶片60g,萨卡巴豆根50g,粉碎,过40目筛,装入渗漉筒,用840mL的70%乙醇溶液溶胀1小时,再加4480mL的70%乙醇溶液进行渗漉,流速2mL/min,得到渗漉液;
(2)称取茜草根50g,粉碎,过60目筛,加400mL的水回流提取3次,合并滤液,得到茜草根滤液;
(3)称取牛膝干茎段50g,粉碎,过60目筛,加入500mL的60%乙醇溶液,超声波提取50min,超声波的功率为300W,过滤,得到牛膝滤液;
(4)合并渗漉液,茜草根滤液,牛膝滤液,减压低温蒸干制成治疗乳腺炎的中药组合物;所述减压低温蒸干的压强为15kPa,温度为12℃。
实施例5
一种治疗乳腺炎的中药组合物,由如下重量份数的原料制成:芍药100g,附子200g,瓜蒌150g,茜草根50g,密叶黑藓60g,蒙迪藤根50g。
上述治疗乳腺炎的中药组合物的制备方法,按照如下步骤进行:
(1)称取干芍药根100g,干附子根200g,瓜蒌干果实150g,密叶黑藓干叶片60g,粉碎,过40目筛,装入渗漉筒,用765mL的70%乙醇溶液溶胀1小时,再加4080mL的70%乙醇溶液进行渗漉,流速2mL/min,得到渗漉液;
(2)称取茜草根50g,粉碎,过60目筛,加400mL的水回流提取3次,合并滤液,得到茜草根滤液;
(3)称取蒙迪藤根50g,粉碎,过60目筛,加入500mL的60%乙醇溶液,超声波提取50min,超声波的功率为300W,过滤,得到牛膝滤液;
(4)合并渗漉液,茜草根滤液,牛膝滤液,减压低温蒸干制成治疗乳腺炎的中药组合物;所述减压低温蒸干的压强为15kPa,温度为12℃。
对比例1
一种治疗乳腺炎的中药组合物,由如下重量份数的原料制成:芍药100g,附子200g,瓜蒌150g,密叶黑藓60g,牛膝50g。
上述治疗乳腺炎的中药组合物的制备方法,按照如下步骤进行:
(1)称取干芍药根100g,干附子根200g,瓜蒌干果实150g,密叶黑藓干叶片60g,粉碎,过40目筛,装入渗漉筒,用765mL的70%乙醇溶液溶胀1小时,再加4080mL的70%乙醇溶液进行渗漉,流速2mL/min,得到渗漉液;

(1)称取茜草根50g,粉碎,过60目筛,加400mL的水回流提取3次,合并滤液,得到茜草根滤液;
(2)减压低温蒸干制成中药粉末;所述减压低温蒸干的压强为15kPa,温度为12℃。
对比例3
一种治疗乳腺炎的中药组合物,由如下重量份数的原料制成:芍药100g,附子200g,瓜蒌150g,茜草根50g,密叶黑藓60g。
上述治疗乳腺炎的中药组合物的制备方法,按照如下步骤进行:

(1)称取蒙迪藤根50g,粉碎,过60目筛,加入500mL的60%乙醇溶液,超声波提取50min,超声波的功率为300W,过滤,得到蒙迪藤滤液;
(2)减压低温蒸干制成中药粉末;所述减压低温蒸干的压强为15kPa,温度为12℃。
实验例1:脂多糖(LPS)诱发的小鼠乳腺炎性损伤修复实验
当机体乳腺组织发生明显的炎性反应时,从血管中渗出大量的嗜中性粒细胞,同时聚集于发生炎症的组织,细胞因子的诱导产生了的该炎性细胞的此种效应。血管内皮细胞由于TNF-α等细胞因子的刺激,粘附分子水平由于血管内皮表达的分泌而上调,从而能够使白细胞粘附于血管内皮细胞上,血管壁通透性随之增强,最后嗜中性粒细胞从血管中渗出,并且逐渐迁移到发生炎症的部位,即乳腺组织发生炎性损伤。
选取7周龄健康的Babl/C小鼠216只,其中144只雌鼠,72只雄鼠。购回后饲养50h以使其适应环境,然后以雌鼠与雄鼠2:1的比例合笼集体饲养,小鼠通过自然交配的方式受孕。雌鼠在分娩后5-7d时使用。小鼠的饲养条件:温度控制在25±1℃左右,相对湿度大约60%,专用配置饲料,小鼠自由采食、饮水,同时给予12h循环光照。
将泌乳5-7天的雌鼠随机分成12组,包括LPS刺激组、实施例1-5及对比例1-4组(10mg/kg体重)、LPS+DEX(地塞米松磷酸钠注射液)组(5mg/kg体重,阳性对照)和空白对照组,12只/组。
分别在乳导管灌注LPS之前1h和灌注之后12h给药,包括实施例1-5及对比例1-4组和LPS+DEX给药组,由腹腔注射。
上述各组泌乳雌鼠在LPS刺激24h后,将其颈椎脱臼处死。分别采集LPS刺激组,实施例1-5及对比例1-4组,LPS+DEX组和空白对照组中雌鼠乳腺组织的新鲜样本。置于-80℃冻存,用于细胞因子的检测。
将-80℃冻存上述各组雌鼠乳腺组织制备成组织匀浆,按照ELISA试剂盒的操作步骤进行TNF-α细胞因子蛋白表达水平的检测。每组3个重复样本,每个样本检测3次,数值取平均值。
检测结果如表1-3所示。

由表1可以看出,实施例1-3的TNF-α细胞因子表达水平与LPS刺激组相比有显著的降低,证明本发明的中药组合物能治疗小鼠的乳腺炎,但是效果不如阳性对照组,对比例2和对比例4TNF-α细胞因子表达水平与LPS刺激组,相当,证明茜草和蒙迪藤单独没有治疗小鼠的乳腺炎的功效,其不能作为治疗乳腺炎的君药使用。

由表2可以看出,实施例4与实施例1相比,TNF-α细胞因子表达水平显著降低,达到了极显著水平(P<0.01),证明萨卡巴豆根具有治疗小鼠的乳腺炎的功效,实施例5与实施例1相比,TNF-α细胞因子表达水平显著降低(P<0.05),虽然蒙迪藤根不能作为君药直接起药效,但是,其可以作为重要的调和药(使药),使得君药的活性成分更快更多的到达病变部位。

由表2可以看出,对比例1与实施例1相比,TNF-α细胞因子表达水平显著升高,虽然茜草根提取物没有直接的药效,但是,对比例1缺乏后,药效有所降低,证明其是具有辅助治疗的臣药;对比例3与与实施例1相比,TNF-α细胞因子表达水平显著升高(P<0.05),再次证明,虽然蒙迪藤根不能作为君药直接起药效,但是,其可以作为重要的调和药(使药),使得君药的活性成分更快更多的到达病变部位。
典型临床实验例:
章某,女,42岁,两胁及乳房胀疼,伴有肿块,局部皮肤发烫、红肿,苔白,乃肝气郁结、气血阻滞、脾胃痰聚所致。服用实施例1的治疗乳腺炎的中药组合物,每天10g,服用1个月,皮肤红肿消失,乳房肿块消失,痊愈。
王某,女,31岁,哺乳期4个月,左乳房疼痛,结肿块,伴恶寒,服用实施例4的治疗乳腺炎的中药组合物,每天10g,服用15天,后寒热减退,肿块消散,痛感消失即愈。
Claims (8)
1.一种治疗乳腺炎的中药组合物,其特征在于,由如下重量份数的原料制成:芍药8-12份,附子15-30份,瓜蒌12-18份,茜草根3-8份,密叶黑藓3-8份,牛膝3-8份。

3.权利要求1所述治疗乳腺炎的中药组合物的制备方法,其特征在于,按照如下步骤进行:
(1)称取干芍药根8-12份,干附子根15-30份,瓜蒌干果实12-18份,密叶黑藓干叶片3-8份,粉碎,过30-50目筛,装入渗漉筒,采用1-2倍重量的乙醇溶液溶胀0.5-1.5小时,再加6-12倍的乙醇溶液进行渗漉,流速1-3mL/min,得到渗漉液;
(2)称取茜草根3-8份,粉碎,过40-80目筛,加5-10倍重量份数的水回流提取3次,合并滤液,得到茜草根滤液;
(3)称取牛膝干茎段3-8份,粉碎,过40-80目筛,加入8-14倍重量份数的50-70%乙醇溶液,超声波提取30-60min,过滤,得到牛膝滤液;
(4)合并渗漉液,茜草根滤液,牛膝滤液,减压低温蒸干制成治疗乳腺炎的中药组合物。
4.根据权利要求3所述治疗乳腺炎的中药组合物的制备方法,其特征在于,所述乙醇溶液为60-80%的乙醇水溶液。
5.根据权利要求3所述治疗乳腺炎的中药组合物的制备方法,其特征在于,所述超声波提取,超声波的功率为200-400W。
6.根据权利要求3所述治疗乳腺炎的中药组合物的制备方法,其特征在于,所述减压低温蒸干的压强为10-20kPa,温度为8-16℃。
7.根据权利要求3所述治疗乳腺炎的中药组合物的制备方法,其特征在于,步骤(2)中还加入萨卡巴豆根3-8份,同茜草根一起回流提取。
8.权利要求1所述治疗乳腺炎的中药组合物,其特征在于,所述中药组合物可以做成片剂,崩解剂,水剂,膏剂或者其他药学上可接受的剂型。
