Abstract
本发明提供了一种治疗乳腺增生结节的中药组合物,制备所述中药组合物的主要原料重量份组成为:黄芪、当归、党参、赤芍、川穹、乌药份、川楝子、青皮、贝母、柴胡、佛手、延胡索、生地、蜈蚣、瓜蒌、生牡蛎、白术、甘草、穿山甲、急性子、三棱、莪术、僵蚕、山慈菇、枸杞子;本发明另一方面还提供了一种治疗乳腺增生结节的中药组合物的制备方法。该中药组合物配方科学合理,各组分协同配伍,能快速发挥药效,具有阴平相火、疏肝理气、软坚散积之功效,经大量试验证明,本发明能够有效改善人体肝气郁结和元气不足,对乳腺增生结节疗效显著,且疗效优异,标本兼顾,疗效肯定,安全方便且无毒副作用,没有任何西药或者激素药成分。

技术领域
本发明涉及中药技术领域,特别涉及一种治疗乳腺增生结节的中药组合物及其制备方法。
背景技术
乳腺增生结节是女性最常见的乳房疾病,其发病率占乳腺疾病的首位。近些年来该病发病率呈逐年上升的趋势,年龄也越来越低龄化。乳腺增生症是正常乳腺小叶生理性增生与复旧不全,乳腺正常结构出现紊乱,属于病理性增生,它是既非炎症又非肿瘤的一类病。多发于30-50岁女性,发病高峰为35-40岁。
很多女性病人以为乳腺增生是常见疾病,从而疏于治疗。实际上,仅有一部分轻度病人通过适度调护后便可自愈;绝大多数病人需通过治疗后才可彻底治愈;又有小部分病人会发展为乳腺癌。
西医治疗诊治乳腺增生病的方法非常多,现阶段西医治疗方案均为针对病因治疗,其诊治乳腺增生的方法如下:
1)性激素类药三苯氧胺
身体内雌性激素含量比较高或乳腺内雌性激素受体敏感性较强,皆有可能引发乳腺增生。三苯氧胺作为合成的抗雌性激素药品,能减小雌性激素对乳腺腺体的刺激。但这一类药品存在一定副面作用,如干扰月经,造成月经血减少、月经延迟或提早等月经不调的情况,且在停止用药后,极易复发。
2)非性激素类治疗药碘剂(碘化钾或复方碘溶液)
可适当缓解疼痛,改善病状,但长期使用可能会影响激素平衡以及甲状腺功能,故应谨慎使用。
3)维生素类(维生素E或维生素A)
见效较慢,临床治疗效果不理想。
4)利尿药
可以作为改善病状的辅助治疗药品,在月经前期乳房胀满时使用可缓解病状,但其只能临时减轻病状,故目前使用不多。
现虽有治疗乳腺增生结节的多种药物,但大多存仅能暂时减缓病痛、复发率高、副作用大、疗效差、见效慢、其疗效并不尽人意,;而中药制备的外用药物中所用原料药成分药效单一,原料药种类较多,有的中药原料药与其他原料药没有发生协同作用,影响了中药组合物的整体疗效。此外,现有的中药组合物多数是将所用原料直接粉碎入药,没有对原料内的活性成分进行提取分离,非有效成分没有除去,杂质较多,使中药组合物的组方过大,不利于患者服用,进一步影响中药组合物的疗效。
发明内容
针对上述现有技术中存在的问题与缺陷,本发明提供了一种治疗乳腺增生结节的中药组合物及其制备方法,该组合物疗效显著、安全方便且无毒副作用,其制备方法高效,提取有效成分效果好,适合工业生产。
本发明具体技术方案如下:
本发明的治疗乳腺增生结节的中药组合物,制备所述中药组合物的主要原料重量份组成为:黄芪1-20份、当归1-20份、党参1-20份、赤芍1-20份、川穹1-20份、乌药1-15份、川楝子1-20份、青皮1-15份、贝母1-20份、柴胡1-20份、佛手1-20份、延胡索1-20份、生地1-20份、蜈蚣1-20份、瓜蒌1-20份、生牡蛎2-25份、白术1-20份、甘草1-15份、穿山甲2-25份、急性子4-30份、三棱2-25份、莪术2-25份、僵蚕1-20份、山慈菇2-25份、枸杞子4-30份。
优选地,制备所述中药组合物的主要原料重量份组成为:黄芪5-15份、当归5-15份、党参5-15份、赤芍5-15份、川穹5-15份、乌药3-12份、川楝子5-15份、青皮3-12份、贝母5-15份、柴胡5-15份、佛手5-15份、延胡索5-15份、生地5-15份、蜈蚣5-15份、瓜蒌5-15份、生牡蛎8-20份、白术5-15份、甘草3-12份、穿山甲8-20份、急性子10-25份、三棱8-20份、莪术8-20份、僵蚕5-15份、山慈菇8-20份、枸杞子10-25份。
优选地,制备所述中药组合物的主要原料重量份组成为:黄芪10份、当归10份、党参10份、赤芍10份、川穹10份、乌药8份、川楝子10份、青皮8份、贝母10份、柴胡10份、佛手10份、延胡索10份、生地10份、蜈蚣10份、瓜蒌10份、生牡蛎12份、白术10份、甘草8份、穿山甲12份、急性子15份、三棱12份、莪术12份、僵蚕10份、山慈菇12份、枸杞子15份。
以上组成原料药的重量是以生药计算的,以重量份作为配比,在生产时可按照比例增大或减少,如大规模生产可以以公斤为单位,或以吨为单位,小规模生产也可以以克或毫克为单位,重量可以增大或者减小,但各组成之间的生药材重量配比的比例不变。且以上重量配比的比例是经过科学筛选得到的,可以相应调整组成的量的配比,增加或减少不超过100%,药效不变。
本发明的治疗乳腺增生结节的中药组合物组方科学合理,其原料药来源如下:
黄芪:为豆科植物蒙古黄芪或膜荚黄芪的干燥根,春、秋二季采挖,除去须根及根头,晒干。甘、温,归肺、脾经。补气固表,利尿托毒,排脓,敛疮生肌,用于气虚乏力,食少便溏,中气下陷,久泻脱肛,便血崩漏,表虚自汗,气虚水肿,痈疽难溃,久溃不敛,血虚痿黄,内热消渴,慢性肾炎蛋白尿,糖尿病。
当归:为伞形科植物当归的干燥根。秋末采挖,除去须根及泥沙,待水分稍蒸发后,捆成小把,上棚,用烟火慢慢熏干。甘、辛、温,归肝、心、脾经。补血活血,调经止痛,润肠通便,用于血虚萎黄,眩晕心悸,月经不调,经闭痛经,虚寒腹痛,肠燥便秘,风湿痹痛,跌扑损伤,痈疽疮疡,酒当归活血通经,用于经闭痛经,风湿痹痛,跌扑损伤。
党参:为桔梗科植物党参、素花党参(西党参)或川党参的干燥根,秋季采挖,洗净,晒干。甘,平,归脾、肺经。补中益气,健脾益肺,用于脾肺虚弱,气短心悸,食少便溏,虚喘咳嗽,内热消渴。
赤芍:为毛茛科植物芍药或川赤芍的干燥根,春、秋二季采挖,除去根茎、须根及泥沙,晒干。苦,微寒,归肝经。清热凉血,散瘀止痛,用于温毒发斑,吐血衄血,目赤肿痛,肝郁胁痛,经闭痛经,症瘕腹痛,跌扑损伤,痈肿疮疡。
川穹:为伞形科植物川穹的干燥根茎,夏季当茎上的节盘显著突出,并略带紫色时采挖,除去泥沙,晒后烘干,再去须根。辛,温,归肝、胆、心包经。活血行气,祛风止痛。用于胸痹痛,胸胁刺痛,跌扑肿痛,月经不调,经闭痛经,癥瘕腹痛,头痛,风湿痹痛。
乌药:为樟科植物乌药的干燥块根,全年均可采挖,除去细根,洗净,趁鲜切片,晒干,或直接晒干。辛,温,归肺、脾、肾、膀胱经。顺气止痛,温肾散寒,用于胸腹胀痛,气逆喘急,膀胱虚冷,遗尿尿频,疝气,痛经。
川楝子:为棟科植物川棟的干燥成熟果实,冬季果实成熟时采收,除去杂质,干燥。苦,寒、有小毒,归肝、小肠、膀胱经。疏肝泄热,行气止痛,杀虫,用于肝郁化火,胸胁、脘腹胀痛,疝气疼痛,虫积腹痛。
青皮:为芸香科植物橘及其栽培变种的干燥幼果或未成熟果实的果皮,5~6月收集自落的幼果,晒干,习称“个青皮”或“青皮子”;7~8月采收未成熟的果实,在果皮上纵剖成四瓣至基部,除尽瓤瓣,晒干,习称“四花青皮”。苦、辛,温,归肝、胆、胃经。疏肝破气,消积化滞,用于胸胁胀痛,疝气,乳核,乳痈,食积腹痛。
贝母:为百合科植物贝母。苦、甘,微寒,归肺、心经。清热润肺,化痰止咳。用于肺热燥咳,干咳少痰,阴虚劳嗽,咯痰带血。
柴胡:为伞形科植物柴胡或狭叶柴胡的干燥根,按性状不同,分别习称“北柴胡”及“南柴胡”,春、秋二季采挖,除去茎叶及泥沙,干燥。苦,微寒,归肝、胆经。和解表里,疏肝,升阳,用于感冒发热,寒热往来,胸胁胀痛,月经不调,子官脱垂,脱肛。
佛手:为芸香科柑橘属植物佛手的干燥果实,秋季果实尚未变黄或变黄时采收,纵切成薄片,晒干或低温干燥。辛、苦、酸,温、归肝、脾、肺经。舒肝理气,和胃止痛,用于肝胃气滞,胸胁胀痛,胃脘痞满,食少呕吐。
延胡索:为罂粟科植物延胡索的干燥块茎,夏初茎叶枯萎时采挖,除去须根,洗净,置沸水中煮至恰无白心时,取出,晒干。辛、苦,温,归肝、脾经。活血,利气,止痛,用于胸胁、脘腹疼痛,经闭痛经,产后瘀阻,跌扑肿痛。
生地:为玄参科植物地黄的新鲜或干燥块根,秋季采挖,除去芦头、须根及泥沙,将地黄缓缓烘焙至约八成干。甘、寒,归心、肝、肾经。清热凉血,养阴,生津,用于热病舌绛烦渴,阴虚内热,骨蒸劳热,内热消渴,吐血,衄血,发斑发疹。
蜈蚣:为蜈蚣科动物少棘巨蜈蚣的干燥体,春、夏二季捕捉,用竹片插入头尾,绷直,干燥。味辛、性温、有毒,肝经。祛风止痉、通络止痛、攻毒散结,主惊风,癫痫,痉挛抽搐,中风口,破伤风,风湿顽痹,偏正头痛,毒蛇咬伤,疮疡,瘰疬。
瓜蒌:为葫芦科植物栝楼或双边栝楼的干燥成熟果,秋季果实成熟时,连果梗剪下,置通风处阴干。甘、微苦、寒,归肺、胃、大肠经。清热涤痰,宽胸散结,润燥滑肠,用于肺热咳嗽,痰浊黄稠,胸痹心痛,结胸痞满,乳痈,肺痈,肠痈肿痛,大便秘结。
生牡蛎:为牡蛎科动物长牡、蛎近江牡蛎的贝壳。咸,微寒,归肝、胆、肾经。重镇安神,潜阳补阴,软坚散结,用于惊悸失眠,眩晕耳鸣,瘰疬痰核,症瘕痞块。煅牡蛎收敛固涩,用于自汗盗汗,遗精崩带,胃痛吞酸。
白术:为菊科植物白术的干燥根茎,冬季下部叶枯黄、上部叶变脆时采挖,除去泥沙,烘干或晒干,再除去须根。苦、甘,温,归脾、胃经。健脾益气,燥湿利水,止汗,安胎,用于脾虚食少,腹胀泄泻,痰饮眩悸,水肿,自汗,胎动不安,土白术健脾,和胃,安胎,用于脾虚食少,泄泻便溏,胎动不安。
甘草:为豆科植物甘草、胀果甘草或光果甘草的干燥,春、秋二季采挖,除去须根,晒干。甘,平,归心、肺、脾、胃经。补脾益气,清热解毒,祛痰止咳,缓急止痛,调和诸药,用于脾胃虚弱,倦怠乏力,心悸气短,咳嗽痰多,脘腹、四肢挛急疼痛,痈肿疮毒,缓解药物毒性、烈性。
穿山甲:为鲮鲤科动物穿山甲的鳞甲,收集鳞甲,洗净,晒干。咸,微寒,归肝、胃经。通经下乳,消肿排脓,搜风通络,用于经闭癓瘕,乳汁不通,痈肿疮毒,关节痹痛,麻木拘挛。
急性子:为凤仙花科植物凤仙花的干燥成熟种子,夏、秋季果实即将成熟时采收,晒干,除去果皮及杂质。微苦、辛、温、有小毒,归肺、肝经。破血软坚,消积。用于症瘕痞块,经闭,噎膈。
三棱:为黑三棱科植物黑三棱的干燥块茎,冬季至次年春采挖,洗净,削去外皮,晒干。辛、苦,平,归肝、脾经。破血行气,消积止痛,用于症瘕痞块,瘀血经闭,食积胀痛。
莪术:为姜科植物蓬莪术、广西莪术或温郁金的干燥根茎,后者习称“温莪术”,冬季茎叶枯萎后采挖,洗净,蒸或煮至透心,晒干或低温干燥后除去须根及杂质。辛、苦,温,归肝、脾经。行气破血,消积止痛,用于燀瘕痞块,瘀血经闭,食积胀痛,早期宫颈癌。
柴胡:为伞形科植物柴胡或狭叶柴胡的干燥根,按性状不同,分别习称“北柴胡”及“南柴胡”,春、秋二季采挖,除去茎叶及泥沙,干燥。苦,微寒,归肝、胆经。和解表里,疏肝,升阳,用于感冒发热,寒热往来,胸胁胀痛,月经不调,子官脱垂,脱肛。
僵蚕:为蚕蛾科昆虫家蚕4~5龄的幼虫感染(或人工接种)白僵菌而致死的幼虫干燥体,多于春、秋季生产,将感染白僵菌病死的蚕干燥。咸、辛,平,归肝、肺、胃经。祛风定惊,化痰散结,用于惊风抽搐,咽喉肿痛,皮肤瘙痒,颌下淋巴结炎,面神经麻痹。
山慈菇:为兰科植物杜鹃兰、独蒜兰或云南独蒜兰的干燥假鳞茎,前者习称“毛慈菇”,后二者习称“冰球子”,夏、秋二季采挖,除去地上部分及泥沙,分开大小置沸水锅中蒸煮至透心,干燥。甘、微辛、凉,归肝、脾经。清热解毒,化痰散结,用于痈肿疔毒,瘰疬痰核,淋巴结结核,蛇虫咬伤。
枸杞子:为茄科植物宁夏枸杞的干燥成熟果实,夏、秋二季果实呈红色时采收,热风烘干,除去果梗。或晾至皮皱后,晒干,除去果梗。甘,平,归肝、肾经。滋补肝肾,益精明目,用于虚劳精亏,腰膝酸痛,眩晕耳鸣,内热消渴,血虚萎黄,目昏不明。
作为本发明的进一步改进,本发明提供了一种治疗褥疮和皮肤溃烂的制剂,所述制剂由中药组合物单独制成或由和中药组合物和药学上可接受的载体组成。
进一步地,所述制剂为口服制剂或外用制剂,所述口服制剂为水提物、片剂、丸剂、水蜜丸、浓缩丸、散剂、胶囊剂、口服液、颗粒剂或保健茶饮口服制剂;所述外用制剂为熏蒸剂、洗剂或膏剂。
进一步地,所述外用制剂为胶囊剂,所述胶囊剂包括重量份数比为1∶1-2.5中药组合物与辅料。
进一步地,所述辅料包括以下重量份数的成分:乳糖5-15份、十二烷基硫酸钠5-15份、甲基苯基环氧丙酸乙酯3-7份、羧甲基淀粉钠3-7份、微晶硅胶10-20份、硬脂酸镁3-7份、乙基环糊精5-15份。
进一步地,所述胶囊填充物包括以下重量份数的成分:中药组合物30份、乳糖10份、十二烷基硫酸钠10份、甲基苯基环氧丙酸乙酯5份、羧甲基淀粉钠5份、微晶硅胶15份、硬脂酸镁5份、乙基环糊精10份。
作为本发明的进一步改进,本发明还提供了一种治疗乳腺增生结节的中药组合物的制备方法,该方法包括如下步骤:
S1、称取处方量的黄芪、当归、党参、赤芍、川穹、乌药、川楝子、青皮、贝母、柴胡、佛手、延胡索、生地13味原料药,粉碎、碾磨、过筛制成细粉,所述筛网的数目为150-180目,加水煎煮3次,第一次12倍水量煎煮3h,第二次10倍水量煎煮2h,第二次8倍水量煎煮1h,合并得煎液,温度50℃条件下浓缩成相对密度为1.30的清膏1,冷却,加75%的乙醇进行醇沉2h,过滤,在50℃下浓缩成相对密度为1.45的干浸膏1;
S2、称取处方量的蜈蚣、瓜蒌、生牡蛎、白术、甘草、穿山甲、急性子、三棱、莪术、僵蚕、山慈菇、枸杞子12味原料药,粉碎、碾磨、过筛制成细粉,所述筛网的数目为60-100目,在搅拌均匀后,加入10倍75%乙醇的回流提取3次,每次1.5h,过滤,合并回流液,浓缩至在55℃下相对密度为1.20的清膏2,用5倍水量将清膏溶解,上大孔树脂柱,依次用水和45%的乙醇洗脱,回收乙醇洗脱液,在55℃下浓缩至相对密度为1.35的干浸膏2;
S3、合并干浸膏1和干浸膏2,将其粉碎成粉,然后置于槽型混合机内混合1h,即得治疗乳腺增生结节的中药组合物。
另外的,本发明还提出了治疗乳腺增生结节的中药组合物作为治疗褥疮和皮肤溃烂的应用。
由此可见,本发明提供的治疗乳腺增生结节的中药组合物,该中药组合物配方科学合理,各组分协同配伍,能快速发挥药效,具有阴平相火、疏肝理气、软坚散积之功效,经大量试验证明,本发明能够有效改善人体肝气郁结和元气不足,对乳腺增生结节疗效显著,且疗效优异,标本兼顾,疗效肯定,安全方便且无毒副作用,没有任何西药或者激素药成分。

本实施例提供了一种治疗乳腺增生结节的中药组合物,该中药组合物包括以下重量克数的原料:
黄芪10g、当归10g、党参10g、赤芍10g、川穹10g、乌药8g、川楝子10g、青皮8g、贝母10g、柴胡10g、佛手10g、延胡索10g、生地10g、蜈蚣10g、瓜蒌10g、生牡蛎12g、白术10g、甘草8g、穿山甲12g、急性子15g、三棱12g、莪术12g、僵蚕10g、山慈菇12g、枸杞子15g。
其中,本实施例提供了一种治疗乳腺增生结节的中药组合物的制备方法,包括如下步骤:
S1、称取处方量的黄芪、当归、党参、赤芍、川穹、乌药、川楝子、青皮、贝母、柴胡、佛手、延胡索、生地13味原料药,粉碎、碾磨、过筛制成细粉,所述筛网的数目为150-180目,加水煎煮3次,第一次12倍水量煎煮3h,第二次10倍水量煎煮2h,第二次8倍水量煎煮1h,合并得煎液,温度50℃条件下浓缩成相对密度为1.30的清膏1,冷却,加75%的乙醇进行醇沉2h,过滤,在50℃下浓缩成相对密度为1.45的干浸膏1;
S2、称取处方量的蜈蚣、瓜蒌、生牡蛎、白术、甘草、穿山甲、急性子、三棱、莪术、僵蚕、山慈菇、枸杞子12味原料药,粉碎、碾磨、过筛制成细粉,所述筛网的数目为60-100目,在搅拌均匀后,加入10倍75%乙醇的回流提取3次,每次1.5h,过滤,合并回流液,浓缩至在55℃下相对密度为1.20的清膏2,用5倍水量将清膏溶解,上大孔树脂柱,依次用水和45%的乙醇洗脱,回收乙醇洗脱液,在55℃下浓缩至相对密度为1.35的干浸膏2;
S3、合并干浸膏1和干浸膏2,将其粉碎成粉,然后置于槽型混合机内混合1h,即得治疗乳腺增生结节的中药组合物。
实施例2:
本实施例提供了一种治疗乳腺增生结节的中药组合物,该中药组合物包括以下重量克数的原料:

本实施例提供一种治疗乳腺增生结节的中药组合物和辅料制成酊剂,该酊剂包括以下重量克数的原料:
黄芪10g、当归10g、党参10g、赤芍10g、川穹10g、乌药8g、川楝子10g、青皮8g、贝母10g、柴胡10g、佛手10g、延胡索10g、生地10g、蜈蚣10g、瓜蒌10g、生牡蛎12g、白术10g、甘草8g、穿山甲12g、急性子15g、三棱12g、莪术12g、僵蚕10g、山慈菇12g、枸杞子15g。
制备方法:将上述药物清洗干净,阴干后置于1000g的75%酒精酒精液中,浸泡15天,滤出药渣,剩余酒液渗滤两次后置于清洁瓶中备用。

试验例1稳定性试验
1.加速试验
取实施例1、实施例6、实施例7、对照例3和对照例4的合剂,分成3批,分别编号为样品1、样品2、样品3、样品4和样品5,在温度40℃±2℃下,相对湿度为75%±5%的条件下放置6个月,在试验期间1个月、2个月、3个月、6个月末分别取样一次,按中国药典中的规定,检测合剂的性状、色泽、气味和主要成分的含量(标示量%)、溶出度以及有关物质,结果发现,样品1、样品2和样品3各项指标均无明显变化,而样品4和样品5颜色变深,出现沉淀,并在6个月的时候均出现轻微异味。
2.长期试验
取实施例1、实施例6、实施例7、对照例3和对照例4的合剂,分成3批,分别编号为样品1、样品2、样品3、样品4和样品5,在温度25℃±2℃下,相对湿度为60%±10%的条件下放置24个月,在试验期间0个月、3个月、6个月、9个月、12个月、18个月、24个月末分别取样一次,按中国药典中的规定,检测合剂的性状、色泽、气味和主要成分的含量(标示量%)、溶出度以及有关物质,结果发现,样品1、样品2和样品3各项指标均无明显变化,而样品4和样品5颜色变深,出现沉淀,并在6个月的时候均出现轻微异味,在12个月时候出现轻微臭味。
结论本发明中药组合物制成的合剂由于辅料的合理选择,药品稳定性好、储存时间长,而十二烷基硫酸钠和甲基苯基环氧丙酸乙酯的加入更显著提高了合剂的稳定性。
试验例2毒性试验
(一)急性毒性试验
1.试验对象:健康SD大鼠
试验组50例,雄鼠,年龄5周龄;
对照组50例,雄鼠,年龄5周龄。
2.试验方法:
试验组:将本发明治疗乳腺增生结节的中药组合物用生理盐水配制成65%浓度的溶液,给大鼠按一次40mL/kg灌胃,一日内连续两次,然后停药,停药后继续观察7天。
对照组:用生理盐水给大鼠按一次40mL/kg灌胃,一日内连续两次,然后停药,停药后继续观察7天。
3.统计方法:分别在试验1天、2天、3天、4天、5天、6天、7天,记录两组大鼠的死亡率及大鼠行为有无明显异常;在观察期结束后,于第8天处死大鼠,进行剖检,记录两组大鼠的脏器异常改变情况。
4.试验结果:
试验结果见表1。

表1的比较结果如下:
由试验组和对照组的比较可知,本发明治疗乳腺增生结节的中药组合物对大鼠没有急性毒性。
5.结论:本发明治疗乳腺增生结节的中药组合物,安全度高,无急性毒性。
(二)长期毒性试验
1.试验对象:健康SD大鼠
试验组50例,雄鼠,年龄5周龄;
对照组50例,雄鼠,年龄5周龄。
2.试验方法:
试验组:将本发明治疗乳腺增生结节的中药组合物溶解在水中灌胃,用药剂量以白鼠体重计,每日0.05g/kg,连续180天,停药后继续观察30天。
对照组:喂食与试验组一样且不含本发明中药组合物的食物,食物量也与试验组一样,并灌胃等量生理盐水,连续180天;随后的30天观察期期间,大鼠与试验组一样对待。
3.统计方法:分别在试验0天、30天、60天、90天、120天、150天、180天、及观察期结束时,记录两组大鼠的死亡率及大鼠行为有无明显异常:在观察期结束后,处死大鼠,进行剖检,记录两组大鼠的饮食、饮水、体重增长和脏器异常改变情况。
4.试验结果:试验结果见表2。

表2的比较结果如下:
由试验组和对照组的比较可知,本发明治疗乳腺增生结节的中药组合物对大鼠无长期毒性;
5.结论:本发明的治疗乳腺增生结节的中药组合物,安全度高,无长期毒性。
(三)结论
由急性毒性试验结果和长期毒性试验结果可知,本发明的治疗乳腺增生结节的中药组合物,安全度高,无毒性。
试验例3抑菌试验
使用下列化合物,采用本领域中经典的滤纸片法来进行抑菌试验。
供试菌种荚膜红细菌ATCC 11166购自上海北诺生物科技有限公司。
培养液
营养琼脂和营养肉汤,购自北京中科晨宇实验设备有限公司。
试验方法
将荚膜红细菌ATCC 11166接种于琼脂平板营养液平面上,接种时均匀密布。
取无菌圆形滤纸片,分别喷洒5mL的实施例1中药组合物溶解成的溶液、5mL的实施例2中药组合物溶解成的溶液,至完全润湿,贴在已接种细菌的琼脂平板营养液平面上。将琼脂平板置于37℃的培养箱内,温育4小时。测量抑菌环的直径。测定3次取平均值。
结论
实施例1中药组合物溶解成的溶液的荚膜红细菌ATCC 11166的抑菌环的平均直径为11.20mm,实施例2中药组合物溶解成的溶液的荚膜红细菌ATCC 11166的抑菌环的平均直径为13.75mm。
治疗乳腺增生结节的效果评价试验
本发明的治疗乳腺增生结节的中药组合物效果如下:
1、临床资料
本发明的各优选实施例已经在500人的规模性测试中获得了验证。
2、疗效评定
痊愈:临床症状全部消失,停药后半年于一年复查未见任何病理性改变:
显效:临床症状全部消失,停药半年后复查病理改变趋于治愈;
无效:临床症状虽有改善,但病理改变无明显好转。
本发明的各优选实施例已经在500人的规模性测试中获得了验证,在用药后,具有疏肝解郁、化痰通络、破瘀散结之功效,对乳腺增生结节有很好的治疗效果,痊愈率在85%以上,显效率在95%以上。
典型病例1:
钱某,女,58岁,双乳痛疼难忍,活动受限,红外透照触诊左乳房内外上象限6×6cm大小肿块,外下象限2×4cm索状肿块,右乳房内外上象限间5×5cm大小肿块,外下象限2×4cm索状肿块,诊断为乳腺囊性增生,伴导管增生。服用了本发明的中药组合物,一个月后,肿块全部消失,随访至今未再复发。
典型病例2:
赵某,女,30岁,左侧乳房外上象限有一个3×3cm大小肿块,外下象限有一个2×4cm条索状肿块;右侧乳房内外象限之间有一个3×3cm大小肿块,外下象限有一个2×3cm大小条索状肿块。服用了本发明的中药组合物,一个月后,肿块全部消失,随访至今未再复发。
典型病例3:
蒋某,女,40岁,左侧乳房内外上象限间6×6cm大小肿块,内下象限散弹状0.5×0.5cm大小肿块七八处,外下象限3×5cm条索状肿块;内外乳房内外象限间5×5cm大小肿块,内下象限散弹状0.5×0.5cm大小肿块七八处,外下象限2×4cm大小条索状肿块。服用了本发明的中药组合物,三个月后,肿块全部消失,随访至今未再复发
虽然,上文中已经用一般性说明、具体实施方式及试验,对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。

1.一种治疗乳腺增生结节的中药组合物,其特征在于,制备所述中药组合物的主要原料重量份组成为:黄芪1-20份、当归1-20份、党参1-20份、赤芍1-20份、川穹1-20份、乌药1-15份、川楝子1-20份、青皮1-15份、贝母1-20份、柴胡1-20份、佛手1-20份、延胡索1-20份、生地1-20份、蜈蚣1-20份、瓜蒌1-20份、生牡蛎2-25份、白术1-20份、甘草1-15份、穿山甲2-25份、急性子4-30份、三棱2-25份、莪术2-25份、僵蚕1-20份、山慈菇2-25份、枸杞子4-30份。
2.如权利要求1所述的治疗乳腺增生结节的中药组合物,其特征在于,制备所述中药组合物的主要原料重量份组成为:黄芪5-15份、当归5-15份、党参5-15份、赤芍5-15份、川穹5-15份、乌药3-12份、川楝子5-15份、青皮3-12份、贝母5-15份、柴胡5-15份、佛手5-15份、延胡索5-15份、生地5-15份、蜈蚣5-15份、瓜蒌5-15份、生牡蛎8-20份、白术5-15份、甘草3-12份、穿山甲8-20份、急性子10-25份、三棱8-20份、莪术8-20份、僵蚕5-15份、山慈菇8-20份、枸杞子10-25份。
3.如权利要求1所述的治疗乳腺增生结节的中药组合物,其特征在于,其特征在于,制备所述中药组合物的主要原料重量份组成为:黄芪10份、当归10份、党参10份、赤芍10份、川穹10份、乌药8份、川楝子10份、青皮8份、贝母10份、柴胡10份、佛手10份、延胡索10份、生地10份、蜈蚣10份、瓜蒌10份、生牡蛎12份、白术10份、甘草8份、穿山甲12份、急性子15份、三棱12份、莪术12份、僵蚕10份、山慈菇12份、枸杞子15份。
4.如权利要求1-3中任一项所述的治疗乳腺增生结节的中药组合物的制剂,其特征在于,所述制剂由中药组合物单独制成或由和中药组合物和药学上可接受的载体组成。
5.如权利要求4所述的制剂,其特征在于,所述制剂为口服制剂或外用制剂,所述口服制剂为水提物、片剂、丸剂、水蜜丸、浓缩丸、散剂、胶囊剂、口服液、颗粒剂或保健茶饮口服制剂;所述外用制剂为熏蒸剂、洗剂或膏剂。
6.如权利要求5所述的制剂,其特征在于,所述外用制剂为胶囊剂,所述胶囊剂包括重量份数比为1∶1-2.5中药组合物与辅料。
7.如权利要求6所述的制剂,其特征在于,所述辅料包括以下重量份数的成分:乳糖5-15份、十二烷基硫酸钠5-15份、甲基苯基环氧丙酸乙酯3-7份、羧甲基淀粉钠3-7份、微晶硅胶10-20份、硬脂酸镁3-7份、乙基环糊精5-15份。
8.如权利要求6或7所述的制剂,其特征在于,所述胶囊填充物包括以下重量份数的成分:中药组合物30份、乳糖10份、十二烷基硫酸钠10份、甲基苯基环氧丙酸乙酯5份、羧甲基淀粉钠5份、微晶硅胶15份、硬脂酸镁5份、乙基环糊精10份。
9.一种如权利要求1-3中任一项所述的治疗乳腺增生结节的中药组合物的制备方法,其特征在于,所述方法包括如下步骤:
S1、称取处方量的黄芪、当归、党参、赤芍、川穹、乌药、川楝子、青皮、贝母、柴胡、佛手、延胡索、生地13味原料药,粉碎、碾磨、过筛制成细粉,所述筛网的数目为150-180目,加水煎煮3次,第一次12倍水量煎煮3h,第二次10倍水量煎煮2h,第二次8倍水量煎煮1h,合并得煎液,温度50℃条件下浓缩成相对密度为1.30的清膏1,冷却,加75%的乙醇进行醇沉2h,过滤,在50℃下浓缩成相对密度为1.45的干浸膏1;
S2、称取处方量的蜈蚣、瓜蒌、生牡蛎、白术、甘草、穿山甲、急性子、三棱、莪术、僵蚕、山慈菇、枸杞子12味原料药,粉碎、碾磨、过筛制成细粉,所述筛网的数目为60-100目,在搅拌均匀后,加入10倍75%乙醇的回流提取3次,每次1.5h,过滤,合并回流液,浓缩至在55℃下相对密度为1.20的清膏2,用5倍水量将清膏溶解,上大孔树脂柱,依次用水和45%的乙醇洗脱,回收乙醇洗脱液,在55℃下浓缩至相对密度为1.35的干浸膏2;
S3、合并干浸膏1和干浸膏2,将其粉碎成粉,然后置于槽型混合机内混合1h,即得治疗乳腺增生结节的中药组合物。
10.一种权利要求1-3任一项所述的中药组合物作为治疗乳腺增生结节药物中的应用。
