Abstract
本发明公开了一种中药复方提取物的制备方法、提取物及在抑菌组合物中的应用,该中药复方提取物的制备方法包括以下步骤:(1)将包含以下重量份组分混合:5‑15份甘草、15‑25份枯芩、5‑15份大黄、15‑25份黄柏和15‑25份苦参,将混合物粉碎打粉;(2)加入蒸馏水浸泡,加入1500‑2500U/g酶水解,在35‑50℃水浴搅拌1‑2h,过滤,将滤液浓缩、干燥,得中药复方提取物;所述酶选自纤维素酶、果胶酶或蛋白酶中的一种或多种。本发明制备的中药提取物在抑菌组合物中,可应用在乳膏、凝胶、混悬液中,抑菌效果好,温和无毒副作用。

技术领域
本发明涉及一种提取物的制备方法及该提取物制备的组合物,特别涉及一种中药复方提取物的制备方法及含有该提取物的抑菌组合物。
背景技术
一些剂型在生产制造和使用的过程中很容易被生活中常见的微生物如大肠杆菌、金黄色葡萄球菌、白色念珠菌等污染,从而发生腐败,因此常需要添加抑菌剂。目前,最常用的抑菌剂如DMDM乙内酰脲,尼泊金甲酯、尼泊金丙酯、凯松等,是通过化学合成得到的。这些抑菌剂存在潜在的风险,对皮肤有一定的慢性毒性,有可能致癌或致敏。因此,以天然中药研究开发出对大肠杆菌等微生物具有抑制作用,而且温和无刺激的天然抑菌成分配方,市场前景非常广阔。
紫草素,从紫草的干燥根中提取得到的天然活性成分,是紫草中特有的一种脂溶性萘醌色素类化合物,具有抗炎、抑菌、止痒及修复等功效。研究发现,紫草素与抗菌药物胡桃醌的化学结构相似。紫草素具有较广的抗菌谱,对金黄色葡萄球菌、蜡状芽孢杆菌等革兰氏阳性菌,和大肠埃希菌、沙门氏菌等革兰氏阴性菌,以及白色念珠菌均具有抑制作用。
甘草,豆科甘草属植物甘草、胀果甘草或光果甘草的干燥根及根茎,具有补脾益气、祛痰止咳、缓急止痛、清热解毒、调和诸药等功效,历代皆为重要药物。甘草主要的生物活性物质是三萜皂苷类和黄酮类,其中甘草素、甘草查尔酮等成分抑菌效果明显,文献显示其对大肠杆菌、双歧杆菌、绿脓杆菌、金黄色葡萄球菌均有抑制作用。
枯芩是规格不同等级黄芩中的一种,多属于野生老根。具有清热燥湿、泻火解毒等功效,常用于治疗湿热痞满、泻痢、黄疸、肺热咳嗽、高热烦渴等。枯芩中含多种黄酮类化合物,主要为黄芩苷、黄芩素、汉黄芩素、汉黄芩苷。研究表明,黄芩无论是单用还是联合使用都表现出极强的抗菌功效。
大黄是多种蓼科大黄属的多年生植物的合称,具有攻积滞、清湿热、抗菌、凉血、祛瘀、解毒等功效。文献显示,大黄的水浸液对金黄色葡萄球菌、大肠杆菌、痢疾杆菌、绿脓杆菌、伤寒沙门菌均有一定的抑制作用,其中对金黄色葡萄球菌、痢疾杆菌、伤寒沙门菌的抑菌效果最强。
黄柏为临床常用中药,来源于芸香科植物黄皮树和黄檗的干燥树皮。黄柏具有抗病原微生物和原虫、解热、抗炎等作用。有研究报道,黄柏对金黄色葡萄球菌、白色葡萄球菌、甲型链球菌、乙型链球菌、变形杆菌、伤寒沙门菌均有明显的抑制作用。
苦参又名枯骨、草槐、地槐等,为豆科植物苦参的干燥根,具有清热燥湿、杀虫止痒、安神等功效。苦参中可分离出23种生物碱,其中主要是氧化苦参碱和苦参碱,另外还有羟基苦参碱、去氢苦参碱等。苦参体外抑菌作用的研究表明,其对大肠杆菌、金黄色葡萄球菌、甲型链球菌、乙型链球菌以及变形杆菌的生长均具有较强的抑制作用。
现有技术中,申请号为201610467423.6的专利《一种抑菌凝胶》,配方添加醋酸洗必泰抑菌成分,虽然抑菌效果强,但是存在低毒性,而且对眼睛、呼吸系统和皮肤有一定的刺激性。申请号为201310165844.7的专利《一种含多种天然抑菌成分的洗手液及制作工艺》,所述的抑菌成分为葡萄籽、石榴籽、石榴皮、大叶女贞树果实、香樟树果实、香樟叶的提取物,但是存在提取方法复杂,且产品抑菌效果未知的问题。
发明内容
发明目的:为了克服以上技术问题,本发明提供了一种中药复方提取物的制备方法。本发明的另一目的是提供了中药复方提取物。本发明还提供了含有该中药复方提取物的抑菌组合物。
技术方案:本发明所述一种中药复方提取物的制备方法,包括以下步骤:(1)将包含以下重量份组分混合:5-15份甘草、15-25份枯芩、5-15份大黄、15-25份黄柏和15-25份苦参,将混合物粉碎打粉得中药粉末,使其能充分接触媒介;(2)加入蒸馏水浸泡,加入1500-2500U/g酶水解,在35-50℃水浴搅拌1-2h,酶解植物组织一定时间,破坏植物的细胞壁,使植物有效成分最大限度溶解分离,过滤,将滤液浓缩、干燥,得中药复方提取物;所述酶选自纤维素酶、果胶酶或蛋白酶中的一种或多种。
为充分提取中药复方的功效成分,提取时,所述蒸馏水为中药粉末的体积的8-10倍量,优选9倍量;所述浸泡时间为1-3h,优选2h;上述水解的酶优选为果胶酶。
优选地,步骤(1)中,包含以下重量份组分:10-15份甘草、20-25份枯芩、10-15份大黄、20-25份黄柏和20-25份苦参。
进一步地,步骤(1)中,包含以下重量份组分:15份甘草、25份枯芩、15份大黄、25份黄柏25份苦参,即当甘草、枯芩、大黄、黄柏和苦参的重量比为15:25:15:25:25时,该中药复方提取物的抑菌效果更优。
上述方法制备的中药复方提取物,用于制备抑菌组合物,该抑菌组合物包含以下重量百分比组分:0.01-0.15%紫草素和2-6%中药复方提取物。
优选地,上述抑菌组合物包含以下重量百分比组分:0.05-0.10%紫草素和3-5%中药复方提取物。
上述抑菌组合物在乳膏、凝胶、混悬液中的应用。
一种抑菌乳膏,包含以下重量百分比组分:2-5%鲸蜡基葡萄糖苷、4-10%硬脂酸、1-3%单硬脂酸甘油酯、1.5-4%十六醇、2-6%棕榈酸异丙酯、2-6%丙二醇、0.01-0.15%紫草素、2-6%中药复方提取物、0.1-0.3%VE、0.1-0.4%甘草酸二钾、0.01-0.05%透明质酸和0.01-0.05%EDTA-2Na。

有益效果:本发明中药复方提取物所用的提取方法与传统方法的水蒸馏法和溶剂浸提法相比,提取难度大幅降低,提取条件温和,既能保证中药有效抑菌成分的活性,又能提高提取物的得率;本发明将制备的中药复方提取物应用于乳膏、凝胶、混悬液中,配方新颖,包括紫草素以及甘草、枯芩、大黄、黄柏、苦参的中药复方提取物,抑菌的有效成分来源于天然植物,抑菌效果好,温和无毒副作用,对大肠杆菌、金黄色葡萄球菌、白色念珠菌均具有良好的抑制作用。
具体实施方式
一、中药复方提取物的制备
制备例1:将重量份为5份甘草、15份枯芩、5份大黄、15份黄柏和15份苦参混合,粉碎打粉得中药粉末,加入中药粉末8倍体积量的蒸馏水,浸泡1h,加入1500U/g纤维素酶水解,在35℃水浴搅拌1h,将滤液浓缩、干燥,得中药复方提取物。
制备例2:将重量份为10份甘草、20份枯芩、10份大黄、20份黄柏和20份苦参混合,粉碎打粉得中药粉末,加入中药粉末10倍体积量的蒸馏水,浸泡3h,加入2500U/g蛋白酶水解,在50℃水浴搅拌2h,将滤液浓缩、干燥,得中药复方提取物。
制备例3:将重量份为15份甘草、25份枯芩、15份大黄、25份黄柏和25份苦参混合,粉碎打粉得中药粉末,加入中药粉末9倍体积量的蒸馏水,浸泡2h,加入2000U/g果胶酶水解,在45℃水浴搅拌1.5h,将滤液浓缩、干燥,得中药复方提取物。
水蒸馏法:将重量份为15份甘草、25份枯芩、15份大黄、25份黄柏和25份苦参混合,粉碎打粉得中药粉末,加入中药粉末9倍体积量的蒸馏水,浸泡2h,在45℃水浴搅拌1.5h,将滤液浓缩、干燥,得中药复方提取物。
溶剂法:将重量份为15份甘草、25份枯芩、15份大黄、25份黄柏和25份苦参混合,粉碎打粉得中药粉末,加入中药粉末9倍体积量的70%乙醇,浸泡2h,在45℃水浴搅拌1.5h,将滤液浓缩、干燥,得中药复方提取物。
二、样品的制备制备例3所制得的中药复方提取物,制备以下样品:
实施例1:一种抑菌乳膏,以重量百分比计,4%鲸蜡基葡萄糖苷,6%硬脂酸,2%单硬脂酸甘油酯,3%十六醇,3%棕榈酸异丙酯,4%丙二醇,0.05%紫草素,3%中药复方提取物,0.2%油溶性VE,0.2%甘草酸二钾,0.03%透明质酸,0.02%EDTA-2Na,余量为水。
实施例2:一种抑菌乳膏,以重量百分比计,4%鲸蜡基葡萄糖苷,6%硬脂酸,2%单硬脂酸甘油酯,3%十六醇,3%棕榈酸异丙酯,4%丙二醇,0.08%紫草素,4%中药复方提取物,0.2%油溶性VE,0.2%甘草酸二钾,0.03%透明质酸,0.02%EDTA-2Na,余量为水。
实施例3:一种抑菌乳膏,以重量百分比计,4%鲸蜡基葡萄糖苷,6%硬脂酸,2%单硬脂酸甘油酯,3%十六醇,3%棕榈酸异丙酯,4%丙二醇,0.10%紫草素,5%中药复方提取物,0.2%油溶性VE,0.2%甘草酸二钾,0.03%透明质酸,0.02%EDTA-2Na,余量为水。
实施例4:一种抑菌乳膏,以重量百分比计,2%鲸蜡基葡萄糖苷,4%硬脂酸,1%单硬脂酸甘油酯,1.5%十六醇,2%棕榈酸异丙酯,2%丙二醇,0.1%紫草素,5%中药复方提取物,0.1%油溶性VE,0.1%甘草酸二钾,0.01%透明质酸,0.01%EDTA-2Na,余量为水。
实施例5:一种抑菌乳膏,以重量百分比计,5%鲸蜡基葡萄糖苷,10%硬脂酸,3%单硬脂酸甘油酯,4%十六醇,6%棕榈酸异丙酯,6%丙二醇,0.1%紫草素,5%中药复方提取物,0.3%油溶性VE,0.4%甘草酸二钾,0.05%透明质酸,0.05%EDTA-2Na,余量为水。
实施例6:一种抑菌凝胶,以重量百分比计,4%丙二醇,0.6%PEG-60氢化蓖麻油,0.5%卡波姆,0.4%角鲨烷,0.5%丙烯酰二甲基牛磺酸铵/VP共聚物,0.05%紫草素,3%中药复方提取物,0.2%甘草酸二钾,0.02%EDTA-2Na,余量为水。
实施例7:一种抑菌凝胶,以重量百分比计,4%丙二醇,0.6%PEG-60氢化蓖麻油,0.5%卡波姆,0.4%角鲨烷,0.5%丙烯酰二甲基牛磺酸铵/VP共聚物,0.08%紫草素,4%中药复方提取物,0.2%甘草酸二钾,0.02%EDTA-2Na,余量为水。

实施例9:一种抑菌凝胶,以重量百分比计,2%丙二醇,0.05%PEG-60氢化蓖麻油,0.2%卡波姆,0.2%角鲨烷,0.2%丙烯酰二甲基牛磺酸铵/VP共聚物,0.1%紫草素,5%中药复方提取物,0.1%甘草酸二钾,0.01%EDTA-2Na,余量为水。
实施例10:一种抑菌凝胶,以重量百分比计,6%丙二醇,1%PEG-60氢化蓖麻油,0.8%卡波姆,0.8%角鲨烷,0.8%丙烯酰二甲基牛磺酸铵/VP共聚物,0.1%紫草素,5%中药复方提取物,0.4%甘草酸二钾,0.05%EDTA-2Na,余量为水。
实施例11:一种抑菌混悬液,以重量百分比计,5%泊洛沙姆,8%甘油,9%丙二醇,18%乙醇,0.05%紫草素,3%中药复方提取物,0.2%甘草酸二钾,0.02%EDTA-2Na,余量为水。
实施例12:一种抑菌混悬液,以重量百分比计,5%泊洛沙姆,8%甘油,9%丙二醇,18%乙醇,0.08%紫草素,4%中药复方提取物,0.2%甘草酸二钾,0.02%EDTA-2Na,余量为水。
实施例13:一种抑菌混悬液,以重量百分比计,5%泊洛沙姆,8%甘油,9%丙二醇,18%乙醇,0.10%紫草素,5%中药复方提取物,0.2%甘草酸二钾,0.02%EDTA-2Na,余量为水。
实施例14:一种抑菌混悬液,以重量百分比计,4%泊洛沙姆,8%甘油,8%丙二醇,15%乙醇,0.1%紫草素,5%中药复方提取物,0.1%甘草酸二钾,0.01%EDTA-2Na,余量为水。
实施例15:一种抑菌混悬液,以重量百分比计,8%泊洛沙姆,10%甘油,10%丙二醇,20%乙醇,0.1%紫草素,5%中药复方提取物,0.4%甘草酸二钾,0.05%EDTA-2Na,余量为水。
对比例1:无中药复方提取物和紫草素,其余同实施例1。
对比例2:无中药复方提取物和紫草素,其余同实施例6。

三、结果评价
3.1制备方法提取效果评价

由表1结果可以看出,本发明利用酶解法,对中药复方利用酶解,可以大幅提高中药复方提取物的提取率,提取条件温和,并且将中药复合物粉碎后,对粉末进行酶解,减少了酶解时间,减少了酶对活性成分的破坏作用。
3.2本发明抑菌效果评价
3.2.1检测方法:参照《GB 15979-2002》中给出的溶出性抗(抑)菌产品抑菌性能试验方法。
3.2.2测定上述实施例1-15及对比例1-3对大肠杆菌、金黄色葡萄球菌、白色念珠菌的抑菌率,具体抑菌率结果如下表1。

由表2可见,本发明中中药复方提取物和紫草素对抑菌效果具有显著的影响,两者组合可以很好的起到抑菌效果。含有抑菌组合物的实施例相比不含有抑菌组合物的对比例,其抑菌率明显提高,说明本发明的抑菌组合物应用在乳膏、凝胶及混悬液中,对大肠杆菌、金黄色葡萄球菌、白色念珠菌均具有良好的抑制作用。
3.3不同组分中药复方提取物对抑菌效果的影响
3.3.1为了进一步验证中药复方提取物中不同组分对抑菌效果的影响,本发明制备例1制备的中药复方提取物制备样品1、样品3和样品5、制备例2制备样品2、样品4和样品6。
样品1:一种抑菌乳膏,以重量百分比计,4%鲸蜡基葡萄糖苷,6%硬脂酸,2%单硬脂酸甘油酯,3%十六醇,3%棕榈酸异丙酯,4%丙二醇,0.10%紫草素,5%中药复方提取物,0.2%油溶性VE,0.2%甘草酸二钾,0.03%透明质酸,0.02%EDTA-2Na,余量为水。
样品2:一种抑菌乳膏,以重量百分比计,4%鲸蜡基葡萄糖苷,6%硬脂酸,2%单硬脂酸甘油酯,3%十六醇,3%棕榈酸异丙酯,4%丙二醇,0.10%紫草素,5%中药复方提取物,0.2%油溶性VE,0.2%甘草酸二钾,0.03%透明质酸,0.02%EDTA-2Na,余量为水。
样品3:一种抑菌凝胶,以重量百分比计,4%丙二醇,0.6%PEG-60氢化蓖麻油,0.5%卡波姆,0.4%角鲨烷,0.5%丙烯酰二甲基牛磺酸铵/VP共聚物,0.10%紫草素,5%中药复方提取物,0.2%甘草酸二钾,0.02%EDTA-2Na,余量为水。
样品4:一种抑菌凝胶,以重量百分比计,4%丙二醇,0.6%PEG-60氢化蓖麻油,0.5%卡波姆,0.4%角鲨烷,0.5%丙烯酰二甲基牛磺酸铵/VP共聚物,0.10%紫草素,5%中药复方提取物,0.2%甘草酸二钾,0.02%EDTA-2Na,余量为水。
样品5:一种抑菌混悬液,以重量百分比计,5%泊洛沙姆,8%甘油,9%丙二醇,18%乙醇,0.10%紫草素,5%中药复方提取物,0.2%甘草酸二钾,0.02%EDTA-2Na,余量为水。
样品6:一种抑菌混悬液,以重量百分比计,5%泊洛沙姆,8%甘油,9%丙二醇,18%乙醇,0.10%紫草素,5%中药复方提取物,0.2%甘草酸二钾,0.02%EDTA-2Na,余量为水。
3.3.2结果评价
将上述制备的样品抑菌效果与实施例部分制备的样品进行抑菌效果对比,样品1、样品2除了中药复方提取物不同,其余组分与实施例3相同;样品3、样品4除了中药复方提取物不同,其余组分与实施例8相同;样品5、样品6除了中药复方提取物不同,其余组分与实施例13相同。

将表3所得的结果与表2中的抑菌效果进行比较,可以发现,当甘草、枯芩、大黄、黄柏和苦参的重量比为15:25:15:25:25时,该中药复方提取物的对大肠杆菌、金黄色葡萄球菌和白色念珠菌抑菌效果均有所增加,取得了更好的抑菌效果。
此外,由以上结果可以看出,本发明的抑菌组合物可同时对上述三种菌起到很好的抑制效果,本发明的抑菌组合物活性成分从天然物质中提取,提取条件温和,提取率高,抑菌谱广,应用范围更广。
Claims (10)
1.一种中药复方提取物的制备方法,其特征在于,包括以下步骤:
(1)将包含以下重量份组分混合:5-15份甘草、15-25份枯芩、5-15份大黄、15-25份黄柏和15-25份苦参,将混合物粉碎打粉;
(2)加入蒸馏水浸泡,加入1500-2500U/g酶水解,在35-50℃水浴搅拌1-2h,过滤,将滤液浓缩、干燥,得中药复方提取物;
所述酶选自纤维素酶、果胶酶或蛋白酶中的一种或多种。
2.根据权利要求1所述的中药复方提取物的制备方法,其特征在于步骤(1)中,包含以下重量份组分:10-15份甘草、20-25份枯芩、10-15份大黄、20-25份黄柏和20-25份苦参。
3.一种由权利要求1-2所述任一中药复方提取物的制备方法制备的中药复方提取物。
4.一种抑菌组合物,其特征在于,包含权利要求3所述的中药复方提取物。
5.根据权利要求4所述的抑菌组合物,其特征在于,包含以下重量百分比组分:0.01-0.15%紫草素和2-6%中药复方提取物。
6.根据权利要求5所述的抑菌组合物,其特征在于,包含以下重量百分比组分:0.05-0.10%紫草素和3-5%中药复方提取物。
7.权利要求4-6所述的任一抑菌组合物在乳膏、凝胶或混悬液中的应用。
8.一种抑菌乳膏,其特征在于,包含以下重量百分比组分:2-5%鲸蜡基葡萄糖苷、4-10%硬脂酸、1-3%单硬脂酸甘油酯、1.5-4%十六醇、2-6%棕榈酸异丙酯、2-6%丙二醇、0.01-0.15%紫草素、2-6%中药复方提取物、0.1-0.3%VE、0.1-0.4%甘草酸二钾、0.01-0.05%透明质酸和0.01-0.05%EDTA-2Na。
9.一种抑菌凝胶,其特征在于,包含以下重量百分比组分:2-6%丙二醇、0.05-1%PEG-60氢化蓖麻油、0.2-0.8%卡波姆、0.2-0.8%角鲨烷、0.2-0.8%丙烯酰二甲基牛磺酸铵/VP共聚物、0.01-0.15%紫草素、2-6%中药复方提取物、0.1-0.4%甘草酸二钾和0.01-0.05%EDTA-2Na。
10.一种抑菌混悬液,其特征在于,包含以下重量百分比组分:4-8%泊洛沙姆、8-10%甘油、8-10%丙二醇、15-20%乙醇、0.01-0.15%紫草素、2-6%中药复方提取物、0.1-0.4%甘草酸二钾和0.01-0.05%EDTA-2Na。
