Abstract
本发明提供一种筛选获得的里氏木霉达宿主细胞,其保藏编号为CCTCC NO:M2016726。本发明的里氏木霉宿主细胞可用于表达一种或多种内源或外源蛋白。本发明再一个方面提供一种里氏木霉重组菌株,是将携带有目的基因的表达载体转化入上述里氏木霉宿主细胞获得的。本发明通过诱变获得的突变菌里氏木霉U11菌丝形态粗短,菌丝体致密且分枝更多,能使发酵菌液的粘度显著降低,达到降低搅拌速度又提高溶氧的目的,更有利于菌株的高密度发酵。里氏木霉U11可以作为一种新型宿主细胞,普遍应用于同源或异源基因的重组表达。

技术领域
本发明涉及基因工程微生物筛选技术领域,具体提供了一种新型木霉宿主细胞及其应用。
背景技术
酶作为生物催化剂可应用于多个领域,酶的产率、产量、品质和功能是酶应用于工业中的重要决定因素。酶的高效表达与多个因素相关:宿主的细胞生长特性、表达水平、胞内/胞外表达模式、翻译后修饰、活性蛋白等。为了实现酶的高效表达,酶表达系统的选择至关重要。重组酶已在多个宿主中成功表达,如大肠杆菌、芽孢杆菌、酵母、丝状真菌、乳酸菌等。由于外源基因或表达宿主性质与功能的多样性以及在细胞培养过程中不能合理监控其生理变化,目的蛋白基因在表达宿主中很难实现高效表达,酶的工业化生产受到限制。
丝状真菌可为表达宿主用于过量生产、分泌蛋白。丝状真菌是具有高效分泌蛋白潜力的真核表达系统,能对蛋白进行翻译后修饰等。随着生物技术的快速发展,传统发酵与其他相关产业开始逐渐重视丝状真菌表达宿主的研究与应用。用于蛋白表达的丝状真菌包括曲霉属、木霉属、青霉属、根霉属、镰刀菌属等。
丝状真菌表达的异源蛋白主要分为2类:工业酶制剂和高等生物的基因产物(蛋白药物等)。丝状真菌的表达水平除了少数蛋白产量偏高外,其他异源蛋白的产量远达不到高水平。为此可以在转录、翻译、翻译后修饰加工、分泌和胞外降解等不同水平进行改造,提高蛋白的表达量。
木霉属表达宿主主要包括:里氏木霉、深绿木霉、绿木霉。在表达系统里氏木霉方面的研究相对较多,但在深绿木霉和绿木霉表达系统方面的研究尚处在起步阶段。
里氏木霉是自然界中广泛分布的腐生性丝状真菌,具有极好的合成蛋白和分泌蛋白的能力,某些高产菌株胞外纤维素酶的主要成分——外切葡聚糖纤维二糖水解酶工的产量可达20~40g/L,此启动子为强启动子。里氏木霉作为微生物具有结构简单、生长迅速、操作简便的优点,同时作为低等真核生物,又具有真核基因表达机制和存在真核蛋白质翻译后修饰加工装置,具有与哺乳动物系统相似的蛋白修饰性能。
里氏木霉培养简单,低廉,发酵罐发酵技术成熟,不易染菌,目前发酵单位可以达到吨级,在工业上用于生产同源或异源的纤维素酶或其它的蛋白质有很多的优势。通过诱变,筛选等系统性进化,开发越来越适合于商品酶生产的木霉宿主是研究的重点。
发明内容
本发明的目的是提供一种筛选获得的里氏木霉达宿主细胞,并将其用于重组表达外源基因;所获得的宿主细胞对外源基因的表达量远高于野生型宿主细胞,应用前景广阔。
本发明首先提供一种里氏木霉宿主细胞,该宿主细胞命名为里氏木霉U11(Trichoderma reesei U11),已于2016年12月7日保藏于中国武汉武汉大学的中国典型培养物保藏中心,保藏编号为CCTCC NO:M2016726。
本发明的里氏木霉宿主细胞可用于有效表达一种或多种内源或外源蛋白。
所述的蛋白选自半纤维素酶、过氧化物酶、蛋白酶、纤维素酶、木聚糖酶、脂肪酶、磷脂酶、酯酶、角质酶、果胶酶、角蛋白酶、还原酶、氧化酶、酚氧化酶、脂加氧酶、木质素酶、支链淀粉酶、鞣酸酶、戊聚糖酶、malanase、β-葡聚糖酶、阿拉伯糖苷酶、透明质酸酶、软骨素酶、漆酶、淀粉酶、葡糖淀粉酶中的任一种或几种。

作为实施例的优选,所述的目的基因为纤维素酶基因或葡萄糖转苷酶基因;
所述纤维素酶基因,其一种核苷酸序列为SEQ ID NO:1,编码的氨基酸序列为SEQID NO:2。
所述的葡萄糖转苷酶基因,其一种核苷酸序列为SEQ ID NO:3,编码的氨基酸序列为SEQ ID NO:4。
所述表达载体,作为实施例的一种具体选择,为PC2G载体。
上述里氏木霉重组菌株用于生产纤维素酶或葡萄糖转苷酶。
本发明通过诱变获得的突变菌里氏木霉U11菌丝形态粗短,菌丝体致密且分枝更多,能使发酵菌液的粘度显著降低,达到降低搅拌速度又提高溶氧的目的,更有利于菌株的高密度发酵。里氏木霉U11可以作为一种新型宿主细胞,普遍应用于内源或异源基因的重组表达。以突变菌里氏木霉U11为宿主细胞构建得到的重组菌株里氏木霉能高效表达NCE5和葡萄糖转苷酶基因,其发酵酶活分别高达412U/mL和3550U/mL,比对照菌株分别提高了43.1%和50.4%。此外,申请人还将糖化酶、葡萄糖氧化酶、果胶酶、淀粉酶等分别在突变菌宿主U11中进行重组表达。与对照宿主里氏木霉U相比,突变菌宿主U11对上述基因的表达量均提高了40-60%,取得了意料不到的效果。

具体实施方式
下面结合实例对本发明的方法做进一步说明。在实施例中未注明具体条件的实验方法,通常可按常规条件,如J.萨姆布鲁克(Sambrook)等编写的《分子克隆实验指南》中所述的条件,或按照制造厂商所建议的条件运行。本领域相关的技术人员可以借助实施例更好地理解和掌握本发明。
实施例1里氏木霉表达宿主的紫外诱变筛选
申请人通过基因敲除的方法,将野生型里氏木霉(Trichoderma reesei)中的纤维素酶基因CBH1、CBH2、EG1和EG2四个基因敲除后,获得一株自身不分泌纤维素酶的里氏木霉菌株,命名为里氏木霉U(Trichoderma reesei U)。
将里氏木霉U接种于新鲜的PDA平板,30℃培养5-7天。待菌落表面变白,产生大量孢子时,吸取5ml无菌水洗脱,获得孢子液,离心后用无菌水重悬,用血球计数板计数,使孢子浓度约为5×107个/ml。取一个转子放入90mm的无菌培养皿,加入10ml稀释好的孢子悬液,在磁力搅拌器上搅拌使孢子液处于均匀状态。在无菌超净工作台中,用功率为9w的紫外灯于垂直距离20cm的上方照射,分别照射60s、90s、120s、150s、180s。将照射后的孢子液稀释10000倍,取其中100ul均匀涂布PDA平板,30℃培养2-3d后计数,以未被紫外照射的孢子液为对照,计算致死率。结果显示,紫外照射120s时,里氏木霉孢子的致死率为90%,因此选取该照射时间进行后续诱变实验。
诱变筛选:取一个转子放入90mm的无菌培养皿中,加入10ml稀释好的孢子悬液(浓度为5×107个/ml)。在磁力搅拌器上搅拌使孢子液处于均匀状态。在无菌超净工作台中,用功率为9w的紫外灯于垂直距离20cm的上方照射,照射120s,然后在黑暗条件下放置30min。将孢子悬液稀释10000倍,取其中100ul均匀涂布于PDA平板,30℃培养2-3d,同时以出发菌里氏木霉U为对照组,采用同样方法涂布PDA平板,30℃培养2-3d。
申请人共涂布了150个PDA平板,每个平板长出约50个菌落。通过观察菌落形态,并与出发菌里氏木霉U进行比较,挑选出菌落形态发生明显变异的突变体203个,分别接种到PDA复筛平板(每个平板均匀接种12个突变体),30℃培养2-3d。
最终,申请人筛选得到38株菌落形态明显变小,菌丝体致密且分枝更多的里氏木霉突变菌。这些突变菌在放大发酵过程中具有菌丝体粗短,溶氧水平高,发酵液黏度低等优点。
实施例2里氏木霉表达宿主的尿嘧啶营养缺陷型筛选
2.1原理:
5-氟乳清酸可以诱导菌体缺失尿嘧啶核苷酸合成途径中的乳清核苷酸转移酶或乳清苷单磷酸脱羧酶,从而使5-氟乳清酸无法形成有毒的物质5-氟尿嘧啶核苷酸,从而产生了对5-氟乳清酸的抗性,其嘧啶核苷酸营养可以通过向培养基中添加尿嘧啶进行补充,因此利用5-氟乳清酸诱导形成的尿嘧啶营养缺陷型菌株可以在含有5-氟乳清酸和尿嘧啶的培养基中生长;而野生型菌株因不具备对5-氟乳清酸的抗性,无法在含有5-氟乳清酸的培养条件下生长。因此常用5-氟乳清酸来筛选尿嘧啶缺陷型的突变株。
2.2筛选方法
分别将实施例1筛选到的38株里氏木霉突变菌的孢子用浓度为0.1%的吐温-20溶液稀释至约1×107个/mL;然后将孢子悬液均匀涂布于含有1.5g/mL 5-氟乳清酸和1.87g/mL尿嘧啶核苷(Uridine)的基本固体培养基(2%葡萄糖,0.5%(NH4)2SO4,1.5%KH2PO4,0.06%MgSO4,0.06%CaCl2,1.5%琼脂)平板上,每一平板涂布约1×106个孢子,避光30℃培养7d以上。结果显示,每种突变菌的平板上都有不同数量的菌落长出,说明这些菌落有可能是对应突变菌的尿嘧啶缺陷型菌株。
分别挑取上述每种突变菌对应平板上长出的菌落,将每个菌落分别涂布于基本培养基平板和含有1.87g/mL Uridine的基本培养基平板进行验证。真正的尿嘧啶缺陷型菌株只能在含有Uridine的基本培养基平板上生长,而在缺少Uridine的基本培养基平板上无法生长。最终,申请人在实施例1中通过紫外诱变获得的38株里氏木霉突变菌中,每株突变菌都对应筛选到1株生长状态相对最好的尿嘧啶缺陷型菌株,共38株,分别命名为里氏木霉U1,U2,U3,U4,……,U38。
为了进一步确定上述38株尿嘧啶缺陷型突变菌株的稳定性,分别将其用液体基本培养基(2%葡萄糖,0.5%(NH4)2SO4,1.5%KH2PO4,0.06%MgSO4,0.06%CaCl2)振荡培养7天,观察其能否生长,重复3次试验。实验结果显示,38株尿嘧啶缺陷型突变菌株在缺少Uridine的液体基本培养基中均无法生长。
实施例3中性纤维素酶基因NCE5在里氏木霉突变菌中的表达
为了验证里氏木霉U1,U2,U3,U4,……,U38对目标基因的表达效率,申请人选择将来源于里氏木霉的中性纤维素酶基因NCE5(其核苷酸序列为SEQ ID NO:1,编码氨基酸序列为SEQ ID NO:2)分别在野生型里氏木霉U和上述突变菌株中进行表达。
3.1提取里氏木霉总基因组DNA
将里氏木霉U1接种摇瓶培养基过夜培养,取适量菌体置于离心管中,13000rpm离心5min,弃上清;加入400μl抽提缓冲液(100mM TrisHCl,100mM EDTA,250mM NaCl,1%SDS);然后加100mg石英砂或玻璃珠、在珠打仪剧烈振荡2min左右;水浴锅65℃水浴20min后加入200μl 10M NH4AC冰浴10min;13000rpm离心10min取上清,然后加入2倍体积的无水乙醇,-20℃放置30min;13000rpm离心10min弃上清,用70%乙醇洗涤2次;晾干,加入适量水溶解于-20℃保存。
3.2基因克隆
以3.1中提取的基因组总DNA为模板,利用引物anl-F和anl-R(TCTAGAGGAGCGGCAGTCCGGCAGCGGCC和AGTTAGTTAGCACTCAAGGG A)进行PCR扩增。PCR扩增条件为95℃4min;94℃30S,59℃40S,72℃1min 30个循环;72℃7min。利用凝胶回收试剂盒回收PCR扩增产物。
3.3测序分析
将3.2中回收的扩增产物连接到PC2G载体,获得克隆载体NCE5-PC2G质粒并送至北京华大基因研究中心进行测序分析。构建好的载体图谱如图1所示。测序结果,扩增产物的基因序列为SEQ ID NO:1,其编码氨基酸序列为SEQ ID NO:2。多个克隆的结果证明没有发生扩增错误。
3.4转化与筛选
3.4.1原生质体制备
接种实施例2获得的里氏木霉突变体U1菌丝于PDA平板上生长7天;切取直径约3cm的菌落置于约60mlYEG(0.5%酵母粉、1%葡萄糖、0.1%Uridine)的液体培养基中,30℃、200rpm振荡培养过夜;多层纱布过滤收集菌丝;将菌丝置于盛有20ml裂解酶液(SigmaL1412)酶解2小时;取出酶解液,加入0.7M NaCl溶液,轻轻摇晃,倒于三层灭菌擦镜纸过滤,收集滤液,3000rpm,离心10min;弃上清,加10-20ml STC液(20%蔗糖,50mM Tris-Cl,50mMCaCl2)悬浮,3000rpm,离心10min;加适量STC悬浮分装(200μl/管,108个/ml)。
3.4.2转化与验证
取10μg NCE5-PC2GDNA加入到200μl原生质体中,接着加入50μl 25%PEG轻轻混匀,室温静置25min;然后分2-3次再加2ml 25%PEG,轻轻混匀,室温静置25min,把原生质体加到50ml左右熔化后冷却至45-55℃的上层半固体培养基(0.1%MgSO4,1%KH2PO4,0.6%(NH4)2SO4,1%葡萄糖,18.3%山梨醇,0.35%琼脂糖),轻轻混匀后倒入下层基础培养基平板(2%葡萄糖,0.5%(NH4)2SO4,1.5%KH2PO4,0.06%MgSO4,0.06%CaCl2,1.5%琼脂),30℃黑暗培养数天至转化子长出。通过摇瓶发酵,选出纤维素酶表达量最高的阳性转化子,命名为里氏木霉U1-NCE5。
采用上述同样方法,分别以里氏木霉突变菌株U2,U3,U4,……,U38为宿主细胞,构建得到重组表达中性纤维素酶NCE5的里氏木霉重组菌株,对应选出表达量最高的阳性转化子,分别命名为里氏木霉U2-NCE5,U3-NCE5,……,U38-NCE5。
同时,以出发菌株里氏木霉U作为对照宿主,采用上述同样方法,构建得到重组表达中性纤维素酶NCE5的里氏木霉重组菌株,对应选出表达量最高的阳性转化子,命名为里氏木霉U-NCE5,作为对照菌株。
实施例4发酵验证
将上述构建的对照菌株里氏木霉U-NCE5,与以突变菌株U1,U2,U3,……,U38为宿主细胞构建得到的重组菌株里氏木霉U1-NCE5,U2-NCE5,……,U38-NCE5分别接种于摇瓶种子培养基(葡萄糖10-30g/L,土豆100-200g/L),30℃,200rpm摇床培养24h之后,将发酵液转入二级种子培养基,继续培养24小时后转入20L发酵罐(培养基为:葡萄糖30-50g/L,乳糖2.0-10g/L,玉米浆20-50g/L,硫酸铵10-30g/L,硫酸镁5-10g/L,磷酸二氢钾15-30g/L),温度控制在30±1℃,pH值控制在5.0±0.2,在发酵罐培养24之后,开始补加液糖诱导菌体产酶,发酵时间约为160h到180h。在发酵过程中,分别取样在显微镜下观察各株菌的菌丝形态。发酵结束后,分别取发酵上清液测定其中的纤维素酶酶活。
(1)酶活测定方法
在50℃、pH值为4.8(中性为pH6.0)的条件下,每分钟从浓度为5mg/ml的羟甲基纤维素钠溶液中降解释放1μmol还原糖所需要的酶量为一个酶活力单位U,还原糖以葡萄糖等量。
取三支试管各加入0.5mL CMC底物,与待测酶液一起50℃水浴预热5min。在第一、二试管中各加入0.5mL待测液,并计时,50℃水浴中反应15min。反应完后在三支试管中各加入1.5mLDNS试剂,并在第三支试管总补加0.5mL的待测酶液。取出并摇匀三支试管后,在沸水浴中反应5min。迅速冷却至室温,用水定到5.0mL。以第三支试管试液为对照在540nm波长条件下测第一、二试管试液的吸光度,吸光度在0.25-0.35之间为宜。待测酶液反应液的吸光度与水平控制酶液反应液吸光度之差的绝对值不超过0.015。
酶活X=(葡萄糖等量值/180/15/0.5)×n
其中:X——酶活力单位,IU/g(mL);
180——葡萄糖从微克换算成微摩尔;
15——待测液与底物的反应时间;
0.5——加入反应的待测酶液量;
n——稀释倍数;
(2)酶活测定结果
以出发菌里氏木霉U作为对照宿主构建得到的对照菌株里氏木霉U-NCE5,其发酵上清液酶活为288U/mL,而以突变菌株U1,U2,U3,……,U38为宿主细胞构建得到的重组菌株里氏木霉U1-NCE5,U2-NCE5,……,U38-NCE5,其发酵酶活约为362~412U/mL,说明纤维素酶基因NCE5在出发菌里氏木霉U以及突变菌株U1,U2,U3,……,U38中均得到有效表达,且在突变菌中的表达量明显高于出发菌。其中,以突变菌U11为宿主细胞构建得到的重组菌株里氏木霉U11-NCE5发酵酶活最高,为412U/mL,比对照菌株提高了43.1%,取得了意料不到的技术效果。
除了中性纤维素酶基因NCE5外,申请人还将纤维素酶基因HGD45、EGⅡ,以及葡萄糖氧化酶等基因分别在上述突变菌宿主U1,U2,U3,……,U38中进行重组表达。与对照宿主里氏木霉U相比,突变菌宿主对上述基因的表达量均得到明显提高,其中尤以突变菌里氏木霉U11的表达量最高。
显微镜观察结果如图2所示,与出发菌相比,突变菌里氏木霉U11的菌丝形态粗短,菌丝体致密且分枝更多,能使发酵菌液的粘度显著降低,达到降低搅拌速度又提高溶氧的目的,更有利于菌株的高密度发酵。
申请人已于2016年12月7日将上述突变菌里氏木霉U11(Trichoderma reeseiU11)保藏于中国武汉武汉大学的中国典型培养物保藏中心,保藏编号为CCTCC NO:M2016726。
实施例5葡萄糖转苷酶基因在里氏木霉U11中的高效表达
根据公共基因数据库中基因序列的记载,申请人人工合成了葡萄糖转苷酶基因,其核苷酸序列为SEQ ID NO:3,其编码的氨基酸序列为SEQ ID NO:4。申请人将该葡萄糖转苷酶基因命名为XyL。
将葡萄糖转苷酶基因片段与PTG载体同时进行双酶切,回收酶切产物,22℃连接过夜,转化大肠杆菌感受态细胞,于LA平板37℃培养过夜。
以PTG通用引物,挑取转化子做菌落PCR,选取阳性转化子送华大基因测序,测序结果显示,无点突变,表达载体构建成功。将表达载体命名为PTG-XyL。
接种里氏木霉U11菌丝于PDA平板上生长7天;切取直径约3cm的菌落置于约60mlYEG(0.5%酵母粉、1%葡萄糖、0.1%Uridine)的液体培养基中,30℃、200rpm振荡培养过夜;多层纱布过滤收集菌丝;将菌丝置于盛有20ml裂解酶液(Sigma L1412)酶解2小时;取出酶解液,加入0.7M NaCl溶液,轻轻摇晃,倒于三层灭菌擦镜纸过滤,收集滤液,3000rpm,离心10min;弃上清,加10-20ml STC液(20%蔗糖,50mM Tris-Cl,50mM CaCl2)悬浮,3000rpm,离心10min;加适量STC悬浮分装(200μl/管,108个/ml)。
取10μg XyL-PC2G DNA加入到200μl原生质体中,接着加入50μl 25%PEG轻轻混匀,室温静置25min;然后分2-3次再加2ml 25%PEG,轻轻混匀,室温静置25min,把原生质体加到50ml左右熔化后冷却至45-55℃的上层半固体培养基(0.1%MgSO4,1%KH2PO4,0.6%(NH4)2SO4,1%葡萄糖,18.3%山梨醇,0.35%琼脂糖),轻轻混匀后倒入下层基础培养基平板(2%葡萄糖,0.5%(NH4)2SO4,1.5%KH2PO4,0.06%MgSO4,0.06%CaCl2,1.5%琼脂),30℃黑暗培养数天至转化子长出。通过摇瓶发酵,选出木聚糖酶表达量最高的阳性转化子,命名为里氏木霉U11-XyL。
同时,以出发菌株里氏木霉U作为对照宿主,采用上述同样方法,构建得到重组表达木聚糖酶XyL的里氏木霉重组菌株,对应选出表达量最高的阳性转化子,命名为里氏木霉U-XyL,作为对照菌株。
将上述构建的对照菌株里氏木霉U-XyL和突变菌株里氏木霉U11-XyL为宿主细胞构建得到的重组菌株里氏木霉U-XyL和里氏木霉U11-XyL分别接种于摇瓶种子培养基(葡萄糖25g/L,土豆150g/L),30℃,200rpm摇床培养24h之后,将发酵液转入二级种子培养基,继续培养24小时后转入20L发酵罐(培养基为:葡萄糖50g/L,乳糖2.0g/L,玉米浆30g/L,硫酸铵20g/L,硫酸镁8g/L,磷酸二氢钾20g/L),温度控制在30±1℃,pH值控制在5.0±0.2,在发酵罐培养24之后,开始补加液糖诱导菌体产酶,发酵时间约为160h到180h。发酵结束后,分别取发酵上清液测定其中的木聚糖酶酶活。
(1)葡萄糖转苷酶酶活单位的定义
在50℃、pH值为4.8的条件下,每分钟从浓度为4mmol/L的对硝基苯酚-α-D-葡萄糖苷溶液中释放1nmol对硝基苯酚所需要的酶量即为一个酶活力单位U。
(2)标准曲线绘制
5mmol/L对硝基苯酚-α-D-葡萄糖苷底物溶液:秤取0.0753g对硝基苯酚-α-D-葡萄糖苷,精确到0.0001g,缓慢加入0.05mol/L pH=4.8的柠檬酸缓冲液至接近50ml,磁力搅拌约10min,用2mol/L的柠檬酸或者氢氧化钠调相应的pH值,最后定容至50ml,现用现配。
秤取对硝基苯酚0.0696g,用0.05mol/L pH=4.8的柠檬酸缓冲液定容至100ml棕色容量瓶中,磁力搅拌30min至完全溶解即得5mmol/L对硝基苯酚溶液。
将5mmol/L对硝基苯酚溶液精确稀释10倍即得对硝基苯酚标准液。将对硝基苯酚标准液分别稀释2、4、6、8、10、12、16倍。
取0.5ml上述对硝基苯酚稀释液(空白对照取缓冲液),加入2ml碳酸钠溶液,加入0.5ml5mmol/L对硝基苯酚-α-D-葡萄糖苷底物溶液,混合均匀,以空白对照调零,以在410nm测定吸光值。以体系中对硝基苯酚含量为横坐标X,吸光值为纵坐标Y,绘制标准曲线Y=kX+b。
(3)酶活测定
取适量底物,50℃预热5min;
取4支15*150试管(1支空白管,3支样品管),分别向4支管中准确加入已稀释好的酶液0.5ml;
将4支试管同时置于50℃水浴中,预热2min;
准确向样品试管中加入0.5ml5mmol/L对硝基苯酚-α-D-葡萄糖苷底物溶液底物溶液,准确计时15min;
迅速向各管中加入2ml碳酸钠溶液,于空白管中加入底物溶液0.5ml,摇匀;
以空白管调零,在分光光度计波长410nm下,用10mm比色杯分别测量。
3样品管中的吸光值取平均值,通过查标准曲线求出对硝基苯酚的含量。
酶活力计算公式:A=X×1/0.5×n/15
A——α-葡萄糖转苷酶酶活力,u/ml;
X——吸光值在标准曲线上查得的对硝基苯酚含量,μmol;
1/0.5——换算成酶液1ml;
n——酶样的稀释倍数;
15——时间换算系数。
结果显示,以出发菌里氏木霉U作为对照宿主构建得到的突变菌株里氏木霉U-XyL,其发酵上清液酶活为2360U/mL,以突变菌U11为宿主细胞构建得到的重组菌株里氏木霉U11-XyL发酵酶活高达3550U/mL,比对照菌株提高了50.4%,取得了意料不到的技术效果。
除了中性纤维素酶基因NCE5和葡萄糖转苷酶外,申请人还将糖化酶、葡萄糖氧化酶、果胶酶、淀粉酶等分别在突变菌宿主U11中进行重组表达。与对照宿主里氏木霉U相比,突变菌宿主U11对上述基因的表达量均提高了40-60%,效果显著。
综上,本发明通过诱变获得的突变菌里氏木霉U11可以作为一种新型宿主细胞,普遍应用于内源或异源基因的重组表达,有利于提高蛋白的产量,降低生产成本。

Claims (6)
1.一种里氏木霉(Trichoderma reesei)U11宿主细胞,其特征在于,所述的宿主细胞的保藏编号为CCTCC NO:M2016726。
2.权利要求1所述的里氏木霉宿主细胞在表达内源或外源蛋白中的应用。
3.如权利要求2所述的应用,其特征在于,所述的蛋白为纤维素酶、葡萄糖转苷酶、糖化酶、葡萄糖氧化酶、果胶酶或淀粉酶中的任一种。
4.一种里氏木霉重组菌株,其特征在于,所述的里氏木霉重组菌株是将携带有目的基因的表达载体转化入权利要求1所述的里氏木霉宿主细胞中获得的。
5.如权利要求4所述的里氏木霉重组菌株,其特征在于,所述的目的基因为编码纤维素酶、葡萄糖转苷酶、糖化酶、葡萄糖氧化酶、果胶酶或淀粉酶中的任一种酶的基因。
6.权利要求5所述的里氏木霉重组菌株在生产纤维素酶、葡萄糖转苷酶、糖化酶、葡萄糖氧化酶、果胶酶或淀粉酶中的应用。
