Abstract
一种猴头菇养胃颗粒及其制备方法,属于猴头菇保健食品技术领域。本发明针对目前市场上具有护胃、养胃的猴头菇相关产品少的问题,提供了一种猴头菇养胃颗粒及其制备方法:具体是将原料猴头菇10重量份、黄芪2~4重量份、山楂1~3重量份和党参2~4重量份按比例混合后进行水煮提取,所述原料混合物与水的料液比为1:(8~10),所述提取温度为80~100℃,提取时间为0.5~2h,提取次数为1~3次;然后经过滤、浓缩、醇沉,获得的沉淀按比例加水溶解后,再按比例加入乳糖20~50重量份、玉米糊精5~15重量份和淀粉0.5~2重量份,制得猴头菇养胃颗粒。本发明适用于猴头菇的加工与利用。

技术领域
本发明属于猴头菇保健食品技术领域,具体涉及一种猴头菇养胃颗粒及其制备方法。
背景技术
猴头菇(Hericium erinaceus)是中国传统的名贵菜肴,是四大名菜(猴头、熊掌、燕窝、鱼翅)之一。其菌肉口感鲜嫩、味道香醇,是鲜美无比的山珍,有“素中荤”之称。猴头菇属于齿菌科菌类,其形体远看像金丝猴头。研究表明,猴头菇中氨基酸含量十分丰富,种类有17种,其中人体所需的占8种,是菌类中的佼佼者。干品猴头菇每百克含蛋白质26.3克,碳水化合物44.9克,脂肪4.2克,细纤维6.4克,磷850毫克,铁18毫克,钙2毫克,核黄素1.89毫克,硫胺素(B1)0.89毫克,胡萝卜素0.01毫克,热量323千卡,是名副其实的高蛋白、低脂肪食品。现代医学研究表明,猴头菇可用于治疗消化不良、胃痛、胃胀及神经衰弱、失眠等病症。
猴头菇不仅可食用,也可作为药材。猴头菇能够养胃,增进食欲,增强胃黏膜屏障机能,大大降低了消化不良、胃溃疡及慢性胃炎的发病率。现代医学研究表明,猴头菇多糖具有提高免疫力、抗衰老、抗肿瘤、降血脂、预防糖尿病等多种作用。猴头菇可以提高淋巴细胞转化率,增加白细胞数量,增强人体对疾病的免疫能力。此外,猴头菇对改善神经衰弱也具有良好效果,对皮肤、肌肉癌亦有明显抗癌功效。猴头菇中含有丰富的牛磺酸和铁元素,有利于缓解体力疲劳和防止贫血,对大脑发育和预防阿尔茨海默症具有一定的疗效。猴头菇多糖是猴头菇的主要有效成分之一,具有多种生理活性,如调节免疫、抗肿瘤、降血糖、保护胃黏膜等。
发明内容
本发明针对目前市场上具有护胃、养胃的猴头菇相关产品少的问题,提供了一种猴头菇养胃颗粒,所述猴头菇养胃颗粒主要是由下述原料制成的:猴头菇、黄芪、山楂和党参。
优选地,所述猴头菇养胃颗粒由以下重量份的原料制成:猴头菇10重量份、黄芪2~4重量份、山楂1~3重量份、党参2~4重量份、乳糖20~50重量份、玉米糊精5~15重量份和淀粉0.5~2重量份。
更优选地,所述猴头菇养胃颗粒由以下重量份的原料制成:猴头菇10重量份、黄芪3重量份、山楂2重量份、党参3重量份、乳糖40重量份、玉米糊精10重量份和淀粉1重量份。
所述淀粉优选为玉米淀粉。
上述猴头菇养胃颗粒的制备方法,包括如下步骤:
(1)清洗:按重量份比例,挑选原料猴头菇、黄芪、山楂和党参,清洗干净;
(2)提取:清洗后的原料,混合后加水浸泡3-5h,然后进行水煮提取,所述原料混合物与水的料液比为1:(8~10),所述提取温度为80~100℃,提取时间为0.5~2h,提取次数为1~3次;
(3)过滤:过滤获得提取液;
(4)制粒:将提取液浓缩后进行醇沉,获得的沉淀按比例加水溶解后,再按重量份比例加入乳糖、玉米糊精和淀粉,制得猴头菇养胃颗粒。
优选地,步骤(2)所述料液比为1:8,提取温度为80℃,提取时间为1h,提取次数为3次。
优选地,步骤(3)所述过滤是指利用3-4层80目的纱布进行过滤。
优选地,步骤(4)所述浓缩温度为60-80℃,浓缩比例为步骤(1)中每10g的猴头菇对应步骤(4)中所述浓缩体积为20ml。
优选地,步骤(4)所述醇沉是指向浓缩后的提取液中加入体积分数为95%的乙醇至终浓度为80%,然后经3000rpm/min离心10min获得沉淀。
优选地,步骤(4)所述加水溶解,其加水的比例为步骤(1)中每10重量份的猴头菇对应步骤(4)中加水的量为5重量份。
优选地,步骤(4)所述猴头菇养胃颗粒粒径大小为10~40目。
有益效果
本发明的主要创新点在于将猴头菇与黄芪、山楂、党参配伍使用。中医认为,猴头菇性平味甘,利五脏,助消化,能够行气消食,健脾开胃,可用于食积不消,脘腹胀痛,脾虚食少,失眠多梦等症状的治疗。黄芪为豆科黄芪属植物黄芪的根,性甘微温,归肺、脾经。具有治疗表虚自汗、阳气虚弱、疮疡窥破、气虚衰弱等功效。山楂是蔷薇科山楂树植物山楂的果实,味酸,性甘微温,归脾、胃、肝经。具有消食健胃、行气散淤、化浊降脂的功效。主治肉食积滞,胃脘胀满,泻痢腹痛,产后淤阻,心腹刺痛,高脂血症等。党参是桔梗科党参属植物党参、素花党参、川党参、管花党参、球花党参、徽毛党参的根,味甘性平。有补中益气、止渴、健脾益肺的功效。可用于治疗脾肺气虚,气血两亏,体倦无力,食少,口渴,久泻,脱肛等病症。
本发明所述的猴头菇养胃颗粒的辅料选择了乳糖、玉米糊精和淀粉,上述三种物质的水溶性较好,有利于制粒,此外,能够起到稀释主要原料猴头菇、黄芪、山楂、党参混合物浓度的作用,并调节口味,微晶纤维素类的物质也可用作本发明的辅料,制成的猴头菇养胃颗粒冲泡效果较差。
本发明制备的猴头菇养胃颗粒,能够明显减轻因急性乙醇刺激导致的胃组织肿胀程度,对急性酒精胃黏膜损伤有明显的保护作用。
根据产品配方原料特性及功效成分的种类,结合对保健食品剂型的调查结果,本发明最终选择剂型为颗粒剂。与国内现有猴头菇加工方式比较,本发明制备的养胃颗粒服用方便,便于携带,产品稳定,口感好,溶出和吸收速度较快,显效迅速。

具体实施方式
下述实施方式所用猴头菇采购于吉林省长春市立新参茸特产店,黄芪,山楂,党参采购于同仁堂药店。
高效旋转式颗粒机:厂家江阴市呈祥制药设备有限公司,型号ZL150G。
其他材料或试剂均通过商业化途径购买获得。
实施例1.猴头菇养胃颗粒的制备方法。
1)清洗:将猴头菇1000g,黄芪200g,山楂100g,党参200g,清洗干净。
2)提取:按照原料与水的料液比1:8的比例,添加12L的饮用水,浸泡3小时,加热到80℃进行提取,提取时间为0.5h,提取1次。
3)过滤:将上述提取混合物经3层80目的纱布过滤,获得提取液。
4)制粒:将过滤获得的提取液经60℃加热浓缩至2L,加体积分数为95%的乙醇至酒精计检测乙醇终浓度为80%,经3000rpm/min离心10min,除去杂质,获得的沉淀加500mL水溶解,加入乳糖2000g、玉米糊精500g、玉米淀粉50g,利用ZL150G颗粒机进行制粒,制得10目大小的猴头菇养胃颗粒,紫外灭菌包装。
实施例2.猴头菇养胃颗粒的制备方法。
1)清洗:将猴头菇1000g,黄芪400g,山楂300g,党参400g,清洗干净。
2)提取:按照原料与水的料液比1:10的比例,添加21L的饮用水,浸泡5小时,加热到80℃进行提取,提取时间为2h,提取3次。
3)提取:将上述提取混合物经4层80目的纱布过滤,获得提取液。
4)过滤:获得的提取液经80℃加热浓缩至2L,加体积分数为95%的乙醇至酒精计检测乙醇终浓度为80%,经3000rpm/min离心10min,除去杂质,获得的沉淀加500mL水溶解,加入乳糖5000g、玉米糊精1500g、玉米淀粉200g,利用ZL150G颗粒机进行制粒,制得40目大小的猴头菇养胃颗粒,紫外灭菌包装。
实施例3.考察猴头菇养胃颗粒的最优制备方法。
本实施例从四因素三水平进行正交试验,以总多糖得率为考察指标,优化猴头多糖提取方法。
各组中原料用量如下:猴头菇100g,黄芪30g,山楂20g,党参30g。
各组提取时间,提取温度,料液比,提取次数如表1所示。

提取方法优化过程中,最终将不同组提取液浓缩到200mL,添加体积分数为95%乙醇至乙醇终浓度为80%乙醇,经3000rpm/min离心10min进行沉淀,经现有的冷冻干燥机,采用-45℃进行冷冻干燥后获得猴头菇多糖,测定猴头菇多糖质量,以猴头菇干重量计,计算多糖得率。从表2中可以看出,影响猴头菇多糖提取工艺的因素大小顺序为提取时间(A)>料液比(C)>提取温度(B)>提取次数(D),提取时间为主要影响因素,其次为料液比。实验结果表面,最佳提取工艺为A2B1C1D3,即将猴头菇用料液比为1:8(kg:L)条件下,80℃提取1h,提取3次。该提取方法能够充分提取猴头菇多糖,相比之下能量消耗小,工艺简单,可操作性强。
表2正交试验安排及试验结果
黄芪、山楂和党参中的主要成分为小分子炔苷类化合物和大分子多糖类物质,因此选取80℃以上热水煮提的方法进行提取。分别对按料液比1:8(kg:L)的比例,检测煮提0.5h,1h,2h时的固形物得率。结果表明,1h和2h提取时的提取物产率无明显差异,并且煮提两次后大部分提取物都能被提取出来。为了在工业化生产过程中节约成本和劳动力,对黄芪和党参的提取方法和猴头菇多糖提取方法保持一致,即料液比1:8(kg:L),80℃煮提1h,共煮提3次。
实施例4.猴头菇养胃颗粒功效性评价。
根据实施例3所述的最佳提取条件,制备猴头菇养胃颗粒:
1)清洗:将猴头菇1000g、黄芪300g、山楂200g、党参300g清洗干净。
2)提取:按照原料与水的料液比1:8的比例,添加14.4L的饮用水,浸泡4小时,加热到80℃进行提取,提取时间为1h,提取3次。
3)过滤:将上述提取混合物经4层80目的纱布过滤,获得提取液。
4)过滤获得的提取液经70℃加热浓缩至2L,加体积分数为95%的乙醇至酒精计检测乙醇终浓度为80%,经3000rpm/min离心10min,除去杂质,获得的沉淀加500mL水溶解,加入乳糖4000g、玉米糊精1000g和玉米淀粉100g,利用颗粒机进行制粒,制得30目大小的猴头菇养胃颗粒,紫外灭菌包装。
根据国家相关政策法规规定,动物功能实验按照《保健食品检验与评价技术规范》(2003版)之“对胃黏膜损伤有辅助保护功能评价方法”,对猴头养胃颗粒进行胃黏膜保护的实验。
(1)实验材料
样品:猴头养胃颗粒。
实验动物:选用辽宁长生生物技术股份有限公司繁殖的清洁级SD健康雄性大鼠40只,体重为180.7~194.8g,生产许可证SCXK(辽)2015-0001,合格编号211002300023526,实验动物在温度20~25℃、相对湿度为40%~70%的环境中饲养。
(2)实验方法
受试样品的人体推荐摄入量为10g/人/日(以60kg体重计),设0.5g/kg·bw/d、0.8g/kg·bw/d、1.6g/kg·bw/d三个剂量组(分别相当于受试样品人体推荐量的3倍、5倍、10倍),选用清洁级SD健康雄性大鼠40只,在动物室内检疫3天,按体重随机分为正常对照组、模型组和猴头颗粒低、中、高剂量组,每组8只。各剂量组灌胃样品30天,对照组给予无菌水,各剂量组灌胃样品30天后,全部动物严格禁食24小时(不禁水),此期间亦禁止给予受试物。除空白对照外,所有试验组动物给予无水乙醇1.0ml/只,1小时后处死动物,暴露完整胃,结扎幽门,灌注适量10%甲醛溶液,固定20min,然后沿胃大弯剪开,洗净胃内容物,展开胃黏膜,在体视解剖显微镜下或肉眼下用游标卡尺测量出血点或出血带的长度和宽度。
(3)实验结果
①猴头养胃颗粒对大鼠体重的影响
由表3可见,在急性胃黏膜损伤模型中,喂养猴头养胃颗粒30天后,各剂量组大鼠的体重增长良好,体重持续增长,但各时期体重与正常组比较差异无显著性差异(P﹥0.05),此项为阴性。

②猴头养胃颗粒对急性胃黏膜损伤中胃重量的影响
表4结果显示,急性乙醇刺激胃黏膜后,大鼠胃组织明显水肿,重量明显增加趋势,差异有统计学意义(P<0.001),而猴头养胃颗粒各剂量组,在给予急性乙醇灌胃后,肿胀程度明显减轻,胃组织重量较模型组明显减少,差异有统计学意义,此项为阳性。

③猴头养胃颗粒对无水乙醇致胃黏膜损伤的病理形态学改变
如图1所示,正常组大鼠胃黏膜腺体排列整齐、均匀、致密;肉眼观察可见单次大剂量给予大鼠无水乙醇造成胃黏膜损伤非常明显,黏膜表面呈现多条条索状出血,严重者表现弥漫性出血;与模型组相比,各给药组大鼠粘膜损较轻;与模型组比较,各剂量组明显减轻无水乙醇对胃黏膜的损伤,出血程度减轻,有的只出现充血而无出血带,此项为阳性。
④猴头养胃颗粒对急性胃黏膜损伤的保护作用
由表5可知,模型组大鼠胃黏膜观察指标中出血损伤积分指数和出血带发生率均高于正常组,差异有统计学意义(P﹤0.001),表明该模型成功。低、中、高剂量实验组出血损伤积分指数和出血发生率较模型组明显降低,有统计学意义(P﹤0.01),出血抑制率分别为19.42%、37.41%、61.87%,表明猴头养胃颗粒对急性酒精胃黏膜损伤有明显的保护作用,此项为阳性。

⑤大鼠胃黏膜组织H&E染色的病理学观察
由胃黏膜组织切片H&E染色图(图2)可知,正常组大鼠胃组织细胞排列紧密、有序,细胞结构清晰,腺体完整,未见明显异常;模型组大鼠胃黏膜在给予乙醇灌胃剌激后,胃黏膜组织遭到破坏,上皮细胞脱落、坏死,腺体排列紊乱,腺体扩张,粘膜下层水肿,炎性细胞浸润;各剂量猴头养胃颗粒组可见少量上皮细胞脱落,损伤情况均轻于模型组,低、中剂量组粘膜损伤稍重于高剂量组,此项为阳性。
猴头养胃颗粒以0.5g/kg·bw/d、0.8g/kg·bw/d、1.6g/kg·bw/d连续给样一个月,结果显示:大鼠体重为阴性,胃重量为阳性,出血发生率、出血损伤积分指数、出血损伤抑制率为阳性,胃组织病理变化为阳性。根据2003版《保健食品检验与评价技术规范》,对具有保护胃黏膜损伤功能的保健食品评价方法规定:受试物一个或一个以上剂量组与模型对照组进行比较,大体观察评分与病理组织学检查评分结果均表明胃黏膜损伤明显改善,可判定该受试样品动物实验结果为阳性。所以在本实验条件下猴头养胃颗粒具有保护胃黏膜损伤的功能。
利用本发明方法,每1kg猴头菇可制得颗粒5~8kg。包装后,10g/袋,储藏在阴凉干燥通风处。温水冲服,每次1袋,每日2次,饭前服用,具有保护酒精造成的胃黏膜损伤及助消化等效果。
Claims (10)
1.一种猴头菇养胃颗粒,其特征在于,所述猴头菇养胃颗粒主要是由下述原料制成的:猴头菇、黄芪、山楂和党参。
2.根据权利要求1所述的一种猴头菇养胃颗粒,其特征在于,所述猴头菇养胃颗粒由以下重量份的原料制成:猴头菇10重量份、黄芪2~4重量份、山楂1~3重量份、党参2~4重量份、乳糖20~50重量份、玉米糊精5~15重量份和淀粉0.5~2重量份。
3.根据权利要求1所述的一种猴头菇养胃颗粒,其特征在于,所述猴头菇养胃颗粒由以下重量份的原料制成:猴头菇10重量份、黄芪3重量份、山楂2重量份、党参3重量份、乳糖40重量份、玉米糊精10重量份和淀粉1重量份。
4.权利要求1或2所述的猴头菇养胃颗粒的制备方法,其特征在于,包括如下步骤:
(1)清洗:按重量份比例,挑选原料猴头菇、黄芪、山楂和党参,清洗干净;
(2)提取:清洗后的原料,混合后加水浸泡3-5h,然后进行水煮提取,所述原料混合物与水的料液比为1:(8~10),所述提取温度为80~100℃,提取时间为0.5~2h,提取次数为1~3次;
(3)过滤:过滤获得提取液;
(4)制粒:将提取液浓缩后进行醇沉,获得的沉淀按比例加水溶解后,再按重量份比例加入乳糖、玉米糊精和淀粉,制得猴头菇养胃颗粒。
5.根据权利要求4所述的猴头菇养胃颗粒的制备方法,其特征在于,步骤(2)所述料液比为1:8,提取温度为80℃,提取时间为1h,提取次数为3次。
6.根据权利要求4所述的猴头菇养胃颗粒的制备方法,其特征在于,步骤(3)所述过滤是指利用3-4层80目的纱布过滤。
7.根据权利要求4所述的猴头菇养胃颗粒的制备方法,其特征在于,步骤(4)所述浓缩温度为60-80℃,浓缩比例为步骤(1)中每10g的猴头菇对应步骤(4)中所述浓缩体积为20ml。
8.根据权利要求4所述的猴头菇养胃颗粒的制备方法,其特征在于,步骤(4)所述醇沉是指向浓缩后的提取液中加入体积分数为95%的乙醇至终浓度为80%,然后经3000rpm/min离心10min获得沉淀。
9.根据权利要求4所述的猴头菇养胃颗粒的制备方法,其特征在于,步骤(4)所述加水溶解,其加水的比例为步骤(1)中每10重量份的猴头菇对应步骤(4)中加水的量为5重量份。
