本发明公开了利用海参活性多肽制备复配物的方法,制备海参活性多肽、制备螯合钙、制备螯合亚铁、复配,将海参匀浆以中性蛋白酶酶解,酶解后超滤获得海参活性多肽,将海参活性多肽分别与氯化钙、氯化亚铁螯合,然后将海参活性多肽螯合钙粉、亚铁螯合肽粉溶解于海参活性多肽溶液中,低温浓缩得海参活性多肽复配物。有益效果为:金属螯合肽具有转运耗能低、转运速度快和不易被饱和等优点,有利于促进金属元素吸收利用,金属螯合肽能抑制小肠刷状缘细胞中肽酶活性,有助于提高金属螯合肽的稳定性,并能减少金属元素间的拮抗作用;同时海参活性多肽被机体吸收后还具有优异的抗氧化损伤、降低血糖、延缓衰老、抗肿瘤、抗疲劳、抗凝血等功能。

技术领域
本发明涉及海参活性多肽复配物领域,尤其是涉及利用海参活性多肽制备复配物的方法。
技术背景
海参,属海参纲(Holothuroidea),是生活在海边至8000米的海洋棘皮动物,距今已有六亿多年的历史,海参以海底藻类和浮游生物为食。 海参全身长满肉刺,广布于世界各海洋中。中国南海沿岸种类较多,约有二十余种海参可供食用。海参同人参、燕窝、鱼翅齐名,是世界八大珍品之一。海参不仅是珍贵的食品,也是名贵的药材。据《本草纲目拾遗》中记载:海参,味甘咸,补肾,益精髓,摄小便,壮阳疗痿,其性温补,足敌人参,故名海参。海参具有提高记忆力、延缓性腺衰老,防止动脉硬化以及抗肿瘤等作用。随着海参价值知识的普及,海参逐渐进入百姓餐桌。海参的营养价值极高,富含聚氨基葡萄糖、粘多糖、海洋生物活性钙、高蛋白、黏蛋白、多肽、胶原蛋白、核酸、海参皂甙、硫酸软骨素、多种维生素、及各种氨基酸和碳水化合物等50多种养分,是不可多得的不含胆固醇的高级滋补品。
现有技术如授权公众号为CN 107279972 B的中国发明专利,公开了一种基于马鲛鱼活性多肽的复配物,该发明方法中的复配物的成分及其重量份为:马鲛鱼活性多肽2-3份、1mol/L的氯化亚铁溶液0.5-1份、抗坏血酸0.1-0.3份、水90-100份。该发明的复配物可通过氨基酸或肽的吸收通道被吸收,提高吸收效率,能够高效快速地合成蛋白质,加快机体吸收金属离子的进程,利用率高,无金属异味,性质稳定,无毒副作用,具有改善缺铁性贫血的功能。但是该发明方法以鱼粉酶解制备活性多肽,不仅操作步骤繁琐,而且耗时较长,耗能较大。
发明内容-
本发明的目的在于提供利用海参活性多肽制备复配物的方法,本制备方法简单易行,制备过程中没有添加毒副物质,海参活性多肽的利用率较高。
本发明针对背景技术中提到的问题,采取的技术方案为:利用海参活性多肽制备复配物的方法,包括:制备海参活性多肽、制备螯合钙、制备螯合亚铁、复配,具体包括以下步骤:
制备海参活性多肽:称取海参样品,按料液比1:15-18匀浆,添加1800-3500U/g中性蛋白酶,再加入中性蛋白酶重量2.3-4.8‰的(R)-1,1’-联萘-2,2’-酚基二异丙氧钛,在酶解温度为55-58℃、酶解pH值为7.5-7.6的条件下酶解1.5-2.0小时,酶解结束后煮沸15-30分钟灭酶;将酶解液经截留分子量为10kD的超滤膜超滤,超滤液经浓缩和喷雾干燥,获得海参活性多肽粉末;(R)-1,1’-联萘-2,2’-酚基二异丙氧钛能够作用于蛋白酶的金属原子活性中心,降低蛋白酶与底物之间的能量壁垒,增大酶与底物之间的反应活性,从而大大提高蛋白酶的酶解活性,加速酶解,海参的水解度可达到52%以上,海参活性多肽的主要氨基酸组成为甘氨酸和丙氨酸,海参活性多肽对DPPH、·OH和·O2-具有较好的清除效果,同时海参活性多肽可以显著降低血清和肝脏中总胆固醇、甘油三酯的浓度,海参活性多肽可以促进NIH/3T3细胞生长,对NIH/3T3细胞胶原蛋白的分泌有显著的促进作用,还具有一定的抗氧化损伤能力、降低血糖、改善糖耐量的功效;
制备螯合钙:将海参活性多肽粉复溶成10-12%的肽液,调整pH为8.8-9.0,按肽钙质量比1:2-4加入无水氯化钙,再加入海参活性多肽质量2-3‰的维生素E,震荡混匀后,在28-30℃恒温螯合,螯合30-45分钟后,加入溶液体积8-10倍的无水乙醇,静置沉淀3-5小时后,在1-4℃、7500-8000r/min温度下离心25-45分钟,取下层沉淀物冻干得海参活性多肽螯合钙粉;螯合物的钙离子含量为56.78mg/g,谷氨酸和天冬氨酸等酸性氨基酸的贡献最大,以Ca-多肽的形式进入细胞,然后在细胞中主动分裂,释放出Ca2+便被细胞利用,从而促进金属离子吸收,是一种优质的补钙剂,同时海参活性多肽也可以增强机体体质;
制备螯合亚铁:将海参活性多肽粉复溶成5-8%的肽液,调节pH值至7.0-7.2,取100-120份肽液,加入200-250份0.8-1.0mol/L的FeCl2溶液,再加入2-5mg的抗坏血酸,在30-34℃温度下震荡合成25-30分钟,于2-4℃温度下10000-12000r/min离心浓缩20-45分钟,弃沉淀,上清液于旋转蒸发器38-40℃真空浓缩,冷冻干燥后即得亚铁螯合肽;亚铁螯合肽易于被机体吸收,避免了无机铁盐的稳定性差、胃肠刺激大、易受到肠内容物干扰、生物利用率低、存在一定毒副作用等缺点,同时海参活性多肽液可以增强机体体质,增强机体抗免疫能力,补充机体所需氨基酸;
复配:称取10-15份海参活性多肽螯合钙粉、20-30份亚铁螯合肽溶解于20-25%海参活性多肽溶液中,置于旋转蒸发器中,以20-22℃真空浓缩,冷冻干燥后粉碎,过100-160目筛即得海参活性多肽复配物;金属螯合肽可利用小肽配位体转运系统进行转运,具有转运耗能低、转运速度快和不易被饱和等优点,有利于促进金属元素吸收利用,金属螯合肽能抑制小肠刷状缘细胞中肽酶活性,有助于完整多肽作为金属元素配体通过小肽转运系统进入肠黏膜细胞,提高金属螯合肽的稳定性,并能减少金属元素间的拮抗作用;同时海参活性多肽具有优异的抗氧化损伤功能,同时可以降低血糖、改善糖耐量,可以有效强化机体免疫系统,还具有延缓衰老、抗肿瘤、抗疲劳、抗凝血等功能,同时也可以为机体补充氨基酸,是一种高效、安全的海参活性多肽复配物。
与现有技术相比,本发明的优点在于:
1)(R)-1,1’-联萘-2,2’-酚基二异丙氧钛能够作用于蛋白酶的金属原子活性中心,降低蛋白酶与底物之间的能量壁垒,增大酶与底物之间的反应活性,从而大大提高蛋白酶的酶解活性,加速酶解,海参的水解度可达到52%以上,海参活性多肽的主要氨基酸组成为甘氨酸和丙氨酸,海参活性多肽对DPPH、·OH和·O2-具有较好的清除效果,同时海参活性多肽可以显著降低血清和肝脏中总胆固醇、甘油三酯的浓度;

具体实施方式
下面通过实施例对本发明方案作进一步说明:
实施例1:
利用海参活性多肽制备复配物的方法,包括以下步骤:
1)称取海参样品,按料液比1:15匀浆,添加1800U/g中性蛋白酶,再加入中性蛋白酶重量2.3‰的(R)-1,1’-联萘-2,2’-酚基二异丙氧钛,在酶解温度为55℃、酶解pH值为7.5的条件下酶解1.5小时,酶解结束后煮沸15分钟灭酶;将酶解液经截留分子量为10kD的超滤膜超滤,超滤液经浓缩和喷雾干燥,获得海参活性多肽粉末;2)将海参活性多肽粉复溶成10%的肽液,调整pH为8.8,按肽钙质量比1:2加入无水氯化钙,再加入海参活性多肽质量2‰的维生素E,震荡混匀后,在28℃恒温螯合,螯合30分钟后,加入溶液体积8倍的无水乙醇,静置沉淀3小时后,在1℃、7500r/min温度下离心25分钟,取下层沉淀物冻干得海参活性多肽螯合钙粉;3)将海参活性多肽粉复溶成5%的肽液,调节pH值至7.0,取100份肽液,加入200份0.8mol/L的FeCl2溶液,再加入2mg的抗坏血酸,在30℃温度下震荡合成25分钟,于2℃温度下10000r/min离心浓缩20分钟,弃沉淀,上清液于旋转蒸发器38℃真空浓缩,冷冻干燥后即得亚铁螯合肽;4)称取10份海参活性多肽螯合钙粉、20份亚铁螯合肽溶解于20%海参活性多肽溶液中,置于旋转蒸发器中,以20℃真空浓缩,冷冻干燥后粉碎,过100目筛即得海参活性多肽复配物。
实施例2:
利用海参活性多肽制备复配物的方法,包括以下步骤:
1)制备海参活性多肽:称取海参样品,按料液比1:18匀浆,添加3500U/g中性蛋白酶,再加入中性蛋白酶重量4.8‰的(R)-1,1’-联萘-2,2’-酚基二异丙氧钛,在酶解温度为58℃、酶解pH值为7.6的条件下酶解1.5-2.0小时,酶解结束后煮沸30分钟灭酶;将酶解液经截留分子量为10kD的超滤膜超滤,超滤液经浓缩和喷雾干燥,获得海参活性多肽粉末;(R)-1,1’-联萘-2,2’-酚基二异丙氧钛能够作用于蛋白酶的金属原子活性中心,降低蛋白酶与底物之间的能量壁垒,增大酶与底物之间的反应活性,从而大大提高蛋白酶的酶解活性,加速酶解,海参的水解度可达到52%以上,海参活性多肽的主要氨基酸组成为甘氨酸和丙氨酸,海参活性多肽对DPPH、·OH和·O2-具有较好的清除效果,同时海参活性多肽可以显著降低血清和肝脏中总胆固醇、甘油三酯的浓度,海参活性多肽可以促进NIH/3T3细胞生长,对NIH/3T3细胞胶原蛋白的分泌有显著的促进作用,还具有一定的抗氧化损伤能力、降低血糖、改善糖耐量的功效;
2)制备螯合钙:将海参活性多肽粉复溶成12%的肽液,调整pH为9.0,按肽钙质量比1:4加入无水氯化钙,再加入海参活性多肽质量3‰的维生素E,震荡混匀后,在30℃恒温螯合,螯合45分钟后,加入溶液体积10倍的无水乙醇,静置沉淀5小时后,在4℃、8000r/min温度下离心45分钟,取下层沉淀物冻干得海参活性多肽螯合钙粉;螯合物的钙离子含量为56.78mg/g,谷氨酸和天冬氨酸等酸性氨基酸的贡献最大,以Ca-多肽的形式进入细胞,然后在细胞中主动分裂,释放出Ca2+便被细胞利用,从而促进金属离子吸收,是一种优质的补钙剂,同时海参活性多肽也可以增强机体体质;
3)制备螯合亚铁:将海参活性多肽粉复溶成8%的肽液,调节pH值至7.2,取120份肽液,加入250份1.0mol/L的FeCl2溶液,再加入5mg的抗坏血酸,在34℃温度下震荡合成30分钟,于4℃温度下12000r/min离心浓缩45分钟,弃沉淀,上清液于旋转蒸发器40℃真空浓缩,冷冻干燥后即得亚铁螯合肽;亚铁螯合肽易于被机体吸收,避免了无机铁盐的稳定性差、胃肠刺激大、易受到肠内容物干扰、生物利用率低、存在一定毒副作用等缺点,同时海参活性多肽液可以增强机体体质,增强机体抗免疫能力,补充机体所需氨基酸;
4)复配:称取15份海参活性多肽螯合钙粉、30份亚铁螯合肽溶解于25%海参活性多肽溶液中,置于旋转蒸发器中,以22℃真空浓缩,冷冻干燥后粉碎,过160目筛即得海参活性多肽复配物;金属螯合肽可利用小肽配位体转运系统进行转运,具有转运耗能低、转运速度快和不易被饱和等优点,有利于促进金属元素吸收利用,金属螯合肽能抑制小肠刷状缘细胞中肽酶活性,有助于完整多肽作为金属元素配体通过小肽转运系统进入肠黏膜细胞,提高金属螯合肽的稳定性,并能减少金属元素间的拮抗作用;同时海参活性多肽具有优异的抗氧化损伤功能,同时可以降低血糖、改善糖耐量,可以有效强化机体免疫系统,还具有延缓衰老、抗肿瘤、抗疲劳、抗凝血等功能,同时也可以为机体补充氨基酸,是一种高效、安全的海参活性多肽复配物。
实施例3:
利用海参活性多肽制备复配物的方法,包括:制备海参活性多肽、制备螯合钙、制备螯合亚铁、复配,具体包括以下步骤:
制备海参活性多肽:称取海参样品,按料液比1:16匀浆,添加2500U/g中性蛋白酶,再加入中性蛋白酶重量3.8‰的(R)-1,1’-联萘-2,2’-酚基二异丙氧钛,在酶解温度为56℃、酶解pH值为7.5的条件下酶解1.5小时,酶解结束后煮沸25分钟灭酶;将酶解液经截留分子量为10kD的超滤膜超滤,超滤液经浓缩和喷雾干燥,获得海参活性多肽粉末;(R)-1,1’-联萘-2,2’-酚基二异丙氧钛能够作用于蛋白酶的金属原子活性中心,降低蛋白酶与底物之间的能量壁垒,增大酶与底物之间的反应活性,从而大大提高蛋白酶的酶解活性,加速酶解,海参的水解度可达到52%以上,海参活性多肽的主要氨基酸组成为甘氨酸和丙氨酸,海参活性多肽对DPPH、·OH和·O2-具有较好的清除效果,同时海参活性多肽可以显著降低血清和肝脏中总胆固醇、甘油三酯的浓度,海参活性多肽可以促进NIH/3T3细胞生长,对NIH/3T3细胞胶原蛋白的分泌有显著的促进作用,还具有一定的抗氧化损伤能力、降低血糖、改善糖耐量的功效;

复配:称取14份海参活性多肽螯合钙粉、24份亚铁螯合肽溶解于22%海参活性多肽溶液中,置于旋转蒸发器中,以20℃真空浓缩,冷冻干燥后粉碎,过140目筛即得海参活性多肽复配物;金属螯合肽可利用小肽配位体转运系统进行转运,具有转运耗能低、转运速度快和不易被饱和等优点,有利于促进金属元素吸收利用,金属螯合肽能抑制小肠刷状缘细胞中肽酶活性,有助于完整多肽作为金属元素配体通过小肽转运系统进入肠黏膜细胞,提高金属螯合肽的稳定性,并能减少金属元素间的拮抗作用;同时海参活性多肽具有优异的抗氧化损伤功能,同时可以降低血糖、改善糖耐量,可以有效强化机体免疫系统,还具有延缓衰老、抗肿瘤、抗疲劳、抗凝血等功能,同时也可以为机体补充氨基酸,是一种高效、安全的海参活性多肽复配物。

1.利用海参活性多肽制备复配物的方法,包括:制备海参活性多肽、制备螯合钙、制备螯合亚铁、复配,其特征在于:所述制备海参活性多肽的步骤为:称取海参样品,按料液比1:15-18匀浆,添加中性蛋白酶、(R)-1,1’-联萘-2,2’-酚基二异丙氧钛酶解,酶解结束后煮沸15-30分钟灭酶;将酶解液经截留分子量为10kD的超滤膜超滤,超滤液经浓缩和喷雾干燥,获得海参活性多肽粉末。
2.根据权利要求1所述的利用海参活性多肽制备复配物的方法,其特征在于:所述制备海参活性多肽步骤中的中性蛋白酶的添加量为1800-3500U/g,(R)-1,1’-联萘-2,2’-酚基二异丙氧钛的添加量为中性蛋白酶重量的2.3-4.8‰。
3.根据权利要求1所述的利用海参活性多肽制备复配物的方法,其特征在于:所述制备海参活性多肽步骤中的酶解温度为55-58℃,酶解pH值为7.5-7.6,酶解时间为1.5-2.0小时。
4.根据权利要求1所述的利用海参活性多肽制备复配物的方法,其特征在于:所述制备螯合钙步骤为:将海参活性多肽粉复溶成10-12%的肽液,调整pH为8.8-9.0,按肽钙质量比1:2-4加入无水氯化钙,再加入海参活性多肽质量2-3‰的维生素E,震荡混匀后恒温螯合,然厚加入溶液体积8-10倍的无水乙醇,静置沉淀3-5小时后,在1-4℃、7500-8000r/min温度下离心25-45分钟,取下层沉淀物冻干得海参活性多肽螯合钙粉。
5.根据权利要求4所述的利用海参活性多肽制备复配物的方法,其特征在于:所述制备螯合钙步骤中的螯合温度为28-30℃,螯合时间为30-45分钟。
6.根据权利要求1所述的利用海参活性多肽制备复配物的方法,其特征在于:所述制备螯合亚铁步骤为:将海参活性多肽粉复溶成5-8%的肽液,调节pH值至7.0-7.2,取100-120份肽液,加入200-250份0.8-1.0mol/L的FeCl2溶液,再加入2-5mg的抗坏血酸,恒温震荡合成后于2-4℃温度下以10000-12000r/min离心浓缩20-45分钟,弃沉淀,上清液于旋转蒸发器38-40℃真空浓缩,冷冻干燥后即得亚铁螯合肽。
7.根据权利要求6所述的利用海参活性多肽制备复配物的方法,其特征在于:所述制备螯合亚铁步骤中的螯合温度为30-34℃,螯合时间为25-30分钟。
8.根据权利要求1所述的利用海参活性多肽制备复配物的方法,其特征在于:所述复配步骤为:称取10-15份海参活性多肽螯合钙粉、20-30份亚铁螯合肽溶解于20-25%海参活性多肽溶液中,置于旋转蒸发器中,以20-22℃真空浓缩,冷冻干燥后粉碎,过100-160目筛即得海参活性多肽复配物。
