技术领域

背景技术
由于大多数用于癌症的常规基因治疗剂通过局部施用(local administration)被注射到身体中,因此就遍布全身的转移性癌症而言,存在可达性降低的限制。因此,在疾病不可察觉地遍布全身,例如转移性癌症的情况下,尽管截至目前一般来说已经从基因治疗中排除了这种疾病,但仍有必要开发能够全身施用的产品以扩大基因治疗市场。
由于多种优点,腺病毒作为基因转移载体已经变得流行,它们越来越多地用于癌症基因治疗,并且最常用于患者临床试验。特别地,通过仅在癌细胞中选择性增殖而能够仅杀死癌细胞的溶瘤腺病毒不仅能够在原发感染细胞中发挥治疗效果,而且由于通过增殖病毒对周围肿瘤细胞的一系列感染和癌细胞杀伤,治疗效果可以像多米诺骨牌效应一样保持扩散,因此也大大提高了癌症治疗效果。同时,当将治疗性基因装载于溶瘤腺病毒中时,治疗性基因仅在癌细胞中高效地受限表达,因此可以协同地改善治疗效果。此外,由于外周正常细胞中溶瘤腺病毒的增殖受到抑制,不会发生细胞杀伤,因此安全性高。出于该原因,治疗性基因负载腺病毒已经作为下一代抗癌疗法而备受关注。

根据本发明的一个示例性实施方案,生物相容性聚合物(biocompatiblepolymer)是包含以下的聚合物:(i)可从免疫反应逃脱的部分(escapable portion fromimmune reactions)、(ii)可带电荷部分(chargeable portion)和(iii)具有二硫键的可生物还原部分(bioreducible portion)。
根据本发明的一个示例性实施方案,可从免疫反应逃脱的部分(escapableportion from immune reactions)是选自以下的任一种:聚乙二醇(polyethyleneglycol,PEG)、聚环氧烷[例如,聚氧化乙烯、聚氧化丙烯或其共聚物(例如,聚环氧乙烷-聚环氧丙烷-聚环氧乙烷共聚物)]、聚苯醚-PEG-聚环氧烷共聚物、聚(甲基丙烯酸甲氧乙酯)、聚(甲基丙烯酰基磷脂酰胆碱)、全氟化聚醚、右旋糖酐和聚乙烯吡咯烷酮。
在本发明的另一个方面中,本发明提供了用于治疗癌症的药物组合物,其包含:(a)(i)选自以下的任一种或更多种材料:SEQ ID NO:1的肽或其经修饰肽、肽或其二聚体、和生物相容性聚合物(biocompatible polymer)、脂质体、泡囊和纳米材料,(ii)治疗有效量的包含病毒DNA或病毒的复合物;和(b)可药用载体。

i.聚合物
广泛用作非病毒载体的基于聚合物的载体的实例包括明胶、壳聚糖(Carreno GB,Duncan R.Evaluation of the biological properties of soluble chitosan andchitosan microspheres.Int J Pharm 148:231-240(1997))、聚-L-赖氨酸(PLL;MaruyamaA,Ishihara T,Kim JS,Kim SW,Akaike T.Nanoparticle DNA carrier with poly(L-lysine)grafted polysaccharide copolymer and poly(D,L-lactide).BioconjugateChem 8:735-742(1997))和聚乙烯胺(PEI;Abdallah B,Hassan A,Benoist C,Goula D,Behr JP,Demeneix BA.A powerful non-virus vector for in vivo gene transferinto the adult mammalian brain:Polyethyleneimine.Human Gene Ther 7:1947-1954(1996))。基于聚合物的载体具有以下优点:免疫应答发生率低和急性毒性小、制备方法简单以及便于批量生产。
ii.脂质体和泡囊
脂质体由分散在水相中的磷脂自动形成。使用脂质体成功地将外源DNA分子递送到细胞中的实例公开于Nicolau和Sene,Biochim.Biophys.Acta,721:185-190(1982)和Nicolau等,Methods Enzymol.,149:157-176(1987)中。同时,使用脂质体转化动物细胞最广泛使用的试剂是Lipofectamine(Gibco BRL)。包含待递送的靶核苷酸序列的脂质体通过机制如内吞作用、吸附到细胞表面或与浆细胞膜融合来与细胞相互作用将待递送的靶核苷酸序列递送到细胞中。
另一方面,虽然脂质体由磷脂制成,但是泡囊(niosome)是由非离子表面活性剂(nonionic surfactant)和胆固醇(cholesterol)的组合组成的双层转运蛋白,其包含极性和非极性材料,并且与通过磷脂作为脂质颗粒载体的脂质体相比,其具有渗透活性且稳定。此外,在储存和处理制备中使用的表面活性剂方面没有特定条件。
iii.纳米颗粒
本文使用的术语“纳米颗粒”是指尺寸为几纳米至几百纳米的生物相容性聚合材料,并且可为例如聚乙二醇(polyetheleneglycol,PEG)、聚乳酸(poly-lactide,PLA)、聚乙交酯(polyglycolide,PGA)、聚-乳酸-共-乙交酯(poly-lactide-co-glycolide,PLGA)、聚-ε-己内酯(poly-ε-carprolactone,PCL)、透明质酸(hValuronic acid,HA)、壳聚糖(chitosan)或血清白蛋白。
由于药物组合物包含上述肽或肽-聚合物复合物作为活性成分,因此将省略共同的内容以避免说明书中的过度复杂。
本文使用的“病毒DNA”包括完整、部分缺失或部分修饰的病毒基因组DNA。
根据本发明的一个示例性实施方案,当病毒DNA是腺病毒DNA(Ad pDNA-聚合物-二聚体肽复合物)时,Ad pDNA:聚合物的重量比可为1∶1至1∶1000,并且Ad pDNA:二聚体肽的重量比可为1∶0.001至1∶1000,但本发明不限于此。
用于本发明的病毒包括某种病毒,并且具体地,是用作基因治疗的病毒。例如,病毒如下,但本发明不限于此。
i.腺病毒
由于中等大小的基因组、方便操作、效价高、宽范围的靶细胞和优异的感染性,腺病毒已被广泛用作基因转移载体。基因组的两个末端都包含100bp至200bp的反向末端重复(inverted terminal repeat,ITR),其是DNA复制和包装所必需的顺式元件。基因组的E1区(E1A和E1B)编码转录蛋白和调节宿主细胞基因转录的蛋白。E2区(E2A和E2B)编码参与病毒DNA复制的蛋白。
在当前开发的腺病毒载体中,缺乏E1区的复制缺陷型腺病毒已被广泛使用。同时,从普通的腺病毒载体中除去E3区域,从而提供外源基因插入位点(Thimmappaya,B.等,Cell,31:543-551(1982);和Riordan,J.R.等,Science,245:1066-1073(1989))。另一方面,将要递送到细胞中的靶核苷酸序列特定地插入到缺失的E1区(E1A区和/或E1B区,优选E1B区)或E3区中,并且更特定地插入到缺失的E1区中。在本文使用的关于病毒基因组序列的术语“缺失”不仅意味着相应序列的完全缺失,而且还意味着其部分缺失。
此外,由于腺病毒可以包装高达约105%的野生型基因组,所以大约2kb可以被额外包装(Ghosh-Choudhury等,EMBO J.,6:1733-1739(1987))。因此,插入腺病毒的上述外源序列可以另外与腺病毒基因组结合。
腺病毒有42种不同的血清型和亚组A至F。其中,亚组C中包含的5型腺病毒是能够获得本发明的腺病毒载体的最合适的起始材料。5型腺病毒的生化和遗传信息是公知的。
通过与附加体相同的方法复制由腺病毒递送的外源基因,因此对宿主细胞具有非常低的遗传毒性。
ii.逆转录病毒
逆转录病毒已经被广泛用作基因转移载体,因为它们自己的基因可以被插入到宿主基因组中,大量的外来遗传物质可以被递送,并且被感染的细胞谱很宽。
为了构建逆转录病毒载体,将待递送的靶核苷酸序列替代逆转录病毒序列插入逆转录病毒基因组中,从而产生不可复制的病毒。为了产生病毒粒子,构建包含gag、pol和env基因但缺乏长末端重复(long terminal repeat,LTR)和Ψ序列的包装细胞系(Mann等,Cell,33:153-159(1983))。当递送包含靶核苷酸序列的重组质粒时,将LTR和Ψ序列引入细胞系,Ψ序列允许产生重组质粒的RNA转录物,将转录物包装入病毒中,并将病毒释放至培养基(Nicolas和Rubinstein″Retrovirus vectors,″In:Vectors:A survey of molecularcloning vectors and their uses,Rodriguez和Denhardt(编辑),Stoneham:Butterworth,494-513(1988))。收集包含重组逆转录病毒的培养基并浓缩以用作基因转移系统。
已经报道了使用第二代逆转录病毒载体的基因转移。根据Kasahara等Science,266:1373-1376(1994),制造莫洛尼鼠白血病病毒(MMLV)的变体,并将红细胞生成素(erythropoietin,EPO)序列插入包膜区域中,从而产生具有新结合特征的嵌合蛋白。本发明的基因转移系统也可以根据上述第二代逆转录病毒载体的构建策略制造。
iii.AAV载体
由于腺相关病毒(AAV)可用于本发明的基因转移系统,因为其能够感染非分裂细胞和多种类型的细胞。美国专利No.5,139,941和No.4,797,368中详细公开了AAV载体的制造和使用的详细描述。
通常地,AAV通过用具有与两个AAV末端重复相邻的靶基因序列的质粒(McLaughlin等,J.Virol.,62:1963-1973(1988);和Samulski等,J.Virol.,63:3822-3828(1989))和包含不具有末端重复的野生型AAV编码序列的表达质粒(McCarty等,J.Virol.,65:2936-2945(1991))转化而制造。
iv.其他病毒载体
其他病毒载体也可以用作本发明的基因转移系统。源自痘苗病毒(Puhlmann M.等,Human Gene Therapy 10:649-657(1999);Ridgeway,“Mammalian expressionvectors,”In:Vectors:A survey of molecular cloning vectors and theiruses.Rodriguez和Denhardt编辑Stoneham:Butterworth,467-492(1988);Baichwal和Sugden,“Vectors for gene transfer derived from animal DNA viruses:Transientand stable expression of transferred genes,”In:Kucherlapati R编辑.Genetransfer.New York:Plenum Press,117-148(1986)以及Coupar等,Gene,68:1-10(1988))、慢病毒(Wang G.等,J.Clin.Invest.104(11):R55-62(1999))或单纯疱疹病毒(Chamber R.等,Proc.Natl.Acad.Sci USA 92:1411-1415(1995))、呼肠孤病毒、痘病毒(GCE,NJL,KrupaM,Esteban M.,The poxvirus vectors MVA and NYVAC as gene delivery systems forvaccination against infectious diseases and cancer Curr Gene Ther 8(2):97-120(2008))、塞姆利基森林病毒的载体也可以用作可以将靶核苷酸序列转移到细胞中的转移系统。
同时,用于本发明的病毒的外源基因序列是诱导癌细胞死亡的癌症治疗基因,最终导致肿瘤变性,并包括肿瘤抑制基因,免疫调节基因[例如:细胞因子基因、趋化因子基因和共刺激因子(costimulatory factor):T细胞活化所必需的辅助分子如B7.1和B7.2)]、抗原基因、自杀基因、细胞毒性基因、细胞抑制基因、促凋亡基因和抗血管生成基因,但本发明不限于此。
自杀基因是表达通过外部因素杀伤细胞或诱导细胞中毒性条件的物质的核酸序列。公知的自杀基因是胸苷激酶(TK)基因(美国专利No.5,631,236号和5,601,818)。表达TK基因产物的细胞对于通过施用丙氧鸟苷(gancyclovir)的选择性死亡敏感。肿瘤抑制基因表示编码抑制肿瘤发生的多肽的基因。肿瘤抑制基因是哺乳动物中天然存在的基因,并且该基因的缺失或失活已被认为是肿瘤发生的先决条件。肿瘤抑制基因的实例包括:APC、DPC4、NF-1、NF-2、MTS1、WT1、BRCA1、BRCA2、VHL、p53、Rb、MMAC-1、MMSC-2、视网膜母细胞瘤基因(Lee等Nature,329:642(1987))、腺瘤性结肠息肉病蛋白质(adenomatous polyposiscoli protein)(美国专利No.5,783,666)、位于染色体3p21.3上的鼻咽肿瘤抑制基因(Cheng等Proc.Nat.Acad.Sci.,95:3042-3047(1998))、缺失结肠癌(DCC)基因、包括MTS1、CDK4、VHL、p110Rb、p16和p21的肿瘤抑制基因INK4a家族成员及其治疗有效片段(例如,p56Rb、p94Rb等)。本领域普通技术人员将理解,除了示例性基因之外的所有已知的抑癌基因均可用于本发明。
本文使用的术语“抗原基因(antigenic gene)”是指在靶细胞中表达并产生可在免疫系统中识别的细胞表面抗原蛋白的核苷酸序列。这种抗原基因的实例包括癌胚抗原(carcinoembryonic antigen,CEA)、前列腺特异性抗原(prostate specific antigen,PSA)、α-胎蛋白(α-feto proteinAFP)和p53(WO 94/02167)。为了容易被免疫系统识别,抗原基因可以与MHC I型抗原结合。
本文使用的术语“细胞毒性基因”是指在细胞中表达并表现出毒性作用的核苷酸序列。此类细胞毒性基因(cytotoxic gene)的实例包括编码假单胞菌外毒素(exotoxin)、蓖麻毒素、白喉毒素等的核苷酸序列。
本文使用的术语“细胞抑制基因”是指在细胞中表达并在细胞周期期间中止细胞周期的核苷酸序列。这种细胞抑制基因(cytostatic gene)的实例可以包括p21、视网膜母细胞瘤基因、E2F-Rb融合蛋白基因、编码细胞周期蛋白依赖性激酶抑制剂的基因(例如,p16、p15、p18和p19)、生长终止特异性同源盒(growth arrest specific homeobox,GAX)基因(WO 97/16459和WO 96/30385),但本发明不限于此。
许多可有效用于治疗多种疾病的治疗基因也可作为通过本发明的腺病毒来帮助抗肿瘤作用的手段来递送。治疗基因的实例包括编码细胞因子(例如,干扰素-α、-β、-γ和-δ)、白细胞介素(例如,IL-1、IL-2、IL-4、IL-6、IL-7、IL-10、IL-12、IL-19和IL-20)和集落刺激因子(例如,GM-CSF和G-CSF)的基因,以及编码趋化因子组(单核细胞趋化蛋白-1(MCP-1)、单核细胞趋化蛋白-2(MCP-2)、单核细胞趋化蛋白-3(MCP-3)、单核细胞趋化蛋白-4(MCP-4))、巨噬细胞炎性蛋白1α(MIP-1α)、巨噬细胞炎性蛋白1β(MIP-1β)、巨噬细胞炎性蛋白1γ(MIP-1γ)、巨噬细胞炎性蛋白3α(MIP-3α)、巨噬细胞炎性蛋白3β(MIP-3β)、EBI1-配体趋化因子(ELC)、巨噬细胞炎性蛋白4(MIP-4)、巨噬细胞炎性蛋白5(MIP-5)、LD78β、RANTES、SIS-g(p500)、胸腺和激活调节趋化因子(TARC)、嗜酸性粒细胞趋化因子(eotaxin)、I-309、人蛋白HCC-1/NCC-2、人蛋白HCC-3和鼠蛋白C10的基因。此外,还包括表达组织纤溶酶原激活剂(tPA)或尿激酶的基因和产生显示持续血栓形成作用以预防高胆固醇血症的LAL的基因。此外,用于治疗囊性纤维化、腺苷脱氨酶缺乏症、病毒例如AIDS以及恶性和炎性疾病和病症的多种多核苷酸是已知的。
本文使用的术语“促凋亡基因(pro-apoptotic gene)”是指被表达以诱导程序性细胞死亡的核苷酸序列。这种促凋亡基因的实例包括p53、腺病毒E3-11.6K(源自Ad2和Ad5)和腺病毒E3-10.5K(源自Ad)、腺病毒E4基因以及编码Fas配体、TNF-、TRAIL、p53途径基因和半胱天冬酶的基因。
本文使用的术语“抗血管生成基因(anti-angiogenic gene)”是指被表达以细胞外释放抗血管生成因子的核苷酸序列。抗血管生成因子的实例包括血管抑制素、血管内皮生长因子(VEGF)抑制剂如Tie2(PNAS,1998,95,8795-800)和内皮抑素。

上述靶核苷酸序列可以从DNA序列数据库例如GenBank或EMBL中获得。
本发明的肽-病毒DNA(或病毒)复合物或肽-聚合物-病毒DNA(或病毒)复合物可用于治疗多种疾病,特别是抗癌治疗。

本文使用的术语“治疗”是指(i)预防癌细胞的形成;(ii)根据癌细胞的移除抑制癌症相关疾病或病症;和(iii)根据癌细胞的移除减轻癌症相关疾病或病症。因此,本文使用的术语“治疗有效量”是指足以实现药物效果的量。
包含在本发明的组合物中的可药用载体通常用于制剂中,并且可包含乳糖、右旋糖、蔗糖、山梨糖醇、甘露糖醇、淀粉、阿拉伯树胶、磷酸钙、藻酸盐、明胶、硅酸钙、微晶纤维素、聚乙烯吡咯烷酮、纤维素、水、糖浆、甲基纤维素、羟基苯甲酸甲酯、羟基苯甲酸丙酯、滑石、硬脂酸镁和矿物油,但本发明不限于此。本发明的药物组合物除了上述组分以外还可包含润滑剂、润湿剂、甜味剂、调味剂、乳化剂、悬浮液、防腐剂等。
本发明的药物组合物可以经肠胃外施用,例如经静脉内、腹膜内、肌内、皮下或局部施用。药物组合物可以经腹膜内施用以治疗卵巢癌,并且可以施用于门静脉以治疗肝癌。在乳腺癌的情况下,药物组合物可以直接注射到肿瘤块中,并且可以通过灌肠直接注射以治疗结肠直肠癌。
药物组合物的合适剂量可以根据参数例如形成方法、施用方法、患者年龄、体重和性别、疾病症状的严重程度、饮食、施用时间、施用途径、排泄速度和反应敏感性而变化,并且用于期望治疗的有效剂量可以由普通技术医师容易地确定和处方。通常地,本发明的药物组合物包含1×105至1×1015pfu/ml的聚合物-病毒复合物,并且通常每隔一天以1×1010pfu注射2周。
本发明的药物组合物可以根据可由本领域普通技术人员容易实施的方法使用可药用载体和/或赋形剂配制之后通过单位剂量包装或多剂量包装来制备。在此,剂型可以是在油或水性介质中的溶液、悬浮液或乳剂、提取物、散剂、颗粒、片剂或胶囊,并且本发明的药物组合物还可以包含分散剂或稳定剂。
本发明的药物组合物可以单独使用或与常规化学疗法或放射疗法组合使用,并且这样的组合方法可以更有效地用于癌症治疗。可以与本发明组合物组合使用的化疗药物包括顺铂、卡铂、丙卡巴肼、氮芥、环磷酰胺、异环磷酰胺、美法仑、苯丁酸氮芥、白消安(bisulfan)、亚硝基脲、更生霉素、道诺霉素、多柔比星、博来霉素、佩里霉素(plicomycin)、丝裂霉素、依托泊苷、他莫昔芬、紫杉醇、反铂(transplatinum)、5-氟尿嘧啶、长春新碱、长春碱和氨甲蝶呤。可以与本发明的组合物组合使用的放射疗法的实例包括X射线辐射和γ射线辐射。
根据本发明的另一个方面,本发明提供了使用包含SEQ ID NO:1的肽或其经修饰肽的组合物用于改善生物活性物质的细胞内递送效率的方法。
根据本发明的又一个方面,本发明提供了使用包含生物相容性聚合物和SEQ IDNO:1的肽或其经修饰肽的组合物用于提高生物活性物质的细胞内递送效率的方法。
本文使用的术语“生物活性物质”是指在本发明的目的方面,具有生物活性的生理功能调节剂和可用于治疗由生理功能的失衡或功能障碍引起的所有疾病的物质。生物活性物质可为核酸、肽、多肽、抗体、化学化合物(化学治疗剂)(chemical compound(chemotherapeutic))或病毒,但本发明不限于此。
由于本发明的方法使用上述肽或肽-聚合物复合物,因此将省略共同的内容以避免说明书中过度复杂。

实施例
实施例1:肽-聚合物复合物的物理特性的分析
意图使用其中9个氨基酸(精氨酸)与肽(SEQ ID NO:1)的C末端连接的肽(VI-9R)和聚合物来增强腺病毒DNA的细胞内递送效率,所述肽(SEQ ID NO:1)由源自腺病毒蛋白VI的13个氨基酸组成。为此,首先将腺病毒DNA添加到PBS缓冲液中,计算重量比之后,添加肽,然后在室温下反应10分钟。之后,再次以多种重量比添加聚合物,并在室温下反应15分钟,由此制备Ad pDNA-聚合物-VI-9R复合物。作为所述聚合物,使用精氨酸接枝的可生物还原的聚(CBA-DAH)聚合物(ABP)和聚酰胺胺(PAMAM,polyamidoamine)树状聚合物-缀合的PAM-ABP,并且其结构如下所示(Journal ofControlled Release,第160卷,592-600,2012)。

之后,意图鉴定通过将肽-聚合物复合物与Ad pDNA结合来由DNA形成复合物的条件。腺病毒质粒DNA(Ad pDNA)与VI-9R的重量比固定为1∶10用于反应,然后将DNA与聚合物的重量比调整为1∶1、1∶5、1∶10、1∶15和1∶20以确认结合Ad pDNA-聚合物-VI-9R复合物的程度。确定形成Ad pDNA-聚合物-VI-9R复合物的最佳条件,并进行凝胶阻滞测定以分析物理特性。
作为分析结果,从DNA:聚合物的重量比为1∶1时起,可以确认DNA缩合通过将腺病毒DNA装载到聚合物-肽复合物中充分发生(图4)。因此,当将VI-9R与聚合物组合使用时,可以确认即使使用少量聚合物,也将腺病毒DNA充分装载到聚合物-肽复合物中。
实施例2:确认Ad pDNA-聚合物-VI-9R复合物的细胞内基因递送效率
在实施例2中,作为聚合物,使用具有生物降解性和pH敏感性的mPEG-b-Pip-CBA(PPCBA),其结构如下所示(Biomaterials,第41卷,53-68,2015)。

为了确认Ad pDNA-PPCBA-VI-9R复合物的细胞内基因递送效率,首先,使用GFP表达性复制缺陷型腺病毒(pdE1/GFP)的质粒DNA将腺病毒DNA添加到PBS缓冲液中,计算重量比之后,将肽添加到同一管中,然后在室温下反应10分钟。之后,添加聚合物,并在室温下反应15分钟,由此制备Ad pDNA-聚合物-VI-9R复合物。在将细胞接种到24孔板之后用pdE1/GFP-PPCBA和pdE1/GFP-PPCBA-VI-9R复合物处理达到60%至70%汇合的293A细胞系,并且在72小时之后,通过荧光显微镜观察GFP表达。
作为观察的结果,在pdE1/GFP-PPCBA的情况下,完全没有观察到GFP表达,但是当用pdE1/GFP-PPCBA-VI-9R复合物处理细胞时,观察到显著提高的GFP表达(图4)。从上述结果可以确认,聚合物与VI-9R缀合的pdE1/GFP-PPCBA-VI-9R复合物能够克服低基因递送效率,这是其中病毒DNA仅与聚合物缀合的复合物(pdE1/GFP-PPPCBA)的限制,并且显著地提高细胞内基因递送效率。
实施例3:确认Ad pDNA-聚合物-VI-9R复合物的病毒增殖能力
在实施例3中,使用PPCBA作为聚合物,并且使用表达GFP的肿瘤选择性溶瘤腺病毒RdB/GFP的病毒DNA,即Ad pDNA(RdB/GFP)作为腺病毒DNA。为了确认Ad pDNA(RdB/GFP)-PPCBA-VI-9R复合物的病毒增殖能力,将A549细胞系接种到12孔板中,在24小时之后,用裸Ad pDNA、Ad pDNA-PEI 25K复合物和Ad pDNA-PPCBA-VI-9R复合物中的每一个处理细胞。在72小时之后,收集培养溶液,通过Q-PCR测定腺病毒颗粒的数目。
作为分析结果,可以确认当仅裸Ad pDNA处理时,测定病毒产量为基础水平(9.2×103VP),并且当Ad pDNA-PPCBA-VI-9R复合物处理时,与其中裸Ad pDNA处理的情况相比,病毒产量(5.5×106VP)提高了596倍(图5)。这表明,与其中Ad pDNA-PEI 25K复合物处理作为阳性对照(1.2×106VP)的情况相比,病毒产量提高了4.6倍。该结果表明由于Ad pDNA-PPCBA-VI-9R复合物的特性,病毒产量提高的可能性非常高,其中在癌细胞中非常成功地进行病毒增殖,然后增殖的腺病毒依次在肿瘤细胞中增殖。
实施例4:通过Ad pDNA-聚合物-VI-9R复合物确认Ad pDNA的细胞摄取效率
在实施例4中,使用PAM-ABP作为聚合物。为了确认通过Ad pDNA(dE1/GFP)-PAM-ABP-VI-9R复合物提高Ad pDNA的细胞摄取效率,将293A细胞系接种到12孔板中并生长至60%至70%汇合,并且在24小时之后,使用FITC-标记的Ad质粒DNA(FITC-pDNA),将细胞用Ad pDNA-PEI、Ad pDNA-PAM-ABP和Ad pDNA-PAM-ABP-VI-9R复合物处理。在2小时之后,吸收到细胞中的FITC-标记-pDNA内容物由FACS进行比较。
作为分析结果,将通过使FITC-标记的Ad pDNA与PEI、PAM-ABP和PAM-ABP/VI-9R(pDNA∶VI-9R的重量比为1∶5或1∶10)反应形成的复合物转染到细胞中,并且在2小时之后,进行FACS。作为分析结果,当使用PAM-ABP时,与用作阳性对照的Ad pDNA-PEI相比,细胞摄取效率显著提高。特别地,当使用Ad pDNA-PAM-ABP-VI-9R复合物时,与仅使用聚合物的PAM-ABP的情况相比,细胞摄取效率提高1.2倍至1.4倍(图6)。这样的结果似乎是由于VI-9R与pDNA和PAM-ABP结合而提高细胞渗透性,导致Ad pDNA-PAM-ABP/VI-9R复合物的细胞摄取效率的提高。
实施例5:Ad pDNA-聚合物-VI-9R复合物的癌细胞的内体逃脱(Endosome escape)效应
在实施例5中,使用PAM-ABP作为聚合物,并且使用pdE1-GFP作为腺病毒DNA。递送到细胞中的生物试剂的内体隔离(endosomal sequestration)作为增强基因递送效率的关键障碍。同时,已知包含胺、磺酰胺或羧酸的聚合物材料表现出使递送到细胞中的生物试剂通过破坏内体膜(endosome membrane)而从内体中逃脱的功能,导致非病毒基因递送效率提高并提高化学治疗剂的核内递送效率。在实施例5中,为了通过VI-9R验证病毒DNA的内体逃脱效应,进行细胞的荧光染色。将HEK293(1×105细胞)细胞系接种到具有盖玻片的6孔板中,并且在24小时之后,将细胞核和内体分别用DAPI和溶酶破碎剂(lysotracker)染色。之后,GFP表达性腺病毒DNA(pdE1-GFP)和PAM-ABP或PAM-ABP-VI-9R复合物的缀合物,即AdpDNA-PAM-ABP或Ad pDNA-PAM-ABP-VI-9R以2μg/mL处理,并且在4小时之后,细胞用4%甲醛溶液固定以使用共聚焦显微镜观察内体逃脱效应。
作为观察的结果(图7),可以看出,在用Ad pDNA-PAM-ABP处理细胞的情况下,溶酶破碎剂和GFP重叠,并且当只有Ad pDNA-PAM-ABP与病毒DNA结合并处理时,病毒DNA从内体中的逃脱效率较低。然而,可以观察到当Ad pDNA-PAM-ABP-VI-9R复合物处理时,GFP和溶酶破碎剂的荧光没有重叠,并且病毒(GFP)被转移到细胞核中,这意味着Ad pDNA-PAM-ABP-VI-9R复合物改善了病毒DNA的内体逃脱效应。这样的结果表明,当单独使用Ad pDNA-PAM-ABP时,由于聚合物-VI-9R复合物的使用,可以克服诸如低内体逃脱效应和基因递送效率降低的限制。
实施例6:Ad蛋白VI-衍生肽的修饰及其物理特性
为了进一步提高在上述实施例中确认的VI-9R的基因递送效率,将腺病毒病毒VI衍生肽(SEQ ID NO:1)以多种形式进行修饰(图8)。即,进一步添加疏水性氨基酸或将常规已知的细胞渗透肽(CPP)TAT进一步与肽的N末端连接。为了分析经修饰肽的物理特性,将每个肽以10μg/ml放入总体积为1ml的比色杯中,使用Zeta-sizer测量溶解于pH 7.4PBS和pH6.4PBS中的肽的表面电荷值。
作为ζ电位分析结果(图9),VI-9R在pH 7.4的环境中显示出16.6±0.41mV的负电荷,并且通过进一步添加R序列,VI-9R肽表现出4.87±0.42mV的正电荷。与VI肽比较,经修饰的VI(mVI)的表面电荷已经变为正值。此外,随着pH降低,添加双极性氨基酸谷氨酸(E)的mVI-TAT-A或mVI-A-TAT的表面电荷增加。
实施例7:Ad pDNA-聚合物-肽复合物的细胞内基因递送效率
在实施例7中,使用PPCBA和PAM-ABP作为聚合物。为了确认Ad pDNA(dE1/GFP)-PPCBA-肽复合物的细胞内基因递送效率,将293A细胞系接种到24孔板中并生长到60%至70%汇合,用6种类型的修饰肽和Ad pDNA-PPCBA-肽复合物中的每一个处理细胞。使用VI-9R、NLS、TAT和鱼精蛋白作为对照,并且用复合物处理,在24小时之后,使用荧光显微镜观察GFP表达。
作为分析结果,当Ad pDNA-PPCBA-VI-9R复合物处理时,与当Ad pDNA或Ad pDNA-PPCTBA处理时比较,GFP表达显著提高。此外,当6种类型的经修饰肽,例如mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT或mVI-G-A-TAT中的每一个在与Ad pDNA-PPCTBA形成复合物之后处理时,与未经修饰的VI-9R时相比,GFP表达提高(图10)。
为了确认使用另外聚合物的Ad pDNA(dE1/GFP)-PAM-ABP-肽复合物(即PAM-ABP)的细胞内基因递送效率,将293A细胞系接种到24孔板中并生长至60%至70%汇合,并且在24小时之后,用使用6种类型的修饰肽mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT和mVI-G-A-TAT形成的Ad pDNA-PAM-ABP-肽复合物中的每一个处理。使用Ad pDNA-PAM-ABP/TAT和Ad pDNA/PEI25K作为对照并用复合物处理,在4天之后,使用荧光显微镜观察GFP表达。
作为分析结果,当用Ad pDNA-PAM-ABP-肽复合物处理时,与用Ad pDNA-PAM-ABP处理时比较,基因递送效率提高(图11)。特别地,与当用Ad pDNA-PAM-ABP/TAT作为阳性对照处理时比较,用其中PAM-ABP与6种类型的修饰肽mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT和mVI-G-A-TAT中的每一个缀合的Ad pDNA-PAM-ABP-肽复合物处理时,基因递送效率显著提高。这种结果表明经修饰肽可以提高Ad pDNA-PPCBA-肽复合物的基因递送效率。
实施例8:通过Ad pDNA-聚合物-肽复合物提高Ad pDNA的细胞摄取效率
在实施例8中,使用PAM-ABP作为聚合物。为了确认Ad pDNA-PAM-ABP-肽复合物的细胞摄取效率,将A549肺癌细胞系接种到12孔板中并生长至60%至70%汇合,并且在24小时之后,用FITC-标记Ad pDNA(dEl/GFP)和PAM-ABP-VI-9R或多种类型的PAM-ABP-修饰肽复合物(mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT和mVI-G-A-TAT)处理。使用AdpDNA-PAM-ABP-NLS和Ad pDNA-PAM-ABP-TAT作为对照并且用复合物处理,在30分钟之后,比较吸收到细胞中的FITC-pDNA的量并通过FACS分析。
与Ad pDNA-PAM-ABP作为对照的情况相比,使用VI-9R和6种类型的经修饰肽(mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT和mVI-G-A-TAT)形成的Ad pDNA-PAM-ABP-肽复合物的细胞摄取效率分别提高了2.4倍、2.6倍、2.5倍、2.0倍、2.1倍、1.6倍和2.1倍(图12)。特别地,与其他阳性对照如Ad pDNA-PAM-ABP/NLS和Ad pDNA-PAM-ABP/TAT相比,它们也显示出显著提高的细胞摄取效率。
实施例9:通过Ad pDNA-肽复合物分析DNA缩合效率
为了确认Ad pDNA-肽复合物的DNA缩合效率,通过PicoGreen染色确认DNA缩合效率。将已知与DNA特异性结合的PicoGreen染料(100μl)与多种形式的Ad pDNA-肽复合物(1μg)反应后,使用微孔板读取仪确认荧光表达的程度,并确认缩合效率。DNA缩合越高,荧光表达的程度越低。
作为分析结果,在Ad pDNA-VI的情况下,几乎不发生DNA缩合,并且在经修饰肽中,可以确认在mVI-TAT、mVI-TAT-A和mVI-A-TAT的情况下,诱导DNA缩合,但是在mVI、mVI-G-TAT-A和mVI-G-A-TAT的情况下,未诱导DNA缩合(图13)。
实施例10:通过肽改善膜裂解活性
内体逃脱是其中DNA通过内体中的膜的裂解逃脱的现象,并且进行PI染色以确认Ad-聚合物-肽复合物的膜的破坏程度。本文使用的每种肽在37℃孵育器中以50nM反应30分钟。之后,100μg的PI在37℃下反应10分钟,培养基更换之后,使用荧光显微镜检查结果。膜裂解程度越高,荧光表达程度越高。
作为分析结果,可以确认当用作为阳性对照的肽TAT处理时,几乎不发生膜裂解,但是当用肽(VI-9R、mVI-TAT-A、mVI-A-TAT和mVI-G-A-TAT)处理时,膜裂解效率显著提高(图14)。为此,预期经修饰肽表现出改善的内体逃脱效应。
实施例11:Ad pDNA-聚合物-肽复合物增加核定位(Nuclear localization)
在实施例11中,使用PAM-ABP作为聚合物。为了比较Ad pDNA-聚合物-肽复合物改善的核定位,将A549细胞系接种到24孔板中。在Ad DNA用FITC标记之后,使用FITC-标记的Ad DNA制备Ad pDNA-PAM-ABP复合物和Ad pDNA-PAM-ABP-肽(mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT、mVI-G-A-TAT、y/、VI-9R、TAT和NLS)复合物,随后在细胞接种后24小时处理这些复合物。Ad pDNA-PAM-ABP-肽复合物处理之后4小时,根据制造商的方案使用核/胞质分级分离试剂盒(Cell Biolabs,San Diego,CA)在Ex/Em=495/519nm下测量细胞质和细胞核。为了相对比较,将各组的细胞质和细胞核的平均值的总和转换为100%。
作为分析结果,与Ad pDNA-PAM-ABP处理时相比,在Ad pDNA-PAM-ABP-肽复合物的所有情况下,可以确认Ad pDNA的核定位显著增加。此外,与阳性对照(Ad pDNA-PAM-ABP-NLS或Ad pDNA-PAM-ABP-TAT复合物)相比,Ad pDNA-PAM-ABP-肽复合物表现出提高的核定位比值(图15)。根据这样的结果,可以确认在Ad pDNA-PAM-ABP-肽复合物的情况下,Ad DNA的核定位通过肽的修饰而增加,导致改善的腺病毒复制。
实施例12:确认Ad pDNA-聚合物-肽复合物的病毒增殖能力和溶瘤能力
在该实例中,为了确认Ad pDNA-聚合物-肽复合物的腺病毒增殖能力和溶瘤能力,使用聚合物PAM-ABP和含有H-Rd19-k35/DCN/shcMet的病毒DNA的d pDNA(其为表达如核心蛋白聚糖(DCN)和shcMet的腺病毒DNA的肿瘤选择性溶瘤腺病毒)制造复合物。将人子宫癌细胞系(Hela细胞系)接种到12孔板中,并且在24小时之后,用裸Ad pDNA、Ad pDNA/PAM-ABP、Ad pDNA/PAM-ABP/肽复合物(其中6种类型的肽,例如mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT和mVI-G-A-TAT与Ad pDNA/PAM-ABP缀合)和对照如VI-9R和与TAT肽的复合物处理。此外,为了验证细胞毒性,用PAM-ABP或单独的肽处理细胞。处理之后四天,收集培养溶液,使用Q-PCR检测腺病毒颗粒的数目,为了验证溶瘤能力,将2mg/ml的PBS中的3-(4,5-二甲基噻唑-2-基)-2,5-二苯基-四唑溴化物(MTT;SigmaAldrich)以200μL添加到上清除去的板的每个孔中,并在37℃下培养4小时。之后,除去上清液,将沉淀物溶解于1.0ml二甲基亚砜中,通过在微孔板读取仪上在540nm处读板来分析吸光度,并将未处理的细胞组分类为阴性对照。
作为分析结果,可以确认当仅用裸Ad pDNA处理Hela细胞系时,病毒产量是1.5×103VP(这是基础水平),使用6种类型的肽形成的Ad pDNA/PAM-ABP/肽复合物的病毒产量,与Ad pDNA/PAM-ABP作为对照的那些相比,除了使用mVI-G-TAT-A的复合物,分别提高约8.0倍、11.6倍、34.1倍、29.5倍和25.6倍。特别地,可以确认与其他对照VI-9R或TAT相比,AdpDNA/PAM-ABP/肽复合物的病毒产量显著提高(图16A)。此外,为了确认溶瘤能力和细胞毒性,在除去上清液的各板用MTT溶液处理时,虽然VI-9R肽表现出高细胞毒性,但是mVI、mVI-G-TAT-A、mVI-A-TAT和mVI-G-A-TAT表现出相对高的细胞毒性,并且作为确认溶瘤能力的结果,与对照Ad pDNA/PAM-ABP相比,使用6种类型的肽形成的Ad pDNA-(PAM-ABP)-肽复合物表现出的溶瘤能力分别提高22.5%、18.7%、29.5%、7.1%、21.7%和26.5%(图16B)。特别地,可以推断使用VI-9R的Ad pDNA/PAM-ABP/肽复合物的最高溶瘤能力是由VI-9R本身毒性诱导的溶瘤效果引起的。
从结果可以看出,在Ad pDNA-(PAM-ABP)-肽复合物中,癌细胞中的病毒增殖非常成功,并且腺病毒在肿瘤细胞中增殖,从而导致癌症-特异性溶瘤能力,因此病毒增殖能力和溶瘤能力极大提高。
实施例13:Ad pDNA-聚合物-肽复合物的病毒增殖能力和溶瘤能力的验证
在本实施例中,为了验证Ad pDNA-聚合物-肽复合物的腺病毒增殖能力,使用聚合物PAM-ABP和Ad pDNA(包含绿色荧光蛋白(GFP)表达性肿瘤选择性腺病毒RdB/GFP之病毒DNA作为腺病毒DNA)。将人肺癌细胞系,例如A549细胞系接种在12孔板中,并且在24小时之后,用裸Ad pDNA、Ad pDNA/PAM-ABP、Ad pDNA/PAM-ABP/肽复合物(其中6种类型的肽,例如mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT和mVI-G-A-TAT与Ad pDNA-(PAM-ABP)缀合)和对照如VI-9R和与TAT肽的复合物处理,。处理之后四天,收集培养溶液,使用Q-PCR确认腺病毒颗粒的数量。
作为分析结果,可以确认,当仅用裸Ad pDNA处理A549细胞系时,病毒产量为1.2×103VP(其为基础水平),并且使用6种类型的肽形成的Ad pDNA-(PAM-ABP)-肽复合物的病毒产量,与对照Ad pDNA/PAM-ABP相比,除了使用mVI和mVI-G-TAT-A的复合物,提高了约1.95、2.01、1.03和10.7倍,并且特别地,与不同对照VI-9R或TAT相比,Ad pDNA-(PAM-ABP)-肽复合物的病毒产量也在显著提高(图17)。
实施例14:确认Ad pDNA-聚合物-二聚体肽复合物提高的细胞内基因递送效率
在该实例中,为了确认Ad pDNA(dE1/GFP)-(PAM-ABP)-二聚体肽复合物的细胞内基因递送效率,将293A细胞系接种到24孔板中,生长至60%至70%汇合,并用经修饰的单体肽、二聚体肽和Ad pDNA-(PAM-ABP)-二聚体肽复合物处理,并且在处理之后48小时,使用荧光显微镜观察GFP表达。
同时,使用空气氧化技术制造二聚体肽。具体地,将2mg的单体肽溶解于2mL的0.1M脱气的碳酸氢铵中,在打开盖子的同时将该混合物在室温下搅拌24小时。之后,使用透析盒分离单体肽,将剩余的反应物冷冻干燥,从而获得粉末。作为对照,使用TAT、用预修饰肽形成的复合物(mVI-G-A-TAT)、PEI25k以及其中PAM-ABP聚合物与Ad pDNA缀合形成的复合物。
此外,确认使用不同聚合物PPCBA-PEI-Arg的Ad pDNA(RdB/GFP)-(PPCBA-PEI-Arg)-肽复合物的细胞内基因递送效率,为此,将293A细胞系接种在24孔板中并生长至60%至70%汇合并且在24小时之后,通过与经修饰的二聚体肽以1∶20或1∶30(Ad pDNA∶PPCBA-PEI-Arg重量比)的比率结合制备Ad pDNA-(PPCBA-PEI-Arg)-二聚体肽,然后用于处理细胞。处理之后四天,使用荧光显微镜观察GFP表达。作为对照,使用Ad pDNA/PEI25K和AdpDNA/lipofectamine。
作为分析结果,当用Ad pDNA-聚合物-二聚体肽复合物处理时,与当用Ad pDNA-PEI25k或Ad pDNA-PAM-ABP处理时相比,GFP表达显著提高。此外,可以确认当mVI-G-A-TAT肽被修饰时,GFP表达与未经修饰肽的GFP表达类似,但是当经修饰的单体肽变成二聚体肽时,则Ad pDNA-聚合物-肽复合物处理与对照相比,GFP表达显著地提高(图18)。
此外,可以确认,当Ad pDNA-(PPCBA-PEI-Arg)-二聚体肽复合物处理由Ad pDNA、聚合物和肽以1∶30∶1的重量比组成时,与对照处理例如Ad pDNA-PEI25K或Ad pDNA-lipofectamine时相比,GFP表达显著提高,并且通常地,当Ad pDNA:聚合物的重量比为30∶1时,与当重量比为20∶1时相比,由此引起的效应是优异的并且结合的二聚体肽的量提高,GFP表达浓度依赖性地提高(图19)。即,从结果可以看出,通过以二聚体形式修饰本发明的肽可以进一步改善细胞内递送效率。
实施例15:Ad pDNA-聚合物-二聚体肽复合物的病毒增殖能力和溶瘤能力的验证
在该实例中,为了确认Ad pDNA-聚合物-二聚体肽复合物的腺病毒增殖能力,使用聚合物PPCBA-PEI-Arg(PPR)和包含肿瘤选择性溶瘤腺病毒RdB/IL-12/GMCSF(表达IL-12和GMCSF作为腺病毒DNA)之病毒DNA的Ad pDNA来制造复合物。将人前列腺癌细胞系,即PC-3细胞系接种在12孔板中,并且在24小时之后,用使用裸Ad pDNA、PPCBA-PEI-Arg(PPR)、二聚体肽、Ad pDNA-lipofectamine、Ad pDNA-PPR、Ad pDNA-PPR-二聚体肽(1∶30∶0.01,1∶30∶0.05[Ad pDNA∶聚合物∶肽重量比])制备的复合物中的每一个处理。处理之后四天,收集培养溶液并且使用Q-PCR鉴定腺病毒颗粒的数目。此外,将仓鼠胰腺癌细胞系,即HapT1细胞系接种在12孔板中,并且在24小时之后,用使用裸Ad pDNA、PPCBA-PEI-Arg(PPR)、二聚体肽、AdpDNA-lipofectamine、Ad pDNA-PPR、Ad pDNA-PPR-二聚体肽(1∶30∶0.01,1∶30∶0.05[AdpDNA∶聚合物∶肽重量比])制备的复合物中的每一个处理。处理之后四天,收集培养溶液,并且使用Q-PCR鉴定腺病毒颗粒的数目。
而且,为了验证溶瘤能力,将200μL的2mg/ml的PBS中的3-(4,5-二甲基噻唑-2-基)-2,5-二苯基-四唑溴化物(MTT;Sigma Aldrich)添加到上清除去的板的每个孔中,并在37℃下培养4小时。之后,除去上清液,将沉淀物溶解于1.0ml的二甲基亚砜中,通过在微孔板读取仪上在540nm处读板来分析吸光度。将未处理的细胞组分类为阴性对照。
作为分析结果,当PC-3细胞系仅用裸Ad DNA处理时,病毒产量为4.6×103VP,这是基础水平。可以确认,当用Ad pDNA-PPR-二聚体肽复合物处理的组中肽和Ad pDNA的重量比为1∶0.01时,病毒产量处于最高水平(9.3×107VP),比用对照Ad pDNA-PPR处理的细胞系高7-3倍(2.4×106VP),并且也比用阳性对照Ad pDNA-lipofectamine复合物处理的细胞系高3.7倍(2.5×107VP)(图20A)。此外,当HapT1细胞系仅用裸Ad pDNA处理时,病毒产量为4.6×103VP,这是基础水平。如上述的结果一样,可以确认当用Ad pDNA-PPR-二聚体肽复合物处理的组中肽和Ad pDNA的重量比为1∶0.01时,病毒产量处于最高水平(6.5×107VP),比用对照Ad pDNA/PPR处理的细胞系高99.4倍(6.5×105VP),并且也比用阳性对照Ad pDNA-lipofectamine复合物处理的细胞系高2.6倍(2.4×107VP)(图21A)。
此外,作为用于验证溶瘤能力的MTT测定的结果,在PC-3细胞系中,当用Ad pDNA-PPR-二聚体肽复合物处理的组中肽和Ad pDNA的重量比为1∶0.01时,溶瘤能力为最高水平57.6%,与用Ad pDNA-lipofectamine复合物或Ad pDNA-PPR组作为对照组处理相比,溶瘤效果分别提高了17.1%和16.2%(图20B)。此外,即使在HapT-1细胞系中,当用Ad pDNA-PPR-二聚体肽复合物处理的组中肽和Ad pDNA的重量比为1∶0.01时,溶瘤能力为最高水平57.3%,与Ad pDNA-lipofectamine复合物处理的组或Ad pDNA-PPR组相比,溶瘤效果分别提高了4%和10.5%(图21B)。
从结果可以看出,当用Ad pDN-/PPR-二聚体肽处理的组中的肽和Ad pDNA的重量比为1∶0.01时,表现出最高的腺病毒增殖能力和最高的溶瘤能力。这样的结果表明AdpDNA-PPR-二聚体肽复合物在癌细胞中表现出非常成功的病毒增殖,腺病毒在肿瘤细胞中增殖从而表现出癌细胞特异性的溶瘤能力,因此病毒增殖能力和溶瘤能力极大提高。
实施例16:腺病毒/肽复合物的细胞内基因递送效率的确认
在该实例中,为了确认腺病毒(dE1/GFP)-肽复合物的细胞内递送效率,将多种细胞系(A549、MCF、H460和U434)接种在24孔板中并且生长至60%至70%汇合,在24小时之后,将8种类型的肽,例如VI、VI-9R、mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT和mVI-G-A-TAT与腺病毒(dE1/GFP)以1∶1×105、1∶1×106和1∶1×107摩尔比缀合,然后以20MOI使用以处理细胞。在处理的48小时之后,使用荧光显微镜观察GFP表达。使用既不与NLS、TAT也不与肽缀合的腺病毒(只有腺病毒)作为使用A549细胞系的实验的对照,并且使用既不与VI、VI-9R、NLS、TAT也不与肽缀合的只有腺病毒的组作为其他细胞系的对照。
将腺病毒(dE1/GFP)/肽复合物转染到细胞中,然后比较基因递送效率。与只有腺病毒的组相比,Ad/肽复合物的基因递送效率通常较高(图22至25)。特别地,可以确认当6种类型的经修饰肽,例如mVI、mVI-TAT、mVI-TAT-A、mVI-G-TAT-A、mVI-A-TAT和mVI-G-A-TAT与Ad缀合时,与Ad/VI和Ad/VI-9R组相比,Ad/肽复合物的基因递送效率显著提高。
因此,可以看出,本发明的肽可以提高核酸如腺病毒DNA的细胞内递送效率,并且还提高病毒本身的细胞内递送效率,并且这种趋势可以通过肽修饰来增强。
实施例17:确认化学治疗剂/肽复合物提高的细胞摄取效率
在该实例中,为了确认化学治疗剂/肽复合物提高的细胞摄取,将U343或H460细胞系接种在12孔板中并生长至70%至80%汇合。具体地,在物理结合(DOX+mVI-G-A-Tat)的情况下,化学治疗剂(即多柔比星和mVI-G-A-TAT肽)放入包含PBS缓冲液的管中,在室温下储存20分钟并在生理条件下反应,然后将5μM的细胞用复合物处理。此外,在化学结合(mVI-G-A-Tat缀合的DOX)的情况下,将DOX(820μg)和N-β-马来酰亚胺丙酸酰肼(BMPH)作为交联剂(1.1当量)添加到800吐的DMSO缓冲液中并混合。相应的反应在黑暗条件下于室温下进行24小时,然后添加肽(4mg)以使DOX∶mVI-G-A-Tat肽的比例为1.1当量,并在室温下反应3小时。最后,使用3.5kDa透析盒去除未反应的化学治疗剂,冻干形成粉末并溶于PBS中,然后用5μM的复合物处理细胞。在处理后5、10、30和120分钟,通过FACS定量细胞多柔比星摄取,并且仅用化学治疗剂处理的组作为对照。
作为用化学治疗剂/肽复合物处理细胞并确认细胞摄取的结果,与仅用化学治疗剂处理的对照相比,化学治疗剂/肽复合物的细胞摄取提高,并且特别地,可以确认,当化学治疗剂与肽物理结合时,初始时间细胞药物摄取的效率极大提高(图26)。
因此,可以看出,细胞化学治疗剂摄取的量通过与化学治疗剂一起细胞内递送经修饰的Ad蛋白VI衍生的肽来提高,导致治疗效果的进一步改善。
实施例18:化学治疗剂/肽复合物提高溶瘤效果的确认
在该实例中,为了确认化学治疗剂/肽复合物提高溶瘤效果,将H460、A549或U343细胞系接种在96孔板中并生长至60%至70%汇合,并且在24小时之后,将化学治疗剂,例如MG132或多柔比星与mVI-G-A-TAT肽物理结合(mVI-G-A-Tat+MG132,DOX+mVI-G-A-Tat)或化学结合(mVI-G-A-Tat缀合的MG123/DOX),然后以0.1μM至5μM处理。在处理之后48小时,通过进行MTT测定确认细胞生存力,将仅用mVI-G-A-TAT肽、MG132或多柔比星处理的组作为对照。
作为确认用化学治疗剂/肽复合物处理细胞并确认溶瘤效果的结果,与仅用化学治疗剂处理的对照组相比,可以确认化学治疗剂/肽复合物的溶瘤效果提高,并且特别地,当化学治疗剂与肽物理结合时,与化学结合相比,可以确认其效果进一步提高(图27和图28)。
因此,可以看出,由化学治疗剂引起的溶瘤能力通过与化学治疗剂一起细胞内递送经修饰的Ad蛋白VI衍生肽来提高。
实施例19:确认通过蛋白质/肽复合物提高了向癌细胞的递送效率
在该实例中,为了确认蛋白质(人血清白蛋白)/肽复合物的细胞内递送效率,将肺癌细胞系,即A549细胞系接种在96孔板中并生长至70%至80%汇合,并且在24小时之后,将FITC-标记的人血清白蛋白(HSA)和mVI-G-A-TAT肽物理结合,然后在0.5μM或5μM下处理(是指实施例17中所述的物理结合过程)。处理之后0.16、0.5、2或6小时,使用微孔板读取仪确认细胞内FITC标记的HSA摄取,将仅用蛋白处理的组作为对照。
作为用蛋白质(HSA)/肽复合物处理细胞并确认复合物的细胞摄取的结果,与仅用蛋白质处理的对照相比,可以确认蛋白质/肽复合物的细胞摄取提高,并且特别地与仅用蛋白质处理的组相比,在用0.5或5μM蛋白质处理的所有实验组中,当也用本发明的肽处理时,表现出细胞内蛋白质递送效率的统计学上的显著提高(图29)。因此,可以看出,通过细胞内递送经修饰的Ad蛋白VI衍生肽和蛋白质,蛋白质的细胞内递送效率进一步提高。
实施例20:确认通过抗体/肽复合物提高的细胞内抗体递送效率
在该实例中,意图鉴定抗体(兔多克隆抗TGFβ1)/肽复合物的细胞内递送效率。转化生长因子β(TGF-β)是存在于细胞质中的蛋白质,已知在癌细胞中过表达,并且对于由外部施用的抗体诱导的抗原-抗体反应,需要对细胞膜具有高渗透性。在该实施例中,为了确认修饰的Ad蛋白VI衍生肽是否可以将抗体递送到细胞中,用抗体(兔多克隆抗TGFβ1)/肽复合物处理细胞而不透化,确认细胞内递送是否发生。具体地,将肺癌细胞系,即A549细胞系接种在腔室载玻片上并生长至70%至80%汇合,并且在24小时之后,将抗体与mVI-G-A-TAT肽以1∶1的重量比物理结合(是指实施例17中所述的物理结合过程),然后在4μg下处理。在处理之后2小时,通过使用Alexa Fluor 488缀合抗体(Alexa Fluor 488山羊抗兔IgG(H+L))的荧光显微镜观察细胞抗体摄取,并将仅用抗体处理的组作为对照。
作为分析结果,在仅用抗体处理的对照中,可以确认大部分抗体未被递送到细胞中,但是大量抗体被递送到用抗体/肽复合物处理的组中的细胞中(图30)。通过其可见,通过细胞内递送经修饰的Ad蛋白VI衍生肽和抗体进一步提高抗体的细胞内递送效率。
以上,已经详细描述了本发明的具体部分。对于本领域的普通技术人员来说显而易见的是,这样的具体描述仅仅是具体实施方案,并且本发明的范围不限于此。因此,本发明的实质范围由所附权利要求及其等同物限定。

Claims (13)
1.用于对生物活性物质进行细胞内递送的组合物,其包含:
由选自SEQ ID NO:4至8的氨基酸序列组成的肽;或来自其的二聚体。
2.根据权利要求1所述的组合物,其中所述生物活性物质是选自以下的任一种:核酸、肽、抗体、化学治疗剂和病毒。
3.根据权利要求1所述的组合物,其还包含:
生物相容性聚合物。
4.根据权利要求3所述的组合物,其中所述生物相容性聚合物是包含以下的聚合物:(i)可从免疫反应逃脱的部分、(ii)可带电荷部分和(iii)具有二硫键的可生物还原部分。
5.根据权利要求4所述的组合物,其中所述可从免疫反应逃脱的部分是选自以下的任一种:聚乙二醇(PEG)、聚环氧烷、聚苯醚-PEG-聚环氧烷共聚物、聚(甲基丙烯酸甲氧乙酯)、聚(甲基丙烯酰基磷脂酰胆碱)、全氟化聚醚、右旋糖酐和聚乙烯吡咯烷酮。
6.用于治疗癌症的药物组合物,其包含:
(a)治疗有效量的复合物,其包含肽、生物相容性聚合物和腺病毒DNA,其中所述肽由选自SEQ ID NO:4至8的氨基酸序列组成;或为来自其的二聚体;以及(b)可药用载体。
7.根据权利要求6所述的药物组合物,其中所述生物相容性聚合物是包含以下的聚合物:(i)可从免疫反应逃脱的部分、(ii)可带电荷部分和(iii)具有二硫键的可生物还原部分。
8.根据权利要求7所述的药物组合物,其中所述可从免疫反应逃脱的部分是选自以下的任一种:聚乙二醇(PEG)、聚环氧烷、聚苯醚-PEG-聚环氧烷共聚物、聚(甲基丙烯酸甲氧乙酯)、聚(甲基丙烯酰基磷脂酰胆碱)、全氟化聚醚、右旋糖酐和聚乙烯吡咯烷酮。
9.根据权利要求6所述的药物组合物,其中所述腺病毒DNA是完全腺病毒基因组DNA、部分缺失的腺病毒基因组DNA或经部分修饰的腺病毒基因组DNA。
10.根据权利要求6所述的药物组合物,其中所述癌症是乳腺癌、肺癌、胃癌、肝癌、血癌、骨癌、胰腺癌、皮肤癌、头颈癌、真皮或眼部黑素瘤、子宫肉瘤、卵巢癌、直肠癌、肛门癌、结肠直肠癌、输卵管癌、子宫内膜癌、子宫颈癌、小肠赘生物、内分泌癌、膀胱癌、喉癌、骨肉瘤、甲状腺癌、脑癌、结肠癌、阴道癌、外阴癌、霍奇金病、食管癌、肾上腺癌、阴茎癌、急性白血病、淋巴细胞淋巴瘤、输尿管癌、中枢神经系统(CNS)肿瘤、脊髓肿瘤、鼻咽癌、甲状旁腺癌、肾癌、软组织肉瘤、尿道肿瘤、前列腺癌、支气管癌或骨髓癌。
11.根据权利要求10所述的药物组合物,其中所述肺癌包括非小细胞肺癌。
12.根据权利要求10所述的药物组合物,其中所述脑癌包括原发性CNS淋巴瘤、脑干胶质瘤或垂体腺瘤。
13.根据权利要求10所述的药物组合物,其中所述肾癌包括肾盂癌。
