本发明涉及草果精油及其乳剂。具体涉及草果精油及其制备方法,以及草果精油乳剂及其制备方法。所述草果精油分离自草果,草果精油及其乳剂具有抑菌和抗菌的作用。
Description
技术领域
本发明涉及植物源的草果精油,其制备方法,及其在抑菌、抗菌中的应用。
背景技术
化学合成制剂目前是空气消杀、病虫害防治、防霉的重要手段,具有防效好、作用快的特点,但是长期、大量、不合理的使用化学制剂,尤其是高毒、高残留化学药剂,造成环境污染、危害人畜健康、防效降低、病菌产生抗药性、农药残留超标等诸多问题。
天然植物是地球上的资源宝库,人类对天然植物源物质的研究已有几千年的历史,是药物的主要来源。近年来,随着人们对植物源制剂天然、健康、低毒、无化学残留等特点的认识,全球市场对天然健康原料产品的需求也日益增加,植物提取物行业的发展越来越快。同时,天然植物源的制剂通常是多种类型成分的混合物,含有多种化学成分,往往含有几十种到上百种成分,微生物不易产生抗药性。因此,近年来,植物源制剂的研究开发和应用逐步得到业内人士的重视,并得到了长足的发展。
目前植物源制剂的开发主要有两种方式,一是从植物中寻找活性成分,作为合成高效低毒新化学制剂的先导化合物,二是直接利用植物资源制成植物源制剂。我国植物资源非常丰富,所以植物源物质和制剂的开发多以第二种方式为主。目前已有部分植物源制剂产品问世,但其中绝大多数为植物源杀虫剂,而植物源杀菌/抑菌剂则较少。
草果(Amomum tsaoko)是姜科(Zingiberaceae)、豆蔻属(Amomum)多年生草本植物,主要分布于我国的云南、广西、贵州等省区。草果具有特殊浓郁的辛辣香味,主要作调味香料,是烹调佐料中的佳品。
发明内容
本发明利用草果的干燥成熟果实作为原料提取精油,获得了具有独特组成的草果精油,所述草果精油具有抑菌、抗菌的活性。
第一方面,本发明提供了一种草果精油,所述草果精油含有以重量计如下含量的特征成分:
(1)1,8-桉叶油素:45-65%;
(2)柠檬醛:1-3.5%。
优选地,以重量计,1,8-桉叶油素的含量为50-63%。优选地,以重量计,1,8-桉叶油素的含量为58-63%。
第二方面,本发明提供了第一方面的草果精油的制备方法,所述方法包括:
(1)称取一定量的干燥草果,敲成小块或粉碎成粗粉;
(2)以干燥草果小块或粗粉作为原料提取草果精油;以及
(3)用挥发油提取器收集草果精油,并计算得油率。
提取草果精油的方法选自:水蒸气蒸馏法、溶剂提取法、CO2超临界萃取技术、亚临界水萃取和微波辅助萃取法。
在采用水蒸气蒸馏法的提取方法中:
以干燥草果小块或粗粉和水的重量比为1:3-1:20,将干燥草果小块或粗粉在水中浸泡0-24小时;然后水蒸气蒸馏提取0.5-10小时;用挥发油提取器收集草果精油,并计算得油率;
优选地,以干燥草果小块或粗粉和水的重量比为1:3-1:10,将干燥草果小块或粗粉在水中浸泡0-4小时;然后水蒸气蒸馏提取3-8小时;用挥发油提取器收集草果精油,并计算得油率。
优选地,以干燥草果小块或粗粉和水的重量比为1:7,将干燥草果小块或粗粉在水中浸泡1小时;然后水蒸气蒸馏提取7小时;用挥发油提取器收集草果精油,并计算得油率。
干燥草果可以是经晾干获得的干燥草果,或者是经烤干获得的干燥草果。
本发明的草果精油的得油率以重量计在1.0%-2.5%的范围内。本发明的草果精油为淡黄色透明油状物,具有特殊的浓郁香味。
本发明的草果精油具有针对选自如下的微生物的抗菌和抑菌效果:大肠杆菌(Escherichia coli)、金黄色葡萄球菌(Staphylococcus aureus)、白色念珠菌(Candidaalbicans)、黄曲霉(Aspergillus flavus)、枝状枝孢霉(Cladosporiumcladosporioides)、波兰青霉(Penicillium polonicum)、立枯丝核(Rhizoctoniasolani),灰葡萄孢菌(Botrytis cinerea),人参链格孢(Alternaria panax),茄病镰刀(Fusarium solani),尖孢镰刀(Fusarium oxysporum),拟参土赤壳(Ilyonectria mors-panacis)和强壮土赤壳(Ilyonectria robusta)。
本发明的草果精油作为抑菌、抗菌的活性成分,可以配制成固体、液体、乳剂、喷雾剂和气雾剂型的抑菌剂。在气雾剂的情况,所用的抛射剂选自氟氯烷烃类、碳氢化合物和压缩气体。
第三方面,本发明提供了草果精油的乳剂型抑菌剂,所述乳剂型抑菌剂按重量计的组成如下:
(1)草果精油10份;
(2)溶剂20-35份;
(3)助剂5-15份;和
(4)水40-55份。
所述溶剂选自:甲醇、乙醇、正丁醇、正丙醇、异丙醇、异丁醇、正戊醇、异戊醇、正己醇、丙酮、二氯甲烷、乙酸乙酯、二甲亚砜、四氢呋喃、二甲基甲酰胺、环己酮、吡咯烷酮、N-甲基吡咯烷酮,或其中两种或多种的混合物。优选地,所述溶剂选自:乙醇、甲醇、丙醇、异丙醇、丁醇、丙三醇、丙酮等,或其中两种或多种的混合物。优选地,所述溶剂为甲醇、乙醇或二者的混合物。
所述助剂为表面活性剂、载体和/或分散剂,所述表面活性剂选自吐温类表面活性剂、司盘类表面活性剂、十二烷基硫酸钠、醇类、丙二醇、聚乙二醇和甘油,所述分散剂选自六偏磷酸钠、焦磷酸钠、聚丙烯酸钠盐和聚乙烯醇,所述载体选自脂质体、环糊精和微囊。优选地,所述助剂为表面活性剂和/或载体,所述表面活性剂选自吐温类、烷基酚聚氧乙烯醚类、司盘类、十二烷基硫酸钠、醇类,所述载体选自脂质体、环糊精和微囊。优选地,所述表面活性剂为吐温类、烷基酚聚氧乙烯醚类。
本发明的草果精油的乳剂型抑菌剂选自:
由草果精油1份,乙醇3.5份,吐温类表面活性剂1.5份和水4份搅拌均匀获得的乳剂;
由草果精油1份,乙醇2份,烷基酚聚氧乙烯醚类表面活性剂1.5份和水5.5份搅拌均匀获得的乳剂;
由草果精油1份,乙醇3.5份,烷基酚聚氧乙烯醚类表面活性剂0.5份和水4份搅拌均匀获得的乳剂;
由草果精油1份,乙醇3.5份,烷基酚聚氧乙烯醚类表面活性剂1.5份,水4份搅拌均匀获得的乳剂;以及
由草果精油1份,乙醇2.5份,甲醇1份,烷基酚聚氧乙烯醚类表面活性剂1.5份和水4份搅拌均匀获得的乳剂。
本发明的草果精油的乳剂型抑菌剂具有针对如下微生物的抗菌和抑菌效果:大肠杆菌(Escherichia coli)、金黄色葡萄球菌(Staphylococcus aureus)、白色念珠菌(Candida albicans)、黄曲霉(Aspergillus flavus)、枝状枝孢霉(Cladosporiumcladosporioides)、波兰青霉(Penicillium polonicum)、立枯丝核(Rhizoctoniasolani),灰葡萄孢菌(Botrytis cinerea),人参链格孢(Alternaria panax),茄病镰刀(Fusarium solani),尖孢镰刀(Fusarium oxysporum),拟参土赤壳(Ilyonectria mors-panacis)和强壮土赤壳(Ilyonectria robusta)。
而且本发明的草果精油乳剂具有相比于草果精油本身预料之外的抗菌、抑菌效果。
本发明的抑菌剂安全无毒、无刺激性,既可用于室内空气及室内设备设施的消杀和清新,也可以用于植物病害防治,还可以用于防霉。

具体实施方式
下面结合实施例及说明书附图对本发明的技术方案做进一步描述。这些实施例仅用于说明本发明而不用于限定本发明的保护范围。
实施例1:草果精油的制备
分别称取晾干及烤干的草果各0.5kg,粉碎成粗粉,在3.5L水中浸泡1h。水蒸气蒸馏提取7h,用挥发油提取器收集草果精油,计算得油率=(得油量/称取草果量)×100%,结果为晾干草果的得油率为2.0%,烤干草果的得油率为1.3%。
实施例2:草果精油的表征
草果精油的成分分析
采用GC/MS法分析由晾干及烤干草果提取的草果精油的成分及含量。将草果精油用正己烷稀释10000倍,进样1μl。GC条件:采用Thermo TR-5MS(30m*0.32mm,0.25μm)色谱柱,柱初温50℃,程序升温6℃/min,升至220℃,然后以10℃/min升至250℃,恒温5min。进样口温度250℃,载气He,载气流量2.0ml/min,分流比20:1。MS条件:EI离子源,扫描模式fullscan,扫描范围m/z 50-600amu,离子源温度250℃,GC/MS接口温度250℃。
草果精油的GC-MS色谱图参见附图1。结果表明,晾干草果提取的草果精油中共检出色谱峰29个,其中鉴定出25种挥发物,占精油总相对含量98.7%,其中1,8-桉叶油素的相对含量为58.11%,柠檬醛的相对含量为3.33%。烤干草果提取的草果精油中第一份样品共检出色谱峰33个,其中鉴定出30种挥发物,占精油总相对含量97.75%,其中1,8-桉叶油素的相对含量为62.73%,柠檬醛的相对含量为2.92%。第二份烤干草果样品提取的草果精油共检出色谱峰32个,其中鉴定出30种挥发物,占精油总相对含量97.12%,其中1,8-桉叶油素的相对含量为50.29%,柠檬醛的相对含量为3.34%。
具体的成分以及含量参见下表1:

由上表可以看出,由三种草果样品获得的三种草果精油中,两种主要成分和含量组成如下:
1,8-桉叶油素:50-63%
柠檬醛:2.9-3.4%。
实施例3:草果精油乳剂抑菌剂的制备
草果精油1份,乙醇3.5份,吐温80 1.5份,水4份,搅拌均匀,即得。
草果精油1份,乙醇2份,烷基酚聚氧乙烯醚(OP10)1.5份,水5.5份,搅拌均匀,即得。
草果精油1份,乙醇3.5份,烷基酚聚氧乙烯醚(OP10)0.5份,水4份,搅拌均匀,即得。
草果精油1份,乙醇3.5份,烷基酚聚氧乙烯醚(OP10)1.5份,水4份,搅拌均匀,即得。
草果精油1份,乙醇2.5份,甲醇1份,烷基酚聚氧乙烯醚(OP10)1.5份,水4份,搅拌均匀,即得。
实施例4:草果精油与草果精油乳剂抑菌剂抑真菌试验
实验菌种:黄曲霉(Aspergillus flavus)和枝状枝孢霉(Cladosporiumcladosporioides)。
草果精油乳剂抑菌剂:采用实施例3中的乳剂,具体配方如下:
草果精油1份,乙醇3.5份,烷基酚聚氧乙烯醚(OP10)1.5份,水4份,搅拌均匀,即得。
毒力测定试验----菌丝生长速率法
1)将草果精油原油按照5000倍的稀释倍数添加至培养基,混匀,冷却,类似地,将草果精油乳剂抑菌剂按500倍(其中草果精油乳剂中草果精油的浓度与草果精油本身稀释5000倍时相同)的稀释倍数添加至培养基,混匀,冷却;
2)将草果精油按照4000倍的稀释倍数添加至培养基,混匀,冷却,类似地,将草果精油乳剂抑菌剂按500倍(其中草果精油乳剂中草果精油的浓度低于草果精油本身稀释4000倍时的浓度)的稀释倍数添加至培养基,混匀,冷却;
3)在以上1)的培养基中接种黄曲霉,在以上2)的培养基中接种枝状枝孢霉,每板接种10μL孢子悬液,6个重复,作为试验样本组;
3)不含药剂的平板分别接种3个菌种,每板接种10μL孢子悬液,6个重复,作为空白对照组;
4)于培养箱25℃培养7天。
评判规定:
测定菌落直径(mm),计算抑菌率,抑菌率=((对照组菌落直径-试验组菌落直径)/对照组菌落直径)×100%。
表2草果精油与草果精油乳剂抑菌剂抑真菌效果的比较
从上述表2结果可以看出,草果精油稀释5000倍时抑制率为39.8%,含10%草果精油的乳剂稀释500倍时达到相同浓度,对黄曲霉的抑制率高达72.2%,抑菌效果优于草果精油本身。草果精油稀释4000倍时对枝状枝孢霉的抑制率为70.19%,含10%草果精油的乳剂稀释500倍时低于此浓度,而抑制率高达100%,抑菌效果也优于草果精油本身。
实施例5:草果精油乳剂抑菌剂对细菌的抑菌作用
依据国家《消毒技术规范(2002年版)》(中华人民共和国卫生部,二00二年十二月)要求进行评价。
实验菌种:
大肠杆菌(Escherichia coli)ATCC8099、金黄色葡萄球菌(Staphylococcusaureus)金黄色亚种ATCC6538和白色念珠菌(Candida albicans)ATCC10231均购买自北京市疾病预防控制中心。
培养基:
营养肉汤培养基用于大肠杆菌和金黄色葡萄球菌的培养;
沙氏培养基用于白色念珠菌的培养。
草果精油乳剂抑菌剂的配方如下:
草果精油1份,乙醇3.5份,吐温80 1.5份,水4份,搅拌均匀,即得。
最小抑菌浓度测定(营养肉汤稀释法)----MIC(Minimal InhibitoryConcentration):
1)制备大肠杆菌、金黄色葡萄球菌和白色念珠菌悬液;
2)将草果精油乳剂用蒸馏水做对倍稀释成不同浓度的受试液,各取2.5mL受试液加入2.5mL双倍浓度的营养肉汤培养基和沙氏液体培养基试管中。浓度设定如下:抑制大肠杆菌的浓度梯度为:0.05%、0.1%、0.2%、0.4%、0.8%;抑制金黄色葡萄球菌的浓度梯度为:0.0625%、0.125%、0.25%、0.5%、1%;抑制白色念珠菌的浓度梯度为1%、2%、4%、8%、16%;
3)以2.5mL蒸馏水加入2.5mL双倍浓度的营养肉汤培养基和沙氏液体培养基试管中作为溶剂对照;
4)取0.1mL含108cfu/mL的大肠杆菌、金黄色葡萄球菌和白色念珠菌悬液接种于以上试管中,作为试验组样本;
5)取0.1mL含108cfu/mL的大肠杆菌、金黄色葡萄球菌和白色念珠菌悬液接种于不含草果精油的试管,作为阳性对照;
6)取0.1mL含108cfu/mL的大肠杆菌、金黄色葡萄球菌和白色念珠菌悬液接种于溶剂对照试管,作为溶剂对照;
7)取2支空白培养基作为阴性对照;
8)将大肠杆菌和金黄色葡萄球的样品组、对照组放置于37℃培养,24小时后观察结果。将白色念珠菌的样品组、对照组放置于28℃培养,48小时后观察结果。
评判规定:
当阳性对照管有细菌生长(浑浊),阴性对照管无菌生长(透明),试验组无菌生长的最高稀释度所对应的抗(抑)菌剂浓度,为该样品对受试菌的MIC。
MIC试验结果如下:草果精油对大肠杆菌的MIC为0.2%,对金黄色葡萄球菌的MIC为0.125%,对白色念珠菌的MIC为4%。
实施例6:草果精油乳剂抑菌剂对真菌的抑菌作用
(1)实验菌种:波兰青霉(Penicillium polonicum)
培养基:马铃薯琼脂培养基
草果精油乳剂抑菌剂的配方如下:
草果精油1份,乙醇3.5份,吐温80 1.5份,水4份,搅拌均匀,即得。
毒力测定试验----EC50(半最大效应浓度):
1)配制波兰青霉菌悬液,菌液浓度为108cfu/mL;
2)将草果精油乳剂以500、200、100、50、25μL/L的终浓度,添加至PDA培养基,混匀,冷却;
3)每个浓度分别接种波兰青霉,每板接种10μL,6个重复,作为试验样本组;
4)不含草果精油制剂的平板分别接种波兰青霉,每板接种10μL,6个重复,作为空白对照组;
5)于培养箱25℃培养7天。
评判规定:
测定菌落直径(mm),计算抑菌率,抑菌率=((对照组菌落直径-试验组菌落直径)/对照组菌落直径)×100%。将抑菌率换算成几率值(Y)、药剂质量浓度(μL/L)换算成对数值(X),求出毒力回归方程(Y=a+bX)和相关系数(R),计算抑制供试菌种生长50%的药剂浓度(EC50)。
草果精油乳剂抑菌剂对波兰青霉的抑菌结果如下表3所示。

(2)实验菌种:黄曲霉(Aspergillus flavus)、枝状枝孢霉(Cladosporiumcladosporioides)和产黄青霉(Penicillium chrysogenum)。
培养基:马铃薯琼脂培养基
草果精油乳剂抑菌剂的配方如下:
草果精油1份,乙醇3.5份,烷基酚聚氧乙烯醚(OP10)1.5份,水4份,搅拌均匀,即得。
毒力测定试验----菌丝生长速率法
1)将草果精油乳剂按500倍、1000倍的稀释倍数添加至培养基,混匀,冷却;
2)每个浓度分别接种3个菌种,每板接种10μL,6个重复,作为试验样本组;
3)不含药剂的平板分别接种3个菌种,每板接种10μL,6个重复,作为空白对照组;
4)于培养箱25℃培养7天。
评判规定:
测定菌落直径(mm),计算抑菌率,抑菌率=((对照组菌落直径-试验组菌落直径)/对照组菌落直径)×100%。
实验结果表明(表4),草果精油乳剂对黄曲霉(Aspergillus flavus)、枝状枝孢霉(Cladosporium cladosporioides)、产黄青霉(Penicillium chrysogenum)均有较强的抑制作用。

实施例7:草果精油乳剂对7种植物病原菌的抑制作用
实验菌种:立枯丝核(Rhizoctonia solani),灰葡萄孢菌(Botrytis cinerea),人参链格孢(Alternaria panax),茄病镰刀(Fusarium solani),尖孢镰刀(Fusariumoxysporum),拟参土赤壳(Ilyonectria mors-panacis),强壮土赤壳(Ilyonectriarobusta)分离自发病西洋参或人参。
草果精油乳剂:采用实施例3中如下配方的乳剂:
草果精油1份,乙醇3.5份,烷基酚聚氧乙烯醚(OP10)1.5份,水4份,搅拌均匀,即得。
培养基:马铃薯琼脂培养基
毒力测定试验----菌丝生长速率法
1)将培养7天的病原菌打成直径6mm的菌饼;
2)将草果精油乳剂按500倍的稀释倍数添加至培养基,混匀,冷却;
3)每板接种1个菌饼,每个浓度5个重复,作为试验样本组;
4)不含药剂的平板分别接种7个菌种,5个重复,作为空白对照组;
5)于培养箱25℃培养5天。
评判规定
测定菌落直径(mm),计算抑菌率,抑菌率=(对照组菌落直径-试验组菌落直径)/(对照组菌落直径-菌饼直径)×100%。
实验结果如下表6所示:

#表示对照组菌落直径在该时间点前达到9cm。
上表6显示草果精油乳剂对7种病原真菌均表现出抑制作用,特别是针对镰刀属和土赤壳属这样难以防治的真菌也表现出优良的抑菌效果。
结果讨论:
从以上实施例可以看出草果精油本身以及以草果精油作为活性成分的草果精油乳剂抑菌剂均有显著的抑制细菌、真菌的作用,而且相对于草果精油本身,乳剂有更显著的抑制真菌效果。
Claims (10)
1.草果精油,所述草果精油含有以重量计如下含量的特征成分:
(1)1,8-桉叶油素:45-65%;
(2)柠檬醛:1-3.5%。
2.根据权利要求1的草果精油,其中以重量计1,8-桉叶油素的含量为50-63%,优选58-63%。
3.权利要求1或2的草果精油的制备方法,所述方法包括如下步骤:
(1)称取一定量的干燥草果,敲成小块或粉碎成粗粉;
(2)以干燥草果小块或粗粉作为原料提取草果精油;以及
(3)用挥发油提取器收集草果精油,并计算得油率;
其中提取草果精油的方法选自:水蒸气蒸馏法、溶剂提取法、CO2超临界萃取技术、亚临界水萃取和微波辅助萃取法。
4.根据权利要求3的方法,在采用水蒸气蒸馏法的提取方法中:
干燥草果小块或粗粉和水的重量比为1:3-1:20,将干燥草果小块或粗粉在水中浸泡0-24小时;然后水蒸气蒸馏提取0.5-10小时;用挥发油提取器收集草果精油,并计算得油率;所述干燥草果可以是经晾干获得的干燥草果,或者是经烤干获得的干燥草果。
5.根据权利要求4的方法,以干燥草果小块或粗粉和水的重量比为1:4-1:10,将干燥草果小块或粗粉在水中浸泡0-4小时;然后水蒸气蒸馏提取3-8小时;用挥发油提取器收集草果精油,并计算得油率;所述干燥草果可以是经晾干获得的干燥草果,或者是经烤干获得的干燥草果。
6.根据权利要求4的方法,以干燥草果小块或粗粉和水的重量比为1:7,将干燥草果小块或粗粉在水中浸泡1小时;然后水蒸气蒸馏提取7小时;用挥发油提取器收集草果精油,并计算得油率;所述干燥草果可以是经晾干获得的干燥草果,或者是经烤干获得的干燥草果。
7.权利要求1或2的草果精油,以及由权利要求3至6任一项的方法制备的草果精油,能配制成固体、液体、乳剂、喷雾剂和气雾剂型的抑菌剂。
8.草果精油的乳剂型抑菌剂,所述乳剂型抑菌剂按重量计的组成如下:
(1)草果精油10份;
(2)溶剂20-35份;
(3)助剂5-15份;和
(4)水40-55份。
9.根据权利要求8的草果精油的乳剂型抑菌剂,所述溶剂选自:甲醇、乙醇、正丁醇、正丙醇、异丙醇、异丁醇、正戊醇、异戊醇、正己醇、丙酮、二氯甲烷、乙酸乙酯、二甲亚砜、四氢呋喃、二甲基甲酰胺、环己酮、吡咯烷酮、N-甲基吡咯烷酮,或其中两种或多种的混合物;优选地,所述溶剂选自:乙醇、甲醇、丙醇、异丙醇、丁醇、丙三醇、丙酮等,或其中两种或多种的混合物;
所述助剂为表面活性剂、载体和/或分散剂,所述表面活性剂选自吐温类表面活性剂、司盘类表面活性剂、十二烷基硫酸钠、醇类、丙二醇、聚乙二醇和甘油,所述分散剂选自六偏磷酸钠、焦磷酸钠、聚丙烯酸钠盐和聚乙烯醇,所述载体选自脂质体、环糊精和微囊,优选地,所述助剂为表面活性剂和/或载体,所述表面活性剂选自吐温类、烷基酚聚氧乙烯醚类、司盘类、十二烷基硫酸钠,所述载体选自脂质体、环糊精和微囊。
10.根据权利要求9的草果精油的乳剂型抑菌剂,其选自:
由草果精油1份,乙醇3.5份,吐温类表面活性剂1.5份和水4份搅拌均匀获得的乳剂;
由草果精油1份,乙醇2份,烷基酚聚氧乙烯醚类表面活性剂1.5份和水5.5份搅拌均匀获得的乳剂;
由草果精油1份,乙醇3.5份,烷基酚聚氧乙烯醚类表面活性剂0.5份和水4份搅拌均匀获得的乳剂;
由草果精油1份,乙醇3.5份,烷基酚聚氧乙烯醚类表面活性剂1.5份和水4份搅拌均匀获得的乳剂;以及
由草果精油1份,乙醇2.5份,甲醇1份,烷基酚聚氧乙烯醚类表面活性剂1.5份和水4份搅拌均匀获得的乳剂。
