本发明公开了一种禽大肠杆菌病活疫苗冻干保护剂,按照重量份数计,它包括海藻糖60‑80份、脱脂乳粉80‑100份、酵母粉2‑3份、蛋白胨3‑6份、葡甘露聚糖0.5‑5份、灭菌水1000份;该保护剂的配伍合理,能够使禽大肠杆菌病活疫苗在冻干过程中以及在运输和储存过程中保持高的活菌率,使疫苗具有良好的稳定性,从而具有较高的效力。本发明适用于制备禽大肠杆菌病活疫苗冻干保护剂。

禽大肠杆菌病活疫苗冻干保护剂
技术领域
本发明属于兽用菌苗领域,涉及一种疫苗冻干保护剂,具体地说是一种禽大肠杆菌病活疫苗冻干保护剂。
背景技术
目前,在我国动物疫苗中,细菌活疫苗占相当大的比例,为了确保细菌活疫苗在冻干过程以及在运输和储存过程中的活菌率,在制备细菌活疫苗的过程中一般会使用冻干保护剂。然而,国内疫苗企业常用的细菌活疫苗冻干保护剂组方简单,保护性能差,活菌率一般来说在冻干后很难超过 55%,如果在 2~8℃条件下保存,保存期只有短短的 3~6 个月,多数需在 -15℃条件下保存;并且随着运输和储存时间的延长,活菌率还会进一步降低,这导致疫苗的稳定性较差,进一步导致疫苗的效力较低,最终导致免疫失败。
发明内容
本发明要解决的技术问题,是提供一种禽大肠杆菌病活疫苗冻干保护剂,该保护剂的配伍合理,能够使禽大肠杆菌病活疫苗在冻干过程中以及在运输和储存过程中保持高的活菌率,使疫苗具有良好的稳定性,从而具有较高的效力。
为解决上述技术问题,本发明所采取的技术方案是:
一种禽大肠杆菌病活疫苗冻干保护剂,按照重量份数计,它包括如下组分:
海藻糖60-80份、脱脂乳粉80-100份、酵母粉2-3份、蛋白胨3-6份、葡甘露聚糖0.5-5份、灭菌水1000份。
作为本发明的一种限定,所述禽大肠杆菌病活疫苗冻干保护剂还包括4-10重量份的氯化钠。
作为本发明的进一步限定,所述禽大肠杆菌病活疫苗冻干保护剂,按照重量份数计,它包括如下组分:
海藻糖80份、脱脂乳粉80份、酵母粉2.5份、蛋白胨5份,氯化钠5份、葡甘露聚糖1份、灭菌水1000份。
本发明还有一种限定,所述禽大肠杆菌病活疫苗冻干保护剂的pH为6.8-7.2。
本发明的禽大肠杆菌病活疫苗冻干保护剂通过如下步骤制备得到:
称取本发明各组分并溶解于灭菌水中,定容并调节 pH 值在6.8-7.2,高压灭菌或过滤除菌,即得。
本发明的禽大肠杆菌病活疫苗冻干保护剂的保存条件为 2-8℃。
本发明的禽大肠杆菌病活疫苗冻干保护剂用于制备禽大肠杆菌病活疫苗,按照如下步骤进行:
禽大肠杆菌活疫苗发酵培养结束后,于3500r/min下离心20min,去上清,菌泥立即加入经过灭菌并预热至37℃的冻干保护剂中,充分混匀;再用冻干保护剂稀释至所需浓度,例如,每 1ml 含菌5×109,得半成品;分装,例如,每西林瓶含 2ml(100羽鸡用剂量),冻干,得成品。
本发明的禽大肠杆菌病活疫苗,是由河北科星药业有限公司制备的重组大肠杆菌BL21(DE3)/pEASY-E1/tsh和本发明提供的禽大肠杆菌病活疫苗冻干保护剂混合后冻干得到。本发明的禽大肠杆菌病活疫苗优选是鸡、火鸡、鸽子用的疫苗。
由于采用了上述的技术方案,本发明与现有技术相比,所取得的技术进步在于:
本发明所提供的禽大肠杆菌病活疫苗冻干保护剂,各组分配伍合理,成本低,能使禽大肠杆菌病活疫苗在冻干过程以及在运输和储存过程中均保持高的活菌率。使用本发明的冻干保护剂制备的禽大肠杆菌病活疫苗活菌减少率低于 30%,在 2-8℃下可放置 1-3个月,活疫苗的活菌数存活率高且具有良好的稳定性,从而具有较高的效力。
本发明适用于制备禽大肠杆菌病活疫苗冻干保护剂。
本发明下面将结合具体实施例作进一步详细说明。
具体实施方式
下述实施例中所用的试剂,如无特殊说明,均为现有的市售试剂,所用的实验方法,如无特殊说明,均为现有的实验方法。
实施例 1 一种禽大肠杆菌病活疫苗冻干保护剂
S1:称取海藻糖80g、脱脂乳粉100g、酵母粉3g、蛋白胨6g、氯化钠6g、葡甘露聚糖5g;
S2:将上述物质溶解于1000g灭菌水中,并用碳酸钠溶液调节 pH 值至 7.2;
S3:高压灭菌,即得冻干保护剂。
实施例2 一种禽大肠杆菌病活疫苗冻干保护剂
S1:称取海藻糖60g、脱脂乳粉80g、酵母粉2g、蛋白胨3g、氯化钠6g、葡甘露聚糖3g;
S2:将上述物质溶解于1000g灭菌水中,并用碳酸钠溶液调节 pH 值至6.8;
S3:高压灭菌,即得冻干保护剂。
实施例3 一种禽大肠杆菌病活疫苗冻干保护剂
S1:称取海藻糖70g、脱脂乳粉90g、酵母粉2.5g、蛋白胨5g、氯化钠10g、葡甘露聚糖1g;
S2:将上述物质溶解于1000g灭菌水中,并用碳酸钠溶液调节 pH 值至7.0;
S3:高压灭菌,即得冻干保护剂。
实施例4 一种禽大肠杆菌病活疫苗冻干保护剂
S1:称取海藻糖80g、脱脂乳粉80g、酵母粉2.5g、蛋白胨5g、氯化钠5g、葡甘露聚糖0.8g;
S2:将上述物质溶解于1000g灭菌水中,并用碳酸钠溶液调节 pH 值至7.1;
S3:高压灭菌,即得冻干保护剂。
实施例5 一种禽大肠杆菌病活疫苗冻干保护剂
S1:称取海藻糖80g、脱脂乳粉80g、酵母粉2g、蛋白胨3g、氯化钠10g、葡甘露聚糖0.5g;
S2:将上述物质溶解于1000g灭菌水中,并用碳酸钠溶液调节 pH 值至7.2;
S3:高压灭菌,即得冻干保护剂。
实施例6 一种禽大肠杆菌病活疫苗冻干保护剂
S1:称取海藻糖70g、脱脂乳粉90g、酵母粉2g、蛋白胨5g、氯化钠8g、葡甘露聚糖5g;
S2:将上述物质溶解于1000g灭菌水中,并用碳酸钠溶液调节 pH 值至7.0;
S3:高压灭菌,即得冻干保护剂。
实施例7 一种禽大肠杆菌病活疫苗冻干保护剂
S1:称取海藻糖75g、脱脂乳粉100g、酵母粉2.8g、蛋白胨6g、氯化钠10g、葡甘露聚糖1g;
S2:将上述物质溶解于1000g灭菌水中,并用碳酸钠溶液调节 pH 值至6.9;
S3:高压灭菌,即得冻干保护剂。
实施例8 对比冻干保护剂
S1:称取蔗糖100g、脱脂乳粉100g、谷氨酸钠120g,山梨醇50g;
S2:将上述物质溶解于1000g灭菌水中,并用氢氧化钠溶液调节 pH 值至7.0;
S3:高压灭菌,即得冻干保护剂。
实施例9 对比冻干保护剂
S1:称取甘油30g、明胶10g、谷氨酸钠100g,甘露醇8g,硫酸铵20g,维生素C 5g;
S2:将上述物质溶解于1000g灭菌水中,并用氢氧化钠溶液调节 pH 值至7.2;
S3:高压除菌,即得。
实施例 10 不同冻干保护剂的性能测试
本实施例对实施例1-9所制备的冻干保护剂进行性能测试,具体如下:
Ⅰ.以M9为培养基,发酵生产禽大肠杆菌病活疫苗,发酵培养结束后,3500r/min下离心20min,去上清,菌泥分别加入实施例1-9制备的冻干保护剂内,充分混匀后,得原液1-9;
Ⅱ.用实施例 1-9 制备的冻干保护剂分别将原液 1-9 稀释至每 1ml 含菌5×109,得半成品1-9;
Ⅲ.分装,每西林瓶含 2ml(100羽鸡用剂量),冻干,得成品。
分别对上述疫苗进行冻干前的活菌数测定,冻干后的活菌数、2-8℃稳定性和 25℃稳定性测定。 结果如下:
(1)疫苗冻干前及冻干后的活菌数测定
分别对上述疫苗冻干前及冻干后的部分样品进行活菌数测定,结果见下表 :
注 :活菌减少率 =(冻干前活菌数 - 冻干后活菌数)/冻干前活菌数
上述结果表明,利用本发明的冻干保护剂(实施例 1-7所提供) 制备的禽大肠杆菌病活疫苗在冻干过程中活菌减少率要明显低于使用对照实施例(实施例 8-9所提供) 制备的疫苗。
(2)疫苗冻干后在 2-8℃的稳定性测试
将上述冻干后的疫苗的部分样品放置在 2-8℃,并分别于放置后的 1、2、3 个月进行活菌数测定,结果见下表:
上述结果表明,利用本发明的冻干保护剂(实施例 1-7所提供) 制备的禽大肠杆菌病活疫苗冻干后在 2-8℃条件下分别放置 1、2、3 个月,活菌数均明显高于使用对照实施例(实施例 8-9所提供) 制备的疫苗。
(3)疫苗冻干后在25℃的稳定性测试
将上述冻干后的疫苗的部分样品放置在 25℃,并分别于7天和 28天后进行活菌数测定,结果见下表:
上述结果表明,利用本发明的冻干保护剂(实施例 1-7所提供) 制备的禽大肠杆菌病活疫苗冻干后在25℃条件下分别放置 7天和 28 天,活菌数要明显高于使用对照实施例(实施例 8-9所提供)制备的疫苗。
综上所述,使用本发明的禽大肠杆菌病活疫苗冻干保护剂与使用目前已报道的细菌活疫苗冻干保护剂相比,在禽大肠杆菌病活疫苗的冻干过程中和在 2-8℃条件下或者在25℃条件下,由前者制备的疫苗的活菌数都明显高于后者。这一结果表明,本发明的冻干保护剂与现有常用冻干保护剂技术相比,疫苗活菌数存活率高且具有良好的稳定性。
实施例1-7,仅是本发明的较佳实施例而已,并非是对本发明所作的其它形式的限定,任何熟悉本专业的技术人员可能利用上述技术内容作为启示加以变更或改型为等同变化的等效实施例。但凡是未脱离本发明权利要求的技术实质,对以上实施例所作出的简单修改、等同变化与改型,仍属于本发明权利要求保护的范围。

Claims (4)

1.一种禽大肠杆菌病活疫苗冻干保护剂,其特征在于按照重量份数计,它包括如下组分:
海藻糖60-80份、脱脂乳粉80-100份、酵母粉2-3份、蛋白胨3-6份、葡甘露聚糖0.5-5份、灭菌水1000份。
2.根据权利要求1所述的禽大肠杆菌病活疫苗冻干保护剂,其特征在于:所述禽大肠杆菌病活疫苗冻干保护剂还包括4-10重量份的氯化钠。
3.根据权利要求2所述的禽大肠杆菌病活疫苗冻干保护剂,其特征在于:按照重量份数计,它包括如下组分:
海藻糖80份、脱脂乳粉80份、酵母粉2.5份、蛋白胨5份,氯化钠5份、葡甘露聚糖1份、灭菌水1000份。
4.根据权利要求1-3任意一项所述的禽大肠杆菌病活疫苗冻干保护剂,其特征在于:所述禽大肠杆菌病活疫苗冻干保护剂的pH为6.8-7.2。
