本发明属于生物医药领域,具体涉及一种用于核酸分子体内递送的、具有肾脏靶向性的嵌段型多肽载体;进一步的,本发明涉及所述嵌段型多肽载体与siRNA、寡聚核苷酸(oligonucleotide)、DNA形成核酸/载体复合物的设计和制备方法;更进一步的,本发明涉及核酸/载体复合物的尺寸大小、稳定性、及可重复性。
背景技术
近二十年来,随着基因组研究的深入,科学家们发现许多疾病(遗传性疾病、癌症等)都与基因异常相关。作为一种革命性的生物医学方案,基因治疗正是在这一基础上发展起来的。利用不同性质的递送载体,人们将正常的人类基因、或具有疾病治疗作用的核酸片段导入目标细胞,纠正基因的表达缺陷,实现疾病治疗的目的。1998年,科学家发现可以以短链小干扰RNA为介质,抑制致病基因的表达,这一发现为疾病的基因治疗提供了一个全新的技术方案。
在基因治疗中,目前的技术瓶颈主要在于如何高效、低毒性地将核酸片段(长链DNA、寡聚核苷酸、siRNA)递送到目标细胞中。常用的递送载体有病毒和非病毒两类。病毒类载体具有较高的基因递送效率,但安全性差,对外源基因的大小有限制,且容易引发较为严重的免疫反应。与之相比,非病毒载体具有较高的安全性,但递送效率较低。
带正电荷的阳离子化合物是最常用的非病毒载体,有聚阳离子、正电磷脂、壳聚糖、白蛋白、树枝状大分子以及阳离子多肽等类型。利用带正电荷的阳离子基团与DNA/RNA上带负电荷的磷酸基团之间的电性作用,将核酸分子压缩,形成核酸/阳离子载体复合物颗粒。这种包载形式使得核酸分子能够顺利通过多个细胞屏障,完成细胞递送过程。尽管关于核酸/阳离子载体复合物的结构、以及复合物的性质与转染效率之间的关系还存在争议,但大量的研究结果表明,这类复合物具有较好的生物相容性和较高的核酸递送效率。
形成稳定的核酸/载体复合物是开展基因治疗的一个先决条件。简单混合的方式所形成的复合物很不稳定,例如寡聚核苷酸与聚赖氨酸形成的复合物(Cui Zheng,LinNiu,Jingjing Yan,Jie Liu,Ying Luo,Dehai Liang,Structure and stability of thecomplex formed by oligonucleotides Phys.Chem.Chem.Phys.2012,14,7352-7359)。复合物的稳定性也会进一步影响核酸分子的递送效率(Jihan Zhou,Jie Liu,Tao Shi,Yuqiong Xia,Ying Luo*,and Dehai Liang*Phase Separation of siRNA/polycationComplex and Its Effect on Transfection Efficiency,soft matter,2013,9,2262-2268)。因此,需要设计能与核酸分子形成结构稳定、尺寸大小合适复合物的递送载体。
作为一种重要的生物大分子,多肽在基因递送应用方面具有许多独特的优势:成熟的固相合成方案能提供结构明确、分布单一的高纯度多肽分子;可以由20种性质各异的氨基酸组成性质丰富的一级序列;可获得beta折叠、alpha螺旋等具有重要生物学活性的二级结构;易于改性或与其他具有组织靶向功能的序列或分子偶联。报道的多肽递送载体大体可分为两类:一类是模拟、改造穿膜肽(cell penetrating peptide)序列的多肽,透膜效果好,但通常具有较大的毒性;另一类是将靶向性多肽、穿膜肽与其他载体混合使用,这类载体的结构和作用方式较为复杂,成药性较差。
发明内容
为了克服现有技术的缺陷,本发明以聚电解质复合理论为基础,设计了一种具有嵌段型结构的多肽分子。该多肽可通过静电作用,与DNA、RNA分子形成稳定的纳米颗粒结构,实现肾脏靶向性核酸递送。
本发明提供的用于siRNA或基因递送载体的多肽,是将数个具有不同功能的多肽序列连接,从而获得的一条嵌段型多肽,包括正电序列嵌段、二级结构嵌段和肾脏靶向性嵌段。
所述正电序列嵌段由带正电的氨基酸残基构成,序列具体为KKKKKKKKKKKKKKKKKKKK。该嵌段的作用是利用其所带的正电荷,与带负电荷的核酸分子发生相互作用,形成核酸/载体复合物的核心。
所述二级结构嵌段能形成β-折叠结构,起到稳定复合物结构的作用,序列具体为VEVKVEVKVEVKVEVKVEVK。
上述正电序列嵌段和二级结构嵌段可以直接连接在一起,也可以由一柔性连接链段将二者连接起来。所述柔性连接链段的作用是将正电序列嵌段与二级结构嵌段以柔性方式连接,组成柔性连接链段的氨基酸可选择甘氨酸(G)、天冬酰胺(N)、谷氨酰胺(Q)和丙氨酸(A)中的一种或多种,优选为甘氨酸(G)。组成柔性连接链段的氨基酸残基数目通常为1-5个,优选为3个。值得注意的是,缺少柔性连接链段可能会造成嵌段多肽的效果略有下降,但是,这一柔性连接链段并不是必需的。
所述肾脏靶向嵌段具有靶向肾脏的作用,序列具体为YGAGAGSGAASGAGAGSGA。
本发明提供的嵌段型多肽能够以不同的N/P比例(阳离子聚合物所带正电与核酸分子所带负电比例),通过简单混合的方式与核酸分子(DNA或RNA)形成大小合适、结构稳定的纳米级核酸/载体复合物,实现核酸分子的肾靶向递送和运送。

本发明的有益效果
本发明提供的嵌段型多肽,能够与核酸分子形成大小合适、稳定性强的纳米颗粒,既可用于递送短链核酸药物(反义寡聚核苷酸,小干扰核酸,微小核糖核酸,其他功能性寡聚核糖核苷酸和脱氧核糖核苷酸),也可用于长链DNA的肾靶向递送。而且,该嵌段型多肽无论在体内还是体外,均具有较小的毒性。

具体实施方式
下面结合具体实施例及附图,进一步阐述本发明。应当理解,这些实施例仅用于说明本发明而不能用于限制本发明的范围。下列实施例中未注明具体条件的实验方法,通常按照常规条件,例如Sambrook等人,分子克隆:实验室手册(New York:Cold Spring HarborLaboratory Press,1989)中所述的条件,或按照制造厂商所建议的条件。
实施例一:实验仪器及样品的配制
1、实验仪器
为了研究正在形成的基因载体的大小和性质,利用动静态多角度联用光散射仪(Brookhaven Instruments Corporation,Holtsville,NY,U.S.A)测定复合物尺寸并跟踪其在溶液中的稳定性。用圆二色谱跟踪多肽二级结构变化。
2、实验材料
以21碱基对的双链小干扰RNA(siRNA)和设计的多肽载体作为实验对象。siRNA购自于广州市锐博生物科技有限公司,多肽由吉尔生化(上海)有限公司合成。siRNA和肾脏靶向性多肽K-peptide的序列如下,其中siRNA-3是以小鼠Lamin A/C基因为靶点的特异性siRNA。

K-peptide(SEQ ID No:7):Ac-KKKKKKKKKKKKKKKKKKKKGGGVEVKVEVKVEVKVEVKVEVKNNNYGAGAGSGAASGAGAGSGA-amide。
3、样品的配制
将siRNA-1、siRNA-2和siRNA-3的正义链和反义链,分别溶解于去除Ca、Mg离子的标准磷酸缓冲液(DPBS)中。分别取每条siRNA等量的正义链和反义链溶液,按照标准方案进行退火,得到双链siRNA。调节siRNA的浓度至1×10-4g/mL。将K-peptide多肽溶解于去除Ca、Mg离子的标准磷酸缓冲液(DPBS)中,至终浓度1×10-4g/mL。利用0.20μm水相过滤器(Sartorius stedim Biotech,Goettingen,Germany),过滤去除siRNA、多肽溶液中的细菌和灰尘。
利用均匀混合方法,将一定量的siRNA溶液加入到一定量的多肽溶液中,以1200rps/min的转速涡旋混匀30秒,使其充分分散,制备siRNA/多肽载体复合物。调节核酸溶液和多肽溶液的用量比例,制备具有不同N/P的siRNA/多肽复合物。将获得的siRNA/多肽复合物在室温下静置30分钟,待其充分稳定后,开展下一步检测或使用。
实施例二:核酸/载体复合物的表征
按照上述方法,分别利用siRNA-1、siRNA-2或siRNA-3与K-peptide多肽混合,制备N/P比为5、10、15、20、40、80的siRNA/多肽复合物。
利用光散射和AFM,对这些复合物的结构进行表征。其流体力学半径的测量值如下:
研究发现,siRNA/多肽复合物颗粒的大小、稳定性与N/P比相关,与所使用的siRNA序列无关。
图1所示的结果为siRNA-1在N/P=20的条件下,与K-peptide多肽复合物的检测结果。这一颗粒在溶液中稳定性很好,颗粒散射光强可以长达上百小时而不变化,相对应的颗粒尺寸也没有变化。(图2)。后续研究均在N/P=20的条件下,制备siRNA/多肽复合物。
随后,我们利用圆二色谱技术,检测了复合物的结构特征。如图3(siRNA-1,N/P=20)所示,多肽载体在没有与寡聚核苷酸形成复合物之前是无规卷曲,没有任何二级结构,而与寡聚核苷酸形成复合物之后,多肽呈现了明显的beta折叠信号,这说明了复合物形成过程中多肽构象发生了变化。
另外,利用液体原子力显微镜原位观察形成复合物的形貌结构。将形成的复合物用移液枪吸取10μL置于云母表面,加入缓冲溶液,直接在溶液中测得复合物结构,可以看到复合物大小与光散射测得的结果一致,直径大小在80nm左右(图4,siRNA-1,N/P=20)。仪器型号为Nanoscope V。
实施例三:肾靶向siRNA递送
购买cy5荧光标记的siRNA-2和siRNA-3(siRNA-2-cy5、siRNA-3-cy5),使其与K-peptide多肽载体结合(N/P=20),形成荧光标记的复合物颗粒。将荧光标记的复合物颗粒、以及未与多肽结合的裸的荧光标记的siRNA通过尾静脉,注射到小鼠体内。使用的小鼠为雄性BALB/c小鼠,鼠龄5-7周,体重18-22g,siRNA的注射剂量为1.0mg/kg。24小时后处死小鼠,进行整体动物的荧光成像观察。与注射了裸siRNA-1-cy5的小鼠相比,整体动物成像显示K-peptide能提高siRNA在体内的分布,尤其是在小鼠肾脏的分布量(图5)。所使用的小动物成像仪为Kodak FX Pro,由Carestream Health公司生产。
随后,我们分离小鼠的主要脏器组织,包括脑、颌下腺、心、肝脏、肾、脾和睾丸。用小动物荧光成像仪测定了各脏器中荧光标记siRNA-3的强度,并进行了定量分析。发现与其他脏器相比,肾脏中荧光信号的强度是最高的(图6)。表明K-peptide多肽能显著提高siRNA在肾脏的富集,是一种具有显著肾靶向效果的siRNA多肽递送载体。
实施例四:K-peptide介导的内源基因的表达沉默
为了评价K-peptide多肽载体是否能介导内源基因的表达沉默,我们选择以LaminA/C基因为靶点的siRNA-3开展研究。使siRNA-3与K-peptide多肽结合,形成siRNA-3/K-peptide复合物颗粒。用PBS、裸siRNA-3、siRNA-3/K-peptide复合物分别处理小鼠。使用的小鼠为雄性BALB/c小鼠,鼠龄5-7周,体重18-22g,siRNA的注射剂量为1.0mg/kg。
48个小时后处死小鼠,获得实验组和对照组小鼠的肾脏组织,提取总RNA。利用Lamin A/C特异的PCR引物,分析经过不同处理的小鼠肾脏中Lamin A/C的表达水平。对比PBS、裸siRNA-3处理的小鼠,分析经由K-peptide递送的siRNA-3对内源基因的表达抑制作用。
实验步骤如下:
1)总RNA提取:利用Trizol试剂,提取肾脏组织中总RNA,并进行定量。
2)RNA逆转录:使用天根生化科技公司的逆转录试剂盒(TIANScript M-MLV,cat:ER104-03)进行RNA样本的逆转录反应,具体按照试剂盒说明书的方法进行。
3)逆转录PCR:使用北京全式金生物科技有限公司的2×Easy Taq@PCR SuperMix试剂(cat:AS111-01),检测样本中目标基因的表达情况。1)取1μL逆转录产物,加入12.5μL2×Easy Taq@PCR SuperMix,0.5μL(10μM)上游引物,0.5μL(10μM)下游引物,最后加入10.5μL去离子水,反应总体积为25μL。2)混匀后短暂离心,置于TECHNE PCR仪中进行PCR扩增反应,反应参数为94℃预变性1分钟,94℃变性30秒,59℃退火30秒,72℃延伸30秒,循环次数为30个循环;72℃,5分钟;随后置于4℃保存。
4)跑胶检测:用2%琼脂糖凝胶检测PCR产物。
5)分析RT-PCR数据,鉴定各组织中Lamin A/C的表达水平。
与PBS或裸siRNA-3处理的小鼠相比,siRNA-3/K-peptide复合物的处理使小鼠肾脏中Lamin A/C的表达水平下降了35%(图7)。这个结果表明K-peptide能将siRNA有效地递送至小鼠肾脏,并抑制目标基因的表达。
使用的PCR扩增引物序列如下:
LaminA/C上游引物(SEQ ID No:11):5′-GATGGAGGGCAATGTCAAGT;
LaminA/C下游引物(SEQ ID No:12):5′-AGAGGTGCAGATGGGAAATG;
β-actin上游引物(SEQ ID No:13):5′-GAAGAGCTATGAGCTGCCTGA;
β-actin下游引物(SEQ ID No:14):5′-CTCATCGTACTCCTGCTTGCT。
实施例五:K-peptide多肽载体的毒性研究
为了评估K-peptide多肽的细胞毒性,我们用具有不同N/P比的siRNA-3/K-peptide复合物处理培养的HEK293细胞中,对照组包括:不经过任何处理的HEK293细胞(CTL),经过siRNA-3/Lipofectamine 2000复合物处理的HEK293细胞(Lipo),和经过不与siRNA复合的K-peptide处理的细胞(L-peptide)。然后用CCK-8试剂盒,检测各种处理方式对细胞存活率的影响。
在实验的前一天,以6×103细胞/孔的接种量将HEK293细胞接种到96孔板中。生长过夜后,利用具有不同N/P比的siRNA-3/K-peptide复合物或对照条件处理细胞,siRNA-3的用量为0.08μg/孔。转染后4小时,每孔加入100μL DMEM,继续培养20小时。
培养结束后,利用Cell Counting Kit-8(CCK-8)试剂(Dojindo MolecorTechnologies,Inc。)检测细胞的活性,具体操作按试剂盒的说明书进行。实验结果如图8所示,表明当N/P比控制在5-40之间时,对培养细胞无显著的毒性作用。
在实施例三和实施例四中,以注射PBS、裸siRNA小鼠为对照,对比研究注射K-peptide复合体的体内毒性。从小鼠体重、摄食量、活动情况、毛色和毛发状态等方面考察,均与对照小鼠一致,未发现任何毒副作用。这表明K-peptide多肽载体具有较好的体内安全性。
Claims (10)
1.一种用作肾脏靶向性siRNA递送的嵌段多肽,包括正电序列嵌段、二级结构嵌段和肾脏靶向嵌段,其中所述正电序列嵌段由带正电的氨基酸残基构成,所述二级结构嵌段为能形成β-折叠的嵌段,所述正电序列嵌段的氨基酸残基数目为10-70,所述二级结构嵌段的氨基酸残基数目为10-40。
2.如权利要求1所述肾脏靶向性siRNA递送多肽,其特征在于,组成所述正电序列嵌段的氨基酸为赖氨酸和/或精氨酸。
3.如权利要求1所述肾脏靶向性siRNA递送多肽,其特征在于,所述二级结构嵌段选自下列序列之一:
VEVKVEVKVEVKVEVKVEVK;
IEIKIEIKIEIKIEIKIEIK;
ELKLELKLELKLELKLELK;
GAGAGSGAGAGSGAGAGS;
KSKLALKLALKAWSAALKLA;或

4.如权利要求1所述肾脏靶向性siRNA递送多肽,其特征在于,所述嵌段多肽还包括一柔性连接链段,该柔性连接链段将所述正电序列嵌段和二级结构嵌段连接起来。
5.如权利要求4所述肾脏靶向性siRNA递送多肽,其特征在于,所述柔性连接链段的氨基酸残基数目为1-5;组成所述柔性连接链段的氨基酸选自甘氨酸、天冬酰胺、谷氨酰胺或丙氨酸中的一种或多种。
6.如权利要求1所述肾脏靶向性siRNA递送多肽,其特征在于,所述肾脏靶向嵌段为YGAGAGSGAASGAGAGSGA。
7.如权利要求1所述肾脏靶向性siRNA递送多肽,其特征在于,所述多肽序列为:
Ac-KKKKKKKKKKKKKKKKKKKKGGGVEVKVEVKVEVKVEVKVEVKNNNYGAGAGSGAASGAGAGSGA-amide。
8.权利要求1-7任一项所述肾脏靶向性siRNA递送多肽在向肾脏递送siRNA中的应用。
9.权利要求1-7任一项所述肾脏靶向性siRNA递送多肽在制备向肾脏递送siRNA药物中的应用。
10.一种组合物,其包含权利要求1所述的肾脏靶向性siRNA递送多肽和有效量的siRNA。
