本发明所采取的技术方案是:
一种兼具缓解疲劳与抗氧化功能的保健营养液,所述保健营养液的原料配方包括以下成分:文蛤提取 物、人参提取物、枸杞提取物、柠檬酸、蔗糖。
作为本发明进一步的方案:每1000mL含有文蛤提取物10-200g,人参提取物5-50g,枸杞提取物 10-100g,柠檬酸3-10g,蔗糖5-10g,以及余量的水。
作为本发明进一步的方案:所述文蛤提取物首先由文蛤软体通过加入水及蛋白酶得到文蛤酶解液,文 蛤酶解液经灭酶处理后离心取上清,喷雾干燥而得。文蛤提取物中,平均分子量3kDa以下的组分不低于 40%。
作为本发明进一步的方案:所述人参提取物、枸杞提取物,由粉碎后的原料加入水,60-100℃浸提1-4 h,离心收集上清。重复以上操作2-4次,并合并上清液,喷雾干燥分别得到人参提取物和枸杞提取物。

实施例1
所述保健营养液按照以下步骤制备:
1)所述文蛤提取物制备如下:将1kg冷冻文蛤肉置于室温下解冻1h,加入适宜比例去离子水,倒 入料理机匀浆。文蛤匀浆倒入清洗灭菌后的酶解罐中,恒温45℃,加入6g酸性蛋白酶,在搅拌 转速50rpm、自然pH条件下进行酶解。酶解2h后结束反应放出酶解液,95℃水浴加热10min 灭酶,冷却后在5000rpm、4℃条件下离心15min留取上清液和残渣。残渣重复上述操作2次, 合并上清液,对上清液进行喷雾干燥,得到文蛤提取物粉末。
2)文蛤提取物经凝胶色谱测定平均分子量,得到平均分子量为1987Da;经高效凝胶色谱分析分子 量分布,其中分子量低于3KDa的组分比例76.3%。
3)所述人参提取物、枸杞提取物制备如下:将1kg原料用搅拌机打碎成粉末,按照料液比1:10加 入去离子水,90℃下搅拌水浴浸提2h,之后在4℃条件下5000rpm离心10min,取上清,沉淀 部分再次加入相同体积的去离子水在相同条件下再次提取1.5-2h,相同条件下离心取上清,合并 上清液后喷雾干燥,分别得到人参提取物、枸杞提取物粉末。
4)按照不同配比,取所述文蛤提取物、人参提取物、枸杞提取物、柠檬酸、蔗糖,加水溶解并定容 至1L,混合均匀后分装包装并灭菌。组合1:文蛤提取物100g、人参提取物20g、枸杞提取物50g、 柠檬酸3g、蔗糖5g;组合2:文蛤提取物10g、人参提取物50g、枸杞提取物100g、柠檬酸10g、 蔗糖5g;组合3:文蛤提取物200g、人参提取物50g、枸杞提取物10g、柠檬酸7g、蔗糖10g。
实施例2
缓解疲劳有效性实验
参照卫生部发布的《保健食品检验与评价技术规范》,针对抗疲劳作用进行动物实验,以6周龄雄性 ICR小鼠100只为测试对象,测试其用药前后负重游泳时间,血清尿素氮、血乳酸、肝糖原4项指标。
(1)将实施例1制备的保健营养液组合1折算成生药材,分为三个剂量组,按0.25g/kg、0.5g/kg、1 g/kg灌胃给药,给药体积20mL/kg。空白对照组灌以相同体积的生理盐水。指标测定如以下步骤:
1)负重游泳时间
每组各取6只小鼠,末次给药30min后,将尾根部负荷5%体重铅皮的小鼠置于游泳箱中游泳, 水深不少于30cm,水温25℃±1℃,记录小鼠自游泳开始至死亡的时间,即小鼠负重游泳时间。 若受试样品组负责游泳时间明显长于对照组,且差异有显著性,可判定该实验结果阳性。
2)血清尿素氮
每组各取6只小鼠,末次给药30min后,在温度为30℃的水中不负重游泳90min,休息60min 后采血(不加抗凝剂),置4℃冰箱约30h,血凝固后离心,取血清备用。采用试剂盒测定血清尿 素。若受试样品组血清尿素明显低于对照组,且差异有显著性,可判定该实验结果阳性。
3)血乳酸测定
每组各取6只小鼠,末次给药30min后采血20μL,然后不负重在温度为30℃的水中游泳10min 后停止。游泳后立即采血20μL,休息20min后再采血20μL。采用试剂盒测定血乳酸含量。以三 个时间点血乳酸曲线下面积来判断,任一试验组的面积小于对照组,且差异有显著性,可判定该 实验结果阳性。
4)肝糖原测定
每组各取6只小鼠,末次给药后30min处死动物,解刨去肝脏,生理盐水漂洗后用滤纸吸干,存 于-80℃冰箱备用。采用试剂盒测定肝糖原含量。若受试样品组肝糖原含量明显高于对照组,且差 异有显著性,可判定该实验结果阳性。
实验结果如表1:

由实验数据来看,所开发的保健营养液中高剂量组能显著延长小鼠负重游泳时间,降低运动后尿素氮 水平,增加肝糖原水平,同时在游泳0min、游泳10min、游泳后休息20min这三个时间点的乳酸曲线下 面积与空白对照组相比显著降低;解剖观察肝、肾、脾等器官组织,病理形态均正常,表明本发明具有良 好的缓解疲劳效果,且安全性较好。
(2)采用如上的实验方法,以6周龄雄性ICR小鼠为测试对象,以实施例1中保健营养液组合1、 组合2、组合3,按照剂量1g/kg分别灌胃给药,给药体积20mL/kg。空白对照组灌以相同体积的生理盐 水。测定负重游泳时间,血清尿素氮、血乳酸、肝糖原4项指标。
实验结果如表2:

由表2数据可得,喂食本发明的保健营养液可以显著延长小鼠负重游泳时间,降低运动后血液尿素氮 水平,增加肝糖原水平,且在游泳0min、游泳10min、游泳后休息20min这三个时间点的乳酸曲线下面 积与空白对照组相比显著降低,说明具有良好的缓解疲劳效果。
实施例3
抗氧化有效性实验
参照卫生部发布的《保健食品检验与评价技术规范》,针对抗氧化作用进行动物实验。选用10月龄健 康状况良好的雄性小鼠32只进行实验。将小鼠随机分为对照组、实验组1、实验组2、实验组3,每组8 只。实验组1、实验组2和实验组3分别喂食实施例1中制得的保健营养食品组合1,组合2和组合3,每 天喂食两次,代替鼠粮,对照组正常饮食。实验结束后采尾血,制备血清。血清中过氧化脂质降解产物丙 二醛(MDA)含量测定采用硫代巴比妥酸(TBA)荧光分光光度法,血清中超氧化物歧化酶(SOD)活力 测定采用羟胺比色法,按SOD测定试剂盒说明书操作。
实验结果如表3:

丙二醛(MDA)是在氧存在条件下,由自由基诱发剂诱发细胞内外超氧阴离子自由基、羟自由基等发生 多种生化成分的过氧化链式反应而逐步形成。自由基及其诱导的脂质过氧化物的长期毒害作用是引起生物 衰老、免疫力下降的重要原因之一。SOD可消除氧自由基对细胞的毒害作用,能减轻和阻断脂质过氧化作 用,保护机体细胞免受自由基的损伤。
表3的数据表明,喂食本发明制得的抗氧化营养组合物,能显著降低雄性小鼠血清中丙二醛(MDA) 的含量,显著提高血清SOD活性,减少机体脂质过氧化作用,从而发挥抗氧化作用。
Claims (6)
1.一种兼具缓解疲劳与抗氧化功能的保健营养液,其特征在于,所述保健营养液的原料配方包括以下成分:文蛤提取物、人参提取物、枸杞提取物、柠檬酸、蔗糖。
2.权利要求1所述的保健营养液的制备方法,其特征在于,每1000mL营养液中含有文蛤提取物10-200g,人参提取物5-50g,枸杞提取物10-100g,柠檬酸3-10g,蔗糖5-10g。
3.如权利要求2所述的方法,其特征在于所述文蛤提取物为采用酶法辅助提取所得:文蛤软体通过加入水及蛋白酶得到文蛤酶解液,灭酶处理后离心取上清,喷雾干燥得到文蛤提取物;其中,平均分子量3kDa以下的组分不低于40%。
4.如权利要求3所述的方法,其特征为,所述的蛋白酶包括但不限于胰蛋白酶、酸性蛋白酶、碱性蛋白酶、木瓜蛋白酶、菠萝蛋白酶、复合蛋白酶、风味蛋白酶、中性蛋白酶中的一种或几种,用量按反应液总体积为100-1000U/mL。
5.如权利要求2所述的方法,其特征在于所述人参提取物、枸杞提取物的制备方法为:粉碎后的原料加入水,60-100℃浸提1-4h,离心收集上清;重复以上操作2-4次,并合并上清液,喷雾干燥分别得到人参提取物和枸杞提取物。
6.如权利要求2所述的方法,其特征在于,按如下步骤制作而成:按照所述配比称取文蛤提取物、人参提取物、枸杞提取物、柠檬酸、蔗糖,溶解混合均匀,并进行包装灭菌。
