Abstract

本发明公开一种葫芦素B在制备抑制缺氧性肺动脉高压药物中的应用。葫芦素B来源丰富、易提取、成本低,对人体无任何副作用,可以有效抑制缺氧性肺动脉高压,预防缺氧性肺动脉高压导致的病理改变,避免现有治疗肺动脉高压的临床药物对人体造成的各种伤害。

葫芦素B在制备抑制缺氧性肺动脉高压药物中的应用
技术领域
本发明涉及葫芦素B的新用途,尤其是葫芦素B在制备抑制缺氧性肺动脉高压药物中的应用。
背景技术
血管收缩、血管壁重建及原位血栓形成这三种因素的综合作用使肺血管阻力进行性升高,即形成肺动脉高压(PAH)。肺动脉高压(PAH)可使肺微小动脉血管痉挛、内膜增生和重构。缺氧(hypoxia)包括环境缺氧及病理性缺氧,尤其是当组织的氧供应不足或利用氧障碍时,导致组织的代谢、功能和形态结构发生异常变化。缺氧性肺动脉高压(HPH)是肺动脉高压中的一个亚型,其病理学改变的主要特征是小动脉壁的肌层增厚与无肌层小动脉肌化,常见于罹患慢性阻塞性肺疾病与久居高原的人群。肺动脉高压是一种进展性疾病,预后较差,会引起右心功能衰竭和死亡。目前用于治疗肺动脉高压的临床药物以下几种药物,副作用较大。
1. 前列腺素类
内皮源性前列腺素I2或前列环素由内皮细胞内花生四烯酸合成前列环素具有很强的扩血管作用并可通过增加细胞内环单磷酸腺苷cAMP的水平抑制血小板聚集,如依前列醇、曲罗尼尔、伊洛前列素等。这类药物半衰期短常需要持续或多次给药,不良反应包括头痛潮红下颌痛和胃肠道不适。
2. 内皮素受体拮抗剂
内皮素受体拮抗剂(波生坦内皮素1等),可抑制内皮素的收缩血管和促进血管平滑肌增殖作用,但是对肝功能有一定的影响,患者服用期间必须每个月监测肝功能。
3. PDE-5抑制剂
PDE-5抑制剂(西地那非、他达拉非等)多用于NYHA心功能II—III级肺动脉高压患者,常见的不良反应有头痛、皮肤潮红和呼吸困难。
葫芦素B (CuB ) 是从葫芦科等植物中分离得到的一类四环三萜类化合物,在葫芦素家族中含量最为丰富,对于护肝、抗菌消炎及抗肿瘤等具有广泛的药理活性,已研发出葫芦素片(含葫芦素B 和葫芦素E)用于肝炎和原发性肝癌的辅助治疗,对人体无任何副作用。但是,迄今为止还没有关于葫芦素B在制备抑制缺氧性肺动脉高压药物中应用的相关报道。
发明内容
本发明是为了解决现有技术所存在的上述技术问题,提供一种葫芦素B在制备抑制缺氧性肺动脉高压药物中的应用。
本发明是发现了葫芦素B具有抑制缺氧性肺动脉高压的作用,从而发明了葫芦素B在制备抑制缺氧性肺动脉高压药物中的应用。
本发明是以来源丰富、易提取、成本低的葫芦素B作为抑制缺氧性肺动脉
高压的药物,对人体无任何副作用,可预防缺氧性肺动脉高压导致的病理改变,
避免现有治疗肺动脉高压的临床药物对人体造成的各种伤害。

具体实施方式
实验:
动物分组:成年昆明小鼠,随机分为四组:
(1)正常+ PBS组:小鼠在正常空气环境中生活,腹腔注射给予PBS,剂量为0.9 mg/kg,每日一次,连续5周;
(2)正常+葫芦素B组:小鼠在正常空气环境中生活,腹腔注射给予用PBS配制的葫芦素B药液,剂量为0.9 mg/kg,每日一次,连续5周;
(3)模型+ PBS组:将小鼠置于低压氧仓(含氧量10%,含氮量90%)内,
连续缺氧4周,在缺氧前一周开始腹腔注射给予PBS,剂量为0.9 mg/kg,每日一次,连续5周;
(4)模型+葫芦素B组:将小鼠置于低压氧仓(含氧量10%,含氮量90%)内,
连续缺氧4周,在缺氧前一周开始腹腔注射给予PBS,剂量为0.9 mg/kg,每日一次,连续5周。
5周结束后,分别测定各组小鼠右心室收缩压及测定右心室肥大指数。
实验结果:
各组小鼠右心室收缩压结果如图1所示。图1中纵坐标RVSP表示右心室收缩压(Rightventricular systolic pressure)。
各组小鼠右心室肥大指数如图2所示。图2中纵坐标RV/(LV+S)表示右心室/(左心室+室间隔)(重量之比)。
结果表示:在缺氧小鼠模型上,葫芦素B可降低肺动脉血管压力,有效地抑制右心室肥大,预防缺氧导致的肺动脉高压。
