技术领域
本发明涉及中医制品及其加工技术,特别是涉及一种用于银屑病的口服制品与制备方法。
技术背景
银屑病又名“牛皮癣”,是一种常见的以皮肤红斑、表面覆盖多层银白色鳞屑,刮去皮屑有薄膜现象和点状出血为特点的慢性复发性皮肤病,边界清楚,全身均可发病,以头皮,四肢伸侧及背部较为常见。该病可见于任何年龄,以青壮年多见,男性多于女性。春冬季节容易复发或加重,而夏秋季多缓解。临床表现以红斑、鳞屑为主。全球有1%-3%的人口受累于银屑病,属中医“白庀”范畴。由于本病发病率高,易于复发,病程长,对患者的身体健康和精神影响甚大,因此银屑病是当前皮肤科领域的重点研究疾病之一。
银屑病的病因和发病机制尚未完全清楚。但一些致病因素或易患病因素已较清楚。银屑病由遗传背景、环境、外伤、感染、药物、合并疾病、饮酒、吸烟、精神创伤等因素诱发或加重。目前的研究认为银屑病是在多基因遗传背景下由T淋巴细胞介导的一种自身免疫性疾病。银屑病患者存在多种免疫细胞(包括角质形成细胞、树枝状细胞、血管内皮细胞、单一核细胞及多形核细胞等)、免疫分子、细胞内信号传导系统等的功能异常。应用分子生物学技术,通过特异性抗体阻断淋巴细胞效应阶段的多种因子所发挥的功能,可以在一定程度上有效的控制银屑病的发生和发展。中医认为本病多由内有血热,外受风邪,而致血热生风,风盛则燥,故皮肤潮红、脱屑;风燥日久,伤阴伤血,而致阴虚血燥,肌肤失养,故皮肤干燥,叠起白屑。
银屑病有多种治疗方法如外用药物治疗、内用药物治疗及物理疗法,中、重度银屑病患者单一疗法效果不明显时,应给予联合、轮换或序贯治疗。目前西医常采用维生素类、抗生素类、抗肿瘤类、重金属类、免疫调节剂或激动剂、作用于神经系统的药物等内服药或用激素、角质形成剂、细胞毒制剂等外用药,并常配合紫外线照射、光化学方法等综合治疗本病。但临床实践证明,上述药物及疗法常会产生毒副作用,并有许多禁忌症。
中医中药治疗寻常型银屑病历史悠久,方法甚多,经验丰富,疗效肯定,疗效与西药相当或优于西药,而毒副作用少见,且不易产生耐药性,可以改善患者机体的整体机能,从多途径来消除疾病和症状。中医中药治疗寻常型银屑病无论从疗效还是安全性方面都显示出了明显的特色和优势。

基于上述领域的需求,本发明提供一种用于银屑病的口服制品及其制备工艺,用于治疗银屑病,疗效好,复发少,使用方便,无毒副作用,及时减缓病情发展,提高患者免疫力,显著改善患者生存质量。

上述任一用于银屑病的口服制品的制备方法,包括如下步骤:
(1)制备原料药的水提取液;
(2)原料药的水提取液与黑木耳菌母种液、银耳菌母种液进行共发酵得到发酵产物;
其特征在于,所述原料药的水提取液的提取工艺如下:在提取容器中,加入富氢水和原料药;水与所述原料药重量比例为3-6:1,在25-35℃、800mpa环境下提取30-50分钟得到所述原料药的水提取液。
上述的制备方法中,优选水与所述原料药重量比例为4-5:1,在28-32℃、800mpa环境下提取30-40分钟得到所述原料药的水提取液,优选地,所述原料药的水提取液是采用富氢水为提取溶剂,所述富氢水为日田天领水。
优选,所述进行共发酵是指在发酵罐中,按30-50:30-50:5-10:5-10的体积比加入所述原料药的水提取液、水、黑木耳菌母种液、银耳菌母种液,发酵到有效固体物含量达80g/L以上时放罐。
进一步优选
按35-45:35-45:6-8:6-8的体积比加入所述原料药的水提取液、水、黑木耳菌母种液、银耳菌母种液。
上述任一制备方法,还包括:放罐后,进入醇沉提取罐,加入冷水浸提渗滤,再煮沸,滤汁去渣;其汁加4倍体积的75度酒精浸润,在≦30℃低温下浸制5小时,利用多糖不溶于乙醇的性质,使多糖从提取液中沉淀出来,升温至50摄氏度回收酒精后,对沉淀再进行≦55℃烘干或喷雾干燥,制得多糖粉。
优选地,所述用于银屑病的口服制品的制备方法还包括富氢水的灭活,将所述富氢水在25-35℃、600mpa环境下灭活5-30分钟。
优选地,将所述富氢水在28-32℃、600mpa环境下灭活7-15分钟。
优选地,所述共发酵时间为72-96小时。
发明人经过认真研究,根据中医对银屑病病因的研究,中药用药的配伍思想,在配伍出的中药原料药基础上,创造性地采用共同发酵的方法联合使用食用菌,通过发酵使得食用菌中的多糖、中药材中的植物多糖的比例大大提高,同时使中药材中其它有效成分分离出来,植物多糖和食用菌多糖协同温和地作用于患者免疫系统,使患者免疫系统逐渐恢复到正常水平,促使免疫系统恢复表皮增生分化异常的识别能力,从而抑制表皮增生的发展;植物/食用菌多糖还有助于损伤组织/细胞的修复。
本发明的制品在制备过程中所用的水为富氢水,英文名为hydrogenRich water,日文是水素水,富氢水是最好的抗氧化物,集高氢量、弱碱性、负电位、小份子水为一体,容易在细胞膜穿梭参与新陈代谢,从而促进细胞排毒,增加了细胞的水合作用。使用富氢水使本发明的口服制品具有很强的还原功能,可以中和身体血液和细胞里的活性氧(自由基),平衡身体酸碱度,有助于抑制银屑病病情的发展,提高患者身体免疫力,并可有效缓解或预防多种并发症。
本发明的另一个主要的贡献在于,还提供了专门针对本发明的口服制品的制备方法,采用富氢水作为溶剂,在常温、超高压(30℃、800mpa)下提取中药成分,能够最大限度地将中药材植物细胞壁内容物提取出来,并保持原有成分,其分子结构只是从大变小,从而保留药效与原料药基本一致。
本发明的口服制品中采用的中药材中多种成分具有清热解毒、祛风除湿、凉血活血、止痒止痛的作用,采用的原料药药理如下:
葛根:为豆科植物野葛Puerarialobata(Willd.)Ohwi或甘葛藤PuerariathomsoniiBenth.的干燥根;学名:Radix Puerariae;别名:葛藤、粉葛、干葛、葛麻藤;功效:解肌退热,生津,透疹,升阳止泻。用于外感发热头痛、项背强痛,口渴,消渴,麻疹不透,热痢,泄泻;高血压颈项强痛。
紫草根:为紫草科植物新疆紫草Arnebiaeuchroma(Royle)Johnst.、紫草LithospermumerythrorhizonSieb.etZucc.或内蒙紫草ArnebiaguttataBunge的干燥根。功效:凉血,活血,解毒透疹。用于血热毒盛,斑疹紫黑,麻疹不透,疮疡,湿疹,水火烫伤。
茜草根:为茜草科植物茜草的根及根茎。别名:血见愁,过山龙,地苏木,活血丹,红龙须根,沙茜秧根,五爪龙,满江红,九龙根,红棵子根,小孩拳,娃娃拳,拉拉秧子根,小活血龙,土丹参,四方红根子,红茜根;功效:行血止血,通经活络,止咳祛痰。治吐血,衄血,尿血,便血,血崩,经闭,风湿痹痛,跌打损伤,瘀滞肿痛,黄疸,慢性气管炎。
白茅根:为禾本科植物白茅ImperatacylindricaBeauv.var.major(Nees)C.E.Hubb.的干燥根茎。别名:丝茅草、茅草、白茅草、茅草根;功效:凉血,止血,清热,利尿。治热病烦渴,吐血,衄血,肺热喘急,胃热哕逆,淋病,小便不利,水肿,黄疸。
生槐花:为豆科植物槐SophorajaponicaL.的花及花蕾。别名:金药树、护房树、豆槐、槐米;功效:凉血止血,清肝明目。用于治疗肠风便血,痔疮下血,赤白痢,血淋,崩漏,吐血,衄血,疮疡肿毒等。
生地:为玄参科植物地黄的块根。学名:Rehmannia glutinosa Libosch;别名:生地黄,野地黄、山烟根等;功效:清热凉血、益阴生津。用于温热病热入营血,壮热神昏,口干舌绛。
鸡血藤:为豆科鸡眼草属植物鸡眼草Kummerowiastriata(Thunb.)Schindl.,以全草入药。学名:Millettia dielsiana;别名:人字草、三叶人字草、掐不齐、老鸦须、铺地锦、白斑鸠窝;功效:清热解毒,健脾利湿。治感冒发热,暑湿吐泻,疟疾,痢疾,传染性肝炎,热淋,白浊。
土茯苓:为百合科植物光叶菝葜的干燥根茎。别名:冷饭团、硬饭头、红土苓;功效:有解毒,除湿,通利关节之功效,主要用于梅毒及汞中毒所致的肢体拘挛,筋骨疼痛;湿热淋浊,带下,痈肿,瘰疬,疥癣。
茵陈:为菊科植物滨蒿ArtemisiascopariaWaldst.etKit.或茵陈蒿ArtemisiacapillarisThunb.的干燥地上部分。学名:ArtemisiacapillarisThunb;别名:绵茵陈、茵陈蒿、白蒿、绒蒿、猴子毛;功效:清湿热,退黄疸。用于黄疸尿少,湿疮瘙痒;传染性黄疸型肝炎。
元参:为玄参科多年生草本植物玄参的根。学名:Scrophularia ningpoensisHemsl.;别名:元参、浙玄参、黑参、乌元参;功效:温热病热和营血、身热、烦渴、舌绛、发斑、骨蒸劳嗽、虚烦不寤、津伤便秘、目涩昏花、咽喉肿痛、瘰疬痰核、痈疽疮毒。
莪术:为姜科植物蓬莪术CurcumaphaeocaulisVal.、
广西莪术CurcumaKwangsiensisS.G.LeeetC.F.Liang或温郁金CurcumawenyujinY.H.ChenetC.Ling的干燥根茎。学名:Curcuma zedoaria(Christm.)Rosc;别名:温莪术、蓬莪术、山姜黄、芋儿七、臭屎姜;功效:行气破血,消积止痛。用于燀瘕痞块,瘀血经闭,食积胀痛;早期宫颈癌。
三棱:为黑三棱科植物黑三棱SparganiumstoloniferumBuch.-Ham.的干燥块茎。别名:黑三棱;功效:破血行气,消积止痛。用于症瘕痞块,瘀血经闭,食积胀痛。
天冬:为百合科植物天门冬Asparaguscochinchinensis(Lour.)Merr.的干燥块根。别名:天门冬、明天冬、天冬草、倪铃、丝冬、赶条蛇、多仔婆;功效:养阴润燥,清肺生津。用于肺燥干咳,顿咳痰黏,咽干口渴,肠燥便秘。
麦冬:为百合科植物麦冬(沿阶草)Ophiopogonjaponicus(Thunb.)Ker-Gawl.的干燥块根。学名:Ophiopogon japonicus(Linn.f.)Ker-Gawl.;别名:麦门冬、沿阶草;功效:养阴生津,润肺清心。用于肺燥干咳。虚痨咳嗽,津伤口渴,心烦失眠,内热消渴,肠燥便秘;咽白喉。
白鲜皮:为芸香科植物白鲜DictamnusdasycarpusTurcz.的干燥根皮。学名:CORTEXDICTAMNI;别名:白藓皮、八股牛、山牡丹、羊鲜草;功效:清热燥湿,祛风解毒。用于湿热疮毒,黄水淋漓,湿疹,风疹,疥癣疮癞,风湿热痹,黄疸尿赤。
蜂房:为胡蜂科昆虫果马蜂Polistes olivaceous(DeGeer)、日本长脚胡蜂Polistesjaponicus Saussure或异腹胡蜂Parapolybia varia Fabricius的巢。功效:攻毒杀虫,祛风止痛。常用于疮疡肿毒,乳痈,瘰疬,皮肤顽癣,鹅掌风,牙痛,风湿痹痛。
金银花:为忍冬科植物忍冬LonicerajaponicaThunb.、红腺忍冬LonicerahypoglaucaMiq.、山银花(毛萼忍冬)LoniceraconfusaDC.或毛花柱忍冬LoniceradasystylaRehd.的干燥花蕾或带初开的花。学名:Lonicera japonica Thunb.;别名:银花、双花、二花、二宝花;功效:清热解毒,凉散风热。用于痈肿疔疮,喉痹,丹毒,热毒血痢,风热感冒,温病发热。
威灵仙:为毛茛科植物威灵仙ClematischinensisOsbeck、棉团铁线莲(山蓼)ClematishexapetalaPall.或东北铁线莲(黑薇)ClematismanshuricaRupr.的干燥根及根茎。学名:Clematis chinensis Osbeck.;别名:铁脚威灵仙、百条根、老虎须、铁扫帚;功效:祛风除湿,通络止痛。用于风湿痹痛,肢体麻木,筋脉拘挛,屈伸不利,骨哽咽喉。
蒲公英:为菊科植物蒲公英TaraxacummongolicumHand.-Mazz.、碱地蒲公英TaraxacumsinicumKitag.或同属数种植物的干燥全草。学名:TaraxacummongolicumHand.-Mazz.;别名:黄花地丁、婆婆丁;功效:清热解毒,消肿散结,利尿通淋。用于疔疮肿毒,乳痈,瘰疠,目赤,咽痛,肺痈,肠痈,湿热黄疸,热淋涩痛。
紫花地丁:为堇菜科植物紫花地丁的全草。学名:Viola yedoensis Makino;别名:野堇菜、光瓣堇菜等;功效:清热解毒,凉血消肿,清热利湿。主治疔疮,痈肿,瘰疬,黄疸,痢疾,腹泻,目赤,喉痹,毒蛇咬伤。
野菊花:为菊科植物野菊ChrysanthemumindicumL.的干燥头状花序。学名:Dendranthemaindicum;别名:野菊、野黄菊、苦薏;功效:清热解毒。用于疔疮痈肿,目赤肿痛,头痛眩晕。
本发明中,采用黑木耳菌母种液、银耳菌母种液与水提取液进行共发酵,使其利五脏、软坚、排毒、止血、活血和胃止泻的功效得以充分利用,临床病例统计显示,本发明的口服制品治疗银屑病功效明显。用于本发明的黑木耳菌母种液、银耳菌母种液,不限于本发明采用的菌种,一般商购获得的试管母种扩繁为液体母种后均可以用于本发明。
黑木耳菌:拉丁学名:Aurcularia auricula,是真菌类担子菌纲木耳科木耳属植物木耳Auriculariaauricula(L.)Underw.,以子实体入药,是一种营养价值丰富且具有多种生物学活性的药食两用真菌;具有凉血,止血之功效。可用于治肠风,血痢,血淋,崩漏,痔疮。其深层发酵产物——黑木耳多糖是发挥作用的主要成分。其所含植物碱具有促进消化道、泌尿道各种腺体分泌的特性,植物碱能协同这些分泌物催化结石,润滑肠道;同时其对血小板的凝集有抑制作用,木耳多糖具有抗衰老活性等。用于本发明的木耳菌母种,不限于本发明采用的菌种,一般商购获得的试管母种扩繁为液体母种后均可以用于本发明。
银耳菌:又名白木耳、雪耳、银耳子,拉丁学名为Tremella,属于真菌类银耳科银耳属,是门担子菌门真菌银耳的子实体,有“菌中之冠”的美称。需声明的是,用于本发明的绣球菌母种,不限于本发明采用的菌种,一般商购获得的试管母种扩繁为液体母种后均可以用于本发明。
本领域技术人员,可以根据本发明的内容得到本发明的制品,并且将该制品作为有效成分,添加药学上可接受的药用辅料:淀粉、蔗糖、乳糖、甘露糖醇、甘油、吐温80、碳酸钙、碳酸氢钙、聚乙二醇、环糊精、高岭土、滑石粉、硬脂酸钙、硬脂酸镁、藻酸盐、明胶、聚乙烯吡咯烷酮、纤维素类及其衍生物、磷脂类辅料等。
本发明的口服制品,还可以是任何可药用的剂型,包括:片剂、糖衣片、薄膜衣片、肠溶衣片、胶囊剂、硬胶囊剂、软胶囊剂、口服液、口含片、颗粒剂、丸剂、散剂、膏剂、丹剂、混悬剂、溶液剂、注射剂、栓剂、喷雾剂、滴剂、贴剂等。本发明更优选的药物剂型为口服剂型。
本发明的口服制品,通过发酵,使中药中的多糖与黑木耳菌、银耳菌中的多种多糖发生綴合,使本发明的药物中含有更高含量、更多种类的多糖綴合物,能够显著增强机体免疫力,经过发酵产生的丰富的多糖具有类似非特异性免疫作用,诱导机体能够主动识别表皮增生分化异常,有利于损伤组织/细胞的修复,并利用药物的协同或抵抗作用,以适应复杂病情的治疗要求,减少不良反应,疗效显著,安全可靠,服用简单,对银屑病有较好的治疗效果。共治疗81例,总有效率达99%。
临床实验统计数据显示,本发明的口服制品,对于银屑病患者,一方面,具有显著的疗效,能及时减缓病情的发展,无毒副作用,另一方面,使用方便,能提高患者免疫力,减少复发情况,显著改善患者生存质量
具体实施方式
以下通过具体实施例描述本发明的制品及其制备工艺。
一、材料:
黑木耳试管母种:购自中国农业微生物菌种保藏管理中心,保藏编号50134规格:试管斜面种(试管规格18mm×180mm,约40毫升)。购买到的或现有技术中记载的其它黑木耳菌株也适用本发明。申请人实验室亦有亦有保存,并制备成试管菌丝母种,自申请日起二十年可向公众发放用于实验研究
银耳试管母种:购自中国农业微生物菌种保藏管理中心,保藏编号51863规格:试管斜面种(试管规格18mm×180mm,约40毫升)。购买到的或现有技术中记载的其它银耳菌株也适用本发明。申请人实验室亦有亦有保存,并制备成试管菌丝母种,自申请日起二十年可向公众发放用于实验研究

二、富氢水:日田天领水。
三、设备:
超高压锅:DEPCOA—500,采用超高压灭菌技术(Ultra-highpressureprocessing),简称UHP;高压食品加工技术(highpressure processing)。
发酵罐:为专利号03270560.3,名称为“食用菌液体菌种培养罐的罐盖及培养罐”的实用新型专利中记载的发酵罐。
注射器:专利号200320126960.X,名称为“食用菌液体注射杆及包含该注射杆的注射器”的实用新型专利中记载的注射器。
四:准备母种液:
1)在18L富氢水中加入硫酸镁20g,磷酸氢二钾50g,磷酸二氢钾50g,蔗糖250g,淀粉250g,加富氢水定容至20L。
2)将液体培养基1注入500ml玻璃瓶内,每瓶400ml,一共得50瓶,置入手提高压锅内在0.012MP/cm2,30min,取出后冷却至28℃~34℃,放入超净工作台内,制得液体培养基。
3)将工具、器皿以及液体培养基放入超净工作台内,用30g高锰酸钾置入盛有50%福尔马林30ml的器皿中,接种用的勾、耙、针、刀放入该器皿中灭菌30min。
4)将试管母种,切成3份,每份分别接入400ml/瓶的液体培养基内,静置120min-150min。
5)在摇床上以20次/min,振荡72-96小时,镜检无杂菌,即得母种液。
实施例1.用于银屑病的1号口服制品
配比好的原料药:葛根20,紫草根20,茜草根20,白茅根20,生槐花20,生地20,鸡血藤20,土茯苓50,茵陈20,元参20,莪术20,三棱20,天冬10,麦冬10,白鲜皮10,蜂房10,金银花10,威灵仙10,蒲公英10,紫花地丁10,野菊花10;
步骤1.水,采用超高压锅在30℃、600mpa环境下灭活10分钟,完成后其中40升通过无菌硅胶管道注入无菌发酵罐内备用。
步骤2.提取中药的水提取液:
配比好的原料药装在100目滤网中,在超高压锅加入富氢水和配比好的原料药,重量比为4:1;
在30℃、800mpa环境下处理30分钟,提取中药原液。
步骤3:共发酵
将提取中药的水提取液注入步骤1准备好的已注入40升水的发酵灌中。
再依次注入黑木耳菌、银耳菌的母种液各7升进行发酵。发酵时间到为72小时,检测有效固体物,含量达80g/L以上时放罐停止发酵。
步骤4.浓缩:放罐后,进入醇沉提取罐,加入冷水浸提渗滤,再煮沸,滤汁去渣;其汁加4倍体积的75度酒精浸润,在≦30℃低温下浸制5小时,利用多糖不溶于乙醇的性质,使多糖从提取液中沉淀出来,升温至50摄氏度回收酒精后,对沉淀再进行≦55℃烘干或喷雾干燥,制得多糖粉。
实施例2.用于银屑病的2号口服制品
配比好的原料药2:葛根15,紫草根25茜草根23,白茅根25,生槐花21,生地15,鸡血藤18,土茯苓60,茵陈15,元参25,莪术15,三棱17,天冬8,麦冬9,白鲜皮15,蜂房10,金银花15,威灵仙8,蒲公英9,紫花地丁15,野菊花8;
步骤1.水,采用超高压锅在25℃、600mpa环境下灭活5分钟,完成后其中50升通过无菌硅胶管道注入无菌发酵罐内备用。
步骤2.提取中药的水提取液:
配比好的原料药装在100目滤网中,在超高压锅加入水和配比好的原料药,重量比为3:1;
在28℃、800mpa环境下处理50分钟,提取中药原液。
步骤3:共发酵
将提取中药的水提取液注入步骤1准备好的已注入50升水的发酵灌中。
再依次注入黑木耳菌、银耳菌的母种液各5升进行发酵。发酵时间到为96小时,检测有效固体物,含量达80g/L以上时放罐停止发酵。
步骤4.浓缩:放罐后,进入醇沉提取罐,加入冷水浸提渗滤,再煮沸,滤汁去渣;其汁加4倍体积的75度酒精浸润,在≦30℃低温下浸制5小时,利用多糖不溶于乙醇的性质,使多糖从提取液中沉淀出来,升温至50摄氏度回收酒精后,对沉淀再进行≦55℃烘干或喷雾干燥,制得多糖粉。
实施例3.用于银屑病的3号口服制品
配比好的原料药3:葛根23,紫草根19,茜草根15,白茅根24,生槐花25,生地21,鸡血藤15,土茯苓45,茵陈17,元参16,莪术18,三棱25,天冬9,麦冬15,白鲜皮12,蜂房8,金银花9,威灵仙11,蒲公英15,紫花地丁9,野菊花9;
步骤1.富氢水(日田天领水),采用超高压锅在28℃、600mpa环境下灭活7分钟,完成后其中30升通过无菌硅胶管道注入无菌发酵罐内备用。
步骤2.提取中药的水提取液:
配比好的原料药装在100目滤网中,在超高压锅加入富氢水和配比好的原料药,重量比为5:1;
在35℃、800mpa环境下处理40分钟,提取中药原液。
步骤3:共发酵
将提取中药的水提取液注入步骤1准备好的已注入30升富氢水的发酵灌中。
再依次注入黑木耳菌、银耳菌的母种液各10升进行发酵。发酵时间到为83小时时,检测有效固体物,含量达80g/L以上时放罐停止发酵。
步骤4.放罐后,进入醇沉提取罐,加入冷水浸提渗滤,再煮沸,滤汁去渣;其汁加4倍体积的75度酒精浸润,在≦30℃低温下浸制5小时,利用多糖不溶于乙醇的性质,使多糖从提取液中沉淀出来,升温至50摄氏度回收酒精后,对沉淀再进行≦55℃烘干或喷雾干燥,制得多糖粉。
实施例4.用于银屑病的4号口服制品
配比好的原料药4:葛根25,紫草根15,茜草根18,白茅根15,生槐花16,生地25,鸡血藤22,土茯苓40,茵陈25,元参24,莪术25,三棱23,天冬15,麦冬13,白鲜皮8,蜂房11,金银花14,威灵仙15,蒲公英11,紫花地丁8,野菊花15;
步骤1.富氢水(日田天领水),采用超高压锅在25℃、600mpa环境下灭活15分钟,完成后其中43升通过无菌硅胶管道注入无菌发酵罐内备用。
步骤2.提取中药的水提取液:
配比好的原料药装在100目滤网中,在超高压锅加入富氢水和配比好的原料药,重量比为3.5:1;
在32℃、800mpa环境下处理35分钟,提取中药原液。
步骤3:共发酵
将提取中药的水提取液注入步骤1准备好的已注入43升富氢水的发酵灌中。
再依次注入黑木耳菌、银耳菌的母种液各7升进行发酵。发酵时间到为72小时,检测有效固体物,含量达80g/L以上时放罐停止发酵。
步骤4.浓缩:放罐后,进入醇沉提取罐,加入冷水浸提渗滤,再煮沸,滤汁去渣;其汁加4倍体积的75度酒精浸润,在≦30℃低温下浸制5小时,利用多糖不溶于乙醇的性质,使多糖从提取液中沉淀出来,升温至50摄氏度回收酒精后,对沉淀再进行≦55℃烘干或喷雾干燥,制得多糖粉。
实施例5.用于银屑病的5号口服制品
配比好的原料药5:葛根17,紫草根21,茜草根25,白茅根19,生槐花15,生地18,鸡血藤25,土茯苓53,茵陈22,元参15,莪术21,三棱15,天冬11,麦冬8,白鲜皮9,蜂房15,金银花8,威灵仙12,蒲公英8,紫花地丁13,野菊花12。
步骤1.富氢水(日田天领水),采用超高压锅在32℃、600mpa环境下灭活10分钟,完成后其中37升通过无菌硅胶管道注入无菌发酵罐内备用。
步骤2.提取中药的水提取液:
配比好的原料药装在100目滤网中,在超高压锅加入富氢水和配比好的原料药,重量比为6:1;
在35℃、800mpa环境下处理42分钟,提取中药原液。
步骤3:共发酵
将提取中药的水提取液注入步骤1准备好的已注入37升富氢水的发酵灌中。
再依次注入黑木耳菌、银耳菌的母种液各6升进行发酵。发酵时间到为72小时时,检测有效固体物,含量达80g/L以上时放罐停止发酵。
步骤4.浓缩:放罐后,进入醇沉提取罐,加入冷水浸提渗滤,再煮沸,滤汁去渣;其汁加4倍体积的75度酒精浸润,在≦30℃低温下浸制5小时,利用多糖不溶于乙醇的性质,使多糖从提取液中沉淀出来,升温至50摄氏度回收酒精后,对沉淀再进行≦55℃烘干或喷雾干燥,制得多糖粉。
疗效观察:
一、临床观察
1.临床资料:
本发明人自2008年至2014年共收治81例病例,其中,男性患者59例,女性患者22例,年龄最小者15岁,最大者69岁。20-50岁之间者58例,占71.8%;50岁以上者17例,20岁以下者6例,病程在一年以上的23人,一年以上、5年以下者19人,10年以上者15人,病程最短者一周多,最长这三十多年。
2.治疗方法
服用本发明口服制品:对收治的银屑病患者随机提供实施例1-5制得的口服制品,每种制品提供给至少15例患者服用。每天8g制品(相当于原药420),早晚各一次,连续服用30-90天。
3.诊断标准;
“治愈”:临床症状体征消失,恢复正常。
“显效”:临床症状体征基本或大部分消失。
“有效”:临床症状明显好转。
“无效”:临床症状无改善。
4.治疗效果:
临床治愈55例,占68%;
显效21例,占26%;
有效4例,占5%;无效1例,总有效率为99%。
本发明服用方便,吸收效果好,无副作用,疗效显著,并有利于患者及时减轻病状。
典型病例:
病例1:女士,28岁
2009年11月6日就诊,2年前发现上肢有少许红色点状皮疹,起白色鳞屑,曾到多家医院就诊,被确诊为银屑病,也曾口服外用多种药物未见效。目前四肢,脱屑、瘙痒明显。检查:皮疹呈点状,色潮红,密布四肢,表面有银白色鳞屑,较厚,基底潮红、浸润。检验:血尿便常规未见异常,肝功、肾功正常。服用本发明实施例1口服制品10天后,皮损变薄,基底色由潮红变淡红,未见新生皮疹,痒减轻;再服用10天后,四肢大片皮损已基本消退,表面鳞屑变薄变淡,但色泽微红,仍有少许瘙痒感;继续服药15天,全身皮损消退,治愈。治疗评价:治愈,2010年10月20日回访,未复发。
病例2:李先生,男,65岁
2010年6月8日出诊,从25岁开始,在头顶部出现少数斑疹,上盖银白色鳞屑,曾多次到医院就诊,并确诊为银屑,后来自己也到多地多家医院及私人诊所就诊,使用多种药物治疗未见好转,此次,因酗酒加感冒,躯干、四肢出现他面积皮疹,渐渐融合成大片,由以头部、两下肢为重,全身自感发烧、奇痒难忍;并不断有新皮疹出现。检查:全头顶覆盖银白色鳞屑,大片脱落,躯干、四肢均有鳞屑大如硬币、小如针尖的皮疹,手抓后鳞屑多,剥离后有出血点。检验:血尿便常规未见异常,肝功、肾功正常。服用本发明实施例2口服制品15天后,病情稳定,未见新皮疹出现,原皮疹鳞屑变薄变淡,有所减少,基底呈暗红色,痒痛感减轻,不再发烧;再服用15天后,全身大片皮损已基本消退,表面鳞屑基本消失;继续服药20天,全身皮损消退,治愈。治疗评价:治愈。治愈后,患者担心会复发,继续用药一个半月。2010年10月25日回访,未复发,2014年11月3日回访,未复发。
病例3:肖先生,男,32岁。
2010年12月13日初诊,患病十多年,全身大面积红斑,覆盖银白色鳞屑,鳞屑较厚,尤以背部、颈后严重,瘙痒尤甚,抓破后有点状出血,伴有怕热。多次就医,确诊为银屑病。检验:血尿便常规未见异常,肝功、肾功正常。服用本发明实施例3口服制品15天后,病情稳定,未见新皮疹出现,原皮疹鳞屑变薄变淡,有所减少,基底呈暗红色,痒痛感减轻;再服用15天后,全身大片皮损已基本消退,表面鳞屑基本消失;继续服药20天,全身皮损消退,治愈。治愈后,患者担心会复发,继续用药一个月。治疗评价:治愈,2011年4月20日回访,未复发,2013年4月26日回访,未复发。
病例4:王某,男,43岁,湖南人。
2011年4月17日出诊,3年前,先有背部出现小的皮损,渐渐地,皮损面积越来越大,被医生诊断为银屑病,曾多次治疗未愈。检查见背部有愈合成片的皮损,基底鲜红浸润,表面有银白色多层性鳞屑,剥离鳞屑后底面有出血点。检验:血尿便常规未见异常,肝功、肾功正常。服用本发明实施例4口服制品一周后,皮损变淡,浸润变薄而软,接着服用一周后,皮损明显变淡变薄,表面鳞屑已基本消失,再服用20天后,皮损处只遗留有褐色的斑片,表面鳞屑已全部消失。治疗评价:治愈,2011年6月14日回访,未复发,2014年6月17日回访,未复发。
病例5:崔某,男,19岁,福建人。
2012年3月2日初诊,2年前开始,全身出现大片红斑,覆盖银白色鳞屑。身上干涩的牛皮斑一块连着一块,尤其一到秋冬季节,病情愈加严重。曾多次就医,并确诊为银屑病。检验:血尿便常规未见异常,肝功、肾功正常。服用本发明实施例5口服制品10天后,病情稳定,未见新皮疹出现,原皮疹鳞屑变薄变淡,有所减少,基底呈暗红色,痒痛感减轻;再服药7天后,全身大片皮损已基本消退,表面鳞屑基本消失;继续服药15天,全身皮损消退,治愈。治疗评价:治愈,2012年5月17日回访,未复发。

1.一种用于银屑病的口服制品,其特征在于:
是以下原料药的水提取液与黑木耳菌、银耳菌母种液进行共发酵所得的产物;
所述原料药具有以下重量份配比:葛根15-25,紫草根15-25,茜草根15-25,白茅根15-25,生槐花15-25,生地15-25,鸡血藤15-25,土茯苓40-60,茵陈15-25,元参15-25,莪术15-25,三棱15-25,天冬8-15,麦冬8-15,白鲜皮8-15,蜂房8-15,金银花8-15,威灵仙8-15,蒲公英8-15,紫花地丁8-15,野菊花8-15。

6.权利要求1-3任一所述的用于银屑病的口服制品的制备方法,包括如下步骤:
(1)制备原料药的水提取液;
(2)原料药的水提取液与黑木耳菌母种液、银耳菌母种液进行共发酵得到发酵产物;
其特征在于,所述原料药的水提取液的提取工艺如下:在提取容器中,加入富氢水和原料药;水与所述原料药重量比例为3-6:1,在25-35℃、800mpa环境下提取30-50分钟得到所述原料药的水提取液。
7.根据权利要求6所述的制备方法,其特征在于:
水与所述原料药重量比例为4-5:1,在28-32℃、800mpa环境下提取30-40分钟得到所述原料药的水提取液。
8.根据权利要求6所述的制备方法,其特征在于:所述进行共发酵是指在发酵罐中,按30-50:30-50:5-10:5-10的体积比加入所述原料药的水提取液、水、黑木耳菌母种液、银耳菌母种液,发酵到有效固体物含量达80g/L以上时放罐。
9.根据权利要求8所述的制备方法,其特征在于:
按35-45:35-45:6-8:6-8的体积比加入所述原料药的水提取液、水、黑木耳菌母种液、银耳菌母种液。
10.根据权利要求6-9任一所述的制备方法,其特征在于:
放罐后,进入醇沉提取罐,加入冷水浸提渗滤,再煮沸,滤汁去渣;其汁加4倍体积的75度酒精浸润,在≦30℃低温下浸制5小时,利用多糖不溶于乙醇的性质,使多糖从提取液中沉淀出来,升温至50摄氏度回收酒精后,对沉淀再进行≦55℃烘干或喷雾干燥,制得多糖粉;
优选的,所述水为富氢水,所述制备方法还包括富氢水的灭活,将所述富氢水在25-35℃、600mpa环境下灭活5-30分钟。
