Abstract
本发明公开了一种三叶青抗菌液及其制备方法,称取组分中各中药,粉碎,置中药提取灌中,加入6~10倍量的60‑80%乙醇,将乙醇与粗粉混合后加温到50‑70℃,恒温24‑48小时,过滤后再次提取,合并两次滤液,用活性炭脱色,获得提取液。将0.2%‑0.4%冰片加入提取液中,充分搅拌至冰片完全溶解,获得混合液,并调节乙醇浓度至35~45%,搅拌均匀后,将其密封静置24h,将静置液过滤,用灌装机进行灌装,即得成品。本发明制备的抗菌液,属于天然抗菌剂,是从天然植物中提取,在生产和使用过程中对人类和环境不产生污染危害,生物相溶性好,适合工业化生产。

技术领域
本发明涉及一种三叶青抗菌液及其制备方法,属于抗菌液技术领域。
背景技术
随着人们生活环境的改善,与各种致病病原微生物(细菌、真菌、酵母菌、藻类及病毒等)的接触仍不可避免,它们的传播和蔓延严重威胁着人类的健康,痱子、痘痘、蚊虫叮咬引起的瘙痒对皮肤黏膜有一定的刺激性,并容易使皮肤创伤而造成感染,从而引发各种疾病。如何有效控制有害细菌的生长和繁殖,显得十分有意义,而抗菌液能够在一定时间内,使某些致病微生物的生长或繁殖保持在必要水平以下的物质。抗菌液在生活中应用日益广泛,其中北美是使用抗菌剂最多的地区,占全球总用量的40%,其次是日本,占据20%,而日本的人均抗菌剂使用量最大,远超北美和欧洲。抗菌剂一般分为无机抗菌剂,有机抗菌剂和天然抗菌剂。无机抗菌剂具有耐热性、持续性、抗菌谱光等优点,但其价格较高,且抗菌的迟效性,不能像有机抗菌剂那样能快速杀死细菌。有机抗菌剂能有效抑制有害细菌、霉菌的产生与繁殖,见效快,但这类抗菌剂热稳定性较差,持久性差,而且通常毒性较大,长时间使用对人体有害。天然抗菌剂,是从天然植物中提取,在生产和使用过程中对人类和环境一般不产生污染危害,生物相溶性好,但抗菌功能有限,很难做到光谱长效。
申请号为201510253667.7的中国发明申请,公开了一种红花酊抗菌液及其制备方法,其特征在于:该抗菌液由下述质量百分比的原料制成:樟脑5-8g,冰片100-110g,细辛15-20g,黄连5-8g,莪术40-45g,三棱40-45g,血竭5-8g,红花40-45g,蟾酥4-6g,无水乙醇7-8kg,乙醚1.0-1.2kg,丙酮1.0-1.0kg,乙基纤维素18.5kg。以上抗菌液的配方组成中有低沸点的乙醚和丙酮存在,极易燃烧或者爆炸,在生产过程中存在极大安全隐患,应极力避免,因此不适宜推广使用。本发明涉及的一种三叶青抗菌液,属于天然抗菌剂,是从天然植物中提取,在生产和使用过程中对人类和环境一般不产生污染危害,生物相溶性好,适合工业化生产。经过科学组方配伍和先进制备工艺,无毒副作用,无环境污染,并克服了天然抗菌剂的抗菌功能有限、持续性差等缺点。
发明内容
本发明的目的在于提供一种抗菌效果好、安全性高、稳定性好及生产工艺先进的三叶青抗菌液及其制备方法,该抗菌液组方配伍合理,生产工艺可操作性程度高,是一种外用的天然抗菌剂,适用于皮肤表层抑菌,对金黄色葡萄球菌、绿脓杆菌、大肠杆菌、堇色毛癣菌等有较好的杀灭作用,抑菌效果明显,对蚊虫叮咬,皮肤瘙痒有较好的消肿止痛止痒的功效,并能祛痱祛痘,对皮肤刺激性小。
为解决上述问题,本发明所采取的技术方案如下:
一种三叶青抗菌液,称取组分中各中药,粉碎,置中药提取灌中,加入6~10倍量的60-80%乙醇,将乙醇与粗粉混合后加温到50-70℃,恒温24-48小时,过滤后再次提取,合并两次滤液,用活性炭脱色,获得提取液。将0.2%-0.4%冰片加入提取液中,充分搅拌至冰片完全溶解,获得混合液,并调节乙醇浓度至35~45%,搅拌均匀后,将其密封静置24h,将静置液过滤,用灌装机进行灌装。
作为上述技术方案的改进,所述称取组分中各中药,粉碎,取4份三叶青,2份苦参,2份白鲜皮,2份黄芩,2份板蓝根,1份青蒿,1份半枝莲,2份地肤子,并粉碎成粗粉,过30目筛。
作为上述技术方案的改进,加入10倍量的70%乙醇,将乙醇与粗粉混合后加温到50℃,恒温48小时,过滤后再次提取,合并两次滤液,用活性炭脱色,获得提取液。
作为上述技术方案的改进,过滤后再次提取,加入8倍量70%乙醇,继续加热至50℃,恒温保持24小时,提取液用活性炭脱色。
作为上述技术方案的改进,脱色液加入0.2%冰片,充分搅拌至冰片溶解,加入纯净水,调整乙醇浓度至35%,密封静置24小时,过滤,获得棕黄色澄清液体,用灌装机灌装即得。
具体地,所述三叶青抗菌液的制备方法由以下步骤构成:
(1)称取各中药,粉碎,置中药提取灌中,加入乙醇,加热提取,提取液过滤后经活性炭脱色,加入冰片,充分搅拌,并调节醇浓度至合适范围,静置后过虑,包装,即得。
(2)所述步骤(1)中,取3-4份三叶青,1-2份苦参,1-2份白鲜皮,1-2份黄芩,1-2份板蓝根,1-2份青蒿,1-2份半枝莲,1-2份地肤子,并粉碎成粗粉,过30目筛。
(3)所述步骤(1)中,加入粗粉总量的6~10倍量的60-80%乙醇,将乙醇与粗粉混合后加温到50-70℃,恒温24-48小时,过滤后再次提取,合并两次滤液,用活性炭脱色,获得提取液;
(4)所述步骤(1)中,将0.2%-0.4%冰片加入提取液中,充分搅拌至冰片完全溶解,获得混合液,并调节乙醇浓度至35~45%,搅拌均匀后,将其密封静置24h;
(5)在所述步骤(1)中,将静置液过滤,用灌装机进行灌装,即得成品。
本发明与现有技术相比较,本发明的实施效果如下:
本发明采用乙醇作为溶剂,低温提取,在生产和使用过程中对人类和环境不产生污染危害,可操作性强,适合工业化生产;一种三叶青抗菌液,其组方配伍合理,各成分含有的多种抑菌、镇痛成分,相辅相成,发挥协同作用,克服了天然抗菌剂的抗菌功能有限、持续性差等缺点。
此外,本发明适用于皮肤表层抑菌,对金黄色葡萄球菌、绿脓杆菌、大肠杆菌、堇色毛癣菌、同心性毛癣菌等有较好的杀灭作用,抑菌效果明显,对蚊虫叮咬,皮肤瘙痒有较好的消肿止痛止痒的功效,并能祛痱祛痘,对皮肤刺激性小。
具体实施方式
下面将结合具体的实施例来说明本发明的内容。
本发明所述的三叶青抗菌液的制备方法,由以下原料组成:3-4份三叶青,1-2份苦参,1-2份白鲜皮,1-2份黄芩,1-2份板蓝根,1-2份青蒿,1-2份半枝莲,1-2份地肤子,0.2-0.4%冰片,60-80%乙醇。
所述三叶青味微苦,性平,具有清热解毒、祛风化痰、活血止痛等功效,主治毒蛇咬伤、扁桃体炎、淋巴结结核、跌打损伤、小儿高热惊厥等疾病。
黄芩味苦,性寒,有清热燥湿,凉血安胎,泻火解毒等功效。黄芩对对痢疾杆菌、伤寒杆菌、副伤寒杆菌、霍乱弧菌、大肠杆菌、变形杆菌、绿脓杆菌、葡萄球菌、溶血性链球菌等有抑制作用,在临床上抗菌性比黄连好,且不产生抗药性。
板蓝根味苦,性寒,有清热解毒、凉血利咽之功效,主要用于治疗温毒发斑、舌绛紫暗、烂喉丹痧等疾病。水浸液对枯草杆菌、金黄色葡萄球菌、八联球菌、大肠杆菌、伤寒杆菌、副伤寒杆菌、痢疾(志贺、弗氏)杆菌、肠炎杆菌等都有不同程度的抑制作用。
苦参味苦,性寒,有清热燥湿,杀虫,利尿之功。用于热痢,湿疹,湿疮,皮肤瘙痒,疥癣麻风等症。其醇提物对金黄色葡萄球菌有较强的抑菌作用;苦参水浸剂对堇色毛癣菌、同心性毛癣菌、许兰毛癣菌、奥杜盎小芽孢癣菌等有抑制作用。
白鲜皮味苦,性寒,具有清热燥湿,祛风解毒之功效。用于风热湿毒所致的风疹、湿疹、疥癣、黄疸、湿热痹。体外试验表明1:4的白鲜皮水浸液对多种致病真菌如堇色毛癣菌、同心性毛癣菌、许兰氏黄癣菌,均有不同程度的抑制作用。
地肤子味辛、苦,性寒,具有清热利湿,祛风止痒功效。用于阴痒带下,风疹,湿疹,皮肤瘙痒。水浸剂(1:3)在试管内对许兰氏黄癣菌,奥社盎氏小芽胞癣菌等皮肤真菌有抑制作用。水浸液用试管稀释法,1:3对许兰黄癣菌、奥杜盎小孢子菌及星形奴卡菌等均有抑制作用。
青蒿味苦、性寒,具有清虚热,除骨蒸,解暑热,截疟,退黄之功效。常用于温邪伤阴,夜热早凉,阴虚发热,骨蒸劳热,暑邪发热,疟疾寒热,湿热黄疸等病症的治疗。其水煎剂对多种细菌有抑制作用。
半枝莲味辛,性平,具有凉血解毒,散瘀止痛,消肿和清热利湿之功效,用于咽喉肿痛、毒蛇咬伤、跌打损伤、疔疮肿毒、水肿、黄疽等症。半枝莲50%煎剂用平板挖沟法,对金黄色葡萄球菌、福氏痢疾杆菌、伤寒杆菌、绿脓杆菌、大肠杆菌有抑制作用。
冰片味辛、性凉,具有开窍醒神,清热散毒,明目退翳的功效。外用能清热消肿、止痒止痛。
乙醇:在三叶青抗菌溶液中起到溶解有效成份的作用,也作为冰片的溶剂,使其便于加入溶剂中。
实施例1:
称取3份三叶青,1份苦参,1份白鲜皮,1份黄芩,1份板蓝根,1份青蒿,1份半枝莲,1份地肤子,分别碎成粗粉,过30目筛,置提取罐中,加入8倍量60%乙醇,加热至50℃,恒温保持24小时,放冷过滤,再加入6倍量60%乙醇,继续加热至50℃,恒温保持24小时,合并两次提取液,用活性炭脱色后加入0.2%冰片,充分搅拌至冰片溶解,加入纯净水,调整乙醇浓度至35%,密封静置24小时,过滤,获得棕黄色澄清液体,灌装即得。
实施例2:
称取3份三叶青,1份苦参,1份白鲜皮,1份黄芩,1份板蓝根,1份青蒿,1份半枝莲,1份地肤子,分别碎成粗粉,过30目筛,置提取罐中,加入8倍量70%乙醇,加热至50℃,恒温保持24小时,放冷过滤,再加入6倍量70%乙醇,继续加热至50℃,恒温保持24小时,合并两次提取液,用活性炭脱色后加入0.3%冰片,充分搅拌至冰片溶解,加入纯净水,调整乙醇浓度至40%,密封静置24小时,过滤,获得棕黄色澄清液体,灌装即得。
实施例3:
称取3份三叶青,1份苦参,1份白鲜皮,1份黄芩,1份板蓝根,1份青蒿,1份半枝莲,1份地肤子,分别碎成粗粉,过30目筛,置提取罐中,加入8倍量80%乙醇,加热至50℃,恒温保持24小时,放冷过滤,再加入6倍量80%乙醇,继续加热至50℃,恒温保持24小时,合并两次提取液,用活性炭脱色后加入0.4%冰片,充分搅拌至冰片溶解,加入纯净水,调整乙醇浓度至45%,密封静置24小时,过滤,获得棕黄色澄清液体,灌装即得。
实施例4:
称取3份三叶青,2份苦参,2份白鲜皮,2份黄芩,2份板蓝根,1份青蒿,1份半枝莲,2份地肤子,分别碎成粗粉,过30目筛,置提取罐中,加入8倍量60%乙醇,加热至50℃,恒温保持36小时,放冷过滤,再加入6倍量60%乙醇,继续加热至50℃,恒温保持36小时,合并两次提取液,用活性炭脱色后加入0.2%冰片,充分搅拌至冰片溶解,加入纯净水,调整乙醇浓度至35%,密封静置24小时,过滤,获得棕黄色澄清液体,灌装即得。
实施例5:
称取3份三叶青,2份苦参,2份白鲜皮,2份黄芩,2份板蓝根,1份青蒿,1份半枝莲,2份地肤子,分别碎成粗粉,过30目筛,置提取罐中,加入8倍量70%乙醇,加热至60℃,恒温保持36小时,放冷过滤,再加入6倍量70%乙醇,继续加热至60℃,恒温保持36小时,合并两次提取液,用活性炭脱色后加入0.3%冰片,充分搅拌至冰片溶解,加入纯净水,调整乙醇浓度至40%,密封静置24小时,过滤,获得棕黄色澄清液体,灌装即得。
实施例6:
称取3份三叶青,2份苦参,2份白鲜皮,2份黄芩,2份板蓝根,1份青蒿,1份半枝莲,2份地肤子,分别碎成粗粉,过30目筛,置提取罐中,加入8倍量80%乙醇,加热至70℃,恒温保持36小时,放冷过滤,再加入6倍量80%乙醇,继续加热至70℃,恒温保持36小时,合并两次提取液,用活性炭脱色后加入0.4%冰片,充分搅拌至冰片溶解,加入纯净水,调整乙醇浓度至45%,密封静置24小时,过滤,获得棕黄色澄清液体,灌装即得。
实施例7:

本发明所述三叶青抗菌液多次皮肤刺激试验:
试验方法:将新西兰家兔的背部脊柱两侧被毛剪掉,去毛范围各位3cm×3cm,涂敷面积2.5cm×2.5cm,取受试物涂敷在一侧皮肤上,另一侧作为对照,在涂抹后4h,用水或无刺激的适宜溶剂清洗,除去残留物;每天涂敷1次,连续涂敷14d;在每次涂抹后24h观察结果,按规范中的相关要求进行评分和分级;微量便于受试物的涂抹和结果观察,必要时应剪毛;对照区的处理方法通试验区。
试验结果,见表1:

结果表明受试物对家兔多次皮肤刺激性检验结果为无刺激性,未见其他毒性。
中和剂鉴定试验和细菌定量杀灭试验:
器材:
①试验菌株:大肠杆菌8099、金黄色葡萄球菌ATCC6538由中国工业微生物种保藏管理中心提供,第4-8代新鲜斜面培养物;
②中和剂:D/E肉汤;
③其他:浓度为3%的有机干扰物,标准硬水等。
方法:
①检测依据:中和剂鉴定试验及定量杀灭试验按照卫生部《消毒技术规范》(2002版)2.1.1.5.5和2.1.1.7.4条;
②中和剂鉴定试验:以该试验样品4倍稀释液和大肠杆菌作用0.5min。试验温度19℃-21℃,试验重复3次;
③定量杀灭试验:以该试验样品原液,作用时间分别为2.5min、5min、7.5min,试验温度19℃-21℃,试验重复3次。
结果:
①中和剂鉴定试验,3次重复试验中,3、4、5三组间回收菌落误差分别为9.77%、10.90%、5.97%。结果见表2。

注:阴性对照均无菌生长。
②对试验结果的杀灭效果
经3次重复试验,结果表明该发明一种三叶青抗菌液对大肠杆菌作用2.5min,杀灭对数值>5.00;对金黄色葡萄球菌作用2.5min、5min、7.5min,平均杀灭对数值分别为2.36、3.95,>5.00。结果见表3。

结论:
①:经3次重复试验,结果表明D/E肉汤中和剂可中和三叶青抗菌液中的杀菌成分,且中和剂及中和产物对试验菌生长及培养基无明显影响。表明该中和剂适用于该试验样品对大肠杆菌、金黄色葡萄球菌的定量杀灭试验。
②:经3次重复试验表明,该发明的三叶青抗菌液对悬液中的大肠杆菌作用2.5min,杀灭对数值>5.00;对金黄色葡萄球菌作用2.5min、5min、7.5min,平均杀灭对数值分别为2.36、3.95,>5.00。

Claims (2)
1.一种三叶青抗菌液,其特征在于:由以下原料组成:10-18份各组分中药、0.2%-0.4%冰片、60-80%乙醇;
所述各组分中药具体包括3-4份三叶青,1-2份苦参,1-2份白鲜皮,1-2份黄芩,1-2份板蓝根,1-2份青蒿,1-2份半枝莲,1-2份地肤子。
2.根据权利要求1所述一种三叶青抗菌液的制备方法,其特征在于:所述方法由以下步骤构成:
(1)称取各中药,粉碎,置中药提取灌中,加入乙醇,加热提取,提取液过滤后经活性炭脱色,加入冰片,充分搅拌,并调节醇浓度至合适范围,静置后过虑,包装,即得;
(2)在所述步骤(1)中,称取3-4份三叶青,1-2份苦参,1-2份白鲜皮,1-2份黄芩,1-2份板蓝根,1-2份青蒿,1-2份半枝莲,1-2份地肤子,并粉碎成粗粉,过30目筛;
(3)在所述步骤(1)中,加入粗粉总量的6~10倍量的60-80%乙醇,将乙醇与粗粉混合后加温到50-70℃,恒温24-48小时,过滤后再次提取,合并两次滤液,用活性炭脱色,获得提取液;
(4)在所述步骤(1)中,将0.2%-0.4%冰片加入提取液中,充分搅拌至冰片完全溶解,获得混合液,并调节乙醇浓度至35~45%,搅拌均匀后,将其密封静置24h;
(5)在所述步骤(1)中,将静置液过滤,用灌装机进行灌装,即得成品。
