本发明涉及中药领域,特别涉及一种检验科用杀菌消毒液及制备方法。
背景技术
检验科是临床医学和基础医学之间的桥梁,每天承担包括病房、门急诊病人、各类体检以及科研的各种人体和动物标本的检测工作,其中包括血液、尿样、粪样等。
在工作过程中,由于医护人员经常接触这些样品,因此,很容易沾染上一些细菌,而检验所使用的杀菌消炎药物如碘酒、酒精、碘伏等大多对人体伤口以及皮肤有刺激性,有的甚至会引起过敏反应,甚至会引发皮炎和湿疹等皮肤疾病。
为此,医疗机构进行医学检验时,检验系统的细菌群系应减少到最低限度,这便对医护人员本身以及检验所需的检验用具进行全方位的彻底的消毒,消除一系列的细菌以及病原菌例如金黄色葡萄球菌、大肠杆菌、伤寒沙门菌、绿脓假单胞菌、普通变形杆菌等,为检验系统提供良好的环境。因此寻找一种杀菌效果好且无刺激性副作用的消毒液,对检验科工作人员及患者都是非常重要的。

一种检验科用杀菌消毒液的制备方法,包括以下工艺步骤:
(1)取土茯苓、水松、盐麸子和茶叶洗净,将醋加入其中,拌匀,文火闷润至醋被吸尽时,置适宜的容器内密闭,用蒸气加热2-4小时,取出干燥,研末,得到过200目的细粉,备用;
(2)取小蜡树研末后,浸入体积浓度为90%酒精溶液,静置4小时,取出上层清液,沉淀下来的浸膏备用;
(3)取蛇含、大蒜、水茴香、八仙草、槐叶和百两金绞汁,滤杂质,留汁
液,备用;
(4)取桉叶、柽柳、浮萍和枣叶放入容器中,加入10倍量蒸馏水浸泡4小时后,加热煮沸4小时,提取;第二次加入8倍量蒸馏水加热煮沸3小时,提取;第三次加入6倍量蒸馏水加热煮沸2小时,提取;合并三次所得提取液,滤过,得滤液备用;
(5)将步骤(1)所得的细粉、步骤(2)所得的浸膏、步骤(3)所得的汁液以及步骤(4)所得的滤液混合后,加入10-12倍量的蒸馏水,搅拌溶解均匀后,加热煮沸0.5小时,至60℃以下,静置1小时,过滤,得滤液,将滤液灌装封存,121℃热压灭菌20分钟,包装,即为所述的检验科用杀菌消毒液。
本发明所用中药的药性如下:
土茯苓:味甘、淡,性平;归肝、胃、脾经。清热除湿,泄浊解毒,通利关节。主梅毒,淋浊,泄泻,筋骨挛痛,脚气,痈肿,疮癣,瘰疬,瘿瘤及汞中毒。《江西草药》载:“杀虫解毒。治瘰疬,小儿疳积。”
水松:味甘、咸,性寒;归膀胱、大肠经。利水消肿,杀虫解毒。主水肿,蛔虫病,中暑,小便不利。《本草经集注》载:“疗溪毒。”现代医学研究证实,水松浸出液驱虫效果较海人草强三倍。水松所含成分丙烯酸有强大的抑菌作用。芳樟醇和牻牛儿醇有抗菌、抗真菌作用。
小蜡树:味苦,性凉;清热利湿,解毒消肿。主感冒发热,肺热咳嗽,咽喉肿痛,口舌生疮,湿热黄疸,痢疾,痈肿疮毒,湿疹,皮炎,跌打损伤,烫伤。现代医学研究证实,小蜡树叶对金黄色葡萄球菌、伤寒杆菌。甲型副伤寒杆菌、绿脓杆菌、大肠杆菌、弗氏痢疾杆菌、肺炎杆菌有极强的抗菌作用。
蛇含:味苦,性寒;归肝、肺经。清热定惊,截疟,止咳化痰,解毒活血。主高热惊风,疟疾,肺热咳嗽,百日咳,痢疾,疮疖肿毒,咽喉肿痛,风火牙痛,带状疱疹,目赤肿痛,虫蛇咬伤,风湿麻木,跌打损伤,月经不调,外伤出血。《草木便方》载:“发汗解肌。治风痰咳嗽,惊痫,洗眼消毒。”
大蒜:味辛,性温;归脾、胃、肺、大肠经。温中行滞,解毒,杀虫。主脘
腹冷痛,痢疾,泄泻,肺痨,百日咳,感冒,痈疖肿毒,肠痈,癣疮,蛇虫咬伤,钩虫病,蛲虫病,带下阴痒,疟疾,喉痹,水肿。《别录》载:“散痈肿蠿疮,除风邪,杀毒气。”现代医学研究证实,生大蒜在试管内对化脓性球菌、结核杆菌、痢疾杆菌、伤寒杆菌、副伤寒杆菌、霍乱弧菌等均有抑制作用;对青、链、氯、金霉素耐药的细菌对大蒜制剂仍敏感。生大蒜水浸液在体外对脑膜炎双球菌有较强的抑菌作用;对多种致病性皮肤真菌(包括白色念珠菌)均有不同程度的抑制作用或杀灭作用;对恙虫热立克次体有很强的抑杀力。
水茴香:味辛,性平;归肺、脾、胃经。清热解表,祛风除湿,止咳止痛。用于感冒,咽喉肿痛,肺热咳嗽,支气管炎,胃痛。外用治天疱疮。《南宁市药物志》载:“燥湿去痰,消肿止痛,杀虫解毒。治气喘咳嗽,小儿奶瘕,疮疖,蛇伤,胃寒痛。”
桉叶:味辛、苦,性寒;归肺、胃、脾、肝经。疏风解表,清热解毒,化痰理气,杀虫止痒。主感冒,高热喘咳,百日咳,脘腹胀痛,腹泻,痢疾,钩、丝虫病,疟疾,风湿痛,湿疹,疥癣,烧烫伤,外伤出血。李承祜《生药学》载:“煎液,治丹毒与其他传染性化脓症。”现代医学研究证实,用柱层析法从蓝桉中分离出一种物质,对革兰氏阳性细菌有抑制作用,在试管内对破伤风杆菌及白喉杆菌毒素有解毒效力。
八仙草:味辛、微苦,性微寒;归少阴、太阴经。清热解毒,利尿通淋,消肿止痛。主痈疽肿毒,乳腺炎,阑尾炎,水肿,感冒发热,痢疾,尿路感染,尿血,牙龈出血,刀伤出血。《昆明民间常用草药》载:"“清热消炎,祛风湿,散瘀血,治痰火热症,风热眼雾眼屎多,风湿跌打疼痛,外伤皮下瘀肿。”现代医学研究证实,100%煎剂用平板纸片法,对金黄色葡萄球菌、大肠杆菌、志贺痢疾杆菌等有抑制作用。
柽柳:味甘、辛,性平;归肺、胃、心经。疏风解表,透疹解毒。主风热感冒,麻疹初起,疹出不透,风湿痹痛,皮肤瘙痒。《东医宝鉴》载:“主疥癣及一切恶疮。”现代医学研究证实,本品煎剂对肺炎球菌、甲型链球菌、白色葡萄球
菌和流感杆菌均有抑制作用。柽柳酮和柽柳醇对耐药金黄色葡萄球菌有较强的抑制作用。
醋:味酸、甘,性温;归肝、胃、经。散瘀消积,止血,安蛔,解毒。主产后血晕,症瘕积聚,吐血,衄血,便血,虫积腹痛,鱼肉菜毒,痈肿疮毒。《本草再新》载:“生用可以消诸毒,行湿气;制用可宣阳,可平肝,敛气镇风,散邪发汗。”现代医学研究证实,乙酸对甲型链球菌、卡他球菌、肺炎链球菌、白色葡萄球菌、流感病毒等致病菌,有很好的抑菌和杀菌作用。
浮萍:味辛,性寒;归肺、膀胱经。发汗解表,透疹止痒,利水消肿,清热解毒。主风热表证,麻疹不透,隐疹瘙痒,水肿,癃闭,疮癣,丹毒,烫伤。《纲目》载:“浮萍,其性轻浮,入肺经,达皮肤,所以能发扬邪汗也。”现代医学研究证实,浮萍抗菌、抗疟实验均为阴性。在实验室及现场对库蚊幼虫及蚊蛹有杀灭作用。
槐叶:味苦,性平;归肝、胃经。清肝泻火,凉血解毒,燥湿杀虫。主小儿惊痫,壮热,肠风,尿血,痔疮,湿疹,疥癣,痈疮疔肿。《食疗本草》载:“主邪气,产难,绝伤。又主瘾疹,牙齿诸风疼。”
盐麸子:味酸、咸,性凉。生津润肺,降火化痰,敛汗止痢。主痰嗽,喉痹,黄疸,盗汗,痢疾,顽癣,痈毒,头风白屑。《纲目》载:“生津降火,化痰,润肺滋肾,消毒,止痢收汗。治风湿,眼病。”
茶叶:味苦、甘,性凉;归心、肺、胃、肝、脾、肾经。清头目,除烦渴,消食,化痰,利尿,解毒。主头痛,目昏,目赤,多睡善寐,感冒,心烦口渴,食积,口臭,痰喘,癫痫,小便不利,泻痢,喉肿,疮疡疖肠,水火烫伤。《日用本草》载:“除烦止渴,解腻清神。炒煎饮,治热毒赤白痢;同芎藭、葱白煎饮,止头痛。”现代医学研究证实,茶叶浸剂或煎剂在试管中,对各型痢疾杆菌皆具有抗菌作用,其抑菌效果与黄连不相上下。一般而言,花茶、绿茶的抗菌效能大于红茶,对志贺氏痢疾杆菌的作用强于其他三型(福氏、施氏、宋内氏)。对沙门氏菌、金黄色葡萄球菌、乙型溶血性链球菌、白喉杆菌、炭疽杆菌、枯草
杆菌、变形杆菌、绿脓杆菌等亦有抑菌作用。
枣叶:味甘,性温。清热解毒。主小儿发热,疮疖,热痱,烂脚,烫火伤。《本草求原》载:“洗疳、痔、疔、烂脚、结毒。枝熬膏消毒。”
百两金:味苦、辛,性凉。清热利咽,祛痰利湿,活血解毒。主咽喉肿痛,咳嗽咯痰不畅,湿热黄疸,小便淋痛,风湿痹痛,跌打损伤,疔疮,无名肿毒,蛇咬伤。《福建中草药》载:“清热利湿。治湿热黄疸,痢疾,白浊,睾丸肿大坠痛,秃疮,疥癣。”现代医学研究证实,百两金对肺炎双球菌,痢疾杆菌有抑制作用。

土茯苓85g 水松83g 小蜡树81g 蛇含78g
大蒜76g 水茴香76g 桉叶73g 八仙草73g
柽柳66g 醋66g 浮萍64g 槐叶60g
盐麸子60g 茶叶58g 枣叶53g 百两金38g。
本发明的制备方法,包括以下工艺步骤:

实验部分
下面将选用本发明进行相应的试验,进一步证明本发明的功效。
一、水溶性试验
1、取本发明消毒液10ml放入容器中,然后加入蒸馏水,搅拌均匀后,过滤。
2、过滤完成后,观察滤纸,滤纸上物明显残留物残留。
3、结论:本发明消毒液极易溶于水,使用后无残留杂质出现。
二、毒性试验
1、取本发明消毒液
1、急性经口性试验:选取质量20-22g的健康昆明种一级小鼠20只,雌雄各半,将本发明消毒液经口灌胃给药,给药观察15天,每天记录小鼠各种表现和死亡情况,计算LD.值。
2、蓄积毒性试验:选取质量180-200g的健康昆明种一级大鼠20只,雌雄各半,将本发明消毒液经口灌胃给药,给药量根据急性经口性试验得出的DL.值,第1-4天,每次给药0.1LD.,以后每四小时递增1.5倍剂量,给药30天,且每四小时称重一次,计算蓄积系数。
3、结果:急性经口性试验小鼠灌药后未见异常反应,未出现死亡,对其进行相应解剖后,组织或内脏未见明显有明显异常改变。
蓄积毒性试验给药20天后,雌雄小鼠均无死亡情况发生,小鼠蓄积系数5,位弱蓄积毒性。
三、眼部刺激性试验
1、选用健康白色家兔4只,体质量2.5-3.0kg,将本发明消毒液原液0.1ml滴入家兔左眼结膜囊内,被动闭合4s后用生理盐水冲洗,右眼作为对照滴入生理盐水。
2、观察家兔6小时、24小时、48小时以及72小时内的眼睛局部变化。
3、结果:家兔染毒组角膜和虹膜正常,结膜未见水肿,有轻微充血,结膜刺激指数为1,按急性眼刺激强度分析强度分级标准,属无刺激性。
四、皮肤刺激性试验
1、选用健康白色家兔4只,体质量2.5-3.0kg,将家兔背部脊柱两侧脱毛分别为3.0cm×3.0cm,不损伤表皮,24小时后,将0.2ml本发明消毒液的原液均匀涂抹在裸露皮肤的左侧,右侧作为对照不涂抹,4小时后,将皮肤擦拭干净,观察皮肤反应,进行刺激评分。
2、结果:家兔裸露皮肤未见有红斑和水肿反应,皮肤刺激反应积分值为0,对照侧皮肤未见异常,按皮肤刺激反应评分标准,反应积分均值为0,本发明消毒液对皮肤无刺激性。
五、稳定性试验
取本发明消毒液50ml,在37℃条件下放置90天后,用于对金黄色葡萄球菌进行杀菌,其杀灭效果和储存前比较无明显差异。
六、抑菌试验
下面对本发明消毒液进行抑菌实验:
1、实验菌种:
挑选实验室具有代表性的菌种,金黄色葡萄球菌和绿脓杆菌。
2、菌种培养基:
营养琼脂培养基和营养肉汤培养基。
3、实验分类:
抑菌实验分实验组和对照组,实验组选用本发明消毒液,对照组选用来苏水。
4、具体实验步骤:
(1)菌液的制备:将上述两种菌种分别接种于两个琼脂培养基上,在370C下培养24小时,分别从上述琼脂培养基中挑取典型菌落分别接种于两个新鲜的营养肉汤培养基中,同温度下,培养20小时,备用;
(2)取本发明消毒液和来苏水各55ml,分别置于稀释容器中,对其分别稀释25倍,将上述两种稀释液溶液分别分置两个容器内,置于无菌条件下,备用;
(3)将上述步骤(1)经营养肉汤培养24小时的实验菌种用无菌的营养肉汤分别稀释50倍,每种菌种分装于两个培养皿中,分别标记对照组和实验组;
(4)将上述步骤(2)所得的两份本发明消毒液的稀释溶液分别滴入实验组培养皿,两份来苏水稀释溶液分别滴入对照组培养皿。
5、试验观察测量:
对上述实验进行观察,观察两组菌种并测量菌种残留面积具体结果。
6、观测测量完毕后,将上述培养皿均置于无菌条件下静置两天,待消毒液完全挥发后,跟踪观察发现,对照组菌落继续繁殖,而实验组菌落无变化。
7、实验结果:
实验组本发明消毒液的杀菌能力远大于对照组消毒液的杀菌能力,并且能够抑制各类菌种继续繁殖。

1.一种检验科用杀菌消毒液,其特征在于:制成所述消毒液有效成分的原料组成及重量份数为:
土茯苓57-87份 水松55-85份 小蜡树53-83份 蛇含52-80份
大蒜50-78份 水茴香50-78份 桉叶47-75份 八仙草45-75份
柽柳40-68份 醋40-68份 浮萍38-66份 槐叶35-62份
盐麸子35-62份 茶叶33-60份 枣叶30-55份 百两金20-40份。
2.根据权利要求1所述的一种检验科用杀菌消毒液,其特征在于:制成所述消毒液有效成分的原料组成及重量份数为:
土茯苓78份 水松76份 小蜡树74份 蛇含72份
大蒜70份 水茴香70份 桉叶65份 八仙草65份
柽柳58份 醋58份 浮萍56份 槐叶53份
盐麸子53份 茶叶51份 枣叶45份 百两金33份。
3.根据权利要求1所述的一种检验科用杀菌消毒液,其特征在于:制成所述消毒液有效成分的原料组成及重量份数为:
土茯苓71份 水松69份 小蜡树67份 蛇含64份
大蒜62份 水茴香62份 桉叶59份 八仙草59份
柽柳52份 醋52份 浮萍50份 槐叶47份
盐麸子47份 茶叶45份 枣叶40份 百两金29份。
4.根据权利要求1所述的一种检验科用杀菌消毒液,其特征在于:制成所述消毒液有效成分的原料组成及重量份数为:
土茯苓64份 水松62份 小蜡树60份 蛇含58份
大蒜56份 水茴香56份 桉叶53份 八仙草53份
柽柳46份 醋46份 浮萍44份 槐叶41份
盐麸子41份 茶叶39份 枣叶35份 百两金24份。
5.一种如权利要求1-4任一项所述的检验科用杀菌消毒液的制备方法,其特征在于:包括以下工艺步骤:
(1)取土茯苓、水松、盐麸子和茶叶洗净,将醋加入其中,拌匀,文火闷润至醋被吸尽时,置适宜的容器内密闭,用蒸气加热2-4小时,取出干燥,研末,得到过200目的细粉,备用;
(2)取小蜡树研末后,浸入体积浓度为90%酒精溶液,静置4小时,取出上层清液,沉淀下来的浸膏备用;
(3)取蛇含、大蒜、水茴香、八仙草、槐叶和百两金绞汁,滤杂质,留汁液,备用;
(4)取桉叶、柽柳、浮萍和枣叶放入容器中,加入10倍量蒸馏水浸泡4小时后,加热煮沸4小时,提取;第二次加入8倍量蒸馏水加热煮沸3小时,提取;第三次加入6倍量蒸馏水加热煮沸2小时,提取;合并三次所得提取液,滤过,得滤液备用;
(5)将步骤(1)所得的细粉、步骤(2)所得的浸膏、步骤(3)所得的汁液以及步骤(4)所得的滤液混合后,加入10-12倍量的蒸馏水,搅拌溶解均匀后,加热煮沸0.5小时,至60℃以下,静置1小时,过滤,得滤液,将滤液灌装封存,121℃热压灭菌20分钟,包装,即为所述的检验科用杀菌消毒液。
