Description

Method for drying and preserving chuanman grass seeds
Technical Field
The invention relates to a method for preserving seaweed seeds, in particular to a method for preserving the seeds of chuanman grass in a drying way.
Background
The Seagrass (seaweeds meadows) ecosystem and the mangrove forest and coral reef ecosystem are considered as three typical offshore marine ecosystems, and the international research result shows that the ecological service value of the Seagrass field is obviously higher than that of the mangrove forest and the coral reef. As used herein, the term "seaweed" refers to the only angiosperm submerged in shallow sea water, which has differentiated root, stem, leaf, flower and seed organs, and is distinct from "seaweed" (e.g., kelp, enteromorpha, etc.) which is a lower plant. The luxurious pasture of seaweeds, like forests on land, is an important sign of a healthy marine environment, being a precious "submarine grassland" or "submarine forest". The sea grass field is a high productivity area, can provide good habitat, seedling raising field and shelter for marine organisms such as fishes, shrimps, crabs and shellfish, and is also beneficial to the inhabitation of sea birds; while providing an important food source for many living beings. The seaweed farm is also one of the most effective carbon capture and sequestration systems on the earth, and the united nations organization considers that the most urgent action for coping with climate change is to maintain and recover the 'blue carbon' sink, and the seaweed, the mangrove forest and the salt marsh are taken priority.
The Sichuan radix Aconiti Kusnezoffii is also called ditch grass or wild duck grass. Belonging to the order Eisenales (Potamogetonales), the family Sichuan cranberries (Ruppiaceae), the genus Sichuan cranberry (Ruppia). The river vine is a submerged angiosperm (i.e. seaweed) in a salty water habitat which is widely distributed in estuaries, coasts and wetlands all over the world. In China, Chuanwan simpleles is distributed from south to Hainan province and from north to Liaoning province (Yu et al, 2014). However, like other seaweed species, the chuanwan grass also has rich ecological value, for example, the chuanwan grass is an important food resource for overwintering eating grass migratory birds and benthic invertebrates; the water quality and the biogeochemical cycle of sediments of the habitat of the estuary coastal zone can be obviously changed; and is also an important primary productivity in coastal ecosystems. In recent decades, due to interference of a plurality of thought factors, the vegetations of the chuanwan grass are greatly reduced, the population range is increasingly reduced, and the work of protecting the chuanwan grass is not smooth.
Since seaweed seeds generally contain higher moisture, and the seeds are easily inactivated after dehydration under the drying condition; therefore, the current common preservation methods of the seaweed seeds are wet preservation, namely, the seaweed seeds are preserved in a low-temperature and high-salinity seawater environment. Although the method can successfully inhibit the seed germination and keep the activity of the seeds, the seeds are easy to infect bacteria and fungi, so that the water needs to be changed regularly and at high frequency in the process of storing the seeds, and the time and labor are consumed. Further, an efficient and simple preservation method is urgently needed, which not only can get rid of the great damage to the seeds caused by the growth of bacteria and fungi in the wet preservation process of the chuanwan sprangletop seeds, but also greatly reduces the physical cost in the preservation process of the seeds.
Disclosure of Invention
The invention aims to provide a simple and convenient method for drying and preserving chuanman grass seeds.
In order to achieve the purpose, the invention adopts the technical scheme that:
a method for storing Sichuan vine grass seeds in a dry manner comprises completely rotting the epicarp of Sichuan vine grass seeds, washing the washed Sichuan vine grass seeds, directly placing the washed Sichuan vine grass seeds into a sealed container filled with a drying agent, and storing the Sichuan vine grass seeds at 5-10 ℃ in a dark place, so that the seeds can be stored under a long-term dry condition;
wherein, the environmental humidity is 33% in the process of light-proof preservation, and the deviation of the preservation humidity is +/-10%.
The chuanwan grass seeds are mature chuanwan grass seeds with completely rotten epicarp.
The water for washing the chuanman grass seeds is natural seawater.
The chuanman grass seeds are fully mature seeds, i.e. seeds with hard, black exocarp.
The culture conditions of the rotten process of the outer pericarp of the chuanman grass seed are room temperature, light-proof and 10-15 salinity, and water is changed periodically.
The humidity of the chuanman grass storage environment is regularly observed, and the drying agent in the storage environment is regularly replaced.
The drying agent is a drying agent commonly used in the field, such as allochroic silica gel and the like.
The invention has the beneficial effects that:
the invention relates to a method for preserving seeds of a chuanwan grass (R.sinensis), which is characterized in that the seeds of the chuanwan grass still keep high activity after slowly losing water under the dry preservation condition at a proper temperature, so that the chuanwan grass seeds are preserved under the dry preservation condition at the proper temperature, the method not only can get rid of the great damage of the seeds caused by the growth of bacteria and fungi in the wet preservation process of the chuanwan grass seeds, but also greatly reduces the physical cost in the preservation process of the seeds; the method specifically comprises the following steps:
the method of the invention is adopted to store the chuanwan grass seeds under the drying condition (33 +/-10%) with certain humidity, the storage temperature is 5-10 ℃ and the condition of keeping out of the sun, thereby reducing the labor cost in the storage process, promoting the seeds to keep the dormancy state without sprouting and improving the retention rate of the chuanwan grass seeds in the storage process; prolonging the preservation time of the seeds of the Sichuan vine; the occurrence frequency of the condition that the chuanman grass seeds are rotten by the growing bacteria in the preservation process is reduced; meanwhile, the higher activity and quality of the chuanman grass seeds can be kept. The method can provide high-quality chuanwan grass seeds for artificial sowing, is beneficial to the recovery and construction of a chuanwan grass sea grass field, and can quickly recover the good ecological environment of the sea.
In the preservation of seaweed seeds, the seeds are generally preserved under high salinity and low temperature conditions. However, in the process of storing the seaweed seeds, the water in the seed storage environment needs to be replaced regularly and at high frequency, so that the growth of fungi and bacteria in the storage environment is inhibited. In the water changing process of wet-method preservation of the chuanman grass seeds, the germinating petals of the seeds are easy to crack along with the sudden rise of the water body temperature, so that the seeds are easy to be infected by bacteria and fungi in the following preservation process and lose vigor. Based on the test, the invention discovers that the chuanwan grass seeds still keep high activity after being dehydrated under the dry preservation condition at the proper temperature (5-10 ℃), and accordingly, the chuanwan grass seeds without the epicarp are directly preserved under the proper temperature dry condition, the method not only can get rid of the great damage to the seeds caused by the growth of bacteria and fungi in the wet preservation process of the chuanwan grass seeds, but also greatly reduces the physical cost in the preservation process of the seeds, and the advantages are shown in the following aspects: firstly, the chuanman grass seeds are preserved under the dry condition, so that the requirement on preservation environment equipment is greatly reduced; secondly, the seeds are preserved in a lower humidity environment, so that the seeds are ensured to have higher activity, the germination of the seeds in the preservation process is inhibited, the preservation time of the chuanwan sprangletop seeds is prolonged, and the purpose of long-term preservation is achieved; finally, as fungi and bacteria can not grow under the dry condition, the seeds can not be rotten in the preservation process, so that the water changing frequency of the chuanman grass seeds in the preservation period is reduced, the labor cost is greatly reduced, the disturbance to the preserved seeds is reduced, and the seeds are prevented from sprouting. In addition, in the seed preservation process, the drying agent in the preservation environment needs to be replaced regularly, the situation that the chuanwan sprawl herbs germinate due to overhigh environment humidity is avoided, and the specific replacement frequency of the drying agent is determined according to the sealing performance of the preservation container.
Drawings
FIG. 1 is a flow chart of the steps of the method for preserving Chuanman grass seeds of the present invention.
Detailed Description
The present invention is described in further detail below with reference to the accompanying drawings.
The method not only ensures that the retention rate of the seeds of the chuanwan grass is high during the storage period, but also ensures that the chuanwan grass seeds still have higher germination activity after long-term storage, and greatly prolongs the storage time of the chuanwan grass seeds.
The preservation method of the invention is shown as the steps in figure 1:
putting mature Sichuan sprangletop (R.sinensis) seeds in a water body with salinity of 15-20 prepared by photophobia, oxygenation and natural seawater and tap water, and cleaning the seeds by using natural seawater after the epicarp of the seeds is completely decayed;
putting the chuanman grass seeds into a sealed plastic box filled with a drying agent, and separating the drying agent and the seeds by using tin foil paper; placing the hygrometers in a sealed plastic box, and then placing the sealed plastic box at 5-10 ℃ for storage in a dark place;
the humidity condition in the sealed box is observed regularly, and the drying agent is replaced regularly.
Example 1
In 2016, the seeds of the chuanman grass are collected in the east-camp coastal water area in 9 months. The culture conditions of the rotten process of the outer pericarp of the chuanman grass seed are room temperature, light-proof and 10-15 salinity, and the water is changed periodically. The seeds used for the test were fully mature seeds, i.e. seeds with a hard, black outer peel. Randomly selecting 2000 mature seeds of Sichuan sprangletop (R. sinensis), uniformly placing in an environment with humidity of 33 +/-10% and temperature conditions of 0 ℃, 5 ℃, 10 ℃, 15 ℃ and 22 ℃, and simultaneously adding a drying agent. The seed preservation time was 9 months. During the period, the change of the environmental humidity in the sealed box is checked every 3 months, and the drying agent is replaced for the processing group with overhigh humidity. And after the seeds are stored, the seed vitality is verified through a germination experiment. (germination experiment: randomly taking 200 seeds from each storage experiment group, dividing into 3 repeated treatment groups, and germinating in an environment with the environmental temperature of 30 ℃ and the salinity of 5).
Wherein, the drying agent is a color-changing silica gel drying agent.
The specific data of the seed germination rate in the identification of the seed retention rate and the seed vigor (germination experiment) related to the invention are obtained through experiments, wherein the specific embodiment of the identification experiment of the seed vigor is as follows: and (3) placing 50 stored chuanwan grass seeds (3 times of repetition) under the optimal germination condition of the chuanwan grass seeds for germination (the optimal germination condition is that the salinity of a water body is 5, the temperature is 30 ℃, and the photoperiod is 12L: 12D), standing and culturing for 10 days, and recording and calculating the germination rate of the seeds.
The method for calculating the seed germination rate in the seed activity test comprises the following steps:
Figure BDA0001417396540000041
the specific germination results are shown in table 1:
TABLE 1 average germination percentage (%) -of seeds in the seed viability test after 9 months of preservation of chuanman grass seeds under different temperature drying conditions
Figure BDA0001417396540000042
As can be seen from table 1, after the chuanwan grass seeds are stored for 9 months in a dry state, the average germination rate of the chuanwan grass seeds is the highest at 5 ℃ and 10 ℃, which is approximately 75%, and is higher than 0 ℃ (the germination rate of the seeds is 55.6%), and other temperature treatment groups. Therefore, the environment temperature is 5-10 ℃, the environment humidity is 33 +/-10%, and the dark environment is a proper drying and storing condition of the chuanman grass seeds.
The application of the invention can obviously improve the preservation time of the seeds and realize the establishment of the chuanwan grass seed bank with less labor cost on the premise of keeping higher activity of the chuanwan grass seeds.
The control of different temperatures in this embodiment adopts the low temperature incubator to keep invariable temperature, product model: KRC-150CA, manufacturer: shanghai Qixin scientific instruments, Inc.; other temperature and illumination demands adopt intelligent illumination incubator to keep invariable temperature and illumination, product model: MGC-250, manufacturer: shanghai, a constant technology, Inc.
