Technical Field
The invention relates to the field of traditional Chinese medicines, in particular to a traditional Chinese medicine composition for treating beriberi and foot odor, a preparation and a preparation method thereof.
Background
Xanthoceras sorbifolia Bunge (name: Xanthoceras sorbifolia Bunge), Sapindaceae, Xanthoceras genus deciduous shrub or small arbor, up to 5m in height; the small branch is brownish red and thick, and the length of the leaf and the stem can reach 30 cm; the two sides of the flower are asymmetrical, the top end is tapered, the base is wedge-shaped, sharp sawteeth are arranged on the edge, inflorescences of the amphoteric flowers are terminal, male inflorescences are axillary and upright, the total pedicel is short, the petals are white, the base is purple red or yellow, the angular appendages of the flower disc are orange yellow, and the filaments are hairless; the capsule length reaches 6 cm; the seeds were black and shiny. Shiny-leaved yellowhorn blossoms in spring and fruits in early autumn.
Xanthoceras sorbifolia is a unique rare woody oil plant in China, is an important woody oil tree species for greening, eating, medicine and manufacturing biofuel, and has been widely concerned by academia in recent years. The medicinal parts of the nutlet, the fruit shell, the carpopodium, the branches and leaves and the like are rich in active ingredients such as triterpenoid saponin, sterol, flavone, coumarin, tartaric acid and the like, and especially the content of the carpopodium and the fruit shell is high.
The triterpenoid saponin belongs to a nonionic surfactant, has a pH value of 6.0-7.0, has very low Critical Micelle Concentration (CMC) and surface tension (gamma CMC), has various biological activities such as good antibacterial, anti-inflammatory, antioxidant and antifungal activities, can be widely applied to the daily chemical industry, can be used as a natural detergent, has a good washing effect, does not contain substances such as phosphorus, aluminum, benzene and the like, does not hurt the human body, does not pollute the water source, is green, environment-friendly, is an ideal raw material for hand sanitizer, shampoo, shower gel, cosmetics and the like, and has great research, development and utilization potentials. The triterpene total saponin crude product has strong antibacterial activity on saccharomyces cerevisiae and candida utilis, and also has relatively strong inhibition effect on saccharomycetes, so that the triterpene total saponin crude product can help to improve skin problems such as whelk, black speck, wrinkle, dry skin, rough skin and the like.
Sterols, flavones, coumarins, and fruit acids can help the skin remove the aged cutin accumulated on the outer layer. Therefore, the xanthoceras sorbifolia bunge is deeply processed to extract the active ingredients such as triterpenoid saponin, sterol, flavone, coumarin, tartaric acid and the like, and has great industrial significance.
In conclusion, the shinyleaf yellowhorn extract has a lot of physiological activities, but the effect of treating certain diseases by adopting a single shinyleaf yellowhorn extract is not obvious and cannot be popularized to the market, for example, aiming at beriberi, the single shinyleaf yellowhorn only has a prevention effect and cannot cure the beriberi. Therefore, the research on the application of the pharmaceutical composition containing the xanthoceras sorbifolia to the treatment of dermatophytosis is a technical problem to be solved urgently.
In view of the above, the present invention is particularly proposed.
Disclosure of Invention
The first purpose of the invention is to provide a traditional Chinese medicine composition for treating beriberi and foot odor, and the two components of the traditional Chinese medicine composition have synergistic effect, thereby having obvious curative effect and higher cure rate on beriberi and foot odor.
The second purpose of the invention is to provide a preparation prepared from the traditional Chinese medicine composition, which is externally applied, convenient to use and free from toxic and side effects.
The third purpose of the invention is to provide a preparation method of the preparation, which has simple process and can effectively protect the activity of the drug effect component.
In order to achieve the above purpose, the invention provides the following technical scheme:

The sophora flavescens is cold in nature, has the effects of clearing heat, eliminating dampness and killing insects, can clear damp heat in lower jiao, kill insects and relieve itching, has a good relieving effect on skin itch, can balance grease secretion, dredge and astringe pores, clear away endotoxin impurities in skin, has rich herbal nutrition, promotes the growth and repair of damaged vascular nerve cells, recovers the activity of subcutaneous capillary cells, and has the effects of beautifying and protecting skin due to the fact that skin is compact and smooth. The matrine and oxymatrine in radix Sophorae Flavescentis have obvious effect of increasing leukocyte, and can be used for treating leukopenia caused by cyclophosphamide, X-ray and cobalt ray irradiation. Radix Sophorae Flavescentis also has antiinflammatory effect, and matrine has inhibitory effect on auricle swelling caused by croton oil in mice, abdominal cavity exudation increase caused by acetic acid in mice, and carrageenan foot pad swelling in rats.
In the invention, the shell and the carpopodium of the xanthoceras sorbifolia bunge are rich in triterpenoid saponin, and have emulsification cleaning effect and strong antibacterial activity; the shell and the carpopodium of the xanthoceras sorbifolia bunge are rich in sterol and flavone, and have the effect of removing aged cuticle, and the shell and the carpopodium of the xanthoceras sorbifolia bunge have the synergistic effect with the matrine in the sophora flavescens by utilizing the characteristics, so that the immunity of the organism can be improved, a plurality of bacteria causing beriberi and foot odor can be killed, the skin state of the place suffering from the beriberi can be improved, the symptoms such as swelling, blister, erosion and the like can be eliminated, and the metabolism of the corresponding skin can be. Therefore, in the composition provided by the invention, the two components have different emphasis points, but by means of the synergistic cooperation of the two components, adverse factors and exogenous harmful bacteria in a human skin metabolic system and an immune system are comprehensively eliminated from multiple dimensions, the immunity of the organism is improved, the bacteria can be killed, the skin metabolism is promoted, and the beriberi and the foot odor are finally and effectively cured.

10 to 60 percent of methanol or 10 to 60 percent of ethanol is used as a solvent, and the mixture is extracted for multiple times at 30 to 90 ℃ according to the material-liquid ratio of 1:8 to 1: 15.
The extract obtained by adopting the solvent extraction has better quality, contains rich active ingredients, not only contains water-soluble substances, but also contains alcohol-soluble substances, and the two ingredients can better improve the treatment effect on the beriberi and the foot odor through synergism.
Preferably, the leaching method comprises the following steps: firstly heating to 30-60 ℃, soaking for 30-60 min, and then heating to 80-90 ℃ for reflux extraction for 20-40 min.
The extraction process comprises two steps of dipping and refluxing, so that water-soluble and alcohol-soluble components are fully released, the materials can be stirred in the dipping process, the flow of the materials is accelerated, and the stirring speed is preferably 20-100 r/min.
Preferably, the method further comprises the following steps after leaching: decolorizing, concentrating, and drying.
Decolorizing, further removing impurities, concentrating, drying, and storing conveniently, and has abundant dosage forms.
Preferably, the decoloring method comprises the following steps: adding 0.1-0.5 times of activated carbon to the solution to be decolorized, and filtering after completing adsorption. Among them, the filtration is preferably carried out in a centrifuge at a rotation speed of preferably 3000 rpm.
Preferably, the method of concentration is: preparing a concentrated solution with the water content of 40-50 wt% under the vacuum degree of-0.01-0.092 MPa and the steam pressure of 0.1-0.5 MPa;
the drying method comprises the following steps: spray drying at the air inlet temperature of 150-220 ℃.
The cooperation of the concentration and drying steps is very important, and the process can not only obtain higher efficiency ratio, but also obtain better product quality.
If the concentrated solution is used as the extract to prepare the spray, 60-90 times of water is added for dilution, and then the spray is applied to feet.
As described above, the quality of the extract is very important for the drug efficacy, and the following qualities of the extract are preferable in the present invention:
the extract comprises saponin, flavone and sterol, and the mass ratio of the saponin to the flavone to the sterol is 15-17: 12-14: 1.6-2.
Wherein the mass ratio of the saponin to the flavone to the sterol can be 15:14:1.6, or 17:12:2, or 17:13:1.8, and the like.
The dosage form suitable for the invention is spray, solid soap or loose powder.
The powder is similar to the spray, and is only characterized in that the spray is prepared, namely the powder is used after being taken, and the powder needs to be prepared at present, namely a patient can dissolve the powder with water and then soak the powder.
The solid soap agent is a multifunctional product, can be used for cleaning feet and treating beriberi and foot odor, has long storage time, and obviously has wider application market.
The solid soap preparation needs to be added with necessary soap base, and auxiliary materials such as essence and/or toner can be added on the basis.
The content of the extract in the soap agent is not too high or too low, preferably 0.1 wt% -10 wt%.
The soap can be prepared by the following steps:
heating and melting the soap base, adding the extract and the rest components, and then carrying out vacuum drying under-0.01 to-0.05 Mpa, grinding and extruding to obtain the soap base.
In summary, compared with the prior art, the invention achieves the following technical effects:
(1) the synergistic effect among the xanthoceras sorbifolia shells, the carpopodium and the sophora flavescens is utilized, so that the treatment effect on beriberi and foot odor is improved, and the treatment period is shortened and the cure rate is improved;
(2) the quality of the extract is improved by improving the extraction process of the composition, so that the treatment effect is further improved;
(3) provides a spray and a soap which are more convenient to use and simpler to prepare, and provides a prerequisite for the wide application of the composition.
Detailed Description
The technical solutions of the present invention will be described clearly and completely with reference to the following detailed description, but those skilled in the art will understand that the following described examples are some, not all, of the examples of the present invention, and are only used for illustrating the present invention, and should not be construed as limiting the scope of the present invention. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention. The examples, in which specific conditions are not specified, were conducted under conventional conditions or conditions recommended by the manufacturer. The reagents or instruments used are not indicated by the manufacturer, and are all conventional products available commercially.
Example 1
Traditional Chinese medicine extract
1. Taking materials: according to the weight percentage, 80 percent of shinyleaf yellowhorn shell and 20 percent of lightyellow sophora root are evenly mixed and crushed into particles with the particle size less than or equal to 5 mm.
2. Putting the raw materials into an extraction tank, adding 60% methanol according to the ratio of material to liquid of 1:15, heating to 30 ℃, extracting for 30 minutes under the condition that the rotating speed of a stirrer is 20 revolutions per minute, heating to 80 ℃, keeping for 40 minutes, and refluxing a methanol solvent; repeating the above operations, and extracting twice.
3. Mixing the two extractive solutions, adding active carbon at a ratio of 10:5 for decolorizing, performing solid-liquid separation in a centrifuge at 3000 rpm, and recovering filtrate.
4. Making the filtrate into concentrated solution with water content of 50% under vacuum degree of-0.092 MPa and steam pressure of 0.5 MPa.
5. Centrifuging the concentrated solution at 150 deg.C in air, and spray drying to obtain powder extract.
As a result:
the mass ratio of saponin, flavone and sterol in the obtained powdery extract is 15.02:14: 1.6.
The extract can be directly used for foot bath, and the specific method comprises:
adding 2 g of the extract into 2L of warm water at 38-45 ℃, soaking feet in the water for 5-10 minutes, and wiping off water on the feet. If the water is used for washing insoles and socks, the effect of removing beriberi and foot odor is better.
Example 2
The only difference from example 1 is that the 60% methanol used in the extraction of step 2 is replaced by water.
As a result:
in the obtained powdery extract, the mass ratio of saponin to flavone to sterol is 12:12: 1.7.
Example 3
The difference from example 1 is that 60% methanol used in the extraction in step 2 is replaced by 10% methanol, and the specific experimental process is as follows:
1. taking materials: according to the weight percentage, 80 percent of shinyleaf yellowhorn shell and 20 percent of lightyellow sophora root are evenly mixed and crushed into particles with the particle size less than or equal to 5 mm.
2. Putting the raw materials into an extraction tank, adding 10% methanol according to the ratio of material to liquid of 1:8, heating to 60 ℃, leaching for 60 minutes under the condition that the rotation speed of a stirrer is 100 revolutions per minute, heating to 90 ℃, keeping for 20 minutes, and refluxing the methanol; repeating the above operations, and extracting twice.
3. Mixing the two extractive solutions, adding active carbon at a ratio of 10:1 for decolorizing, performing solid-liquid separation in a centrifuge at 3000 rpm, and recovering filtrate.
4. Making the filtrate into concentrated solution with water content of 40% under vacuum degree of-0.01 MPa and steam pressure of 0.5 MPa.
5. Centrifuging the concentrated solution at 220 deg.C in air, and spray drying to obtain powder extract.
As a result:
in the obtained powdery extract, the mass ratio of saponin to flavone to sterol is 17:12: 2.
Example 4
The difference from example 1 is that 60% methanol used in the extraction in step 2 is replaced by 10% ethanol, and the specific experimental process is as follows:
1. taking materials: according to the weight percentage, the shiny-leaved yellowhorn shells and the stems account for 80 percent, and the lightyellow sophora root accounts for 20 percent, are evenly mixed and crushed into particles with the diameter less than or equal to 5 mm.
2. Putting the raw materials into an extraction tank, adding 10% ethanol solvent according to the ratio of material to liquid of 1:10, heating to 60 ℃, extracting for 80 minutes under the condition that the rotating speed of a stirrer is 40 revolutions per minute, heating to 90 ℃, keeping for 20 minutes, and refluxing the ethanol solvent; repeating the above operations, and extracting twice.
3. Mixing the two extractive solutions, adding active carbon at a ratio of 10:5 for decolorizing, performing solid-liquid separation in a centrifuge at 3000 rpm, and recovering filtrate.
4. Making the filtrate into concentrated solution with water content of 40% under vacuum degree of-0.01 MPa and steam pressure of 0.5 MPa.
5. Centrifuging the concentrated solution at 150 deg.C in air, and spray drying to obtain powder extract.
Example 5
The difference from example 1 is that 60% methanol used in the extraction in step 2 is replaced by 60% ethanol, and the specific experimental process is as follows:
1. taking materials: according to the weight percentage, the shinyleaf yellowhorn shell accounts for 90 percent, the lightyellow sophora root accounts for 10 percent, the mixture is evenly mixed, and the mixture is crushed into particles with the diameter less than or equal to 5 mm.
2. Putting the raw materials into an extraction tank, adding 60% ethanol solvent according to the ratio of material to liquid of 1:8, heating to 60 ℃, extracting for 90 minutes under the condition that the rotating speed of a stirrer is 60 revolutions per minute, heating to 80 ℃, keeping for 40 minutes, and refluxing the methanol or ethanol solvent; repeating the above operations, and extracting twice.
3. Mixing the two extractive solutions, adding active carbon at a ratio of 10:5 for decolorizing, performing solid-liquid separation in a centrifuge at 3000 rpm, and recovering filtrate.
4. The filtrate is made into concentrated solution with the water content of 40 percent to 50 percent under the vacuum degree of-0.092 MPa and the steam pressure of 0.1.
5. Centrifuging the concentrated solution at 150 deg.C in air, and spray drying to obtain powder extract.
Example 6
The difference from the embodiment 1 is only that the mixture ratio of the xanthoceras sorbifolia shells to the sophora flavescens is 92: 8.

The difference from the embodiment 1 is only that the mixture ratio of the xanthoceras sorbifolia shells to the sophora flavescens is 85: 15.
examples 8 to 14
The spray for treating beriberi and foot odor comprises the following components:
deionized water was added to the powdery extracts obtained in examples 1 to 7 in an amount of 90% by weight, and the mixture was filled in an extrusion type spray bottle after being mixed uniformly to obtain a spray having a function of removing beriberi and foot odor, and the spray was applied to the area between the toes, the upper surface of the foot, and the sole of the foot to achieve the effect of removing beriberi and foot odor.
Example 15
The soap for treating beriberi and foot odor comprises the following components:
adding 0.1 percent of the extract obtained in the example 1, the essence for the soap and the color-mixing raw material for the soap into a proportioning cylinder added with the soap base, keeping the temperature at 60-80 ℃ under the condition that the rotating speed of a stirrer is 40 revolutions per minute; stirring for 60min, vacuum drying at-0.05 Mpa, grinding with three-roller mill, extruding into strip machine, printing, and packaging; the soap with beriberi and foot odor removing function can be prepared.
Comparative example 1
The only difference from example 8 is that all the lightyellow sophora root was replaced with xanthoceras sorbifolia husk.
Comparative example 2
The only difference from example 8 is that all of the xanthoceras sorbifolia shells were replaced with sophora flavescens.

Comparative example 4
The difference from example 1 is only that the weight ratio of the xanthoceras sorbifolia shells to the sophora flavescens is 75: 25.
Experiment of antifungal Effect
Strain: trichophyton rubrum, Trichophyton gypseum, Epidermophyton floccosum, Microsporum canis, and Candida albicans, wherein the culture medium is modified Sabouraud's medium.
The method comprises the following steps: after each strain is subcultured, 0.2mL of the strain is diluted by sterilized normal saline and then is uniformly inoculated on a Shashi plate culture medium; the drugs to be tested are put on the flat plate in an equal weight mode; and (3) placing the flat plate in a thermostat at 28 ℃ for culturing, and after the negative control is full of fungi around, starting to measure the diameter of the bacteriostatic ring and judge the bacteriostatic effect.
The results are shown in Table 1.

The results show that the invention has obvious bacteriostatic effect on fungi causing beriberi and foot odor, and is obviously higher than that of comparative examples, especially a single component.
Example of clinical experience
1. For men, 20 years old, they prefer to wear shoes with higher sealing performance such as sneakers and sneakers, but have severe foot odor. 2 g of the extract powder (example 1) is added into 2L of warm water, feet are soaked for 5-10 minutes, the odor is basically eliminated after three days, and the odor is completely eliminated after seven days. The foot odor is not caused after the foot odor is used once a week till now for half a year.
2. For a male, the male is 83 years old, beriberi is suffered for years, white skin appears among toes, and the whole foot is difficult to endure due to alternate weather changes every season. The spray of the extract in the embodiment 8 is used for spraying and coating the extract among toes, on the upper surface of feet and on the sole of feet, after the spray is continuously used for five days, the beriberi symptoms are obviously relieved, and after ten days, the white skin among the toes disappears; the keratinized old skin of the sole of the foot basically disappears; after continuous use for one month, the medicine is used once a week instead. The itching of the whole foot does not occur after the change of seasons of autumn, winter and spring.
In addition, the invention also counts the cure rate of the foot odor of the examples 8 to 15, which is 100 percent.
