本发明公开一种饲用蛋白酶制剂保存载体及其制备方法与应用,属于饲用酶制剂领域。该保存载体包含以下按质量百分比计的组分:滑石粉40%~65%,膨润土20%~30%,二氧化硅3%~10%,玉米芯粉8.9%~15%,β‑环糊精1%~4.7%,无水柠檬酸0.1%~0.3%。能够很好的吸附及释放蛋白酶,且产品经过重压及长期存放不会分层结块,分散均匀,活性持久;在保存液体深层发酵经喷雾干燥的饲用蛋白酶制剂时,具有很好的分散性,对蛋白酶具有充分的保护作用。同时也提高了酶的释放,易于检出且结果准确,降低了检测标准误差。保证了蛋白酶在饲料生产中长期稳定的应用。该方法工艺流程简单,操作方便,易于实施。

本发明属于饲用酶制剂领域,具体涉及一种饲用蛋白酶制剂保存载体及其制备方法与应用。
背景技术
随着科学技术的不断发展,饲用蛋白酶制剂得到了迅速普及,因其能够节约蛋白的作用而被重视,尤其是近年来畜牧行业饲料资源紧张、成本上涨,食品安全问题频发,环境污染日益严重,酶制剂作为一种绿色、安全、无污染的添加剂呈现出越来越明显的价值与作用。蛋白酶作为消化酶主要以复合酶的形式在饲料中使用。然而,饲用蛋白酶作用效果与其活力保存密切相关。
蛋白酶广泛存在于动物、植物和微生物中,饲料工业上应用的蛋白酶目前主要来源于微生物。通过微生物发酵方式获得,目前饲用蛋白酶的生产多采用液体深层发酵方法,经过喷雾干燥技术,制成高浓度酶粉,此发酵方法生产的蛋白酶具有纯度高、污染小、活性强等优点。但是在储存运输过程中容易吸潮结块,酶活力下降速度快,给生产和应用带来了难题,因此急需开发一种能够防结块、酶活保存稳定的饲用蛋白酶保存载体,为饲料生产及畜牧业带来益处。
发明内容
为了克服现有技术的缺点与不足,本发明的首要目的在于提供一种饲用蛋白酶制剂保存载体。该保存载体适用于液体深层发酵喷雾干燥。
本发明的另一目的在于提供上述饲用蛋白酶制剂保存载体的制备方法。该方法工艺流程简单,操作方便,易于实施。
本发明的再一目的在于提供上述饲用蛋白酶制剂保存载体的应用。
本发明的目的通过下述技术方案实现:
一种饲用蛋白酶制剂保存载体,包含以下按质量百分比计的组分:滑石粉40%~65%,膨润土20%~30%,二氧化硅3%~10%,玉米芯粉8.9%~15%,β-环糊精1%~4.7%,无水柠檬酸0.1%~0.3%。

一种饲用蛋白酶制剂保存载体,优选包含以下按质量百分比计的组分:
滑石粉60%,膨润土20%,二氧化硅9%,玉米芯粉8.9%,β-环糊精2%,无水柠檬酸0.1%;
优选的,所述的滑石粉、膨润土、二氧化硅、玉米芯粉、β-环糊精的水分质量含量优选均在10%以下;
更优选的,所述的滑石粉、膨润土、二氧化硅、玉米芯粉、β-环糊精的水分质量含量为:滑石粉3%,膨润土8%,二氧化硅5%,玉米芯粉8%,β-环糊精5%。
所述的饲用蛋白酶制剂保存载体的制备方法,包括以下具体步骤:
(1)将滑石粉、膨润土、二氧化硅、玉米芯粉、β-环糊精和无水柠檬酸分别粉碎,过筛;
(2)按上述比例称取步骤(1)中过筛后的滑石粉、膨润土、二氧化硅、玉米芯粉、β-环糊精和无水柠檬酸,按其添加量从低到高逐级混合,搅拌均匀,制成饲用蛋白酶制剂保存载体。
步骤(1)中所述的粉碎优选是用T130X加强超细磨粉机进行粉碎;
步骤(1)中所述的过筛优选为使用200目筛过筛;
步骤(2)中所述的搅拌均匀优选是用立式KX-500型电动计量干粉搅拌机,在450~500r/min下搅拌10~20min。
所述的饲用蛋白酶制剂保存载体在保存饲用蛋白酶制剂中的应用,优选包含如下步骤:将饲用蛋白酶制剂与保存载体按质量比1:(1~9)复配,制备成蛋白酶活力为10000~150000U/g的饲用蛋白酶。
所述的饲用蛋白酶制剂优选为液体深层发酵经喷雾干燥后的饲用蛋白酶制剂;
所述的饲用蛋白酶制剂优选为中性蛋白酶、酸性蛋白酶和碱性蛋白酶中的至少一种;
所述的饲用蛋白酶制剂的酶活力为100000~300000U/g。
所述的保存载体的复配量要与所要保存的蛋白酶活力比重相当,且保存载体与蛋白酶制剂的配比根据实际生产需要配备。
本发明相对于现有技术,具有如下的优点及效果:
(1)本发明的保存载体原料滑石粉、膨润土、二氧化硅均为无机原料,且含有许多动物生长所需的常量元素和微量元素,均具有较好的吸附和分散能力。其中膨润土具有吸附性、膨胀性、分散性和润滑性,能提高饲料的适口性和改进饲料的松散性,并能延缓饲料通过动物消化道的时间,加强饲料在消化道内的消化吸收作用,并对消化道内的重金属有害气体细菌等有强烈的吸附作用,从而起到对动物的保健作用,由于膨润土是碱性物质,在动物的消化道内能够中和消化道内的酸起到缓冲作用,可增强动物的食欲;
所述保存载体原料β-环糊精可以掩盖或驱除发酵后的蛋白酶产品中的不良异味,提高产品适口性;
所述保存载体原料玉米芯粉具有很好的吸附性和较强的化学稳定性,是优良的有机载体。

下面结合实施例对本发明作进一步详细的描述,但本发明的实施方式不限于此。
实施例1
一种饲用蛋白酶制剂保存载体,各成分所占的质量百分比分别为:滑石粉60%,膨润土20%,二氧化硅9%,玉米芯粉8.9%,β-环糊精2%,无水柠檬酸0.1%。各成分其水分含量分别为:滑石粉3%,膨润土8%,二氧化硅5%,玉米芯粉8%,β-环糊精5%。
所述的饲用蛋白酶制剂保存载体的制备方法,包括如下步骤:
(1)将滑石粉、膨润土、二氧化硅、玉米芯粉、β-环糊精和无水柠檬酸分别粉碎,过200目筛,使其细度达到200目;
(2)过200目筛后,按上述配方分别称取各组分,各组分按其添加量从低到高逐级混合,用立式KX-500型电动计量干粉搅拌机,在500r/min下搅拌10min,搅拌均匀,制成饲用蛋白酶制剂保存载体。

(1)将滑石粉、膨润土、二氧化硅、玉米芯粉、β-环糊精和无水柠檬酸分别粉碎,过200目筛,使其细度达到200目;

所述的饲用蛋白酶制剂保存载体的制备方法,包括如下步骤:
(1)将滑石粉、膨润土、二氧化硅、玉米芯粉、β-环糊精和无水柠檬酸分别粉碎,过200目筛,使其细度达到200目;
(2)过200目筛后,按上述配方分别称取各组分,各组分按其添加量从低到高逐级混合,用立式KX-500型电动计量干粉搅拌机,在450r/min下搅拌20min,搅拌均匀,制成饲用蛋白酶制剂保存载体。
效果实施例
根据本发明饲用蛋白酶制剂保存载体及制备方法,将此保存载体分别应用于液体深层发酵经喷雾干燥后的酸性蛋白酶、中性蛋白酶及碱性蛋白酶制剂(以上蛋白酶制剂购买自云南博仕奥生物技术有限公司)。根据生产需要将酶活力为100000~300000U/g的饲用蛋白酶制剂与保存载体分别按质量比1:(1~9)复配,制备成酶活力为10000~150000U/g的饲用蛋白酶制剂。定期取样观察并测定酶活力。试验结果表明,使用本发明保存载体的蛋白酶制剂适口性好,无发酵异味,分散性强,不吸潮,不结块,保质期长,活性稳定,室温保存12个月各蛋白酶相对剩余酶活力仍在93%以上。如表1,2,3所示。

将饲用蛋白酶制剂与权利要求1~4任一项所述的饲用蛋白酶制剂保存载体按质量比1:(1~9)复配,制备成蛋白酶活力为10000~150000U/g的饲用蛋白酶。
10.根据权利要求9所述的应用,其特征在于:
所述的饲用蛋白酶制剂为中性蛋白酶、酸性蛋白酶和碱性蛋白酶中的至少一种;
所述的饲用蛋白酶制剂的酶活力为100000~300000U/g。
