技术领域

发明背景
经由B7:CD28共刺激分子家族的信号强烈地参与T细胞反应的活化、抑制和调节。已知属于B7:CD28共刺激分子家族的PD-1(程序性死亡1)分子与PD-L1和PD-L2反应,并负调控T细胞反应(非专利文献1)。首先,据认为在T细胞上表达的PD-1与在抗原呈递细胞或癌细胞上表达的PD-L1,或者在抗原呈递细胞上的PD-L2反应以抑制T细胞的活化。

优选实施方案详述
本文中,“T细胞的经增强的功能”意为改良所述T细胞的效应子功能。所述T细胞的经增强的功能(其不限制本发明),包括在所述T细胞的增殖速率上的提高、在细胞因子产量上的增加、或细胞毒性的改良。此外,所述T细胞的经增强的功能包括在受抑制状态例如无反应性(无应答性)状态或休眠状态下所述T细胞的耐受性的消除和抑制,更确切地说,所述T细胞从所述受抑制状态转变到所述T细胞对来自外界的刺激作出反应的状态。
本发明中,“无反应性”包括免疫细胞对刺激(例如,通过活化受体或细胞因子的刺激)的无应答性。无反应性可因例如暴露于免疫抑制剂或暴露于高剂量的抗原而发生。这种无反应性通常是抗原特异性的,和甚至在完成暴露于所耐受的抗原之后仍持续。例如,T细胞中的无反应性的特点在于不产生细胞因子(例如,白细胞介素(IL)-2)。T细胞的无反应性发生在当T细胞暴露于抗原时缺乏第二个信号(共刺激信号)的情况下接收到第一个信号(经由TCR或CD-3的信号)的时候。

(1)本发明的用于增强T细胞功能的方法和具有经增强的功能的T细胞。
本发明用于增强T细胞功能的方法是包括抑制所述T细胞的PD-L1和/或PD-L2表达的步骤的方法。另外,本发明的具有经增强的功能的T细胞是通过本发明用于增强T细胞功能的方法获得的细胞。本发明的增强T细胞功能的方法有可能改善T细胞的效应子功能,包括T细胞增殖速率的提高,细胞因子产量的增加或细胞毒性的改良。此外,消除或抑制T细胞的受抑制状态例如无反应性状态或休眠状态,改善T细胞对来自外界的刺激的敏感性。本发明如此获得的具有经增强的功能的T细胞可用于治疗或预防癌症、传染病或自身免疫性疾病中。
在本发明的方法中,可离体或体内实施抑制T细胞的PD-L1和/或PD-L2表达的步骤。
其中离体实施所述步骤的本发明方法包括下述步骤:(a)制备T细胞或包含T细胞的细胞群体,和(b)抑制步骤(a)中获得的T细胞的PD-L1和/或PD-L2表达,以制备具有经增强的功能的T细胞。所述方法可应用于包括过继免疫疗法在内的细胞免疫疗法,并且是有用的。步骤(a)可为从活体中分离T细胞或包含T细胞的细胞群体的步骤,或制备已确立的T细胞或包含T细胞的细胞群体的步骤。此外,本发明的方法可包括从细胞群体中将包含T细胞的细胞群体分离出以制备细胞亚群的步骤和/或培养和刺激所述细胞群体的步骤。这些步骤可在步骤(a)和步骤(b)之前、之后的任意阶段或与步骤(a)和步骤(b)同时进行。在其中离体进行抑制T细胞的PD-L1和/或PD-L2表达的步骤的本发明方法中,可通过适当的方式(例如,细胞亚群的制备,或所述细胞群体的培养或刺激)调控所述方法所适用的细胞的量和种类。因此,可减少有害事件(例如自身免疫,其为当将抑制PD-L1和/或PD-L2表达的物质或抑制信号传送的物质施用于活体时所关注的),因此它是非常有用的。
可实施将细胞群体与包含T细胞的细胞亚群分离的步骤,例如,通过经由密度梯度离心的单核细胞部分的分级,或通过将T细胞的表面标记用作指标的分离方法。所述表面标记的实例包括CD3、CD8和CD4,并且根据这些表面标记的分离方法在本领域中已知。例如,可如下实施所述步骤:通过将抗CD8抗体已固定在其上的载体(例如珠粒或培养容器)与包含T细胞的细胞群体混合,然后回收与所述载体结合的CD8阳性T细胞。对于抗CD8抗体固定于其上的珠粒,例如可适宜地使用CD8MicroBeads(由Miltenyi Biotec制造)、DynabeadsM450CD8(由Invitrogen制造),和Eligix抗CD8mAb包被的镍颗粒(由Biotransplant制造)。这也与将CD4用作指标的实现相同,例如可适宜地使用CD4MicroBeads(由Miltenyi Biotec制造)、Dynabeads M-450CD4(由Invitrogen制造)。
可通过选择适当的已知培养条件(这取决于细胞群体)来实施培养细胞群体和包含T细胞的细胞亚群的步骤。另外,在刺激细胞群体的步骤中,可将已知的蛋白质和化学成分等加入培养基以进行培养。例如,可将细胞因子、趋化因子或其他成分加入培养基。本文中,对所述细胞因子没有特定限制,只要它可对T细胞起作用即可,它的实例包括IL-2、IFN-γ、转化生长因子(TGF)-β、IL-15、IL-7、IFN-α、IL-12、CD40L、和IL-27。从增强细胞免疫的角度出发,特别适宜使用IL-2、IFN-γ或IL-12,而从改善体内转移的T细胞存活的角度出发,适宜地使用IL-7、IL-15或IL-21。另外,对所述趋化因子没有特定限制,只要它对T细胞起作用和显示迁移活性即可,其实例包括RANTES、CCL21、MIP1α、MIP1β、CCL19、CXCL12、IP-10和MIG。可通过针对存在于T细胞表面上的分子例如,CD3、CD28或CD44的配体和/或针对所述分子的抗体的存在来实施对细胞群体的刺激。此外,可通过在细胞表面与其它淋巴细胞(例如呈递靶肽(例如得自癌抗原的肽)的抗原呈递细胞(树突状细胞))接触来刺激细胞群体。
当在体内实施抑制T细胞的PD-L1和/或PD-L2表达的步骤时,所述步骤通过向活体施用后述的特异地抑制其表达的方法来实施。因此,可使用合适的药物递送系统(例如脂质体、微粒、微囊体等)。此外,例如,利用将T细胞的表面标记用作指标的递送系统,可特异地抑制T细胞的PD-L1和/或PD-L2表达。
抑制T细胞的PD-L1和/或PD-L2表达的方法的实例包括使用核酸例如针对PD-L1和/或PD-L2的siRNA、反义核苷酸、和核酶。
本发明中使用siRNA是为了选择性地抑制PD-L1和/或PD-L2的表达的目的,并基于利用双链RNA分子的RNA干扰(RNAi),所述双链RNA分子具有与转录自编码PD-L1和/或PD-L2的基因的mRNA的核苷酸序列同源的序列和与所述核苷酸序列互补的序列。本文中“具有与转录自编码PD-L1和/或PD-L2的基因的mRNA的核苷酸序列同源的序列和与所述核苷酸序列互补的序列”不仅指每个序列均与所述mRNA的核苷酸序列同源或互补的事实,而且还包括每个序列在使得发挥所需功能的这样的范围内大致同源或互补的事实。所述双链RNA分子被称作siRNA(短干扰RNA)。所述siRNA可为与转录自编码PD-L1和/或PD-L2的基因的mRNA的一个区域同源/互补的siRNA中的一种,或可为包含多个与不同区域同源/互补的RNA分子的siRNA。
本发明中使用的siRNA的链长的实例从在哺乳动物细胞中抑制干扰素反应的角度出发包括这样的链长,其中组成所述siRNA的双链体的其中一链具有13-29个碱基对的链长,优选15-25个碱基对的链长,进一步优选20-25个碱基对的链长。另外,所述链长的所有核苷酸序列可得自PD-L1和/或PD-L2的mRNA的核苷酸序列,或其部分可得自所述核苷酸序列。此外,从在哺乳动物细胞中RNA干扰的有效性的角度出发,本发明所用的siRNA可为例如具有2-4个核苷酸突出于3’-末端侧的单链区的双链RNA形状,进一步优选具有2个核苷酸突出于3’-末端侧的单链区的双链RNA形状。作为突出的单链区,例示连续2-4个核苷酸的脱氧胸苷残基(TT、TTT或TTTT)。
本发明所用的siRNA主要由核糖核苷酸构成,其部分可包含不同于核糖核苷酸的核苷酸,例如,脱氧核糖核苷酸、脱氧核糖核苷酸的衍生物、核糖核苷酸的衍生物,等等。可通过已知的化学合成方法合成siRNA,但不特定限制所述方法。可使用适当的模板核酸酶促地(例如,使用RNA聚合酶)制备siRNA。本发明中所用的siRNA可呈可在分子中形成双链体的单链RNA形式,例示具有的茎-环结构(短发夹结构:sh结构)的单链RNA,所述茎环结构具有siRNA部分作为茎和任意序列作为环。作为所述任意序列,例示1-30个核苷酸的序列,优选使用1-25个核苷酸的序列,进一步优选使用5-22个核苷酸。
可基于需要抑制其表达的基因序列适宜地设计siRNA序列。已报道许多siRNA设计算法(参见,例如,WO 2004/0455543、和WO 2004/048566),也可使用市售的软件。另外,有许多根据需要抑制其表达的基因序列的信息设计siRNA、合成并提供所述siRNA的公司。因此,本领域技术人员可基于需要抑制其表达的基因序列容易地获得siRNA。作为本发明中使用的siRNA,可使用任一种,只要它是选择性地抑制PD-L1和/或PD-L2的表达的siRNA即可。例如,在非具体限制的情况下,可将包含核苷酸序列SEQ ID NO:1或SEQ ID NO:9的siRNA用于PD-L1,可将包含核苷酸序列SEQ ID NO:2或SEQ ID NO:11的siRNA用于PD-L2。
在本发明中,可将反义核苷酸用于抑制PD-L1和/或PD-L2的表达。将所述反义核苷酸用于抑制蛋白质的表达,例如,通过直接干扰PD-L1和/或PD-L2的mRNA分子的翻译,通过RNA降解酶H降解mRNA,通过干扰mRNA的5’加帽,通过掩蔽5’帽,通过防止翻译因子与mRNA结合,或通过抑制mRNA的多腺苷化。通过反义核苷酸和PD-L1和/或PD-L2的mRNA之间的杂交抑制蛋白质表达。为了降低作为反义核苷酸的靶标的mRNA的稳定性或降解所述mRNA,选择在所述mRNA上的特定靶定位点。当鉴定出一个或多个靶位点时,设计出具有与所述靶位点充分互补(更确切地说,其在生理条件下充分杂交并具有足够的特异性)的核苷酸序列的核苷酸。
作为本发明的方法中使用的反义核苷酸,例如,例示具有链长为8-100个核苷酸,优选10-80个核苷酸,更优选14-35个核苷酸的核苷酸。
在本发明中,可将核酸(例如用于抑制PD-L1和/或PD-L2的表达的siRNA和反义核苷酸)直接导入细胞,或可将经设计使得在细胞中转录所述siRNA或所述反义核苷酸的核酸构建体导入细胞。当直接导入细胞时,可适宜地使用用于导入核酸的试剂例如TransIT-TKO(由Mirus制造)和人T细胞Nucleofector Kit(Amaxa)。另一方面,当使用其中转录RNA分子的核酸构建体时,可使用功能性连接于启动子下游的核酸构建体,所述启动子在T细胞中在可进行转录编码siRNA或反义核苷酸的核酸的情况下能够发挥功能,但该核酸构建体不具体限制本发明。另外,为了实现基因的有效转录,与启动子或转录起始位点协作的其它调控序列(例如,增强子序列或终止子序列)可存在于所述核酸构建体中。另外,为了通过同源重组将待导入T细胞染色体的对象插入的目的,例如可将基因安排在包含核苷酸序列的侧翼序列之间,每个所述核苷酸序列与染色体中基因的所需靶插入位点的两侧核苷酸序列具有同源性。
在所述核酸构建体中所用的启动子不受具体限制,只要它可在哺乳动物细胞中起作用即可,其实例包括RNA聚合酶II启动子、RNA聚合酶III启动子和人工可调控的启动子。RNA聚合酶II启动子的实例包括CMV启动子。另外,基于RNA聚合酶III的启动子的实例包括tRNA启动子、U6snRNA启动子和组蛋白H1启动子。作为可用四环素人工调控的启动子,例示可用四环素调控的启动子,其实例包括四环素调控的U6启动子和TR启动子。另外,所述启动子和Cre-loxP系统的组合使得有可能更严格地控制转录。
可构建用于转录siRNA的核酸构建体以使可抑制目标基因功能的双链RNA的有义链和反义链通过下述系统转录:(A)串联型,其中编码有义RNA的核酸和编码反义RNA的核酸分别连接在两个不同的启动子的下游,并将这两个转录单位安排在正向方向,分别转录所述有义RNA和所述反义RNA,(B)以下类型,其中将编码有义RNA的核酸和编码反义RNA的核酸呈正向方向分别安排在一个启动子的下游,并转录具有茎-环型(或短发夹型)的RNA,其中直接连接或用环连接所述有义RNA和所述反义RNA,或(C)反向型,其中将启动子分别安排在编码有义链和反义链的核酸(分别具有两条链)的两端侧,并用不同的启动子转录两条RNA链。本发明中,可根据使用条件(例如,细胞种类、有义序列或反义序列的种类等)使用串联型、茎-环型或反向型。
可将本发明使用的核酸构建体掺入适当的载体(例如,质粒载体或病毒载体)以在细胞中更稳定地发挥效果。此外,可将本发明的核酸构建体掺入细胞的染色体DNA上。所述质粒载体的实例包括但不具体限于,piGENE tRNA质粒(商标名,由iGENE制造)、siLentGene(由Promega制造)和pSEC Hygro载体(由Ambion制造)。为了将所述质粒载体导入细胞中,可使用利用载体(例如脂质体或配体-聚赖氨酸)的方法、磷酸钙法、电穿孔法、基因枪法,等等。
不具体限制病毒载体,通常使用在基因导入方法中使用的已知病毒载体,例如逆转录病毒载体(包括慢病毒载体、假型载体)、腺病毒载体、腺伴随病毒载体、猿猴病毒载体、痘苗病毒载体或仙台病毒载体。特别优选地,使用逆转录病毒载体、腺病毒载体或慢病毒载体。作为病毒载体,优选其中复制能力缺陷以致其不能在被感染细胞中自主复制的病毒载体。另外,在基因导入时,也可使用提高基因导入效率的物质例如retronectin(注册商标,由TAKARA BIO INC.制造)。市售腺病毒载体的实例包括Knockout Adenoviral RNAi系统(由Clonetech制造),市售逆转录病毒载体的实例分别包括pSINsi载体(由TAKARA BIOINC.制造)和pSIREN-RetroQ载体(由Clonetech制造)。在病毒载体的情况下,可利用病毒感染细胞的能力将其导入目标细胞。
本发明的方法中,在T细胞上实施抑制PD-L1和/或PD-L2表达的方法。更确切地说,在步骤(b)中,例如,可使用辅助性T细胞、抑制性T细胞、调控T细胞、细胞毒性T细胞(CTL)、幼稚T细胞、记忆T细胞、表达TCRα和β链的αβT细胞或表达TCRγ和δ链的γδT细胞,或包括这些细胞的细胞群体、血液(外周血、脐带血等)或骨髓液。这些T细胞可得自哺乳动物,或可得自人或非人哺乳动物。
可通过定量测定PD-L1和/或PD-L2蛋白,或定量测定转录自PD-L1和/或PD-L2基因的RNA来确证对PD-L1和/或PD-L2表达的抑制。
可在每一步骤的之前和之后的多个时间点利用下述方法评价本发明方法中T细胞的功能增强:细胞因子测定法、抗原特异性细胞测定法(四聚体测定法)、增殖测定法、溶细胞性细胞测定法或者使用重组肿瘤相关抗原或免疫原性片段或抗原衍生肽的体内迟发型超敏反应测试。用于测量免疫应答的增加的其它方法的实例包括迟发型超敏反应测试、使用肽主要组织相容性基因复合体四聚体的流式细胞术、淋巴细胞增殖测定法、酶联免疫吸附测定法、酶联免疫斑点测定法、细胞因子流式细胞术、直接细胞毒性测定法、通过定量逆转录酶聚合酶链式反应对细胞因子mRNA的测定法、或当前用于测量T细胞反应的测定法(例如有限稀释法)。
与其中不抑制PD-L1和/或PD-L2表达的对照T细胞相比,本发明的具有经增强的功能的T细胞在T细胞功能方面增强了。与对照T细胞相比,本发明的具有经增强的功能的T细胞在例如IFN-γ的产量上增加了。与对照T细胞相比,本发明的T细胞在IFN-γ产量方面增加达10%或更多,优选20%或更多,更优选30%或更多。
本发明一方面的实例除包括对PD-L1和/或PD-L2的表达的抑制之外,还包括增强进一步T细胞功能的方法。例如,将编码识别所需抗原的TCR的基因导入T细胞,使得有可能获得被赋予针对所述抗原的特异性和其效应子功能经改善的T细胞。所述抗原的实例包括但不具体限于:肿瘤抗原、微生物和病毒衍生抗原。关于针对多种抗原的TCR,已知其氨基酸序列和编码所述氨基酸序列的基因的核苷酸序列,基于载体信息和载体向T细胞的导入构建用于TCR表达的载体,使得有可能制备发挥抗原特异性作用的T细胞。因此,同时将用于抑制PD-L1和/或PD-L2的表达的核酸构建体导入T细胞,或将所述构建体掺入用于TCR表达的载体中,使得有可能增强抗原特异性T细胞的效应子功能。
当将包含α和β链的TCR在T细胞中人工表达时,得自所述T细胞自身拥有的内源TCR基因的α和β链之间的错配等,可阻碍所需抗原特异性的赋予。例如,完成导入的TCR基因的密码子转换,或利用siRNA敲减内源TCR基因(其在WO 2008/153029中公开),使得有可能制备获得了高抗原特异性的T细胞。也可利用本发明的用于增强T细胞功能的方法制备这样的T细胞。
(2)使用本发明具有经增强的功能的T细胞的治疗方法或预防方法
本发明的治疗方法或预防方法具有的特征为给活体施用通过(1)中的本发明用于增强功能的方法制备的T细胞。更确切地说,本发明的治疗方法或预防方法包括下述步骤:(c)给活体施用步骤(b)中获得的功能增强的T细胞。不特定限制本发明功能增强的T细胞所施予的疾病,只要它是对所述T细胞显示敏感性的疾病即可,其实例包括癌症(白血病、实体瘤,等等)、肝炎、流感、传染病,其起因是病毒例如HIV、细菌或真菌,例如结核病、MRSA、VRE、深部真菌病。另外,也可将本发明的细胞用于骨髓移植、或辐射后传染病的预防、或为缓解复发性白血病的供体淋巴细胞输注。此外,所述细胞增强调控T细胞的功能,并可用于预防或治疗自身免疫性疾病中。
在本发明的方法中,可皮内、肌内、皮下、腹膜内、动脉内、静脉内(包括通过留置导管进行的方法)、瘤内、给受试者施用功能增强的T细胞或将其施用到传入淋巴管里。
在本发明的治疗方法或预防方法中,待施用的功能增强的T细胞可为活体自身的细胞,或可为交叉衍生的细胞。
在首次施用本发明的功能增强的T细胞之前,或在治疗起始后的多个时间点上,可使用抗原特异性细胞测定法、增值测定法、溶细胞性细胞测定法、或使用重组肿瘤相关抗原或免疫原性片段或抗原衍生肽的体内迟发型超敏反应测试,在活体中评价本发明实施中所诱导的免疫应答。用于测量免疫应答的增加的其它方法的实例包括迟发型超敏反应测试、使用肽主要组织相容性基因复合体四聚体的流式细胞术、淋巴细胞增殖测定法、酶联免疫吸附测定法、酶联免疫斑点测定法、细胞因子流式细胞术、直接细胞毒性测定法、通过定量逆转录酶聚合酶链式反应对细胞因子mRNA的测量,或当前用于测量T细胞反应的测定法(例如有限稀释法)。此外,可通过活体具有的肿瘤质量、直径或恶性程度,受试者的感染病毒量或感染细菌量,或者受试者的存活率或存活期,来评价免疫应答。
在本发明的方法中,待施予活体的功能增强的T细胞量的实例包括但不具体限于,每成年人每天优选1×106至1×1012个细胞/天,更优选1×107至5×1011个细胞/天,进一步优选1×108至2×1011个细胞/天。
(3)用于疾病的包含本发明功能增强的T细胞的治疗剂
通过(1)中本发明用于增强功能的方法制备的功能增强的T细胞和包含所述T细胞作为活性成分的组合物可用作治疗剂,用于对所述T细胞敏感的疾病(例如,前述的癌症、肝炎或传染病)。
可如下制备本发明的治疗剂,例如通过将通过本发明方法制备的T细胞作为活性成分,与例如已知的有机或无机的载体、赋形剂、稳定剂等(按照制药领域中已知的方法,其适合于胃肠外施用)混合。另外,其可按照本发明(2)中的治疗方法使用。
实施例
以下将通过实施例进一步具体描述本发明,但本发明并非仅限于下述实施例的范围。
实施例1在CD8阳性T细胞中PD-L1的表达的确证
根据WO 2007/032255,制备HLA-A24限制性MAGE-A 4143-151-特异性细胞毒性CD8阳性T细胞克隆2-28(Miyahara Y,和另外13人,Clin.Cancer Res.,第11卷,第5581-5589页(2005),在下文中,被称作CD8阳性T细胞克隆2-28)和MAGE-A4143-151肽。将所述CD8阳性T细胞克隆2-28在37℃下与自身类淋巴母细胞细胞系(LCL)共培养,所述LCL已用80Gly X射线照射并且已经用MAGE-A4143-151肽脉冲,在0天、1天、2天和3天后用抗人PD-L1抗体(BDbioscience制造)将细胞表面的PD-L1染色,并用流式细胞术分析。图1显示用流式细胞仪分析的结果。培养1天后PD-L1的表达最高。
实施例2通过RNA干扰抑制CD8阳性T细胞的PD-L1和PD-L2的表达
将具有SEQ ID NO:1所述序列的对PD-L1特异的siRNA、具有SEQ ID NO:2所述序列的对PD-L2特异的siRNA或阴性对照siRNA(全部由Invitrogen制造)通过电穿孔导入CD8阳性T细胞克隆2-28。
将其中导入有针对PD-L1的siRNA的细胞培养3天,然后在37℃下与经80Gly X射线照射和经MAGE-A4143-151肽脉冲的自身LCL共培养。第二天,用抗人PD-L1抗体将其染色,用流式细胞仪分析在细胞表面的PD-L1的表达。
关于其中导入有针对PD-L2的siRNA的细胞,在导入后的2天和3天后,用抗人PD-L2抗体(BD bioscience制造)将所述细胞染色并用流式细胞仪分析细胞中的PD-L2的表达。
分别地在图2中显示PD-L1的分析结果,和在图3中显示PD-L2的分析结果。在其中不导入siRNA的细胞(不经处理)和其中导入有阴性对照siRNA的细胞(si对照)中,未见对PD-L1或PD-L2表达的抑制。在其中导入有对PD-L1特异的siRNA的细胞(si PD-L1)和其中导入有对PD-L2特异的siRNA的细胞(si PD-L2)中,分别发现对PD-L1和PD-L2的表达的抑制。
实施例3通过RNA干扰促进表达PD-L1和PD-L2的CD8阳性T细胞产生IFN-γ
通过电穿孔将阴性对照siRNA、具有SEQ ID NO:3所述序列的对PD-1特异的siRNA(Invitrogen制造)、具有SEQ ID NO:1所述序列的对PD-L1特异的siRNA或具有SEQ ID NO:2所述序列的对PD-L2特异的siRNA,导入CD8阳性T细胞克隆2-28。在培养3天后,将此细胞与其中已用MAGE-A4143-151肽脉冲自身LCL的细胞以4:1或2:1比例混合,在37℃下共培养所述混合物,在第二天通过ELISA方法测量各培养上清液中的IFN-γ的浓度。
图4中显示各细胞上清液中IFN-γ的浓度。与其中导入有阴性对照siRNA的细胞相比,在其中导入有对PD-1特异的siRNA的细胞(siPD-1)中,在用两种混合比例的培养时未见IFN-γ浓度的改变。与其中导入有阴性对照siRNA的细胞和其中导入有对PD-1特异的siRNA的细胞相比,分别在其中导入有对PD-L1特异的siRNA的细胞和其中导入有对PD-L2特异的siRNA的细胞中,明显见到IFN-γ浓度的增加。
实施例4通过RNA干扰促进表达PD-L1和PD-L2的CD4阳性T细胞表达IFN-γ
将阴性对照siRNA、对PD-1特异的具有SEQ ID NO:3所述序列的siRNA、对PD-L1特异的具有SEQ ID NO:1所述序列的siRNA或对PD-L1特异的具有SEQ ID NO:2所述序列的siRNA,通过电穿孔导入MAGE-A446-66-特异性CD4阳性T细胞克隆5,将其培养3天。将此细胞与其中已用MAGE-A446-66肽脉冲自身LCL的细胞以2:1混合,在37℃下培养所述混合物,在第二天通过ELISA方法测量各培养上清液中的IFN-γ浓度。
图5中显示各细胞上清液中IFN-γ的浓度。在其中导入有阴性对照siRNA的细胞和其中导入有对PD-1特异的siRNA的细胞中,未见IFN-γ的表达。在其中导入有对PD-L1特异的siRNA的细胞和其中导入有对PD-L2特异的siRNA的细胞中,明显见到IFN-γ的表达。
实施例5针对抑制PD-L1和PD-L2的表达选择siRNA
将各自具有SEQ ID NO:8或9(sh831、sh832)所述序列的对PD-L1特异的siRNA,或各自具有SEQ ID NO:10或11(sh833、sh834)所述序列的对PD-L2特异的siRNA(全部由TAKARA BIO INC.制造)或实施例2中使用的阴性对照siRNA,通过电穿孔导入LCL,在37℃下将其培养2天,从每个细胞中提取出总RNA,通过利用QuanTitect SYBR PCR Kit(QIAGEN制造)的实时RT-PCR测量PD-L1和PD-L2的表达,选择具有强的表达抑制能力的siRNA。
分别地,SEQ ID Nos.:12和13中显示用于测量PD-L1表达的实时RT-PCR的引物序列,SEQ ID Nos.:14和15中显示用于测量PD-L2的表达的实时RT-PCR的引物序列,SEQ IDNos.:16和17中显示作为表达的测量对象的人GAPDH(甘油醛-3-磷酸脱氢酶)的实时RT-PCR的引物序列。
图6中显示PD-L1的实时RT-PCR的结果,图7中显示PD-L2的实时RT-PCR的结果。在所述图中,纵坐标显示与人GAPDH的表达相关的PD-L1或PD-L2的表达的比例。根据此结果,选择了SEQ ID NO:9中显示的sh832作为对PD-L1特异的siRNA,和选择SEQ ID NO:11中显示的sh834作为对PD-L2特异的siRNA。
实施例6表达密码子转换型TCR的逆转录病毒载体的制备
根据WO 2008/153029,制备pMS-Ma2和pMS-Pb2。这些载体包括基于MSCV(鼠干细胞病毒)的逆转录病毒载体系统。此外,那些载体包含分别编码识别肿瘤抗原MAGE-A4的TCRα和β链的基因,在这些基因中,转换密码子使得表达不受后述的用siRNA对TCR的RNA干扰抑制(在下文中,其中密码子经转换的基因被称作密码子转换型)。
实施例7表达密码子转换型TCR和siRNA的逆转录病毒载体的制备
从实施例6中制备的pMS-Ma2中用MluI和BglII切下密码子转换型TCRα基因,并制备出TCRα-MluI/BglII。将siRNA簇PDL1-PDL2-TCRα-TCRβ(SEQ ID NO:4中显示的人工合成基因)用BglII和NotI消化,并联同TCRα-MluI/BglII克隆至pMS-Pb2的MluI-NotI位点,以制备MS-aPb1-siPDL_1/2_siTCR载体(载体1)。所述载体1表达密码子转换型TCR,并可表达SEQID Nos.:9、11、18和19中显示的针对PD-L1、PD-L2、TCRα和TCRβ的siRNA。在它们之中,针对TCRα和TCRβ的siRNA抑制内源野生型TCR的表达,并不抑制所述密码子转换型TCR的表达。
此外,制备SEQ ID NO:5中显示的siRNA簇PDL1-PDL2人工合成基因、SEQ ID NO:6中显示的siRNA簇PDL1人工合成基因和SEQ ID NO:7中显示的siRNA簇PDL2人工合成基因,然后与在载体1中一样地制备MS-aPb1-siPDL_1/2载体(载体2)、MS-aPb1-siPDL_1载体(载体3)和MS-aPb1-siPDL_2载体(载体4)。表1显示各个载体表达的基因的概况。

实施例8表达密码子转换型TCR和siRNA的逆转录病毒溶液的制备
用实施例7中制备的载体1-4转化大肠杆菌JM109,和用QIAGEN Plasmid Midi Kit(QIAGEN制造)纯化质粒DNA以提供用于转染的DNA。
根据产品方案使用逆转录病毒包装试剂盒Eco(Retrovirus Packing Kit Eco)(TAKARA BIO INC.制造),将用于转染的各种制备的DNA转染至293T细胞中以获得各种同向性病毒上清液。用0.45μm过滤器(Milex HV,Millipore制造)过滤此上清液,通过使用5-二甲基-1,5-二氮十一亚甲基聚甲溴化物的方法[Kawai等人,Mol.Cell.Biol.,第4卷,第1172页,(1984)],使此上清液感染PG13细胞(ATCC CRL-10686),回收该细胞的所得培养上清液,并使用0.45μm过滤器过滤以制备逆转录病毒溶液。
实施例9用共表达密码子转换型TCR和siRNA的逆转录病毒载体感染人外周血单核细胞
根据手册使用Retronectin(注册商标,TAKARA BIO INC.制造),将分离自人外周血的外周血单核细胞(PBMC)用实施例8中制备的逆转录病毒溶液感染两次,以制备共表达密码子转换型TCR和siRNA导入外周血单核细胞。在第二次病毒感染的3天后,回收所述细胞,用FastPure DNA试剂盒(TAKARA BIO INC.)提取基因组,用原病毒拷贝数检测引物组(Proviral Copy Number Detection Primer Set)(TAKARA BIO INC.制造)和CycleavePCRCore试剂盒(TAKARA BIO INC.制造)计算各细胞的基因组中的平均原病毒拷贝数。用包含所述载体1-4的病毒溶液感染的PBMC的原病毒拷贝数分别为4.6个拷贝/细胞、2.2个拷贝/细胞、3.2个拷贝/细胞和4.5个拷贝/细胞。然后,从第二次病毒感染的5天后,回收所述细胞,用HLA-A2402 MAGE-A4四聚体PE(MBL制造)和人CD8-FITC CONJUGATE(Becton、Dickinson制造)染色,和通过流式细胞仪确定CD8阳性且四聚体阳性的细胞的比例。
图8中显示MAGE-A4四聚体-阳性细胞的比例。横坐标表示导入的逆转录病毒载体,纵坐标表示MAGE-A4四聚体-阳性细胞的比例(%)。如图8所示,通过各逆转录病毒载体的导入确证通过基因导入人外周血单核细胞的抗MAGE-A4TCRα/β复合蛋白的表达。另外,针对由所述逆转录病毒载体表达的PD-L1和PD-L2的siRNA的表达并不对所导入的密码子转换型TCR的表达产生很大的影响。此外,针对TCRα和TCRβ的siRNAs通过用siRNA抑制野生型内源TCR基因的表达的作用提高了密码子转换型TCRα/β复合蛋白表达的比例。
工业适用性
本发明提供了增强T细胞功能的方法,和具有经增强的功能的T细胞。此T细胞增强对癌症的免疫应答,并可用于有效的癌症免疫疗法以及传染病或自身免疫性疾病的治疗和预防。

Claims (9)
1. 一种用于增强T细胞功能的方法,包括抑制所述T细胞的程序性死亡-1配体1 (PD-L1)和/或程序性死亡-1配体2 (PD-L2)的表达的步骤,其中所述步骤离体进行,并且其中通过针对PD-L1和/或PD-L2的siRNA抑制PD-L1和/或PD-L2的表达,所述siRNA直接导入T细胞中或在T细胞中转录,其中与其中不抑制PD-L1和/或PD-L2表达的对照T细胞相比,所述功能增强是T细胞干扰素-γ (IFN-γ)的产生增强达30%或更多。
2.权利要求1的用于增强T细胞功能的方法,其中所述T细胞是细胞毒性T细胞或者调节性T细胞。
3.权利要求2的用于增强T细胞功能的方法,其中所述调节性T细胞是辅助性T细胞、调控T细胞或Th17细胞。
4. 一种具有经增强的功能的T细胞,其在PD-L1和/或PD-L2的表达方面受到抑制,其中通过针对PD-L1和/或PD-L2的siRNA抑制PD-L1和/或PD-L2的表达,所述siRNA直接导入T细胞中或在T细胞中转录,其中与其中不抑制PD-L1和/或PD-L2表达的对照T细胞相比,所述功能增强是T细胞干扰素-γ (IFN-γ)的产生增强达30%或更多。
5.权利要求4的具有经增强的功能的T细胞,其中所述T细胞是细胞毒性T细胞或调节性T细胞。
6.权利要求5的具有经增强的功能的T细胞,其中所述调节性T细胞是辅助性T细胞、调控T细胞或Th17细胞。
7.一种用于对权利要求4-6中任一项的具有经增强功能的T细胞显示敏感性的疾病的治疗剂,其包含所述T细胞作为活性成分。
8.权利要求1的用于增强T细胞功能的方法,其还包括将编码识别所需抗原的TCR的基因导入所述T细胞中。
9.权利要求4的具有经增强的功能的T细胞,其中将编码识别所需抗原的TCR的基因导入所述T细胞中。
