包封在聚乙烯中的染料已添加至牙膏、漱口液和其他口腔护理组合物以提供着色的斑点或其他多彩的装饰性图案,以便改善这些产品的消费者接受性。虽然聚乙烯已用于包封在美观产品中使用的染料,但公众日益关注的问题是塑料微粒可因其不易于可生物降解而造成全球性污染。
可生物降解聚合物诸如聚乳酸在本领域中是已知的。聚乳酸及其降解产物,即H2O和CO2对人身体既没有毒性又没有致癌性。因此,这种聚合物可用于生物医学应用中。
例如,已报道聚乳酸聚合物为可用于药物诸如小分子和蛋白质的载体。这些聚合物经由水解的降解特性有益于将药物释放到患者中。然而,此类降解特征对于在储存和使用过程中不希望降解的含水产品似乎是不可取的。因此,在本领域中仍期望在不使用聚乙烯的情况下向消费者提供美观着色的口腔护理产品。
发明内容
本公开涉及一种稳定性口腔护理组合物,其包含口服可接受的媒介物和由可生物降解聚合物包封的着色剂。
本文还提供了一种制备口腔护理组合物的方法,其包括:(a)将水混合到口服可接受的媒介物中以形成混合物,以及将包封在可生物降解聚合物中的着色剂分散到在(a)中形成的混合物中以形成稳定性口腔护理组合物。
本公开还提供了一种促进口腔健康的方法,所述方法包括:使口腔表面与稳定性口腔护理组合物接触,所述稳定性口腔护理组合物包含口服可接受的媒介物和由可生物降解聚合物包封的着色剂。

除非另外规定,否则本文和在本说明书中其他处表达的所有百分比和量应理解为是指重量百分比。给定量是基于材料的有效重量计。
组合物
本发明人意外地认识到,由可生物降解聚合物包封的着色剂可用于提供甚至在组合物含有水时也不渗色或浸析的美观的稳定性口腔护理组合物。如本文所用,术语“包封”意指用可生物降解聚合物包围着色剂的粒子或小滴以提供尺寸在下述范围内的微胶囊,所述范围为例如约10μm至约1000μm,更通常约100μm至约500μm,甚至更通常约100μm至约300μm,还更通常约230μm。术语“微胶囊”是指微粒,所述微粒包括例如充当壁形成材料的可生物降解聚合物或共聚物壳和位于微粒的芯内的被包封材料例如着色剂。如本文所用,“微胶囊”还涵盖分散在没有独特外壁相的聚合物中的均相着色剂颗粒。微胶囊可以为任何形状,例如球形、其他几何形状或不规则形状。
如本文所述,分散在本发明稳定性口腔护理组合物中的本发明包封的着色剂展示出向稳定性口腔护理组合物的媒介物中的极少迁移。因此,着色剂与组合物的其他成分最小程度地反应,并且着色剂颜色的完整性被保持。例如,着色剂不像在与媒介物中组分接触时可能发生的那样进行褪色或变色。此外,着色剂能够在稳定性口腔护理组合物的媒介物内分散为离散粒子,诸如斑点或其他图案,而不渗色或浸析到媒介物中。
可生物降解聚合物
在一些实施方案中,着色剂包封在可生物降解聚合物中,所述可生物降解聚合物具有的熔点在约80℃至约180℃,更通常地在约130℃至约150℃之间,甚至更通常地在约140℃至约150℃的范围内,在从喷嘴喷洒出时能够形成细小滴,在室温下为固体并且在加热时液化而不被破坏。可生物降解聚合物在与水或其他成分(诸如标准地存在于口腔护理组合物中的风味剂、甘油、山梨醇、表面活性剂和其他材料)接触时是稳定的。在一些实施方案中,聚合物是可分散在合适溶质或霜剂等中的聚合物,已被形成为例如微胶囊或微球。
如本文所用术语“可生物降解的”是指含有可通过水解、酶和/或细菌进行裂解以形成较低分子量物质的化学键的聚合物。本公开中所述的可生物降解聚合物涵盖符合ASTM6400中规定的可生物降解性标准的那些。更具体地,ASTM 6400标准被设计用于测试塑性材料在暴露于实验室条件下的受控堆肥环境时的有氧生物降解的程度和速率。所述测试旨在模拟公认的城市和工业需氧堆肥设施中的堆肥条件。使用的参数是用于确定塑料和由塑料制备的产品是否令人满意地堆肥(包括以与已知的可堆肥材料相当的速率生物降解)所需的那些参数。此外,需要ASTM 6400规范中的特性来确保这些材料的降解不减少由堆肥过程产生的堆肥的价值或效用。例如,ASTM 6400标准陈述到,当在受控的堆肥环境中十二周之后,在2.0-mm筛网上筛分之后留存不超过测试材料的原始干重的10%时,指示测试材料的崩解性令人满意。此外,当与阳性对照诸如纤维素相比时,在测试周期结束时,至少60%的有机碳可转化为二氧化碳。此外,应观察到对堆肥的质量没有不利影响。例如,对于两种不同的植物种类,样品堆肥的发芽速率和植物生物质应不小于90%的对应的对照堆肥的发芽速率和植物生物质。此外,重金属必须满足如由ASTM 6400指南所指出的可接受水平。
本文所用的术语“聚合物”是赋予其本领域中所用的普通含义,即,包括由共价键连接的一个或多个重复单元(单体)的分子结构。重复单位可以全部是相同的,或在一些情况下,可以有多于一种类型的重复单元存在于聚合物内。如果一种以上类型的重复单元存在于聚合物内,那么聚合物被认为是“共聚物”。形成共聚物的重复单元可以按任何方式来布置。例如,重复单元可以按随机顺序、交替顺序或呈“嵌段”共聚物形式来布置,所述嵌段共聚物即,包含各自包含第一重复单元(例如,第一嵌段)的一个或多个区,和各自包含第二重复单元(例如,第二嵌段)的一个或多个区等。嵌段共聚物可以具有两个(二嵌段共聚物)、三个(三嵌段共聚物),或更多数量的不同嵌段。
用于包封本公开的着色剂的可生物降解聚合物包括但不限于聚乳酸、聚己内酯、聚乙醇酸、聚(丙交酯-共-乙交酯)聚合物(PLGA)及其共聚物、衍生物和混合物、聚酸酐、聚(原酸)酯、聚氨酯、聚(丁酸)、聚(戊酸)、聚丙交酯-共-己内酯)及其共混物和共聚物。
在一些实施方案中,基于聚乳酸的聚合物用作包封材料。如本文所用,“基于聚乳酸的聚合物”为具有至少约20重量%或更多的聚乳酸的均聚物或共聚物。在一些实施方案中,基于聚乳酸的聚合物具有按重量计至少约50%、至少约60%、至少约75%、至少约80%或至少约90%的聚乳酸。
术语“聚乳酸”包括以下四种形态上不同的聚乳酸聚合物中的任何一种或多种:D-聚乳酸、L-聚乳酸、D,L-聚乳酸和内消旋-聚乳酸。“D-聚乳酸”和“L-聚乳酸”分别是右旋聚乳酸和左旋聚乳酸,并且两者均是在透射穿过聚合物时使光向量旋转的光学活性聚合物。“D,L-聚乳酸”为外消旋聚合物,即具有确定构象的D-聚乳酸单元和L-聚乳酸单元的D-聚乳酸和L-聚乳酸的共聚物。“内消旋聚乳酸”为D-聚乳酸和L-聚乳酸的无规共聚物。
在一些实施方案中,合适的本公开的可生物降解的基于聚乳酸的聚合物包括包含诸如ε-己内酯乙交酯、对二氧杂环己酮、戊酸或丁酸的单体的共聚物。在一些实施方案中,本公开的基于聚乳酸的聚合物与另一种可生物降解聚合物共混或共聚合。此类合适的可生物降解聚合物包括但不限于聚己内酯、聚乙醇酸、聚(丙交酯-共-乙交酯)聚合物(PLGA)及其共聚物、衍生物和混合物、聚酸酐、聚(原酸)酯、聚氨酯、聚(丁酸)、聚(戊酸)、聚丙交酯-共-己内酯)及其共混物、共聚物和衍生物。如本文所用,“衍生物”包括具有取代基的、附加的化学基团和由本领域技术人员常规进行的其他修饰的聚合物。
在一些实施方案中,聚交酯聚合物具有在约100,000至约500,000kDa范围内的重均分子量(Mw)。在其他实施方案中,重均分子量在100,000至300,000道尔顿的范围内,并且在又其他实施方案中在100,000至约250,000道尔顿的范围内。
在一些实施方案中,聚乳酸聚合物为具有75∶25的聚乳酸(PLA):聚乙交酯(PGA)重量比、具有76,000-116,000道尔顿分子量的共聚物。在其他实施方案中,聚乳酸聚合物为具有85∶15的PLA∶PGA重量比、具有190,000-240,000道尔顿分子量的共聚物。
制备聚乳酸聚合物的方法在本领域中是熟知的。例如,聚乳酸聚合物可通过例如如本文所述的D-或L-聚乳酸或其组合的缩聚(包括溶液缩聚和熔融缩聚),更通常地,由丙交酯即聚乳酸的环状二聚体的开环聚合制备。聚乳酸聚合物的化学修饰可通过以下方式进行:使乳酸与其他单体共聚合或通过交联或添加有机或无机填料/纤维材料以修改物理特征和/或降低总体的材料成本。
有机填料的非限制性实例包括木粉、种子、聚合物粒子、未胶凝淀粉颗粒、软木、明胶、锯屑、磨碎的聚合物材料、琼脂基材料等。
有机填料的实例包括碳酸钙、二氧化钛、二氧化硅、滑石、云母、砂、碎石、压碎的岩石、矾土、花岗石、石灰石、砂岩、玻璃珠、气凝胶、干凝胶、粘土、氧化铝、高岭土、微球、中空玻璃球、多孔性陶瓷球、石膏二水合物、不溶性盐、碳酸镁、氢氧化钙、铝酸钙、碳酸镁、陶瓷材料、火山灰材料、盐、锆化合物、硬硅钙石(结晶硅酸钙凝胶)、轻量膨胀的粘土、珍珠岩、蛭石、水化的或未水化的水凝水泥粒子、符石、沸石、片状剥落岩石(exfoliated rock)、矿山、矿物质等。在一些实施方案中,在本公开的可生物降解聚合物中使用10重量%至30重量%,诸如15-30%,诸如15%至25%的有机纤维、无机纤维或纤维。
可掺入到可生物降解组合物中的纤维的非限制性实例包括天然存在的有机纤维,诸如从木材、植物叶和植物茎提取的纤维素纤维。这些有机纤维可衍生自棉花、木材纤维(硬木或软木纤维,其实例包括南方硬木和南方松木)、亚麻、蕉麻、剑麻、苎麻、大麻和蔗渣。此外,还可使用由玻璃、石墨、二氧化硅、陶瓷、岩棉或金属材料形成的无机纤维。
合适的可商购获得的聚乳酸聚合物可例如从美国Cargill Dow以、从日本MitsuiChemicals以LACEA、从日本Shimadzu Seisakusho以Lacty获得。在某些实施方案中,聚乳酸聚合物包括可购自Cargill Dow的PLA NatureWorksTM 5729B、2002D和5040D。
着色剂
如上所提出,本文所述的可生物降解聚合物包封着色剂。如本文所用,术语“着色剂”是指向另一种物质赋予颜色的呈干粉或液体形式的物质。一般来讲,着色剂包括染料、色淀、颜料或其组合。在一些实施方案中,也可将云母、云母基珠光颜料、滑石、碳酸钙和二氧化硅用作着色剂。更通常地,着色剂为染料、色淀或一种或多种染料和/或一种或多种色淀的组合。
如本文所用,术语“染料”是指有机物质,其在含水介质中基本上是水溶性的,在所述含水介质中染料保持化学稳定。“色淀”是通过用惰性粘结剂或媒染剂(通常是金属盐)沉淀染料制造的。通常使用的盐包括硫酸钡、硫酸钙、氢氧化铝和铝氧化物(矾土)。在一些实施方案中,使用了铝色淀。
“颜料”是向另一种物质赋予颜色的合成的或天然的水不溶性物质。
与本公开的稳定性口腔护理组合物一起使用的染料通常为目前根据《联邦食品、药品和化妆品法案》而被认证用于食品和摄取性药品中的食品颜色添加剂,包括成各种比例的染料诸如FD&C蓝1号、FD&C蓝2号、FD&C绿3号、D&C绿5号、D&C橙5号、FD&C红3号、D&C红21号、D&C红22号、D&C红27号、D&C红28号、D&C红30号、D&C红40号、FD&C黄5号、FD&C黄6号和D&C黄10号及其混合物。
可与本公开的本发明稳定性口腔组合物一起使用的色淀包括但不限于FD&C蓝1号铝色淀、FD&C蓝2号铝色淀、FD&C绿3号铝色淀、FD&C红3号铝色淀、FD&C红40号铝色淀、FD&C黄5号铝色淀、FD&C黄6号铝色淀。在某些实施方案中,着色剂选自FD&C蓝1号铝色淀、FD&C黄5号铝色淀和FD&C红40号铝色淀。
在一些实施方案中,着色剂为颜料。颜料包括无毒、水不溶性无机颜料,诸如二氧化钛和氧化铬绿、群青蓝和粉红以及氧化铁,例如胭脂树(Annatto)提取物、叶绿酸铜络合物、角黄素、β-胡萝卜素、合成的铁氧化物、TiO2及其混合物。
适合用于本文的染料和色淀、其结构和特性是本领域中技术人员所熟知的,参见例如Kirk-Othmer Encyclopedia of Chemical Technology,第3版,第6页,第561-596页,所述文献以引用的方式整体并入本文。
在一些实施方案中,组合物是稳定的。如本文所用“稳定”意指着色剂向本发明口腔护理组合物的媒介物中的迁移通过可生物降解包衣而被抑制、缓和或阻止一段时间,使得导致着色剂变化(例如着色剂的褪色)的着色剂与组合物中其他成分的反应视觉上观察不到。“稳定”也是指缺乏或抑制了包封的着色剂浸析或渗色或迁移到本发明稳定性洁齿剂组合物中的能力或者缺乏或抑制了包封的着色剂浸析或渗色或迁移到本稳定性洁齿剂组合物中的倾向,使得在视觉上观察时可使包封的着色剂的离散粒子或图案在媒介物中保持完整性。如本文所用,“视觉上观察不到”或“视觉上不可观察到的”意指当用肉眼观察时,包封在可生物降解聚合物中的着色剂未可视化为或不可观察到向媒介物中渗色或浸析,或者未可视化为或不可观察到着色剂的颜色变化。
在各种实施方案中,包封在可生物降解聚合物中的着色剂的迁移在下述范围内的温度下被抑制并且在视觉上观察不到:约20℃至约50℃,例如约20℃至约40℃或约30℃或约40℃。
在一些实施方案中,着色剂向媒介物中的迁移在一定持续时间或时间段内被抑制并且在视觉上不可观察到,所述一定持续时间或时间段诸如约一天、约一周、约一个月、约两个月、约三个月、约四个月、约六个月、约一年、约两年或约三年或更长。
如本文所用的“稳定”还涵盖指定量的着色剂向本公开的口腔护理组合物的本发明媒介物中进行指定的持续时间的迁移,其中在约20℃至约50℃,例如约20℃至约40℃或约30℃或约40℃的范围内的温度下所述量为约0%、小于约1%、小于约2%、小于约3%、小于约4%、小于约5%、小于约10%或小于约20%,其中所述持续时间为约一天、约一周、约一个月、约两个月、约三个月、约四个月、约六个月、约一年、约两年或约三年或更长。
包封在可生物降解聚合物中并且适合与本发明稳定性口腔护理组合物一起使用的着色剂是可商购获得的。例如,由聚乳酸和保护性包衣包围的FD&C蓝1号色淀(EcoBlue50BL1)可从Micro Powders,Inc.,Tarrytown,NY购买。
在一些实施方案中,包封在可生物降解聚合物中的着色剂可使用本领域中熟知的方法制备,所述方法包括但不限于喷雾干燥、界面聚合、热熔融包封、相分离包封(溶剂去除和溶剂蒸发)、自发性乳液和凝胶,参见例如Kirk-Othmer Encyclopedia of ChemicalTechnology,Third Edition,第125卷第470-493页(1981),所述文献以引用的方式整体并入本文。
可使着色剂诸如本公开的色淀或染料包封在聚合物(诸如聚乳酸)中所采用的方法的实例是通过将所需量的着色剂分散在已被热软化的聚合物中来形成液体组合物。例如色淀或染料与聚合物(诸如聚乳酸)的比率在约0.1%至约9%,更通常约0.1%至约6%,甚至更通常约0.1-3%的范围内。在一些实施方案中,颜料着色剂与可生物降解聚合物的比率或云母、滑石、碳酸钙和二氧化硅与可生物降解聚合物(诸如聚乳酸)的比率在1%至50%,更通常地10%至50%或甚至更通常地30%至50%的范围内。
在一些实施方案中,着色剂的所需量为使得本发明口腔护理组合物中着色剂的最终浓度高达20%的着色剂量,例如按重量计约0.1%至约12%、约0.1%至约10%、约3%至约10%、约0.1至约5%、约0.1%至约2%或约0.1%至约1%。
可搅拌液体聚合物分散体以使得液体聚合物沉积在着色剂材料的每个实体上,从而形成具有液体聚合物壁的小滴。然后将分散体冷却并研磨以提供于其中包封着色剂例如染料或色淀的固体粒子。聚合物粒子的粒度分布可在约10μm至约1000μm的范围内。
口服可接受的媒介物
如本文所述的包封的着色剂与口服可接受的媒介物混合形成口腔护理组合物。如本文所用的“口腔护理组合物”是指这样的组合物,其在口腔中保留足够的时间以接触基本上所有的牙表面和/或口腔组织,达到口腔活性。本发明的口腔护理组合物可呈各种形式,包括漱口液或洁齿剂。洁齿剂可包括牙用片剂、牙膏(牙霜剂)、牙粉、凝胶、液体、液状凝胶或本领域中技术人员已知的任何其他形式。
如本文所用,“口服可接受的媒介物”是指安全用于本公开的组合物中的材料或材料组合,与合理的效/险比相称,任何所需的活性成分可在保留显著功效的同时与所述效/风险比相关联。
在一些实施方案中,口服可接受的媒介物是高水含量的口服可接受的媒介物并且可包含任何已知成分或添加剂。例如,媒介物可包括水、甘油、山梨醇和氯化钠的液体混合物。在此类实施方案中,水的存在量通常占口腔组合物的至少约20重量%,并且通常约25重量%至50重量%,更通常约40%至75%,甚至更通常在77重量%至88重量%的范围内。在商业上合适的口腔组合物的制备中采用的水应优选为去离子的并且不含机杂质。
在一些实施方案中,口服可接受的媒介物包含结构剂,所述结构剂诸如在结构化漱口液,即包含提供流变学和结构化有益效果(例如,如通过静态屈服应力测量的)的材料的漱口液中使用。此类结构剂包括但不限于可在水合时溶胀或膨胀形成独立的微凝胶粒子的随机分散体的聚合物或树胶结构剂。聚合物或树胶结构剂的实例包括:结冷胶、果胶、藻酸盐、阿拉伯半乳聚糖、角叉菜胶(caageenan)、瓜尔豆胶、鼠李糖胶(rhamsan gum)、红藻胶(furcellaran gum)、羧甲基纤维素和纤维素。还参见PCT公开号WO 2014088534,其以引用的方式整体并入本文。在一些实施方案中,结构化剂可用于提供0.20至400Pa的静态屈服应力。
在一些实施方案中,口服可接受的媒介物包含0.05重量%至1.00重量%的黄原胶。在某些实施方案中,特别是当口服可接受的媒介物用作漱口液时,媒介物包含0.01重量%至0.25重量%的黄原胶、0.01重量%至0.30重量%的低酰基结冷胶或其组合。
在一些实施方案中,媒介物是低水含量的口服可接受的媒介物并且可包含任何已知成分或添加剂。例如,媒介物可包括水、甘油和山梨醇的液体混合物。在一些实施方案中,稳定性口腔组合物的水含量小于约20%、小于约10%、小于约8%、小于约4%、小于约2%、小于约1%或小于约0.1%w/w。
在一些实施方案中,口服可接受的载体是基本上无水的。适合用于此类实施方案的低湿润剂/媒介物包括但不限于聚乙二醇,诸如PEG400、PEG600;PEG/PPG共聚物诸如PEG/PPG 38/8共聚物、PEG/PPG-1 16/66共聚物(分别由BASF公司以L4370和L1220销售)。在一些实施方案中,湿润剂/媒介物的存在总量为2%至55%w/w,并且通常为2%至35%w/w。丙二醇或甘油的存在量还可以为0%至15%w/w.。
在一些实施方案中,口腔护理组合物的粘度在200cps至1,000,000cps的范围内。例如,漱口液的粘度可在200至1000cps的范围内,液状凝胶牙膏的粘度可在约5,000至60,000cps的范围内,并且洁齿剂的粘度可在约60,000cps至1,000,000cps的范围内。
在一些实施方案中,可添加表面活性剂来增强制剂的稳定性、帮助通过去垢剂清洁口腔表面,并且在搅拌时例如在用本公开的口腔护理组合物刷洗或淋洗过程中提供泡沫。表面活性剂通过将防龋齿剂完全分散遍及例如口腔来实现增加预防作用。在各种实施方案中,合适的表面活性剂可用作表面活性剂、乳化剂和/或泡沫调节剂。
可使用任何口服可接受的表面活性剂,它们大部分是阴离子型、非离子型或两性离子型的。合适的阴离子型表面活性剂包括但不限于C8-20烷基硫酸酯、磺化的C8-20脂肪酸甘油单酯的水溶性盐、肌氨酸盐、牛磺酸盐等。这些和其他种类的例示性实例包括十二烷基硫酸钠、椰油基甘油单酯磺酸钠、月桂基肌氨酸钠、月桂基羟乙基磺酸钠(sodium laurylisoethionate)、月桂醇聚醚羰酸钠(sodium laureth carboxylate)和十二烷基苯磺酸钠。合适的非离子型表面活性剂包括但不限于泊洛沙姆(诸如泊洛沙姆(polyoxomer)407)、聚氧乙烯失水山梨醇酯(诸如聚山梨醇酯20)、脂肪醇聚氧乙烯醚、烷基酚聚氧乙烯醚、叔胺氧化物、叔膦氧化物、二烷基亚砜等。合适的两性型表面活性剂包括但不限于具有阴离子基团(诸如羰酸根、硫酸根、磺酸根、磷酸根或膦酸根)的C8-20脂肪族仲胺和叔胺的衍生物。合适的实例为椰油酰胺基丙基椰油酰氨基丙基。
在一些实施方案中,一种或多种表面活性剂的存在总量可以为约1.8%至约2%w/w。在一些实施方案中,一种或多种表面活性剂的存在总量可以为约1.9%至约2%w/w。在一些实施方案中,一种或多种表面活性剂的存在总量可以为约2%w/w。
在一些实施方案中,组合物任选包含增稠剂。可以使用任何口服可接受的增稠剂,其无限制地包括卡波姆(也称为羧乙烯基聚合物)、角叉菜胶(也称为爱尔兰藓)并且更具体地-角叉菜胶(I-角叉菜胶)、高分子量聚乙二醇(诸如CARBOWAX,可从Dow ChemicalCompany获得)、纤维素聚合物(诸如羟乙基纤维素、羧甲基纤维素(CMC)及其盐,例如CMC钠)、天然树胶(诸如低酰基结冷胶、黄原胶、刺梧桐胶、阿拉伯胶和黄蓍胶)、胶体硅酸镁铝,和胶体二氧化硅和/或增稠剂二氧化硅及其混合物。
在一些实施方案中,增稠剂二氧化硅与本发明口服可接受的媒介物一起使用。在某些实施方案中,增稠剂二氧化硅为具有高表面积和内部孔体积的合成的无定形沉淀材料,以提供约50ml或更大/20克二氧化硅的水吸附和约200ml或更大/100g二氧化硅的油吸附(根据ASTM D281方法)。可使用的增稠剂二氧化硅的实例为165、163和153(来自Huber);200和22S(来自Evonik);15和SM 660(来自W.R.Grace&Co.);和Tixocil 43B(来自Rhodia)。

在一些实施方案中,一种或多种调味剂任选的存在总量为约0.01%至约5%w/w。在一些实施方案中,一种或多种调味剂任选的存在总量为约0.05%至约2%w/w。在一些实施方案中,一种或多种调味剂任选的存在总量为约0.1%至约2.5%w/w。在一些实施方案中,一种或多种调味剂任选的存在总量为约0.1%至约0.5%w/w。在一些实施方案中,一种或多种调味剂任选的存在总量为约2.25%w/w。
可用于本文中的甜味剂包括口服可接受的天然或人造、营养性或非营养性甜味剂。此类甜味剂包括右旋糖、聚右旋糖、蔗糖、麦芽糖、糊精、干的转化糖、甘露糖、木糖、核糖、果糖、左旋糖、半乳糖、玉米糖浆(包括高果糖玉米糖浆和玉米糖浆固体)、部分水解的淀粉、氢化的淀粉水解产物、山梨糖醇、甘露醇、木糖醇、麦芽糖醇、异麦芽酮糖醇、阿司帕坦、新特姆(neotame)、糖精及其盐、三氯蔗糖、基于二肽的强增甜剂、环己氨基磺酸盐(cyclamate)、二氢查耳酮及其混合物。一些实施方案中任选地包含一种或多种甜味剂。在一些实施方案中,一种或多种任选的甜味剂的存在总量为约0.005%至约5%w/w。在一些实施方案中,一种或多种任选的甜味剂的存在总量为约0.01%至约1%w/w。
合适的防腐剂诸如苯甲酸钠在口服可接受的媒介物中的存在量可以为基于组合物的总重量的例如约0%至约0.2%,并且通常约0.05%至约0.10%。
活性成分
本公开的稳定性口腔护理组合物任选地包含一种或多种活性材料,所述一种或多种活性材料可操作用于预防或治疗口腔的硬组织或软组织的病状或病症、预防或治疗生理病症或病状,或提供美容有益效果。
本公开的一些实施方案包括牙齿研磨剂或牙齿研磨药剂的组合。如本文所用,术语“研磨剂”或“研磨药剂”还包括常常称之为“抛光剂”的材料。可使用任何口服可接受的研磨剂,但是通常应选择研磨剂的类型、细度(粒度)和量使得牙齿釉质在正常使用组合物中不被过度地磨损。合适的研磨剂包括但不限于二氧化硅(呈硅胶、水合二氧化硅或沉淀二氧化硅的形式)、氧化铝、不溶性磷酸盐、碳酸钙、树脂的研磨剂诸如脲醛缩合产物等。

如果存在,则研磨剂的平均粒度通常为约0.1μm至约30μm、例如约1μm至约20μm或约5μm至约15μm。在一些实施方案中,一种或多种研磨剂的存在总量为约0.1%至约40%w/w。在一些实施方案中,研磨剂为焦磷酸钙。在一些实施方案中,焦磷酸钙的存在量为约5%至约50%w/w。
在本公开的各种实施方案中,口腔护理组合物包含抗牙结石剂。合适的抗牙结石剂非限制地包括磷酸盐和聚磷酸盐(例如焦磷酸盐)、聚氨基丙磺酸(AMPS)、六偏磷酸盐、柠檬酸锌三水合物、多肽、聚烯烃磺酸盐、聚烯烃磷酸盐、二磷酸盐。在一些实施方案中,抗牙结石剂的存在量为约0.1%至约30%w/w。在一些实施方案中,稳定性口腔护理组合物包含抗牙结石剂的组合物。在一些优选实施方案中,使用焦磷酸四钠(TSPP)和三聚磷酸钠(STPP)用作抗牙结石剂。在一些实施方案中,抗牙结石剂包含约1%至约2%w/w的TSPP和约0%至约7%w/w的STPP。

在一些实施方案中,口腔组合物包含增白剂。可在本发明的稳定性口腔护理组合物中使用任何已知或开发的增白剂。在各种实施方案中,增白剂为氧化剂、还原剂或其组合。在最广泛的含义中,“氧化剂”旨在包括在正常且认可的使用期间可接受来自口腔环境中的另一种分子的电子而对口腔没有有害或不可接受的损害影响的那些化合物。
适合与本发明稳定性口腔护理组合物一起使用的氧化剂包括过氧化物、金属亚氯酸盐和过硫酸盐。示例性过氧化物包括氢过氧化物、过氧化氢、碱金属和碱土金属的过氧化物、有机过氧化合物、过氧酸、其药学上可接受的盐及其混合物。

在一些实施方案中,固体增白剂诸如过硼酸钠、过氧化脲、亚氯酸钙、亚氯酸钡、亚氯酸镁、亚氯酸锂、亚氯酸钠、亚氯酸钾或其混合物用于本文所述的稳定性口腔护理组合物中。
在一些实施方案中,增白剂与例如聚合物诸如PVP(聚(N-乙烯基吡咯烷酮)结合。例如在美国专利号5,122,370中公开了合适的PVP络合物,所述专利的内容以引用的方式整体并入本文。
在一些实施方案中,稳定性口腔护理组合物任选地包含氟离子。在一些实施方案中,氟离子源选自:氟化物、单氟磷酸盐(MFP)和氟硅酸盐。在一些实施方案中,一种或多种释放氟化物离子的化合物任选地以提供100至20,000ppm、200至5,000ppm或500至2,500ppm总氟离子的量存在。
组合物还可包含亚锡离子或亚锡离子源以减轻钙损失。合适的亚锡离子源无限制地包括氟化亚锡、其他卤化亚锡诸如二水二氯化锡、焦磷酸亚锡、有机亚锡羧酸盐诸如甲酸亚锡、乙酸亚锡、葡萄糖酸亚锡、乳酸亚锡、酒石酸亚锡、草酸亚锡、丙二酸亚锡和柠檬酸亚锡、乙醛酸亚锡(stannous ethylene glyoxide)等等。一种或多种亚锡离子源任选和例示性的存在总量为约0.01%至约10%,例如约0.1%至约7%或约1%至约5%。
本公开的组合物任选地包含抗微生物(例如抗菌)剂,诸如十六烷基氯化吡啶鎓(CPC)。可用的抗菌剂的例示性列表提供于授予Gaffar等的美国专利号5,776,435中,所述专利的内容以引用的方式并入本文。一种或多种抗微生物剂任选存在的抗菌有效总量通常为约0.05%至约10%,例如约0.1%至约3%、0.075%至约3%。
pH调节剂等也可掺入到本发明口腔护理组合物中。在一些实施方案中,组合物的pH维持在约5至约10.5、诸如约5至约9、诸如约7至约9、诸如约5至约7.5或诸如约5.5至约7的范围内。在一些实施方案中,例如含有碳酸钙或不溶性磷酸盐研磨剂的口腔护理组合物,组合物的pH维持在约8.5至10.5的范围内。
方法
在各种实施方案中,本公开提供了制备口腔护理组合物的方法,所述方法包括将水混合到口服可接受的媒介物中以形成混合物(a)以及将包封在可生物降解聚合物中的着色剂混合到在(a)中形成的混合物中以形成口腔护理组合物。
例如,在一些实施方案中,可将所需增稠剂与水合并并且混合形成均匀混合物。然后可添加甜味剂和其他活性成分诸如抗牙结石剂并且混合为均匀混合物,然后混合甜味剂和表面活性剂。然后由本发明的可生物降解聚合物包封的着色剂可分散遍及含有水和其他成分的口服可接受的媒介物。
包封的着色剂可根据尺寸和/或颜色变化,并且可分散遍及含水的媒介物,使得稳定性口腔护理组合物看起来含有斑点、斑点状条纹、波纹状条纹、直条纹、斑点、点或由包封的着色剂形成的其他图案。在其他实施方案中,包封的着色剂可被配制并且以足以使得整个本发明口腔护理组合物视觉上看起来为一种单色的量添加、
本文还提供了用于促进人或动物受试者的口腔健康的方法,所述方法包括使口腔表面与本公开的口腔护理组合物接触,所述口腔护理组合物包含赋予稳定性口腔护理组合物所需外观的至少一种包封的着色剂。如本文所用,短语“促进口腔健康”涵盖预防、养护、逆转、减弱、减轻、改善、最小化、抑制或停止人或动物的一种或多种口腔病状的有害影响。短语“口腔病状”包括但不限于牙斑、牙垢、色斑、口臭、齿龈炎、牙周炎及其组合。
如本文所用的“动物受试者”包括非人哺乳动物诸如犬科、猫科和马科动物。稳定性口腔护理组合物与哺乳动物受试者的口腔表面接触,从而以高效方式促进口腔健康。
在各种实施方案中,根据本公开制备的稳定性口腔护理组合物可定期向口腔表面施用,例如基于每天,每天至少一次持续多天,或另选地每隔两天或三天。在一些实施方案中,每天向表面施用稳定性口腔护理组合物1至3次,持续至少2周至8周、4个月至3年、或更多至一生。
本文所述的实例和其他实施方案是示例性的并且不旨在限制描述本公开的组合物和方法的完全范围。可在本公开的范围内进行具体实施方案、材料、组合物和方法的等同改变、修改和变型,具有基本上类似的结果。
实施例
实施例1-结构化漱口液
通过混合下表1中所述的成分制备三种漱口液制剂A、B和C。如表1所示的配方A和B用黄原胶(0.110重量%,配方A和0.090重量%,配方B)和结冷胶(0.147重量%,配方A和0.122重量%,配方B)的组合进行结构化。配方C不含树胶组分。使用NaCl以达到在255至595(表1以厘泊(cps)报道,其中1cps=0.001Pa·s)范围内的粘度。将包封在聚乳酸中的0.5重量%的FD&C蓝色1号铝色淀(直径大约为230μm)添加到每种制剂。包封的粒子获自MicroPowders,lnc.,Tarrytown,NY。
使用布氏粘度计型号HADV-II+Pro(可购自Brookfield EngineeringLaboratories,Middleboro,MA)在25℃下以1rpm测量漱口液制剂的粘度。由于制剂的低粘度,使用了V73心轴,其足以大至实现较大准确性的粘度。以预设的每分钟转数(RPM)旋转心轴。根据如针对仪器所指定的最大值(Tmax)的%确定扭矩(扭矩%)(对于HADVII+Pro,Tmax=1.437mN·m)。仅具有10%与100%之间的扭矩%的那些测量值认为是有效的。使用用于库埃特几何的熟知公式将原始数据(扭矩%和RPM)转换为剪切应力(SS)和剪切速率(SR)值。假设导叶(心轴)如其围绕圆筒一样进行运转(即,假设叶片之间的样品如固体块一样运动)。如下方式测定粘度:

表1中的粘度为1RPM下的SS/SR值。在测试中,RPM以对数模式以20个档级从0.5扫至200,每档级10秒。
SS(SR)函数用Casson方程进行拟合,SS=(Yn+(V0*SR)n)1/n,其中Y、V0和n是拟合参数。仅拟合向上弯曲曲线的单调增加部分(即其中SS(SR)的第一导数和第二导数是正的,通常在1RPM以上)。表1中报道的“屈服应力”(YS)是指Y参数。
如表1中所指出的,表现出至少0.3Pa的YS值的微结构化漱口液(制剂A和B)在提供均匀的在连续相中的粒子悬浮液方面是有效的,但是与漱口液的连续相浓度1.0g/cm3相比,包封的粒子的密度高(大约1.26g/cm3)。因此,如表1中所示的制剂A和B表现出有效水平的结冷胶和黄原胶材料以为包封的粒子的均匀悬浮液提供必要的屈服应力。相反,未用黄原胶和结冷胶结构化的配方C不能够实现长效的PLA粒子悬浮。
意外地,PLA斑点在与受控室温下或在加速条件(40℃和75%湿度)下老化三个月后保持完整。因此,如本公开中描述的包封的着色剂产生稳定性组合物,甚至在组合物具有高水含量时也是如此。

实施例2.含水凝胶状牙膏
具有漱口液口气清新组分和牙膏清洁组分的含水凝胶状牙膏通过混合表2中所述的成分制备。将0.3重量%PLA斑点添加至制剂。粘度和YS值如上所述那样确定。
如表2中所指出的,制剂的最终粘度在120,000至270,000cps的范围内。pH在7.0至9.0的范围内。
PLA斑点具有与牙膏的连续相类似的比重值。在三个月的储存时间段期间,保持了PLA斑点的均匀分布。在不受理论限制的情况下,通过低浓度的二氧化硅增稠剂和黄原胶提供均匀分布的斑点,所述二氧化硅增稠剂和黄原胶有助于制剂的静态屈服应力(YS)。在于加速条件下老化三个月之后PLA斑点在这种制剂中保持完整。

实施例3.含水牙膏和凝胶
将具有白色(TiO2)、蓝色(FD&C蓝色淀1、绿色(FD&C蓝铝色淀、D&C黄5色淀的组合和红色(FD&C 40/色淀)的PLA斑点配制在含水牙膏或凝胶制剂中以评估稳健性和/或这些制剂与不同成分的相容性。如表3中所示的制剂E-H通过混合指定的成分制备。如上所述那样确定粘度和YS,不同的是由于制剂的较高粘度而使用更小(V74)的导叶叶片以确保不超过仪器的最大扭矩能力。
如表3中所指出的,制剂的最终粘度在226,000至554,000cps的范围内并且pH在7.33至8.05的范围内。还如表中指出的那样,在受控室温和加速的高温条件下的三个月的老化评价产生可接受的化学和物理稳定的最终制剂。具体地,从PLA包封的斑点到牙膏媒介物中的颜色迁移在视觉上观察不到。此外,在储存时间段期间,保持了PLA斑点的均匀分布。这种均匀性很可能是由于黄原胶、羧甲基纤维素、二氧化硅增稠剂和/或甲基乙烯基醚/马来酸酐共聚物的存在,它们在制剂中提供了足够的粘度和静态屈服应力特性。

实施例4.具有PLA斑点的非含水牙膏
通过合并下表4中指定的成分配制含有增白剂、过氧化氢的非含水牙膏。粘度和YS值如在上文实施例3中所述那样确定。
如表中所指出的制剂“I”产生具有蓝色斑点样美观的部分半透明凝胶。第二制剂,制剂“J”产生具有蓝色斑点状美观的白色膏剂。最终粘度在382,000至683,000cps的范围内。pH在7.9与8.9之间。
在于25℃下、于60%相对湿度下和在加速老化条件(40℃,在75%相对湿度下)老化三个月之后,制剂展示出可接受的化学和物理稳定性。此外,在视觉上仅观察到从PLA包封的着色剂到牙膏媒介物中的极少颜色迁移。因此,PLA甚至在1%过氧化氢存在下也是稳定的。虽然仅评价了FD&C色1号铝色淀着色剂,但是也预期含有其他色淀和颜料颜色诸如D&C黄5铝色淀、FD&C红40铝色淀和氧化钛的PLA粒子因包封在PLA基质中而是适当稳定的。

Claims (19)
1.一种稳定性口腔护理组合物,其包含:

由可生物降解聚合物包封的着色剂。

包封在可生物降解聚合物中的着色剂。
