Abstract

本发明提供了一种脱细胞生物羊膜的制备方法,包括羊膜的清洗、化学处理和冷冻干燥。该方法是将新鲜羊膜用生理盐水清洗干净,洗去羊膜上的残留血液,再一次用过氧化氢、丙酮、十二烷基硫酸钠、酒精处理后,在‑70℃下的环境中深低温冷冻16‑25小时,然后将其放入冷冻干燥机中,按照设定好的冻干曲线进行冷冻干燥。辐照灭菌后得到脱细胞生物羊膜的成品。本发明使用过氧化氢、丙酮、十二烷基硫酸钠、酒精联合处理羊膜,可以有效地除去羊膜中的上皮细胞层、纤维膜细胞层和海绵层具有免疫原性的成分,同时保留了其基底膜和致密层,较好的保留了羊膜的生物活性和生物力学性能;另一方面通过冷冻干燥处理可以进一步的减少脱细胞生物羊膜的免疫原性,降低宿主细胞的免疫排斥反应。另外,本发明设计的脱细胞生物羊膜的制备方法简单易行,方便规模化生产操作。

Description

一种脱细胞生物羊膜的制备方法
技术领域
发明设涉及生物材料领域,具体设计一种同种异体植入性生物材料的制备方法。
背景技术
羊膜是胎膜的最内层,属于生物膜中的一种,是一类天然细胞外基质类生物衍生材料。羊膜由滋养细胞层分化而来,表面光滑,半透明,有韧性及弹性,厚度为0.02-0.50mm,主要由I、III 型纤维胶原蛋白构成,含有少量的VI、V 型胶原,其中含有多种生长因子,如成纤维细胞生长因子-2(fibroblast growth factor-2,FGF-2)、转移生长因子-β(transforming growth-β,TGF-β)、促进血管生成的血管内皮生长因子(VEGF),还含有纤维连接蛋白(fibronectin,FN) 等。羊膜不表达HLA-A、B、C 及DR 抗原或β2 微球蛋白,透明且无血管、淋巴和神经组织,其抗原性远低于普通组织。大量研究表明,羊膜具有良好的细胞相容性和组织相容性,并具有部位特异的组织再生能力,为一新型的生物衍生材料。
羊膜作为生物材料应用于基础及临床已经有多年的历史。早在20 世纪初期就有用含羊膜的胎膜供体移植于烧伤和溃疡创面的报道,随后在颅脑外科、腹腔外科、妇产科及眼科均有其应用的报道。具体来说其主要用途有:①用于眼睛表面疾病如角膜修复、先天性青光眼、角膜溃疡等;②用于修补鼓膜损伤;③用于外科手术后预防粘连;④用于烧伤后表面覆盖等。
然而,目前对羊膜的处理大多含有使用机械力刮除方法,使羊膜存在细胞残留、机械性能不足以及降解周期较短等缺陷,限制了其临床应用范围。根据临床报道,由于羊膜细胞的存在,在眼科,羊膜植片可能会发生植片排斥反应、植片脱落、移位溶解、感染等并发症,其安全性存在一定的风险。
发明内容
本发明的目的在于提供一种脱细胞生物羊膜的制备方法,摒弃了传统的机械力刮除的方法,采用了相对温和的化学处理方法,一方面能有效、彻底地除去羊膜中细胞残留等免疫原,另一方面还能较好的保留羊膜的生物活性和生物力学性能。
本发明的技术方案是:一种脱细胞生物羊膜的制备方法,包括羊膜的清洗、化学处理和冷冻干燥,其特征在于,该方法按以下步骤进行:
(1)将新鲜羊膜用生理盐水反复清洗,直至无残留血液;
(2)将步骤(1)所得的羊膜用过氧化氢溶液浸泡2小时;
(3)将步骤(2)所得的羊膜用分析纯的丙酮浸泡,水浴恒温震荡脱去脂肪成分;
(4)将步骤(3)所得的羊膜用十二烷基硫酸钠溶液浸泡16-20小时;
(5)将步骤(4)所得的羊膜用酒精浸泡16-20小时;
(6)将步骤(5)所得的羊膜用大量纯化水反复清洗至末道清洗液pH值为6.5-7.5;
(7)将步骤(6)所得的羊膜放入-70℃下的环境中冷冻16-25小时;
(8)将步骤(7)所得的羊膜放入冷冻干燥机中进行冷冻干燥;
(9)将步骤(8)所得的羊膜进行辐照灭菌。
根据本发明的脱细胞生物羊膜的制备方法,较好的是,所述过氧化氢的质量分数为5%-25%。
根据本发明的脱细胞生物羊膜的制备方法,在一个优选的实施方案中,所述水浴恒温震荡的温度为25-37℃。
步骤(4)中所述十二烷基硫酸钠的质量分数为0.1%-0.5%。
根据本发明的脱细胞生物羊膜的制备方法,较好的是,所述酒精的质量分数为75-85%。
根据本发明的脱细胞生物羊膜的制备方法,在一个优选的实施方案中,所述冷冻干燥时间为24小时。
根据本发明的脱细胞生物羊膜的制备方法,较好的是,所述辐照灭菌的剂量为20-30kGy。
本发明的技术原理:摒弃了机械力刮除的传统方法,采用了相对温和的化学处理的方法,可以有效地除去羊膜中的上皮细胞层、纤维母细胞层和海绵层,达到消除或减弱抗原性的目的,从而减弱羊膜在移植时引起的免疫排斥反应;同时又能有效地保留基底膜中的各种胶原蛋白,可以较好地保留羊膜的生物活性。另外采用辐照的方式灭菌,能够进一步降低羊膜的免疫原性,使植入的羊膜能够被宿主细胞所适应,大大降低免疫排斥反应。
本发明的有益效果在于将新鲜羊膜用生理盐水清洗干净,洗去羊膜上的残留血液,再依次用过氧化氢、丙酮、十二烷基硫酸钠、酒精处理,能够有效地去除羊膜中的细胞膜和细胞基质等免疫原,大大降低了羊膜的免疫原性,同时通过深低温冷冻和冷冻干燥处理,可以进一步的降低羊膜的免疫原性,一方面能够降低免疫排斥反应,另一方面还能较好的保留羊膜的生物活性和生物力学性能。此外,本发明设计的化学处理方法步骤简单移行,有利于实现规模化生产操作。
具体实施方式
为使本领域技术人员更好地理解本发明的技术方案,下面结合实施例对本发明作进一步详细描述。
实施例1
将新鲜羊膜先用大量生理盐水冲洗至无残留血迹,将其放入质量体积比为5:1的、质量分数为5%的过氧化氢溶液中浸泡2小时;然后羊膜放入质量体积比为5:1的分析纯的丙酮中,水浴恒温震荡2小时,脱去脂肪成分;再将所得的羊膜用质量体积比为5:1的、质量分数为0.15%的十二烷基硫酸钠溶液浸泡18小时;再用质量体积比为5:1的、质量分数为75%的酒精浸泡18小时脱去水分后用大量的纯化水反复冲洗羊膜直至末道清洗液的pH值在6.5-7.5之间。
将清洗完成后的羊膜平铺在不锈钢冻干盘上面,放入-70℃以下的环境中冷冻24小时,然后将其放入冷冻干燥机中进行冷冻干燥。冷冻干燥结束后在万级洁净环境中将其分割和包装,最后将其进行辐照灭菌(剂量为25kGy),即可得到脱细胞生物羊膜成品。
实施例2
将新鲜羊膜先用大量生理盐水冲洗至无残留血迹,将其放入质量体积比为6:1的、质量分数为5%的过氧化氢溶液中浸泡2小时;然后羊膜放入质量体积比为6:1的分析纯的丙酮中,水浴恒温震荡2小时,脱去脂肪成分;再将所得的羊膜用质量体积比为6:1的、质量分数为0.2%的十二烷基硫酸钠溶液浸泡18小时;再用质量体积比为5:1的、质量分数为75%的酒精浸泡18小时脱去水分后用大量的纯化水反复冲洗羊膜直至末道清洗液的pH值在6.5-7.5之间。
将清洗完成后的羊膜平铺在不锈钢冻干盘上面,放入-70℃以下的环境中冷冻24小时,然后将其放入冷冻干燥机中进行冷冻干燥。冷冻干燥结束后在万级洁净环境中将其分割和包装,最后将其进行辐照灭菌(剂量为25kGy),即可得到脱细胞生物羊膜成品。
实施例3

Claims (8)

1.一种脱细胞生物羊膜的制备方法,包括羊膜的清洗、化学处理和冷冻干燥,其特征在于,该方法按以下步骤进行:
(1)将新鲜羊膜用生理盐水反复清洗,直至无残留血液;
(2)将步骤(1)所得的羊膜用体积质量比为5-8:1比例的过氧化氢溶液浸泡2小时;
(3)将步骤(2)所得的羊膜用体积质量比为5-8:1比例的分析纯的丙酮浸泡,水浴恒温震荡脱去脂肪成分;
(4)将步骤(3)所得的羊膜用体积质量比为5-8:1比例的十二烷基硫酸钠溶液浸泡16-20小时;
(5)将步骤(4)所得的羊膜用体积质量比为5-8:1比例的酒精浸泡16-20小时;
(6)将步骤(5)所得的羊膜用大量纯化水反复清洗至末道清洗液pH值为6.5-7.5;
(7)将步骤(6)所得的羊膜放入-70℃下的环境中冷冻16-25小时;
(8)将步骤(7)所得的羊膜放入冷冻干燥机中进行冷冻干燥;
(9)将步骤(8)所得的羊膜进行辐照灭菌。
2.根据权利要求所述的脱细胞生物羊膜的制备方法,其特征在于,所述过氧化氢溶液的质量分数为5%-25%。
3.根据权利要求所述的脱细胞生物羊膜的制备方法,其特征在于,所述水浴恒温震荡的温度为25-37℃。
4.根据权利要求所述的脱细胞生物羊膜的制备方法,其特征在于,所述水浴恒温震荡的时间为2-4小时。
5.根据权利要求所述的脱细胞生物羊膜的制备方法,其特征在于,所述十二烷基硫酸钠的质量分数为0.1%-0.5%。
6.根据权利要求所述的脱细胞生物羊膜的制备方法,其特征在于,所述酒精的质量分数为75-85%。
7.根据权利要求所述的脱细胞生物羊膜的制备方法,其特征在于,所述冷冻干燥时间为24小时。
8.根据权利要求所述的脱细胞生物羊膜的制备方法,其特征在于,所述辐照灭菌的剂量为20-30kGy。
