Abstract
本发明公开了一种抗衰老组合物及其在精华液中的应用,抗衰老组合物为精选三种植物原料,巴戟天根提取物为君,甘草提取物为臣,姜黄提取物为佐使,经有效比例复配,通过清除自由基,抑制络氨酸酶活性,阻止黑色素形成,吸收紫外线,营养滋润肌肤的途径,实现其显著的抗衰老功效。并将该组合物添加到抗衰老精华液中,赋予精华液显著的抗衰老功能,兼具营养与滋润,减少皮肤皱纹,延缓皮肤衰老,令皮肤富有弹性。

技术领域
本发明涉及化妆品领域,具体涉及一种抗衰老组合物及其应用。
背景技术
皮肤时时刻刻都在老化,其弹性慢慢降低,并变得越来越薄、越来越脆弱和松弛,给人的直观印象就是长皱纹。皮肤老化一般有自然老化和光老化。前者主要指随着年龄增长,由机体内存在的如遗传、重力、内分泌及免疫功能等不可抗因素引起的老化;后者主要是由于紫外线的作用,加快了自然衰老的进程,而气候变化以及环境污染等一系列环境暴露对皮肤的光老化也有着直接的影响。抗衰老的途径,主要包括:保护皮肤免受外界环境刺激;清除细胞内的多余自由基;对皮肤细胞进行修复和补充营养。皮肤抗衰老是防止皮肤因时间推移而发生逐渐性的功能和器质性退性改变,表现为防止出现皱纹、干燥、起屑、松弛和色斑。抗衰老化妆品就是实现抗衰老功效的化妆品,是重要的功效型化妆品之一。
抗衰老化妆品大致分为三类,分别为保湿型化妆品、抗氧化化妆品以及生物活性化妆品。不同化妆品中的活性原料不同,作用机制一般可将活性原料分为保湿类、清除自由基类、吸收紫外线类以及细胞修复类。将有抗衰老功效的天然植物提取物添加到化妆品中,赋予产品良好的抗衰老功效,是抗衰老产品的一个趋势。
中国专利申请CN105310948A公开了一种抗衰老面膜,将25克白芨、25克白芨、25克甘草加水浸泡,加热煎煮1小时,后加入7克金银花、8克槐米、10克黄氏煎煮2小时,得浓缩液,最后加入40克蜂蜜、30克人参提取液、200克面粉和10克维生素C。该发明通过简单的煎煮工艺,中药的有效成分的浸提率低,造成浪费等问题。
目前,市售的抗衰老产品种类繁多,大都添加维生素类抗氧化剂,但这类抗氧化剂稳定性较差;或者添加一些有害的美白亮肤成分,长期使用,对人体有一定的损害。
发明内容
本发明的目的在于提供一种含有巴戟天根提取物、甘草提取物、姜黄提取物的抗衰老组合物,并将其应用到精华液中,使精华液安全,有效对抗自由基、延缓皮肤衰老。
本发明通过以下方案来实现:
一种抗衰老组合物,由以下重量份的物质组成:巴戟天根提取物0.01-3份、甘草提取物0.1-1.5份、姜黄提取物0.1-2份。
本发明提供的一种抗衰老组合物中,巴戟天根提取物的制备方法为:
(1)取新鲜巴戟天根,洗净干燥,粉碎成粗粒,粗粒用水煎煮提取2-3次,煎煮时间每次2-3h,合并两次滤液,静置2-4h,过滤,滤液备用;
(2)将步骤(1)所得滤液减压浓缩得稠浸膏,将稠浸膏进行喷雾干燥(入口温度180℃,出口温度70℃),将干燥后的浸膏粉碎成65-80目大小的细粉,即得所述巴戟天根提取物。
本发明提供的一种抗衰老组合物中,甘草提取物的制备方法为:
(1)将干燥甘草粉碎成平均粒径为20-70目的甘草原料粉末;
(2)将步骤(1)中得到的甘草原料粉末放入提取容器中,向提取容器中加入10-15倍的70-82%乙醇溶液,回流提取3-5次,每次2-3小时,浓缩,干燥,粉碎后即得到甘草提取物。
本发明提供的一种抗衰老组合物中,姜黄提取物的制备方法为:取姜黄粉碎成粗粉,加8-10倍量去离子水煎煮2次,第一次2小时,第二次1.5小时,过滤,合并滤液,浓缩至55-65℃测相对密度为1.30,冷却至室温,加入68-80%乙醇溶液,静置过夜,取上清液减压浓缩至相对密度为1.25的稠膏,70-80℃减压干燥,即得姜黄提取物。
本发明还提供抗衰老组合物在制备化妆品中的应用。
所述化妆品为精华液。
一种抗衰精华液包括以下成分:抗衰老组合物0.21-6.5份、增稠剂0.2-0.5份、保湿剂5-15份、防腐剂0.2-0.5份、去离子水至100份。
所述增稠剂为卡波姆、羧甲基纤维素纳、黄原胶中的至少一种。
所述保湿剂为甘油、丙二醇、山梨醇、1,3丁二醇中的至少一种;所述防腐剂为山梨酸钾、苯氧乙醇中的一种。
本发明抗衰精华液的制备方法,包括以下步骤:
(1)将增稠剂、保湿剂和去离子水加入搅拌锅中,加热至80-85℃,搅拌溶解完全;
(2)降温至40-45℃,加入抗衰老组合物、防腐剂,搅拌完全出料。
对本发明的植物进行功效介绍:
巴戟天又名巴戟、鸡眼藤、黑藤钻、糠藤、三角藤,为茜草科多年生攀援木质藤本植物,肉质根入药,是我国著名的四大南药之一。其主要化学成分有:糖类、蒽醌类、环烯醚萜苷类、有机酸类、微量元素、氨基酸和甾醇类等。近年来研究表明,巴戟天具有多方面的药理作用,如提高机体免疫力、抗肿瘤、抗氧化、强壮骨骼、抗炎镇痛、补血及促进造血干细胞增殖和分化等作用。其中抗氧化作用表现在对羟自由基及超氧阴离子自由基均有良好的清除作用。享有“南国人参”之称。
甘草又名乌拉尔甘草,是一种补益中草药。含有甘草酸等三萜衍生物,富含黄酮类、木质素、香豆素和多种氨基酸,其提取物具有防晒、消炎、抑菌、抗氧、润肤、有效清除超氧离子和羟基自由基的作用。可通过抑制酪氨酸酶和多巴色素互变酶的活性,阻碍5,6-二羟基吲哚的聚合,以此来阻止黑色素的形成。
姜黄又名郁金、宝鼎香,富含姜黄素、姜黄酮醇、挥发油、芳樟醇、豆甾醇、倍半萜类等化合物。有降血脂、抗肿瘤、抗炎、抗病原微生物、抗氧化等药理作用。其提取物的抗氧化表现为能清除氧自由基而发挥抗氧化活性。
本发明具有以下有益效果:(1)本发明的抗衰老组合物精选三种植物原料,巴戟天根提取物为君,甘草提取物为臣,姜黄提取物为佐使,经有效比例复配,通过清除自由基,抑制络氨酸酶活性,阻止黑色素形成,吸收紫外线,营养滋润肌肤的途径,实现其显著的抗衰老功效;(2)添加了小分子的甘油和丙二醇,进一步加强了精华液的补水锁水功能,丙二醇有促渗透作用,可促进营养成分在皮肤的吸收,从而促进胶原蛋白的生长,可增强皮肤的弹性;(3)本发明添加植物成分天然安全、环保,有效成分浸提率高,能赋予精华液显著的抗衰老功效。

实施例1~5中巴戟天根提取物、甘草提取物和姜黄提取物制备方法如下:
巴戟天根提取物的制备方法为:
(1)取新鲜巴戟天根,洗净干燥,粉碎成粗粒,粗粒用水煎煮提取2-3次,煎煮时间每次2-3h,合并两次滤液,静置2-4h,过滤,滤液备用;

本发明抗衰老组合物清除DPPH能力实验
方法和设备
植物组合物(自制);95%乙醇(国药集团化学试剂有限公司);DPPH(日本东京化成株式会社,分析纯);AB204-E电子分析天平(METTLE TOLEDO);UV-1600紫外分光光度计(METTLE TOLEDO)。
DPPH在无水乙醇和乙醇-水体系中呈现黑紫色,分别在517nm和525nm下有最大吸收。自由基清除剂能够使紫色逐渐退却,根据吸光度的变化测定出DPPH的含量和反应自由基清除剂对DPPH的清除能力。
精密称取DPPH3.00mg,用95%乙醇溶解并定容至100ml,DPPH的溶度为20mg/L,取4mlDPPH标准溶液,放置于试管中,加入1ml大麦提取物溶液,放置30min后,在517nm测定吸光度A。根据下列公式测定DPPH清除率;
DPPH的清除率=[1-(Ai-Aj)/Ac]×100%
Ai:提取液+4mlDPPH溶液的吸光度
Aj:提取液+4ml95%乙醇的吸光度
Ac:1ml95%乙醇+4mlDPPH溶液的吸光度
对DPPH清除能力以清除率表示,清除率越高表示清除能力越强,将大麦提取物样品用95%乙醇分别稀释到5%,10%,15%,20%,25%,检测各个浓度清除DPPH的能力。结果见图1。
从图1中可以看出本发明抗衰老对DPPH有清除能力,并且随着浓度增加清除度逐渐增高,浓度达到15%之后清除度趋于稳定,清除率达到85%。
本发明抗衰老组合物抑制络氨酸酶酶实验
方法和设备
本发明抗衰老组合物;ph=6.8磷酸缓冲溶液(PBS溶液)、氢氧化钠(国药集团化学试剂有限公司);络氨酸酶,Tyrosinase EC1.14.18.19(signa);L-DOPA(AcrosOrganics);AB204-E电子分析天平(METTLE TOLEDO);UV-1600紫外分光光度计(METTLE TOLEDO)。
络氨酸转化成黑色素的过程中,络氨酸酶的催化作用主要发生在络氨酸酶转化为多巴及多巴转化成多巴醌的过程,多巴醌为一种有色物质,在475nm处有吸收,可利用分光光度计测量含量。L-DOPA为底物,测定本发明抗衰老组合物的络氨酸酶抑制活性。
表二待测样品成分
按照表二所示准确吸取本发明抗衰老组合物提取液、0.15%的多巴溶液,充分混合均匀,在30℃水浴中孵育10分钟后加入300μ/mL的络氨酸酶溶液,继续培育十分钟,立即在475nm测定其吸光值。
络氨酸酶抑制率(I)的计算公式如下:
I=[(A-B)-(C-D)]/[A-B]×100%
A:未加抗衰老组合物加酶的混合液所测的吸光度A(A);
B:未加抗衰老组合物且未加加酶的混合液所测的吸光度A(B);
C:加抗衰老组合物和加酶的混合液的、所测的吸光度A(C);
D:加抗衰老组合物未加酶的混合液所测的吸光度A(D)。
将抗衰老组合物样品用PBS分别稀释到5%,10%,15%,20%,25%检测各个浓度抑制络氨酸酶活性。结果见图2
有图2可见抗衰老组合物浓度在5%到20%的时候,抗衰老组合物抑制络氨酸酶活性随着计量的增长而增长,当抑制络氨酸酶活性溶度达到20%时,络氨酸酶抑制率可达到85%。
对实施例1-5和对比例1-6制备的产品进行试用检测效果,同时以一款市售的宣称具有良好抗衰老功效的精华液为对照组。测试对象:各实验组均为20名女性(25-50岁);测试区域:前臂内侧和鱼尾纹区域;使用:每天两次,持续使用4周;测试项目:皮肤弹性、皮肤水嫩、皱纹深度(鱼尾纹区域)。试用结果见下表二。
由表二可见实施例1~5皮肤弹性、水嫩和皱纹深度改善明显的人数明显多于对比例1~6和对照组,说明本发明抗衰老组合物所制精华液有显著的改善皮肤弹性、皮肤水嫩、和皱纹深度的效果;对比例1皮肤弹性明显改善8人,皮肤水嫩明显改善6人,皱纹深度明显改善5人,明显低于其他实施例和对比例,说明本发明抗衰老组合物为抗衰老精华液主要有效成分;对比例2的抗衰老精华液未加入巴戟天提取物,其皮肤弹性、水嫩和皱纹深度改善明显的人数分别为10人、11人、11人远低于实施例1皮肤弹性、水嫩和皱纹深度改善明显的人数;对比例3和对比例4分别为未添加甘草提取物和姜黄提取物,其皮肤弹性、水嫩和皱纹深度改善明显的人数均明显低于实施例1,对比例2~4与实施例1对比可见在本发明抗衰老组合物中,巴戟天提取物、甘草提取物和姜黄提取物协同增效。

1.一种抗衰老组合物,其特征在于,所述组合物由以下重量份的物质组成:
巴戟天根提取物0.01-3份
甘草提取物0.1-1.5份
姜黄提取物0.1-2份。
2.根据权利要求1所述的一种抗衰老组合物,其特征在于,所述巴戟天根提取物的制备方法为:
(1)取新鲜巴戟天根,洗净干燥,粉碎成粗粒,粗粒用水煎煮提取2-3次,煎煮时间每次2-3h,合并两次滤液,静置2-4h,过滤,滤液备用;
(2)将步骤(1)所得滤液减压浓缩得稠浸膏,将稠浸膏进行喷雾干燥(入口温度180℃,出口温度70℃),将干燥后的浸膏粉碎成65-80目大小的细粉,即得所述巴戟天根提取物。

4.根据权利要求1所述的一种抗衰老组合物,其特征在于,所述姜黄提取物的制备方法为:取姜黄粉碎成粗粉,加8-10倍量去离子水煎煮2次,第一次2小时,第二次1.5小时,过滤,合并滤液,浓缩至55-65℃测相对密度为1.30,冷却至室温,加入68-80%乙醇溶液,静置过夜,取上清液减压浓缩至相对密度为1.25的稠膏,70-80℃减压干燥,即得姜黄提取物。
5.根据权利要求1-4任一项所述的抗衰老组合物在制备化妆品中的应用。
6.根据权利要求5所述的应用,其特征在于,所述化妆品为精华液。
7.一种抗衰精华液,其特征在于,所述精华液包括以下成分:

增稠剂0.2-0.5份
保湿剂5-15份
防腐剂0.2-0.5份
去离子水至100份。
8.根据权利要求7所述的一种抗衰精华液,其特征在于,所述增稠剂为卡波姆、羧甲基纤维素纳、黄原胶中的至少一种。
9.根据权利要求7所述的一种抗衰精华液,其特征在于,所述保湿剂为甘油、丙二醇、山梨醇、1,3丁二醇中的至少一种;所述防腐剂为山梨酸钾、苯氧乙醇中的一种。
10.一种如权利要求7所述的抗衰精华液的制备方法,其特征在于,包括以下步骤:
(1)将增稠剂、保湿剂和去离子水加入搅拌锅中,加热至80-85℃,搅拌溶解完全;
(2)降温至40-45℃,加入抗衰老组合物、防腐剂,搅拌完全出料。
