背景技术
中药有效成分的提取分离是中药生产过程中的关键环节，其工艺特点、工艺流程的选择和设备配置都直接关系到被提取有效成分的数量和质量，从而影响到产品质量的稳定性和有效性等。因此探明中药提取、分离技术的机制、优化提取工艺参数，明确应用前景等逐渐成为中药生产和研究的重点内容。随着我国中药现代化进程的不断推进，中药提取分离新技术的基础和应用研究十分活跃，并取得了长足的进步。传统的中药有效成分的提取方法有煎煮法、回流提取法、超声辅助提取技术、超临界流体萃取技术、微波辅助萃取技术，但这些方法由于大多数中药有效成分含量低，难于富集，许多有效成分热敏，易水解等因素，使得传统分离与富集技术获得的产品收率低，同时成本高、耗时长、工艺复杂、消耗溶剂多、能耗高、成本大、一些溶剂毒副作用较大等缺点。
提取植物样品的提取过程为两个过程，一个是植物样品浸润、其中溶质溶解的过程，另一个是溶质与溶液之间的扩散过程。超高压能够快速、高效地提取天然产物有效成分，主要是超高压能改变基质材料的组织结构，大大减小目标成分的扩散阻力，同时超高压力差是目标成分扩散的传质动力，其独特的提取机理可以从升压、保压和卸压3个阶段加以说明：升压阶段使压力在极短的时间内(一般小于1 min)由常压升到几百兆帕，而细胞内部的压力却很小(1个大气压)，由于渗透压差极大，在渗透过程中溶剂易形成湍流，使细胞边界层变薄、细胞膜发生疏松、破碎等结构变化，增大固液接触面积，减小有效成分由细胞内部向外扩散的传质阻力。升压阶段引起压力的快速变化，可以改变体系的体积，进而推动了化学平衡发生移动。快速渗透到细胞内部的溶剂与有效成分充分接触，在较短的时间内实现有效成分溶解平衡，因此保压阶段一般在几分钟内完成。卸压在几秒内完成使组织细胞的压力从几百兆帕迅速减小为零，溶解了有效成分的溶液在超高反向压力差的作用下形成强大的湍流。卸压时间越短，细胞内流体扩散时产生的冲击力越强，湍动效应越强，有效成分扩散的传质阻力越小，溶解了有效成分的溶剂快速转移到细胞外，达到快速、高效提取的目的。基于超高压技术的上述原理，与传统的浸渍法、渗漉法、煎煮法、热回流提取法、索氏提取法等常规提取方法，以及近年来研究和开发的超声提取法、微波提取法、超临界流体萃取法等方法相比，超高压提取技术具有独特的优势：提取时间短、能耗低、大分子物质溶出少、有效成分提取率高以及可避免热效应引起的有效成分结构改变、损失和生理活性降低等，同时由于超高压提取是在密闭环境中进行，没有溶剂挥发，不会造成环境污染。井凤、刘峰等在《超高压技术在中药有效成分提取中的应用》中对超高压提取的原理、特点、在中药提取中的应用以及技术存在问题进行分析研究，为在实际生产中更好的利用超高压技术提供理论依据。
发明内容
本发明目的是为了有效减少中药提取时间，增加提取效率，稳定有效成分，防止资源浪费，提供一种祛病延年长寿酥及其制备方法，生产出能够祛病延年、口感适宜、食用方便的中药。
本发明是通过以下技术方案实现的：
一种祛病延年长寿酥，它是由下述组分及其重量份制成
茯苓50-55、陈酒100-120、蜂蜜100-120、当归10-12、川芎10-12、白芍10-12、熟地10-12、黄芪30-35、人参20-23、白术10-12、炙甘草25-30、生姜10-12、红枣30-35、何首乌30-35、杜仲10-12、丹参10-12。
所述的一种祛病延年长寿酥，是由下列步骤制成的：
1）、将茯苓清洗干净，研磨成粉，文火炒制后加入陈酒密封2-3天；
2）、将杜仲清洗干净，切片后加入质量分数为30-32%的食盐，并加入15-20倍量的30-50%的乙醇溶液，密封浸泡1-2天，干燥后粗粉碎；
3）、分别将当归、川芎、白芍、黄芪、人参、白术、炙甘草、生姜、何首乌、丹参润洗干净，粗粉碎后混合加入料液比为1:35-50的70-75%乙醇溶液，于200-230MPa条件下提取5-7分钟，过滤分别收集滤渣和滤液；
4）、将步骤3所得滤渣与步骤2所得杜仲混合加入料液比为1:10-12的70-75%乙醇溶液，于600-650MPa压强条件下提取5-7分钟，过滤收集滤液，并将所得滤液与步骤3所得滤液混合，回收乙醇，并将其浓缩成浸膏状；
5）、将熟地、红枣混合，捣碎研粉，过80-100目筛，再与步骤1所得物、步骤4所得浸膏、蜂蜜混合，提炼成酥。
