背景技术
我国是中草药的发源地,它在中华人类发展健康史上发挥着重要作用,随着时代的发展和社会的进步,科学创新,中草药的应用又进入了一个新的领域空间。近年来,我国的养兔业发展迅速,而发病率随着生产规模的扩大而随之升高,使生产性能降低,影响了养殖业的健康发展。为解决发病率高的问题,抗生素大量应用,使兔肉生长缓慢、品质下降、生产性能出现故障,同时对人体健康造成了伤害。
发明内容
为增强肉兔的免疫力,缩短其生长周期,本发明提供一种液体中草药生物饲料添加剂及其制备方法。
本发明的目的是以下述方式实现的:
一种液体中草药生物饲料添加剂,由中草药和专用菌种制备而成,所述中草药包括以下重量份数的原料药:连翘20-40份,黄连5-15份,黄芪10-20份,黄柏5-15份,苦参8-15份,山楂20-40份,桂皮2-10份,菜菔子10-30份,丁香2-8份,茵陈10-20份,车前5-15份,扁畜10-20份,薄荷2-10份,鱼腥草10-20份,金银花20-40份,淫羊藿10-20份,青蒿10-20份,艾蒿10-20份,艾叶5-15份,射干10-20份,蛤蟆草8-15份,甘草5-10份;所述专用菌种是从健康家兔盲肠内分离出的有益活性物质。
所述中草药包括以下重量份数的原料药:连翘30份,黄连10份,黄芪15份,黄柏10份,苦参12份,山楂30份,桂皮6份,菜菔子20份,丁香6份,茵陈15份,车前12份,扁畜15份,薄荷6份,鱼腥草15份,金银花30份,淫羊藿15份,青蒿15份,艾蒿15份,艾叶10份,射干15份,蛤蟆草12份,甘草10份。
上述液体中草药生物饲料添加剂的制备方法,具体步骤如下:
(1)专用菌种的选择培养:采集健康家兔盲肠内容物,采用固体MRS肉汤培养基对盲肠内容物中的细菌分别进行有氧培养和厌氧培养,得到需氧培养菌和厌氧培养菌;
(2)将中草药洗净,晒干或风干,将各味中草药的根、茎、叶、果、皮、籽和花进行归属分类,在5-20℃下存放备用;
(3)将不同中草药的根、茎、叶、果、皮、籽和花粉粹成60目粉面,分别放进砂锅内泡制10-30分钟,再进行熬制,花叶类、皮果籽类和根茎类分开熬制,花叶类熬制时间为20-30分钟,皮果籽类熬制时间为30-40分钟,根茎类熬制时间为40-60分钟,直至熬制成的中草药药液浓度为波美度2-3度,pH值5-6.5;
(4)将熬制成的中草药药液放在阴凉处自然降温沉淀,先用双层纱布过滤一遍,将药渣过滤掉,得到初滤中草药药液,再将初滤中草药药液用多层纱布过滤3-5遍,得到精滤中草药药液,然后将得到的精滤中草药药液置于消毒室内,于5-15℃下进行紫外线杀菌30-60分钟;
(5)将步骤(1)培养成的专用菌种接种到步骤(4)得到的中草药药液中进行培养,培养温度为5-10℃,培养5-7天后,培养温度升至15-20℃,继续培养5-8天,整个培养过程要求暗光培养。
所述需氧培养菌包括大肠杆菌属、粪球菌属、肠球菌属、显核菌属、短乳杆菌属、布氏乳杆菌属、蜡状芽孢杆菌、唾液乳杆菌和柠檬酸细菌属,厌氧培养菌包括梭杆菌属、双歧杆菌属、韦荣氏球菌属、乳杆菌属、明串球菌属和乳球菌属。
所述固体MRS肉汤培养基为:牛肉膏10g、蛋白陈10g、酵母浸粉5g、葡萄糖20g、吐温801mL、磷酸氢二钾2g、乙酸钠4g、柠檬酸氢二铵2g、水合硫酸镁0.2g、水合硫酸锰0.2g、琼脂20g、半胱氨酸0.3g、蒸馏水1000mL。
所述厌氧培养采用干燥器法:计算好容器体积,根据其体积称取焦性没食子酸和配制氢氧化钠溶液(1000mL空间用焦性没食子酸10g、10%氢氧化钠溶液100mL);然后将氢氧化钠溶液倒入干燥器底部,盛有焦性没食子酸的平皿轻轻浮于液面之上,放好隔板,将接种好的平板置于隔板上,并在隔板上点燃一支蜡烛,把干燥器盖盖上密封。轻轻摇动干燥器,使焦性没食子酸和氢氧化钠溶液混合作用。待蜡烛熄灭后置于37℃温箱2-3d,取出观察。
本发明选用原料药的功效简述如下:
连翘:性味苦、凉,归经:入心、肝、胆经,治温热,丹毒,斑疹,痈疡肿毒,瘰疬,小便淋闭,主治外感风热,温病初起,热病高热、斑疹,疮疡肿毒,瘰疬,丹毒、乳痈等病症。
黄连:清热燥湿,泻火解毒,清肠止泻,平肝明目。现代药理研究认为:1、具有广谱抗菌作用;抑制细菌、真菌、病毒、真菌、抗原虫;2、止泻,健胃,提高胃液,胆汁分泌,促进胃肠运动;3、利胆,增加胆汁形成和促进胆红素排泄;4、免疫调节,对白血病细胞有抑制,抗血小板聚集。
黄芪:性甘、温;归肺、脾经,具有增强免疫系统、延缓衰老,抗氧化、增强心血管系统、抗血栓、抗病毒、抗炎及镇痛等作用;益气升阳,固表止汗,利水消肿,托毒生肌;主治一切气虚血亏之证,如脾虚泄泻,肺虚咳嗽,脱肛,子宫下垂,自汗,盗汗,水肿,血痹,痈疽难溃或久溃不敛等。
黄柏:性寒、味苦,归肾、膀胱经,清热燥湿,泻火解毒,除骨蒸清虚热,用于湿热泻痢,黄疸,带下,热淋,脚气,痿辟,骨蒸劳热,盗汗,遗精,疮疡肿毒,湿疹瘙痒,黄柏用于阴虚火旺,盗汗骨蒸。
苦参:性寒,有清热燥湿,杀虫的功效,苦参醚提物及醇提物对金黄色葡萄球菌有较强的抑菌作用;苦参水浸剂对堇色毛癣菌、同心性毛癣菌、许兰毛癣菌、奥杜盎小芽孢癣菌等有抑制作用。
山楂:性酸、甘,微温;归脾、胃、肝经,具有促消化、降压、降脂、抗氧化等作用;消食健胃,行气散淤;主治饮食积滞,脘腹胀痛,泄泻痢疾等。
桂皮:味辛甘、性热,入肾、脾、膀胱经;有补元阳,暖脾胃,除积冷,通脉止痛和止泻的功效;主治命门火衰、肢冷脉微、亡阳虚脱、腹痛泄泻、寒疝、腰膝冷痛、经闭、阴疽流注、虚阳浮越之上热下寒等症。
莱菔子:又名萝卜子、萝白子、菜头子等,为十字花科植物萝卜的成熟种子,入脾、胃、肺经,能消食除胀,功效显著,有“冲墙倒壁”之称,可用于用于治疗饮食停滞、脘腹胀痛、大便秘结、积滞泻痢、痰壅喘咳等。
丁香:味辛,性温,温中、暖肾、降逆。治呃逆、呕吐、反胃、痢疾、心腹冷痛、痃癖、疝气、癣症,治疗牙痛、支气管炎,神经痛、胃酸,抗呼吸系统及泌尿系统感染,减轻痢疾所造成之不适与疼痛,改善衰弱体质与贫血,催情(性无能、冷感),驱虫。促进血液循环,治疗皮肤溃疡及伤口发炎,治疗疥癣,改善粗糙肌肤。
茵陈:利湿退黄用于黄疸;现代常用于急、慢性病毒性黄疸型肝炎、肝硬化等;
车前:甘,寒,归手太阳,阳明经,利水渗湿中药,主治:主小便不利、淋浊带下、水肿胀满、暑湿泻痢、目赤障翳、痰热咳喘。
扁畜:性味甘,平。归膀胱经。功效是清湿热,利小便。主治热淋涩痛,霍乱,黄疸,产后淤痛,痔疮肿痛,妇女阴蚀。
薄荷:味辛,性凉,归肺、肝经,有发散风热,清利咽喉,透疹解毒,疏肝解郁和止痒等功效,适用于感冒发热、头痛、咽喉肿痛、无汗、风火赤眼、风疹、皮肤发痒、疝痛、下痢及瘰疬等症,外用有轻微的止痛作用,用于神经痛等。
鱼腥草:性辛,微寒;归肺、膀胱、大肠经,具有抗菌、抗病毒、免疫增强、利尿作用;清热解毒,排脓消痈,利尿通淋;主治肺痈吐脓,肺热咳喘。
金银花:清热解毒、凉散风热。其中金银花挥发性成分可清热解暑,并清头目。
淫羊藿:辛、甘,温。归肝、肾经。补肾阳,强筋骨,祛风湿。用于阳痿遗精,筋骨痿软,风湿痹痛,麻木拘挛;更年期高血压。
青蒿:本品为菊科植物青蒿的全草,性苦、寒,有清热解暑、除蒸、截疟等功效。近年来在鸡球虫病的防治上,取得了较好的效果。陈蒲丹等研究表明,青蒿及其提取物体外能有效的抑制球虫卵囊孢子化,青蒿注射液和青蒿颗粒剂的抗球虫指数分别为164.69和155.86,能有效地增加感染鸡的成活率和相对增重率,并能减轻球虫感染引起的盲肠病变,减少血便,具有较好的抗球虫效果。本品并能增强免疫功能,增强巨噬细胞吞噬能力,提高淋巴细胞的转化率,增加脾脏重量。
艾蒿:味苦、辛,性温。归肝、脾、肾经,芳香、苦燥辛散,能理气血、温经脉、逐寒湿、止冷痛,为妇科要药。用治脘腹冷痛,经寒不调,宫冷不孕等症状。
艾叶:苦辛,温,入脾、肝、肾经。理气血,逐寒湿;温经,止血,安胎。治心腹冷痛,泄泻转筋,久痢,吐衄,下血,月经不调,崩漏,带下,胎动不安,痈疡,疥癣。
射干:苦,寒,入肺、肝经,能清能降,功能降火解毒,散血肿,利咽喉,又能清肺散结,脾胃虚寒、气血虚损、病无实热者忌用。
蛤蟆草:性 味:性凉,味苦、辛,主治:清热,解毒,凉血,利尿,用于咽喉肿痛、支气管炎、肾炎水肿、痈肿;外治乳腺炎、痔疮肿痛、出血。
甘草:性平,味甘,归十二经,有解毒、祛痰、止痛、解痉以至抗癌等药理作用。在中医上,甘草补脾益气,滋咳润肺,缓急解毒,调和百药。临床应用分“生用”与“蜜炙”之别。生用主治咽喉肿痛,痛疽疮疡,胃肠道溃疡以及解药毒、食物中毒等;蜜炙主治脾胃功能减退,大便溏薄,乏力发热以及咳嗽、心悸等。
相对于现有技术,将本发明提供的液体中草药生物饲料添加剂添加到肉兔的饲料中,不再使用其他药物,肉兔养殖期间没有病害发生,与没有使用液体中草药生物饲料添加剂的肉兔相比,成活率由93%提高到了99.8%,肉兔生长周期短,肉质好,医药成本也从原来的1元/只降低到了0.06元/只。本发明配方科学合理,相辅相成,将这些液体中草药生物饲料添加剂与专用菌种结合使用,具有抗菌消毒、健胃消食、促进消化吸收、增强机体免疫力的功效,还能够提高饲料利用率和肉兔成活率,改善肉兔肉质风味。
具体实施方式
专用菌种是从健康家兔盲肠内分离出的有益活性物质,菌液在与其乳酸菌、酵母菌等有益菌在无有害细菌环境下,接种到基质中进行培养,在15-20度温度条件下7天培养成功。将培养成功的有益菌种存放在5-10度环境中,可做生产备用。
通过对培养基及培养条件的筛选,筛选出适合家兔用的益生菌菌种和适用范围广的益生菌培养基,为研制兔专用益生菌奠定基础。
1材料与方法
1.1家兔盲肠内容物的采集
实验兔为来自舞钢盛元牧业有限公司硕丰养殖基地的健康法国伊拉兔青年兔5只,经临床剖检无任何病变,采集其盲肠内容物。
1.2培养基的制备
本试验采用固体MRS肉汤培养基及其特殊培养基。配制方法:准确称量下列各种成分,补水至原来体积,用1mol/L NaOH溶液调整pH至7.2-7.4,115℃灭菌30min。
普通培养基MRS):牛肉膏10g、蛋白陈10g、酵母浸粉5g、葡萄糖20g、吐温801mL、磷酸氢二钾2g、乙酸钠4g、柠檬酸氢二铵2g、水合硫酸镁0.2g、水合硫酸锰0.2g、琼脂20g、半胱氨酸0.3g、蒸馏水1000mL。
1.3盲肠内容物细菌的培养
1.3.1有氧培养 将接种好的平板置于37℃恒温箱种培养24~48h.
1.3.厌氧培养 采用干燥器法。计算好容器体积,根据其体积称取焦性没食子酸和配制氢氧化钠溶液(1000mL空间用焦性没食子酸10g、10%氢氧化钠溶液100mL);然后将氢氧化钠溶液倒入干燥器底部,盛有焦性没食子酸的平皿轻轻浮于液面之上,放好隔板,将接种好的平板置于隔板上,并在隔板上点燃一支蜡烛,把干燥器盖盖上密封。轻轻摇动干燥器,使焦性没食子酸和氢氧化钠溶液混合作用。待蜡烛熄灭后置于37℃温箱2-3d,取出观察。
综合鉴定结果
需氧培养菌:大肠杆菌属,粪球菌属,肠球菌属,显核菌属,短乳杆菌属,布氏乳杆菌属,蜡状芽孢杆菌,唾液乳杆菌,柠檬酸细菌属,其他未定。
厌氧培养菌:梭杆菌属,双歧杆菌属,韦荣氏球菌属,乳杆菌属,明串球菌属,乳球菌属,其他未定。

上述液体中草药生物饲料添加剂的制备方法,具体步骤如下:
(1)专用菌种的选择培养:采集健康家兔盲肠内容物,采用固体MRS肉汤培养基对盲肠内容物中的细菌分别进行有氧培养和厌氧培养,得到需氧培养菌和厌氧培养菌;

(4)将熬制成的中草药药液放在阴凉处自然降温沉淀,先用双层纱布过滤一遍,将药渣过滤掉,得到初滤中草药药液,再将初滤中草药药液用多层纱布过滤4遍,得到精滤中草药药液,然后将得到的精滤中草药药液置于消毒室内,于8℃下进行紫外线杀菌40分钟;
(5)将步骤(1)培养成的专用菌种接种到步骤(4)得到的中草药药液中进行培养,培养温度为6℃,培养6天后,培养温度升至16℃,继续培养6天,整个培养过程要求暗光培养。
厌氧培养采用干燥器法:计算好容器体积,根据其体积称取焦性没食子酸和配制氢氧化钠溶液(1000mL空间用焦性没食子酸10g、10%氢氧化钠溶液100mL);然后将氢氧化钠溶液倒入干燥器底部,盛有焦性没食子酸的平皿轻轻浮于液面之上,放好隔板,将接种好的平板置于隔板上,并在隔板上点燃一支蜡烛,把干燥器盖盖上密封。轻轻摇动干燥器,使焦性没食子酸和氢氧化钠溶液混合作用。待蜡烛熄灭后置于37℃温箱3d,取出观察。

一种液体中草药生物饲料添加剂,由中草药和专用菌种制备而成,中草药包括以下重量份数的原料药:连翘24g,黄连7g,黄芪12g,黄柏7g,苦参10g,山楂24g,桂皮4g,菜菔子14g,丁香4g,茵陈12g,车前7g,扁畜12g,薄荷4g,鱼腥草12g,金银花24g,淫羊藿12g,青蒿12g,艾蒿12g,艾叶7g,射干12g,蛤蟆草10g,甘草6g;所述专用菌种是从健康家兔盲肠内分离出的有益活性物质。
上述液体中草药生物饲料添加剂的制备方法,具体步骤如下:
(1)专用菌种的选择培养:采集健康家兔盲肠内容物,采用固体MRS肉汤培养基对盲肠内容物中的细菌分别进行有氧培养和厌氧培养,得到需氧培养菌和厌氧培养菌;
(2)将中草药洗净,晒干或风干,将各味中草药的根、茎、叶、果、皮、籽和花进行归属分类,在12℃下存放备用;
(3)将不同中草药的根、茎、叶、果、皮、籽和花粉粹成60目粉面,分别放进砂锅内泡制20分钟,再进行熬制,花叶类、皮果籽类和根茎类分开熬制,花叶类熬制时间为25分钟,皮果籽类熬制时间为35分钟,根茎类熬制时间为50分钟,直至熬制成的中草药药液浓度为波美度2.5度,pH值5.8;
(4)将熬制成的中草药药液放在阴凉处自然降温沉淀,先用双层纱布过滤一遍,将药渣过滤掉,得到初滤中草药药液,再将初滤中草药药液用多层纱布过滤5遍,得到精滤中草药药液,然后将得到的精滤中草药药液置于消毒室内,于10℃下进行紫外线杀菌45分钟;
(5)将步骤(1)培养成的专用菌种接种到步骤(4)得到的中草药药液中进行培养,培养温度为7℃,培养7天后,培养温度升至18℃,继续培养7天,整个培养过程要求暗光培养。

一种液体中草药生物饲料添加剂,由中草药和专用菌种制备而成,中草药包括以下重量份数的原料药:连翘26g,黄连8g,黄芪13g,黄柏8g,苦参11g,山楂26g,桂皮5g,菜菔子16g,丁香5g,茵陈13g,车前8g,扁畜13g,薄荷5g,鱼腥草13g,金银花26g,淫羊藿13g,青蒿13g,艾蒿13g,艾叶8g,射干13g,蛤蟆草11g,甘草6.5g;所述专用菌种是从健康家兔盲肠内分离出的有益活性物质。
上述液体中草药生物饲料添加剂的制备方法,具体步骤如下:

(3)将不同中草药的根、茎、叶、果、皮、籽和花粉粹成60目粉面,分别放进砂锅内泡制25分钟,再进行熬制,花叶类、皮果籽类和根茎类分开熬制,花叶类熬制时间为28分钟,皮果籽类熬制时间为38分钟,根茎类熬制时间为55分钟,直至熬制成的中草药药液浓度为波美度2.8度,pH值6;
(4)将熬制成的中草药药液放在阴凉处自然降温沉淀,先用双层纱布过滤一遍,将药渣过滤掉,得到初滤中草药药液,再将初滤中草药药液用多层纱布过滤4遍,得到精滤中草药药液,然后将得到的精滤中草药药液置于消毒室内,于12℃下进行紫外线杀菌50分钟;
(5)将步骤(1)培养成的专用菌种接种到步骤(4)得到的中草药药液中进行培养,培养温度为8℃,培养6天后,培养温度升至19℃,继续培养8天,整个培养过程要求暗光培养。

(4)将熬制成的中草药药液放在阴凉处自然降温沉淀,先用双层纱布过滤一遍,将药渣过滤掉,得到初滤中草药药液,再将初滤中草药药液用多层纱布过滤3遍,得到精滤中草药药液,然后将得到的精滤中草药药液置于消毒室内,于15℃下进行紫外线杀菌60分钟;
(5)将步骤(1)培养成的专用菌种接种到步骤(4)得到的中草药药液中进行培养,培养温度为10℃,培养5天后,培养温度升至20℃,继续培养7天,整个培养过程要求暗光培养。

利用本发明制备的液体中草药生物饲料养育肥兔的试验:
试验选取28日龄断奶兔、伊拉肉兔、商品兔共3000只,将其随机分为三组,每组公母比例为1:1,每组数量均为1000只,在同一个农场,同等环境同等条件下进行,每笼装四只,预试时间为2天,正式试验时间为30天,试验期间管理人员管理方法不变,饲料不变,自由饮水,每天打扫卫生一次,3天消毒一次,室温5-25℃,湿度50-75%,整个试验期间开窗透风,实验组在饮水中分别添加不同比例的液体中草药生物饲料1%-2%,饲料按日龄阶段增加,饲喂次数每天两次,验收时将食盒的余料收起称重,做好填写记录。
小结:
1.试验期内发病死亡率2%,验收实数980只,成活率98%;
2.试前总重量1350斤,单只体重1.35±斤,验收日总重为4508斤,单只平均体重4.6斤,平均每只每天增重0.108±斤;
3.试验期内实耗饲料7700斤,实增重量3158斤,料肉比为2.438:1;
验收数据统计:育肥成活率98%,日增重高于0.1±斤,料肉比低于2.5±1。
小结:
1.试验期内发病死亡率和淘汰率共3%,验收实数970只,成活率97%;
2.试前总重量1362±斤,单只体重1.36±斤,验收日总重为4410±斤,单只平均体重4.55斤,平均每只每天增重0.106±斤;
3.试验期内实耗饲料7760±斤,实增重量3048±斤,料肉比为2.55±:1;
验收数据统计:育肥成活率97%,日增重高于0.1±斤,料肉比高于2.5±1。
小结:
1.试验期内发病死亡率5%,验收实数950只,成活率95%;
2.试前总重量1350±斤,单只体重1.35±斤,验收日总重为4038±斤,单只平均体重4.25斤,平均每只每天增重0.095±斤;
3.试验期内实耗饲料7800±斤,实增重量2688±斤,料肉比为2.9±:1;
4.验收数据统计:育肥成活率95%,日增重高于0.1±斤,料肉比高于2.5±1。
硕丰绿色养兔生产标准化管理与液体中草药生物饲料保健流程:
小结:在饮水中添加液体中草药生物饲料,种公兔精子密度明显提高,个体增大。
小结:在饮水中添加液体中草药生物饲料,种母兔发情明显,受孕率高,母兔机体免疫力提高。
小结:在不同的生长发育阶段,使用液体中草药生物饲料,仔兔从出生到出栏只需60日龄就可以达到出栏体重,育肥期成活率98%,出栏合格率99.8%以上。
以上所述的仅是本发明的优选实施方式,应当指出,对于本领域的技术人员来说,在不脱离本发明整体构思前提下,还可以作出若干改变和改进,这些也应该视为本发明的保护范围。
Claims (6)
1.一种液体中草药生物饲料添加剂,其特征在于:由中草药和专用菌种制备而成,所述中草药包括以下重量份数的原料药:连翘20-40份,黄连5-15份,黄芪10-20份,黄柏5-15份,苦参8-15份,山楂20-40份,桂皮2-10份,菜菔子10-30份,丁香2-8份,茵陈10-20份,车前5-15份,扁畜10-20份,薄荷2-10份,鱼腥草10-20份,金银花20-40份,淫羊藿10-20份,青蒿10-20份,艾蒿10-20份,艾叶5-15份,射干10-20份,蛤蟆草8-15份,甘草5-10份;所述专用菌种是从健康家兔盲肠内分离出的有益活性物质。
2.根据权利要求1所述的液体中草药生物饲料添加剂,其特征在于:所述中草药包括以下重量份数的原料药:连翘30份,黄连10份,黄芪15份,黄柏10份,苦参12份,山楂30份,桂皮6份,菜菔子20份,丁香6份,茵陈15份,车前12份,扁畜15份,薄荷6份,鱼腥草15份,金银花30份,淫羊藿15份,青蒿15份,艾蒿15份,艾叶10份,射干15份,蛤蟆草12份,甘草10份。
3.如权利要求1或2所述的液体中草药生物饲料添加剂的制备方法,其特征在于:具体步骤如下:
(1)专用菌种的选择培养:采集健康家兔盲肠内容物,采用固体MRS肉汤培养基对盲肠内容物中的细菌分别进行有氧培养和厌氧培养,得到需氧培养菌和厌氧培养菌;
(2)将中草药洗净,晒干或风干,将各味中草药的根、茎、叶、果、皮、籽和花进行归属分类,在5-20℃下存放备用;
(3)将不同中草药的根、茎、叶、果、皮、籽和花粉粹成60目粉面,分别放进砂锅内泡制10-30分钟,再进行熬制,花叶类、皮果籽类和根茎类分开熬制,花叶类熬制时间为20-30分钟,皮果籽类熬制时间为30-40分钟,根茎类熬制时间为40-60分钟,直至熬制成的中草药药液浓度为波美度2-3度,pH值5-6.5;
(4)将熬制成的中草药药液放在阴凉处自然降温沉淀,先用双层纱布过滤一遍,将药渣过滤掉,得到初滤中草药药液,再将初滤中草药药液用多层纱布过滤3-5遍,得到精滤中草药药液,然后将得到的精滤中草药药液置于消毒室内,于5-15℃下进行紫外线杀菌30-60分钟;
(5)将步骤(1)培养成的专用菌种接种到步骤(4)得到的中草药药液中进行培养,培养温度为5-10℃,培养5-7天后,培养温度升至15-20℃,继续培养5-8天,整个培养过程要求暗光培养。
4.根据权利要求3所述的液体中草药生物饲料添加剂的制备方法,其特征在于:所述需氧培养菌包括大肠杆菌属、粪球菌属、肠球菌属、显核菌属、短乳杆菌属、布氏乳杆菌属、蜡状芽孢杆菌、唾液乳杆菌和柠檬酸细菌属,厌氧培养菌包括梭杆菌属、双歧杆菌属、韦荣氏球菌属、乳杆菌属、明串球菌属和乳球菌属。
5.根据权利要求3所述的液体中草药生物饲料添加剂的制备方法,其特征在于:所述固体MRS肉汤培养基为:牛肉膏10g、蛋白陈10g、酵母浸粉5g、葡萄糖20g、吐温801mL、磷酸氢二钾2g、乙酸钠4g、柠檬酸氢二铵2g、水合硫酸镁0.2g、水合硫酸锰0.2g、琼脂20g、半胱氨酸0.3g、蒸馏水1000mL。
6.根据权利要求3所述的液体中草药生物饲料添加剂的制备方法,其特征在于:所述厌氧培养采用干燥器法:计算好容器体积,根据其体积称取焦性没食子酸和配制氢氧化钠溶液(1000mL空间用焦性没食子酸10g、10%氢氧化钠溶液100mL);然后将氢氧化钠溶液倒入干燥器底部,盛有焦性没食子酸的平皿轻轻浮于液面之上,放好隔板,将接种好的平板置于隔板上,并在隔板上点燃一支蜡烛,把干燥器盖盖上密封,轻轻摇动干燥器,使焦性没食子酸和氢氧化钠溶液混合作用,待蜡烛熄灭后置于37℃温箱2-3d,取出观察。
