Abstract
本发明提供了一种用于治疗猪痢疾的药物组合物及其制备方法,其中药物组合物包括以下原料药材:乌梅、细辛、干姜、当归、蜀椒、地锦草、赤石脂、罂粟壳、黄柏、黄连、黄芩、党参、翻百草、白术、甘草、白芍、老虎须、肉豆蔻、木香、诃子、马齿苋、瓶尔小草、梅花草。本发明的有益效果是:本发明的药物组合物能清热凉血解毒、调气行血止痢,在治疗猪痢疾方面具有质优价廉,安全高效、操作简便、低毒、无药物残留及寄生虫顾虑等优势特点,还能够对体重增长也有很大的帮助。

本发明涉及兽药技术领域,特别涉及一种用于治疗猪痢疾的药物组合物及其制备方法。
背景技术
猪痢疾是由致病性猪痢疾蛇形螺旋体引起猪的一种严重的肠道传染病,又称血痢。主要临诊症状为严重的黏液性和黏液性出血性下痢,急性型以出血性下痢为主,亚急性和慢性以黏液性腹泻为主。剖检病理特征为大肠黏膜发生卡他性、出血性及坏死性炎症。本病在世界上许多养猪国家和地区都有发生的报道,中国于1978年10月由美国进口的种猪首次发现本病,上海市畜牧兽医研究所经系列试验确诊为猪痢疾。其后在中国疫情扩大、蔓延,现已有20多个省、市发生本病,已是普遍存在的常见传染病之一,由于猪只死亡、生长率降低、饲料报酬率低、药物及防疫费用增加等,给养猪业造成很大的经济损失。
猪痢疾蛇形螺旋体,又称为猪痢疾短螺旋体,属于蛇形螺旋体属成员,存在于病猪的病变肠段黏膜、肠内容物及排出的粪便中。该病菌革兰氏染色阴性,苯胺染料或姬姆萨染色液着色良好,组织切片以镀银染色为好,可见两端尖锐、形如双雁翼状,菌体长6~8μm,宽0.32~0.38μm,有4~6个弯曲,新鲜病料在暗视野显微镜下,可见活泼的以长轴为中心旋转运动,在电子显微镜下可见细胞壁与外膜之间有7~9条轴丝,轴丝在靠近细胞中部发生重叠,具有运动性和溶血性,在暗视野显微镜下,可见其活泼的蛇样运动。本菌为严格的厌氧菌,对培养条件要求较严格,一般不做培养。猪痢疾蛇形螺旋体对结肠、盲肠的致病性不依赖于其他微生物,但肠内固有的厌氧微生物则是本菌定居的必要条件和导致病变严重。所以,用本菌口服感染无菌猪时,不发生症状和病变。
猪痢疾蛇形螺旋体对外界环境抵抗力较强,在密闭猪舍粪尿沟中可存活30天,土壤中4℃时能存活102天,粪便中5℃时存活61天,25℃时存活7天,37℃时很快死亡。对阳光照射、加热和干燥敏感。兽医实践中常用的消毒药和常用浓度,如过氧乙酸、氢氧化钠、煤酚皂等可迅速将其杀死。
病猪和带菌猪是主要的传染源,康复猪带菌率高,带菌时间可长达数月,经常随着粪便排出大量病菌,污染饲料、饮水、猪圈、饲槽、用具、周围环境、运输工具及母猪躯体(包括奶头),经消化道感染,健康猪吃入污染的饲料、饮水感染,仔猪出生后通过消化道感染,在断奶前后发病。 在自然情况下,只引起猪发病。不同品种、不同年龄的猪均可感染,以2~3月龄幼猪发生最多。小猪的发病率和病死率比大猪高。一般发病率70%~80%,病死率30%~60%。
本病的特征性病变主要在大肠(结肠、盲肠),尤其时回、盲肠接合部,而小肠一般没有病变。剖检见病尸明显脱水,显著消瘦,被毛粗刚和被粪便污染。急性期病猪的大肠壁和大肠系膜充血、水肿,肠系膜淋巴结也因发炎而肿大。结膜粘膜下的淋巴小节肿胀,隆突于粘膜表面。粘膜明显肿胀,被覆有大量混有血液的黏液。当病情进一步发时,大肠壁水肿减轻,而粘膜表层形成一层出血性纤维蛋白伪膜。剥去假膜,肠粘膜表面有广泛的糜烂和潜在性溃疡。当病变转为慢性时,粘膜面常被覆一层致密的纤维素性渗出物。本病的病变分布部位不定,病轻时仅侵害部分肠段,反之则可分布于整个大肠部分,而病的后期,病变区扩大,常呈广泛分布。
加强卫生管理对本病有一定预防作用,一般常用西药如下:痢菌净按0.5%水溶液0.5毫升/千克体重,作肌肉注射;或2.5-5.0毫克/千克体重,内服,每天2次,连续3天为1个疗程。四环素按100-200克/吨饲料,连喂3-5天。吠喃哩酮300克/吨饲料,连喂14天,硫酸新霉素300克/吨饲料,连喂3-5天。氯霉素50毫克/千克体重(最大剂量不超过5克),连服3-5天。下述药物按50千克体重给予,链霉素1-2克每天2次,连服2-3天,或1-2克,每天2次,肌肉注射。红霉素0.5-0.7克,每天2次肌肉注射,连用2-3天,对剧烈下痢者还应采用补液、强心等对症治疗。现有的治疗方法,效果不甚理想,并且会造成药物残留。
我国对痢疾疾病的研究历史悠久,古代医家认为痢疾是因外感时行疫毒,内伤饮食而致邪蕴肠腑,气血壅滞,传导失司,以腹痛腹泻,里急后重.,排赤白脓血便为主要临床表现的具有传染性的外感疾病。古代亦称“肠游”、“滞下”等,含有肠腑“闭滞不利”的意思。本病为最常见的肠道传染病之一,一年四季均可发病,但以夏秋季节为最多,可散在发生,也可形成流行,常因急骤发病,高热惊厥,厥脱昏迷而导致死亡,故须积极防治。
《内经》称本病为“肠游”,对本病的病因、症状、预后等方面都有所论述,如《素问,至真要大论》说:“火淫所胜,……民病泄注赤白,……腹痛溺赤,甚为血便。”指出本病的病因与气候有关,症状为腹痛,便下赤白。汉《金匮要略·呕吐哕下利病脉证并治》将本病与泄泻合称“下利”,制定了寒热不同的白头翁汤和桃花汤治疗本病,开创了痢疾的辨证论治,两方一直为后世医家所喜用。隋《诸病源候论》有“赤白痢”、“血痢”、“脓血痢”、“热痢”等20余种痢候记载,对本病的临床表现和病因、病机已有较深刻的认识。唐《备急千金要方》称本病为“滞下”,宋《严氏济生方》正式启用“痢疾”之病名:“今之所谓痢疾者,古所谓滞下是也”,一直沿用至今。金元时期,《丹溪心法》明确指出本病具有流行性、传染性:“时疫作痢,一方一家之内,上下传染相似”,并论述痢疾的病因以“湿热为本”。清代,出现了痢疾专著,如《痢疾论》《痢证论》等,对痢疾理论和临床进行了系统总结,学术上也有所创新。
中医药对各类型痢疾有良好的疗效,尤其是久痢,在辨证的基础上,采用内服中药或灌肠疗法,常能收到显着的效果。
发明内容
本发明所要解决的技术问题是提供一种用于治疗猪痢疾的药物组合物及其制备方法,使其具有清热凉血解毒、调气行血止痢功能的同时,提高猪的抗病能力,增强其免疫力,对生长有促进作用,在治疗猪痢疾疾病方面有着快捷有效、廉价安全、绿色无污染、纯天然性、无毒害性、无药物残留等优势特点。
为解决上述问题,本发明提供了一种具有清热凉血解毒、调气行血止痢功能的用于治疗猪痢疾疾病的药物组合物,其中,药物组合物包括以下原料药材:乌梅、细辛、干姜、当归、蜀椒、地锦草、赤石脂、罂粟壳、黄柏、黄连、黄芩、党参、翻百草、白术、甘草、白芍、老虎须、肉豆蔻、木香、诃子、马齿苋、瓶尔小草、梅花草。

为了解决上述技术问题,本发明还提供了一种用于治疗猪痢疾的药物组合物的制备方法,其中,所述药物组合物的剂型为片剂,其制备方法包括以下步骤:
a、将乌梅、细辛、干姜和当归放入3-5倍量乙醇中,加热回流提取2次,每次1.5-2小时,提取活性成份,将2次提取液合并静置为第一提取液;
b、将剩余原料放入3-6倍量水中,加热回流提取3次,每次35-50分钟,将3次提取液合并静置为第二提取液;
c、将第一提取液和第二提取液合并,减压回收乙醇并浓缩至药液浓度为0.3-0.5g生药/ml,抽滤后,滤液的相对密度约为20℃时1.04-1.1;上述滤液经体积为10L的大孔吸附树脂柱,先用10倍树脂柱体积的去离子水或蒸馏水洗脱,再用4倍树脂柱体积的80-90%乙醇洗脱,收集乙醇洗脱液,去除溶剂,得到原料药;加入淀粉或糊精压片。
其中,所述药物组合物的剂型为注射剂,其制备方法包括以下步骤:
a.将所述原料药材按比例混合,搅拌粉碎,加入相对于原料药质量之和2-4倍水,煎煮1-2个小时,煎煮液过滤,滤渣再次加入相对于原料药质量之和1-3倍水,再次煎煮1.5-2个小时,煎煮液过滤,将过滤液合并,加入2-4g活性炭,煮沸50-70分钟,自然冷却后,经离心、抽滤,得到滤液,加热浓缩至浓度为1g/ml-5g/ml的药液;
b. 取所述药液15ml,在其中依次加入0.1-0.4ml吐温-80、60-90mg亚硫酸钠,混合均匀后加注射用水至10ml,并用氢氧化钠调pH值至7.4-7.6,最后分装于西林瓶中,经沸水浴加热灭菌后,即得注射剂。
本发明的有益效果是:本发明的药物组合物具有治疗猪痢疾清热凉血解毒、调气行血止痢的同时,能够提高猪的增强其免疫力,在治疗猪痢疾方面具有安全、高效、快捷、方便、无药残、无毒害等优势特点,并且对仔猪的生长也有一定促进作用。
具体实施方式
中药治疗痢疾在我国有着源远流长的历史,早在《内经》时期,就出现了有关痢疾的论述,中兽医活灵活现地把中医中有关痢疾的治疗用于动物身上,理论原则主要有以下三个方面:1.祛邪导滞痢疾的基本病机是邪气壅滞肠中,只有祛除邪气之壅滞,才能恢复肠腑传导之职,避免气血之凝滞,脂膜血络之损伤,故为治本之法。因此,清除肠中之湿热、疫毒、冷积、饮食等滞邪颇为重要。常用祛湿、清热、温中、解毒、消食、导滞、通下等法,以达祛邪导滞之目的。2.调气和血即是顺畅肠腑凝滞之气血,祛除腐败之脂脓,恢复肠道传送功能,促进损伤之脂膜血络尽早修复,以改善腹痛、里急后重、下痢脓血等临床症状。正如刘河间所说:“调气则后重自除,行血则便脓自愈”。常采用理气行滞、凉血止血、活血化瘀、去腐生肌等治法。3,顾护胃气“人以胃气为本,而治痢尤要”。这是由于治疗实证初期、湿热痢、疫毒痢的方药之中,苦寒之晶较多,长时间大剂量使用,有损伤胃气之弊。因此,治痢应注意顾护胃气,并贯穿于治痢的始终。
虚证痢疾应扶正祛邪。因虚证久痢,虚实错杂,若单纯补益,则滞积不去,贸然予以通导,又恐伤正气,故应虚实兼顾,扶正祛邪。中焦气虚,阳气不振者,应温养阳气;阴液亏虚者,应养阴清肠;久痢滑脱者,可佐固脱治疗。
此外,古今学者提出有关治疗痢疾之禁忌,如忌过早补涩,以免关门留寇,病势缠绵不已;忌峻下攻伐,忌分利小便,以免重伤阴津,戕害正气等,都值得临床时参考借鉴。
总之,痢疾的治疗,热痢清之,寒痢温之,初痢则通之,久痢虚则补之。寒热交错者,清温并用;虚实夹杂者,通涩兼施。赤多者重用血药,白多者重用气药。始终把握祛邪与扶正的辨证关系、顾护胃气贯穿于治疗的全过程。
因此选用原料药材:乌梅、细辛、干姜、当归、蜀椒、地锦草、赤石脂、罂粟壳、黄柏、黄连、黄芩、党参、翻百草、白术、甘草、白芍、老虎须、肉豆蔻、木香、诃子、马齿苋、瓶尔小草、梅花草,还包括鸦胆子、败酱草、秦皮、苦参、麦瓶草、笔仔草、凌霄花,以清热凉血解毒、调气行血止痢为主要治疗原则治疗猪痢疾。
其中,乌梅为君药,涩肠止泻之功显著;赤石脂、鸦胆子、秦皮、苦参、罂粟壳、麦瓶草、笔仔草、凌霄花清热解毒,止痢担当主要作用,君臣相须为用,以加强止痢之功;黄柏、黄连、黄芩清热凉血,清三焦之火,宜于地锦草、翻百草、马齿苋同用,共奏清热解毒之效;木香、诃子、细辛、白芍、党参调畅气机,常与细辛、干姜、当归、白术等药配伍活血调气,同时也可增强泻火之力;佐以肉豆蔻、老虎须、干姜、蜀椒温补肾阳,固护本源;用甘草、瓶尔小草、梅花草固护胃气,调和诸药,共奏清热凉血解毒、调气行血止痢之效。
现将各味中药分述如下:
乌梅:性味酸、涩,平。归肝、脾、肺、大肠经。《本草纲目》:敛肺涩肠,治久嗽,泻痢,反胃噎膈,蛔厥吐利,消肿,涌痰,杀虫,解鱼毒、马汗毒、硫黄毒。该品酸涩,主要功用敛肺,涩肠,生津,安蛔。用于肺虚久咳;虚热烦渴;久疟;久泻;痢疾;便血;尿血;血崩;蛔厥腹痛;呕吐;钩虫病。
细辛:辛,温。归心、肺、肾经。用于气虚多汗,血虚头痛,阴虚咳嗽等忌服,反藜芦。细辛经水蒸气蒸馏可得精油,在化妆品、医药等行业用途很多。以辽细辛浸膏为添加剂原料,广泛用于日用产品,如肥皂、化妆品、牙膏等,具有抗菌、消炎、止血、镇痛等疗效。含细辛的兽药用于治咳嗽喘、便秘;含细辛的农药作杀虫剂和杀菌剂;由于细辛中所含挥发油具有特殊芳香气味,国外已开发使用细辛作为建筑材料的防蛀填料和防蚊驱虫原料。
干姜:味辛,性热。归脾、胃、心、肺经。功效:温中散寒,回阳通脉,燥湿消痰,温肺化饮。主治脘腹冷痛、呕吐、泄泻、亡阳厥逆、寒饮喘咳、寒湿痹痛《得配本草》:"干姜,入手少阴、足太阴经气分;炮姜,入足太阴经血分。"
当归:甘、辛、温[入心、肝、肺三经。功效有补血;活血;调经止痛;润燥滑肠主治证为:血虚诸证;月经不调;经闭;痛经;症瘕结聚;崩漏;虚寒腹痛;痿痹;肌肤麻木;肠燥便难;赤痢后重;痈疽疮疡;跌扑损伤。本品为良好的补血药,故用于因血虚引起的各种证候;本品为妇科调经要药,故用于女人月经不调、经闭、痛经等证;本品因有补血活血、消肿止痛、排脓生肌之功效,故可用于跌打损伤、痈疽疮疡等证;本品因有补血润肠的功效,故可用于血虚、肠湿阻中满及大便溏泄者慎服。现代药理研究,有明显的抗炎作用。
蜀椒:辛、温。除各种肉类的腥气;促进唾液分泌,增加食欲;使血管扩张,从而起到降低血压的作用;服花椒水能去除寄生虫。现代药理研究果含挥发油(为柠檬烯、枯醇、香叶醇等)、淄醇,不饱和有机酸。有芳香健胃、温中散寒、除湿止痛、杀虫解毒、止痒解腥的功效。
地锦草:性味辛、平,归肝、大肠经。喜温暖湿润气候,稍耐荫蔽,较耐湿。以疏松肥沃、排水良好的砂质壤上或壤上栽培为宜。可与玉米间作。《单方验方调查资料选编》治细菌性痢疾。《乾坤生意》治血痢不止。现代药理研究其含没食子酸、没食子甲脂、槲皮甙、槲皮素、肌醇和鞣酸等,没食子酸为其主要抗菌有效成分。
赤石脂:性味甘、涩、温,归大肠、胃经。《日华子本草》:“治泻痢,血崩带下,吐血衄血,并涩精淋沥,安心,镇五脏,除烦,疗惊悸,排脓,治疮疖痔瘘,养脾气,壮筋骨,补虚损。”用于治疗涩肠,止血,收湿,生肌。治久泻,久痢,便血,脱肛,遗精,崩漏,带下,溃疡不敛等病。
罂粟壳:性味平;酸、涩;归肺、大肠、肾经;《本草纲目》:“罂子粟壳,酸主收涩,故初病不可用之。泄泻下痢既久,则气败不固而肠滑肛,咳嗽诸病既久,则气散不收而肺胀痛剧,故俱宜此涩之、固之、收之、敛之”。《本草经疏》:“若肺家火热盛,与夫风寒外邪未散者,误用则咳愈增而难治。如肠胃积滞尚多,湿热方炽,命门火盛, 湿热下流为遗精者,误用之则邪气无从而泄,或腹痛不可当,或攻入手足骨节,肿痛不能动,或遍身发肿,或呕吐不下食,或头面俱肿,或精窍闭塞,水道不通,变证百出而淹延不起矣,可不慎哉!”功能有敛肺、涩肠、止痛。用于久咳、久泻,脱肛,脘腹疼痛。《本草求真》:“功专敛肺涩肠固肾,凡久泻、久痢脱肛、久嗽气乏,并心腹筋骨诸痛者最宜”。
黄柏:味苦,性寒。归肾、膀胱、大肠经。《汤液本草》:足大阳经引经药,足少阴经之剂。《医学入门》:足少阴、手厥阴本药,足大阳引经药。功效有清热,燥湿,泻火,解毒。治热痢,泄泻,消渴,黄疸,痿躄,梦遗,淋浊,痔疮,便血,赤白带下,骨蒸劳热,目赤肿痛,口舌生疮,疮疡肿毒。《伤寒论》白头翁汤主治热毒痢疾。黄柏具有抑制免疫反应的作用,其活性物质为黄柏碱和木兰花碱。黄柏可以对抗多种因素所致的炎症反应。黄柏对金黄色葡萄球菌感染的皮肤有明显的抗炎作用。
黄连:清热燥湿,泻火解毒。用于湿热痞满,呕吐吞酸,泻痢,黄疸,高热神昏,心火亢盛,心烦不寐,血热吐衄,目赤,牙痛,消渴,痈肿疔疮;外治湿疹,湿疮,耳道流脓。酒黄连善清上焦火热。用于目赤,口疮。姜黄连清胃和胃止呕。用于寒热互结,湿热中阻,痞满呕吐。萸黄连舒肝和胃止呕。用于肝胃不和,呕吐吞酸。《本草纲目》:黄连大苦大寒,用之降火燥湿,中病即当止,岂可久服,使肃杀之令常行,而伐其生发冲和之气乎《本草衍义》:今人多用治痢,盖执以苦燥之义。
黄芩:性味:苦,寒。入心、肺、胆、大肠经《本草纲目》:治风热湿热头疼,奔豚热痛,火咳,肺痿喉腥,诸失血。《本草正》:枯者清上焦之火,消痰利气,定喘嗽,止失血,退往来寒热,风热湿热,头痛,解瘟疫,清咽,疗肺痿肺痈,乳痈发背,尤祛肌表之热,故治斑疹、鼠瘘,疮疡、赤眼;实者凉下焦之热,能除赤痢,热蓄膀胱,五淋涩痛,大肠闭结,便血、漏血。泻实火,除湿热,止血,安胎。治壮热烦渴,肺热咳嗽,湿热泻痢,黄疸,热淋,吐、衄、崩、漏,目赤肿痛,胎动不安,痈肿疔疮。现代药理研究黄芩具有抗菌作用抗炎抗变态反应作用,对中枢神经系统的作用,对心血管的作用抗血小板聚集及抗凝,降血脂保肝、利胆、抗氧化抗癌,抗真菌作用。
党参:性味甘,平归脾、肺经。《本草正义》:党参力能补脾养胃,润肺生津,腱运中气,本与人参不甚相远。其尤 可贵者,则健脾运而不燥,滋胃阴而不湿,润肺而不犯寒凉,养血而不偏滋腻,鼓舞清阳, 振动中气 ,而无刚燥之弊。主要的功用是补中,益气,生津。治脾胃虚弱,气血两亏,体倦无力,食少,口渴,久泻,脱肛。
翻白草: 苦,寒。归胃、大肠经。《本草纲目》:治吐血,下血,崩中,疟疾,痈疮。翻白草的主要功用有:清热,解毒,止血,消肿。治痢疾,疟疾,肺痈,咳血,吐血,下血,崩漏,痈肿,疮癣,瘰疬结核。经现代医学临床验证,翻百草与琼珍灵芝搭配煮水喝有治疗糖尿病的作用,主要有降血糖,治疗失眠,防治并发症等作用,也可加入地骨皮。全草煎剂用平板打洞法,对志贺痢疾杆菌及福氏痢疾杆菌有抑制作用。翻白草中提得成分富马酸、没食子酸、原儿茶酸、槲皮素、柚皮素、山柰素、间苯二酸,对福氏和志贺痢疾杆菌具有抑菌作用,尤以没食子酸和槲皮素活性最强,最低抑菌浓度分别为59ppm/ml和37ppm/ml。
白术:味苦;甘;性温。归脾、胃经。可健脾益气,燥湿利水,止汗,安胎。用于脾虚食少,腹胀泄泻,痰饮眩悸,水肿,自汗,胎动不安。土白术健脾,和胃,安胎。用于脾虚食少,泄泻便溏,胎动不安。水浸液在试管内对絮状表皮癣菌、星形奴卡氏菌有抑制作用。煎剂对脑膜炎球菌亦有抑制作用。近报道白术煎剂和四君子汤对伤寒杆菌、甲型副伤寒杆菌、福氏痢疾杆菌、大肠杆菌、绿脓杆菌等均有不同程度的抑菌作用,而无杀菌作用。
甘草:生甘,平;炙甘,温。入脾、胃、肺经。.用于胃痛、腹痛及腓肠肌挛急疼痛等。另外,在许多处方中也常用本品调和诸药。西医药理发现,甘草剂有抗炎和抗变态反应的功能,因此在西医临床上主要作为缓和剂。缓解咳嗽,祛痰,治疗咽痛喉炎;甘草或甘草次酸有去氧皮质酮类作用,对慢性肾上腺皮质功能减退症有良好功效;甘草制剂能促进胃部粘液形成和分泌,延长上皮细胞寿命,有抗炎活性,常用于慢性溃疡和十二指肠溃疡的治疗;甘草的黄酮具有消炎、解痉和抗酸作用;甘草也是人丹的主要原料之一。
白芍:性凉,味苦酸,微寒,具有补血养血、平抑肝阳、柔肝止痛、敛阴止汗等功效,适用于阴虚发热、月经不调、胸腹胁肋疼痛、四肢挛急,泻痢腹痛、自汗盗汗、崩漏、带下等症。白芍的药理成分芍药甙有抗菌、解热、抗炎、增加冠状动脉流量、改善心肌营养血流、扩张血管、对抗急性心肌缺血、抑制血小板聚集、镇静、镇痛、解痉、抗溃疡、调节血糖的作用。白芍煎剂能抑制痢疾杆菌、肺炎链球菌、大肠杆菌、伤寒杆菌、溶血性链球菌、绿脓杆菌等。
老虎须:别名箭根薯、蒟蒻薯、山大黄、老虎花。蛛丝草科蒟蒻薯属植物长须果Tacca chantrieri Andre以根状茎入药。全年可采,洗净切片,鲜用或晒干。性味苦、辛,凉。功能主治清热解毒,理气止痛。用于肠炎,痢疾,消化不良,肝炎,胃、十二指肠溃疡,流行性感冒,咽喉肿痛,扁桃体炎,肺炎,疟疾,疮疡肿毒,烧烫伤。
肉豆蔻:性味辛;苦;温。脾;胃;大肠经。《本草经疏》:大肠素有火热及中暑热泄暴注,肠风下血,胃火齿痛及湿热积滞方盛,滞下初起,皆不宜服。功用有温中涩肠;行气消食。主虚泻;冷痢;脘腹胀痛;食少呕吐。种皮捣碎加入凉菜或其他腌渍品中作为调味食用;种子含固体油,可供工业用油抗菌作用。其化学成分甲基异丁香酚对金黄色葡萄球菌和肺炎双球菌,MIC分别为0.4mg/ml和0.6mg/ml。马拉巴酮B对金黄色葡萄球菌、枯草杆菌和坚忍链球菌有较强的抑菌作用,前两者的MIC为1ug/ml,后者为2ug/ml,马拉巴酮C对枯草杆菌的MIC为2ug/ml。
木香:性味:辛味辛;苦;性温。归脾;大肠、三焦经《本草纲目》:木香,乃三焦气分之药,能升降诸气。诸气膹郁,皆属于肺,故上焦气滞用之者,乃金郁则泄之也;中气不运,皆属于脾,故中焦气滞宜之者,脾胃喜芳香也;大肠气滞则后重,膀胱气不化则癃淋,肝气郁则为痛,故下焦气滞者宜之,乃塞者通之也。功效:行气,止痛,健脾,消食。主治:行气止痛;调中导滞。主胞胁胀满足;脘腹胀痛;嗳吐泄泻;痢疾后重。用于胸脘胀痛、泻痢后重、食积不消、不思饮食;中气不省;突发耳聋;蛇虫咬伤;牙痛。
诃子:性味;苦、酸、涩、平。归肺、大肠经。诃子主要功效有敛肺;涩肠;下气;利咽;收敛、止泻;久泻;久痢;脱肛;喘咳痰嗽;久咳失音。诃子、罂粟壳皆涩肠止泻、收敛固脱 且诃子下气消胀;干姜温中祛寒以助脾胃之阳气;陈皮理气和胃燥湿,有温中祛寒、涩肠止泻、理气之功效 用于治疗脾胃虚寒、泻痢日久不愈之脘腹冷痛、便下脓血白多赤少、甚或脱肛者。实验表明诃子具有抗病原微生物,解痉抗菌作用:体外试验证明,对4-5种痢疾杆菌都有效,尤以诃子壳为佳。诃子水煎剂(100%)除对各种痢疾杆菌有效外,且对绿脓杆菌、白喉杆菌作用较强,对金黄色葡萄球菌、大肠杆菌、肺炎球菌、溶血性链球菌、变形杆菌、鼠伤寒杆菌亦有作用(纸片法);对痢疾杆菌、金黄色葡萄球菌、绿脓杆菌的有效浓度分别为1:32,1:128,1:64(试管法)。诃子在体外有良好的抗伤寒杆菌的作用。用盐酸、乙醚提取的乙醇提取物具有更高的抗菌及抗真菌作用。
马齿苋:性味酸、寒。归肝、大肠经。《唐本草》:主诸肿瘘疣目,捣揩之;饮汁主反胃,诸淋,金疮血流,破血癖症癖,小儿尤良;用汁洗紧唇、面疱、马汗、射工毒涂之瘥。功效有清热解毒,散血消肿。治热痢脓血,热淋,血淋,带下,痈肿恶疮,丹毒,痕疬。用于湿热所致的腹泻、痢疾。亦用于便血、子宫出血,有止血作用。马齿苋对急、慢性菌痢的疗效,与其它治痢药物如磺胺脒、合霉素等相仿,对急性病例的有效率在90%以上,对慢性病例的有效率亦在60%上下。含有蛋白质、脂肪、碳水化合物、膳食纤维、钙、磷、铁、铜、胡萝卜素、维生素B1、维生素B2、尼克酸、维生素C等多种营养成分,尤其是维生素A、维生素C、核黄素等维生素和钙、铁等矿物质。其ω-3脂肪酸含量在绿叶菜中占首位。
鸦胆子:味苦,性寒,有清热解毒、止痢疾等功用。鸦胆子含生物碱、糖甙、酚性成分和一种羟基羧酸称鸦胆子酸等。鸦胆子仁含脂肪油。油中不皂化物,内含挥发油少许;皂化物,内含油酸、亚油酸、硬脂酸、棕榈酸等。种子中含多种结构上类似苦木素的苦味成分:鸦胆子苦醇、鸦胆子素A、B、C、D、E、F、G、H等。含鸦胆子甙。从茎中亦分离鉴定了鸦胆子苦醇,并有没食子酸乙酸大黄酚、大黄酚甙和β-谷甾醇,其中鸦胆子苦醇有较强的抗癌活性。又报道,尚分得鸦胆子苦烯。去油鸦胆子水浸液和乙醚浸膏加入感染粪便,能杀灭阿米巴原虫,鸦胆子油无作用。鸦胆子仁水浸液(1%)在体外15~20min内可杀死阿米巴原虫,1:1000水溶液可抑制其生长,鸦胆子仁口服或浸剂灌肠对阿米巴痢疾有良好效果。在体外,鸦胆子丁醇提取物、苦木素、鸦胆子苦素C和鸦胆亭均有明显抑制溶组织内阿米巴原虫的作用。
败酱草:性味:性凉,味辛、苦。用治肠痈(急性阑尾炎);用治肺痈(肺脓疡);用治疮痈肿毒。《神农本草经》列为上品,为常用的清热解毒药。其性味辛、苦、微寒,有清热解毒、消痈排脓、祛瘀止痛之功。
秦皮:性味:味苦,寒。归肝经、胆经、大肠经。功能:清热燥湿,清肝明目,收涩止痢,止带。主治:用于热毒泻痢,带下阴痒,肝热目赤肿痛,目生翳障。
苦参:性味味苦;性寒。归肝;肾;大肠;小肠经;膀胱;心经;膀胱经。《本草纲目》:苦参、黄柏之苦寒,皆能补肾,盖取其苦燥湿,寒除热也。热生风,湿生虫,故又能治风杀虫。惟肾水弱而相火胜者用之相宜,若火衰精冷,真元不足,及年高之人不可用也。张从正亦云,凡药皆毒也,虽甘草、苦参,不可不谓之毒,久服则五味各归其脏,必有偏胜气增之患,诸药皆然,学者当触类而长之可也,至于饮食亦然。又按《史记》云,太仓公淳于意医齐大夫病龋齿,灸左手阳明脉,以苦参汤日漱三升,出入慎风,五、六日愈,此亦取其去风气湿热杀虫之义。苦参具有抗菌消炎的作用,对痤疮杆菌、金黄色葡萄球菌、大肠杆菌均有抑制作用,能利尿、抗过敏、镇痛、平喘、祛痰。清热燥湿;祛风杀虫。主湿热泻痢;肠风便血;黄疸;小便不利;水肿;带下;阴痒;疥癣;麻风;皮肤瘙痒;湿毒疮疡。
瓶尔小草:性味微甘、酸,凉。功能主治清热解毒,消肿止痛。用于小儿肺炎,脘腹胀痛,毒蛇咬伤,疔疮肿毒;外用治急性结膜炎,角膜云翳,眼睑缘炎。
梅花草:生于山坡、林边、山沟、隰草地。分布东北及河北、山西、陕西、甘肃、内蒙古等地。全草含山柰酚、芸香甙、金丝桃甙。茎尚含槲皮素的葡萄糖甙。根含生物碱0.085~0.09%。性味苦,凉。功能主治清热凉血,消肿解毒。治黄疸型肝炎,脉管炎,疮痈肿毒。
麦瓶草:化学成份种子含淀粉等。性味甘微苦,凉。《陕西中草药》:"味淡微苦,性凉。"功能主治养阴,和血。治虚劳咳嗽,咯血。衄血,月经不调。①《民间常用草药汇编》:"养阴除热。治虚弱咳嗽。"②《陕西中草药》:"止血,调经活血。治鼻衄,吐血,月经不调。"
笔仔草:性味甘,凉。①《福建民间草药》:"甘,平,无毒。"②《闽南民间草药》:"甘,寒。"③广州部队《常用中草药手册》:"甘淡,凉。"归经《闽东本草》:"入脾、肾、膀胱三经。"功能主治清热,利水。治热病烦渴,泄泻,黄疸型肝炎,糖尿病,淋蚀,尿血,小便不利。①《福建民间草药》:俐小便,通淋闭,止消渴,除烦热。"②《广东中药》Ⅱ:"清热,利水,通淋,凉血。主治淋浊,肝热,疳积,小儿尿短。"③《闽东本草》:"治发热口渴,肠风泄泻,恶心呕吐,小便不利,目赤。"④广州部队《常用中草药手册》:"治尿路感染,肾炎,水肿,感冒高热,黄疸型肝炎,糖尿病。"
凌霄花:性味酸,寒。①《本经》:"酸,微寒。"②《吴普本草》:"神农、雷公、歧伯:辛。扁鹊:苦咸。黄帝:甘,无毒。"③《履巉岩本草》:"味辛,有毒。"④《本草备要》:"甘酸而寒。"归经入肝经。①《纲目》:"手、足厥阴经,行血分。"②《雷公炮制药性解》:"入脾、肝二经。"③《本草再新》:"入肾经。"功能主治凉血去瘀。治血滞经闭,症瘕,血热风痒,酒齄鼻。①《本经》:"主妇人产乳余疾,崩中,症瘕,血闭,寒热羸瘦。"②《药性论》:"主热风,风痫,大小便不利,肠中结实,止产后崩血不定,淋沥。"③《日华子本草》:"治酒齄,热毒风,刺风,妇人血膈,游风,崩中,带下。"④《履巉岩本草》:"降诸草毒。"⑤《医林纂要》:"缓肝风,泻肝热。治肝风巅顶痛。"⑥《天宝本草》:"行血通经:治跌打损伤,痰火脚气。"
以下采用实施例来详细说明本发明的实施方式,借此对本发明如何应用技术手段来解决技术问题,并达成技术效果的实现过程能充分理解并据以实施。
实施例1
药物组合物1
乌梅30份、细辛4份、干姜15份、当归20份、蜀椒5份、地锦草17份、赤石脂15份、罂粟壳6份、黄柏18份、黄连13份、黄芩15份、党参28份、翻百草15份、白术17份、甘草104份、白芍30份、老虎须4份、肉豆蔻16份、木香20份、诃子17份、马齿苋15份、瓶尔小草27份、梅花草17份。
其制备过程包括:
a、将乌梅、细辛、干姜和当归放入3倍量乙醇中,加热回流提取2次,每次2小时,提取活性成份,将2次提取液合并静置为第一提取液;
b、将剩余原料放入6倍量水中,加热回流提取3次,每次50分钟,将3次提取液合并静置为第二提取液;
c、将第一提取液和第二提取液合并,减压回收乙醇并浓缩至药液浓度为0.5g生药/ml,抽滤后,滤液的相对密度约为20℃时1.04;上述滤液经体积为10L的大孔吸附树脂柱,先用10倍树脂柱体积的去离子水或蒸馏水洗脱,再用4倍树脂柱体积的90%乙醇洗脱,收集乙醇洗脱液,去除溶剂,得到原料药;加入淀粉或糊精压片。
实施例2
药物组合物2
乌梅26份、细辛5份、干姜16份、当归20份、蜀椒5份、地锦草16份、赤石脂18份、罂粟壳5份、黄柏18份、黄连13份、黄芩15份、党参28份、翻百草15份、白术17份、甘草15份、白芍28份、老虎须4份、肉豆蔻14份、木香18份、诃子17份、马齿苋15份、瓶尔小草26份、梅花草15份、鸦胆子5份、败酱草15份、秦皮14份、苦参15份、麦瓶草18份、笔仔草15份、凌霄花23份。
其制备过程包括:
a.将所述原料药材按比例混合,搅拌粉碎,加入相对于原料药质量之和2倍水,煎煮2个小时,煎煮液过滤,滤渣再次加入相对于原料药质量之和3倍水,再次煎煮2个小时,煎煮液过滤,将过滤液合并,加入4g活性炭,煮沸70分钟,自然冷却后,经离心、抽滤,得到滤液,加热浓缩至浓度为5g/ml的药液;
b. 取所述药液15ml,在其中依次加入0.4ml吐温-80、90mg亚硫酸钠,混合均匀后加注射用水至10ml,并用氢氧化钠调pH值至7.4,最后分装于西林瓶中,经沸水浴加热灭菌后,即得注射剂。
实施例3
药物组合物3
乌梅30份、细辛4份、干姜15份、当归20份、蜀椒5份、地锦草17份、赤石脂15份、罂粟壳6份、黄柏18份、黄连13份、黄芩15份、党参28份、翻百草15份、白术17份、甘草104份、白芍30份、老虎须4份、肉豆蔻16份、木香20份、诃子17份、马齿苋15份、瓶尔小草27份、梅花草17份、鸦胆子5份、败酱草15份、秦皮14份、苦参17份、麦瓶草20份、笔仔草15份、凌霄花24份。
其制备过程为:
将所述原料药材按比例混合,搅拌粉碎,加入相对于原料药质量之和2倍水,煎煮2个小时,煎煮液过滤,滤渣再次加入相对于原料药质量之和2倍水,再次煎煮2个小时,煎煮液过滤,将过滤液合并,加热浓缩,获得生药浓度为9g/ml的汤剂。
毒性试验
①急性毒性试验:选择白鼠作为实验对象,以本发明实施例1的药物组合物1为对象,临床日用量的150倍给白鼠大肠内投药,观察7日。结果所有动物健存,其活动、饮食、毛发、排泄物未发现异常,未见毒性反应。对白鼠阴道和破损皮肤进行刺激性试验,未见有刺激性反应和其它不良反应。毒性试验证明,该药未有毒性作用和不良反应,临床应用安全可靠。
②蓄积毒性试验:选用20只健康小白鼠,雌、雄各10只,以本发明实施例2的药物组合物2为对象,剂量设计每5天为一期,每期剂量分别为0.10LD50 、0.15LD50 、0.22LD50 、0.34LD 50 、0.50LD50 ,此样品的雌性、雄性小鼠的LD 50均为5000m份/k份BW(五个剂量为500、750、1100、1700、2500m份/k份BW)。每5天称体重一次,调整给药量,按0.1ml/10份BW经口灌胃。20天后,动物无死亡情况发生,试验结束,按递增剂量给受试物20天后,雌性、雄性动物均无死亡情况发生。说明,本发明的药物组合物不具有毒性作用,临床应用安全可靠。
对比试验
沈阳某养猪场养殖猪约1500头,2012年5月,约有400头猪出现猪痢疾的症状:剖检见病尸明显脱水,显著消瘦,被毛粗刚和被粪便污染。急性期病猪的大肠壁和大肠系膜充血、水肿,肠系膜淋巴结也因发炎而肿大。结膜粘膜下的淋巴小节肿胀,隆突于粘膜表面。粘膜明显肿胀,被覆有大量混有血液的黏液。当病情进一步发时,大肠壁水肿减轻,而粘膜表层形成一层出血性纤维蛋白伪膜。剥去假膜,肠粘膜表面有广泛的糜烂和潜在性溃疡。当病变转为慢性时,粘膜面常被覆一层致密的纤维素性渗出物。
选择200头患病猪,隔离养殖,并随机分为四组,实施例1组、实施例2组、实施例3组和对照组,每组50头。
实施例1组采用本发明实施例1的药物组合物1片剂进行治疗,将片剂按照3:100的比例混入传统饲料中,正常饲喂,3天为1个疗程,进行1个疗程。
实施例2组采用本发明实施例2的药物组合物2注射剂进行治疗,将注射剂肌肉注射一日一次,每次5ml,3天为1个疗程,进行1个疗程。
实施例3组采用本发明实施例3的药物组合物3汤剂进行治疗,将汤剂按照3:100的比例混入传统饲料中,正常饲喂,3天为1个疗程,进行1个疗程。
对照组:采用痢菌净进行治疗,痢菌净按0.5%水溶液0.5毫升/千克体重,作肌肉注射;或2.5-5毫克/千克体重,内服,每天2次,3天为1个疗程,进行1个疗程。
记录实验过程的治疗情况。治愈:症状消失,各方面恢复正常;有效:症状减轻,各方面基本恢复正常;无效:症状未见好转,更厉害。参见表1。
记录实验前后的猪的平均体重,计算出平均体重增长情况。参见表2。
表1为各组的治疗情况对比(总共50头)
参见表1可以看出,实施例1组、实施例2组和实施例3组的治疗猪的治愈数都要高于对照组的猪的治愈数,可以得知,本发明的药物组合物,治疗猪痢疾上,治疗效果好,疗效确切,很好的改善病情。
表2为各组的猪的平均增长体重对比
参见表2可以看出,实施例1组、实施例2组和实施例3组的治疗猪痢疾的同时,增长速度都要快于对照组,保持了猪的正常生长。
综上所述,本发明药物组合物,在治疗猪痢疾上,治疗效果好,疗效确切,并且保证了猪的正常生长。
举例:广州市某养猪场养猪500头,2013年8月,200头出现猪痢疾症状,随机选择样品解剖:在大肠(结肠、盲肠),尤其时回、盲肠接合部,而小肠一般没有病变。剖检见病尸明显脱水,显著消瘦,被毛粗刚和被粪便污染。急性期病猪的大肠壁和大肠系膜充血、水肿,肠系膜淋巴结也因发炎而肿大。结膜粘膜下的淋巴小节肿胀,隆突于粘膜表面。粘膜明显肿胀,被覆有大量混有血液的黏液。当病情进一步发时,大肠壁水肿减轻,而粘膜表层形成一层出血性纤维蛋白伪膜。剥去假膜,肠粘膜表面有广泛的糜烂和潜在性溃疡。选择60头患病猪进行隔离治疗,采用本发明实施例2的药物组合物2注射剂进行治疗,将注射剂肌肉注射一日一次,每次5ml,3天为1个疗程,进行1个疗程,治愈的为60头,治愈率100%。体重在治疗期间增长3.25kg。

1.一种用于治疗猪痢疾的药物组合物,其特征在于,所述药物组合物包括以下原料药材: 乌梅、细辛、干姜、当归、蜀椒、地锦草、赤石脂、罂粟壳、黄柏、黄连、黄芩、党参、翻百草、白术、甘草、白芍、老虎须、肉豆蔻、木香、诃子、马齿苋、瓶尔小草、梅花草。
2.根据权利要求1所述的用于治疗猪痢疾的药物组合物,其特征在于,所述药物组合物中各原料药材的重量份数为:乌梅25-30重量份、细辛3-7重量份、干姜10-20重量份、当归15-25重量份、蜀椒4-6重量份、地锦草15-20重量份、赤石脂10-25重量份、罂粟壳3-8重量份、黄柏15-28重量份、黄连10-16重量份、黄芩10-20重量份、党参25-35重量份、翻百草10-20重量份、白术15-20重量份、甘草10-18重量份、白芍25-35重量份、老虎须2-7重量份、肉豆蔻10-20重量份、木香15-25重量份、诃子15-20重量份、马齿苋10-18重量份、瓶尔小草25-30重量份、梅花草10-20重量份。

7.根据权利要求1-6任一项所述的用于治疗猪痢疾的药物组合物,其特征在于,所述药物组合物的剂型为片剂,其制备方法包括以下步骤:
a、将乌梅、细辛、干姜和当归放入3-5倍量乙醇中,加热回流提取2次,每次1.5-2小时,提取活性成份,将2次提取液合并静置为第一提取液;
b、将剩余原料放入3-6倍量水中,加热回流提取3次,每次35-50分钟,将3次提取液合并静置为第二提取液;
c、将第一提取液和第二提取液合并,减压回收乙醇并浓缩至药液浓度为0.3-0.5g生药/ml,抽滤后,滤液的相对密度约为20℃时1.04-1.1;上述滤液经体积为10L的大孔吸附树脂柱,先用10倍树脂柱体积的去离子水或蒸馏水洗脱,再用4倍树脂柱体积的80-90%乙醇洗脱,收集乙醇洗脱液,去除溶剂,得到原料药;加入淀粉或糊精压片。
8.根据权利要求1-6任一项所述的用于治疗猪痢疾的药物组合物,其特征在于,所述药物组合物的剂型为注射剂,其制备方法包括以下步骤:
a.将所述原料药材按比例混合,搅拌粉碎,加入相对于原料药质量之和2-4倍水,煎煮1-2个小时,煎煮液过滤,滤渣再次加入相对于原料药质量之和1-3倍水,再次煎煮1.5-2个小时,煎煮液过滤,将过滤液合并,加入2-4g活性炭,煮沸50-70分钟,自然冷却后,经离心、抽滤,得到滤液,加热浓缩至浓度为1g/ml-5g/ml的药液;
b. 取所述药液15ml,在其中依次加入0.1-0.4ml吐温-80、60-90mg亚硫酸钠,混合均匀后加注射用水至10ml,并用氢氧化钠调pH值至7.4-7.6,最后分装于西林瓶中,经沸水浴加热灭菌后,即得注射剂。
9.一种如权利要求1-6任一项所述的用于治疗猪痢疾的药物组合物的制备方法,其特征在于,所述药物组合物的剂型为片剂,其制备方法包括以下步骤:
a、将乌梅、细辛、干姜和当归放入3-5倍量乙醇中,加热回流提取2次,每次1.5-2小时,提取活性成份,将2次提取液合并静置为第一提取液;
b、将剩余原料放入3-6倍量水中,加热回流提取3次,每次35-50分钟,将3次提取液合并静置为第二提取液;
c、将第一提取液和第二提取液合并,减压回收乙醇并浓缩至药液浓度为0.3-0.5g生药/ml,抽滤后,滤液的相对密度约为20℃时1.04-1.1;上述滤液经体积为10L的大孔吸附树脂柱,先用10倍树脂柱体积的去离子水或蒸馏水洗脱,再用4倍树脂柱体积的80-90%乙醇洗脱,收集乙醇洗脱液,去除溶剂,得到原料药;加入淀粉或糊精压片。
10.一种如权利要求1-6任一项所述的用于治疗猪痢疾的药物组合物的制备方法,其特征在于,所述药物组合物的剂型为注射剂,其制备方法包括以下步骤:
a.将所述原料药材按比例混合,搅拌粉碎,加入相对于原料药质量之和2-4倍水,煎煮1-2个小时,煎煮液过滤,滤渣再次加入相对于原料药质量之和1-3倍水,再次煎煮1.5-2个小时,煎煮液过滤,将过滤液合并,加入2-4g活性炭,煮沸50-70分钟,自然冷却后,经离心、抽滤,得到滤液,加热浓缩至浓度为1g/ml-5g/ml的药液;
b. 取所述药液15ml,在其中依次加入0.1-0.4ml吐温-80、60-90mg亚硫酸钠,混合均匀后加注射用水至10ml,并用氢氧化钠调pH值至7.4-7.6,最后分装于西林瓶中,经沸水浴加热灭菌后,即得注射剂。
