本发明公开了一种伊红染色液,伊红染色液包括以下质量份数的组分:水溶性伊红12份、水1200份、氯化钠0.4份、1mol/L的柠檬酸水溶液1.3份、无水乙醇20份、纳米银0.01份、丙三醇19份、浓度为5mmol/L的硼酸溶液2份、硬脂酸2份、鲸蜡醇0.3份、异丙醇胺1份、吐温20 2份、尿囊素0.8份、羟苯乙酯1份、丙烯酸树脂6份、生育酚乙酸酯0.8份、十二烷基苯磺酸钠0.001份和脂肪酸甘油酯0.001份。本发明不仅满足病理学试剂环保的要求,而且染色效果优于传统的伊红染色液。
Description
技术领域
本发明涉及一种伊红染色液。
背景技术
组织病理学的诞生已有150多年的历史,伊红溶液是常用的病理学试剂之一,其主要用来对组织切片染色,目前的伊红染色液中大都含有甲醛、二甲苯、氧化汞、氨水等,有毒害化学试剂,不仅对工作人员的身体造成了伤害,而且也对环境造成了污染。
发明内容
本发明的一个目的是解决至少上述问题,并提供至少后面将说明的优点。
本发明还有一个目的是提供一种伊红染色液,其不仅满足病理学试剂环保的要求,而且染色效果优于传统的伊红染色液。
为了实现根据本发明的这些目的和其它优点,提供了一种伊红染色液,伊红染色液包括以下质量份数的组分:
水溶性伊红12份、水1200份、氯化钠0.4份、1mol/L的柠檬酸水溶液1.3份、无水乙醇20份、纳米银0.01份、丙三醇19份、浓度为5mmol/L的硼酸溶液2份、硬脂酸2份、鲸蜡醇0.3份、异丙醇胺1份、吐温20 2份、尿囊素0.8份、羟苯乙酯1份、丙烯酸树脂6份、生育酚乙酸酯0.8份、十二烷基苯磺酸钠0.001份和脂肪酸甘油酯0.001份;
优选的是,所述的伊红染色液中,
制备伊红染色液:将氯化钠加入到温度40℃的水中,搅拌均匀,升温至50℃,加入水溶性伊红和柠檬酸水溶液,搅拌均匀,边搅拌边加入纳米银,在温度为30℃的水浴中,水浴超声10min,得到第一混合液;
将硼酸溶液加入到无水乙醇中,搅拌均匀,加入十二烷基苯磺酸钠和脂肪酸甘油酯,在水浴温度为30℃下,水浴超声2min,加入丙三醇、硬脂酸、鲸蜡醇、异丙醇胺、吐温20、尿囊素、羟苯乙酯、丙烯酸树脂和生育酚乙酸酯,在水浴温度为40℃下,水浴超声1min,得到第二混合液;
边搅拌将第二混合加入至第一混合液中,在水浴温度为30℃下,水浴超声2min,即得到伊红染色液。
本发明至少包括以下有益效果:在伊红染色液中:加入的无水的乙醇起到了对各组分的良好溶解,同时加入硼酸溶液,进一步促进组分的溶解,硬脂酸和鲸蜡醇其可以保证染色液的稳定性;十二烷基苯磺酸钠和脂肪酸甘油酯有效的保证了各组分之间良好的混匀效果;异丙醇胺具有良好的抗静电能力,同时其具有良好的吸湿性;尿囊素具有良好的防腐性和抗氧化性,可以有效的保持组织新鲜防止腐败变质;羟苯乙酯具有良好的抗菌性,能有效防止切片变质;吐温20可以为非离子表面活性剂,其能有效的增加羟苯乙酯的水溶性;生育酚乙酸酯能有效的防止切片氧化,同时本发明中加入了纳米银,其不仅具有练好的杀菌抗腐作用,且具有良好的吸附力和穿透力,其能有效的吸附伊红染料,并能将其均匀传播到组织各处,且不会影响对组织的观察。
本发明设计的制备方法中采用分步在不同的温度下加入组分,其不仅保证了各个溶液的均质性,而且能防止结块等现象的出现,同时采用超声的方法增加了各组分之间的混合以及组分的溶解性。

实施例1、
一种伊红染色液,包括:
水溶性伊红12g、水1200g、氯化钠0.4g、1mol/L的柠檬酸水溶液1.3g、无水乙醇20g、纳米银0.01g、丙三醇19g、浓度为5mmol/L的硼酸溶液2g、硬脂酸2g、鲸蜡醇0.3g、异丙醇胺1g、吐温20 2g、尿囊素0.8g、羟苯乙酯1g、丙烯酸树脂6g、生育酚乙酸酯0.8g、十二烷基苯磺酸钠0.001g和脂肪酸甘油酯0.001g。
实施例2、
一种实施例1中所述的伊红染色液的制备方法:将氯化钠加入到温度40℃的水中,搅拌均匀,升温至50℃,加入水溶性伊红和柠檬酸水溶液,搅拌均匀,边搅拌边加入纳米银,在温度为30℃的水浴中,水浴超声10min,得到第一混合液;
将硼酸溶液加入到无水乙醇中,搅拌均匀,加入十二烷基苯磺酸钠和脂肪酸甘油酯,在水浴温度为30℃下,水浴超声2min,加入丙三醇、硬脂酸、鲸蜡醇、异丙醇胺、吐温20、尿囊素、羟苯乙酯、丙烯酸树脂和生育酚乙酸酯,在水浴温度为40℃下,水浴超声1min,得到第二混合液;
边搅拌将第二混合加入至第一混合液中,在水浴温度为30℃下,水浴超声2min,即得到伊红染色液。

1.一种伊红染色液,其特征在于,伊红染色液包括以下质量份数的组分:
水溶性伊红12份、水1200份、氯化钠0.4份、1mol/L的柠檬酸水溶液1.3份、无水乙醇20份、纳米银0.01份、丙三醇19份、浓度为5mmol/L的硼酸溶液2份、硬脂酸2份、鲸蜡醇0.3份、异丙醇胺1份、吐温20 2份、尿囊素0.8份、羟苯乙酯1份、丙烯酸树脂6份、生育酚乙酸酯0.8份、十二烷基苯磺酸钠0.001份和脂肪酸甘油酯0.001份。
2.如权利要求1所述的伊红染色液,其特征在于,
制备伊红染色液:将氯化钠加入到温度40℃的水中,搅拌均匀,升温至50℃,加入水溶性伊红和柠檬酸水溶液,搅拌均匀,边搅拌边加入纳米银,在温度为30℃的水浴中,水浴超声10min,得到第一混合液;
将硼酸溶液加入到无水乙醇中,搅拌均匀,加入十二烷基苯磺酸钠和脂肪酸甘油酯,在水浴温度为30℃下,水浴超声2min,加入丙三醇、硬脂酸、鲸蜡醇、异丙醇胺、吐温20、尿囊素、羟苯乙酯、丙烯酸树脂和生育酚乙酸酯,在水浴温度为40℃下,水浴超声1min,得到第二混合液;
