Abstract
本发明提供了一种皇冠草人工种子制备方法,将皇冠草幼嫩叶片作为外植体,在附加有BA(6‑苄氨基嘌呤)和ZT(玉米素)的MS培养基上培养30天后,获得的皇冠草的成熟体细胞胚,在无菌条件下将体细胞胚切下,并加入到含有MS培养基的3%海藻酸钠溶液中混合,用无菌滴管将含有体细胞胚的包埋液滴入到2%的氯化钙溶液中,半小时后用无菌水进行漂洗,即可制成皇冠草人工种子。本发明解决了皇冠草在培植过程中繁殖率较低的问题,以叶片作为外植体材料易于获得,不受季节影响,繁殖速度快,成活率高。

技术领域
本发明属于植物人工种子生产技术领域,具体涉及一种皇冠草人工种子的制备方法。
背景技术
皇冠草又称亚马逊剑草,原产于南美洲,是一种大型多年生沉水性水草,因叶子宽大、叶形优美多用于水族箱造景。皇冠草以分株繁殖为主,繁殖水温需保持在20-26℃。当匍匐茎上散生的嫩芽长至6-8cm的时候,带根掰下另行栽种。但是这种繁殖方式的繁殖率太低,且对环境的要求较高,现有的繁殖方式不能满足实际需要。
人工种子是指将植物离体培养中产生的体细胞胚或者能发育成完整植株的分生组织包埋在含有营养物质和具有保护功能的外壳内形成,在适宜条件下发芽出苗的颗粒体。人工种子在适宜环境条件下,与天然种子具有相同的繁殖功能,同时人工种子可以不受某些环境因素限制,实现大规模高效率生产,从而达到快速繁育的目的。
发明内容
本发明所要解决的技术问题是提供一种皇冠草人工种子的制备方法,克服自身繁殖效率低的限制,以实现大规模大批量繁育。
未解决上述技术问题,本发明采用的具体技术方案如下:
一种皇冠草人工种子的制备方法,其特征在于包括以下步骤:
(1)消毒:皇冠草幼嫩叶片用自来水冲洗干净后,在超净工作台上,将其叶片浸泡在70%酒精40s,无菌水冲洗两次后,用3%次氯酸钠表面消毒5分钟,用无菌蒸溜水冲洗4次。
(2)组织培养:叶片切成5mm2左右,接种到MS培养基上,叶片背面朝下,培养温度为25℃,光照周期16h:8h,培养30天。所述MS培养基上含有3%的蔗糖、0.8%的琼脂、0.8mg/LBA(6-苄氨基嘌呤)和1.1mg/L ZT(玉米素)。
(3)包埋:在超净工作台上,将体细胞胚剥离,用镊子置于盛3%海藻酸钠和1/5MS+1mg/L吲哚乙酸(IAA)的溶液中,将体细胞胚和包埋液混合。用口径为5mm的滴管吸起将混合液滴入2%的氯化钙溶液中,经过30分钟形成凝胶状颗粒。
(4)清洗:用无菌水漂洗凝胶颗粒4次即可得到无菌的人工种子。
本发明的有益效果如下:
1、本发明提供的皇冠草人工种子制备方法,制作过程简单,不受季节影响,成本低廉。
2、发明提供的皇冠草人工种子制备方法,可以实现皇冠草种子的款速无性繁殖,种子发芽率高,成活率好,极大地缩短了繁殖时间。

下面结合实施例对本发明作进一步详细描述,但是下述实施例并不限制本发明的范围,此外相关领域的普通技术人员对本发明做的任何改动,只要不脱离本发明的实质,都将等价落在本发明权利要去书所限定的范围内。
实施例
选取皇冠草匍匐茎上散生的幼嫩叶,用镊子掰下备用。
(1)皇冠草幼嫩叶片用自来水冲洗干净后,在超净工作台上,将其叶片浸泡在70%酒精40s,无菌水冲洗两次后,用3%次氯酸钠表面消毒5分钟,用无菌蒸溜水冲洗4次。
(2)叶片切成5mm2左右,接种到含有0.8mg/L BA(6-苄氨基嘌呤)、1.1mg/L ZT(玉米素)、3%的蔗糖和0.8%的琼脂的MS培养基上。叶片背面朝下,培养温度为25℃,光照周期16h:8h,培养30天。
(3)培养30天后,在超净工作台上,将体细胞胚剥离,用镊子置于盛3%海藻酸钠和1/5MS+1mg/L吲哚乙酸(IAA)的包埋溶液中,将体细胞胚和包埋液混合。用口径为5mm的滴管吸混合液滴入2%的氯化钙溶液中,经过30分钟形成凝胶状颗粒。
(4)用无菌水漂洗凝胶颗粒4次即可得到无菌的人工种子。
Claims (1)
1.一种皇冠草人工种子的制备方法,其特征在于,它包括以下步骤:
(1)皇冠草幼嫩叶片用自来水冲洗干净后,在超净工作台上,将其叶片浸泡在70%酒精40s,无菌水冲洗两次后,用3%次氯酸钠表面消毒5分钟,用无菌蒸溜水冲洗4次;
(2)叶片切成5mm2,接种到含有0.8mg/L BA(6-苄氨基嘌呤)、1.1mg/L ZT(玉米素)、3%的蔗糖和0.8%的琼脂的MS培养基上;叶片背面朝下,培养温度为25℃,光照周期16h:8h,培养30天;
(3)在超净工作台上,将培养好的体细胞胚剥离,用镊子置于盛3%海藻酸钠和1/5MS+1mg/L吲哚乙酸(IAA)的包埋液中,将体细胞胚和包埋液混合;用口径为5mm的滴管吸起混合液滴入2%的氯化钙溶液中,经过30分钟形成凝胶状颗粒;
(4)用无菌水漂洗凝胶颗粒4次,即得到无菌的人工种子。
