Abstract

本发明涉及一种用于提高益生菌生物活性的包埋微胶囊,该微胶囊以果胶和葡萄糖为核心材料包埋益生菌,并以氯化钙为包芯材料,该微胶囊可以促进益生菌的体外存活期及体内的生长,还可以靶向定位传递益生菌到肠道内的作用部位,减少肠道内酸性环境及消化酶对益生菌的损害,以达到提高益生菌功效的作用。

一种用于提高益生菌生物活性的包埋微胶囊
技术领域
本发明涉及微生物制剂辅料的开发及应用技术领域,尤其涉及一种用于提高益生菌生物活性的包埋微胶囊。
背景技术
口服是益生菌常用的给药途径,在人体内结肠是益生菌起作用的一个重要部位。口服益生菌后,益生菌需通过胃及小肠才能达到结肠,胃和肠道内的消化酶对益生菌的活性有很大的破坏作用。例如,有实验证明,在模拟胃微环境中,当PH值低于3时,乳酸杆菌将无法生长及存活。
众所周知,包埋系统是目前研究最广泛的一种用于保护益生菌的技术。目前应用的包埋材料包括壳聚糖、海藻酸钠、卡拉胶、明胶、乳清蛋白、和淀粉。这些包埋材料对益生菌有一定的保护作用。但是,此作用未达到使益生菌发货最大效应的程度。例如,最常用的海藻酸钠保护。用海藻酸钠包埋的益生菌仍不能在PH<2的环境中生存。PH<2在人体胃中是一种常见的状态。目前有些包埋系统需要在高湿加热高温的条件下进行制备,制备过程中对益生菌有很大的杀伤力。
目前,益生菌对维护人体健康及疾病防治的作用已被逐步发现,这些研究成果非常支持广泛应用益生菌。因此,开发新的益生菌包埋技术,生产出包埋微胶囊,对于益生菌的应用将起到促进作用。
发明内容
本发明的目的是针对现有技术中存在的上述问题,提供一种用于提高益生菌生物活性的包埋微胶囊,制得的微胶囊可以促进益生菌的体外存活期及体内的生长,还可以靶向定位传递益生菌到肠道内的作用部位,减少肠道内酸性环境及消化酶对益生菌的损害,以达到提高益生菌功效的作用。
本发明解决其技术问题所采用的技术方案是:一种用于提高益生菌生物活性的包埋微胶囊,该微胶囊以果胶和葡萄糖为核心材料包埋益生菌,并以氯化钙为包芯材料。
进一步的,所述微胶囊的粒径为1mm~1.5mm。
进一步的,所述益生菌为乳杆菌或者双歧杆菌属内或者属间的一种或者两种以上。
与现有技术相比,本发明产生的有益效果是:该包埋微胶囊以果胶和葡萄糖为核心材料,并利用氯化钙为包芯材料的封装系统,采用冷冻干燥制备出微粒。果胶是食品级,非易消化的多糖,提取自柑橘、苹果和其他高等植物的细胞壁组织。首先,果胶含有大量D-半乳糖醛酸,通过1,4糖苷键;其次,果胶还是中性糖,例如L-鼠李糖,可插入或者连接到主链;再次,果胶是酸性环境中聚合体大分子,具有抗蛋白酶和淀粉酶的特性;另外,果胶溶解于水,制备过程中无需有机溶剂。而葡萄糖可促进益生菌的生长。
该包埋微胶囊可以延长益生菌(包括乳酸干菌或者双歧杆菌)的体外存活期,达到延长保质期的目的。被包埋的益生菌还具有对低酸环境和消化酶的抵抗能力。老鼠服用该包埋微胶囊可以提高防治结肠炎的能力。所以,这种包埋技术可提高益生菌的保存期和应用疗效。

具体实施方式
下面结合实施例对本发明做进一步说明:
如图1所示,本发明提供的包埋LGG微胶囊颗粒,体积为10uL,标尺=2;
如图2所示,本发明提供的包埋后的LGG在细菌培养液中浮育培养前(C)和经过3天培养,LGG数目明显增加(D)。箭头指向:LGG,放大倍数:A=70,B,C,D=10,000。箭头指向:LGG。
如图3所示,给小鼠喂已经在室温下储藏一个月的包埋的LGG(1x108LGG/包埋颗粒,每天一次),未经室温下储藏非包埋LGG(108在500L PBS中/天)以及无LGG颗粒后,利用ELISA方法检测粪便中p40含量,结果表明包埋LGG微胶囊颗粒饲喂后的小鼠,粪便中检测出的p40含量明显高与未包埋的LGG,表明包埋的LGG能够在结肠中产生更多的活性蛋白。
如图4和图5所示,给小鼠喂包埋的LGG(1x108LGG/包埋颗粒,每天一次),无LGG颗粒(1粒/每天),或者非包埋LGG(108在500μL中,PBS/天)。3天后,在饮水中给DSS(3%),连服4天,诱发肠损伤和结肠炎。以饮水小鼠为对照组。图4中的箭头表示显示溃烂和炎性细胞浸润,图5为炎症分数,0为正常。图4和图5表明,在喂包埋的LGG小鼠中,DSS诱发的肠损伤和结肠炎明显减轻。
该包埋微胶囊可以延长益生菌(包括乳酸干菌或者双歧杆菌)的体外存活期,达到延长保质期的目的。被包埋的益生菌还具有对低酸环境和消化酶的抵抗能力。老鼠服用该包埋微胶囊可以提高防治结肠炎的能力。所以,这种包埋技术可提高益生菌的保存期和应用疗效。

1.一种用于提高益生菌生物活性的包埋微胶囊,其特征在于:该微胶囊以果胶和葡萄糖为核心材料包埋益生菌,并以氯化钙为包芯材料。
2.根据权利要求1所述的用于提高益生菌生物活性的包埋微胶囊,其特征在于:所述微胶囊的粒径为1mm~1.5mm。
3.根据权利要求1所述的用于提高益生菌生物活性的包埋微胶囊,其特征在于:所述益生菌为乳杆菌或者双歧杆菌属内或者属间的一种或者两种以上。
