Abstract
本发明属于保健品领域,涉及含有青梅提取物的组合物在制备减肥保健品中的用途,本发明含有青梅提取物的组合物包括以下重量份数的制备原料:青梅提取物13‑19份、柠檬片11‑16份、绿豆9‑14份、苦瓜7‑11份、人参7‑11份、枸杞子5‑9份、茵陈6‑13份、栀子4‑9份、郁金香3‑8份和泽泻6‑12份。与现有技术相比,本发明含有青梅提取物的组合物对食欲无影响,在改善肥胖方面具有效果好、健康减肥等技术优势;并且其制备工艺简单、成本低、普遍适应性好、易于推广应用。

技术领域
本发明属于保健品,尤其涉及含有青梅提取物的组合物在制备减肥保健品中的用途。
背景技术
肥胖症是一组常见的代谢症群。当人体进食热量多于消耗热量时,多余热量以脂肪形式储存于体内,其量超过正常生理需要量,且达到一定值时即演变为肥胖症。正常男性成人脂肪组织重量约占体重的15%-8%,女性约占20%-25%。随年龄增长,体脂所占比例也相应增加。因体脂增加使体重超过标准体重20%或体重指数[BMI=体重(kg)/(身高)2(m2)]大于24者称为肥胖症。如无明显病因可寻者称单纯性肥胖症;具有明确病因者称为继发性肥胖症。
中医认为肥胖病的起因,有四大方面:(1)先天禀赋有关,如陈修园说:“大抵素禀之盛”,即现代医学所述的遗传因素;(2)嗜食膏粱厚味,饮食超量,营养成分,供过于求,即《黄帝内经》所述“肥贵人,膏粱之疾也”;(3)嗜卧少动,使体能消耗明显降低,致营养过剩,使脂肪充于肌肤而致肥胖;(4)脏腑功能失调,肝郁气滞、脾虚失运、肾虚气化失职,内伤久病,痰浊内生,或外受湿邪,使痰湿蓄积体内而肥胖。肥胖者多表现为本虚标实之证,形体表象壮实、实际却正气不足。本为气虚,标为湿、痰、疲,临床应据证而辨。《素问·阴阳应象大论》:“年四十而阴气自半也,起居衰矣,年五十而体重……”。人至中年肥胖多发,中医认为中年以后身体由盛转衰,脏腑功能渐弱,活动减少且代谢功能降低,故而发胖。此外,因过食肥甘、膏粱厚味之品,生活安逸、好坐、好静,“形不动则精不流,精不流则气郁”,气血流通缓慢,脾胃功能减弱,致水谷精微失于输布,反化膏脂与水湿,积于肌肤,形成肥胖。李东垣提出“脾胃旺”的人能食而胖,过食亦可伤脾,水湿内停,郁而化热,湿热溢于肌肤而现肥胖。若七情变化超出人体生理调节范围,必定影响饮食起居,肝气郁滞,肝胆疏泄失于调畅,脾胃运化功能障碍,影响气机之升降转输,且胆汁不能正常泌输精汁、净油化脂,则池脂内聚发为肥胖。脾肾气虚,肝胆失调,造成痰独、膏脂、水湿停聚,使气机失畅,脉道不利,又造成气滞或血癖。
中国专利申请200710065446.2公开了一种具有减肥作用的保健品,其主要由莱菔子、熟大黄、蒲公英和薄荷等原料制成。该发明保健品用于减肥具有一定的效果,但是效果见效慢,服用周期过长。
中国专利申请200710065447.7公开了一种具有减肥作用的保健品,其主要由番泻叶、制大黄、决明子和茯苓等原料制成。该发明保健品价格低廉,疗效好,但是服用过程中常出现腹泻、眩晕和疲倦等副作用。
发明内容
本发明要解决的技术问题是提供一种含有青梅提取物的组合物,该组合物对正常食欲无影响,用于减肥,具有起效快、副作用小、无反弹现象等优点,可用于制备减肥保健品。
本发明含有青梅提取物的组合物包括以下重量份数的制备原料:青梅提取物13-19份、柠檬片11-16份、绿豆9-14份、苦瓜7-11份、人参7-11份、枸杞子5-9份、茵陈6-13份、栀子4-9份、郁金香3-8份和泽泻6-12份。
进一步地,所述含有青梅提取物的组合物包括以下重量份数的制备原料:青梅提取物13份、柠檬片11份、绿豆9份、苦瓜7份、人参7份、枸杞子5份、茵陈6份、栀子4份、郁金香3份和泽泻6份。

进一步地,所述青梅提取物的制备方法包括如下步骤:
称取青梅,洗净后沥干其表面水分,去皮去核后,加入其质量6-8倍量的体积分数为75%的乙醇回流提取4-6小时,过滤,滤渣加4-6倍量的体积分数为75%的乙醇回流提取2-4小时,过滤,将2次滤液合并,减压蒸馏回收乙醇后3000rpm条件下离心15-30分钟,分离上清液,上清液减压浓缩至原体积1/8-1/10,得青梅提取物。
进一步地,本发明所述青梅提取物的组合物的制备方法包括如下步骤:
S1:称取青梅,洗净后沥干其表面水分,去皮去核后加入其质量6-8倍量的体积分数为75%的乙醇回流提取4-6小时,过滤,滤渣加4-6倍量的体积分数为75%的乙醇回流提取2-4小时,过滤,将两次滤液合并,减压蒸馏回收乙醇后3000rpm条件下离心15-30分钟,分离上清液,上清液减压浓缩至原体积1/8-1/10,得青梅提取物;
S2:称取柠檬片,将柠檬片粉碎,加入其质量8-12倍重量的水煎煮2-4小时;过滤,得到粗提液和柠檬片残渣,将粗提液1500-3000rpm条件下离心10-20分钟,分离上清液,上清液减压浓缩至60℃环境下相对密度为1.20-1.25的浸膏,得到柠檬片水提物;将柠檬片残渣,加入4-6倍量体积分数为50-70%的乙醇回流提取1-2次,每次1-2小时,过滤,合并滤液,减压蒸馏回收乙醇后在1500-3000rpm条件下离心10-20分钟,分离上清液,上清液减压浓缩至60℃环境下相对密度为1.20-1.25的浸膏,得到柠檬片醇提物;将柠檬片水提物和醇提物合并混匀,得柠檬片提取物;
S3:称取绿豆、苦瓜、人参、枸杞子、茵陈、栀子、郁金香和泽泻,粉碎,加入药材总重量6-8倍量的水煎煮3-4h,过滤;滤渣加入药材总重量4-6倍量的水煎煮1-2h,过滤;合并两次滤液,减压浓缩至65℃环境下相对密度为1.20-1.25的浸膏,减压干燥,粉碎后过80-100目筛,得粉末I;
S4:将S1得到的青梅提取物、S2得到柠檬片提取物和S3得到的粉末I充分混匀,减压浓缩至60℃环境下相对密度为1.25-1.35的浸膏,添加辅料制备成常见的保健食品。
本发明保健食品组合物所用原料来源和功效:
青梅:本品为蔷薇科植物梅的近成熟果实。味酸、涩,性平;归肝、脾、肺、大肠经;青梅止咳、涩肠止泻、除烦静心、生津止渴;主要用于肺虚久咳,久痢滑肠,虚热消渴。
柠檬片:本品为芸香科柑橘属植物干燥成熟果实。味酸,性平;归脾、胃经;化痰止咳,生津健胃;用于支气管炎,百日咳,食欲不振,维生素丙缺乏症,中暑烦渴。
绿豆:本品为豆科菜豆属植物绿豆。味甘,性寒;归心、胃经;清热解毒,消暑;用于暑热烦渴,疮毒痈肿等症。
苦瓜:本品为葫芦科苦瓜属植物苦瓜。味苦,性凉;归心、脾、胃经;清暑涤热,明目,解毒;用于中暑发热,牙痛,肠炎,痢疾,便血;外用治痱子,疔疮疖肿。
人参:本品为五加科植物人参的干燥根。味甘、微苦,性平;归脾、肺、心经;大补元气,复脉固脱,补脾益肺,生津,安神。用于体虚欲脱,肢冷脉微,脾虚食少,肺虚喘咳,津伤口渴,内热消渴,久病虚羸,惊悸肥胖。
枸杞子:本品为茄科植物宁夏枸杞的干燥成熟果实。味甘,性平;归肝、肾经;滋补肝肾,益精明目。主要用于虚劳精亏,腰膝酸痛,眩晕耳鸣,内热消渴,血虚萎黄,目昏不明。
茵陈:本品为菊科植物茵陈蒿的幼嫩茎叶。味苦,性平;归肝、脾、膀胱经;清热利湿;主要用于治湿热黄疸,小便不利,风痒疮疥。
栀子:本品为茜草科植物栀子的干燥成熟果实,其根也可入药。味苦;性寒。归心、肺、三焦经。泻火除烦,清热利尿,凉血解毒;主要用于热病心烦,黄疸尿赤,血淋涩痛,血热吐衄,目赤肿痛,火毒疮疡;外治扭挫伤痛。
郁金香:本品为百合科植物郁金香的花。味苦、辛,性平;归肺经;化湿辟秽;主要用于脾胃湿浊;胸脘满闷,呕逆腹痛,口臭苔腻。
泽泻:本品为泽泻科植物泽泻的干燥块茎。味甘,性寒;利小便,清湿热;主要用于小便不利,水肿胀满,泄泻尿少,痰饮眩晕,热淋涩痛。
本发明提供的青梅保健食品组合物中,青梅提取物能刺激唾液腺分泌较多的腮腺素,促进新陈代谢,还能促进肠胃蠕动,帮助排泄;柠檬片、绿豆、苦瓜具有生津健脾、清热解毒的功效,能够溶解多余的脂肪,清除肝、肾、肺等器官中的废物和毒素,净化血液,改善血质,促进新陈代谢;人参能够显著降低小肠和肝脏中脂肪和胆固醇;栀子、郁金香、泽泻、茵陈具有润肠通便、舒筋通脉、燥湿健脾、功毒散结的功效。本发明各原料成分配伍合理,相辅相成,协同作用,共同达到渗湿利水、利尿消肿、益气健脾、泻热通腑、化痰降浊的功效,用于减肥效果显著。
与现有技术相比,本发明技术优势如下:
1、本发明含有青梅提取物的组合物,副作用小,具有渗湿利水、利尿消肿、益气健脾、泻热通腑、化痰降浊的功效,对肥胖改善效果明显、稳定。
2、动物试验结果显示,本发明含有青梅提取物的组合物对肥胖大鼠的食欲无影响,同时可以显著抑制肥胖大鼠体重的增加及降低其体脂比,减肥功能显著,可用于制备减肥保健品。

实施例1:
本发明实施例1含有青梅提取物的组合物包括以下重量份数的制备原料:
青梅提取物13份、柠檬片11份、绿豆9份、苦瓜7份、人参7份、枸杞子5份、茵陈6份、栀子4份、郁金香3份和泽泻6份、黄芪2份及茯苓6份。
制备方法如下:
S1:称取青梅,洗净后沥干其表面水分,去皮去核后加入其质量6倍量的体积分数为75%的乙醇回流提取6小时,过滤,滤渣加4倍量的体积分数为75%的乙醇回流提取4小时,过滤,将两次滤液合并,减压蒸馏回收乙醇后3000rpm条件下离心20分钟,分离上清液,上清液减压浓缩至原体积1/10,得青梅提取物;
S2:称取柠檬片,将柠檬片粉碎,加入其质量12倍重量的水煎煮3小时;过滤,得到粗提液和柠檬片残渣,将粗提液1500rpm条件下离心15分钟,分离上清液,上清液减压浓缩至60℃环境下相对密度为1.20的浸膏,得到柠檬片水提物;将柠檬片残渣,加入6倍量体积分数为60%的乙醇回流提取2次,每次2小时,过滤,合并滤液,减压蒸馏回收乙醇后在3000rpm条件下离心10分钟,分离上清液,上清液减压浓缩至60℃环境下相对密度为1.20的浸膏,得到柠檬片醇提物;将柠檬片水提物和醇提物合并混匀,得柠檬片提取物;
S3:称取绿豆、苦瓜、人参、枸杞子、茵陈、栀子、郁金香和泽泻,粉碎,加入药材总重量8倍量的水煎煮4h,过滤;滤渣加入药材总重量6倍量的水煎煮2h,过滤;合并两次滤液,减压浓缩至65℃环境下相对密度为1.20的浸膏,减压干燥,粉碎后过100目筛,得粉末I;
S4:将S1得到的青梅提取物、S2得到柠檬片提取物和S3得到的粉末I充分混匀,减压浓缩至60℃环境下相对密度为1.30的浸膏,添加辅料制备成颗粒剂。
实施例2:
本发明实施例2含有青梅提取物的组合物包括以下重量份数的制备原料:
青梅提取物19份、柠檬片16份、绿豆14份、苦瓜11份、人参11份、枸杞子9份、茵陈13份、栀子9份、郁金香8份和泽泻12份。
制备方法同实施例1。
实施例3:
本发明实施例3含有青梅提取物的组合物包括以下重量份数的制备原料:
青梅提取物15份、柠檬片13份、绿豆11份、苦瓜9份、人参8份、枸杞子7份、茵陈8份、栀子6份、郁金香6份和泽泻9份。
制备方法同实施例1。
对比例1:
本发明对比例1含有青梅提取物的组合物包括以下重量份数:青梅提取物15份、麦冬13份、绿豆11份、苦瓜9份、人参8份、枸杞子7份、茵陈8份、栀子6份、郁金香6份和泽泻9份。

与实施例3的区别在于,将柠檬片替换为麦冬。
对比例2:
本发明对比例2含有青梅提取物的组合物包括以下重量份数:青梅提取物20份、柠檬片13份、绿豆11份、苦瓜9份、人参8份、枸杞子7份、栀子6份、郁金香6份和泽泻9份。
制备方法同实施例3。
与实施例3的区别在于,没有添加茵陈,增加了青梅提取物和柠檬片的用量。
本发明含有青梅提取物的组合物对肥胖大鼠的研究试验
1.试验动物:清洁级健康Sprague-Dawley(SD)雄性大鼠90只,体重75-90g(由广东省医学实验动物中心提供)。随机分为2组,模型构建组80只,正常对照组10只。
2.试验动物模型建立:90只大鼠适应性喂养1周之后,模型构建组80只SD大鼠饲喂高热量饲料(基础饲料55%,猪油15%,奶粉10%,白糖10%,豆芽5%,鸡蛋10%),正常对照组10只SD大鼠喂基础饲料。喂养8周后,高热量饲料喂养的大鼠体重超出基础饲料喂养的大鼠体重20%,即造模成功。最终构建成功72只肥胖大鼠模型,随机选取70只肥胖大鼠,按体重随机分为7组,分别为模型对照组、阳性对照组、实施例1-3组、对比例1-2组,每组10只。
3.试验方法
各组大鼠的给药量如下:
正常对照组:灌胃给予等体积的生理盐水;
模型对照组:灌胃给予等体积的生理盐水;
阳性对照组:汤臣倍健纤巧纤纤胶囊(国食健字G20050006,汤臣倍健股份有限公司),去胶囊壳取内容物用水制成混悬液,取1组肥胖大鼠,按0.6g/kg的剂量每天灌胃1次,连续30天。
实施例1组:将本发明实施例1制得的颗粒剂,用水制成溶液,按2g/kg的剂量每天灌胃1次,连续30天。
实施例2组:试验药物为本发明实施例2制得的颗粒剂,给药方案同实施例1组。
实施例3组:试验药物为本发明实施例3制得的颗粒剂,给药方案同实施例1组。
对比例1组:试验药物为本发明对比例1制得的颗粒剂,给药方案同实施例1组。
对比例2组:试验药物为本发明对比例2制得的颗粒剂,给药方案同实施例1组。
连续灌胃给予相应受试样品30天,给药期间每日记录肥胖型大鼠的给食量、剩食量及撒食量,计算其进食量,观察药物对大鼠食欲的影响。每周定时称量体重1次,试验结束时计算大鼠体重增加量。试验结束时解剖大鼠,取体脂(睾丸及肾周围脂肪垫)并称重,计算体脂比(脂肪垫湿重与体重之比)。
4.试验结果:
1.各给药组对肥胖型大鼠进食量、体重的影响,试验结果如表1所示

注:与模型对照组比较,*P<0.05,**P<0.01,与阳性对照组比较,#P<0.05。
由表1可知,各给药组肥胖型大鼠的食欲稍有下降,但与模型组比较,无显著性差异(P>0.05),说明本发明含有青梅提取物的组合物不是通过降低食欲来实现的。与模型对照组相比,正常对照组大鼠体重增加无显著性差异,而实施例1、2、3组和阳性对照组肥胖大鼠的体重增加缓慢(P<0.05),其中实施例3组体重增加增加最为缓慢(P<0.01)。而对比例1组、对比例2组肥胖大鼠的体重增加量与模型对照组相比无统计学意义(P>0.05)。
2.各给药组对肥胖大鼠体脂比的影响,试验结果如表2所示

注:与正常对照组比较,**P<0.01;与模型对照组比较,#P<0.05,##P<0.01,与阳性对照组比较,^P<0.05。
由表2可知,与正常对照组相比,模型对照组大鼠体脂比明显升高(P<0.01),与模型对照组相比,实施例1、2、3组和阳性对照组肥胖大鼠的体脂比明显降低(P<0.05),其中实施例3组体脂比降低最为明显(P<0.01)。而对比例1组、对比例2组肥胖大鼠的体脂比与模型对照组相比无显著性差异(P>0.05)。
综上所述,本发明含有青梅提取物的组合物对肥胖大鼠的食欲无影响,同时可以显著抑制肥胖大鼠体重的增加及降低其体脂比,具有显著的减肥功能,是一种健康的减肥药物,可用于制备减肥保健品。同时,由试验结果可知,本发明实施例3为本发明最佳实施例。
Claims (7)
1.含有青梅提取物的组合物在制备减肥保健品中的用途,其特征在于,所述含有青梅提取物的组合物包括以下重量份数的制备原料:
青梅提取物13-19份、柠檬片11-16份、绿豆9-14份、苦瓜7-11份、人参7-11份、枸杞子5-9份、茵陈6-13份、栀子4-9份、郁金香3-8份和泽泻6-12份。
2.如权利要求1所述的用途,其特征在于,所述含有青梅提取物的组合物包括以下重量份数的制备原料:
青梅提取物13份、柠檬片11份、绿豆9份、苦瓜7份、人参7份、枸杞子5份、茵陈6份、栀子4份、郁金香3份和泽泻6份。
3.如权利要求1所述的用途,其特征在于,所述含有青梅提取物的组合物包括以下重量份数的制备原料:
青梅提取物19份、柠檬片16份、绿豆14份、苦瓜11份、人参11份、枸杞子9份、茵陈13份、栀子9份、郁金香8份和泽泻12份。
4.如权利要求1所述的用途,其特征在于,所述含有青梅提取物的组合物包括以下重量份数的制备原料:
青梅提取物15份、柠檬片13份、绿豆11份、苦瓜9份、人参8份、枸杞子7份、茵陈8份、栀子6份、郁金香6份和泽泻9份。
5.如权利要求1所述的用途,其特征在于,所述含有青梅提取物的组合物被制成颗粒剂、胶囊剂、丸剂、口服液或片剂。
6.如权利要求1-5任一所述的用途,其特征在于,所述青梅提取物的制备方法包括如下步骤:
称取青梅,洗净后沥干其表面水分,去皮去核后,加入其质量6-8倍量的体积分数为75%的乙醇回流提取4-6小时,过滤,滤渣加4-6倍量的体积分数为75%的乙醇回流提取2-4小时,过滤,将2次滤液合并,减压蒸馏回收乙醇后3000rpm条件下离心15-30分钟,分离上清液,上清液减压浓缩至原体积1/8-1/10,得青梅提取物。
7.如权利要求1-5任一所述的用途,其特征在于,所述含有青梅提取物的组合物的制备方法包括如下步骤:
S1:称取青梅,洗净后沥干其表面水分,去皮去核后加入其质量6-8倍量的体积分数为75%的乙醇回流提取4-6小时,过滤,滤渣加4-6倍量的体积分数为75%的乙醇回流提取2-4小时,过滤,将两次滤液合并,减压蒸馏回收乙醇后3000rpm条件下离心15-30分钟,分离上清液,上清液减压浓缩至原体积1/8-1/10,得青梅提取物;
S2:称取柠檬片,将柠檬片粉碎,加入其质量8-12倍重量的水煎煮2-4小时;过滤,得到粗提液和柠檬片残渣,将粗提液1500-3000rpm条件下离心10-20分钟,分离上清液,上清液减压浓缩至60℃环境下相对密度为1.20-1.25的浸膏,得到柠檬片水提物;将柠檬片残渣,加入4-6倍量体积分数为50-70%的乙醇回流提取1-2次,每次1-2小时,过滤,合并滤液,减压蒸馏回收乙醇后在1500-3000rpm条件下离心10-20分钟,分离上清液,上清液减压浓缩至60℃环境下相对密度为1.20-1.25的浸膏,得到柠檬片醇提物;将柠檬片水提物和醇提物合并混匀,得柠檬片提取物;
S3:称取绿豆、苦瓜、人参、枸杞子、茵陈、栀子、郁金香和泽泻,粉碎,加入药材总重量6-8倍量的水煎煮3-4h,过滤;滤渣加入药材总重量4-6倍量的水煎煮1-2h,过滤;合并两次滤液,减压浓缩至65℃环境下相对密度为1.20-1.25的浸膏,减压干燥,粉碎后过80-100目筛,得粉末I;
S4:将S1得到的青梅提取物、S2得到柠檬片提取物和S3得到的粉末I充分混匀,减压浓缩至60℃环境下相对密度为1.25-1.35的浸膏,添加辅料制备成常见的保健食品。
