Abstract

本发明提供一种梅花鹿用超数排卵聚乙烯吡咯烷酮FSH复合缓释注射剂,将FSH、酸性木糖酶、纤维素酶和庆大霉素四种物质均匀地分散在聚乙烯吡咯烷酮内,形成缓释剂;通过FSH、酸性木糖酶、纤维素酶和庆大霉素等药物的协同增效作用,能使目标物质及间接产生的营养物质源源不断均匀的作用于从卵巢细胞上,从而促使卵泡增长和成熟,从而达到超数排卵的良好效果,成熟的卵子质量优良;该剂型采用静脉注射降低应激反应、减轻疼痛而造成的激烈挣扎。

技术领域
本发明提供一种梅花鹿用超数排卵聚乙烯吡咯烷酮FSH复合缓释注射剂,属于家畜医药制备技术领域。
背景技术
FSH是促卵泡素的英文简称,它是一种糖蛋白。FSH的生理作用是促进卵巢卵泡的生长;促进卵泡颗粒细胞的增生和雌激素的合成和分泌,刺激卵泡细胞上LH受体的产生。FSH被用于母畜的超数排卵。
FSH是促进卵泡发育的激素,而酸性木糖酶和纤维素酶的添加促进了卵泡所需的各种营养成分的释放,上述这些药物只有与聚乙烯吡络烷酮一起制成缓释剂,方可减缓PMSH及酸性木糖酶和纤维素酶释放速度和释放的均一度,以保证卵巢卵泡短时间营养和增长的需要,以达到卵泡同时生长目的而抑制了卵泡优势现象,再佐以促排3号,使这些成熟卵子几乎同时排除,进而达到了超数排卵的目的。上述的材料均是促进母畜排卵妊娠的常用药物,但在实际使用中,运用静脉注射可以把这种缓释剂的作用发挥到很好的水准,如果采用非静脉注射的其他注射法,超数排卵效果不佳。
发明内容
本发明提供一种梅花鹿用超数排卵聚乙烯吡咯烷酮FSH复合缓释注射剂,用于对梅花鹿的排卵进行超数促排。
本发明进一步提供一种梅花鹿用超数排卵聚乙烯吡咯烷酮FSH复合缓释注射剂的制备方法,适用于工业化生产。
本发明所述的一种梅花鹿用超数排卵聚乙烯吡咯烷酮FSH复合缓释注射剂,其特征在于是由以下药物按重量份数比制成的:
FSH:1200~1500国际单位、聚乙烯吡咯烷酮:1.2~1.6克、酸性木糖酶:0.8~1.3克、纤维素酶:0.6~0.9克、庆大霉素:5~8万单位、重蒸馏水21~24毫升。
本发明所述的一种梅花鹿用超数排卵聚乙烯吡咯烷酮FSH复合缓释注射剂的制备方法,包括以下步骤:
按比例称取上述药物,先将FSH和酸性木糖酶和纤维素酶加入聚乙烯吡咯烷酮,再加入重蒸馏水,用充分混匀后,再加庆大霉素,混合均匀制成超数排卵缓释剂。
用法用量:静脉注射;每天注射剂量为2.0~2.5毫升。
本发明将FSH、酸性木糖酶、纤维素酶和庆大霉素四种物质均匀地分散在聚乙烯吡咯烷酮内,形成缓释剂;通过梅花鹿耳缘静脉注射,形成一个相对慢性释放的缓释剂,它在短时间内持续不断地向血液里均匀地释放FSH、酸性木糖酶、纤维素酶和庆大霉素,使这些物质通过血液循环系统运送到身体的各个毛细血管网中,FSH在到达卵巢后会通过卵巢的毛细血管作用在卵巢的未成熟的卵泡上,经过持续不断的作用,它在短时间内迅速使卵巢的卵泡细胞同时大量增殖,大量增殖的卵泡需要从所处的周围环境中获取大量的营养物质包括各种糖分等,酸性木糖酶和纤维素酶通过血液循环作用到消化系统使食物中的营养成分得到充分消化进入血液,通过血液循环给卵泡提供充足的营养,使之迅速成熟有了一定保障,庆大霉素在卵泡周围起到了强效的保健作用,最终对卵巢卵泡的排卵创伤有一定的疗效,这三种物质持续协同作用的结果使卵巢上的卵泡内的卵子大量的同时增长并逐渐成熟。结合使用前列烯醇使卵泡细胞周围的脂肪溶解,激发了成熟卵子脱离卵泡腔的目的,从而达到了良好的超数排卵结果,前列烯醇按照市面销售的按照说明使用即可。
本发明的积极效果在于:通过FSH、酸性木糖酶、纤维素酶和庆大霉素等药物的协同增效作用,能使目标物质及间接产生的营养物质源源不断均匀的作用于从卵巢细胞上,从而促使卵泡增长和成熟,从而达到超数排卵的良好效果,成熟的卵子质量优良;该剂型采用静脉注射降低应激反应、减轻疼痛而造成的激烈挣扎。
具体实施方式
实施例1
将1300国际单位的FSH和0.8克酸性木糖酶和0.9克纤维素酶加入1.2克聚乙烯吡咯烷酮,再加入21毫升重蒸馏水,用充分混匀后,再加5万单位庆大霉素,混合均匀制成超数排卵缓释剂。其释放效果可以保证一天24小时均匀释放该法使用的药物。
实施例2
将1400国际单位的FSH和1.1克酸性木糖酶和0.8克纤维素酶加入1.4克聚乙烯吡咯烷酮(PVP),再加入22毫升重蒸馏水,用充分混匀后,再6万单位加庆大霉素,混合均匀制成超数排卵缓释剂。其释放效果可以保证一天24小时均匀释放该法使用的药物。
实施例3
将1500国际单位的FSH和1.3克酸性木糖酶和0.6克纤维素酶加入1.6克聚乙烯吡咯烷酮,再加入24毫升重蒸馏水,用充分混匀后,再加8万单位庆大霉素,混合均匀制成超数排卵剂。其释放效果可以保证一天24小时均匀释放该法使用的药物。
以下试验例表明本发明对于梅花鹿超数排卵的效果:
试验例
1.材料与方法缓释
生理盐水,75%的酒精棉球,庆大霉素,重蒸馏水,手术刀,剪子,镊子,消毒纱布,兔手术台,1、2、5、10毫升注射器,50毫升及10毫升的烧杯,表面皿,连续变倍实体显微镜。
2.实验设计
把未怀孕的成年母梅花鹿分成三组,每组10只,分别为采用本法的超数排卵组、常法超数排卵组和对照组
3.本发明实验组母梅花鹿的超数排卵处理
在试验第1~3天上午9~10时,分别采用耳缘静脉注射本法的超数排卵缓释剂2.0~2.5毫升,连续3天同一时间进行,在第4天注射一针促排3号,在第5天对供试母梅花鹿麻醉,解剖;对每只梅花鹿的卵巢排卵点计数,然后实施输卵管伞冲胚术,用灭菌烧杯回收卵子,之后再放在表面皿内在显微镜下捡卵,最后得出镜检的卵子数。
常法超数排卵:把FSH直接用重蒸馏水稀释,给每只梅花鹿注射与本法中等量的FHS,整个超数排卵过程同上;而对照组不做任何超数排卵处理。

总结:通过上述3组实施例结果表明:采取本法的超数排卵效果(卵巢排卵数平均值(45、49、47)和镜检卵子平均检出数(39、43、42)明显优于FSH的超排效果,其相应的数据为(12、14、14)和(7、8、7)。本发明各个物质协同配伍能使目标物质及间接产生的营养物质源源不断均匀的作用于从卵巢细胞上,从而促使卵泡增长和成熟,从而达到超数排卵的良好效果,成熟的卵子质量优良;该剂型采用静脉注射降低应激反应、减轻疼痛而造成的激烈挣扎(最好提供相应的数据试验)。

Claims (2)

1.一种梅花鹿用超数排卵聚乙烯吡咯烷酮FSH复合缓释注射剂,其特征在于是由以下药物按重量份数比制成的:
FSH:1200~1500国际单位、聚乙烯吡咯烷酮:1.2~1.6克、酸性木糖酶:0.8~1.3克、纤维素酶:0.6~0.9克、庆大霉素:5~8万单位、重蒸馏水21~24毫升。
2.根据权利要求1所述的一种梅花鹿用超数排卵聚乙烯吡咯烷酮FSH复合缓释注射剂的制备方法,包括以下步骤:
按比例称取上述药物,先将FSH和酸性木糖酶和纤维素酶加入聚乙烯吡咯烷酮,再加入重蒸馏水,用充分混匀后,再加庆大霉素,混合均匀制成超数排卵缓释剂。
