技术领域
本发明涉及细胞生物学和免疫学技术,特别涉及生物制品领域免疫佐剂的体外细胞筛选技术,具体涉及鸡胚成纤维细胞在体外筛选CpG寡聚脱氧核苷酸活性分子中的应用。
背景技术
CpG寡脱氧核苷酸(CpG oligonucleotide,CpG ODN)是含有未甲基化的CpG二核苷酸基序的核苷酸序列,能够在机体内诱生强烈的免疫反应。CpG ODN首先激活Toll样受体9/21(toll-like receptor 9/21,TLR-9/21),而后经一系列信号转导诱导机体B细胞增殖分化,刺激自然杀伤(NK)细胞的免疫保护活性;促进树突状细胞、单核细胞、巨噬细胞等的抗原递呈功能;激发T细胞活性,产生免疫调节级联反应。产生的效应物质中除免疫球蛋白外,还包括Th1和Th2型细胞因子、前炎症细胞因子以及趋化因子,具有显著增强抗原体液免疫和细胞免疫的作用。研究表明,人工合成的含未甲基化CpG基序的CpG ODN和细菌DNA一样,同样具有很强的免疫作用。
CpG ODN的免疫刺激作用具有种属特异性,鸡的最佳刺激基序为5‘-GTCGTT-3’。天然的CpG ODN进入体内和细胞后极不稳定,易受细胞内和血清中核酸水解酶的降解,B型CpGODN半衰期仅5min左右,C型也不过60min,因此在人工合成时多将CpG ODN进行部分或者全链的硫代修饰以防止序列的降解。然而,序列的硫代修饰往往会产生非特异性的细胞毒性作用,影响细胞存活率,干扰试验结果。目前,体外筛选和研究CpG ODN多用外周血分离的单核细胞或者脾脏分离的淋巴细胞,其作用效应显著,但耐受细胞毒性的阈值较低,当CpGODN达到20μg/mL(鸡脾脏细胞浓度为5*106)时即出现细胞毒性的抑制作用,极大限制了CpGODN最优工作浓度的全面筛选。
发明内容
为了解决以上现有技术中淋巴细胞用于CpG ODN体外筛选和研究时耐受细胞毒性的阈值较低,无法全面准确获得最优工作浓度的问题,本发明提供了一种应用耐受力强的鸡胚成纤维细胞(chicken embryo fibroblast,CEF)进行CpG ODN体外筛选的方法。当CpGODN浓度大于80μg/mL(鸡脾脏细胞浓度为5*106)时,可作为宿主免疫应答研究起始点的鸡胚成纤维细胞(chicken embryo fibroblast,CEF)才表现出细胞损伤,增殖率受到影响。基于此,我们以耐受力强的CEF为模型,通过测定细胞增殖和免疫应答效果进行CpG ODN活性分子的筛选,旨在建立利用CEF体外筛选CpG ODN的体系,为体外筛选CpG ODN提供更适合的手段和方法。
本发明的目的是首先确定CpG ODN刺激CEF产生的免疫反应能够正确体现序列结构对其活性的影响。
本发明的另一目的是提供一种优于利用鸡淋巴细胞体外筛选CpG ODN活性分子的新方法。
本发明所述的确定CpG ODN刺激CEF产生的免疫反应能够正确体现序列结构对其活性的影响是通过测定不同序列CpG ODN对CEF的增殖效果和相关免疫基因mRNA表达量的变化来实现的。
本发明是通过以下步骤得到的:
鸡胚成纤维细胞在体外筛选CpG寡聚脱氧核苷酸活性分子中的应用。
所述的应用,具体操作为待鸡胚成纤维细胞生长为单层细胞后与CpG寡聚脱氧核苷酸共孵育,放入细胞培养箱培养。
所述的应用,鸡胚成纤维细胞对不同结构CpG寡聚脱氧核苷酸刺激所产生的增殖效果与淋巴细胞具有相同的规律。
所述的应用,优选CpG寡聚脱氧核苷酸序列为序列表中序列1-9中任一项序列。

本发明的有益效果:
CEF用于CpG ODN的体外筛选不仅避免了核酸序列细胞毒性的干扰问题,对硫代CpG寡聚脱氧核苷酸的耐受浓度为80μg/mL,扩大了CpG ODN的浓度筛选范围,还正确区分体现了CpG ODN的结构与功能,与淋巴细胞相比,其免疫反应更强,更适合于CpG ODN免疫佐剂的体外筛选。

具体实施方式
下面结合具体实施例对本发明进行进一步说明:
实施例1
1.1测定不同序列CpG ODN对CEF的增殖效果。根据CpG ODN的设计原则共设计序列9条,其中A型2条,B型3条,C型3条,阳性、阴性对照各1条,序列如表1,分别对应序列表中的序列1-9。

然后将制备好的CEF密度调整为1*107个/mL,加入96孔细胞板,每孔100μL,置于37℃、5%CO2细胞培养箱中培养24h,使CEF贴壁并生长成单层细胞。将CpG ODN1-9用细胞培养液稀释后分别加入细胞孔中,每孔终体积120μL,CpG工作浓度选择20μg/mL(便于与淋巴细胞比较),轻轻混匀后放入细胞培养箱培养48h。每条CpG ODN重复5孔,同时添加PBS替代CpGODN的阴性对照和无细胞的DMEM空白对照。在培养终止前4h,每个细胞孔加入MTT 10μL,继续孵育4h,取出后弃上层营养液,按100μL/孔加入DMSO,置于摇床上晃动10min,使结晶紫完全溶解。然后酶标仪上测定各孔OD490的值。
各CpG ODN作用于CEF的效果以刺激指数(stimulation index,SI)来衡量,其计算方法为SICpG ODN=(ODCpG ODN-OD空白对照)/(OD阴性孔-OD空白对照)。计算不同序列CpG ODN刺激下CEF增殖样品的OD490平均值,并得到相应的刺激指数。如表2所示,与无活性的CpG ODN9、阴性对照和空白对照相比,CpG ODN1-8均可有效刺激细胞增殖,SI指数接近2,与CpG ODN9差异显著(P<0.05),其中CpG ODN4、CpG ODN6刺激效果最好,指数分别达到2.349、2.376。

CpG ODN序列中凡含CpG基序的均能够刺激CEF的增殖,CpG ODN9因序列中CG碱基倒置而丧失刺激活性,甚至低于阴性对照值。CpG ODN6和CpG ODN4刺激指数最高,其序列内部均含有回文序列,且序列5’端以TCG开头,不同之处在于CpG ODN6的回文序列在5’端且包含4个CpG基序,而CpG ODN4回文序列靠近中部且只含3个CpG基序,所以CpG ODN6的活性强于CpG ODN4。CpG ODN5也含有回文序列,与CpG ODN4不同在于回文序列中含GTCGTT,但刺激活性并未增强,回文序列中的GTCGTT不起促进作用。CpG ODN1即CpG2006,其对鸡的淋巴细胞具有很好的刺激作用,作用于CEF的刺激效果也很明显,与淋巴细胞反应一致。CpG ODN2比CpG ODN1减少一组GTCGTT,其活性也相应降低。多个TCG重复序列有助于增强CpG ODN对B细胞和NK细胞的刺激作用,CpG ODN3即为7个TCG的简单重复,具有良好的刺激活性,这与已有的淋巴细胞研究结果一致。磷酸骨架的CpG ODN序列3’端的poly G尾能与单核细胞表面的清道夫受体结合,它所形成的四聚体结构可以使CpG ODN的结合和吞入过程变得更加容易,CpG ODN7和CpG ODN8的序列3’端含有poly G尾,虽然未以TCG开头,但其刺激活性依然有所增强,高于CpG ODN1。此外,CpG ODN7相比CpG ODN8序列中还加入了12bp的回文序列,对活性也有较好的提升作用。可见,CEF对不同序列CpG ODN刺激所产生的增殖效果与淋巴细胞具有相同的规律,具有很好的区分度,准确反应了CpG ODN的结构特征。
2.2测定不同序列CpG ODN刺激CEF相关免疫基因mRNA表达量的变化。荧光定量检测目的基因有IL-2、IL-6、IL-12、IFN-α、IFN-γ和TLR-21,β-Actin为内参基因,引物序列如表3,分别对应序列表中的序列10-23。

CpG ODN刺激CEF的操作同CEF增殖试验,同时设置PBS未处理组对照。待作用24h后,取出细胞反复冻融3次,使细胞从板上脱落,吸出后进行RNA的提取。应用Trizol法提取细胞的总RNA,然后进行反转录。向11.5μL的RNA溶液中加入1μL 25pmol/L的Oligo(dT)18,70℃保温10min后,冰上急冷2min。然后加入5*M-MLV Buffer 4μL,dNTP Mixture(10mM)2μL,RTase M-MLV 0.5μL,RNase Inhibitor(RRI)1μL,反应条件为:室温10min,42℃1h,70℃15min。获得反转录一链用于荧光定量PCR的检测。检测采用相对定量方法,以β-actin为内参基因。7个基因PCR反应条件相同,95℃预变性2min;95℃10s,56℃15s,72℃15s,共45个循环;95℃15s,60℃1min,每个样品3个重复。使用-ΔΔCt方法计算基因表达量变化倍数。
Th细胞因子IL-2、IL-12、IL-6在不同CpG ODN的刺激下表达量均有所升高。在CpGODN4和CpG ODN6的作用下,IL-2表达量提升25倍以上,除阴性对照(CpG ODN9)外,与其他组相比差异显著(P<0.05);CpG ODN1与CpG ODN5作用相当,提升17倍左右;CpG ODN2、CpGODN3、CpG ODN7则在10倍上下(图1)。IL6和IL-12的表达量变化相似:在CpG ODN4、CpG ODN6的作用下变化最大,与其他组差异显著(P<0.05);B型CpG ODN作用强于强A型,但差异不显著;A型CpG ODN作用较弱(图2、图3)。
不同序列的CpG ODN对IFN-α的表达均有明显的上调作用,A型和C型CpG ODN(除CpG ODN 5)作用最强,上调至少50倍,与其他组差异显著(P<0.05),其中CpG ODN7和CpGODN8作用最强,约为B型CpG ODN的2倍(图4)。CpG ODN4和CpG ODN6能够上调IFN-γ表达量近1000倍,与CpG ODN1、CpG ODN2、CpG ODN5、CpG ODN7和CpG ODN8差异显著(P<0.05),与CpG ODN3差异极其显著(P<0.01)(图5)。
作为CpG ODN的受体,TLR-21的表达量明显上调,CpG ODN6升高最多,在150倍以上,其次为CpG ODN4,CpG ODN1、CpG ODN2、CpG ODN5则上调100倍左右,与剩余序列差异显著(P<0.05);CpG ODN8上调最少,为25倍。(图6)
A型CpG ODN主要作用于树突状细胞,分泌IFN-α,但不作用于Β细胞;B型CpG ODN作用于B细胞,分泌IgM、IL-6等,促进树突状细胞成熟;C型CpG ODN则能够分别作用于树突状细胞和Β细胞,所以由活性T细胞表达的IL-2,C型诱导的表达量要高于B型而显著高于A型,回文序列的存在作用显著,但CpG ODN5回文序列内部的GTCGTT序列未起促进作用。此外,B型表达量高于A型的另一原因是CpG ODN进行了硫代修饰,作用于B细胞的能力增强而对DC细胞的作用很弱。对于IL-6和IL-12,C型和B型CpG ODN能够有效增强B细胞活性而提高其表达量,但A型序列则不能,说明C型和B型CpG ODN能够直接作用于表达TLR-21分子的B细胞。但从各型CpG ODN内部不同序列刺激结果来看,刺激单元数量和回文序列位置有增强活性的作用,与IL-2情况类似,CpG ODN5回文序列内部的GTCGTT序列作用不明显。
A型和C型CpG ODN能够诱导大量Ⅰ型干扰素的产生,前者因polyG的存在,而后者则是回文序列的作用。实验结果表明,部分硫代修饰的A型CpG ODN能够直接作用于树突状细胞,而C型CpG ODN则能够促进单核细胞向树突状细胞转化,加速成熟单核细胞转化成为功能性树突状细胞,引发强烈抗原特异性细胞免疫。各型CpG ODN不同序列差异不显著。
C型CpG ODN能够有效刺激NK细胞产生IFN-γ,尤其以CpG ODN4和CpG ODN6刺激效果最强,提升表达量接近1000倍,与其他序列产生显著差异,这与Ballas等在小鼠淋巴细胞上获得的结论一致,原因在于C型CpG ODN被TLR-21阳性细胞所识别,分泌了大量的IL-6或者IL-12,刺激活化了NK细胞,从而产生大量的IFN-γ。
具有免疫活性的CpG ODN能够有效刺激TLR-21的表达,并引发机体的固有和获得性免疫反应。作为CpG ODN进入禽类细胞和机体唯一的通路,TLR-21表达量多少能够反应后续免疫反应的强弱。CpG ODN1-8作用于细胞后,TLR-21表达量明显上调,表明A、B、C三种类型CpG ODN均能够激活TLR-21信号通路,其中以C型CpG ODN作用最为明显,A型最弱,B型居中,这与他们所作用的不同细胞类型和识别激活模式相关。C型CpG ODN兼具A型、B型的作用,免疫反应相对快速、强烈,TLR-21表达量也证实了这一点。
可见,CEF受到不同序列CpG ODN刺激后免疫因子的表达量变化能够准确反应CpGODN的结构差别,并与淋巴细胞的变化规律相同。
本发明所述的优于现行操作的体外筛选CpG ODN的新方法是通过CEF代替淋巴细胞进行CpG ODN的体外筛选,并与淋巴细胞方法进行比较实现的。利用CEF代替淋巴细胞进行CpG ODN的体外筛选,除了能够避免核酸序列修饰的细胞毒性对研究的影响、实现CpGODN高浓度区间最优工作浓度进行筛选外,还能正确体现不同序列CpG ODN的活性区别。CEF刺激指数准确反映了CpG ODN的结构特征,只是刺激指数较淋巴细胞偏低一些,但对于CpGODN活性的区分度良好。
通过对CpG ODN刺激细胞的相关免疫基因的表达量的测定,以CpG ODN1为例,IL-6、IL-12、IFN-α、IFN-γ、TLR-21表达量在刺激作用后分别上调了25倍、30倍、48倍、960倍、110倍。而在已报道相关研究中,涉及CpG2006或者CpG2007刺激细胞或机体后相关细胞因子的变化倍数中,最多上调倍数为IL-6(20倍)、IL-12(6倍)、IFN-α(4倍)、IFN-γ(1000倍)、TLR-21(100倍)。可知,CEF各检测基因的表达增加量高于或接近于淋巴细胞的增加量,表明CEF受到CpG ODN刺激后引发的免疫反应强于淋巴细胞。所以,在刺激细胞因子表达量变化方面,CEF可视为体外筛选CpG ODN的“强化版淋巴细胞”。
实施例2
将制备的CEF密度调整为5*106个/mL,加入96孔细胞板,每孔100μL,置于37℃、5%CO2细胞培养箱中培养24h,使CEF贴壁并生长成单层细胞。将CpG ODN2006用细胞培养液稀释成不同浓度后加入不同细胞孔中,工作浓度分别为400μg/mL、200μg/mL、150μg/mL、100μg/mL、80μg/mL、50μg/mL、30μg/mL、20μg/mL、10μg/mL、5μg/mL、2.5μg/mL,每个浓度重复6孔,每孔终体积200μL,同时添加PBS替代CpG ODN的阴性对照。轻轻混匀后放入细胞培养箱继续培养72h。在培养终止前4h,每个细胞孔加入MTT 10μL,继续孵育4h,取出后弃上层营养液,按100μL/孔加入DMSO,置于摇床上晃动10min,使结晶紫完全溶解。然后酶标仪上测定各孔OD570的值。各CpG ODN浓度对CEF的毒性以细胞增殖率来衡量,其计算方法为:增殖率(100%)=(OD试验组-OD对照组)/OD对照组*100%,以阴性对照值为参照,低于阴性对照值表示产生细胞毒性。
通过计算各浓度硫代CpG ODN作用下CEF的增殖率可知,随着浓度的增大,增值率先上升后下降:当浓度处于2.5-20μg/mL之间时,对CEF刺激效果良好,增殖率持续上升,至20μg/mL时达到最高值;当浓度大于20μg/mL时,增殖率呈下降趋势,达到80μg/mL时,细胞增殖率为3.32%,而浓度增加至100μg/mL时增殖率为负数,表明在此浓度下CEF未增殖,表现出一定程度的细胞毒性作用;浓度继续增大时,增殖率进一步降低。结果提示,CEF对硫代CpG ODN的耐受浓度为80μg/mL。
综上所述,CEF用于CpG ODN的体外筛选不仅避免了核酸序列细胞毒性的干扰问题,扩大CpG ODN的浓度筛选范围,还正确区分体现了CpG ODN的结构与功能,与淋巴细胞相比,其免疫反应更强,结合自身的诸多优点,更适合于CpG ODN免疫佐剂的体外筛选。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受实施例的限制,其它任何未背离本发明的精神实质与原理下所做的改变、修饰、组合、替代、简化均应为等效替换方式,都包含在本发明的保护范围之内。

Claims (8)
1.一种鸡胚成纤维细胞在体外筛选CpG寡聚脱氧核苷酸活性分子中的应用,鸡胚成纤维细胞对硫代CpG寡聚脱氧核苷酸的耐受浓度为80μg/mL。
2.根据权利要求1所述的应用,其特征在于待鸡胚成纤维细胞生长为单层细胞后与CpG寡聚脱氧核苷酸共孵育,放入细胞培养箱培养。
3.根据权利要求1或2所述的应用,其特征在于鸡胚成纤维细胞对CpG寡聚脱氧核苷酸刺激所产生的增殖效果与淋巴细胞具有相同的规律。
4.根据权利要求1或2所述的应用,其特征在于CpG寡聚脱氧核苷酸序列为序列表中序列1-9中任一项序列。
5.根据权利要求1或2所述的应用,其特征在于CpG寡聚脱氧核苷酸工作浓度20μg/mL。
6.根据权利要求1或2所述的应用,其特征在于鸡胚成纤维细胞受到CpG寡聚脱氧核苷酸刺激后引发的免疫反应强于淋巴细胞。
7.根据权利要求1或2所述的应用,其特征在于CpG寡聚脱氧核苷酸刺激鸡胚成纤维细胞后,免疫基因的表达增加量高于或接近于使用淋巴细胞的表达增加量。
8.根据权利要求7所述的应用,其特征在于免疫基因为IL-2、IL-6、IL-12、IFN-α、IFN-γ或TLR-21基因。
